In-situ extrusion 3D printing with tea polyphenol crosslinking for Hyaluronic acid sodium salt -based composite hydrogel scaffolds

Reyihanguli Alimu,Lanlan Dong,Patiguli Aihemaiti,Houfeng Jiang,Wurikaixi Aiyiti,Cijun Shuai

High-performance hydrogel biomaterials hold considerable promise for advanced wound care. However, the suboptimal mechanical properties of conventional hydrogel materials limit their practical application. In this study, Hyaluronic acid sodium salt (HA), xanthan gum (XG), and N-acryloyl-glycinamide (NAGA) hydrogels with porous structures were successfully fabricated using in-situ extrusion 3D printing technology, and a functionalization strategy involving tea polyphenol (TP) immersion was proposed to enhance material properties through additional hydrogen bonding. Systematic optimization of TP concentration and immersion time led to significant improvements in the resulting hydrogel: tensile strain increased from 95 % to 266 %, tensile stress improved 6.8-fold, and compressive stress increased 10-fold, with an elastic modulus of 0.68 MPa and toughness of 1.2 MJ/m³ . Furthermore, TP-treated hydrogels exhibited excellent biocompatibility and strong antioxidant activity and antibacterial activity. This study provides a simple yet effective secondary functionalization approach for enhancing the performance of 3D-printed hydrogels, offering a promising approach for multifunctional hydrogel biomaterials with integrated mechanical strength and biocompatibility. These findings open new avenues for advanced wound care systems and personalized medicine applications.

Tailored Iron–Polyphenol interfacial networks in bio-based aerogels for solar-driven desalination with life cycle assessment Insights

Yuan Tian,Pan Wu,Xiong Yang,Yafei Shi,Gary Owens,Kewu Pi

Tailoring interfacial architectures in porous photothermal materials is critical for advancing solar-driven desalination technologies. Herein, we present a fully bio-based aerogel engineered through an iron-polyphenol coordination strategy inspired by the traditional Chinese “iron-tannin dyeing” method. Liquid-phase tannic acid (TA) extract and solid-phase cellulose from Chinese gallnut were co-assembled with sodium alginate, followed by Fe 3+ /Ca 2+ dual-ion crosslinking and freeze-drying, yielding a TA@SA@Fe 3+ aerogel with hierarchical porosity and adaptive interfacial characteristics. The tailored coordination network enhances light absorption (95.7 %), hydrophilicity, and a high solar evaporation rate of 2.0 kg·m –2 ·h –1 under 1 sun illumination. Its interconnected pores promote salt diffusion, ensuring long-term outdoor operation. Furthermore, the material exhibits strong adhesion on diverse substrates—including melamine sponges and geotextiles—supporting modular integration into scalable desalination systems. A comprehensive life cycle assessment (LCA) confirms the sustainability advantage of this system, with a total emission of only 2.4  kg CO 2 -equivalent per kilogram of material, significantly lower than conventional alternatives. This work offers a green, cost-effective, and application-ready strategy for designing interfacial aerogels, providing a promising platform for next-generation solar-thermal water purification technologies.

Microwave and enzyme co-assisted aqueous two-phase extraction of polyphenol and lutein from marigold (Tagetes erecta L.) flower

Xiao-Qian Fu,Na Ma,Wei-Peng Sun,Yan-Yan Dang

In this study, a novel method of simultaneous extraction and enrichment of total polyphenol (TP) and lutein from marigold flower was developed by microwave and enzyme co-assisted aqueous two-phase extraction (ATPE) (MEAATPE). In this study, ethanol/ammonium sulfate was chosen to construct aqueous two-phase system (ATPS) due to its fine partition and recycling behaviors. The optimal MEAATPE condition was obtained, that is, 28% (w/w), ethanol/20% (w/w) ammonium sulfate, 0.45 U/g enzyme concentration, enzymolysis time of 150 min, enzymolysis temperature of 45 °C, microwave time of 120 s, and microwave power of 270 W, by investigating the effects of ATPS composition and other factors on the yield of target products. Under this optimal condition, the yields of TP and lutein reached 84.61 and 7.32 mg/g with the corresponding recovery rates of 95.35% and 99.85%, respectively. Compared with enzyme-assisted ATPE, microwave-assisted ATPE, ATPE, and Soxhlet extraction (SE), the extraction yield of TP by MEAATPE was the highest, which is 71.14% higher than that of SE. Scanning electron microscopy analysis further showed an optative pretreatment effect of marigold flower powders. Thus, MEAATPE was an efficient, rapid, and environmental-friendly method used for the extraction of natural products.

Polyphenol from millet bran increases the sensitivity of colorectal cancer cells to oxaliplatin by blocking the ganglioside GM3 catabolism

Xiaoli Zhang,Shuhua Shan,Jiangying Shi,Hanqing Li,Zhuoyu Li

Colorectal cancer (CRC) is an aggressive malignancy with very limited therapeutic approaches. Drug resistance develops as a frequent characteristic in many patients with CRC, which leads to a decrease in the therapeutic efficacy of anticancer agents. Our previous evidences showed that bound polyphenol from millet bran (BPIS) possesses the potential of inhibiting cancer cell proliferation, and its main anticancer components are ferulic acid (FA) and p-coumaric acid (p-CA). In the present study, we found that BPIS significantly increases the sensitivity of human drug-resistant CRC cell line to oxaliplatin (OXA), a commonly used chemotherapy drug against CRC. Mechanistically, we indicated that BPIS significantly impairs the expression of a gene encoding multidrug resistance protein 1 (MDR1), a well-known permeability glycoprotein (P-gp), by preventing ganglioside GM3 catabolism. Neuraminidase 3 (NEU3) is a key enzyme catalyzing the conversion of ganglioside GM3 to ceramide trihexosides (Gb3), whose expression is increased in drug-resistant HCT-116/L cells. BPIS treatment increased GM3 level, but reduced Gb3 and P-gp levels by inhibiting NEU3 expression, which subsequently boosted the chemotherapy sensitivity of drug-resistant HCT-116/L cells to OXA. These findings reveal that BPIS increases the chemo-sensitivity by remodeling NEU3-mediated ganglioside GM3 catabolism, and it may be applied as a novel drug for facilitating the effectiveness of chemotherapeutic agents in CRC.

Exploration of Natural Protein–Polysaccharide–Polyphenol Ternary Complexes from Grape Pomace for Clean-Label Pickering Emulsions Through pH Adjustment

Zixuan Du,Zhengyang Jia,Jianyu Yang,Yue Zhao,Jiachen Zang,Guanghua Zhao

Grape pomace represents a major organic solid waste generated by the wine industry, but its application has been largely unexplored. On the other hand, so far, stable and sustainable raw materials for producing stable, edible Pickering emulsifiers suitable for the food industry have been lacking. To solve these problems, this study established a mild but effective co-extraction method to obtain protein–polysaccharide–polyphenol ternary complexes (GPTCs) from grape pomace. Subsequently, these complexes were directly developed into an edible Pickering emulsifier by a pH-controlled method. Results showed that a series of properties related to the Pickering emulsifier, such as particle size, surface charge, wettability, and interfacial adsorption behavior, could be easily controlled by adjusting the solution’s pH. Consequently, the GPTC prepared at pH 7.0 exhibited optimal emulsifying performance. The resulting particles had an average particle size of approximately 111 nm, and stabilized Pickering emulsions with a volume-weighted mean oil droplet diameter (D [4,3]) of 9.49 μm, indicating high emulsion stability. Collectively, this study provided an actionable approach for the green, high-value utilization of wine byproducts by establishing a pH-responsive design framework for edible Pickering emulsifiers.

Sequential high hydrostatic pressure and polyphenol oxidase treatments modulate the structure, digestion behavior, and antibody-binding immunoreactivity of amandin

Weiman Gao,Jingyue Zhang,Lining Ren,Shijing Jiang,Xinyu Ma,Guangjiao Ru,Zihan Lin,Yuhang Zhang,Aidong Sun

BACKGROUND Amandin (Pru du 6), a major almond allergen, exhibits immunochemical recognition that is influenced by protein conformation and the accessibility of antibody-recognized regions. This study investigated whether processing sequence governs the effects of high hydrostatic pressure (HHP) and polyphenol oxidase (PPO) on the structural, digestive, and commercial antibody-based immunoreactivity of amandin. RESULTS HHP and PPO conditions were optimized before individual and sequential treatments were compared. Among the tested pressures, 400 MPa produced the lowest commercial antibody-based relative immunoreactivity and was selected for the sequential experiments. HHP pretreatment followed by PPO treatment produced the greatest reduction in antibody-binding immunoreactivity and the most pronounced accumulation of high-molecular-weight material. Spectroscopic and physicochemical analyses showed altered secondary structure, fluorescence behavior, surface hydrophobicity, and sulfhydryl accessibility. Molecular docking supported a potential non-covalent PPO–amandin interaction but was not interpreted as evidence of aggregate formation. HHP-PPO treatment also generated a more interconnected aggregate morphology, increased radical-scavenging activity, and maintained comparatively low antibody-binding immunoreactivity during simulated gastrointestinal digestion despite a lower degree of hydrolysis. CONCLUSION Processing sequence was a key determinant of amandin structural remodeling and antibody-binding immunoreactivity. The observed structural changes are consistent with HHP pretreatment influencing subsequent PPO-mediated association, although the exact intermolecular linkages and antibody-recognized regions were not determined. Because the enzyme-linked immunosorbent assay used commercial anti-almond-protein antibodies rather than patient immunoglobulin E, further validation with allergic sera and biological models is required before conclusions regarding allergenicity can be made. © 2026 Society of Chemical Industry.

Developing Metal–Polyphenol Network-Based Single-Protein Particle Encapsulation System for Sustained Release of Whey Protein to Mitigate Sarcopenia

Yafei Zhang,Yuning Zhang,Yongjian Ai,Yao Lu,Heng Quan,Zhen Zhang,Yuqi Wang,Yujia Luo,Xiaoxu Zhang,Yingying Lin,Sijia Song,Huiyuan Guo

Sarcopenia has emerged as a major health challenge in the aging population. Whey protein (WP)—rich in branched-chain amino acids—is an important nutritional supplement for mitigating sarcopenia. However, the rapid digestion of WP limits its therapeutic efficacy. Developing a sustained-release system for WP may enhance its effectiveness in mitigating sarcopenia. Herein, a novel “single-protein particle encapsulation” (SPPE) system is developed to protect WP and control its release. Specifically, the “molecular glue” metal–polyphenol networks (MPNs) are assembled onto the surface of individual polymerized WP isolate (PWPI) particles as an intermediate layer and bridged with the saccharide hyaluronic acid (HA) to construct PWPI@MPNs/HA. The results showed that PWPI@MPNs/HA precisely regulates the release rate of PWPI in the gastrointestinal tract. In a murine sarcopenia model, PWPI@MPNs25/HA significantly improves lean body content, grip strength, grid hanging time, exhaustion time, and exhaustion distance. The underlying molecular mechanism involves activation of the PI3K/AKT/mTOR signaling pathway and suppression of the muscle atrophy factors MuRF-1 and Atrogin-1 in the ubiquitin–proteasome system via PWPI@MPNs/HA. The SPPE system developed in this study may have broad future applications in protein protection and delivery.

A systematic study of green wheat kernel starch: Multi-scale structure, physicochemical properties, polyphenol binding and Pickering emulsifying capacities

Benguo Liu,Shijie Zhang,Qiqi Liu,Kangyi Zhang

This study explored the structural and physicochemical properties of green wheat kernel starch (GWKS) and their effect on Pickering emulsion performance with polyphenol complexes. Compared to mature wheat starch (WS), GWKS had lower amylose content (28.20 ± 0.27%), greater branching (4.91%), and longer amylopectin chains, resulting in more double-helix structures but lower crystallinity. These traits gave GWKS better swelling power and thermal stability but weaker gel strength and slower retrogradation. For polyphenol-starch complexes, tannic acid (TA) bound strongly to GWKS, improving interfacial wettability (contact angle ∼90°), while gallic acid (GA) and epigallocatechin gallate (EGCG) failed to stabilize GWKS-based emulsions. GWKS-TA emulsion showed superior stability, with fine droplets (D 50  = 24.19 μm) and a strong elastic gel network, outperforming WS-based emulsions. The structural predominance of the branched-chain network in immature starch governs its intrinsic properties and underpins the enhanced stability of GWKS-TA emulsions, offering new pathways for advanced processing and high-value applications.

Facile synthesis of polyphenol-based spherical adsorbent for simultaneous adsorption and reduction of hexavalent chromium in an aqueous environment

Jianxue Li,Kun Zhao,Ke Meng,Lei Tan,Zhimin Chen,Jianfu Chen,Shaohua Liu,Jianwei Fu

Polyphenol-based materials have been regarded as a promising type of adsorbent due to their potential to simultaneously adsorb and reduce Cr(VI) in water. Herein, we report a new tea polyphenol-gallic acid-based adsorbent (TPGA), which can be facilely synthesized via polymerization using tea polyphenols and gallic acid as comonomers, and histidine and formaldehyde as crosslinking agents. Under the optimum conditions of an adsorbent dosage of 0.25 g L -1 , an initial solution pH of 2.0, and a contact time of 600 min, TPGA exhibited a maximum adsorption capacity of 408 mg g −1 for Cr(VI) at 298 K. A removal rate of over 89 % for Cr(VI) at a concentration of 100 mg L -1 was achieved. The adsorption kinetics of Cr(VI) conformed to the pseudo-second-order model and the adsorption isotherm data was in line with the Langmuir model. The adsorption process was spontaneous, endothermic, and accompanied by an increase in entropy. Multiple analysis including Zeta potential, FTIR, and XPS demonstrated that the rich phenolic hydroxyl groups on the surface of TPGA served as electron donors to reduce 63.92 % of Cr(VI) to Cr(III), and the excess hydroxyl groups could coordinate with the generated Cr(III), achieving concurrent adsorption and reduction of chromium pollutants in water.

Hemoglobin-decorated metal polyphenol network platform featuring antibacterial, antioxidant, and oxygen carrying properties for promoting infected diabetic wound healing

Haonan Hu,Guitao Tan,Huan He,Qinqin Zhang,Jing Tu

Chronic wounds have emerged as a formidable challenge in diabetes management, leading to high morbidity and mortality. The diabetic wound healing is often impaired due to recurrent bacterial infections, excessive reactive oxygen species (ROS), and severe hypoxia. Thus, we developed a multifunctional nanoplatform that exhibited remarkable antibacterial activity, ROS scavenging and oxygen-carrying properties. Metal-phenol network (MPN) as core material was synthesized by assembling tannic acid (TA) with Fe 2 ⁺ via coordination and hydrophobic interactions. Subsequently, hemoglobin (Hb) was decorated onto MPN surface through cation-π interactions. MPN protected Hb against oxidation and preserved its physiological function, facilitating oxygen transport. Moreover, MPN-Hb displayed robust and broad-spectrum antioxidant activity that can effectively scavenge ROS and reactive nitrogen species. Under near-infrared (NIR) irradiation, MPN-Hb demonstrated excellent bactericidal activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), effectively eradicated biofilms through hyperthermia. In vivo studies have shown that MPN-Hb with good biocompatibility could effectively eliminate bacteria at the wound site, mitigate the inflammatory response, and promote collagen deposition and angiogenesis, thereby accelerating wound healing. Overall, this multifunctional nanoplatform offers a straightforward, safe, and efficient therapeutic strategy for treating chronic diabetic wounds.

The dietary polyphenol Resveratrol attenuates High-Fat Diet-induced testicular dysfunction via modulating the Gut-Microbiota-Testis metabolic axis

Fujia Chen,Siqiang Li,Shuaihui Jia,Mengjie Lu,Lumei Dai,Shuang Guo,Xiyu Hu,Hongwei Guo,Enzhong Li

Resveratrol, a dietary polyphenol from grapes and berries, was evaluated for its protection against high-fat diet (HFD)-induced testicular dysfunction via the gut-microbiota-testis axis. In mice, resveratrol improved sperm quality and systemic metabolism. It reshaped gut microbiota (enriching Bifidobacterium) and normalized testicular metabolites, including the antioxidant 1,4-dithiothreitol and membrane-stabilizing heptadecanoic acid. Crucially, network pharmacology predicted specific targets, TNF-α, IL-1β, PTGS2, HRAS, and the TCA cycle enzyme SDHB, which led us to hypothesize and validate that resveratrol enhances mitochondrial TCA cycle activity (upregulating SDHB, ACO2, CS, MDH2). Concurrently, it suppressed the NF-κB pathway and promoted repair processes. Molecular docking confirmed stable binding to these targets. This work demonstrates that dietary resveratrol alleviates testicular impairment, a mechanism pinpointed through computational prediction, supporting its role as a nutritional intervention for male subfertility.

Protein co-assembled silk fibroin-whey protein isolate hydrogels mediated by metal-polyphenol coordination: Formation mechanism and antimicrobial activity

Hongyan Fang,Chang Liu,Yibo Liu,Qian Li,Jingqian Wang,Feng Jin,Ning Liu

The advancement of functional materials through the utilization of food proteins has increasing attention within the realm of food science and technology. In this research, it was reported that two food-grade namely silk fibroin (SF) and whey protein isolate (WPI), are capable of coagulating under physical action. They were conjugated with oxidized guar gum (OGG), epigallocatechin gallate (EGCG), and silver ion (Ag + ) to construct a hydrogel system. The dynamic Schiff base linkage between the amino group of SF-WPI complex and the aldehyde group of OGG, along with the metal-polyphenol coordination between Ag + and EGCG, facilitated the formation of the hydrogel. The findings regarding the antibacterial activity of the hydrogel demonstrated that it exerted antibacterial effects against Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus . Moreover, the hydrogel in this research also exhibited remarkable antioxidant capacity within the cell model, along with excellent adhesive and injectable properties. The integration of protein co-assembly, dynamic covalent cross-linking, and metal coordination provides a novel approach for the fabrication of multifunctional food-protein-based hydrogels. The results of this study indicate that this hydrogel holds potential applications in food-contact antibacterial materials or biomedical domain.
