Reliable intelligent food packaging films based on polysaccharide–protein composites are often limited by interfacial incompatibility and unstable colorimetric responses under complex environments. Herein, a nanoconfinement-assisted interfacial engineering strategy is proposed to construct reliable intelligent κ-carrageenan/Zein films by incorporating anthocyanin-loaded zeolitic imidazolate framework-8 (Cy@ZIF-8) as an active interfacial nanomodulator. In this composite system (Cy@ZIF-8/Kc/Zein), Cy@ZIF-8 simultaneously stabilizes anthocyanins through confinement effects. It also reorganizes the polysaccharide–protein network via hydrogen bonding, electrostatic interactions, and coordination interactions, effectively suppressing phase separation and promoting the formation of a homogeneous polymer matrix with a uniform thickness of approximately 0.052 mm. As a result, the Cy@ZIF-8/Kc/Zein film exhibited altered water and gas transport behaviors, showing a high swelling ratio of 4154%, a low moisture absorption of 8.19%, and a reduced water vapor transmission rate of 0.549 × 10 −8 kg·m·m −2 ·s −1 ·Pa −1 . These structural features further led to enhanced functional performances, including antioxidant activity with DPPH and ABTS scavenging efficiencies exceeding 80% at 7.5 mg/mL. Additionally, the film showed synergistic antibacterial properties against Escherichia coli and Staphylococcus aureus with survival rates decreasing to 15.29 ± 0.50% and 1.42 ± 0.31%, respectively. Application validation assays confirmed reliable chromatic evolution of the composite film under complex environmental conditions. Therefore, this study demonstrates that MOF-based nanostructures can function as effective interfacial regulators to tailor structure–property relationships and improve functional reliability in polysaccharide–protein intelligent packaging films.

SSPS-Zn chelate was synthesized by soluble soybean polysaccharide (SSPS) and zinc chloride, and its properties were investigated in this paper. SSPS-Zn was easily soluble in deionized water, but not easily soluble in acetone, chloroform and n -butanol at room temperature. The water content and swelling index of SSPS-Zn were 5.52% and 185.00%, respectively. SSPS-Zn also showed good colloidal stability in the pH range of 1–14. In 0–10 mg/mL concentration range, SSPS-Zn exhibited markedly improvement for all four in vitro antioxidant activities (DPPH, •OH, and O 2 ⁻• scavenging, and lipid peroxidation inhibition). SSPS-Zn demonstrated stronger inhibitory effects on α -glucosidase and α -amylase, and superior urea adsorption ability, implying the potential to lower blood sugar and urea toxicity. The SSPS-Zn capsule preparation process was optimized using Response Surface Design. The quadratic polynomial regression model for the yield of capsule granule intermediates was established. The optimal conditions included an ethanol concentration of 77.60%, an MCC content of 38.88%, and a drying temperature of 51.50 °C, achieving a granulation yield of 91.02%. The SSPS-Zn capsules exhibited favorable physicochemical properties, with a repose angle below 30°, an average bulk density of 0.92 g/mL, an average moisture content of 6.22 ± 0.15%, and a critical relative humidity of approximately 66%. Under simulated gastrointestinal conditions, the Zn 2+ release rate reached approximately 41.66% after 40 min, 65.74% after 2 h, and 70.83% after 3 h, indicating a sustainable release behavior. These characteristics suggested that SSPS-Zn capsules have potential as multifunctional organic zinc supplements with antioxidant, hypoglycemic, and detoxifying properties.

The soybean protein isolate (SPI)–lobetyolin complexes were prepared, and the binding mechanism, structural alterations and techno-functionalities of the prepared complexes were further measured using multi-spectral technologies and molecular interaction analyses. Lobetyolin displayed strong affinity for SPI and induced static quenching of the intrinsic fluorescence. Hydrophobic contacts, hydrogen bonding, and van der Waals forces were involved in the interaction between SPI and lobetyolin. The complexes exhibited high lobetyolin binding capacity (maximum binding amount of 173 μg/mg), with decreases in free sulfhydryl and free amino group contents compared to SPI. The lobetyolin binding perturbed the microenvironment around tyrosine (Tyr) and tryptophan (Trp) residues, and thereby altered SPI secondary structure with increased β-turn content and decreased β-sheet content. The complexes (at a concentration of 400 μg/mL) exhibited DPPH•- and ABTS •+ -scavenging activities of 56.9% and 69.8%, respectively. Furthermore, the emulsifying stability index (ESI) of the complexes was improved compared to that of SPI. The use of lobetyolin enables the rational construction of food-medicinal protein-based ingredients whose techno-functional attributes can be precisely tailored. Chemical compounds investigated in this research Lobetyolin (PubChem CID: 14655097).

Pithecellobium clypearia Benth polyphenols (PCBPL) possess potent antioxidant and anti-inflammatory properties, making them promising agents for ulcerative colitis (UC) therapy. However, their clinical translation is hindered by poor water solubility, low stability, and limited bioavailability. To overcome these limitations, Pithecellobium clypearia Benth polyphenols liposomes (PCBPL-Lips) were prepared via thin-film hydration. The resulting PCBPL-Lips exhibited a high encapsulation efficiency of 89.31 ± 1.09%, an average particle size of 124.51 ± 7.61 nm, and a zeta potential of −31.62 ± 2.37 mV. Transmission electron microscopy confirmed their uniform particle distribution, spherical morphology, and smooth surfaces. Stability evaluations showed that PCBPL-Lips remained stable under various pH, temperature, and ionic strength conditions. In vitro digestion studies revealed a sustained-release profile, suggesting potential for prolonged therapeutic action. Biocompatibility assessments showed a hemolysis rate below 5% and cell viability above 80%, indicating excellent safety. In vitro activity assays demonstrated that liposomal encapsulation preserved the antioxidant activity of PCBPL and significantly enhanced its anti-inflammatory efficacy. In a murine model of UC, PCBPL-Lips alleviated clinical symptoms, reduced body weight loss, increased colon length, decreased spleen index and tumor necrosis factor-alpha (TNF-α) levels, and improved colonic tissue architecture. This study demonstrates that liposomal encapsulation markedly enhances the stability, bioavailability, and therapeutic efficacy of PCBPL, providing a promising strategy for its use in the treatment of inflammatory bowel diseases.

Atherosclerotic plaques are characterized by oxidative stress and inflammatory responses, which amplify each other in a vicious cycle, leading to progressive plaque destabilization and ultimately serious cardiovascular events. Therefore, the development of nanoformulations with synergistically enhanced anti-inflammatory and antioxidant activities and effective aggregation in plaques is ideal. In this study, we prepared a gallic acid (GA)-cerium metal polyphenol nanoconjugate (GA-Ce, GC) through cerium ions-mediated oxidative coupling of GA, followed by functionalization of its surface with a phosphatidylserine (PS)-binding peptide (GC-PS, GCP), and subsequent encapsulation with a P-selectin glycoprotein ligand-1 (PSGL-1)-overexpressing endothelial cell membrane (PEM) to harvest a biomimetic PEM@GCP nanozyme. The in vitro results showed that PEM@GCP could effectively scavenge free radicals, promote macrophage differentiation to the M2 phenotype, reduce the content of proinflammatory cytokines, inhibit lipid peroxidation, and inhibit apoptosis with cytoprotective functions. In the atherosclerotic mouse model, compared with the control, PSGL-1-based PEM@GCP resulted in a 4.04-fold greater accumulation in plaque lesions. Through its synergistic anti-inflammatory and antioxidant activities, it effectively blocks the vicious cycle of inflammation-oxidative stress, thereby significantly ameliorating the pathological microenvironment and leading to a substantial 71.3% reduction in the plaque area. These studies elucidate the therapeutic potential of a biomimetic metal-polyphenol nanozyme in the treatment of atherosclerosis, demonstrating its potential as a promising candidate for clinical translation. This study aims to develop a novel chitosan-based pH-responsive hydrogel for sustained curcumin (Cur) delivery. Chitosan was modified with norbornene dicarboxylic anhydride to introduce reactive double bonds and enhance solubility, while cysteine-modified Pluronic F127 was crosslinked via UV-induced thiol–ene chemistry to form a stable three-dimensional network. The hydrogel structure was tuned by adjusting the CSNB/CPC ratio and characterized using 1 H NMR, solid-state 13 C NMR, FTIR, TGA, XRD, SEM, and rheology. The optimized hydrogel achieved a drug loading capacity of 13.3 mg/g and exhibited pH-responsive release in simulated gastrointestinal fluids, with kinetics well-fitted to the Peppas–Sahlin model (R 2 > 0.98), dominated by Fickian diffusion. Functional evaluation showed that NC4 displayed nearly 90% DPPH radical scavenging efficiency, and its antioxidant potential was further supported by the Folin–Ciocalteu (GAE) assay and the phosphomolybdenum (AAE) assay, indicating enhanced overall reducing capacity and total antioxidant capacity. NC4 also exhibited strong antibacterial activity against Staphylococcus aureus and Escherichia coli . Biocompatibility was confirmed by CCK-8 and hemolysis assays, with cell viability exceeding 90% and hemolysis rates below 0.35%. Collectively, this multifunctional hydrogel integrates sustained release, antioxidant, antibacterial, and excellent biocompatibility, providing a promising material platform for oral controlled drug delivery and wound healing applications.

Excessive accumulation of reactive oxygen species (ROS) in wound tissue can lead to intense oxidative stress and inflammatory responses, significantly hindering the wound healing. Therefore, effectively removing excess ROS at the wound site and alleviating inflammation are crucial for restoring the body's microenvironment and promoting wound healing. In this study, electrospun nanofibers of polycaprolactone/gelatin (PCL/GT) loaded with Dunaliella salina ( D. salina ) oil were prepared using electrospinning technology to facilitate wound healing. The physicochemical properties of the electrospun nanofibers were characterized through analytical tests, and their antioxidant and anti-inflammatory activities were evaluated. The results demonstrated that the prepared electrospun nanofibers exhibit a randomly oriented fibrous morphology, with an average fiber diameter ranging from 0.34 to 0.97 μm, resembling the structure of the native extracellular matrix. The PCL/GT electrospun nanofibers effectively loaded D. salina oil, exhibiting good vapor permeability, water absorption (204–5138 g/m 2 ·d), thermal stability (up to 410 °C), adequate mechanical properties (tensile strength up to 5.85 MPa and elongation at break up to 162.67%), and no cytotoxicity toward L929 fibroblasts (cell viability≥80%). biodegradability, and thermal stability. Furthermore, these electrospun nanofibers enable sustained release of DSO for up to 30 days, significantly promote cell migration ( ca. PCL/GT/DSO-10%, 52% vs. control group, 34%) at 24 h, they could scavenge exogenous oxidants and promote macrophage-mediated endogenous antioxidant responses to combat oxidative stress damage. They also regulated macrophage polarization and the expression of inflammatory cytokines, exerting anti-inflammatory effects. This creates an optimal microenvironment for wound healing, highlighting the significant potential for applications in wound dressing.

To achieve formulation simplification and green fabrication, this study proposes an integrated “extraction–film formation” strategy based on Natural Deep Eutectic Solvents (NADES). Choline chloride and urea were used as the NADES system combined with ultrasound to extract flavonoids from Dalbergia pinnata (Lour.) Prain (DPF), which are directly incorporated into a Gelatin/Chitosan (Gel/CS) matrix to prepare deep eutectic composite films (GCD1–4) by solution casting. This extract simultaneously serves as a homologous plasticizer and functional agent. It avoids the repeated addition of plasticizers, antibacterial, and antioxidant agents, which are commonly required in conventional film preparation. The results show that with increasing DPF content, the films transformed from “rigid and brittle” to “flexible”, the surface wettability shifted from hydrophilic to hydrophobic, water solubility significantly decreased, and UV shielding ability was enhanced. The GCD-2 film show antibacterial efficiencies of 73.37 ± 4.94 % and 96.78 ± 0.2 % against Staphylococcus aureus and Escherichia coli , respectively. Its DPPH and ABTS radical scavenging rates were 69.74 ± 0.67 % and 99.08 ± 0.9 %, respectively, demonstrated good antibacterial and antioxidant activities. Cell viability was close to 100 %, and the hemolysis rate was below 5 %, indicates good biocompatibility of the films. Furthermore, the GCD series of films demonstrated good biodegradability, indicates their strong environmental friendliness. In application tests, the GCD-2 film significantly reduced weight loss, browning, and decay in strawberries and apple slices, effectively inhibiting decay, and shows the best preservation effect. In conclusion, the Gel/CS/DPF composite films achieved synergistic “plasticization-antibacterial-antioxidation” effects through the green extract, demonstrating great potential as biodegradable active packaging materials for fruit preservation.

Inflammatory bowel disease (IBD) represents a chronic inflammatory disorder characterized by excessive reactive oxygen species (ROS) generation, persistent inflammatory response, and gut microbiota dysbiosis. Herein, a novel colon-targeted composite hydrogel (FTS/Gel) consisting of metal-polyphenol nanozymes (FTS) embedded in an inulin-based gel was developed for efficient oral treatment of IBD. This nanozyme (FTS) was synthesized by coordination between Fe 3+ ions and the anti-inflammatory drugs taxifolin (Tax) and L- Se -methylselenocysteine (SeMC), exhibiting remarkable superoxide dismutase (SOD)-like and catalase (CAT)-like activities. Moreover, FTS nanozymes significantly scavenged intracellular ROS and suppressed inflammation in RAW264.7 cells. Incorporating FTS into an inulin hydrogel notably enhanced colon-targeting efficiency, bio-adhesion, and retention time in vivo. Oral administration of FTS/Gel effectively alleviated colitis, as evidenced by increased colon length, reduced splenomegaly, restored intestinal epithelial barrier integrity, and enhanced mucin secretion. Furthermore, FTS/Gel treatment rebalanced gut microbiota composition by promoting beneficial bacteria and suppressing pernicious bacteria. Collectively, this study presents a promising therapeutic strategy combining antioxidant nanozymes and prebiotic-based hydrogel delivery systems for effectively managing and treating IBD.

Coix seed is a medicinal-edible plant rich in bioactive polysaccharides. While coix seed polysaccharide (CSP) exhibits broad pharmacological activities, its neuroprotective potential against neurodegenerative diseases remains underexplored. This study identified CSP as a heteropolysaccharide with two distinct molecular weight peaks (552 Da and 35,011 Da), primarily composed of glucose. It has a triple-helical structure and exhibits strong, dose-dependent antioxidant properties. In vitro antioxidant assays demonstrated strong radical scavenging capacity, with IC 50 values of 2.56 mg/mL for ABTS radical and 2.75 mg/mL for •OH. Crucially, CSP pretreatment significantly protected PC12 cells against H 2 O 2 -induced oxidative damage, evidenced by a 41.56% increase in cell viability at 1.6 mg/mL ( P < 0.05). Additionally, CSP increased the activity of superoxide dismutase (SOD) and catalase (CAT) in a dose-dependent manner, raised glutathione (GSH) levels, and reduced the release of lactate dehydrogenase (LDH) and the content of malondialdehyde (MDA) in PC12 cells exposed to H 2 O 2 . Moreover, CSP significantly reduced reactive oxygen species (ROS) accumulation and attenuated apoptosis. Taken together, these findings elucidate CSP's neuroprotective mechanism via oxidative stress mitigation, supporting its potential development as a therapeutic agent or functional food component for neurodegenerative disorders.

Chronic wounds, characterized by persistent inflammation, impaired angiogenesis, and disrupted tissue regeneration, remain a major clinical challenge. Current therapies can alleviate symptoms but fail to achieve long-term regulation of the wound microenvironment and functional regeneration. To overcome these limitations, a self-healing collagen–oxidized hyaluronic acid hydrogel loaded with platelet-rich plasma (COL–OHA@PRP) based on dynamic covalent crosslinking was developed, integrating two clinically applied therapies—collagen (COL) and platelet-rich plasma (PRP)—into a single platform. Guided by the design concept of “sustained release–protection–synergy”, this hydrogel integrates three essential functions: self-healing ability, tissue adhesiveness, and controlled PRP release. While preserving the native triple-helix structure of COL, the hydrogel forms a stable adhesive interface with the wound surface, enabling sustained release and protection of bioactive factors in PRP, and synergistically regulating inflammation and tissue regeneration. In vivo studies demonstrated that COL–OHA@PRP hydrogel continuously released multiple bioactive factors, effectively attenuated inflammation and oxidative stress, promoted angiogenesis and COL remodeling, and accelerated wound closure and re-epithelialization. This hydrogel demonstrated sustained therapeutic efficacy following a single administration in a diabetic wound animal model, highlighting its potential as a promising biomaterial platform for further translational investigation in chronic wound treatment.

Background: Chronic diabetic wounds, particularly diabetic foot ulcers (DFUs), are prone to recurrent infection and delayed healing, resulting in substantial morbidity, mortality, and economic burden. Multifunctional wound dressings that combine antibacterial, antioxidant, conductive, and self-healing properties may help to address the complex microenvironment of chronic diabetic wounds. Methods: In this study, ε-poly-L-lysine and amino-terminated polyethylene glycol were grafted onto carboxylated single-walled carbon nanotubes (SWCNTs) via amide coupling to obtain ε-PL-CNT-PEG. Aminated chondroitin sulfate (CS-ADH) and a catechol–metal coordination complex of protocatechualdehyde and Fe 3+ (PA@Fe) were then used to construct a dynamic covalently cross-linked hydrogel network through Schiff-base chemistry. The obtained hydrogels (Gel0–3, Gel4) were characterized for photothermal performance, rheological behavior, microstructure, swelling/degradation, adhesiveness, antioxidant capacity, electrical conductivity, cytocompatibility, hemocompatibility, and antibacterial activity in the presence and absence of near-infrared (NIR, 808 nm) irradiation. Results: ε-PL-CNT-PEG showed good aqueous dispersibility, NIR-induced photothermal conversion, and improved cytocompatibility after surface modification. Incorporation of ε-PL-CNT-PEG into the PA@Fe/CS-ADH network yielded conductive hydrogels with porous microstructures and storage modulus (G′) higher than loss modulus (G′′) over the tested frequency range, indicating stable gel-like behavior. The hydrogels exhibited self-healing under alternating strain and macroscopic rejoining after cutting. Swelling and degradation studies demonstrated pH-dependent degradation, with faster degradation in mildly acidic conditions (pH 5.0), mimicking infected chronic diabetic wounds. The hydrogels adhered to diverse substrates and tolerated joint movements. Gel4 showed notable DPPH• and H 2 O 2 scavenging (≈65% and ≈60%, respectively, within several hours). The electrical conductivity was 0.19 ± 0.0X mS/cm for Gel0–3 and 0.21 ± 0.0Y mS/cm for Gel4 (mean ± SD, n = 3), falling within the range reported for human skin. In vitro, NIH3T3 cells maintained >90% viability in the presence of hydrogel extracts, and hemolysis ratios remained below 5%. Hydrogels containing ε-PL-CNT-PEG displayed enhanced antibacterial effects against Escherichia coli and Staphylococcus aureus , and NIR irradiation further reduced bacterial survival, with some formulations achieving near-complete inhibition under low-power (0.2–0.3 W/cm 2 ) 808 nm irradiation. Conclusions: A dynamic, conductive hydrogel based on PA@Fe, CS-ADH, and ε-PL-CNT-PEG was successfully developed. The hydrogel combines photothermal antibacterial activity, antioxidant capacity, electrical conductivity, self-healing behavior, adhesiveness, cytocompatibility, and hemocompatibility. These properties suggest potential for application as a wound dressing for chronic diabetic wounds, including diabetic foot ulcers, although further in vivo studies are required to validate therapeutic efficacy.
