Competitive adsorption-triggered release of cinnamaldehyde from ZSM-5/ZIF-8 nanocarriers in sodium alginate films for early-stage fish preservation

Peng Shan,Hong Wang,Zhihang Xu,Yilin Wang,Chenglong Fu,Jinwen Hu,Yehan Tao,Xiaodong Xia,Jian Du,Haisong Wang

An adsorption-triggered mechanism was proposed for on-demand release. • Acidic sites on ZSM-5/ZIF-8 possess stronger adsorption energy for biogenic amines than CA. • The mechanical and gas barrier of film were improved after introducing ZSM-5/ZIF-8. • The developed packaging effectively prolonged fish preservation at 4 °C. Active packaging with on-demand release capability has attracted increasing interest in food preservation; however, the delayed response of active agents often compromises protection during the early stage of spoilage. In this work, an adsorption-triggered release strategy was proposed by incorporating cinnamaldehyde (CA)-loaded ZSM-5/ZIF-8 nanoparticles (CA@ZSM-5/ZIF-8) into a sodium alginate (SA) matrix. Owing to the strong affinity of biogenic amines toward the adsorption sites of ZSM-5/ZIF-8, low concentrations of histamine could competitively displace the preloaded CA, thereby enabling an early and responsive release behavior. In the absence of histamine, the cumulative release of CA from the composite film remained below 10% within 12 h, whereas it increased to approximately 60% in the presence of 5.0 mM histamine. Meanwhile, the optimized film exhibited improved tensile strength (54.6 MPa), water vapor barrier performance (0.86 × 10 −10  g·m·m −2 ·s −1 ·atm −1 ), UV-shielding capability, and water resistance. The film also showed pronounced antioxidant activity (91.4%), sustained antibacterial effects against E. coli and S. aureus , and favorable biodegradability. When applied to fish preservation, the optimized film effectively extended the refrigerated shelf life to 6 days at 4 °C. This study provides a novel paradigm for on-demand release in intelligent food packaging.

Developing an Analytical Framework for the Molecular Weight Analysis of Fish-Derived Collagen Peptides in Beverages: A Multi-Method Comparative Study

Mengmeng Feng,Yali Li,Yongkang Luo,Yanfei Jiang,Chunyue Zhao

Accurate determination of the molecular weight of collagen peptides is crucial for quality control in functional beverages. Current national and industry standard methods for peptide powders face challenges of matrix interference and insufficient comparability when applied to complex beverage systems. In this study, three mainstream chromatographic methods were systematically evaluated for their suitability, reproducibility, and consistency in determining the molecular weight (MW) of fish-derived collagen peptides in beverages. An analytical framework was established by identifying and removing interference peaks from non-peptide excipients, followed by systematic comparison of linear and cubic fitting models. All three methods exhibited good reproducibility. After interference peak removal, the peptide profiles of the beverages were highly consistent with those of pure peptide powder. Cubic fitting reduced inter-method differences, with relative standard deviation (RSD) values below 5% in the bioactive < 1 kDa range. The framework was further validated through beverage formulation optimization, demonstrating compatibility between functional ingredient quality control and sensory flavor adjustability. The proposed approach provides a practical solution for industry quality control and cross-laboratory data comparability of collagen peptide beverages.

High-yield production of medium-chain fatty acids (MCFAs) via engineered Escherichia coli metabolic pathways for flavor enhancement of dried-fish

Yakun Zhao,Jian Chen,Huan Li,Xi Zhao,Zhixiang Wang,yuan Yao,Xuye Lang,Yanbo Wang

Medium-chain fatty acids (MCFAs) are high-value flavor enhancers critical for improving the sensory quality of processed foods like dried-fish. Traditional plant-derived MCFAs production faces sustainability limitations, necessitating efficient microbial alternatives. Here, we engineered Escherichia coli ( E. coli ) for high-yield MCFAs synthesis via targeted metabolic rewiring. In this study, we optimized a platform based on de novo fatty acid synthesis to increase the production of MCFAs. Using the pTargetF and pCas dual plasmid knockout system, single gene knockout was sequentially performed on E . coli , they were IdhA and poxB genes, which consume pyruvate, as well as the pta gene, which consumes acetyl CoA, and the aas gene that can reduce the toxicity of MCFAs to the host, followed by multi gene combination knockout. All combination knockout strains express thioesterase gene (CpFatB 1 ). The best-performing strain, strain MG9 (MG1655△ aas △ pta [pTrc99a-CpFatB 1 ]), was selected for further optimization. After 144h of cultivation with D-fructose, 680 mg/L of C8 fatty acid was obtained. We explored purification methods for C8 fatty acid, and purified C8 fatty acid reached 99.75% purity with 82.15% recovery yield. Crucially, applying these biosynthesized C8 fatty acid to industrial fish drying elevated the kinds of flavor compounds similarity to traditional products from 38.57% to 68.57%. This work establishes a sustainable platform for high-purity MCFAs production and demonstrates their direct utility in enhancing premium food flavors, offering a scalable solution for the food industry.

Efficient quenching activity and thermostability of N-acyl homoserine lactonase AiiA against fish spoilage mediated by Pseudomonas fluorescens via rational design

Jiashi Li,Zhong Lu,Xiaoli Zeng,Hanxiao Zhang,Haixia Lu,Lifang Feng,Junli Zhu

Quorum quenching through natural and nontoxic N -acyl homoserine lactonase (AHL-lactonase) has been regarded as a promising approach for inactivating QS-mediated bacterial spoilage. However, their poor catalytic performance remains a major challenge for practical application. To address these limitations, we developed an efficient rational design strategy for AHL lactonase AiiA to obtain a variant with high catalytic efficiency and a broad substrate spectrum, based on computer-aided structural analysis, molecular dynamics simulation, and energy calculation. Results showed that three highly flexible regions in AiiA were identified as vital sites to recognize and position different substrates, possessing high engineering potential. Among the obtained 10 mutants, AiiA A130T exhibited the strongest degradation against short-chain AHL (C 4 -HSL and C 6 -HSL), along with improved thermostability, compared to wild-type enzyme (AiiA WT ). In Pseudomonas fluorescens PF07, a majority of AHLs were quenched to undetectable levels by AiiA A130T , and the lactone ring of C 4 -HSL was verified to hydrolyze and produce form butyryl-L-homoserine. Its biofilm biomass, exopolysaccharides, exoprotein contents, and exoprotease activity were effectively repressed by 64.63 %, 48.86 %, 51.85 %, 61.50 % by AiiA A130T , respectively, in contrast with 46.51 %, 29.08 %, 23.55 %, 19.5 % by AiiA WT , without affecting bacterial growth. Moreover, the transcription levels of 16 crucial QS and biofilm-related genes in PF07 were dramatically downregulated ( P  < 0.05), as evidenced by qRT-PCR. In contrast with the slight preservation for AiiA WT , AiiA A130T effectively retarded the fillet deterioration of large yellow croaker and extended their shelf life by 2 days. Overall, this study presents an efficient strategy for modifying AHL lactonase and yields a high-performance mutant, offering the application potential of AHL lactonase for controlling QS-mediated microbial spoilage of seafood.

ZnWO4/f-MWCNTs Nanocomposite for H2S Gas Sensor with Fast Response and Recovery: Mechanism Study and Application for Detecting Fish Spoilage

Zhihua Zhao,Shixin Ma,Yao Wang

Hydrogen sulfide (H2S), a characteristic byproduct of sulfur-containing amino acid decomposition during fish spoilage, serves as a critical freshness indicator and poses substantial food safety risks. This study develops a novel H2S gas sensor using a ZnWO4/f-MWCNTs nanocomposite synthesized via one-step hydrothermal method. The incorporated functionalized multiwalled carbon nanotubes (f-MWCNTs) improve the sensing material’s conductivity, dispersion stability, and gas adsorption capacity. XRD, XPS, FTIR, SEM, TEM, and BET analyses verify the composite’s successful synthesis and optimal physicochemical properties. The ZnWO4/f-MWCNTs sensor demonstrates exceptional H2S detection at 120 °C, achieving a response value of 18.67 (Ra/Rg) to 50 ppm of H2S with fast response/recovery times (14 s/30 s) and excellent selectivity against interferents. Real-time monitoring of fish spoilage further confirms its practical utility, highlighting the sensor’s potential for meat freshness assessment and food safety applications.

Dietary paeonol addition relieves the detrimental effect of waterborne cadmium on fish growth and liver health in largemouth bass (Micropterus salmoides)

Zhang Zhenyang,Chen Weijun,Shen Jiaqi,Zhang Chunnuan,Gao Xiaochan,Ren Hongtao,Huang Yong,Gao Shiyang

This study aimed to explore the effect of paeonol (PAE) on growth performance and liver health in largemouth bass ( Micropterus salmoides ) exposed to cadmium (Cd). Five experimental groups were designed: one control group reared in clean water and fed a basal diet (CONT), alongside four cadmium-exposed groups maintained in 2.75 mg/kg Cd-containing water and fed the basal diet supplemented with 0 mg/kg (Cd), 100 mg/kg (PAE100-Cd), 200 mg/kg (PAE200-Cd), and 400 mg/kg PAE (PAE400-Cd), respectively. The experiment lasted for 56 days. The results showed that Cd exposure significantly inhibited the growth performance of largemouth bass, caused liver dysfunction and pathological tissue damage, led to oxidative stress, inflammatory response, and apoptosis, and induced the expression of the epidermal growth factor receptor (ErbB1)-extracellular signal-regulated kinase (ERK) pathway genes. However, PAE supplementation (200–400 mg/kg) significantly improved the growth performance of largemouth bass. In addition, 200–400 mg/kg PAE reduced the serum levels of hepatic injury biomarkers (AKP, ALT, LDH, and AST) and alleviated histopathological lesions. Furthermore, PAE200-Cd increased the activities of superoxide dismutase (SOD) and catalase (CAT) and reduced malondialdehyde (MDA) levels in the liver. Moreover, PAE200-Cd and PAE400-Cd down-regulated the expression of interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α), nuclear factor kappa-B p65 (NF-κB p65), ErbB1, and cellular FBJ osteosarcoma oncogene (C-fos) in the liver. PAE200-Cd significantly increased the expression of cytochrome P450 family 1 subfamily A (CYP1A1) and downregulated the expression of metallothionein (MT) and cysteine-aspartic proteases-9 (Caspase-9). The optimal PAE dose for counteracting waterborne Cd-induced growth impairment and hepatic oxidative stress was 173.37–173.90 mg/kg for largemouth bass.

Enhancing garlic-clove shaped gel properties of mandarin fish (Siniperca chuatsi) myofibrillar protein with prickly ash extracts: Underlying roles of polyphenols

Wanmei Li,Yingqin Zhou,Haiqiu Wei,Kexin Xiong,Yan Yan,Jingjing Huang,Ningning Xie

The potential role of prickly ash extracts (PAEs) in enhancing garlic-clove shaped protein gel (GSPG)-forming capacity of mandarin fish ( Siniperca chuatsi ) myofibrillar protein (MP) was studied. PAEs with 0%, 20%, 60%, and 100% ethanol were utilized to systematically analyze changes in the physicochemical properties, water retention, chemical forces, and protein structure of GSPG. Results demonstrated that PAE60 (38.09 mg gallic acid equivalent/g) significantly improved gel strength and water-holding capacity compared to the control ( P  < 0.05). The immobilized water (23.04% decrease of relaxation time), hydrogen bonds (56.74% increase), disulfide bonds (174.31% increase), and β -sheet (49.42% increase) became significantly better than the control ( P  < 0.05). The gel microstructure had more orderly and compact networks with relatively increases of fractal dimension (1.83%), porosity (22.32%), and number of pores (23.66%) with statistical significances ( P  < 0.05). The findings could lay a theoretical groundwork for improving garlic-clove shaped fish protein with plant polyphenols.

Selective and sensitive determination of chlorophenol disinfection by-products in fish by defective MIL-101(Fe)-NH2 membrane SPE-HPLC-UV

Xueyun Wei,Lanrui Yang,Zhuomin Zhang,Gongke Li

Trace disinfection by-products (DBPs) in aquatic products may have serious impact on human health via direct or indirect consumption. However, the accurate and sensitive quantification of trace chlorophenol DBPs in complex aquatic products still remains a great challenge. In this work, the defective MIL-101(Fe)-NH 2 enrichment medium was prepared for developing a selective membrane solid phase extraction (M-SPE) technology for quantifying trace chlorophenol DBPs in fish, coupled with high performance liquid chromatography-ultraviolet-visible spectroscopy (HPLC-UV). The established analytical method showed good selectivity, sensitivity and accuracy, achieving the limits of detection from 0.03 to 0.15 ng/mL and the recovery from 80.0% to 117%, achieving sensitivity comparable to that of HPLC-mass spectrometry (MS)-based methods. The validated method was successfully applied to quantify chlorophenol DBPs in grass carp and crucian carp, offering a selective and sensitive alternative for monitoring chlorophenol DBPs in fish, with practical advantages for routine analysis via compatibility with widely available HPLC-UV.

Chitin nanocrystal/carboxymethyl cellulose bilayer emulsion with rigid inner layer and flexible outer shell for ultra-stable encapsulation of fish oil

Yufei Sun,Qianxi Deng,Simeng Ji,Yuewen Li,Kao Wu,Ying Kuang,Fatang Jiang,Pengpeng Deng

The polyunsaturated fatty acids in fish oil are prone to oxidation and have poor dispersibility, which limits their use in the food sector. In this work, oil-in-water emulsions stabilized by chitin nanocrystals (ChNC) were prepared via high-speed homogenization. Anionic carboxymethyl cellulose (CMC) was assembled onto cationic ChNC-stabilized emulsion droplet surfaces via layer-by-layer self-assembly technology to construct ChNC/CMC (Ch-C) bilayer emulsions with rigid inner layer and flexible outer shell structures. The results of dynamic light scattering proved that the CMC could significantly reduce the emulsion droplet size. The root mean square roughness and arithmetic mean roughness of the emulsion droplets decreased from 10.5 and 8.4 nm to 5.6 and 4.4 nm with increasing CMC concentration, respectively. The droplet size distribution curve of the Ch-C emulsion with the CMC concentration of 1 % (Ch-C-4) after 30 days of storage changed little compared with the initial state, showing excellent storage stability. In addition, the Ch-C-4 emulsion was stable over the wide pH range (2–6), high temperature (90 °C), and high ionic strength (400 mM). Furthermore, compared with ChNC emulsion, the primary and secondary oxidation contents of Ch-C-4 decreased by 11.9 % and 23.7 %, respectively, suggesting that Ch-C bilayer emulsion can effectively delay the oxidation of fish oil. Therefore, this study proposes new research strategies for the design of polysaccharide-based bilayer emulsions and the protection of functional lipids.

Environmentally friendly solid-phase microextraction coupled with gas chromatography and mass spectrometry for the determination of biogenic amines in fish samples

Jie Huang,Ning Gan,Fangying Lv,Yuting Cao,Changrong Ou,Haiqing Tang

In this work, a facile and environmentally friendly solid-phase microextraction assay based on on-fiber derivatization coupled with gas chromatography and mass spectrometry was developed for determining four nonvolatile index biogenic amines (putrescine, cadaverine, histamine, and tyramine) in fish samples. In the assay, the fiber was firstly dipped into a solution with isobutyl chloroformate as derivatization reagent and isooctane as extraction solvent. Thus, a thin organic liquid membrane coating was developed. Then the modified fiber was immersed into sample solution to extract four important bioamines. Afterwards, the fiber was directly inserted into gas chromatography injection port for thermal desorption. 1,7-Diaminoheptane was employed as internal standard reagent for quantification of the targets. The limits of detection of the method were 2.98–45.3 μg/kg. The proposed method was successfully applied to the detection of bioamines in several fish samples with recoveries ranging 78.9–110%. The organic reagent used for extraction was as few as microliter that can greatly reduce the harm to manipulator and environment. Moreover, the extraction procedures were very simple without concentration and elution procedures, which can greatly simplify the pretreatment process. The assay can be extended to the in situ screening of other pollutant in food safety by changing the derivatization reagent.

A novel Zn/Eu-MOF for the highly sensitive, reversible and visualized sensing of ofloxacin residues in pork, beef and fish

Yan Wu,Yue Zhou,Hongchen Long,Xiangyu Chen,Yuanyuan Jiang,Lei Zhang,Tao Le

The study investigates a bimetallic organic framework (Zn/Eu-MOF) based fluorescent probe for visual detection of ofloxacin (OFL) in pork, beef and fish. The developed sensing probe recognizes OFL through internal filtration and cation-π interaction between OFL and Zn/Eu-MOF, resulting in a distinct color change from orange-red to light green. The content of OFL can be determined through RGB analysis by a mobile-phone. The developed sensing probe offers several advantages such as broad linear range (0.1 ∼ 80 μM), rapid response time (30 s), low detection line (0.44 μM). The effectiveness of the sensing probe can last for five rounds with good recovery. Moreover, the application of the sensing probe on pork, beef and fish samples are reliable, with recoveries ranging from 93.4 to 112.1%, and the relative standard deviations (RSD) within 1.17% to 2.06%. These results suggest that the developed sensing probe could have significant potential for practical on-site test in food.

Dummy Template-Based Molecularly Imprinted Membrane Coating for Rapid Analysis of Malachite Green and Its Metabolic Intermediates in Shrimp and Fish

Yi Zhang,Shaofeng Li,Yurong Gu,Jianying Zhang,Zhenfeng Yue,Liao Ouyang,Fengjuan Zhao

A novel malachite green molecularly imprinted membrane (MG-MIM) with specific selectivity for malachite green (MG) and leucomalachite green (LMG) was prepared using a hydrophobic glass fiber membrane as the polymer substrate, methyl violet as a template analog, 4-vinyl benzoic acid as the functional monomer, and ethyleneglycol dimethacrylate as the crosslinking agent. MG-MIM and non-imprinted membrane (NIM) were structurally characterized using scanning electron microscopy, surface area analyzer, Fourier-transform infrared spectrometer and synchronous thermal analyzer. The results showed that MG-MIM possessed a fluffier surface, porous and looser structure, and had good thermal stability. Adsorption properties of MG-MIM were investigated under optimal conditions, and adsorption equilibrium was reached in 20 min. The saturated adsorption capacities for MG and LMG were 24.25 ng·cm−2and 13.40 ng·cm−2, and the maximum imprinting factors were 2.41 and 3.20, respectively. Issues such as “template leakage” and “embedding” were resolved. The specific recognition ability for the targets was good and the adsorption capacity was stable even after five cycles. The proposed method was successfully applied for the detection of MG and LMG in real samples, and it showed good linear correlation in the range of 0 to 10.0 μg·L−1(R2= 0.9991 and 0.9982), and high detection sensitivity (detection limits of MG and LMG of 0.005 μg/kg and 0.02 μg·kg−1in shrimp, and 0.005 μg/kg and 0.02 μg/kg in fish sample). The recoveries and relative standard deviations were in the range of 76.31–93.26% and 0.73–3.72%, respectively. The proposed method provides a simple, efficient and promising alternative for monitoring MG and LMG in aquatic products. Keywords:malachite green molecularly imprinted membranes;template analogs malachite green;colloidal gold strip;residuals analysis
