An antibacterial, multifunctional nanogel for efficient treatment of neutrophilic asthma

Xu Zuo,Xiaoping Guo,Dan Zhao,Yinuo Gu,Zheng Zou,Yuanyuan Shen,Chaoliang He,Caina Xu,Yan Rong,Fang Wang

Asthma is a chronic and heterogeneous disease affecting the lungs and respiratory tract. In particular, the neutrophil subtype of asthma was described as persistent, more severe, and corticosteroid-resistant. Growing evidence suggested that nontypeable Haemophilus influenzae (NTHi) infection contributes to the development of neutrophilic asthma, exacerbating clinical symptoms and increasing the associated medical burden. In this work, arginine-grafted chitosan (CS-Arg) was ionically cross-linked with tris(2-carboxyethyl) phosphine (TCEP), and a highly-efficient antimicrobial agent, poly-ε-L-Lysine (ε-PLL), was incorporated to prepare ε-PLL/CS-Arg/TCEP (ECAT) composite nanogels. The results showed that ECAT nanogels exhibited highly effective inhibition against the proliferation of NTHi, Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ). In addition, ECAT nanogels could effectively inhibit the formation of mucins aggregates in vitro , suggesting that the nanogel might have the potential to destroy mucin in respiratory disease. Furthermore, in the ovalbumin (OVA)/NTHi-induced Balb/c mice model of neutrophilic asthma, the number of neutrophils in the alveolar lavage fluid and the percentage of inflammatory cells in the blood were effectively reduced by exposure to tower nebulized administration of ECAT nanogels, and reversing airway hyperresponsiveness (AHR) and reducing inflammation in neutrophilic asthma mice. In conclusion, the construction of ECAT nanogels was a feasible anti-infective and anti-inflammatory therapeutic strategy, which demonstrated strong potential in the clinical treatment of neutrophilic asthma.

An aldolase-dependent phloroglucinol degradation pathway in Collinsella sp. zg1085

Yating Li,Tong Xu,Yanqin Tu,Tong Li,Yifeng Wei,Yan Zhou

Phloroglucinol (1,3,5-trihydroxybenzene) is a key intermediate in the degradation of polyphenols such as flavonoids and hydrolysable tannins and can be used by certain bacteria as a carbon and energy source for growth. The identification of enzymes that participate in the fermentation of phloroglucinol to acetate and butyrate in Clostridia was recently reported. In this study, we present the discovery and characterization of a novel metabolic pathway for phloroglucinol degradation in the bacterium Collinsella sp. zg1085, from marmot respiratory tract. In both the Clostridial and Collinsella pathways, phloroglucinol is first reduced to dihydrophoroglucinol by the NADPH-dependent phloroglucinol reductase (PGR), followed by ring opening to form (S)-3-hydroxy-5-oxohexanoate by a Mn2+-dependent dihydrophloroglucinol cyclohydrolase (DPGC). In the Collinsella pathway, (S)-3-hydroxy-5-oxohexanoate is then cleaved to form malonate semialdehyde and acetone by a newly identified aldolase (HOHA). Finally, a NADP+-dependent malonate-semialdehyde dehydrogenase converts malonate semialdehyde to CO2 and acetyl-CoA, an intermediate in carbon and energy metabolism. Recombinant expression of the Collinsella PGR, DPGC, and HOHA in E. coli enabled the conversion of phloroglucinol into acetone, providing support for the proposed pathway. Experiments with Olsenella profusa, another bacterium containing the gene cluster of interest, show that the PGR, DPGC, HOHA, and MSDH are induced by phloroglucinol. Our findings add to the variety of metabolic pathways for the degradation of phloroglucinol, a widely distributed phenolic compound, in the anaerobic microbiome.

An aqueous phase pre-crosslinking modulated nanofiltration membrane preparation process

Gu Kaifeng,Fang Chuanjie,Wang Jianyu,Zhu Liping

Nanofiltration (NF) is one of the most promising separation technologies of this century. It plays a major role in the chemical, medical, food, and other fields. The separation and permeation performance of NF membranes is closely related to microstructure. In this study, aqueous phase amine monomers are pre-crosslinked during the preparation of NF membranes. Trimesic acid (TMA) is used as a cross-linking agent with polyethyleneimine (PEI), whereby the resulting aqueous-phase monomer is interfacially polymerized with the organic-phase monomer to form a separation layer with internal lithium-ion permeation channels. The physicochemical structure of nanofiltration membranes is analyzed in detail. The pre-crosslinking process is optimized and the NF membrane performance can achieve 97% rejection of MgCl 2 , 15% rejection of LiCl, and 34 L m −2  h −1 permeate flux. The resulting NF membranes also show stable desalination performance in tests with mixed salt solutions. This study provides a new nanofiltration membrane structure and performance optimization scheme, which is favorable for magnesium-lithium separation systems.

An Endogenous Proton-Powered Adaptive Nanomotor for Treating Muscle Atrophy

Ming Liu,Zhicun Liu,Xiangkai Qiao,Cheng Chen,Hongtu Guo,Hao Gu,Junbo Li,Tiedong Sun

Nanomotors driven by endogenous enzymes are favored in biology and pharmacy due to their spontaneous driving and efficient biocatalytic activity, and have potential applications in the treatment of clinical diseases that are highly dependent on targeted effects. For diseases such as muscle atrophy, using energy molecules such as ATP to improve cellular metabolism is a relatively efficient treatment method. However, traditional adenosine triphosphate (ATP) therapies for muscle atrophy face limitations due to instability under physiological conditions and poor targeting efficiency. To address these challenges, we developed an endogenous proton-gradient-driven ATP transport motor (ATM), a nanomotor integrating chloroplast-derived FoF1-ATPase with a biocompatible flask-shaped organic shell (FOS). The ATM is synthesized by vacuum-injecting phospholipid-embedded FoF1-ATPase nanothylakoids into ribose-based FOS, enabling autonomous propulsion in acidic microenvironments through proton-driven negative chemotaxis (directional movement away from regions of higher proton concentration). This nanomotor converts proton gradients into ATP synthesis, directly replenishing cellular energy deficits in atrophic tissues. In vitro studies demonstrated high biocompatibility (>90% cell viability at 150 μg/mL) and pH-responsive motility, achieving speeds up to 4.32 μm/s under physiological gradients (ΔpH = 3). In vivo experiments using dexamethasone-induced muscle atrophy mice revealed that ATM treatment accelerated weight recovery and restored normal muscle morphology, with treated mice exhibiting cell sizes comparable to healthy controls (30–40 μm vs. 15–25 μm in untreated). These results highlight the ATM’s potential as a precision therapeutic platform for metabolic disorders, leveraging the natural enzyme functionality and synthetic material design to enhance efficacy while minimizing systemic toxicity.

An engineered superdurable cellulosic radiative cooling – Power generation wearable metafabric

Chenyang Cai,Yibo Wang,Xiaodan Wu,Wanquan Cai,Zechang Wei,Yu Fu

Incorporating zero-energy-input cooling-power generation technology into personal thermal management (PTM) systems is a promising solution for thermal energy utilization but remains a huge challenge due to lacking of effective assembly technology. In this work, a sustainable, cost-effective, superdurable, and nanostructured cellulosic metafabric (CWF) is reported that exhibits well radiative cooling function to achieve high-performance power generation. Through tailoring the viscoelasticity and optical performance in the electrospinning process, this metafabric demonstrates a unique micro/nano structure and anchored MOF assembly, 95.7 % solar reflectivity, 0.94 infrared emissivity, high mechanical strength, washability, surface hydrophobicity, and UV resistance can be achieved. This metafabric exhibited high cooling efficiency of 5.7 °C during direct sunlight in summer and anti-washing and UV stability. Furtherly, we coupled the CWF (as the cold sink) with a commercial thermoelectric device to assemble a power generation device. The device can output average voltage of 200 mV with an average temperature gradient of ≈20.1 °C under stimulated sunlight and shows superdurable power generation performance even after washing or folding (7000 cycles) use. An outdoor test demonstrated it can deliver a high output voltage of 519 mV in Nanjing, China. The cooling metafabirc shows great promise for practical applications in wearable power generation devices.

An E-skin for handwriting input at human-machine interface

Huacui Xiang,Zhijian Li,Zhou Bai,Haiwei Wu,Guodong Liu,Hongwei Zhou,Hanbin Liu

Wearable human–machine interaction remains a huge challenge when various flexible electronics have been developed to date, for which the information input seems more difficult than the information output such as display devices. Here in this work, an e-skin for handwriting input was disclosed based on a double network hydrogel of gluten protein and polyvinyl alcohol (PVA) that is cross-linked by borax. The hydrogel can tightly adhere onto human skin, being stretchable, self-healable, and working properly even under breakage, which perfectly match the requirements of the e-skin for human–machine interaction. Under the assistant of deep learning, the handwriting of letter and words on it can be recognized with high accuracy of above 89%, even under tensile state or destruction. In addition, this flexible device is biocompatible and biodegradable benefiting from the protein and PVA networks, which make it suitable as a wearable e-skin and being free of electronic wastes. The finding of this work may open a window for the development of e-skin for the two-way human–machine interaction.

An electrochemical signal ‘off–on’ sensing platform for microRNA detection

Huanshun Yin,Yunlei Zhou,Chuanxia Chen,Lusheng Zhu,Shiyun Ai

The abnormal expression of microRNAs (miRNAs) in many solid tumors makes miRNAs potential biomarkers for disease diagnosis and highlights the need for the sensitive and selective detection of miRNAs. In the present work, an ‘off-on’ signaling genosensor platform for miRNA-21 detection was well developed. This tactic was based on a locked nucleic acid-integrated nucleic acid hairpin probe, a biotin-labeled bridge DNA–AuNPs–bio-barcode signal amplification unit and enzymatic signal amplification. The test is simple, fast and ultrasensitive with a linear range of 0.01–700 pM. The detection limit was estimated to be 6 fM. The overexpression of miRNA-21 was confirmed in total RNA extracted from human hepatocarcinoma cells BEL-7402 and human HeLa cells compared with the control sample extracted from normal human hepatic L02 cells. This method does not need miRNA-21 labeling, isolation, enrichment or PCR amplification. The performance of the assay developed here could satisfy the need for rapid, easy, sensitive and specific early cancer diagnosis in clinical diagnostics.

A jelly-like artificial muscle for an untethered underwater robot

Xinge Li,Dingnan Rao,Mingqi Zhang,Yaoting Xue,Xunuo Cao,Shunyu Yin,Jie-Wei Wong,Fanghao Zhou,Tuck-Whye Wong,Xuxu Yang,Tiefeng Li

Underwater robots desire versatile, water-adaptive, untethered, and rapidly actuating soft artificial muscles. Existing artificial muscles compromise between high water compatibility and untethered rapid actuation. Taking inspiration from the watery and fast contraction characteristics of jellyfish, we present a jelly-like muscle design that features an extremely soft (elastic modulus, 66.9 kPa) and watery (water content, 83.3%) structure. The jelly-like muscle can instantly respond to electrical stimuli and provide underwater propulsion. The muscle can shift propulsion direction by simply tuning the voltage signal and quickly recover from electromechanical failures through repair. An untethered robot can be assembled by stimulating the muscle with an onboard power system. The robot reaches a maximum speed of 0.91 cm/s (0.3 body length per second) and has an endurance of 15.7 min with a 500-mAh lithium-ion battery. Robots can be constructed in different forms by tailoring the jelly-like muscle, which is promising for various applications.

An intelligent re-shieldable targeting system for enhanced tumor accumulation

Zhenpeng Hu,Jinlong Ma,Fei Fu,Chen Cui,Xiaomin Li,Xinyu Wang,Wei Wang,Yeda Wan,Zhi Yuan

Programmed ligand targeting strategy promotes the blood circulation stability of nanoparticles by shielding the ligand. However, the irreversible shielding causes the deshielded nanoparticles to be easily recognized and cleared by the reticuloendothelial system (RES), impeding their further retention in the tumor. Here, we for the first time prove the superiority of the intelligent re-shieldable targeting system that is based on the pH-responsive self-assembly/disassembly of gold nanoparticles . The system can enhance the stability of gold nanoparticles in the blood circulation (2.6-fold at 24   h), reduce uptake by the RES (35% lower) and improve tumor accumulation (41% higher by analysis of gold content in tumor) effectively compared with the conventional irreversible system. Furthermore, preliminary study indicates that the system could be applied as computed tomography contrast agent in tumor imaging. The in vivo validity of the intelligent re-shieldable targeting system provides inspiration for the design of nanomaterials for cancer diagnosis and treatment.

Prenylation of Flavanones by an Aromatic Prenyltransferase from Fusarium globosum

Dingtao Tang,Jiajie Quan,Zhengjiao Gao,Bingfeng He,Yu Hou,Peipei Fan,Meidong Pan,Jiali Yang

Prenylation increases the structural diversity and biological activity of flavonoids. In this study, an aromatic prenyltransferase, FgPT1, was identified fromFusarium globosum. This enzyme was demonstrated to specifically catalyze the prenylation of flavanones, including naringenin, hesperitin, eriodictyol, liquiritigenin, rac-pinocembrin, and dihydrogenistein, and exhibited no activity toward other types of flavonoids, including chalcones, flavonols, isoflavonoids, and flavonols. Ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) and nuclear magnetic resonance (NMR) analysis indicated that the majority of prenylated products were 6-Cprenyl flavanones, with the exception of liquiritigenin, which was additionally transformed to 4′-Oprenyl liquiritigenin. Enzyme kinetic analysis suggested that FgPT1 exhibited the highest catalytic efficiency towards naringenin, with akcat/KMvalue determined as 61.92 s−1M−1, and the lowest catalytic efficiency towards liquiritigenin, with akcat/KMof 1.18 s−1M−1. Biochemical characterization suggested that FgPT1 functioned as a metal-dependent enzyme with optimal activity in the presence of Ba2+at pH 7.5 and 30 °C. Site-directed mutagenesis resulted in a series of mutants, including A325V with impaired prenylation activity and V116I, V181I, and V194I with enhanced activity. V194I displayed the highest enzymatic activity with a nine-fold increase compared to wild-type FgPT1.

Arriving an ultralow wear using cellulose/two-dimensional material composites

Xuan Yin,Dingyao Zhang,Bing Zhang

The development of environmentally friendly solid lubricants with exceptional wear resistance is imperative to address the escalating environmental concerns and performance limitations of conventional lubricants in demanding tribological applications. This study systematically investigated the wear resistance of hydroxypropyl methylcellulose (HPMC)/tungsten disulfide (WS2)/graphene composites under normal applied loads (2 and 4 N) and varying solid lubricant contents (stoichiometric ratios 0.2 referred to CWG-0.2 and 10 referred to CWG-10). Quantitative tribological tests revealed that the wear rate of HPMC composites exhibited distinct load dependence at fixed lubricant concentrations. Notably, CWG-0.2 and CWG-10 composites achieved an ultra-low wear rate below 10-10 mm3, representing an approximately 95% reduction compared to pristine HPMC (10-8 mm3). Surface characterization demonstrated that localized carbon phase clusters and interconnected carbon skeleton chains governed the ultra-low wear transition. Prolonged sliding (>10,000 cycles) induced the formation of a 10-50 nm-thick transfer film comprising WS2 nanoflakes and a hybrid amorphous phase (C-O-W-S), as confirmed by X-ray photoelectron spectroscopy and high resolution transmission electron microscopy. The exceptional performance, quantified through rigorous parametric analysis, positions HPMC composites as sustainable solid lubricants for precision machinery, aerospace bearings, and biodegradable micro-electromechanical systems requiring eco-friendly superlubricity lubricants.

Direct ammonia and dihydroxyacetone production in an unbiased photoelectrochemical cell

Guo Xiaotian,Gao Rui-Ting,Ren Shijie,Nguyen Nhat Truong,Chen Haojie,Wu Limin,Wang Lei

Photoelectrochemical production of ammonia usually suffers from a low solar-to-ammonia efficiency and a high overpotential, which influences the bias-free operation of sustainable photoelectrochemistry. Herein, we realize solar-driven ammonia production from waste nitrate by constructing copper-osmium catalysts deposited on the Sb 2 (S,Se) 3 semiconductor, enabling optimized photo-carrier transport pathways and a beneficial co-adsorption configuration of *NO 3 -H 2 O moieties. The photocathode reaches a photocurrent density of 5.6 mA cm −2 at 0 V RHE with a low onset potential of 0.86 V RHE and a Faradaic efficiency of 96.98% at 0.6 V RHE under AM 1.5 G illumination. We further employ glycerol oxidation reaction on ruthenium doped bismuth oxide catalyst decorated on titanium oxide photoanode, requiring an onset potential of 0.3 V RHE to enable bias-free operation. The unbiased photoelectrochemical system shows Faradaic efficiencies of over 97% for ammonia products and above 77% for glycerol oxidation product under AM 1.5 G illumination. The large-sized photoelectrodes maintain a stability for 24 h without noticeable degradation. Our works indicate that unassisted and stable PEC ammonia production is feasible with in situ glycerol valorization using the photoanode and photocathode.
