Hyperoside inhibits pancreatic lipase activity in vitro and reduces fat accumulation in vivo

Xinyue Zhang,Dan Li,Kexin Wang,Jiao Xie,Yaojie Liu,Tianxin Wang,Suwen Liu,Qun Huang,Qingbin Guo,Hao Wang

Hyperoside, the main component of many anti-obesity plants, might exhibit a lipase inhibition effect to reduce fat accumulation. The anti-obesity effect of hyperoside was investigated by studying its inhibitory effect and mechanism on pancreatic lipase in vitro and evaluating its ability to reduce lipid accumulation in vivo. Hyperoside is a mixed-type inhibitor of lipase with an IC50 of 0.67 ± 0.02 mmol L−in vitro. Hyperoside changed the secondary conformation of lipase, increased the α-helix content, and changed the microenvironment of lipase through static quenching. The interaction between hyperoside and lipase results from a strong binding spontaneous exothermic reaction, mainly through hydrogen bonding, van der Waals force and electrostatic force. Hyperoside protected hepatic lipid accumulation and adipose tissue hypertrophy and reduced the expression of inflammatory factors in high-fat diet-induced rats. Moreover, hyperoside had a good inhibitory effect on lipase activity in serum and increased fecal fat excretion, thereby reducing lipid absorption in vivo. This study provides theoretical support for the research and development of hyperoside in fat-reducing functional foods.

Role of Berberine Thermosensitive Hydrogel in Periodontitis via PI3K/AKT Pathway In Vitro

Chang Wang,Chang Liu,Chen Liang,Xingyuan Qu,Xinying Zou,Siyu Du,Qian Zhang,Lei Wang

Periodontitis is a long-term inflammatory illness and a leading contributor to tooth loss in humans. Due to the influence of the anatomic parameters of teeth, such as root bifurcation lesions and the depth of the periodontal pocket, basic periodontal treatment on its own often does not completely obliterate flora microorganisms. As a consequence, topical medication has become a significant supplement in the treatment of chronic periodontitis. Berberine (BBR) has various pharmacological effects, such as hypoglycemic, antitumor, antiarrhythmic, anti-inflammatory, etc. The target of our project is to develop a safe and non-toxic carrier that can effectively release berberine, which can significantly reduce periodontal tissue inflammation, and to investigate whether berberine thermosensitive hydrogel can exert anti-inflammatory and osteogenic effects by modulating phosphatifylinositol-3-kinase/Protein Kinase B (PI3K/AKT) signaling pathway. Consequently, firstly berberine temperature-sensitive hydrogel was prepared, and its characterizations showed that the mixed solution gelated within 3 min under 37 °C with a hole diameter of 10–130 µm, and the accumulation of berberine release amounted to 89.99% at 21 days. CCK-8 and live-dead cell staining results indicated that this hydrogel was not biotoxic, and it is also presumed that the optimum concentration of berberine is 5 µM, which was selected for subsequent experiments. Real-time polymerase chain reaction (qRT-PCR) and Western blotting (WB)results demonstrated that inflammatory factors, as well as protein levels, were significantly reduced in the berberine-loaded hydrogel group, and LY294002 (PI3K inhibitor) could enhance this effect (p< 0.05). In the berberine-loaded hydrogel group, osteogenesis-related factor levels and protein profiles were visibly increased, along with an increase in alkaline phosphatase expression, which was inhibited by LY294002 (p< 0.05). Therefore, berberine thermosensitive hydrogel may be an effective treatment for periodontitis, and it may exert anti-inflammatory and osteogenic effects through the PI3K/AKT signaling pathway. Keywords:periodontitis;berberine;thermosensitive hydrogel;anti-inflammatory;osteogenesis

Fluoroquinolone Residues in the Environment Rapidly Induce Heritable Fluoroquinolone Resistance in Escherichia coli

Hebin Liang,Jiayu Zhang,Jiahui Hu,Xiaoyan Li,Bing Li

Extensive antibiotic use increases the environmental presence of their residues and may accelerate the development of antibiotic resistance, although this remains poorly understood at environmentally relevant concentrations. Herein, susceptible Escherichia coli K12 was continuously exposed to five antibiotics at such concentrations for 100 days. The de novo-evolved mutants rapidly obtained fluoroquinolone resistance within 10 days, as indicated by the 4- and 16-fold augmentation of minimum inhibitory concentrations against enrofloxacin and ciprofloxacin, respectively. Moreover, the mutants maintained heritable fluoroquinolone resistance after the withdrawal of antibiotics for 30 days. Genomic analysis identified Asp87Gly or Ser83Leu substitutions in the gyrA gene in the mutants. Transcriptomics data showed that the transcriptional response of the mutants to fluoroquinolones was primarily involved in biofilm formation, cellular motility, porin, oxidative stress defense, and energy metabolism. Homologous recombination and molecular docking revealed that mutations of gyrA primarily mainly conferred fluoroquinolone resistance, while mutations at different positions of gyrA likely endowed different fluoroquinolone resistance levels. Collectively, this study revealed that environmentally relevant concentrations of antibiotics could rapidly induce heritable antibiotic resistance; therefore, the discharge of antibiotics into the environment should be rigorously controlled to prevent the development of antibiotic resistance.

Regulation of Cat8 in energy metabolic balance and glucose tolerance in Saccharomyces cerevisiae

Deng Hong,Du Zhengda,Lu Surui,Wang Zhaoyue,He Xiuping

Abstract Cat8 is a C6 zinc cluster transcription activator in yeast. It is generally recognized that the transcription of CAT8 is inhibited and that Cat8 is inactive in the presence of high concentrations of glucose. However, our recent study found that constitutively overexpressed Cat8 played a regulatory role in Saccharomyces cerevisiae in the presence of 20 g/L glucose . To explore the regulatory network of Cat8 at high glucose concentrations, CAT8 was both overexpressed and deleted in this study. Cell growth and glucose consumption in different media were significantly accelerated by the deletion of CAT8 , while the lag period was greatly shortened. RNA-seq and genetic modification showed that the deletion of CAT8 changed the type of energy metabolism in yeast cells. Many genes related to the mitochondrial respiratory chain were downregulated, resulting in a reduction in aerobic respiration and the tricarboxylic acid cycle. Meanwhile, both the energy supply of anaerobic ethanol fermentation and the Crabtree effect of S. cerevisiae were enhanced by the deletion of CAT8 . CAT8 knockout cells show a higher sugar uptake rate, a higher cell growth rate, and higher tolerance to glucose than the wild-type strain YS58. This study expands the understanding of the regulatory network of Cat8 and provides guidance for modulating yeast cell growth. Key points • The deletion of CAT8 promoted cell growth of S. cerevisiae. • Transcriptome analysis revealed the regulation network of Cat8 under 1% glucose condition. • CAT8 deletion increases the glucose tolerance of cells by enhancing the Crabtree effect.

Divergence in the distribution of di(2-ethylhexyl) phthalate (DEHP) in two soils

Wang Qiutang,Xu Yanwen,Du Wenchao,Yin Ying,Wu Xuan,Sun Feifei,Ji Rong,Guo Hongyan

Understanding the distribution of di(2-ethylhexyl) phthalate (DEHP) is necessary for future risk evaluation of DEHP in agricultural soils. This study used 14 C-labeled DEHP to examine its volatilization, mineralization, extractable residues, and non-extractable residues (NERs) incubated in Chinese typical red and black soil with/without Brassica chinensis L. Results showed that after incubated for 60 days, 46.3% and 95.4% of DEHP were mineralized or transformed into NERs in red and black soil, respectively. The distribution of DEHP in humic substances as NER descended in order: humin > fulvic acids > humic acids. DEHP in black soil was more bioavailable, with 6.8% of initial applied radioactivity left as extractable residues at the end of incubation when compared with red soil (54.5%). Planting restrained the mineralization of DEHP by 18.5% and promoted the extractable residues of DEHP by 1.5% for black soil, but no such restrain was observed in red soil. These findings provide valuable information for understanding the distribution of DEHP in different soils and develop the understanding for the risk assessments of PAEs in typical soils. Influence of interfacial properties/structure on oxygen diffusion in oil-in-water emulsions

Yanlei Li,Fangfang Chen,Zhiming Gao,Wei Xiang,Yuehan Wu,Bing Hu,Xuewen Ni,Katsuyoshi Nishinari,Yapeng Fang

Oxygen diffusion played an important role in the lipid oxidation of food emulsions . In this study, a simple method was developed to quantitatively observe the oxygen diffusion in the oil–water biphasic system, and it was further applied to investigate the relationship between the oxygen diffusion and lipid oxidation in O/W emulsions. Various factors that related to the emulsion oxidation were considered, from their influence on the oxygen diffusion and lipid oxidation in the emulsions. Results showed that there was obvious correlation between the oxygen diffusion and lipid oxidation in O/W emulsions, which reveals the inhibition of oxygen diffusion could apparently slow down the lipid oxidation. Moreover, the changes of oil phase, water phase and interfacial layer of the emulsions, which were related to the oxygen diffusion, could improve the oxidative stability of the emulsions effectively. Our findings are helpful for deep understanding the mechanisms of the lipid oxidation in food emulsions.

Potential of Tamarind Shell Extract against Oxidative Stress In Vivo and In Vitro

Weixi Li,Rongping Huang,Shaocong Han,Xiyou Li,Haibiao Gong,Qiongyi Zhang,Changyu Yan,Yifang Li,Rongrong He

Tamarind shell is rich in flavonoids and exhibits good biological activities. In this study, we aimed to analyze the chemical composition of tamarind shell extract (TSE), and to investigate antioxidant capacity of TSE in vitro and in vivo. The tamarind shells were extracted with 95% ethanol refluxing extraction, and chemical constituents were determined by ultra-performance chromatography–electrospray tandem mass spectrometry (UPLC-MS/MS). The free radical scavenging activity of TSE in vitro was evaluated using the oxygen radical absorbance capacity (ORAC) method. The antioxidative effects of TSE were further assessed in 2,2-azobis (2-amidinopropane) dihydrochloride (AAPH)-stimulated ADTC5 cells andtert-butyl hydroperoxide (t-BHP)-exposed zebrafish. A total of eight flavonoids were detected in TSE, including (+)-catechin, taxifolin, myricetin, eriodictyol, luteolin, morin, apigenin, and naringenin, with the contents of 5.287, 8.419, 4.042, 6.583, 3.421, 4.651, 0.2027, and 0.6234 mg/g, respectively. The ORAC assay revealed TSE and these flavonoids had strong free radical scavenging activity in vitro. In addition, TSE significantly decreased the ROS and MDA levels but restored the SOD activity in AAPH-treated ATDC5 cells andt-BHP-exposed zebrafish. The flavonoids also showed excellent antioxidative activities against oxidative damage in ATDC5 cells and zebrafish. Overall, the study suggests the free radical scavenging capacity and antioxidant potential of TSE and its primary flavonoids in vitro and in vivo and will provide a theoretical basis for the development and utilization of tamarind shell. Keywords:tamarin shell extract;flavonoids;oxidative stress;free radicals scavenging;AAPH;tert-butyl hydroperoxide;zebrafish

Calcium and L-glutamate present the opposite role in managing arsenic in barley

Fanrong Zeng,Muhammad Mudassir Nazir,Temoor Ahmed,Muhammad Noman,Shafaqat Ali,Muhammad Rizwan,Mohammad Shah Alam,Jonas Lwalaba Wa Lwalaba,Guoping Zhang

Arsenic contamination in agricultural soils has posed tremendous threat to sustainable crop production and human health via food chain. Calcium and Glutamate have been well-documented in metal(loid)s detoxification, but it is poorly understood how they regulate arsenic-induced toxicity to plants. In this study, the effect of glutamate and calcium at high concentration on arsenic toxicity and accumulation in barley seedling was accessed in terms of plant growth, photosynthetic efficacy, arsenic uptake, translocation and accumulation, antioxidant defense, nutrient uptake and the expression of As transporters. Our results have demonstrated that calcium could effectively ameliorate arsenic toxicity to barley seedlings, which is mainly attributed to its beneficial effect on increasing nutrient uptake, reducing the aboveground arsenic accumulation and enhancing antioxidative defense capacity. However, it is unexpected that glutamate considerably exacerbated the arsenic toxicity to barley seedlings. More importantly, for the first time, glutamate was observed to tremendously facilitate the root-to-shoot translocation of arsenic by 41.8- to 60.8-fold, leading to 90% of the total amount of As accumulating in barley shoots. The reason of this phenomenon can be well explained by the glutamate-triggered enormous upregulation of genes involved in arsenic uptake ( HvPHT1;1 , HvPHR2 and HvNIP3;2 ), reduction ( HvHAC1;1 ), translocation ( HvABCC7 , HvNIP1;1 and HvNIP3;3 ) and intracellular sequestration ( HvABCC1 ). These findings suggest that calcium and glutamate function as the opposite player in managing arsenic, with calcium being an effective alleviator of arsenic stress to ensure the safe production of crops; while glutamate being a highly efficient phytoextraction agent for phytoremediation of arsenate-contaminated soils.

Compositional variations in algal organic matter during distinct growth phases in karst water

Tian Liye,Zhang Zhiwei,Wang Zhikang,Zhang Ping,Xiong Chao,Kuang Ye,Peng Xingyi,Yu Mengxin,Qian Yu

Inland surface water plays an important role in global carbon cycling, which responds to transformation between dissolved inorganic carbon (DIC) and dissolved organic carbon (DOC). Studies have shown that algae in karst lakes and reservoirs can convert DIC to organic matter (OM) and form stable carbon sinks via photosynthesis. However, the pathways of conversion of inorganic carbon to organic carbon during algal growth remain unclear and need further investigation. In this study, spectroscopic techniques were applied to investigate the variations in algal organic matter (AOM) composition in the growth metabolism of Chlorella vulgaris and Scenedesmus obliquus under simulated karst water condition. The results showed that algal extracellular organic matter (EOM) contained high DIC concentration during the adaptation phase, which formed the carbon source for algal photosynthesis. In addition, DOC in algae increased after entering the stationary phase, while more OM was released into water. As algal growth proceeded, the amino groups in EOM were consumed to produce more aromatic protein-like material, while more lipid material was produced in intracellular organic matter (IOM). The spectral characterization results could intuitively determine AOM dynamics in different growth stages of algae, which can be used for establishing effective approaches for detecting organic carbon variations and responding to regional carbon cycling in karst water.

Jujuboside A ameliorates cognitive deficiency in delirium through promoting hippocampal E4BP4 in mice

Du Jianhao,Zhang Fugui,Chen Min,Xiao Yifei,Zhang Li,Dong Linlin,Dong Dong,Wu Baojian

ObjectiveDelirium (acute brain syndrome) is a common and serious neuropsychiatric disorder characterized by an acute decline in cognitive function. However, there is no effective treatment clinically. Here we investigated the potential effect of jujuboside A (JuA, a natural triterpenoid saponin) on cognitive impairment in delirium.MethodsDelirium models of mice were established by injecting lipopolysaccharide (LPS) plus midazolam and implementing a jet lag protocol. Novel object recognition test and Y maze test were used to evaluate the effects of JuA on delirium-associated cognitive impairment. The mRNA and protein levels of relevant clock factors and inflammatory factors were measured by qPCR and Western blotting. Hippocampal Iba1+ intensity was determined by immunofluorescence staining.Key findingsJuA ameliorated delirium (particularly delirium-associated cognitive impairment) in mice, which was proved by the behavioural tests, including a preference for new objects, an increase of spontaneous alternation and improvement of locomotor activity. Furthermore, JuA inhibited the expression of ERK1/2, p-p65, TNFα and IL-1β in hippocampus, and repressed microglial activation in delirious mice. This was attributed to the increased expression of E4BP4 (a negative regulator of ERK1/2 cascade and microglial activation). Moreover, loss of E4bp4 in mice abrogated the effects of JuA on delirium as well as on ERK1/2 cascade and microglial activation in the hippocampus of delirious mice. Additionally, JuA treatment increased the expression of E4BP4 and decreased the expression of p-p65, TNFα and IL-1β in LPS-stimulated BV2 cells, supporting a protective effect of JuA on delirium.ConclusionsJuA protects against delirium-associated cognitive impairment through promoting hippocampal E4BP4 in mice. Our findings are of great significance to the drug development of JuA against delirium and related disorders.

Positive Regulation of Acetate in Adipocyte Differentiation and Lipid Deposition in Obese Mice

Changbao Sun,Ang Li,Huan Wang,Jiage Ma,Juncai Hou

Acetate is associated with adipocyte differentiation and lipid deposition. To further develop this scientific point, obese mice on a high-fat diet were given an intragastric administration of acetate for 8 weeks and mouse adipose mesenchymal stem cells (mAMSCs) were treated with acetate for 24 h. The results showed that the body weight, food intake, Lee’s index, adipose tissue coefficient, liver index, blood lipid levels, insulin resistance, pro-inflammatory factors levels and fatty lesions in liver and adipose tissue in obese mice treated with acetate increased markedly, while anti-inflammatory factors levels and liver function decreased significantly (p< 0.05). The mRNA expression levels of PPAR-γ, C/EBP-α, SREBP, AFABP, FAS, ACC-1, SCD-1, LPL, LEPR, GPR41 and GPR43 genes in adipose tissue and mAMSCs were significantly increased, while the mRNA expression levels of HSL, CPT-1, CPT-2, AMPK, AdipoR1 and AdipoR2 genes were significantly reduced (p< 0.05). Except for AMPK-α signaling pathway proteins, the phosphorylation levels of p38 MAPK, ERK1/2, JNK and mTOR were significantly increased (p< 0.05) and these changes were dose-dependent. The findings indicated that acetate played a positive role in regulating adipocyte differentiation and lipid deposition by activating MAPKs and mTOR signaling pathways (the expression up-regulation of genes such as PPAR-γ, C/EBP-α and SREBP-1, etc.) and inhibiting the AMPK signaling pathway (the expression down-regulation of genes such as HSL, CPT-1 and AMPK-α, etc.).Keywords:obesity;lipid metabolism;acetate;adipocyte differentiation;lipid deposition

Detection of carbendazim in oranges with metal grating integrated microfluidic sensor in terahertz

Yanqun Tong,Liuxin Ding,Kuiguo Han,Xiaobo Zou,Shiyan Wang,Zhang Wen,Yunxia Ye,Xudong Ren

A novel microfluidic metal grating integrated terahertz sensor has been designed, which is composed of a metal microstructure array-dielectric layer-metal layer, where the dielectric layer is the microfluidic channel carrying the analyte. By adjusting the structural parameters of the metal grating sensor, a highly confined electromagnetic field can be obtained in the microfluidic channel, thereby significantly enhancing the interaction between the analyte and the terahertz wave and improving the terahertz detection sensitivity. The metal grating described in this paper is composed of an array of square holes, which is manufactured using laser micromachining technology, so that the measurement method is simplified and improved. The results show that the addition of different concentrations of carbendazim solution resulted in a redshift of the overall spectrum, with the highest sensitivity reaching 8.773 GHz/mg L−1, which is about eight times more sensitive than the traditional terahertz transmission sensor. The relative error of using this method to determine carbendazim levels in orange juice samples was less than 5.3%. The terahertz time-domain spectroscopy technology combined with the metal grating integrated microfluidic sensor can improve the sensitivity of sample detection and realize the rapid detection and analysis of trace elements.
