Migration detection of six aromatic amines in polyamide food contact materials by HPLC after molecularly imprinted polymer pipette tip solid phase extraction

Yang Liu,Xueping Dang,Shicheng Zhang,Yuling Hu,Huaixia Chen

Some food contact materials (FCMs) may contain aromatic amines (AAs), which would migrate to food and pose a serious threat to human health. An extraction and determination method for AAs was established using pipette tip solid-phase extraction (PTSPE) combined with high performance liquid chromatography-photodiode array detector (HPLC-PAD). The PTSPE was based on molecularly imprinted polymer (MIP) monolith, which was synthesized in situ polymerization using benzidine as template molecule and 4-vinylpyridine as functional monomer . It was applied for selective extraction of six AAs from FCMs, including benzidine, 3,3′-dimethyl-4,4′-diaminodiphenylmethane, 2-methoxy-5-methylaniline, 3,3′-dimethoxybenzidine, 4,4′-methylenedianiline and 4-aminobiphenyl. The MIP monolith showed highly specific recognition and good adsorption ability for the six AAs, the imprinting factor for 4,4′-methylenedianiline can reach 4.29 and the adsorption capacity for 4-aminobiphenyl can reach 112.3 mg g −1 . The linear range of the determination method for the six AAs was 0.60–500 µg kg −1 (R 2 >0.997), the limits of quantitation was 0.60–4.50 µg kg −1 and the limits of detection (S/N = 3) was 0.20–1.50 µg kg −1 . The MIP-PTSPE-HPLC-PAD method was applied for the migration detection of six AAs from polyamide kitchenware and the absolute recovery were 81.7–110.9%.

Graphene oxide-based a network porous poly (trially isocyanurate-co-methacrylate) monolithic column for HPLC separation of aromatic molecular and lipopeptide antibiotics

Yuanyuan Guo,Helong Si,Hongya Li,Xinyao Zhao,Yuxin Zhao,Shuna Li,Quan Wang,Baocheng Zhu

Graphene oxide (GO) was introduced into a monolithic column and a network porous poly (GO-co-TAIC-co-MMA) monolith was prepared by redox polymerization. The internal morphology and pore size distribution of the polymer were observed by scanning electron microscopy, and the nitrogen adsorption-desorption and mercury intrusion methods. After optimization, 8 kinds of aromatic compounds were effectively separated in 5 min, and the theoretical plates number of the monolithic column exceeded 33, 070 plates m −1 . Five kinds of main ingredients were separated from the traditional Chinese medicine ( Schisandra ) ingredients and 26 peaks were successfully separated from the fermentation broth containing natural lipopeptide antibiotics. The addition of GO material enhanced the interaction between the compound and the monolithic column, increased the binding sites, improved the uniformity of the internal pore structure of the monolithic column, and improved the separation performance of the monolith. Methodologic validation of five ingredients in Schisandra showed that the correlation coefficients of the linear regressions were in the range of 0.9987–0.9997. The intra- and inter-day values of the relative standard deviation for precision were in the range of 0.6–4.1% and 1.1–4.8%, respectively. The values of accuracy (expressed as recovery) were in the range of 97.7–103.2%, 100.5–105.0%, 98.2–101.8%, 101.3–104.1%, and 101.2–103.3% for the 5 ingredients in order. In terms of the relative standard deviation of the retention time, the reproducibility of the monolithic column M1 was <3.7%. The monolithic column based on GO has great potential in chromatographic separation.

Potential Multifunctional Bioactive Compounds from Dysosma versipellis Explored by Bioaffinity Ultrafiltration-HPLC/MS with Topo I, Topo II, COX-2 and ACE2

Huixia Feng,Guilin Chen,Yongli Zhang,Mingquan Guo

Background Dysosma versipellis ( D. versipellis ) has been traditionally used as a folk medicine for ages. However, the specific phytochemicals responsible for their correlated anti-inflammatory, anti-proliferative and antiviral activities remain unknown. Purpose This study aimed to explore the specific active components in D. versipellis responsible for its potential anti-inflammatory, anti-proliferative, and antiviral effects, and further elucidate the corresponding mechanisms of action. Methods Bioaffinity ultrafiltration coupled to liquid chromatography–mass spectrometry (UF-LC/MS) was firstly hired to fast screen for the anti-inflammatory, anti-proliferative and antiviral compounds from rhizomes of D. versipellis , and then further validation was conducted using in vitro inhibition assays and molecular docking. Results A total of 12, 12, 9 and 12 phytochemicals with considerable affinities to Topo I, Topo II, COX-2 and ACE2 were fished out, respectively. The anti-proliferative assay in vitro indicated that podophyllotoxin and quercetin exhibited comparably strong inhibitory rates on A549 and HT-29 cells compared with 5-FU and etoposide. Meanwhile, kaempferol displayed prominent dose-dependent inhibition against COX-2 with IC 50 value at 0.36 ± 0.02 μM lower than indomethacin at 0.73 ± 0.07 μM. Furthermore, quercetin exerted stronger inhibitory effect against ACE2 with IC 50 value at 104.79 ± 8.26 μM comparable to quercetin 3- O -glucoside at 135.25 ± 6.54 μM. Conclusion We firstly showcased an experimental investigation on the correlations between bioactive phytochemicals of D. versipellis and their multiple drug targets reflecting its potential pharmacological activities, and further constructed a multi-target and multi-component network to decipher its empirical traditional applications. It could not only offer a reliable and valuable experimental basis to better comprehend the curative effects of D. versipellis but also provide more new insights and strategies for other traditional medicinal plants.

Novel hydrophobic deep eutectic solvent for vortex-assisted liquid phase microextraction of common acaricides in fruit juice followed by HPLC-UV determination

Setareh Rostami-Javanroudi,Masoud Moradi,Kiomars Sharafi,Nazir Fattahi

In the present research, several novel and natural hydrophobic deep eutectic solvents (DESs) were prepared using methyltrioctylammonium chloride (MTOAC) as the hydrogen bond acceptor (HBA) and different types of straight chain alcohols as hydrogen bond donors (HBDs). One of the DESs composed of MTOAC and n-butanol was advantageously used to develop a vortex-assisted liquid phase microextraction (VALPME) method combined with high-performance liquid chromatography-ultraviolet detection (HPLC-UV) for the determination of common acaricides in fruit juice samples. Several important parameters influencing extraction efficiency were investigated and optimized, including the type and volume of DES, sample solution pH, effect of salt addition and, extraction and vortex time. Under optimal experimental conditions, the method showed good linearity with the correlation coefficients (R2) of 0.9986–0.9991 in the linear range of 2–300 μg L−1, low limits of detection of 0.5–1 μg L−1 and acceptable extraction recoveries in the range of 85–93%. The proposed method was successfully applied for the extraction and preconcentration of trace acaricides in real fruit juice samples, and the results demonstrated the potential of the synthesized DESs for the extraction and determination of contaminants in aqueous samples.

A facile online multi-gear capacitively coupled contactless conductivity detector for an automatic and wide range monitoring of high salt in HPLC

Weiwen Liu,Ziqi Liang,Yuanyu Wang,Jun Cao,Qiang Zhang,Xiaoping Liu,Yuxing Wang,Chengxi Cao

Sensing the electrolyte solution or aqueous–organic mixture has attracted much interest in chemical separation, pharmaceutical engineering, bioprocess, and biochemical experiments. However, reports on online contactless sensor with automatic and wide range sensing of high content electrolyte have been rarely presented. Herein, a facile model and theory of online multi-gear capacitively coupled contactless conductivity detection (M-C4D) sensor was proposed using one excitation electrode and multiple detection electrodes. Further, the relevant digital computation based on the M-C4D theory was developed for parameter optimization: the electrode gap of 5–150 mm, inner radius of 0.25–0.75 mm, electrode length of 10–60 mm, and frequency of 40–250 kHz using MATLAB. To demonstrate the model, theory, and digital computation, liquid chromatography (LC) was chosen as the model of bioprocess, and the sensor was designed and used as an online sensing device for the automatic monitoring of high salt elution in LC. The experiments showed that (i) the detection results were in agreement with the digital data, validating the digital computation, theory, and model of M-C4D and (ii) the monitoring data of M-C4D were in agreement with those via the traditional meter, further validating the model and theory. Finally, the developed sensor was applied to the automated detection of high salt gradient in LC. In contrast to the currently used meters and C4D, the developed M-C4D sensor had the following merits: (i) facile and automatic online detection avoiding cumbersome manual switching of detector heads, (ii) fair linear range of 0.015–20 mS cm−1 (equivalently 0.1–159 mM KCl) that does not fit the range of traditional C4D, and (iii) fair accuracy of less than 1.50% relative error. All these results indicate that the developed model, theory, and sensor have potential for the process monitoring of high content electrolytes transfer in biochemical engineering and clinic pre-warning.

Simultaneous and rapid screening and determination of twelve azo dyes illegally added into food products by using chemometrics-assisted HPLC-DAD strategy

Ming-Yue Dong,Hai-Long Wu,Wan-Jun Long,Tong Wang,Ru-Qin Yu

Edible colorants, as an essential external condition of food products, can increase people’s appetite. Some businesses have used some cheap and illegal dyes instead of edible colorants to gain benefits. It not only deceives consumers, but also seriously endangers their health. In this work, two chemometric algorithms assisted high-performance liquid chromatography with diode array detection (HPLC-DAD) were proposed for screening and quantifying twelve azo dyes illegally added into different food products. All analytes were successfully eluted within 6.5 min. With the aid of the prominent “second-order advantage” of the chemometric algorithms, the overlapping peaks and unknown interferences were solved. The spiked average recoveries obtained by alternating trilinear decomposition (ATLD) and multivariate curve resolution-alternating least squares (MCR-ALS) were 82.8–111.5% with standard deviation (SD) less than 6.5% and 78.7–122.6% with SD less than 18.7%, respectively. The limits of detection were 0.01–2.56 mg kg −1 and 0.01–2.95 mg kg −1 , respectively. The results indicated that two algorithms had practical value for the simultaneous and rapid screening and determination of azo dyes in food products. Compared with other methods based on HPLC, the proposed method could flexibly handle different interferences, and had the advantages of high efficiency, simple operation and being environmentally friendly.

Covalent organic framework-LZU1@PEI@Fe3O4-based magnetic dispersive micro-solid phase extraction of tetracyclines from environmental water prior to HPLC analysis

Ying Zhang,Yuhan Ning,Wanliang Liao,Weiping Wang,Ai-Jun Wang

In this work, COF-LZU1@PEI@Fe3O4 was synthesized by immobilization of COF-LZU1 onto the surface of polyethyleneimine-functionalized Fe3O4 nanoparticles (PEI@Fe3O4) and employed as an adsorbent for magnetic dispersive micro-solid phase extraction of tetracyclines (TCs). COF-LZU1@PEI@Fe3O4 was characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction and nitrogen adsorption–desorption isotherms analysis. The optimal extraction and desorption conditions were as follows: 15.00 mL sample solution (pH 7.0) extracted with 5.0 mg of adsorbent for 30 min at 30 °C, and then desorbed with 1.50 mL methanol/acetonitrile/0.02 mol L−1 oxalic acid solution (v/v, 1 : 2 : 7). Good linearities were obtained between the peak area and TC concentration ranging from 5–500 μg L−1 with correlation coefficients (R2) higher than 0.9992 and limits of detection lower than 0.51 μg L−1. The relative standard deviations (RSDs) of intra-day and inter-day were less than 5.7% and 7.4%, respectively. The developed method was successfully applied to the determination of TCs in environmental water samples with recoveries in the range of 87.0–113.8% and RSDs less than 5.1%, suggesting great potential of COF-LZU1@PEI@Fe3O4 for efficient extraction and analysis of trace TCs in water samples.

Development of hydrophilic magnetic molecularly imprinted polymers for the dispersive solid-phase extraction of sulfonamides from animal-derived samples before HPLC detection

Xiaolei Zhao,Lei Lü,Mengzhen Zhu,Huilin Liu,Jinxing He,Fuping Zheng

Highly hydrophilic magnetic molecularly imprinted polymers were prepared through a surface imprinting technique for dispersive solid-phase extraction coupled with high-performance liquid chromatography to detect trace levels of ten sulfonamides in animal-derived samples. The obtained imprinted polymers were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, thermogravimetric analysis, and adsorption experiments, which exhibited excellent specific adsorption for template sulfamethazine in aqueous solution. Moreover, the adsorption process could be completed within 25 min. Under the optimum conditions, the method exhibits good linear performance in the range of 5–to 10 mg/L, limits of detection ranging from 0.57 to 1.50 μg/L, and good recoveries of 85.09–110.93% in the spiked samples (chicken, cow milk, and goat milk). The proposed detection method not only avoids the use of organic solvents but also simplifies the pretreatment procedure via excellent magnetic properties. Furthermore, the method shows great potential for the rapid detection of drug residues.

Task specific monolith for magnetic field-reinforced in-tube solid phase microextraction of mercury species in waters prior to online HPLC quantification

Xiaochong Song,Jiangyi Wu,Jinling Pang,Yuanfei Wu,Xiaojia Huang

In this study, a novel sorbent based on task specific monolith doped with Fe 3 O 4 was in situ fabricated in capillary and acted as the extraction medium of magnetic field-reinforced in-tube solid phase microextraction (MFR/IT-SPME) to trap and preconcentrate mercury species which were coordinated with dithizone to form chelates. Various characterization technologies evidenced that the obtained monolithic adsorbent presented porous and super paramagnetic properties, and possessed abundant functional groups. Results evidenced that the implementation of magnetic field during extraction stages enhanced the extraction efficiency of studied Hg chelates from 48.5% to 75.3% to 69.9–94.4%. Under the optimized extraction parameters, the introduced MFR/IT-SPME was online coupled to HPLC/DAD to quantify mercury species at ultra-trace levels in various water samples. Limits of detection varied from 0.0067 μg/L to 0.016 μg/L, and the RSDs for precision were below 7.5%. Additionally, related extraction mechanism was deduced and revealed multiple forces co-contributed to the enrichment. The reliability and accuracy of suggested online approach for speciation analysis of mercury was well proved by confirmatory experiments.

Layered porous organic frameworks as a novel adsorbent for the solid phase extraction of chlorophenols prior to their determination by HPLC-DAD

Wang Qianqian,Li Gaigai,Wang Chun,Wu Qiuhua,Wang Zhi

A layered porous organic framework (L-POF) with high surface area and porous volume was used as a novel adsorbent for adsorption of chlorophenols (CPs). The L-POF showed much higher adsorption capacity for four CPs (2-CP, 3-CP, 2,3-CP, 2,4-CP) compared with corresponding amorphous POF and some commercial adsorbents. Then, an L-POF-based solid-phase extraction method combined with high-performance liquid chromatography for analyzing CPs at the wavelength of 280 nm in water and honey samples was established. Under the optimum conditions, four CPs exhibited good linearity in the range 0.10–80.0 ng mL −1 for water and 2–400 ng g −1 for honey samples with determination coefficient r 2  > 0.996. The method has low limits of detection of 0.03–0.10 ng mL −1 for water and 0.5–1.0 ng g −1 for honey samples. The method presented satisfactory precision and accuracy with relative standard deviations ≤ 7.8% and recoveries of 80–115%. High enrichment factors of 123–190 for water and 63–97 for honey were obtained. The validated method was successfully used to determine CPs in water and honey samples. The L-POF also displayed prominent adsorption performance for many other dyes and organic contaminants. This suggests that it can be an ideal candidate as adsorbent for wide application in separation and analysis. Graphical Abstract

Dissipation behavior, residues distribution and dietary risk assessment of tembotrione and its metabolite in maize via QuEChERS using HPLC-MS/MS technique

Yue Su,Weijun Wang,Jiye Hu,Xiaolu Liu

The dissipation and residues of tembotrione in corn field application were investigated using liquid chromatography tandem mass spectrometry (LC-MS/MS) method. The average recoveries of tembotrione in maize, corncob, and straw were in the ranges of 98–107% with relative standard deviations (RSDs ≤9.3%), respectively. The recoveries of M5 was in the ranges of 90–108% in all three matrices of maize, with RSDs were 3.3–12.8%. The LODs for tembotrione and M5 in maize were 0.85 μg/L and 1.0 μg/L, 0.84 μg/L and 0.43 μg/L in corncob, 0.94 μg/L and 1.5 μg/L in straw, respectively. The LOQs of the method in maize grain, corncob and straw were 0.01, 0.01 and 0.05 mg/kg for both analytes, respectively. The dissipation of tembotrione in straw was in compliance with the first-order dynamic equation, with half-lives of 1.18–1.23 days at Beijing and Heilongjiang. Total residue of tembotrione in maize grain and corncob matrix were both below 0.02 mg/kg, lower than the max residue limit (MRL) recommended by european food safety authority (EFSA). Risk quotients (RQs) of this pesticide was assessed via comparing national estimated daily intake with acceptable daily intake . The dietary intake risk of tembotrione residue in maize was very low for all groups of Chinese residents. These data could provide scientific data and strategies and facilitate Chinese government to establish the MRLs of tembotrione.

Determination of fluoroquinolones in foods using ionic liquid modified Fe3O4/MWCNTs as the adsorbent for magnetic solid phase extraction coupled with HPLC

Meng Zhang,Jie Chen,Faqiong Zhao,Baizhao Zeng

In this work, a simple and sensitive method based on Fe3O4/multi-walled carbon nanotubes/ionic liquid (Fe3O4/MWCNTs/IL) as the adsorbent for magnetic solid phase extraction coupled with high performance liquid chromatography was developed for the determination of trace fluoroquinolones in foods. After a simple one-pot solvothermal synthesis, Fe3O4/MWCNTs were further modified with an amine-terminated ionic liquid to enhance their dispersibility and extraction capacity. The morphology, structure and magnetic properties of the composite adsorbent were characterized by transmission electron microscopy, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy, X-ray diffraction spectrometry and vibrating-sample magnetometry. Under the optimized extraction and detection conditions, the studied fluoroquinolones were enriched effectively and separated well and their UV signals were linear to their concentrations in the range of 4–1000 ng mL−1, with correlation coefficients ranging from 0.9958 to 0.9990. The enrichment factors were 29.1–43.9 fold and the limits of detection were 0.33–0.78 ng mL−1. The spiked recoveries were 85.4–105.9% for milk and 85.2–103.7% for pork samples, with relative standard deviations of 0.8–5.7%. The developed method provided a fast, sensitive and reliable determination platform for fluoroquinolones in complex real samples.
