Chitosan-based hydrogel dressings with antibacterial and antioxidant for wound healing

Yongyan Yang,Ying Ma,Haodong Wang,Chongruihan Li,Chaoqun Li,Ruiting Zhang,Shuangling Zhong,Wenqi He,Xuejun Cui

Bacterial infection and free radical oxidative stress at the wound site could easily cause cascade inflammation and hinder the healing process of the wound. In this study, chitosan-cysteine-gallic acid (CCG) hydrogel with antibacterial and antioxidant properties was synthesized by chitosan (CS), cysteine (Cys), and gallic acid (GA) for a preliminary evaluation of its therapeutic efficacy in a mouse model of full-layer skin defect. In vitro analysis showed that the CCG hydrogel had good antibacterial activity and blood compatibility. In vivo , the CCG hydrogel wound dressings accelerated wound healing, stimulate angiogenesis, increase collagen deposition and anti-inflammatory factor expression. The CCG hydrogel wound dressing is designed to promote the regeneration of damaged skin tissue and is expected to become a potential candidate for clinical treatment.

Biodegradable aminated alginate-chitosan porous polymer for efficient indoor formaldehyde capture

Furong Li,Tao Zhu,Bo Yuan,Jun Yang,Qiuge Ju,Weifang Li,Zefu Ye,Xudong Xu,Zhujun Zhu

Formaldehyde, even at low concentrations, poses a significant indoor pollutant risk, underscoring the need for efficient and sustainable sorbents. This study introduces a novel, multifunctional, and fully biodegradable adsorbent derived from waste shrimp shells. The shells were converted into soluble chitosan, which was then physically crosslinked with branched amine-modified alginate, resulting in a porous biuret-modified alginate/chitosan (BCC) polymer. BCC, with a 6 wt% biuret content, demonstrated exceptional adsorption performance for low-concentration formaldehyde, achieving a capacity of 1.97 mg·g −1 —159% higher than alginate and 149 % greater than activated carbon. The adsorption kinetics followed the pseudo-second-order model, and the Sips isotherm model suggested a chemical adsorption-dominated mechanism, with physical adsorption acting as a supplementary process. Additionally, BCC exhibited excellent reusability, maintaining high adsorption efficiency after four regeneration cycles, and incorporated a colorimetric signaling function, with a detection threshold of 0.07 mg·m −3 and a sensitivity of 0.02 mg·m −3 . These findings provide valuable insights and a theoretical basis for developing effective, safe, and environmentally friendly formaldehyde adsorbents.

Boosting CO2 transport in mixed matrix membranes by chitosan-MOF networks

Jiangnan Wang,Long Li,Jinli Zhang,Xueqin Li

The chitosan (CS)-grafted UiO-66-NH 2 (U6N) (CSGUN) was synthesized and incorporated into Pebax matrix to fabricate mixed matrix membranes (MMMs) for boosting CO 2 transport. The excellent CO 2 separation performance was exhibited for MMMs due to the following reasons: U6N with the suitable porosity and abundant CO 2 transport carriers can provide a unique “expressway” for CO 2 to boost CO 2 transport through MMMs. The grafting CS in CSGUN can connect Pebax (regular roads) and U6N (expressway), acting as an “on-ramp” between them to enrich CO 2 and guide CO 2 through U6N in an orderly manner. Thus, the constructed “on-ramp” and “expressway” in MMMs can improve CO 2 separation performance. Furthermore, the CS-MOF networks were formed in MMMs through using CS molecular chains to connect U6N for enhancing CO 2 separation performance of MMMs. Therefore, Pebax/CSGUN MMMs exhibited the excellent CO 2 separation performance with the CO 2 permeability of 558 ± 7 Barrer and the CO 2 /CH 4 selectivity of 37.5 ± 0.7. The study indicated that MMMs with CS-MOF bridged networks had great potential for boosting CO 2 transport by combining “expressways” and “on-ramp”.

Algae-inspired chitosan-pullulan-based multifunctional hydrogel for enhanced wound healing

Taishan Liu,Huan Lei,Linlin Qu,Chenhui Zhu,Xiaoxuan Ma,Daidi Fan

Chronic wounds caused by hyperglycaemia, hypoxia and bacterial infections are common complications in diabetic patients and chronic wound repair is extremely challenging in clinical practice. A series of hydrogels QPMP with good antioxidant and antimicrobial functions were prepared based on quaternized chitosan (QCS), oxidized pullulan polysaccharide (OP), dopamine-coated polypyrrole (PPY@PDA), and Chlorella vulgaris . The Schiff base cross-linking between the quaternized chitosan (QCS) and oxidized pullulan polysaccharide (OP) constitutes the basic skeleton of the hydrogel, and imparts a certain antimicrobial ability to the hydrogel. Chlorella vulgaris continuously produced oxygen under light conditions to relieve wound hypoxia and promote wound healing. The incorporation of PPY@PDA gave the hydrogel near-infrared (NIR) irradiation-assisted bactericidal activity and antioxidant activity, and as a conductive hydrogel, the hydrogel can be used to sense wound exudate and temperature changes, which can help to achieve the integration of diagnosis and treatment of wound healing. Most importantly, in a chronic wound model, the QPMP hydrogel was more effective in controlling the level of wound inflammation and promoting collagen deposition, angiogenesis, and early wound closure compared to the HeraDerm dressing. Therefore, this conductive oxygen-producing hydrogel is extremely beneficial for chronic wound healing in diabetes.

Chitosan matrix with three dimensionally ordered macroporous structure for nimodipine release

Yuling Xie,Yanli Liu,Yan Wang,Siling Wang,Tongying Jiang

Three dimensionally ordered macroporous (3DOM) chitosan (3D-CS) matrix with interconnected pores in the nanometer range was developed as a drug carrier for the first time. 3D-CS was prepared using a template-assisted assembly and characterized by SEM, TGA, N 2 adsorption and FT-IR. As a model drug, nimodipine (NMDP) was incorporated into the pores of 3D-CS matrix. The solid state properties of NMDP-loaded samples were characterized by SEM, XRD, DSC and FT-IR. Dissolution studies showed that release behavior of the drug was markedly affected by the particle size of the matrix. With a relatively small matrix particle size, formulations of NMDP-3D-CS-0.5 and NMDP-3D-CS-1 exhibited rapid release patterns. However, on increasing the amount of carrier, release rate of the drug decreased. The pH-dependent slow-release characteristic of 3D-CS matrix delivery system was demonstrated by investigating the release behavior of NMDP at different pH values.

A chondroitin sulfate/chitosan composite coating for anticoagulation on cardiovascular implants

Bin Chu,Yibin Huang,Yifan Chen,Song Wang,Xiaoli Li

Developing durable anticoagulant coatings for cardiovascular implants remains a major research challenge. Heparin, the current gold-standard anticoagulant, faces challenges due to rapid elution from coated surfaces. This article reports a new heparin-alternative anticoagulant coating system engineered through rational design of a dual-functional chondroitin sulfate derivative (methacrylated/aldehyde-modified chondroitin sulfate, CSMAO) and sulfonated chitosan (SCS). The hierarchical architecture was constructed via sequential layer-by-layer (LbL) assembly on polydopamine (PDA)-functionalized 316 L stainless steel substrates. The material characterization methods included Fourier transform infrared (FTIR) spectroscopy, nuclear magnetic resonance (NMR), and X-ray photoelectron spectroscopy (XPS). The grafting rate of glycidyl methacrylate was 20.05 %, with an aldehyde substitution degree of 48.61 %, and the sulfonation degree of SCS reached 46.96 %. The composite coating exhibited excellent biocompatibility and anticoagulant properties. Coagulation profiling indicated comparable extrinsic pathway regulation to that of heparin controls along with superior intrinsic pathway inhibition (P < 0.01). This rationally designed coating system shows translational promise for improving the hemocompatibility of cardiovascular implants.

An Organic–Inorganic Hybrid Hydrogel Based on Chitosan for Effective Hemostasis

Xiaoyan Song,Siyuan Yu,Junxia Deng,Jia Liu,Jinfeng Xing

The ideal hemostatic dressing should not only achieve the effect of hemostasis but also maintain a suitable moist environment and sufficient physical and mechanical strength and avoid secondary injury to the wound. Here, a stretchable, adhesive, and antibacterial hydrogel (SAQH) for wound hemostasis is facilely prepared by photopolymerization under green LED irradiation, in which acrylamide (AAM) and methacryloylethylsulfobetaine (SBMA) are used as the skeleton and quaternized carboxymethyl chitosan (QCCS) is used as the procoagulant component. The introduction of the macromolecular cross-linking agent methacryloyl hyaluronic acid (HA-GMA) enhances biocompatibility and avoids the addition of small molecular toxic cross-linking agents. The introduction of inorganic nanoparticle hydroxyapatite (HAP) into the hydrogel system not only improves the mechanical properties of the polymer network but also releases calcium ions from HAP to achieve a good coagulation-promoting effect. SAQH hydrogels have excellent mechanical properties (the maximum stress can reach 237 kPa), good adaptability, sufficient tissue adhesion, and effective antibacterial properties. In the mouse liver injury model, hydrogels can adhere to the bleeding site to reduce blood loss, and they will not cause secondary bleeding when removed. This research develops a potential hemostatic hydrogel with broad applicable prospects.

Antifungal properties and preservation applications of resin acid modified chitosan derivatives

Zhuo Xu,Bin Wu,Chunhua Wu,Qian Chen,Kaimeng Xu,Zhengjun Shi,Xiaoping Rao,Yanfei Niu

In order to obtain natural bio-antimicrobial agents with better antifungal activity, the present study compared the antifungal properties of different structural natural resin acid derivatives (dehydroabietic acid (DHA), acrylpimaric acid (APA), and maleopimaric anhydride (MPA)) grafted with chitosan C 2 -OH and investigated the relationship between structure and activity. The structures of the derivatives were determined by FTIR and 1 H NMR. The antifungal properties were determined by mycelial growth rate method. The results demonstrated that CS-DHA (CSDA) exhibited the capacity to inhibit the majority of the tested fungi with notable efficacy, with an optimal antifungal effect observed at 97.53% against Alternaria solani . CS-APA (CSAA) demonstrated a comparatively weaker antifungal effect in comparison to CSDA, yet it exhibited a stronger effect when compared to CS-MPA (CSMA). SEM, TEM and relative permeability of cell membranes proved that the derivatives inhibited fungal growth by destroying the cell wall structure. The in vivo experiments proved that the derivatives all have the effect of protecting chili peppers, which has some application prospects. The best curative effect of CSAA on chili was observed at 1.5 mg/mL concentration (21.04%). At 3 mg/mL concentration, CSDA showed the best protective effect (29.48%) against chili peppers, which was closest to the effect of Imazalil at the same concentration. Meanwhile, the three derivatives had good preservation effect on cherry tomatoes, and the longest one could extend the shelf life by 6 days.

Gentamicin modified chitosan film with improved antibacterial property and cell biocompatibility

Yang Liu,Peihong Ji,Huilin Lv,Yong Qin,Linhong Deng

Gentamicin modified chitosan film (CS-GT) was produced using a three-step procedure comprising: (i) the chitosan solution was air-dried to form a chitosan (CS) film, (ii) using citric acid to generate the amide and carboxyl groups on the surface of CS, (iii) the CS with surface carboxyl groups was modified by grafting of gentamicin. After modification, this CS-GT film has excellent hydrophilicity and biocompatibility. It is very evident that the gentamicin grafting treatment significantly improves the antibacterial properties of the CS film. Our preliminary results suggest that this novel gentamicin modified chitosan film, which can be prepared in large quantities and at low cost, should have potential application in biomedical applications.

Synthesis and characterization of magnetic chitosan microspheres as coking wastewater adsorbents

Yunlong Zhao,Hao Peng,Lei Zhang,Wanwan Fu,Man Zhang,Zhishan Chen,Longfei Li,Yutao Chen

As a new type of polymer functional material, magnetic chitosan has been widely used in wastewater treatment. In this study, magnetic chitosan microspheres (MCMs) were prepared by reversed-phase microemulsion cross-linking technology, and the adsorption capacity of MCMs to organic pollutants with high chemical oxygen demand (COD) in coking wastewater was studied. Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM) and energy dispersive spectrometer (EDS), brunauer - emmett - teller (BET), vibrating sample magnetometer (VSM) were characterized. The effects of Fe 3 O 4 magnetic nanoparticles (MNPs), pH and the amount of adsorbent on the adsorption properties of MCMs were investigated. The following conclusions are obtained: the MCMs prepared in this study are successfully encapsulated, and the MCMs have good magnetic properties; Meanwhile, the removal performance of COD is higher. When the addition amount of Fe 3 O 4 is 10 % of the mass fraction of chitosan, the maximum adsorption amount of COD by MCMs can reach 261.82 mg/g under the conditions of pH 7.5, reaction temperature 298 K and adsorption time 42 h. The adsorption kinetics can be described by a quasi-second-order kinetic model. In addition, after 4 times of adsorption-desorption processes, the adsorbent still has more than 40 % COD adsorption, and has good adsorption performance. In general, because of the high adsorption efficiency, strong regeneration and excellent magnetic separation performance, MCMs have practical application potential in sewage treatment, biomedicine and other fields.

Sulfonamide modified chitosan oligosaccharide with high nematicidal activity against Meloidogyne incognita

Zhaoqian Fan,Kun Gao,Linsong Wang,Yukun Qin,Song Liu,Ronge Xing,Huahua Yu,Kecheng Li,Pengcheng Li

Chitosan oligosaccharide (COS) modification is a feasible way to develop novel green nematicides. This study involved the synthesis of various COS sulfonamide derivatives via hydroxylated protection and deprotection, which were then characterized using NMR, FTIR, MS, elemental analysis, XRD, and TG/DTG. In vitro experiments found that COS-alkyl sulfonamide derivatives ( S6 and S11 – S13 ) exhibited high mortality (>98 % at 1 mg/mL) against Meloidogyne incognita second-instar larvaes (J2s) among the derivatives. S6 can cause vacuole-like structures in the middle and tail regions of the nematode body and effectively inhibit egg hatching. In vivo tests have found that S6 has well control effects and low plant toxicity. Additionally, the structure-activity studies revealed that S6 with a high degree of substitution, a low molecular weight, and a sulfonyl bond on the amino group of the COS backbone exhibited increased nematicidal activity. The sulfonamide group is a potential active group for developing COS-based nematicides.

Synergistic phototherapy using chitosan-enhanced antimonene nanosheets for effective cancer treatment

Zhang Ziying,Liu Feng,Li Jiale,Wang Bing

Photothermal therapy (PTT), as a non-invasive and selective treatment strategy, has garnered extensive research interest. Photothermal agents (PTAs) are critical components of PTT, whose light-absorbing and thermosensitive properties enable effective conversion of light energy into heat, creating localized high-temperature regions. However, PTAs often face challenges with degradation in vivo, and standalone PTT is insufficient for complete tumor cell ablation. In this study, we successfully developed a biodegradable nanoplatform for synergistic photothermal-photodynamic therapy. This platform is based on degradable antimonene nanosheets, further functionalized with a chitosan coating, and loaded with the photosensitizer Ce6. In vitro experiments demonstrated that this nanoplatform exhibits excellent biocompatibility and biodegradability. Upon laser excitation, the platform induces localized thermal effects for cell ablation and promotes reactive oxygen species generation, leading to superior anti-tumor efficacy compared to monotherapies. These findings suggest that this multifunctional nanoplatform can significantly enhance the therapeutic efficiency of synergistic phototherapy, presenting a promising candidate for cancer treatment.
