Intercalated binding of anthracycline anticancer drugs to DNA is associated with anticancer mechanisms. The i-motif, a cytosine-rich DNA secondary structure found in the promoter region of oncogenes, is a primary target for anticancer drugs action. Therefore, it is significant to study the interaction between anthracycline drugs and i-motif for cancer therapy using simple, rapid and low-cost analytical methods. Here, we utilized α-HL protein nanopore, which offers high sensitivity and high signal-to-noise ratio to investigate the conformational transition of human telomere i-motif induced by various anthracycline drugs at the single-molecule level. Our results revealed for the first time that anthracyclines drugs could induced the unfolding of i-motif into hairpin DNA. Additionally, our experiment captured the current signal of the intermediate state during the unfolding of i-motif into hairpin DNA, a signal difficult to obtain by conventional analytical tools such as circular dichroism (CD) and UV–visible spectroscopy (UV–vis). This research provides a novel tool for investigating the interaction between small molecule and DNA and lays the foundation for future screening of drugs acting on i-motif targets, as well as analyzing their therapeutic mechanism.

Excessive ultraviolet radiation is one of the most common external factors causing skin photoaging, which leads to collagen degradation, wrinkle formation, pigmentation, and is even associated with skin disorders such as seborrheic keratosis and skin tumors. Although various strategies have been developed to treat photoaging skin; however, they suffer from poor stability, risk of allergic reactions, and prolonged recovery periods. In this study, we develop a novel highly stable and bioactive yak type I collagen-based hydrogels dressing to promote the rejuvenation of photoaged skin. The YIC gel demonstrated excellent stability over a six-month period, and maintained the intact triple-helical structure of collagen. It also displayed shear-thinning behavior and notable ductility. In vitro cell experiments confirmed its favorable biocompatibility and bioactivity.Non-invasive Combo evaluations demonstrated the remarkable ability of YIC gel to restore dermal density, elasticity, hydration, and transepidermal water loss to healthy levels, surpassing the efficacy of the commercial product. Histological analysis revealed YIC gel significantly reduced epidermal thickness, promoted the deposition and regeneration of collagen, demonstrating greater efficacy than the commercial product. Antioxidant indicators further highlighted the antioxidant properties of YIC gel, including reduced MDA content, increased SOD activity and GSH content. Immunofluorescence staining suggested that the YIC gel may enhance collagen synthesis through modulation of the TGF-β/Smad signaling pathway. This YIC gel offers valuable insights into its protective effects against skin photoaging and highlights its promising applications in cosmetic formulations.

The cGAS-STING pathway mediates the innate immune response to cytosolic DNA, contributing to surveillance against microbial invasion or cellular damage. Once activated, STING recruits TBK1 at the trans-Golgi network (TGN), which in turn phosphorylates IRF3 to induce type I interferon (IFN-I) expression. In contrast to STING, little is known about how TBK1 is transported to the TGN for activation. Here, we show that multiple TGN tethering factors, a group of proteins involved in vesicle capturing, are indispensable for STING-IFN-I signaling. Deletion of TBC1D23, a recently reported tethering factor, in mice impairs the STING-IFN-I signaling, but with insignificant effect on STING-NF-κB signaling. Mechanistically, TBC1D23 interacts with TBK1 via the WASH complex subunit FAM21 and promotes its endosome-to-TGN translocation. Furthermore, multiple TGN tethering factors were reduced in aged mice and senescent fibroblasts. In summary, our study uncovers that TGN tethering factors are key regulators of the STING-IFN-I signaling and suggests that their reduction in senescence may produce aberrant STING signaling.

Graphical The inclusion of carboxylic acid ligands significantly impacts the structural arrangement of the metal core in Cu(I) alkynyl clusters, thereby affecting their fluorescence properties. Furthermore, the incorporation of ferrocene into these clusters has been noted to induce modifications in the electrochemical performance of the Cu(I) alkynyl clusters. Surface ligands play a pivotal role in the functional design of atomically precise Cu(I) nanoclusters. They act as protective agents, ensuring the stability of the Cu(I) clusters, while also influencing their structure and properties. This study delves into the synthesis, structure, and properties of innovative Cu(I) nanoclusters co-stabilized by carboxylic acid and alkynyl ligands. The findings reveal that these surface ligands wield a significant impact on the cluster's structure and can even modulate the luminescent characteristics of the Cu(I) alkynyl clusters. Moreover, density-functional theory (DFT) calculations shed light on how different carboxylic acid ligands affect the UV-visible absorptivity of paired Cu(I) clusters. In addition, ferrocene carboxylic acids were employed as protective ligands to impart electrochemical properties to the Cu(I) clusters. This research presents an effective methodology for synthesizing atomically precise Cu(I) clusters shielded by carboxylic acid ligands.

Parkinson's disease (PD) is one of the most common neurodegenerative diseases. Lewy bodies that are formed by the aggregated α-synuclein are a major pathological feature of PD. Salvia miltiorrhiza has been used as food and as a traditional medicine for centuries in China, with tanshinone I (TAN I) and tanshinone IIA (TAN IIA) as its major bioactive ingredients. Here, we investigated the effects of TAN I and TAN IIA on α-synuclein aggregation both in vitro and in a transgenic Caenorhabditis elegans PD model (NL5901). We demonstrated that TAN I and TAN IIA inhibited the aggregation of α-synuclein as demonstrated by the prolonged lag time and the reduced thioflavin-T fluorescence intensity; TAN I and TAN IIA also disaggregated preformed mature fibrils in vitro. Moreover, the presence of TAN I or TAN IIA affected the secondary structural transformation of α-synuclein from unstructured coils to β-sheets, and alleviated the membrane disruption caused by aggregated α-synuclein in vitro. Besides, the immuno-dot-blot assay indicated that TAN I and TAN IIA reduce the formation of oligomers and fibrils. We further found that TAN I and TAN IIA extended the life span of NL5901, a strain of transgenic C. elegans that expresses human α-synuclein, possibly by attenuating the aggregation of α-synuclein. Taken together, our results suggested that TAN I and TAN IIA may be explored further as potential candidates for the prevention and treatment of PD.

In this work, we presented the preparation of magnetic carbon nanotubes (MCNTs) functionalized with molecularly imprinted polymers (MIPs) for effective removal of aristolochic acid I (AAI) in traditional Chinese medicine (TCM). MCNTs@AAI-MIPs was obtained via a facile and environmental friendly sol-gel process. Firstly, MCNTs were synthesized by a solvothermal method. Then, the template molecules were self-assembled with the functional monomer phenyltrimethoxysilane (PTMOS) in the presence of ethanol and water. Finally, AAI-MIPs film was coated on the MCNTs to obtain product MCNTs@AAI-MIPs using tetraethyl-orthosilicate (TEOS) as cross-linker. The morphology and structure of prepared MIPs were characterized by Fourier transform infrared spectroscopy (FT-IR), transmission electron microscopy (TEM), X-ray diffraction (XRD), nitrogen gas adsorption and vibrating sample magnetometer (VSM). The adsorption properties were demonstrated by kinetic, isothermal and selective adsorption experiments. The results showed that the imprinted nanocomposites exhibited fast separation rate (10 s), high adsorption capacity (18.54 μg∙mg −1 ), short kinetic equilibrium time (15 min), and good selectivity to template molecule with imprinting factor (IF) of 3.17. A regression equation (y=57294x-4734.1) with good linearity was obtained in the concentration range of 0.1–200 μg∙mg −1 for AAI with a correlation coefficient (R 2 ) of 0.9998. The limit of detection (LOD, S/N=3) was 0.034 μg∙mg -1 . Moreover, high recoveries ranged from 80% to 110% (RSD=3.27%-8.16%) were received in spiked TCM samples. The results suggested that the proposed MCNTs@AAI-MIPs could efficiently and specifically capture AAI from an actual complex TCM samples.

The copper(I)-catalyzed azide–alkyne cycloaddition (CuAAC) click reaction has drawn increasing attention in the field of analytical science. However, the poor stability of Cu(I) usually hinders not only the simplicity of the click reaction but also its applications in precise analyses. Therefore, the development of a nanocatalyst containing stable Cu(I) is of great significance for broadening the application of CuAAC-based assays. Herein, inspired by the active center structure of natural multicopper oxidases (MCOs), we successfully prepared a novel nanocatalyst containing abundant stable Cu(I) as an artificial “clickase” (namely, CCN) by using glutathione to stabilize Cu(I). The stability and enzyme-like catalytic activity in the CuAAC reaction of the prepared CCN clickase were studied, and the catalytic mechanism of the CCN clickase-mediated CuAAC reaction between 3-azide-7-hydroxycoumarin (Azide 1) and propargyl alcohol (Alkyne 2) was also revealed. Compared with the existing solid CuO nanocatalysts used in CuAAC-based assays, CCN clickases exhibited plenty of superior properties (including high stability, excellent catalytic activity, no requirements of dissolution and reducing agents/radical initiator during the detection, well-defined porosities benefiting the substrate diffusion, and good biocompatibility), which can greatly increase the reaction efficiency and shorten the detection time. Encouraged by these remarkable performances, CCN clickases were used as labels to establish a new catalytic click fluorescence immunoassay for foodborne pathogens. Notably, the proposed CCN clickase-based immunoassay exhibited high analytical performances for the quantification of Salmonella enteritidis in the linear range of 102–106 CFU/mL with a limit of detection as low as 11 CFU/mL. The developed method has also been used in the determination of S. enteritidis in food samples, showing its great potential in the detection of foodborne pathogens.

Photoelectrochemical cathodic protection is emerging as a green and environmental method to protect metals against corrosion. Graphitic carbon nitride (g-C 3 N 4 ) shows great application potential in this area because of its high efficiency, high stability and low cost. However, the pristine g-C 3 N 4 shows amphoteric properties, resulting in its failure in providing photoelectrochemical cathodic protection for the coupled metal in NaCl solution. In this work, a K&I co-doping technique was performed to modulate the band structure of g-C 3 N 4 , and an n-type g-C 3 N 4 (n-C 3 N 4 ) was obtained. The changes in the molecular structure were studied by SEM, HRTEM , XRD , EDS, XPS and FT-IR technologies. The n-type semiconductor characteristics were verified by the photoinduced i-V curves, SKP potential distributions and pH drift techniques. The n-C 3 N 4 can generate positive photocurrent in the whole investigated potential range, making it able to provide photoelectrochemical cathodic protection for the coupled 316L stainless steel in NaCl solution. This work is very helpful for promoting the application of g-C 3 N 4 in the fields of the photoelectric conversion and the photoelectrochemical cathodic protection.

The first derivatives of gelatin and type I collagen fluorescence spectra were characterized in order to describe the effect of free radicals on pyridinoline (PYD) cross-links. The different gas saturation conditions were used to investigate the effect of different free radicals. An analysis of first derivative fluorescence spectra suggests that PYD cross-link fluorescence emission is composed of three peaks in gelatin, but only two in type I collagen. The PYD cross-link was photo-degraded more than other gases in the presence of O 2 . This suggests that the singlet oxygen ( 1 O 2 ) plays a key role when using photodynamic therapy to treat skin fibrosis disease with Hypocrellin B (HB).

Early intervention in acute myocardial infarction can minimize myocardial damage and improve patient survival. Herein, a low-cost device-free portable immunobiosensing platform for flexible monitoring of immediate myocardial infarction is reported. CuS-Pt nanofragments (CuS-Pt NFs) with high photothermal conversion efficiency (≈26.41%) are synthesized by liquid-phase polarity-mediated synthesis. The CuS NFs are loaded in situ with platinum (Pt) nanoreactors using a solvothermal reduction strategy, which is employed to enhance the efficiency of gas production. The resulting CuS-Pt nanocatalysts are encapsulated within liposomes for signal cascade amplification. Specifically, cardiac troponin I (cTn I), a target biomarker in serum, is captured on pre-modified microtiter plates and formed into a classical sandwich model. The thermo-chemically kinetically enhanced CuS-Pt reactor is released through a one-step chemical treatment and transferred to a closed gas generator. Under the excitation of a near-infrared laser emitter, the internal pressure in the gas generator device increases with time and drives the carbon quantum dot solution in the connected hose. The moving distance shows a correlation with the target concentration. This work provides a new implementation for the development of low-cost, efficient pressure immunosensors without the requirement of a readout device.

As a key biomarker in clinical diagnostics, cardiac troponin I facilitates the early diagnosis of acute myocardial infarction. This study developed a rapid quantitative method for detecting cardiac troponin I in blood, aiming to establish its quantitative correlation with myocardial infarction area by accurately quantifying the serum cTnI concentration in patients suffering from acute myocardial infarction. We utilized a cTnI-specific aptamer as the recognition element. Gold nanoparticles exhibit salt-induced aggregation with visible color change. The aptamer protects gold nanoparticles from high-salt aggregation. Through systematic optimization, we established a gold nanoparticle-based colorimetric assay leveraging this anti-aggregation mechanism. Results demonstrated that under optimal conditions—200 mM NaCl and 800 nM aptamer concentration—the developed assay exhibited a promising linear response (R 2 = 0.942) across a cardiac troponin I concentration spanning from 0.1 to 0.8 μg/mL. According to the standard calibration method, the detection limit was estimated as 0.1 μg/mL. We observed recovery rates ranging from 105.6 % to 118.5 %. This research presents a highly sensitive, specific, and rapid point-of-care detection methodology for cardiac troponin I, offering enhanced field-testing efficiency and providing a low-cost alternative for serum cardiac troponin I detection. Moreover, it lays the groundwork for the development of a novel methodology enabling dynamic and quantitative assessment of infarct size in patients suffering from myocardial infarction.
