A ddPCR platform based on a microfluidic chip with a dual-function flow-focusing structure for sample-to-result DNA quantification analysis

xiaoliang zhang,Shun Wang,Jinxian Wang,Xiaojie Sun,Jinbing Xue,Zhenya Wang,Tianhang Yang,Liangfei Weng,Bidou Wang,Gangyin Luo

Droplet digital PCR (ddPCR) is a powerful method for absolute nucleic acid quantification with high precision and accuracy. However, complicated operational steps have hampered the use and diffusion of ddPCR. Therefore, an automated, easy-to-use, low-sample-consumption, and portable ddPCR platform is urgently needed. This paper proposes a microfluidic ddPCR platform based on a microfluidic chip that can realize the sample-to-result function by switching the rotary valve, achieving the dual function of the flow-focusing structure for droplet generation and readout. Sample, generation oil, and analysis oil were pre-added to the reservoirs. Droplets were generated due to focusing flow, and after passing through the integrated temporary storage bin in the rotary valve, the droplets and oil subsequently entered the collecting tube, improving the droplet-to-oil volume ratio for enhanced thermal cycle performance. Droplets with an average diameter of 107.44 μm and a CV of 2.38% were generated using our chip under the optimal pressures. High-performance thermal cycling was achieved through improvements of the droplet-to-oil volume ratio of the sample, the integrated heating lid, the pure copper heating base, and the temperature-controlling algorithm. Gradient quantification experiments were conducted for the HER2 and CEP17 genes extracted from breast cancer cells, yielding strong linear correlations with R2 values of 0.9996 for FAM and 0.9989 for CY5. Moreover, pronounced linearity was obtained between the detected concentrations of HER2 and CEP17, indicated by a slope of 1.0091 and an R² of 0.9997, signifying consistent HER2:CEP17 ratios across various sample dilutions. The outcomes of the quantitative analysis, encompassing the dynamic range and the consistency of the HER2:CEP17 ratio using our ddPCR platform, meet the standards required for breast cancer assessment and therapy. Our ddPCR platform is automated, portable, and capable of stable droplet generation, high-efficiency amplification, realization of the sample-to-result function based on dual-function flow-focusing structure, and accuracy absolute quantification, underscoring its significant potential for ddPCR analysis in clinical diagnostics.

Simultaneously responsive microfluidic chip aptasensor for determination of kanamycin, aflatoxin M1, and 17β-estradiol based on magnetic tripartite DNA assembly nanostructure probes

He Liyong,Shen Zhipeng,Wang Jiaqi,Zeng Jin,Wang Wenhai,Wu Huihui,Wang Qiqin,Gan Ning

The authors describe a microfluidic chip-based aptasensor platform combined with magnetic tripartite DNA structure-functionalized nanocomposites to achieve simultaneous determination of kanamycin (KANA), aflatoxin M1 (AFM1), and 17β-estradiol (E2) in milk. The two-duplex tripartite DNA nanostructure was first assembled on the surface of magnetic beads. When the aptamer on the probes recognized the specific target, the aptamer-target would be released into the supernatant. The pre-primer@circular DNA template structure initiates rolling circle amplification (RCA) by phi29 polymerase. After magnetic separation, the magnetic nanocomposites were added into a solution containing three different lengths of complementary strands to the RCA products. The number of complementary strands significantly decrease, and this can be quantitated by the microfluidic chip. Further, the employment of magnetic nanocomposites and microfluidic chip not only resolve the complex matrix interference, but also dramatically enhances the determination selectivity and sensitivity. This aptasensor allows for determination of KANA, AFM1, and E2 with limits of detection as low as 0.32 pg mL −1 , 0.95 pg mL −1 , and 6.8 pg mL −1 , respectively. This novel method exhibits the advantages of excellent stability and fast response time (< 3 min on microfluidic chip platform) for simultaneous determination of KANA, AFM1, and E2 in milk samples and ensures food safety. Combining field-amplified sample stacking with moving reaction boundary electrophoresis on a paper chip for the preconcentration and separation of metal ions

Liangfei Ouyang,Qian Liu,Heng Liang

A common drawback of paper-based separation devices is their poor detection limit. In this study, we combined field-amplified sample stacking with moving reaction boundary electrophoresis on a paper chip with six array channels for the parallel separation and concentration of multiple samples. With a new hyphenated technique, the brown I 2 from the Fe 3+ /I − oxidation–reduction reaction emerged near the boundary between the dilute ethylene diamine tetraacetic acid and potassium iodide and highly concentrated KCl solutions. For the separation and concentration of three components, Cr 3+ , Cu 2+ , and Fe 3+ , the Fe 3+ detection limit was improved at least 266-fold by comparing the hyphenated technique with moving reaction boundary electrophoresis. The detection limit of Fe 3+ was found to be as low as 0.34 ng (20 μM) on the paper chip. We also demonstrated the analysis of a real sample of four metal ions, with detection limits as follows: 0.16 μg Cr 3+ , 1.5 μg Ni 2+ , 0.64 μg Cu 2+ , and 1.5 μg Co 2+ . The synergy of field-amplified sample stacking and moving reaction boundary electrophoresis in the micron paper-based array channels dramatically improved the detection limit and throughput of paper-based electrophoresis.

Construction of a pumpless gravity-driven vascularized Skin-on-a-Chip for the study of hepatocytotoxicity in percutaneous exposure to exogenous chemicals

Zhou Su,Li Rui,Sun Jie,Gu Minyang,Gao Dan,Tang Liming,Zhu Jiangbo

The utilization of existing Skin-on-a-Chip (SoC) is constrained by the complex structures, the multiplicity of auxiliary devices, and the inability to evaluate exogenous chemicals that are hepatotoxic after percutaneous metabolism. In this study, a gravity-driven SoC without any auxiliary devices was constructed for the hepatocytotoxicity study of exogenous chemicals. The SoC possesses 3 layers of culture chambers, from top to bottom, for human skin equivalent (HSE), Human Umbilical Vein Endothelial Cells (HUVEC) and hepatocytes (HepG2), and the maintenance and expression capacity of the corresponding cells on the SoC were verified by specificity parameters. The reactivity of the SoC to exogenous chemicals was verified by 2-aminofluorene (2-AF). The SoC can realistically simulate the in vivo exposure process of exogenous chemicals that are percutaneously exposed and metabolized into the bloodstream and then to the liver to produce toxicity, and it can achieve the same effects on transcriptome as those of animal tests at lower exposure levels while examining multiple toxicological targets of the skin, vascular endothelial cells, and hepatocytes. Both in terms of species similarity, the principles of reduction, replacement and refinement (3R), or the level of exposure suggest that the present SoC has a degree of replacement for animal models in assessing exogenous chemicals, especially those that are hepatotoxic after percutaneous metabolism.

Construction of a two-dimensional magnetic solid phase extraction microfluidic chip/tandem mass spectrometry platform for the determination of cytarabine in plasma

Yangkun Sun,Yu Han,Linghao Zhao,Yuanzheng Xiang,Wenjing Xiong,Hongyang Zhang,Min Zhang,Fanghong Ning,Ping Hu

A novel concept of two-dimensional magnetic solid phase extraction microfluidic chip (2D-MSPE-chip) was proposed, in which two or even more different separation mechanisms of solid phase extraction units were designed and constructed on microfluidic chips in series. The synthesized Fe 3 O 4 -UiO-66-C18 was used in the first dimension to retain protein and phospholipid through hydrophobic effect and Zr O P coordination, which could almost eliminate the interference of the matrix effect on the mass spectrum. The second dimensional material utilized Fe 3 O 4 -UiO-66-NH 2 to enable cytarabine enrichment via π–π and hydrogen bonding. The parameters of 2D MSPE-Chip/tandem MS method such as desorption solvent (5 % acetic acid acetonitrile), loading solvent (20 % acetonitrile), and flow rate for extraction and desorption (10 μL/min) were select. The LOD of the established method for cytarabine in plasma was 0.13 ng/mL, and the linear range was between 2.0 ng/mL and 1000 ng/mL with correlation coefficient (R 2  = 0.9991). The matrix effect was −5.0 %. The recovery rate of real samples ranged from 95.5 % to 104.8 %, with RSD ≤ 5.2 %. The chip design was simple and efficient and could be reused, thereby decreasing analysis expenses. It held considerable potential for application in rapid clinical diagnostics and precision medicine.

A rapid microfluidic paper-based chip sensor using ratiometric fluorescence and molecularly imprinted polymers for visual detection of sulfadiazine in actual samples

Yuanzheng Cai,Lingling Cao,Hongmei Cai,Wenqi Yang,Hongjie Lu,Abulaite Adila,Baolin Zhang,Yang Cao,Weihong Huang,Wanzhen Xu,Wenming Yang

In recent years, the widespread use of sulfonamide antibiotics has led to increasing concerns over antibiotic residues. To address the challenges, this study developed a rapid and highly sensitive detection method based on ratiometric fluorescence. By employing nitrogen-doped carbon quantum dots (N-BCDs) and biomass-derived carbon quantum dots (RCDs), single-channel and ratiometric fluorescence sensors were constructed. The incorporation of molecularly imprinted polymers (MIPs) enhanced the specificity and sensitivity of target recognition. Experimental results demonstrated that the ratiometric fluorescence sensor exhibited superior anti-interference capability and detection stability compared to the single-channel sensor, achieving a detection limit as low as 10.41 nM, significantly outperforming the single-channel sensor's limit of 25.94 nM. To enable rapid on-site detection, the ratiometric fluorescence sensor was integrated with a microfluidic paper-based chip, resulting in a visual detection platform. Moreover, a microfluidic paper-based chip with a spatially rotational structure was developed, integrating smartphone photo-recognition functionality to convert fluorescence ratio changes into RGB values, enabling rapid visual detection of SDZ. In practical applications involving milk and water samples, the sensor achieved recovery rates between 98.70 % and 108.70 %, demonstrating excellent performance. This ratiometric fluorescence microfluidic paper-based chip sensor offers simplicity, high sensitivity, and strong selectivity, making it suitable for non-professional users to perform rapid on-site detection. It shows great potential for applications in food safety and environmental monitoring.

A Miniaturized Electrochemical SERS Chip with 3D Nanoporous Gold Interface for Qualitative and Quantitative Analysis of Organic Pollutants and their Oxidation Intermediates

Xu Wang,Yong Yan,Huan Zhou,Xinyu Liu,Riyi Zhang,Dong Wang,Peter Schaaf,Guangsheng Guo,Xiayan Wang

Polycyclic aromatic hydrocarbons (PAHs) and organophosphorus pesticides (OPPs) are typical pollutants characterized by a wide variety of types, difficulty in degradation, and significant differences in mutagenic and carcinogenic toxicity. However, due to their distinct physicochemical properties, it remains challenging to simultaneously analyze PAHs and OPPs under real environmental conditions within the same simplified detection systems and accurately assess their oxidation-reduction processes. In this study, electrochemical surface-enhanced Raman scattering (EC-SERS), utilizing a microfluidic chip of self-made 3D nanoporous Au loaded with Ag nanoparticles, is employed to analyze PAHs and OPPs in the environment and investigate their oxidation-reduction processes, which displayed excellent sensitivity and selectivity. The chip successfully analyzed three types of PAHs and four types of OPPs, achieving the lowest detection limit of 0.01 ppb, and strong linearity within a certain range. The recovery in real water samples reached 102.89% with a relative standard deviation (RSD) of 13.36%. Furthermore, this system successfully identified different types of pollutants and their concentrations in mixed samples with high accuracy. Meanwhile, preliminary insights into the electrochemical oxidation and degradation processes of these pollutants are provided, offering a first understanding of their transformation mechanisms.

Multi-component hybrid films based on covalent post-synthetic functionalization of silicon chip using both ZIF-90 and lanthanide complexes for luminescence tuning

Jing Ma,Bing Yan

We prepared multi-component luminescent hybrid films based on a silicon chip by a covalent functionalization strategy of both metal–organic frameworks (ZIF-90) and lanthanide complexes. Herein the Si chip is firstly covalently modified by aminopropyltriethoxysilane (APTES) via a hydrolysis and condensation reaction, and then ZIF-90 is covalently grafted onto the surface of Si chip through imine condensation with APTES to form the hybrid films (Si-APTES-ZIF-90). Ternary lanthanide complexes with 3-methacryloxypropyltrimethoxysilane (MPTMS) and Phen (1,10-phenanthroline) are introduced to the above film systems through MPTMS as a covalent linker. The final multi-component hybrid films (Phen-Ln-MPTMS-Si-APTES-ZIF-90) display the typical Ln3+ (Ln = Eu, Tb, Sm and Dy) luminescence through the energy transfer from the MPTMS-Si-APTES-ZIF-90 host. Moreover, fine tuning of the luminescence color for these films is discussed by adjusting the excitation wavelength. Phen-Ln-MPTMS-Si-APTES-ZIF-90 (Ln = Eu, Sm, Dy) can exhibit white color emission closely by selective excitation. In addition, the consolidation stability under airflow of the film and the luminescence stability of the film under certain humidity environments are discussed.

ZnSe quantum dot based ion imprinting technology for fluorescence detecting cadmium and lead ions on a three-dimensional rotary paper-based microfluidic chip

Junrui Zhou,Bowei Li,Anjin Qi,Yajun Shi,Ji Qi,Huizhong Xu,Lingxin Chen

In this study, a newly fluorescent ZnSe quantum dots (QDs) with ion imprinting technology was firstly realized on the three-dimensional (3D) rotary paper–based microfluidic chip platform which can be used to realize specific and multiplexed detection of Cadmium ions (Cd 2+ ) and Lead ions (Pb 2+ ). Compared to CdTe quantum dots, ZnSe quantum dots are less toxic and more environmental friendly. In addition, this design improved the portability of the device by transferred the liquid phase of ZnSe QDs@ion imprinted polymers to solid glass fiber paper. Moreover, the 3D rotary microfluidic chip ( μ PADs) showed great advantages including low cost, simple and fast facile operation, multiplexed detection, and showed good sensitivity and selectivity. Under optimal experiment conditions, our proposed method was enabled to realize specific and multi-channel determination of Cd 2+ and Pb 2+ ions. The developed sensor of Cd 2+ μ PADs provided a linear response from 1 to 70 μg/L with a lower detection limit of 0.245 μg/L, and Pb 2+ μ PADs provided a linear response from 1 to 60 μg/L with a lower detection limit of 0.335 μg/L, respectively. Excitingly, this newly designed 3D rotary μ PADs exhibited quantitative information conveniently, which showed the promising application prospects to rapid testing target metal ions in environmental in the future.

A dual-mode paper-based chip for real-time detection of histamine in fish meat via Lewis base-induced in situ fluorescence system

Yidan Zhang,Yuhua Lai,Yifei Ma,Lin Ge,Jinhua Liu

Since excess histamine (His) in seafood poses a significant health threat, the development of intelligent, on-site, and visual multimodal methods for detecting fish freshness has attracted widespread attention. In this study, we developed a Lewis base-induced paper-based chip coupled with smartphone RGB (Red Green Blue) recognition for the real-time colorimetric/fluorescent dual-mode detection of His in fish meat. The sensing mechanism was based on the catalytic role of His in rapidly forming a green fluorescent compound (GFC) from dopamine and 3-ethylamino-4-methylphenol. Interestingly, the introduction of 1% Tween (TW) 80 was found to significantly enhance the fluorescence signal and real-time stability of the system, achieving an exceptional detection limit of 16.67 nM for His. By immobilizing the GFC probe on Whatman 1 filter paper, a paper-based test strip for His was further developed. His quantification is achieved through RGB analysis with a smartphone, allowing visual and real-time assessment of fish meat freshness directly in the field. The developed sensor exhibits outstanding selectivity, stability, and sensitivity, and has been successfully applied to monitor His levels in fish meat samples (crucian, saury and golden pomfret) over time, providing a reliable means to prevent His poisoning. Therefore, this work presents a robust, low-cost, and user-friendly point-of-care testing platform that holds significant potential for applications in intelligent food monitoring and environmental sensing.

Jiyong Yin,Jiangping Niu,Junsheng Huo,Jing Sun,Jian Huang,Chaoqun Sun

We attempted to construct and evaluate a novel detection method to realize simultaneous detection based on a multiplex liquid protein chip technique for nine nutrition-and-health-related protein markers to meet the requirement of an accurate, simultaneous and comprehensive analysis of the proteomics of nutrition and health. The lower limits of detection, biological limits of detection and regression equations of serum ferritin (SF), soluble transferrin receptor (sTfR), c-reactive protein (CRP), retinol-binding protein4 (RBP4), apolipoprotein B (ApoB), alpha-fetoprotein (AFP), prealbumin (PA), carcino-embryonic antigen (CEA) and D-Dimmer (D-D) were determined after a series of optimal experiments. Then, the results of the methodological evaluation for this novel method indicated that the accuracies were between 70.12% and 127.07%, the within-run precisions were between 0.85% and 7.31%, the between-run precisions were between 3.53% and 19.07%, the correlation coefficients between this method and other methods were above 0.504 (p< 0.05), and the direct bilirubin (DBIL) of low concentration and the indirect bilirubin (IBIL) of high concentration could not interfere with the detected results of nine indicators. The novel multiplex detection method, which can increase accuracy and improve the ability of comprehensive analysis, can basically meet the requirement of detection and the diagnosis of the proteomics of nutrition and health. Keywords:liquid protein chip;nutrition and health;protein marker;multiplex detection

Vat photopolymerization-based 3D printing of complex-shaped and high-performance Al2O3 ceramic tool with chip-breaking grooves: Cutting performance and wear mechanism

Haidong Wu,Wei Liu,Yuerui Xu,Lifu Lin,Yehua Li,Shanghua Wu

Due to the processing of alumina ceramic cutting tools with complex shapes using traditional methods is difficult and time-consuming, vat-photopolymerization-based 3D printing was adopted to fabricate Al2O3 ceramic cutting tools with grooves for the first time. Subsequently, cutting performance evaluation and wear mechanism analysis were conducted. The relative density, Vickers hardness, and bending strength of the alumina cutting tools were determined. The effects of the cutting speed, feed rate, and cutting depth on the cutting performance and wear mechanism of the cutting tools were systematically investigated. In addition, two commercial cutting tools, namely cemented carbide and ceramic tools without grooves, were used for comparison. The cutting speed has the highest influence on the cutting performance, whereas the cutting depth has the least influence. The cutting performance of the prepared alumina cutting inserts with chip breaker grooves superior to that those without chip-breaking grooves and that of the cemented carbide tools. The wear mechanisms of the prepared alumina cutting tools and commercial tools were determined to be abrasive and adhesive wear, and those of the cemented carbide tools were adhesive wear and breakage. This work opens a new avenue for the future preparation of high-performance and complex-shaped ceramic cutting tools.
