Carbonaceous materials (CMs) have been applied extensively for enhancing the catalytic performance of environmental catalysts, however, the self-catalytic mechanism of CMs for groundwater remediation is rarely investigated. Herein, we unveiled the catalytic ability of various CMs via Fe(III) reduction through polyvinyl alcohol-coated calcium peroxide nanoparticles (PVA@nCP) for trichloroethylene (TCE) removal. Among selected CMs (graphite (G), biochar (BC) and activated carbon (AC)), BC and AC showed enhancement of TCE removal of 89% and 98% via both adsorption and catalytic degradation. BET and SEM analyses showed a higher adsorption capacity of AC (27.8%) than others. The generation of solution-Fe(II) and surface-Fe(II) revealed the reduction of Fe(III) on CMs-surface. The role of O-containing groups was investigated by the FTIR technique and XPS quantified the 52% and 57% surface-Fe(II) in BC and AC systems, respectively. EPR and quenching tests confirmed that both solution and surface-bound species (HO•, O 2 − • and 1 O 2 ) contributed to TCE degradation. Acidic pH condition encouraged TCE removal and the presence of HCO 3 − negatively affected TCE removal than other inorganic ions. Both schemes (PVA@nCP/Fe(III)/BC and PVA@nCP/Fe(III)/AC) exhibited promising results in the actual groundwater, surfactant-amended solution, and removal of other chlorinated-pollutants, opening a new direction towards green environmental remediation for prolonged benefits.

The purpose of present study was to develop a long-acting drug-in-adhesive patch of rivastigmine (RVS) to achieve controlled release under high drug loading. Formulation factors including ion-pair, pressure sensitive adhesive (PSA), drug-loading and permeation enhancers were investigated through in vitro skin permeation experiments. Optimized patch was evaluated by pharmacokinetic study. The mechanism of controlled release was studied by FTIR, Raman, DSC , rheology study and molecular modeling . The optimized patch composed of RVS-SA (equal to 30% RVS), 15% POCC as permeation enhancer and AAOH as PSA matrix. The RVS in optimized patch was basically permeated at a uniform rate, and the ratio of the skin permeation amount (2803.38 ± 153.85 μg/cm 2 ) in 72 hours to that of the control group (1000.89 ± 62.45 μg/cm 2 ) was 2.8. The plasma concentration of RVS was stable for 72 hours in vivo ( AUC optimized = 5721.30 ± 1994.87 h ng/mL, MRT 0-t = 29.55 ± 2.49 h), and C max was significantly controlled. The results of the study on the controlled release mechanism showed that the addition of counter ion formed hydrogen bonds with RVS and PSA respectively, which reduced the fluidity and molecular mobility of PSA, and enhanced the interaction between RVS and PSA, thus achieving the purpose of long-acting effect. In conclusion, long-acting drug-in-adhesive patch of RVS was developed, and provided a new idea for the long term drug delivery of Alzheimer's disease .

A facile methodology was explored by using glutaraldehyde as a cross-linking reagent for in situ modification of polyvinyl alcohol (PVA) electrospun nanofibers doped with monodisperse silver nanoparticles (AgNPs) via the one-pot reactions. The hydroxyl groups along the PVA molecule chains can serve as both the reactive sites and stabilizers for AgNPs. Meanwhile, the cross-linking degree can be easily tuned by controlling the charged amounts of glutaraldehyde to obtain either partial or cured cross-linked PVA nanofibrous mats doped with AgNPs. It was revealed that such different cross-linking degrees could effectively control the release contents and rates of the embedded Ag to the surrounding aqueous solution. Furthermore, such release behavior was also found to be pH-responsive and acid-labile due to the formation of acetal groups during the cross-linking reactions. Besides both the partial and cured cross-linked PVA doped with Ag nanoparticles can still bear good antibacterial efficacy against S. aureus while have low cytotoxicity against mouse embryo fibroblasts (NIH3T3), human embryonic kidney cells (293T) and human histiocytic lymphoma cells (U937).

Gold nanodevices have attracted extensive interest in the detection of specific targets within cells. However, constructing gold sensing devices that can be activated by the simulation of remote applications remains a huge challenge. Here, we report a Au nanoparticle (AuNP)-capped cage fluorescent biosensor based on controlled-release and Exonuclease III (Exo III) assisted cyclic enzymatic amplification that can be activated by adenosine triphosphate (ATP). In the system, AuNPs were used as the building blocks to cap the pores of Au nanocages (AuNCs) loaded with Rhodamine B (RhB) molecules through the hybridization of DNA. The RhB fluorescent molecules were finally released with the help of Exo III in the presence of ATP for detection purposes. Ultimately, the biosensor leads to a wide linear ATP detection range from 1.0 × 10−9 to 1.0 × 10−7 M with a limit of detection (LOD) down to 0.88 nM. In addition, it also has good selectivity for ATP to distinguish between ATP and ATP analogues such as cytidine triphosphate (CTP), guanosine triphosphate (GTP), and uridine triphosphate (UTP). Therefore, as a convenient and sensitive biosensor, it is expected to be widely used in the biomedical field.

In this paper, an adenosine-5′-triphosphate ( ATP ) controlled-release strategy to construct a fluorescence sensing platform has been designed. In the sensing platform, because of ATP aptamer and singlestranded DNA ( ssDNA )-linked mesoporous silica nanoparticles ( Si-MPs ) were hybridized, the pores of Si-MPs were blocked with Au nanoparticles ( AuNPs ) modified with ATP aptamer. Carboxy fluorescein was plugged in channels of Si-MPs. In the presence of target molecule ATP, the ATP aptamer combined with ATP and the AuNPs got away from the pore of the surface of Si-MPs modified by ssDNA. 5-Carboxyfluorescein molecule was released to allow the fluorescent detection. By monitoring the fluorescence at 518 nm, ATP could be quantitatively detected with a detection limit of 6 × 10 –8 M. The linear response range was 6 × 10 –8 to 1 × 10 –6 M. This assay was also able to discriminate ATP from its analogs. The controlled-release aptamer-based biosensor could have an effective application in human breast cancer MCF-7 cells.

In this contribution, we reported a novel strategy to synthesize porous molecularly imprinted polymers (MIPs) of anticancer drug of doxorubicin hydrochloride (DOX) using reduction-cleavable hyperbranched polymers containing disulfide bonds (ds-HP-alkyne). The porous MIPs (MIP-DOX-HP) possess enhanced specific surface area and porosity by the incorporation of cleavable hyperbranched polymers from the copper(I)–catalyzed azide–alkyne click chemistry. In comparison to MIPs synthesized without using ds-HP-alkyne, MIP-DOX-HP exhibits more regular and open porous structures, which increase the loading and controlled release abilities of DOX anticancer drug. Under optimized pH condition, the total cumulative release amount of DOX at equilibrium is as high as 2134 and 1249 µg for MIP-DOX-HP and MIP-DOX, respectively. The effective and robust strategy for synthesizing MIPs using cleavable hyperbranched polymers is helpful for extending applications of MIPs in drug delivery and target-activated release systems.

A successful drug delivery device for cancer chemotherapy should ideally be able to load drugs highly, bring the drug preferentially into tumor cells and reduce its distribution in normal tissue to enhance therapeutic efficacy. To this purpose, a novel protocol for DOX-loaded PEGylated nanodiamond (ND-PEG–DOX/NaAc, NPDA) was fabricated using sodium acetate medium. The NPDA nanoparticles exhibited a maximum loading efficiency (99 wt%) with ultra-low drug leakage (7 wt%). Examination by confocal microscope and flow cytometer showed that the NPDA uptake by cells was time-dependent, with a slow and sustained drug release from the lysosomes at a low pH. Also, when CHO (a normal cell) and MCF-7 (a cancer cell) were treated with NPDA, the results demonstrated that NPDA preferentially accumulated much more in tumor cells than in normal cells, which implied that NPDA has the ability to selectively kill tumor cells. In addition, NPDA can inhibit the migration and proliferation of tumor cells and change the cell cycle compared to the free drug. Outcomes from this work suggest that NPDA would be a promising drug delivery platform and the preparation of such a drug delivery system will also have implications in improving the biomedical applications of smart nanodiamond carriers.

In this work, PEG–PLLA electrospun fibers were developed as a new controlled release system for macrolide antibiotic drug brefeldin A (BFA). SEM and XRD analyses of the BFA-loaded PEG–PLLA fibers revealed that the average diameter of fibers was below 950 nm with smooth surfaces, and the drug was well incorporated into the fibers in amorphous form. The release profiles of BFA in PBS were measured by HPLC, demonstrating that the controlled release of BFA could be gained for long time. The in vitro antitumor activity against human liver carcinoma HepG2 cells of the fibers containing 3%, 6%, 9%, 12% and 15% BFA were examined by MTT method, and the results showed that cell growth inhibition rates at 72 h were 64%, 77%, 80%, 81% and 85%, respectively. These results strongly suggested that the BFA/PEG–PLLA fibers had an effect of controlled release of BFA and were suitable for postoperative chemotherapy of cancers.

Betaine ester-shell functionalized hyperbranched polyethylenimines (BEHPEI) were synthesized by Menschutkin reaction between per-N-methylated polyethylenimine (MeHPEI) and alkyl bromoacetate. BEHPEI could play dual antimicrobial roles that were, the betaine ester-shell acted as contact-based antibacterial polycations and the drug-loaded BEHPEI could controllably release the drugs due to the cleavage of the betaine ester groups under weak alkaline condition. The BEHPEI exhibited high transport capacities that per gram of BEHPEI could encapsulate 0.24–2.67 g of dyes or drugs. The model drug release experiment employing methyl orange (MO) showed that the drug release of MO-loaded BEHPEI complex occurred slowly in weak alkaline solutions and the release rate was controlled by varying pH, while the complex kept very stable under weak acid condition (pH = 3.0–7.0). BEHPEI generated from long chain alkyl bromoacetate was compatible with organic resins implying the possible usage in antimicrobial fibers and coatings. Another BEHPEI obtained from short-chain alkyl bromoacetate was water soluble and maybe used in lotion formula. The hydrolysis of BEHPEI afforded zwitterionic shell.

Magnetic microspheres, 2-hydroxypropyltrimethyl ammonium chloride chitosan/Fe 3 O 4 /halloysite nanotubes/ofloxacin (HACC/Fe 3 O 4 /HNTs/OFL), for the controlled release of OFL were prepared by in situ crosslinking with glutaraldehyde in the spray-drying process. The magnetic microspheres were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, thermogravimetric analysis and a magnetometer. Various parameters influencing the encapsulation efficiency, drug loading and in vitro controlled release properties of the magnetic microspheres for OFL were also studied. Many stripes were formed and some tubular HNTs could be seen at higher magnification on the surface of the HACC/Fe 3 O 4 /HNTs/OFL magnetic microspheres. The magnetic microspheres show superparamagnetic property and fast magnetic response. The encapsulation efficiency and the cumulative release of OFL are closely related to HACC concentration, HNTs contents and crosslinking density. The release of OFL follows the first-order kinetics.

A novel graphene oxide (GO)-based nanocarrier has been designed for the targeting and pH-responsive controlled release of anti-cancer drugs via the classic amidation of the carboxyl groups of carboxylated graphene oxide (CG) with the amine end-groups of functional poly(ethylene glycol) (PEG) terminated with an amino group and a folic acid group (FA–PEG–NH2). The carboxylated graphene oxide conjugated folate-terminated poly(ethylene glycol) (CG–PEG–FA) nanocarrier containing 44.4 wt% of functional PEG brushes exhibits stable dispersibility in PBS media, outstanding cytocompatibility, high drug-loading capacity (0.3993 mg mg−1 for DOX) via π–π stacking interactions, perfect folate receptor-targeting and pH-activated controlled release properties, demonstrating that the nanocarrier could be a promising drug delivery system (DDS) for cancer therapy.

The hairy poly(methacrylic acid- co -divinylbenzene)- g -poly( N -isopropylacrylamide) (P(MAA- co -DVB)- g -PNIPAm) nanocapsules with pH-responsive P(MAA- co -DVB) inner shell and temperature-responsive PNIPAm brushes were prepared by combined distillation–precipitation copolymerization and surface thiol-ene click grafting reaction using 3-(trimethoxysilyl)propyl methacrylate-modified silica (SiO 2 -MPS) nanospheres as a sacrificial core material. The well-defined PNIPAm was synthesized by a reversible addition fragmentation chain transfer (RAFT) polymerization. The chain end was converted to a thiol by chemical reduction. The PNIPAm was integrated into the nanocapsules via thiol-ene click reaction. The surface thiol-ene click reaction conduced to tunable grafting density of PNIPAm brushes. The grafting densities decreased from 0.70 chains nm −2 to 0.15 chains nm −2 with increasing the molecular weight of grafted PNIPAm chains. Using water soluble doxorubicin hydrochloride (DOX·HCl) as a model molecular, the tunable shell permeability of the nanocapsule was investigated in detail. The permeability constant can be tuned by controlling the thickness of the P(MAA- co -DVB) inner shell, the grafting density of PNIPAm brushes, and the environmental pH and temperature. The tunable shell permeability of these nanocapsules results in the release of the loaded guest molecules with manipulable releasing kinetics. © 2014 Wiley Periodicals, Inc. J. Polym.
