This paper described that Fe3O4 nanoparticles were encapsulated with methyl methacrylate (MMA) using linolenic acid (LA) as a crosslinking agent, after which the resulting polymethyl methacrylate (PMMA) embed Fe3O4 nanoparticles (PMMA/Fe3O4) were coated with silica, forming SiO2/(PMMA/Fe3O4) core–shell structure particles. Then these magnetic nanoparticles (MNPs) were applied in the developed system of chemiluminescent magnetic enzyme-linked immunoassay. E. coli O157:H7 was sandwiched between rabbits anti-E. coli O157:H7 polyclonal antibody-coated magnetite nanoparticles (immunomagnetic nanoparticles or IMNPs) and mouse anti-E. coli O157:H7 monoclonal antibody (E. coli O157-McAb). Commercial alkaline phosphatase conjugated horse anti-mouse immunoglobulin (ALP-Ab) was used to bond with the monoclonal antibody, finally the chemiluminescent signals were detected by adding 3-(2′-spiroadamantane)-4-methoxy-4-(3″-phosphoryloxy)phenyl-1,2-dioxetane (AMPPD) which was the substrate reagent of ALP. The specificity and sensitivity of this system for detecting E. coli O157:H7 were researched. The results indicated that this method was of good specificity when using E. coli Top 10F′ and Vibrio cholera as negative controls. The detection limit was 103 cells mL−1 when the antigen solution was 1 mL, and the procedure duration was about 3 h.

Functional semiconductor nanomaterials modified TiO 2 fibrous membranes with desirable heterostructures, good mechanical properties, and superior visible-light-driven photocatalytic activity would have broad applications in environmental remediation; however, constructing such fantastic fibrous membranes still remains an enormous challenge. Herein, we fabricated a soft and heterostructured g-C 3 N 4 @Co-TiO 2 (CNCT) nanofibrous membranes by a facile electrospinning approach and subsequent thermal polymerization process. The ultrathin g-C 3 N 4 nanoshell was in situ synthesized and uniformly wrapped onto Co-TiO 2 nanofiber to form core-shell quantum heterojunction , and the thickness and loading amount of g-C 3 N 4 nanoshell can be precisely controlled through simply regulating the content of precursor (melamine). Benefiting from the three-dimensional porous networks, enhanced visible-light response, and the effective charge transfer induced by uniform and compact heterojunction, the as-synthesized CNCT membranes exhibited a prominent photodegradation efficiency of 90.8% within 60 min towards tetracycline hydrochloride, and also displayed excellent antibacterial performance with a 6 log inactivation of E. coil after 90 min visible light exposure. Moreover, the stable core-shell structures and robust mechanical strength also endow the membranes with favorable reversibility and easy-recycling ability. This study may open up new avenues for designing and constructing flexible high-performance photocatalytic membranes for water purification.

A fiber optic surface plasmon resonance (FOSPR) sensor was developed for detection of Escherichia coli O157:H7 ( E. coli O157:H7) in water and juice, based on antimicrobial peptides (AMP), Magainin I, as recognition elements and silver nanoparticles-reduced graphene oxide (AgNPs-rGO) nanocomposites assisted signal amplification . The uniform AgNPs-rGO was fixed on the surface of optical fiber and covered with gold film. Not only was the SPR response greatly enhanced, but also the AgNPs was prevented from being oxidized. The FOSPR showed a sensitivity of about 1.5 times higher than that fabricated only with gold film. In the assay, Magainin I, immobilized on the surface of gold film, could specifically capture E. coli O157:H7, resulting in the wavelength shift of the SPR absorption peak. Under the optimized conditions, the SPR resonance wavelength exhibited a good linear relationship with natural logarithm of the target bacteria concentration in the range of 1.0 × 10 3 to 5.0 × 10 7 cfu/mL with the detection limit of 5.0 × 10 2 cfu/mL (S/N = 3). The FOSPR sensor showed good specificity for E. coli O157:H7 detection compared to other bacteria similar to the target bacterial species . Furthermore, the FOSPR sensor was successfully applied to the detection of E. coli O157:H7 in water, fruit and vegetable juice with the satisfactory recoveries of 88–110%. This assay for E. coli O157:H7 detection possesses high sensitivity, good selectivity , reproducibility and stability. In addition, the AMP based SPR biosensing methodology could be extended to detect a wide variety of foodborne pathogens. Therefore, the versatile method might become a potential alternative tool in food analysis and early clinical diagnosis.

Advanced oxidation processes are often applied to a variety of refractory wastewater, but rarely is the electro-Fenton process combined with the activated persulfate process applied to printing and dyeing wastewaters. In this study, we have delicately designed a new advanced oxidation process for the degradation of persistent pollutants in wastewater. The zero-valent iron (ZVI) activated peroxymonosulfate process coupled with an electro-Fenton (EF-PMS) oxidation process is applied to treat methylene blue (MB) wastewater. Its main advantage is to generate more reactive oxygen species (ROS) and reduce the cost of oxidants. Experimentally, it was found that the degradation rate of the MB by the EF-PMS process was significantly higher than that before the coupling, and the mineralization rate can reach 56.84% within 2 h. According to free radical scavenging experiments and electron spin resonance (ESR) spectroscopy, the main ROS in the EF-PMS process were identified as ˙OH, SO4˙− and 1O2, where free radical oxidation played an important role in the removal of MB, accounting for 70.39% of the total removal efficiency. The mechanism of ROS generation and action in the EF-PMS process was investigated, and the pathway of MB degradation was determined by the identification of intermediates. The EF-PMS process was also effective in degrading MB in different water quality backgrounds as well as for process reuse. The removal efficiency of other organic compounds was also evaluated. The results indicate that the EF-PMS process has good potential for application.

Glycans play important roles in the activity and function of monoclonal antibodies (mAbs). In this study, an isotope labeling method for the relative quantitative analysis of glycans in cetuximab, a chimeric human/mouse IgG1 monoclonal antibody that specifically targets epidermal growth factor receptor, via hydrophilic interaction LC-ultra-high-performance LC-HRMS was established based on Pronase E digestion. To this aim, novel isotope MS probes, i.e., 3-benzoyl-2-oxothiazolidine-4-carboxylic acid (d0-BOTC) and 3-(2,3,4,5,6-pentadeuterio-benzoyl)-2-oxothiazolidine-4-carboxylate acid (d5-BOTC), which include a carboxyl group to target the amino functional group in glycosylamine, were developed. The nonspecific Pronase E enzyme could simultaneously digest the peptide bound to the N - and O -glycans into glycosylamine having only one amino acid. Since the mass difference between the light- and heavy-labeled glycans was 5.0 Da, the relative abundance of their MS peaks was used to achieve the qualitative and relative quantitative analysis of glycans. Sialylglycopeptide was used as a complex glycan model to validate the accuracy of the method. The results demonstrated the good linearity (R 2 ≥ 0.9994) between the experimentally detected MS intensity ratios and the theoretical molar ratios of the d0-BOTC to the corresponding d5-BOTC derivatives in the dynamic range of 0.03-10 and 0.03-20 of three orders magnitude for the d5-BOTC/d0-BOTC ratios. The reproducibility was between 0.16% and 10.70%, and the limit of detection was 13 fmol. The feasibility of the relative quantification method was investigated by analyzing the glycan content in cetuximab, finding good consistency between experimental and theoretical molar ratios (5:1, 3:1, 1:1, 1:3, 1:5) of d0/d5-BOTC-labeled glycans. Finally, 13 glycans were successfully identified in cetuximab by applying this method using an in-house Tracefinder database. This study provides a novel strategy for the high throughput analysis, identification, and functional study of glycans in mAbs.

H 2 O 2 production by 2 electron water oxidation reaction (2e - WOR) is a promising approach, but still lacks mechanistic studies especially from a dynamic point of view. The interfacial water behavior on the solid/liquid interface is believed to play a crucial role in WOR activity. Here, F modification was used to accommodate the structure and dynamic process of interfacial water on WO 3 surface and boost the 2e- WOR activity. Electrochemical measurement, Raman spectra and computational techniques were used to investigate the mechanism. F modification was proved to guide the dipole moments of interfacial water molecules towards WO 3 surface, meaning more difficult to flip interfacial water molecules to perform the first step of WOR, in turn resulting in easy desorption of HO* and O-O*, thus benefiting for 2 e - WOR. This work paves a novel insight to understand mechanism of 2 e - WOR and rationally design the electrocatalysts for H 2 O 2 production from water.

8-Epidiosbulbin E acetate (EEA), a furan-containing diterpenoid lactone , is one of main component of Dioscorea bulbifera L. (DBL). It has been reported that EEA induces severe hepatotoxicity in mice and that its hepatotoxicity is associated with metabolic activation. The present study demonstrated that exposure to EEA (50, 100 or 200 μM) induced DNA damage, including significant DNA fragmentation, increases of tail DNA and olive tail moment, H2AX phosphorylation and PARP-1 activation, in cultured mouse primary hepatocytes. Similar observation was obtained in mice administered EEA at 50, 100 or 200 mg/kg. Pre-treatment with 10 μM ketoconazole (KTC), 200 μM vitamin C (VC), or 200 μM glutathione ethyl ester (GSH-OEt) reversed the over-production of reactive oxygen species (ROS) induced by EEA and attenuated susceptibility of hepatocytes to EEA-induced cytotoxicity and DNA damage in mouse primary hepatocytes. In contrast, pre-treatment with 1.0 mM L-buthionine sulfoximine (BSO) potentiated over-production of ROS, cytotoxicity and DNA damage induced by EEA. In summary, EEA induced DNA damage in cultured primary hepatocytes and the liver of mice. ROS, possibly along with DNA alkylation , participated in the observed DNA damage.

BACKGROUND The relative low stability, reusability and activity of enzymes made the industrial production of vitamin E succinate (VES) can only be performed with complex processes and high cost using chemical methods. To address these issues, in the present study, an ionic liquids (ILs) modification strategy was developed to improve the activity and stability of lipases in VES synthesis. RESULTS The results showed that the [1-butyl-3-methyl imidazole] [ N -acetyl- l -proline] ILs modified Candida rugosa lipase (CRL) has the highest modification degree (48.28%), activity (774 U g −1 ), thermostability and solvent tolerance in three selected modifiers. Additionally, after reaction condition optimization, the highest yield of VES can be improved to 95.18% at 45 °C for 15 h, which was significantly improved compared to some previous studies. CONCLUSION In the present study, a high-efficiency VES synthesis strategy was successfully developed via modification of lipase. Moreover, the mechanism by which ILs modification can enhance the activity and stability of lipase was investigated via both experimental and computational-aided methods. Molecular dynamics simulation suggested that ILs modification changed the geometry of Phe344 from flat to upright, which significantly reshaped and enhanced the size of substrate binding pocket of CRL. It is also agreement with our circular dichroism and fluorescence spectroscopy results, which suggested that the modification changed the secondary structure of CRL to a certain extent. The larger pocket also endowed the suitable binding pose of succinate, which made the hydrogen bonds between succinate and active site Ser209 become stronger, and thus improving the yield of VES. © 2023 Society of Chemical Industry.

Both plastic (e.g., polyvinyl chloride (PVC)) solid waste and bacteria/heavy metal ions-containing wastewater pose a severe threat to the environment, which must be addressed simultaneously by developing highly efficient and sustainable strategies. Novel cationic PVC nanofibrous membranes with high filtration and adsorption capability were fabricated by electrospinning and surface grafting, and then employed for the removal of bacteria (e.g., E. coli ) and hexavalent chromium (Cr (VI)) ions for wastewater treatment. A cationic pendant, methylimidazolium, was covalently anchored on the surface of PVC nanofibers and verified using FTIR-ATR, NMR, and SEM measurements. As a result, the optimized cationic PVC nanofibrous membrane (PVC-MIM) exhibited remarkable filtration efficiency for E. coli (LRV ≥ 6.1) from contaminated water, and a maximum adsorption capacity of 285.7 mg/g against Cr (VI). Kinetics, isotherms, and thermodynamics of the nanofibrous PVC-MIM membrane in the adsorption process were extensively investigated. The separation mechanism was proposed based on size exclusion, charged interactions, and hydrogen bonding/coordination between the membrane and the targets. Used sewer pipes and electric cables were employed as raw materials to fabricate modified nanofibrous membranes (0.9 g) to then manufacture of a spiral-wound PVC-MIM nanofibrous membrane cartridge. The resulting system exhibited a retention rate as high as 100 % when 24 L of simulated tanning wastewater containing concentrated suspended solids, oil, ammonia–nitrogen, chloride, and other pollutants were tested for a dynamic adsorption process. Furthermore, the nanofibrous membrane cartridge could be recycled ten times while the regeneration ratio remained as high as 91.0 %.

To overcome cancer drug resistance and to reverse tumor immunosuppression, a vitamin E-tocopheryl polyethylene glycol succinate (TPGS) decorated polymer-based drug delivery system was developed. The effects of the drug delivery system on diverse proteins involved in cell apoptosis, cancer invasion, metastasis and immunosuppression were investigated to provides a more comprehensive understanding of TPGS modified drug delivery systems. Curcumin (CUR), a hydrophobic drug, was loaded in star poly(DL-lactide) with a cholic acid core, and then decorated with TPGS shell to obtain [email protected] /star poly(DL-lactide) nanoparticles ( [email protected] ). Cholic acid cored star poly(DL-lactide) is featured by a rapid degradation rate with a surface erosion characteristic. TPGS not only reverses tumor drug resistance through downregulating P-gp expression of tumor cells but also enhances the anti-tumor efficiency through combined TPGS/CUR therapeutic actions on reversal of tumor immunosuppression mediated by CD47 and PD-L1. As compared with the CUR loaded nanoparticles without TPGS decoration, [email protected] with TPGS decoration results in significantly enhanced intracellular drug accumulation to efficiently induce cell apoptosis in drug resistant tumor cells. More importantly, [email protected] possesses considerably improved efficacy in upregulating p53 and p21 as well as downregulating β-catenin, MMP-9, Snail, CD47 and PD-L1, indicating the TPGS decorated drug delivery system inhibits tumor development and metastasis much more efficiently.

Background The tick Haemaphysalis longicornis (Neumann) is a well-known vector of numerous pathogens of veterinary and medical importance. Various control strategies, including the use of synthetic pesticides, have been developed to control this tick species. However, demand for effective and safe alternative pesticides is increasing due to the adverse effects associated with the intensive and injudicious use of synthetic pesticides, which include undesirable effects on non-target species and environmental pollution. Hence, the acaricidal activity of the extract and the essential oil of Cinnamomum cassia (Chinese cinnamon) and their major components, and the underlying mechanisms of this activity, were evaluated against unfed larvae and nymphs of H . longicornis . Methods The components of the extract and essential oil of C. cassia were determined by gas chromatography-mass spectrometry, and their larvicidal and nymphicidal activity were evaluated using the larval and nymphal packet test. The underlying detoxification mechanism was elucidated by targeting in vivo esterase and monooxygenase activity, and the toxicological effect was assessed on non-target Tenebrio molitor and Harmonia axyridis by topical application in open Petri dishes. Results ( E )-cinnamaldehyde was the predominant component of the extract (50.79%) and essential oil (89.95%). The 50% lethal concentration (LC 50 ) for larvae and nymphs treated with the extract was 11.56 and 49.18 mg/mL, respectively. The essential oil, ( E )-cinnamaldehyde and fenvalerate exhibited acaricidal activity, with LC 50 values of 3.81, 3.15, and 0.14 mg/mL, respectively, against the larvae, and 21.31, 16.93, and 1.89 mg/mL, respectively, against the nymphs. ( E )-cinnamaldehyde significantly increased esterase and monooxygenase activity in both larvae and nymphs. Unlike fenvalerate, C . cassia essential oil and ( E )-cinnamaldehyde did not cause mortality of T . molitor or H . axyridis adults. Conclusions This study demonstrates that C . cassia essential oil and ( E )-cinnamaldehyde have the potential to be developed into botanical-based larvicidal and nymphicidal agents for tick control. Ischemia/reperfusion (I/R) induces prolongation of QT interval and action potential duration (APD), which is a major cardiac electrical disorder in patients with arrhythmias. However, the mechanism of QT interval prolongation induced by I/R remains unclear. In the present study, we hypothesized that the rapid component of delayed rectifier potassium ( I Kr ) channel plays an important role in I/R-induced QT interval prolongation. We observed a marked attenuation of I Kr and a significant prolongation of action potential duration (APD) in a simulated I/R system with sodium dithionite (Na 2 S 2 O 4 ) in ventricular myocytes of guinea pigs. The I Kr current density was inhibited by 64% and APD increased by 87% respectively. Moreover, the inhibition of I Kr is primarily ascribed to overproduction of reactive oxygen species (ROS) by I/R, which can be partly reversed by antioxidant vitamin E (100 μmol/L). The value of I Kr tail current density increased from 0.516 ± 0.040 pA/pF in I/R to 0.939 ± 0.091 pA/pF when treated with vitamin E. Moreover, we also demonstrated that QTc interval was increased by I/R and reversed by Vitamin E in isolated guinea pig hearts. In conclusion, the inhibition of I Kr is one of the underlying mechanisms of prolongation of QT interval and APD in I/R. Vitamin E might have a benefit in coronary reperfusion therapy .
