Exploration of the Microstructure and Rheological Properties of Sodium Alginate-Pectin-Whey Protein Isolate Stabilized Β-Carotene Emulsions: To Improve Stability and Achieve Gastrointestinal Sustained Release

Haoxin Ye,Tingshuai Chen,Min Huang,Gerui Ren,Qunfang Lei,Wenjun Fang,Hujun Xie

Sodium alginate (SA)-pectin (PEC)-whey protein isolate (WPI) complexes were used as an emulsifier to prepare β-carotene emulsions, and the encapsulation efficiency for β-carotene was up to 93.08%. The confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM) images showed that the SA-PEC-WPI emulsion had a compact network structure. The SA-PEC-WPI emulsion exhibited shear-thinning behavior and was in a semi-dilute or weak network state. The SA-PEC-WPI stabilized β-carotene emulsion had better thermal, physical and chemical stability. A small amount of β-carotene (19.46 ± 1.33%) was released from SA-PEC-WPI stabilized β-carotene emulsion in simulated gastric digestion, while a large amount of β-carotene (90.33 ± 1.58%) was released in simulated intestinal digestion. Fourier transform infrared (FTIR) experiments indicated that the formation of SA-PEC-WPI stabilized β-carotene emulsion was attributed to the electrostatic and hydrogen bonding interactions between WPI and SA or PEC, and the hydrophobic interactions between β-carotene and WPI. These results can facilitate the design of polysaccharide-protein stabilized emulsions with high encapsulation efficiency and stability for nutraceutical delivery in food and supplement products.Keywords:β-carotene;polysaccharide;protein;encapsulation;emulsion

An injectable and adaptable system for the sustained release of hydrogen sulfide for targeted diabetic wound therapy by improving the microenvironment of inflammation regulation and angiogenesis

Hao Zhang,Xianzhen Dong,Yuhang Liu,Ping Duan,Changjiang Liu,Kun Liu,Yifeng Yu,Xinyue Liang,Honglian Dai,Aixi Yu

The combined effects of persistent chronic inflammation, oxidative stress, microcirculation disorders, and dysregulated cellular energy metabolism often hinder the repair of diabetic skin wounds. Traditional treatment methods are typically insufficient in simultaneously addressing these complex factors, resulting in delayed wound healing and a high propensity for recurrence and chronic ulceration. This study developed an innovative strategy based on reactive oxygen species (ROS)–responsive nanoparticles loaded with an ultraviolet (UV)-light-responsive hydrogen sulfide (H 2 S) donor. This approach leverages the endogenous ROS present in diabetic wounds and external UV light as dual triggers to facilitate the controlled and stepwise release of H 2 S. The material design explicitly targets the critical challenges in diabetic wound repair, including the inhibition of chronic inflammation, oxidative stress reduction, microcirculation improvement, and support of cellular energy metabolism, thereby significantly accelerating wound healing. This adaptive release of signaling molecules effectively modulates the wound regeneration microenvironment, enhancing the repair process and offering a promising solution for diabetic skin wound management. Temperature-sensitive polyurethane (TSPU) film incorporated with carvacrol and cinnamyl aldehyde: antimicrobial activity, sustained release kinetics and potential use as food packaging for Cantonese-style moon cake

Hao Dong,Jiapeng He,Kaijun Xiao,Chao Li

Summary Temperature-sensitive polyurethane (TSPU) films incorporated with carvacrol and cinnamyl aldehyde were prepared for the potential use of food packaging. The antimicrobial properties and sustained release kinetics of carvacrol and cinnamyl aldehyde in TSPU film were investigated. Results indicated that cinnamyl aldehyde and carvacrol had favourable antimicrobial properties at relatively low addition ratio. The diffusion and release of carvacrol were linearly related to temperature and its addition ratio. The release rates of carvacrol from TSPU film increased from 0.6% to 2.2% with the increase of addition ratio and temperature. The first-order kinetic equation could be used to describe its diffusion and sustained release process. TSPU films could significantly prolong the shelf life of Cantonese-style moon cakes by effectively inhibiting microbial growth and decreasing lipid oxidation comparing with commonly used polyethylene food packaging. Results obtained in the present work can provide technical guide of sustained release food packaging films with antimicrobial properties.

Development of a novel resin-based dental material with dual biocidal modes and sustained release of Ag+ ions based on photocurable core-shell AgBr/cationic polymer nanocomposites

Cao Weiwei,Zhang Yu,Wang Xi,Chen Yinyan,Li Qiang,Xing Xiaodong,Xiao Yuhong,Peng Xuefeng,Ye Zhiwen

Abstract Research on the incorporation of cutting-edge nano-antibacterial agent for designing dental materials with potent and long-lasting antibacterial property is demanding and provoking work. In this study, a novel resin-based dental material containing photocurable core-shell AgBr/cationic polymer nanocomposite (AgBr/BHPVP) was designed and developed. The shell of polymerizable cationic polymer not only provided non-releasing antibacterial capability for dental resins, but also had the potential to polymerize with other methacrylate monomers and prevented nanoparticles from aggregating in the resin matrix. As a result, incorporation of AgBr/BHPVP nanocomposites did not adversely affect the flexural strength and modulus but greatly increased the Vicker’s hardness of resin disks. By continuing to release Ag + ions without the impact of anaerobic environment, resins containing AgBr/BHPVP nanoparticles are particularly suitable to combat anaerobic cariogenic bacteria. By reason of the combined bactericidal effect of the contact-killing cationic polymers and the releasing-killing Ag + ions, AgBr/BHPVP-containing resin disks had potent bactericidal activity against S. mutans . The long-lasting antibacterial activity was also achieved through the sustained release of Ag + ions due to the core-shell structure of the nanocomposites. The results of macrophage cytotoxicity showed that the cell viability of dental resins loading less than 1.0 wt% AgBr/BHPVP was close to that of neat resins. The AgBr/BHPVP-containing dental resin with dual bactericidal capability and long term antimicrobial effect is a promising material aimed at preventing second caries and prolonging the longevity of resin composite restorations. Preparation of Magadiite-Sodium Alginate Drug Carrier Composite by Pickering-Emulsion-Templated-Encapsulation Method and Its Properties of Sustained Release Mechanism by Baker–Lonsdale and Korsmeyer–Peppas Model

Ge Mingliang,Li Xinxiang,Li Yueying,Jahangir Alam S. M.,Gui Yuee,Huang Yongchao,Cao Luoxiang,Liang Guodong,Hu Guoqing

In this study, the nanohybrid drug carrier were synthesized by Pickering emulsion-templated encapsulation (PETE) method to control the sustained-released properties of the nanohybrid drug carrier; magadiite-cetyltriphenyl phosphonium bromide (MAG-CTPB-KH550) and sodium alginate (NaC 6 H 7 O 6 ) was dissolved in the aqueous phase but metronidazole (C 6 H 9 N 3 O 3 ) was dissolved in the ethyl acetate (CH 3 COOC 2 H 5 ) of the oil phase; both the oil phase and the aqueous phase were mixed and dispersed to prepared organically-modified magadiite-sodium alginate (MAG–CTPB–KH550/SA) nanohybrid drug carrier. X-ray diffraction (XRD), Flourier transform infrared spectrometry (FTIR) and scanning electron microscopy (SEM) results were shown that the most of Sodium alginate (SA) were encapsulated into the MAG–CTPB–KH550 but a few of SA were intercalated into the inner space layers of MAG–CTPB–KH550, metronidazole was combined with carrier materials through physical apparent adsorption, ion exchange and electrostatic interaction. In vitro result, it was showed that the slow release was shown less than 10% content of Sodium alginate; whereas, it was reduced the initial release percentage of Metronidazole but it was extended the sustained-released time. To reach at equilibrium, the sustained-released effects of the drug carrier were prepared with 10% of Sodium Alginate for 32 h and the maximum cumulative release percentage was 93.42% for 24 h. First order model, Baker–Lonsdale model and Korsmeyer–Peppas model were fitted to study the slow-release mechanism; the correlation coefficients (R 2 ) of the three models were found over 0.9; thus, it was well described the release kinetics behavior of drug carrier composites. The slow-release mechanisms of the drug carrier were performed swelling and dissolving but the barrier effects of the lamina that were reduced the dissolution percentage of metronidazole. Influence of PEG-PLGA nanoparticles on the pore structure of injectable and self-healing hydrogels based on carboxymethyl chitosan/aldehyde sodium hyaluronate for prolonged sustained release of valsartan

Chihao Gao,Lixia Pan,Jie Song,Min Wei,Haozhi Sun,Feng Su,Xin Shen,Suming Li

Valsartan is almost insoluble, resulting in very low bioavailability. Preparing prolonged sustained-release systems to achieve valsartan drug delivery is essential. Herein, two types of poly(ethylene glycol)-b-poly(lactide-co-glycolide) (PEG-PLGA) nanoparticles loading valsartan and carboxymethyl chitosan/aldehyde sodium hyaluronate hydrogels were prepared. The nanoparticles and hydrogel composites(NPs@Gel) were obtained by physical mixing. The NPs@Gel composites have swelling ratios within the scope from 157.87% to 206.51% and mass loss within the scope from 12.85% to 17.32%, influenced by incorporating PEG-PLGA nanoparticles. Moreover, the NPs@Gel composites had properties of self-healing and injection proven by rheological measurements. Importantly, adding nanoparticles could significantly reduce the swelling ratios of the hydrogels by shrinking the void fraction and enhancing the mechanical properties. Meanwhile, NPs@Gel composites also present excellent biocompatibility as evidenced by hemolysis test and MTT assay. Therefore, a potential prolonged sustained-release system of NPs@Gel for the delivery of valsartan was achieved.

Intelligently thermoresponsive flower-like hollow nano-ruthenium system for sustained release of nerve growth factor to inhibit hyperphosphorylation of tau and neuronal damage for the treatment of Alzheimer's disease

Hui Zhou,Youcong Gong,Yanan Liu,Anlian Huang,Xufeng Zhu,Jiawei Liu,Guanglong Yuan,Li Zhang,Ji-an Wei,Jie Liu

Alzheimer's disease (AD) seriously affects human health and life and lacks effective treatments. The lessons of many clinical trial failures suggest that targeting amyloid beta to treat AD is difficult, and finding new targets is an important direction for AD drug research. The neurofibrillary tangles formed by hyperphosphorylation of tau protein induce the production of cytotoxic reactive oxygen species (ROS) and cause neuronal apoptosis . Therefore, inhibition of hyperphosphorylation of tau protein and reduction of neuronal damage have become promising methods for the treatment of AD. We herein designed a novel nanocomposite with high stability and good biocompatibility by using flower-shaped hollow nano-ruthenium (Ru NPs) as a carrier, loading nerve growth factor (NGF) and sealing with phase change material (PCM). Due to its excellent photothermal effect, under the near-infrared (NIR) irradiation, the nanocomposite could effectively penetrate the blood-brain barrier (BBB) and respond to phase changes in the lesion area, releasing NGF, which inhibited tau hyperphosphorylation, reduced oxidative stress , and more importantly restored nerve damage and maintained neuronal morphology, thereby significantly improving learning and memory in AD mice. Thus, the experimental results indicate that multifunctional nanocomposites may be a promising drug in the treatment of AD.

A scalable microfluidic-blow-spinning platform for fabricating food-grade γ-cyclodextrin metal–organic-framework nanofiber composites with sustained release of thymol: a preservation case study of fresh-cut fruits

Zi-An Deng,Yibo Liu,Wenjing Shi,Jiayue Jin,Fengjun Guo,Di Wu,Qingjun Liu,Saowapa Chaiwong,Kunsong Chen

Introduction Developing advanced functional composites by integrating metal–organic frameworks (MOFs) with polymeric nanofibers are important research in materials science. This strategy of incorporating MOFs into polymeric nanofibers offers a promising route to fabricate multifunctional materials with controlled release and antimicrobial capabilities. However, most existing MOFs are metal-based, which poses potential metal ion leaching risks in food-contact scenarios; besides, achieving uniform MOF dispersion in polymeric matrices and enabling scalable fabrication of such composite nanofibers remain challenges. Objectives The present study aimed to develop a novel and scalable fabrication platform for food-grade MOF-nanofiber composites by integrating γ-cyclodextrin metal–organic frameworks (γ-CD-MOFs) with rapid microfluidic-blow-spinning (MBS). Methods γ-CD-MOF was synthesized using γ-cyclodextrin as a biomolecular ligand, followed by adsorption of thymol (THY) through electrostatic interactions. Molecular docking was conducted to elucidate the encapsulation and stabilization mechanisms. The THY-loaded γ-CD-MOF was incorporated into polycaprolactone (PCL) nanofibers via MBS to produce THY@γ-CD-MOF/PCL nanofiber films. Their physicochemical properties, mechanical performance, sustain release behavior, antimicrobial activity, biocompatibility, and preservation efficacy in fresh-cut fruits were evaluated. Results γ-CD-MOF achieved 78.3 % THY encapsulation. Docking revealed hydrogen bonding between the phenolic hydroxyl group of THY and hydroxyl groups of dual γ-CD units (1.781 Å), shorter than typical hydrogen bonds (∼3.5 Å), contributing to complex stability. MOFs were well-dispersed, and their incorporation enhanced the thermal stability, mechanical strength, and hydrophilicity of the nanofibers. The nanofiber films exhibited broad-spectrum antimicrobial activity, sustained THY release at 4 °C and 25°C, and good biocompatibility, effectively suppressing browning and firmness loss in fresh-cut apples. Conclusion The MBS-based platform enabled efficient fabrication of active food-grade γ-CD-MOF-loaded nanofiber preservation materials. THY encapsulation via hydrogen bonding and hydrophobic interactions provided sustained release and antimicrobial functionality, significantly extending the freshness of fresh-cut apples.

N-acetyl-L-cysteine-modified macrophage membrane-coated VEGF sustained-release nanoparticles for treatment of myocardial infarction: A biomimetic nano-buffer for neutralization of detrimental factors and promotion of mature angiogenesis

Ke Zhu,Kun Wang,Yu Yao,Yinxia Zhu,Ziyang Zhu,Wenyuan Wang,Ruijie Qian,Zhiping Deng,Jun Zhao,Yunli Shen,Fuqiang Shao

Myocardial infarction (MI) is a severe cardiovascular event arising from vascular thrombosis, causing cardiomyocyte fatality. Neovascularization, facilitated by Vascular Endothelial Growth Factor (VEGF), is a crucial therapy that delivers oxygen and nutrients to the surviving myocardium. After MI, a substantial influx of macrophages of the inflammatory phenotype infiltrates the infarcted myocardium. these macrophages secrete factors that exacerbate myocardial injury and orchestrate neovascular regression. A biomimetic nano-buffer system was developed in this study, consisting of VEGF-encapsulated nanoparticles coated with N-acetyl-L-cysteine (NAC)-modified macrophage membrane. These nanoparticles accumulate at the infarcted myocardium by exploiting the macrophage membrane's biomimetic inflammation chemotaxis. It effectively neutralizes detrimental factors in the infarcted myocardium while simultaneously enabling the controlled release of VEGF nanoparticles to promote angiogenesis within the affected tissue. What sets our research apart is the profound impact of the nano-buffer on macrophages, accomplished by its efficient scavenging of reactive oxygen species. This action triggers a vital response—the secretion of PDGF by macrophages—which significantly enhances the maturation of nascent blood vessels. The cumulative effect of these actions creates an exceptionally favorable milieu, fostering neovascularization. This study highlights the potential of the nano-buffer system in treating MI.

Preparation of Gum Arabic–Maltose–Pea Protein Isolate Complexes for 1−Octacosanol Microcapsule: Improved Storage Stability, Sustained Release in the Gastrointestinal Tract, and Its Effect on the Lipid Metabolism of High−Fat−Diet−Induced Obesity Mice

Yin-Yi Ding,Yuxiang Pan,Wanyue Zhang,Yijing Sheng,Yanyun Cao,Zhenyu Gu,Qing Shen,Qingcheng Wang,Xi Chen

1-Octacosanol (Octa) is a natural compound with several beneficial properties. However, its poor water solubility and metabolism in the digestive tract reduce its efficacy. The Octa-GA-Malt-PPI microcapsule was prepared as follows: gum Arabic (GA):maltose (Malt):pea protein isolate (PPI) = 2:1:2; core:shell = 1:7.5; emulsification temperature 70 °C; pH 9.0. An in vitro simulated gastrointestinal tract was used to analyze the digestion behavior. C57BL/6 mice were selected to establish an obesity model induced by a high-fat diet (HFD) to evaluate the effect of Octa monomer and the microcapsule. The diffusivity in water and storage stability of Octa improved after encapsulation. The microcapsule was ascribed to electrostatic interactions, hydrogen bonding, and hydrophobic interactions. The sustained release of Octa from the microcapsule was observed in a simulated gastrointestinal tract. Compared with Octa monomer, the microcapsule was more effective in alleviating the symptoms of weight gain, hypertension, and hyperlipidemia induced by HFD in mice. In conclusion, the construction of microcapsule structure can improve the dispersibility and stability of Octa in water, achieve sustained release of Octa in the gastrointestinal tract, and improve its efficiency in alleviating the effects of HFD on the body.Keywords:1-Octacosanol;microcapsule;storage stability;sustained release;HFD-induced obesity mice
