Chronic wounds have emerged as a formidable challenge in diabetes management, leading to high morbidity and mortality. The diabetic wound healing is often impaired due to recurrent bacterial infections, excessive reactive oxygen species (ROS), and severe hypoxia. Thus, we developed a multifunctional nanoplatform that exhibited remarkable antibacterial activity, ROS scavenging and oxygen-carrying properties. Metal-phenol network (MPN) as core material was synthesized by assembling tannic acid (TA) with Fe 2 ⁺ via coordination and hydrophobic interactions. Subsequently, hemoglobin (Hb) was decorated onto MPN surface through cation-π interactions. MPN protected Hb against oxidation and preserved its physiological function, facilitating oxygen transport. Moreover, MPN-Hb displayed robust and broad-spectrum antioxidant activity that can effectively scavenge ROS and reactive nitrogen species. Under near-infrared (NIR) irradiation, MPN-Hb demonstrated excellent bactericidal activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), effectively eradicated biofilms through hyperthermia. In vivo studies have shown that MPN-Hb with good biocompatibility could effectively eliminate bacteria at the wound site, mitigate the inflammatory response, and promote collagen deposition and angiogenesis, thereby accelerating wound healing. Overall, this multifunctional nanoplatform offers a straightforward, safe, and efficient therapeutic strategy for treating chronic diabetic wounds.

Grape pomace represents a major organic solid waste generated by the wine industry, but its application has been largely unexplored. On the other hand, so far, stable and sustainable raw materials for producing stable, edible Pickering emulsifiers suitable for the food industry have been lacking. To solve these problems, this study established a mild but effective co-extraction method to obtain protein–polysaccharide–polyphenol ternary complexes (GPTCs) from grape pomace. Subsequently, these complexes were directly developed into an edible Pickering emulsifier by a pH-controlled method. Results showed that a series of properties related to the Pickering emulsifier, such as particle size, surface charge, wettability, and interfacial adsorption behavior, could be easily controlled by adjusting the solution’s pH. Consequently, the GPTC prepared at pH 7.0 exhibited optimal emulsifying performance. The resulting particles had an average particle size of approximately 111 nm, and stabilized Pickering emulsions with a volume-weighted mean oil droplet diameter (D [4,3]) of 9.49 μm, indicating high emulsion stability. Collectively, this study provided an actionable approach for the green, high-value utilization of wine byproducts by establishing a pH-responsive design framework for edible Pickering emulsifiers.

Colorectal cancer (CRC) is an aggressive malignancy with very limited therapeutic approaches. Drug resistance develops as a frequent characteristic in many patients with CRC, which leads to a decrease in the therapeutic efficacy of anticancer agents. Our previous evidences showed that bound polyphenol from millet bran (BPIS) possesses the potential of inhibiting cancer cell proliferation, and its main anticancer components are ferulic acid (FA) and p-coumaric acid (p-CA). In the present study, we found that BPIS significantly increases the sensitivity of human drug-resistant CRC cell line to oxaliplatin (OXA), a commonly used chemotherapy drug against CRC. Mechanistically, we indicated that BPIS significantly impairs the expression of a gene encoding multidrug resistance protein 1 (MDR1), a well-known permeability glycoprotein (P-gp), by preventing ganglioside GM3 catabolism. Neuraminidase 3 (NEU3) is a key enzyme catalyzing the conversion of ganglioside GM3 to ceramide trihexosides (Gb3), whose expression is increased in drug-resistant HCT-116/L cells. BPIS treatment increased GM3 level, but reduced Gb3 and P-gp levels by inhibiting NEU3 expression, which subsequently boosted the chemotherapy sensitivity of drug-resistant HCT-116/L cells to OXA. These findings reveal that BPIS increases the chemo-sensitivity by remodeling NEU3-mediated ganglioside GM3 catabolism, and it may be applied as a novel drug for facilitating the effectiveness of chemotherapeutic agents in CRC.

Resveratrol, a dietary polyphenol from grapes and berries, was evaluated for its protection against high-fat diet (HFD)-induced testicular dysfunction via the gut-microbiota-testis axis. In mice, resveratrol improved sperm quality and systemic metabolism. It reshaped gut microbiota (enriching Bifidobacterium) and normalized testicular metabolites, including the antioxidant 1,4-dithiothreitol and membrane-stabilizing heptadecanoic acid. Crucially, network pharmacology predicted specific targets, TNF-α, IL-1β, PTGS2, HRAS, and the TCA cycle enzyme SDHB, which led us to hypothesize and validate that resveratrol enhances mitochondrial TCA cycle activity (upregulating SDHB, ACO2, CS, MDH2). Concurrently, it suppressed the NF-κB pathway and promoted repair processes. Molecular docking confirmed stable binding to these targets. This work demonstrates that dietary resveratrol alleviates testicular impairment, a mechanism pinpointed through computational prediction, supporting its role as a nutritional intervention for male subfertility.

Skin diseases affect approximately 30%–70% of the global population and significantly reduce quality of life. Although polysaccharide-based dissolving microneedles (DMNs) are a minimally invasive transdermal delivery platform with strong clinical potential, their loose molecular packing and weak intermolecular interactions often lead to insufficient mechanical strength, increasing the risk of bending or fracture during insertion into pathological skin. In this study, a metal–polyphenol network (MPN)-mediated dual-enhancement strategy was developed by incorporating an EGCG–iron ion self-assembled precursor into a hyaluronic acid (HA) matrix to generate the preferred MPN-enhanced DMNs (PMN). MPN incorporation regulated the molecular organization of the HA polysaccharide matrix through synergistic hydrogen bonding and metal–ligand coordination interactions, leading to enhanced structural integrity and mechanical robustness. Under 808 nm near-infrared irradiation (NIR), PMN generated localized photothermal heating that disrupted dense tissue barriers and promoted molecular transport, resulting in a 2.67-fold increase in transdermal drug penetration compared with conventional DMNs. In melanoma-bearing mice model, PMN combined with NIR irradiation achieves an 83.5% tumor inhibition rate in a B16 melanoma model with good biosafety. Overall, this work provides mechanistic insights into the structure–property relationship of MPN-regulated polysaccharide matrix and offers a versatile strategy for constructing mechanically robust and multifunctional polysaccharide-based transdermal delivery systems. To investigate the digestive behavior of extruded starch-polyphenols system, extruded sweet potato starch vermicelli (ESPSV) was used as a model. The multi-scale structure, starch digestibility, polyphenol release, digestive enzyme activity during digestion and their correlation of ESPSV supplemented with matcha (MT), green tea extract (GTE), tea polyphenols (TP) and epigallocatechin gallate (EGCG) (at 1% polyphenol level) were discussed. Results showed that tea products in whatever form could retard starch digestion, with EGCG working best. The predicted glycemic index (pGI) of ESPSV was decreased from 82.50 to 65.46 after adding EGCG. Starch formed larger molecular aggregates with tea products under extrusion, showing a “B + V” type pattern. The order of V-type crystals content was EGCG + ESPSV (1.41) > TP + ESPSV (1.50) > GTE + ESPSV (1.88) > MT + ESPSV (2.62) > ESPSV (3.20). Under external pressure, EGCG, as tea monomer, was more likely to enter the spiral cavity of amylose and form V-type inclusion complex. Notably, polyphenols released during digestion could still reduce digestive enzyme activity , with a 15.53% decrease in EGCG + ESPSV compared to ESPSV. This was verified by correlation analysis, where RDS content (0.961, p < 0.01 ) and pGI (0.966, p < 0.01 ) were highly significantly correlated with the enzyme activity. Furthermore, tea products did not break or even enhance the quality of ESPSV as the final product.

Previous research had proved that collagen fiber can be used as separation material for removing caffeine from tea polyphenol extract effectively. But the recovery rate of catechin compounds from collagen fiber column was quite low by using water and ethanol as eluent because of excessively strong hydrogen bond interaction between collagen fiber and polyphenols. To solve the problem, collagen fiber was hydrophobically modified in this work by using silane coupling agents with alkyl chains (butyl, octyl, dodecyl and octadecyl). FT-IR, DSC, SEM and EDS Mapping proved the successful grafting of alkyl chains on collagen fiber, leading to the significantly enhanced hydrophobicity of collagen fiber. Static adsorption showed that the adsorption capacity of EGCG decreased when the grafted alkyl chain length increased mainly relying on the weakened hydrogen bond interaction. Dynamic adsorption and elution behavior also indicated less retention capacity of EGCG on collagen fiber with higher hydrophobicity. In column separation of simulated tea polyphenol extract containing caffeine and catechin compounds, the collagen fiber grafted with octadecyls (CF(C18)) presented excellent separation efficiency for caffeine and catechin compounds, and simultaneously the catechin compounds were highly recovered. The recovery rates of EGC and EGCG were 2–7 times higher than those separated by pristine collagen fiber column and glutaradehyde crosslinked collagen fiber column developed in our previous work. In the reusing round of CF(C18) column, the chromatogram was almost the same as the first round, showing excellent reusability. In fact, the separation performance of CF(C18) to caffeine and catechin compounds was comprehensively superior to Sephadex LH-20, presenting great practical application potential in effectively removing caffeine from tea polyphenol extract and recovering catechin compounds.

High-purity eicosapentaenoic acid (EPA) ethyl ester (EPA-EE) can be produced from an integrated technique consisting of saponification, ethyl esterification, urea complexation, molecular distillation and column separation. In order to improve the purity and inhibit oxidation, tea polyphenol palmitate (TPP) was added before the procedure of ethyl esterification. Furthermore, through the optimization of process parameters, 2:1 (mass ratio of urea to fish oil, g/g), 6 h (crystallization time) and 4:1 (mass ratio of ethyl alcohol to urea, g/g) were found to be the optimum conditions in the procedure of urea complexation. Distillate (fraction collection), 115 °C (distillation temperature) and one stage (the number of stages) were found to be the optimum conditions for the procedure of molecular distillation. With the addition of TPP and the above optimum conditions, high-purity (96.95%) EPA-EE was finally obtained after column separation.

The inhibitory activity of hesperetin on polyphenol oxidase (PPO) and their interaction characteristics were investigated using multiple spectroscopic methods and computational simulation. Hesperetin, a mixed inhibitor, reversibly inhibited PPO activity, and its half-maximum inhibitory concentration (IC50) values on monophenolase and diphenolase were 80.8 ± 1.4 μM and 776.0 ± 15.5 μM, respectively. Multivariate curve resolution–alternate least squares (MCR–ALS) analysis suggested PPO interacted with hesperetin and formed PPO–hesperetin complex. Hesperetin statically quenched PPO’s endogenous fluorescence, and hydrophobic interactions mainly drove their binding. Hesperetin affected the polarity of the microenvironment around the Trp residues in PPO, but had no effect on that around Tyr residues. Circular dichroism (CD) results showed that hesperetin increased α-helix content and decreased β-fold and random coil contents, thus tightening PPO’s structure. Molecular docking showed that hesperetin entered the hydrophobic cavity of PPO, bound near the dinuclear copper active center, interacted with Val283, Phe264, His85, Asn260, Val248, and His263 via hydrophobic interactions, formed hydrogen bonds with Met280, His89, and His259 residues and also interacted with Phe292, His61, Phe90, Glu256, His244, Asn260, Phe264, and Gly281 via van der Waals forces. The molecular dynamics simulation results also demonstrated that the addition of hesperetin reduced the stability and hydrophobicity of PPO and increased PPO’s structural denseness. Thus, the inhibition of hesperetin on PPO may be because hesperetin bound near the active center of PPO, interacted with the surrounding residues, occupied the binding site for substrate, and induced the changes in PPO’s secondary structure, thus inhibiting the catalytic activity of PPO. This study may provide novel views for the inhibition of hesperetin on PPO and theoretical guidance for developing flavonoids as new and efficient PPO inhibitors. Keywords:hesperetin;polyphenol oxidase;inhibition mechanism;conformational change;computational simulation

Plant polyphenols can form functional coatings on various materials through self-polymerization. In this paper, a series of modified capillary columns, which possess diversity of charge characteristics for modulating electroosmotic flow (EOF), were prepared by one-step co-deposition of gallic acid (GA), a plant-derived polyphenol monomer , and branched polyethyleneimine (PEI). The physicochemical properties of the prepared columns were characterized by Fourier transform infrared spectroscopy (FT-IR), UV–Vis spectroscopy and scanning electron microscopy (SEM). The magnitude and direction of EOF of GA/PEI co-deposited columns were modulated by changing a series of coating parameters, such as post-incubation of FeCl 3 , co-deposition time, and deposited amounts of GA and PEI with different relative molecular mass (PEI-600, PEI-1800, PEI-10000, and PEI-70000). Furthermore, the separation efficiencies of the prepared GA/PEI co-deposited columns were evaluated by separations of small molecules , including organic acids, polar nucleotides, phenols, nucleic acid bases and nucleosides . Results indicated that modulating of EOF plays an important role in enhancing the separation performance and reversing the elution order of the analytes. Finally, the developed method was successfully applied to quantitative analysis of acidic compounds in four real samples. The recoveries were in the range of 73.5%–85.8% for citric acid , benzoic acid , sorbic acid , salicylic acid and ascorbic acid in beverage and fruit samples, 101.6%–104.9% for cinnamic acid , vanillic acid , and ferulic acid in Angelica sinensis sample, while 84.6%–97.8% for guanosine-5′-monophosphate, uridine-5′-monophosphate, cytosine-5′- monophosphate and adenosine-5′-monophosphate in Cordyceps samples. These results indicated that the co-deposition of plant polyphenol-inspired GA/PEI coatings can provide new opportunities for EOF modulation of capillary electrophoresis .

The main purpose of this work was to investigate the influence of typical thermal sterilization approaches (pasteurization, high-temperature sterilization) on the structure, in vitro digestibility , and antioxidant activity of soy protein isolate (SPI)/black soybean seed coat extract (BE) complexes at pH 7.0. The results of zeta potential and particle size demonstrated that the addition of BE was contributed to inhibit protein thermal aggregation. Heat sterilization resulted in protein unfolding revealed by UV–Vis, circular dichroism , and fluorescence spectroscopy analysis. The increase of BE led to the reduction of fluorescence intensity and surface hydrophobicity of SPI. Moreover, the formation of SPI/BE complexes inhibited protein digestion in vitro, while thermal sterilization promoted protein digestion. The SPI/BE complexes showed a strong radical scavenging ability both before and after thermal treatment.

Postfermented Pu-erh tea (PE) protects against metabolic syndrome (MS), but little is known regarding its underlying mechanisms. Animal experiments were performed to determine whether the gut microbiota mediated the improvement in diet-induced MS by PE and its main active components (PEAC). We confirmed that PE altered the body composition and energy efficiency, attenuated metabolic endotoxemia and systemic and multiple-tissue inflammation, and improved the glucose and lipid metabolism disorder in high-fat diet (HFD)-fed mice via multiple pathways. Notably, PE promoted the lipid oxidation and browning of white adipose tissue (WAT) in HFD-fed mice. Polyphenols and caffeine (CAF) played critical roles in improving these parameters. Meanwhile, PE remodeled the disrupted intestinal homeostasis that was induced by the HFD. Many metabolic changes observed in the mice were significantly correlated with alterations in specific gut bacteria. Akkermansia muciniphila and Faecalibacterium prausnitzii were speculated to be the key gut bacterial links between the PEAC treatment and MS at the genus and species levels. Interestingly, A. muciniphila administration altered body composition and energy efficiency, promoted the browning of WAT, and improved the lipid and glucose metabolism disorder in the HFD-fed mice, whereas F. prausnitzii administration reduced the HFD-induced liver and intestinal inflammatory responses. In summary, polyphenol- and CAF-rich PE improved diet-induced MS, and this effect was associated with a remodeling of the gut microbiota.
