The Yellow Light Irradiation Promotes the Formation of a Distinct Floral Aroma in Steamed Green Tea

Muxue Qin,Jingtao Zhou,Junyu Zhu,Bernard Ntezimana,Zehua Sun,Fuwei Guo,Jinjun Zhang,Youyue Zhang,Zhi Yu,Yuqiong Chen,De Zhang,Dejiang Ni

The effect of light on the aroma of green tea during the withering process is still unclear. The impact of yellow light withering on the aroma of steamed green tea was investigated. Results showed that yellow light irradiation enhanced the floral quality of the tea. Aroma dilution analysis was performed on the sample with optimal aroma under yellow light treatment (Y68) and its control (N68) using GC-O/MS, resulting in 49 aroma active compounds with flavor dilution factors ranging from 64 to 8192 were identified. Through odor activity value analysis as well as recombination and omission experiments, 8 and 7 key odorants were identified in Y68 and N68, respectively, with 5 differentiating metabolites. Addition experiments confirmed that the difference in floral intensity between Y68 and N68 was mainly caused by linalool and indole, while the differences in fresh aroma and tender aroma intensity were respectively attributed to (Z)-3-hexenol and dimethyl sulfide.

Taste Modulation of White Tea by Red/Blue-LED-Assisted Withering Revealed via Non-Volatile Metabolomics

Dan Wu,Yongyi Deng,Jiabao Xing,Lianghua Wen,Jiawei Ma,Dubin Dong,Fanrong Cao

Background: Red/blue- light-emitting diode (LED)-assisted withering provides a controllable spectral input to steer tea quality, yet metabolite-level evidence linking spectrum composition to quantitative taste phenotypes in white tea remains insufficient. Methods: Fresh leaves were withered under supplemental red/blue LEDs—S0, S1, S2, S3, S4, and S5—and the resulting white teas were evaluated by quantitative descriptive analysis (QDA), untargeted metabolomics, weighted gene co-expression network analysis (WGCNA), and high-performance liquid chromatography (HPLC) quantification of caffeine, gallic acid, and eight catechin monomers. Results: Red/blue-mixed spectrum enhanced the overall sensory quality relative to the incandescent lamp; S3 maximized sweetness and freshness, whereas S4 minimized bitterness and astringency and achieved the highest overall score. Untargeted metabolomics showed the largest deviation for S0 vs. S4. A total of 18 common metabolites were identified between the S0 and light-supplemented withering treatments, dominated by saccharides and related derivatives. WGCNA linked a saccharide-centered module to higher sweetness/freshness/smoothness and a lipid-oxylipin-centered module to stronger bitterness/astringency. HPLC independently confirmed that S4 contained the lowest catechins and caffeine, supporting its reduced bitter/astringent attributes. Conclusions: Overall, the application of mixed red-blue spectra offered a promising approach to enhancing the palatability of white tea by coordinately intensifying saccharide metabolism while simultaneously suppressing key bitter and astringent components. Our study provided a scientific basis for standardizing white tea processing to enhance sensory quality.

Amelioration of colitis by donor microbiota modulated with tea active ingredients: A fecal microbiota transplantation study

Kang Wei,Lanlan Peng,Qiurong Wei,Yang Wei,Li Wang,Yuzhi Zhu,Yanxiang Wei,Xinlin Wei

Fecal microbiota transplantation (FMT) using donor microbiota modulated by tea represents an emerging approach to ameliorate ulcerative colitis (UC). This study aims to demonstrate the amelioration of dextran sulfate sodium (DSS)-induced colitis by donor microbiota modulated with tea-derived bioactive ingredients—tea polyphenols (TPP), tea polysaccharides (TPS), or theabrownin (TB). Following transplantation of fecal microbiota modulated by TPP, TPS, or TB from donors into colitis mice, significant reductions in Disease Activity Index (DAI) were observed, alongside attenuated intestinal histopathological damage. Additionally, gut barrier integrity was enhanced, as indicated by upregulated expression of occludin and Muc2. Fecal microbiota modulated by TPP and TPS showed superior efficacy in alleviating colitis compared with TB-modulated microbiota. The mechanism involved that marked enrichment of beneficial genera, including Akkermansia (TPP-enriched) and Allobaculum / Lactobacillus (TPS-enriched), which promoted the biosynthesis of short-chain fatty acids (SCFAs) and secondary bile acids, while reducing primary bile acids levels. These metabolic changes enhanced intestinal barrier function and suppressed suppressing pro-inflammatory cytokines through the modulation of TLR4/NF-κB p65 and Nrf2/ARE signaling pathways. This study not only elucidates the mechanism of tea bioactive ingredients-modulated donor microbiota alleviates colitis through regulation of the gut-microbiota-metabolite axis, but also provides a foundation for FMT-based strategies in colitis intervention.

Multidimensional Analysis Reveals the Flavor Quality Formation Mechanism During the Primary Pile Fermentation of Dark Tea

Dunchao Wu,Yufei He,Juanshu Wen,Hongfa Zheng,Xi Zhao,Penghui Yu,Ni Zhong,Li Niu,Shi Li,Yong Lin,Hao Huang,Zhonghua Liu

Pile fermentation is a crucial process for developing the characteristic mellow taste and aged aroma of dark tea, yet the internal quality transformation mechanism of this process is still unclear. This study employed a high-sensitivity analytical platform based on gas chromatography–mass spectrometry (GC-MS) to systematically investigate the dynamic interplay between key chemical components, enzyme activities, and volatile compounds during the pile fermentation of primary dark tea. Our findings revealed a significant decrease in ester-type catechins, crude protein, and protopectin, alongside a notable accumulation of non-ester-type catechins, gallic acid, and soluble components. The multi-enzyme system—comprising PPO/POD, pectinase/cellulase, and protease—cooperatively drove the oxidation of phenols, cell wall degradation, and the release of aromatic precursors. This was complemented by GC-MS analysis, which identified and quantified 103 volatile compounds across nine chemical classes. The total content of volatile compounds increased significantly, with alcohols, esters, and aldehydes/ketones being the dominant groups. Floral and fruity compounds such as linalool and geraniol accumulated continuously, while esters exhibited an initial increase followed by a decrease. Notably, carotenoid degradation products, including β-ionone, were significantly enriched during the later stages. This study revealed a “oxidation–hydrolysis–reconstruction” metabolic mechanism co-driven by microbial activity and a multi-enzyme system, providing a theoretical foundation for the precise regulation of pile fermentation and targeted quality improvement of primary dark tea.

Detection of catechins in tea beverages using a novel electrochemical sensor based on cyclodextrin nanosponges composite

Yuhong Li,Xiaoling Chen,Yueli Lin,Yaqi Yang,Li Zhang,Pengcheng Zhao,Chenxi Wang,Junjie Fei,Yixi Xie

Catechin (Cat) is one of the primary functional components of tea, and its accurate detection is significant for understanding the health benefits of tea. In this study, cyclodextrin nanosponges (CDNS) were prepared and employed to combine with graphene quantum dots (GQDs) and carboxylated multi-walled carbon nanotubes (MWCNTs-COOH) to form a CDNS/GQDs/MWCNTs-COOH composite. Scanning electron microscope and X-ray diffraction were used to characterize the composite which was used to build a new electrochemical Cat sensor. The abundance of cavities, intermolecular gaps, and surface active groups of CD molecules endowed CDNS with remarkable Cat enrichment capacity. Due to the outstanding biocompatibility of GQDs, the good electrocatalytic activity of MWCNTs-COOH, and the enrichment capacity of CDNS, the composite modified glass carbon electrode (GCE) exhibited excellent electrochemical sensing performance. The CDNS/GQDs/MWCNTs-COOH/GCE showed a sensitive peak current response to catechins in the linear range between 2 × 10 −8 and 1.2 × 10 −6 M, with a minimum limit of detection (S/N = 3) of 4.45 nM. The sensor has also been successfully applied for catechin detection in tea beverages.

Medicinal plant acid-treatment for a healthier herb tea and recycling of the spent herb residue

Hong-Wen Gao,Dong-Dong Ma,Gang Xu

Many kinds of medicinal plants are used in the treatment of disease, and a proportion of these plants grow tenaciously in soils contaminated with heavy metals. An example of one such medicinal plant is the herb Thlaspi rotundifolium L., which accumulates selenium (Se), and has anti-oxidation, anti-aging, anti-cancer and immunity-adjusting properties, but also simultaneously hyperaccumulates harmful metals such as Cd and Zn from the soil. An interesting model named ‘digging metals@plant then backfilling’ has been suggested, where the metals are removed from the plant by acid treatment, then the nano-pores imprinted with the heavy metals are backfilled from the drinking water. Experimental studies of this idea indicate that it can remove unwanted substances from the herb itself and also from the drinking water, ensuring the safety of the herb tea. Additionally, the Se normally embedded in the herb was not noticeably lost. Thus, this model provides a sustainable solution for the disposal of heavy metals using metal-accumulating plants.

Comprehensive investigation on flavonoids metabolites of Longjing tea in different cultivars, geographical origins, and storage time

Xiao-Lan Yu,Jia Li,Yanqing Yang,Jiayi Zhu,Haibo Yuan,Yongwen Jiang

In this study, four kinds of Longjing tea, the famous flat green tea and the protected geographical indication product in China, were used to explore the quality difference of the same green tea due to the cultivar, geographic origin, and storage time under the premise of consistent picking conditions and processing technology using the widely targeted metabolomics. Results showed that 483 flavonoid metabolites with 10 subgroups of flavonoids were screened and 118 differential flavonoid metabolites were identified. The number and subgroups of differential flavonoid metabolites produced by different cultivars of Longjing tea were the largest, followed by storage time, and third by the geographic origin. Glycosidification and methylation or methoxylation were the main structural modifications of differential flavonoid metabolites. This study has enriched the understanding of the effects of the cultivar, the geographic origin, and the storage time on the flavonoid metabolic profiles of Longjing tea, and provided worthy information for the traceability of green tea.

Lateral flow analysis of Pb (II) in green tea integrated with ionic imprinted paper-based chip

Yiwen Lou,Wanqin Chen,Yating Gan,Ying Zhou,Haipeng Ye,Weiwei Huan,Yiming Zhang

A high-performance lateral flow and paper-based microfluidic device ( μ PAD) was constructed to develop a rapid read-out analytical platform for heavy metal elements (Pb (II)) in green tea with complex matrices. The entire analytical device was integrated with several functional parts, including a liquid flow channel and an enrichment visualization zone prepared by the molecularly imprinted polymer (MIP) protocol. Under the optimal conditions, the Pb (II) content in tea products can be sensitively and specifically analyzed using an in situ visualization readout. Herein, a rapid visual read-out analysis of the lead level can be achieved by overcoming the other co-existing interfering elements in the traditional green tea sample. The ionic imprinted paper chip can effectively capture the Pb (II) of the digested sample solution, and a visualization sensing process occurred between the metal elements and a chelating visual reagent with high sensitivity and selectivity in situ . Owing to the stable visualization performance, the constructed chip demonstrated good contrast, with a low background and easily recognizable colored spots. The linear range was 2.5–100 mg kg −1 , and the limit of detection for Pb (II) in traditional green tea was 1.375 mg kg −1 . In addition, other co-existing metal elements did not interfere with the analysis because of the appropriate pretreatment and selectivity of the paper-based chip. Furthermore, the analytical results for tea samples were verified by atomic absorption spectrophotometer (AAS) tests, which demonstrated the robustness and reliability of the proposed method. Therefore, the high-performance visualization lateral flow- μ PAD is a promising platform for the rapid analysis of metal element levels in food products.

Network pharmacology-based prediction and experimental validation of the anti-hyperuricemic effects of oolong tea polyphenols

Fenqi Lin,Duoer Lu,Jia Chen,Dongming Lu,Xiangjun Liu,Shiyu Lin,Langming Wu,Leisi Sun,Yueji Luo

ObjectiveThis study aimed to identify candidate therapeutic targets of oolong tea polyphenols (TP) against hyperuricemia (HUA) using network pharmacology and bioinformatics, and to validate the predicted molecular mechanism through in vivo experimentation.MethodsDrug and disease targets were retrieved from public databases, and overlapping targets were identified by Venn diagram analysis. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed on the shared targets, and a protein-protein interaction (PPI) network was constructed to identify hub genes. For in vivo validation, an HUA mouse model was established by 15 days of oral potassium oxonate (PO) administration. Model mice then received TP by gavage at low (0.5 g⋅kg−1⋅d−1), medium (1 g⋅kg−1⋅d−1), or high (2 g⋅kg−1⋅d−1) doses for an additional 15 days. Serum biochemical markers, histopathological changes, and pathway-related protein expression were assessed by enzyme-linked immunosorbent assay (ELISA), hematoxylin and eosin (HE) staining, and western blot analysis, respectively.ResultsNetwork pharmacology analysis identified 59 overlapping targets between TP and HUA; GO and KEGG enrichment analyses revealed that these targets were primarily associated with hormone metabolism and the PI3K-AKT signaling pathway. In the animal experiment, TP dose-dependently reduced serum uric acid (SUA) levels in hyperuricemic mice. At the molecular level, low and medium doses of TP suppressed phosphorylation of phosphatidylinositol 3-kinase (PI3K), protein kinase B (AKT), and mammalian target of rapamycin (mTOR), whereas the high dose paradoxically activated this pathway and concomitantly elevated interleukin-1β levels. These findings indicate that TP modulates uric acid metabolism through a non-monotonic, dose-dependent mechanism.ConclusionBy combining network pharmacology with animal experiments, this study identified the PI3K/AKT/mTOR signaling pathway as a likely mediator of the anti-hyperuricemic action of oolong tea polyphenols (TP). A medium dose of TP achieved the most balanced outcome, attenuating inflammation and preserving hepatic and renal architecture; the high dose, by contrast, paradoxically elevated interleukin-1β (IL-1β) and overactivated PI3K/AKT/mTOR signaling, underscoring the importance of dose calibration. These data suggest that a medium dose of TP may represent a feasible dietary strategy against hyperuricemia. Further work—including monomer identification, direct target validation, and clinical evaluation—is warranted to confirm and extend these preclinical findings.

Isoindole-functionalized carbon dots as a highly sensitive fluorescent probe for manganese detection in tea samples

Aqian Chu,Guoqiang Xiang,Yaming Sun,Lijun He,Wenjie Zhao

Isoindole-CDs probe synthesized via rapidly gram-scale self-exothermic method. • Isoindole groups enhance oxidation by Mn(VII), enabling ultra-sensitive detection. • Achieved LOD of 2.45 ng mL −1 for Mn(VII) in various tea samples. Manganese (Mn) is an essential trace element for human health, but excessive intake can lead to neurotoxicity. Tea is a major dietary source of Mn, necessitating sensitive and reliable methods for its quantification. In this work, a novel fluorescent probe based on isoindole-functionalized carbon dots (Iso-CDs) was developed for the highly sensitive detection of total Mn in tea samples. The Iso-CDs were synthesized via a two-step strategy: first, amino-rich nitrogen-doped carbon dots (NH₂-CDs) were rapidly prepared on a gram scale using a self-exothermic reaction with locust powder as the precursor; then, the surface amino groups were reacted with o-phthalaldehyde (OPA) and 4-mercaptophenylboronic acid (MPBA) to construct electron-rich isoindole moieties on the CD surface. The resulting Iso-CDs exhibited strong blue fluorescence at 427 nm. The isoindole group is more susceptible to oxidation by Mn(VII) compared to the original amino groups, enabling an efficient fluorescence quenching response. Under optimized conditions, the fluorescence quenching ratio showed a good linear relationship with Mn(VII) concentration in the range of 10–800 ng mL −1 , with a detection limit as low as 2.45 ng mL −1 . The proposed method was successfully applied to the determination of total Mn in various tea samples, yielding satisfactory recoveries. This work provides a rational surface engineering strategy for designing highly sensitive CDs-based fluorescent probes and demonstrates its practical potential for food safety monitoring. Black tea ethanol soluble fraction restores intestinal homeostasis by modulating PI3K/AKT signaling and gut microbiota

Shi Yuxuan,Sun Jiayi,Chen Lin,Xu Shuang,Guo Shasha,Xu Anan,Ye Qun,Wang Yuefei,Xu Ping

While black tea’s oxidized polyphenols hold potential for gut health, the specific protective mechanisms of its ethanol-soluble fraction (BES), particularly regarding host-microbiota interactions, remain poorly defined. This study elucidated the dual mechanisms through which BES ameliorates dextran sulfate sodium (DSS)-induced colitis in mice. By integrating network pharmacology, molecular docking/dynamics simulations, RT-qPCR, and Western blot analysis, we identified the PI3K/AKT signaling axis as a potential host target. Our findings suggest that key flavonoid constituents stably bind to critical nodes, potentially downregulating the expression and activation of pathway components. Concurrently, 16S rRNA sequencing and metabolomics revealed that BES reshaped gut microbiota by suppressing Akkermansia and enriching SCFA producers such as Lactobacillus , restoring fecal SCFA levels. Fecal microbiota transplantation confirmed that BES-modulated microbiota alone transferred protection. Thus, BES acts concertedly through direct PI3K/AKT inhibition and indirect microbiota-SCFA modulation, providing a mechanistic basis for targeted use of black tea fractions in intestinal inflammation.

Selective separation of hematite from quartz in reverse flotation using tea polyphenols: Experimental and molecular insights

Mingzhen Wu,Jiushuai Deng,Jiaozhong Cai,Hao Zhang

TP enables the selective flotation separation of hematite from quartz. • TP is selectively chemisorbed onto the hematite surface. • TP is a natural, biodegradable, and water-soluble depressant. This paper investigated the selective depression of hematite by tea polyphenols (TP) in a cationic collector dodecylamine (DDA) system. Micro-flotation results indicate that, under natural pH conditions, when the DDA concentration was 2 × 10 -4 mol/L and the TP concentration was 2 × 10 -3 mol/L, the artificially mixed ore achieved satisfactory flotation indexes: an iron grade of 65.68% and a recovery of 87.75%. Contact angle test results indicate that TP could significantly reduce the hydrophobicity of hematite surfaces while exerting a minor effect on quartz. FTIR and XPS analyses indicate that TP was chemisorbed on the surface of hematite. Frontier molecular orbital analysis shows that the phenolic hydroxyl group served as the primary reactive site of the TP molecule. Molecular dynamics simulation results reveal that TP exhibited stronger interaction with hematite than with quartz, with interaction energies of approximately −56.44 kcal/mol and −27.80 kcal/mol, respectively. Therefore, TP, as a selective and biodegradable depressant for hematite, is expected to be applicable to the flotation separation of hematite and quartz.
