To improve the catalytic epoxidation activity of conventional TS-1 zeolite under acid-free conditions, a series of TS-1-supported zinc ( x Zn/TS-1) catalysts were prepared and evaluated in the epoxidation of 1-hexene and methyl oleate (MO) with H 2 O 2 oxidant. The microstructure of x Zn/TS-1 was investigated using various characterization techniques, and the results showed that the introduction of Zn species was not only well dispersed on the surface of TS-1 but also did not disrupt the crystal structure of TS-1 and tetrahedral framework titanium. Furthermore, the Ti-O-Zn structure can be formed between Zn and Ti species through O-atomic coordination, which improves the vulnerability of the Ti-O bond to attack the H 2 O 2 molecule, and ultimately facilitates the epoxidation reaction between 1-hexene and MO. In addition, the 2% Zn/TS-1 catalysts showed optimal conversions of 45.7 % and 75.41 % for 1-hexene and MO, respectively, and exhibited good catalytic stability and regeneration during the epoxidation process.

Background: Inhibitor of growth 4 (ING4) level was reported to be decreased in head and neck squamous cell carcinoma (HNSC) tissue, however, it is unknown whether and how ING4 participates in regulating the development of oral squamous cell carcinoma (OSCC). Objective: This study
aimed to investigate the role and mechanism of ING4 in OSCC. Methods: ING4 was forced to up- or down-regulated in two OSCC cell lines, and its effects on the malignant behavior of OSCC cells were investigated in vitro. The ubiquitination level of NF-kB p65 in ING4 upregulated cells was
measured by co-immunoprecipitation. Moreover, the effects of ING4 on the methylation level of ALDH1A2 were evaluated by methylation-specific polymerase chain reaction (MSP) assay. The role of ING4 in OSCC growth in vivo was observed in nude mice. Results: Our results showed that the expression
of ING4 in OSCC cell lines was lower than that in normal oral keratinocyte cells. In vitro, ING4 overexpression inhibited the proliferation, migration, and invasion of OSCC cell lines and ING4 silencing exhibited opposite results. We also demonstrated that ING4 overexpression promoted the
ubiquitination and degradation of P65 and reduced DNA methyltransferase 1 (DNMT1) expression and Aldehyde dehydrogenase 1A2 (ALDH1A2) methylation. Moreover, overexpression of p65 rescued the suppression of malignant behavior, induced by ING4 overexpression. In addition, ING4 negatively regulated
the growth of OSCC xenograft tumors in vivo. Conclusion: Our data evidenced that ING4 played a tumor-repressing role in OSCC in vivo and in vitro via NF-ΚB/DNMT1/ALDH1A2 axis.

A rapid and sensitive assessment of the toxicity of oxygenated polycyclic aromatic hydrocarbons (OPAHs), widely distributed persistent organic pollutants in the environment, is crucial for human health. In this study, using high-performance liquid chromatography, the separation and detection of four purines, xanthine (X), guanine (G), adenine (A), and hypoxanthine (HX) in cells were performed. The aim was to evaluate the cytotoxicity of three OPAHs, namely 1,4-benzoquinone (1,4-BQ), 1,2-naphthoquinone (1,2-NQ) and 9,10-phenanthrenequinone (9,10-PQ), with higher environmental concentrations, from the perspective of purine nucleotide metabolism in human skin fibroblast cells (HFF-1). The results revealed that the levels of G and A were low in HFF-1 cells, while the levels of HX and X showed a dose-response relationship with persistent organic pollutants concentration. With increased concentration of the three persistent organic pollutants, the purine metabolism in HFF-1 cells weakened, and the impact of the three persistent organic pollutants on purine metabolism in cells was in the order of 9,10-PQ > 1,4-BQ > 1,2-NQ. This study provided valuable insights into the toxic mechanisms of 1,4-BQ, 1,2-NQ and 9,10-PQ, contributing to the formulation of relevant protective measures and the safeguarding of human health.

Purpose Photothermal therapy (PTT) is a promising anticancer treatment that involves inducing thermal ablation and enhancing antitumor immune responses. However, it is difficult to completely eradicate tumor foci through thermal ablation alone. Additionally, the PTT elicited antitumor immune responses are often insufficient to prevent tumor recurrence or metastasis, due to the presence of an immunosuppressive microenvironment. Therefore, combining photothermal and immunotherapy is believed to be a more effective treatment approach as it can modulate the immune microenvironment and amplify the post-ablation immune response. Methods Herein, the indoleamine 2, 3‐dioxygenase‐1 inhibitors (1-MT) loaded copper (I) phosphide nanocomposites (Cu3P/1-MT NPs) are prepared for PTT and immunotherapy. The thermal variations of the Cu3P/1-MT NPs solution under different conditions were measured. The cellular cytotoxicity and immunogenic cell death (ICD) induction efficiency of Cu3P/1-MT NPs were analyzed by cell counting kit-8 assay and flow cytometry in 4T1 cells. And the immune response and antitumor therapeutic efficacy of Cu3P/1-MT NPs were evaluated in 4T1-tumor bearing mice. Results Even at low energy of laser irradiation, Cu3P/1-MT NPs remarkably enhanced PTT efficacy and induced immunogenic tumor cell death. Particularly, the tumor-associated antigens (TAAs) could help promote the maturation of dendritic cells (DCs) and antigen presentation, which further activates infiltration of CD8+ T cells through synergistically inhibiting the indoleamine 2, 3‐dioxygenase‐1. Additionally, Cu3P/1-MT NPs decreased the suppressive immune cells such as regulatory T cells (Tregs) and M2 macrophages, indicating an immune suppression modulation effect. Conclusion Cu3P/1-MT nanocomposites with excellent photothermal conversion efficiency and immunomodulatory properties were prepared. In addition to enhanced the PTT efficacy and induced immunogenic tumor cell death, it also modulated the immunosuppressive microenvironment. Thereby, this study is expected to offer a practical and convenient approach to amplify the antitumor therapeutic efficiency with photothermal-immunotherapy.

Summary Caspase-4 is an intracellular sensor for cytosolic bacterial lipopolysaccharide (LPS) and underlies infection-elicited pyroptosis. It is unclear whether and how caspase-4 detects host-derived factors to trigger pyroptosis. Here we show that mitochondrial permeability transition (MPT) activates caspase-4 by promoting the assembly of a protein complex, which we term the Apaf-1 pyroptosome, for the execution of facilitated pyroptosis. MPT, when induced by bile acids, calcium overload, or an adenine nucleotide translocator 1 (ANT1) activator, triggers assembly of the pyroptosome comprised of Apaf-1 and caspase-4 with a stoichiometry ratio of 7:2. Unlike the direct cleavage of gasdermin D (GSDMD) by caspase-4 upon LPS ligation, caspase-4 activated in the Apaf-1 pyroptosome proceeds to cleave caspase-3 and thereby GSDME to induce pyroptosis. Caspase-4-initiated and GSDME-executed pyroptosis underlies cholestatic liver failure. These findings identify Apaf-1 pyroptosome as a pivotal machinery for cells sensing MPT signals and may shed light on understanding how cells execute intrinsic pyroptosis under sterile conditions.

In this work, the densities of CO2-loaded and unloaded alkanolamine (1) + N, N-dimethylacetamide (DMAC) (2) mixtures were determined at temperature of (288.15 to 323.15) K and 101 kPa. The alkanolamines were primary amines, 3-amino-1-propanol (3A1P) and 1-amino-2-propanol (1A2P), and tertiary amine, 3-dimethylamino-1-propanol (3DMA1P). There CO2-unloaded 3A1P/1A2P/3DMA1P + DMAC binary systems with the whole concentration and two CO2-loaded 3A1P/1A2P + DMAC systems with alkanolamine mass fraction of (0.2, 0.3 and 0.4) were investigated. Based on the density values of CO2-unloaded binary systems, the thermal expansion coefficient (αp), excess molar volume (VmE), and apparent molar volume (Vφ,i) were calculated and discussed in terms of intermolecular interactions and spatial structure. The VmE values were positive under the studied conditions and increased with the temperature, and the maximum followed the decreasing order of 1A2P + DMAC > 3DMA1P + DMAC > 3A1P + DMAC. Additionally, the VmE data were well fitted with the Redlich-Kister equation with a small standard deviation (σ ≤ 0.024). For the CO2-loaded systems, the density values were correlated with temperature and concentration using different empirical equations, and the average relative deviations (ARD%) were less than 0.299%.

Metallo-β-lactamases (MBLs) are significant threats to humans because they deteriorate many kinds of β-lactam antibiotics and are key enzymes responsible for multi-drug resistance of bacterial pathogens. As a result of in vitro screening, two compounds were identified as potent inhibitors of two kinds of MBLs: imipenemase (IMP-1) and New Delhi metallo-β-lactamase (NDM-1). The binding structure of one of the identified compounds was clarified by an X-ray crystal analysis in complex with IMP-1, in which two possible binding poses were observed. Molecular dynamics (MD) simulations were performed by building two calculation models from the respective binding poses. The compound was stably bound to the catalytic site during the simulation in one pose. The binding model between NDM-1 and the compound was constructed for MD simulation. Calculation results for NDM-1 were similar to those of IMP-1. The simulation suggested that the binding of the identified inhibitory compound was also durable in the catalytic site of NDM-1. The compound will be a sound basis for the development of the inhibitors for MBLs.

Introduction Although Phellinus igniarius has been shown to treat hyperuricemia (HUA) and gout, a self-inflammatory disease caused by purine metabolism disorders, the underlying mechanisms remain unclear. Polysaccharides are among the main components of P. igniarius with anti-inflammatory and immunoregulatory properties. Methods Here, we examined the therapeutic effects of polysaccharide SH-P-1-1 isolated from P. igniarius on HUA and gout, and explored the underlying mechanism, focusing on gut microbiota and metabolite regulation. Results SH-P-1-1 significantly decreased uric acid and creatinine levels, reduced xanthine oxidase and adenosine deaminase activities, alleviated kidney damage, and reduced urate deposition in joints in model rats. It increased the abundance of Blautia and Muribaculaceae but reduced that of the Lachnospiraceae_NK4A136_group , Lactobacillus , and Turicibacter . SH-P-1-1 also significantly modulated the metabolic profile and was closely related to some metabolic pathways, such as tryptophan metabolism, relevant to HUA and gout. The beneficial activities of SH-P-1-1 correlated with gut microbiota composition and differential metabolites, including short-chain fatty acids. Discussion Overall, this study demonstrates the potential of SH-P-1-1 as a natural supplement for preventing and managing HUA and gout.

Bioethanol upgrading to higher alcohols (C4–10–OH) involves several sequential reaction steps occurring at multiple different active sites. Therefore, precisely engineering heterogeneous catalysts with appropriate local chemical structures of active sites and spatial environments is highly desired for efficient ethanol tandem conversion. Herein, we constructed a novel Cu-based catalyst featuring atomically dispersed Cu sites confined in a hierarchical silicalite-1@La2O3@silicalite-1 architecture, where single Cu atoms were confined in the micropores of surface silicalite-1 (S-1) zeolite nanoislands dispersed on the outer La2O3 layer encapsulating a hollow S-1 zeolite core. By modulating the content of Cu and La species, such a constructed multihierarchical Cu@S-1@La2O3@S-1 catalyst exhibited excellent performance in ethanol conversion to higher alcohols, along with an impressively high ethanol conversion of 74.9% and C4–10–OH selectivity of 85.0%, as well as an unprecedentedly high production rate of higher alcohols (10.575 mol·gCu–1·h–1) at 275 °C, far exceeding those over state-of-the-art Cu-based catalysts. It was elucidated that surface single-atom Cu sites first notably promoted the adsorption and activation of ethanol and thus initiated its dehydrogenation to produce acetaldehyde, and subsequently, acetaldehyde intermediates migrated to the outer La2O3 layer featuring abundant medium-strength basic and acidic sites, thereby facilitating the condensation of acetaldehydes and their further dehydration. This work provides an innovative approach for rationally designing high-performance supported Cu catalysts with a spatiotemporal decoupling structure applied in advanced heterogeneous tandem catalytic processes through the design of site-specific confinement within hierarchical structures.

Cocrystal hydrate accounts for a small percentage of cocrystals of poorly water-soluble drugs, and still lack the explanation on its cause of formation. In this study, a cocrystal hydrate with a novel phosphodiesterase-4 (PDE-4) inhibitor, chlorbipram, is prepared and characterized using multi-techniques. The dissolution behavior, ternary phase diagram and theoretical calculations are further performed and discussed. Chlorbipram: m-Hydroxybenzoic acid (ChBP-MHBA, 1: 1) cocrystal hydrate presents approximate equivalent dissolution (1.61 mg/mL) in comparison with ChBP (1.90 mg/mL) in isopropanol/water (1: 1, v/v) solution. Ternary phase diagram plotted in ethyl acetate solvent suggests that ChBP-MHBA cocrystal hydrate has an optimum mass percentage range from (2.91%: 2.64%: 94.45%) to (3.20%: 10.74%: 86.06%) among ChBP, MHBA and ethyl acetate. There are two lattice waters in each unit cell of ChBP-MHBA cocrystal hydrate, accounting for 4.5% proportion of the cell volume using contact surface calculations. Molecular Electrostatic Potential Surface (MEPS) investigations indicate that the strong hydrogen bonds are formed within the cocrystal lattice, which are between the global minimum values of ChBP/MHBA and the global maximum values of lattice water. In summary, lattice water plays an important role in the formation of cocrystal hydrate, providing hydrogen atom donor and forming hydrogen bonds to support its three-dimensional crystal structure.

Oleuropein (OLE) is a secoiridoid glycoside that mainly exists in olives with multifaceted health benefits. The present study aimed to investigate the stress resistance and lifespan extension effects of OLE inCaenorhabditis elegans. The results showed that OLE could significantly prolong the lifespan ofC. elegansby 22.29%. Treatment with OLE also significantly increased the survival rates of worms against lethal heat shock and oxidative stress. Meanwhile, OLE supplementation increased the expression and activity of antioxidant enzymes and suppressed the generation of malondialdehyde in nematodes. In addition, the results from mutants implied that OLE might mediate longevity and stress resistance via DAF-16/FoxO, which played a vital role in the insulin/IGF-1 signaling (IIS) pathway. To further identify the molecular targets of OLE, mRNA level and loss-of-function mutants of IIS-associated genes were investigated. The data revealed that OLE activated IIS by down-regulating the upstream components,daf-2andage-1. Furthermore, another stress response and longevity pathway in parallel to DAF-16, SKN-1/Nrf2, was also shown to involve in OLE-induced beneficial effects. Collectively, these results provide the theoretical basis that OLE could enhance the stress resistance and increase the lifespan ofC. elegansthrough the IIS and SKN-1/Nrf2 signaling pathways. Keywords:oleuropein;Caenorhabditis elegans;longevity;stress resistance

The enzymatic reaction–distillation coupling process is a promising technology to realize the chiral resolution of (R/S)-1-phenylethanol with ethyl butyrate and ethanol as acyl donor and byproduct, respectively. In order to design such a coupling process, the vapor–liquid equilibrium (VLE) data are necessary. However, these data were incomplete in literatures. In this work, the isobaric VLE experiments were conducted at 101.3 kPa for the binary systems of 1-phenylethanol + ethyl butyrate, 1-phenylethanol + ethanol, 1-phenylethyl butyrate + ethyl butyrate, 1-phenylethyl butyrate + ethanol, and 1-phenylethyl butyrate +1-pheny ethanol. Additionally, the saturated vapor pressure of 1-phenylethyl butyrate was measured in the temperature range 420–500 K and correlated with the Antoine equation. Both the Non-Random Two Liquid (NRTL) and Wilson models were used for the correlation of experimental VLE data. Both models showed satisfactory accuracy, although the Wilson model performed slightly better than the NRTL model. This work would provide important thermodynamic information for the design of enzymatic reaction–distillation coupling process to realize the chiral resolution of (R/S)-1-phenylethanol.
