Catalytic synthesis of vitamin E acetate using metal organic framework material ZIF-8

Mingzhe Li,Yuyang Zhou,Hua Zhou,Dingqiang Lu

Vitamin E is an important fat soluble antioxidant, its acetate is the main derivative product of vitamin E. Using metal organic framework material ZIF-8 as a catalyst, vitamin E acetate (VEA) was prepared by catalyzing the esterification reaction of D-α-tocopherol and acetic anhydride in a solvent-free system. Firstly, the preparation conditions of metal organic framework materials were studied: different imidazole ligands, different zinc ion ligands, molar concentration ratio of Zn2+ to 2-methylimidazole(2-mIM), and preparation time. The results showed that the catalytic performance of ZIF-8 was optimal when Zn(CH3COO)2·2H2O was used as the zinc source, 2-mIM was used as the imidazole ligand, the molar concentration ratio of Zn2+ / 2-mIM was 1:20, and the preparation time was 1 hour. The yield of vitamin E acetate was 86.85%. The synthesis process of VEA was optimized by single factor experiments such as substrate molar ratio, reaction time, dosage of nano enzyme, reaction temperature and so on. The results showed that the yield of vitamin E acetate reached 96.31% under the optimal conditions of n (D-α-tocopherol) =1mmol, [n (D-α-tocopherol) : n (Acetic anhydride)] =1:15, reaction time 24 h, ZIF-8 dosage 0.05 g, reaction temperature 50 °C.

Leveraging Diffusion Kinetics to Reverse Propane/Propylene Adsorption in Zeolitic Imidazolate Framework-8

Linghe Yang,Ying Liu,Fang Zheng,Fuxing Shen,Baojian Liu,Rajamani Krishna,Zhiguo Zhang,Qiwei Yang,Qilong Ren,Zongbi Bao

The separation challenge posed by propylene/propane mixtures arises from their nearly identical molecular sizes and physicochemical properties. Metal–organic frameworks (MOFs) have demonstrated potential in addressing this challenge through the precision tailoring of pore sizes and surface chemistry. However, introducing modifications at the molecular level remains a considerable hurdle. This work presents an approach to reversibly tune the propylene/propane adsorption preference in zeolitic imidazolate framework-8 (ZIF-8) by manipulating the particle size and gas flow rate. Systematically increasing the ZIF-8 crystals from 9 to 224 μm restricts propane diffusion, thereby reversing its preferential adsorption over propylene. Furthermore, raising the gas flow rate of mixed propylene/propane shifts the rate-determining breakthrough step from thermodynamic equilibrium to kinetics, again reversing the adsorption preference in a particular ZIF-8 sample. We propose “dynamic selectivity (Sd(t))” as a concept that incorporates both thermodynamic and kinetic factors to elucidate these unexpected findings. Moreover, the driving force equation, grounded on the concept of Sd(t), has improved the precision and stability of the computational simulation for fixed-bed adsorption processes. This work underscores the potential of diffusion-based modulation, implemented through manageable external changes, as a viable strategy to optimize separation performance in porous adsorbent materials.

Multiple synergistic antibacterial melamine-impregnated paper based on nano Ag-doped ZIF-8

Jinrong He,Wei Qu,Yun Feng,Jinrui Jiang,Jiaming Luo,Yuzhang Wu,Limin Peng

Melamine-impregnated paper, a decorative material in furniture and interior design, could potentially impede the spread of indoor bacteria if it possesses antibacterial properties. Here, nano Ag-doped ZIF-8 (Ag@ZIF-8) was synthesized using ion exchange and in-situ chemical reduction strategies, followed by incorporating it into melamine-impregnated paper. The ion exchange mechanism of Zn 2+ and Ag + during ion exchange reactions was investigated. The structure of Ag@ZIF-8 was well-defined, with Ag nanoparticles evenly distributed on the surface of ZIF-8. Ag@ZIF-8 showed good thermal stability at 400 °C, meeting the requirements for the hot-pressing process of the melamine-impregnated paper. Ag@ZIF-8 exhibited enhanced ROS generation under visible light compare to ZIF-8, which was attributed to its reinforced light absorption and charge carrier separation facilitated by the localized surface plasmon resonance (LSPR) effect. The antibacterial activity of Ag@ZIF-8 was better than ZIF-8, owing to the multiple-synergistic antibacterial mechanisms of the ion release and ROS generation. Specifically, the 10-Ag@ZIF-8 exhibited MIC of 128 μg/mL and 64 μg/mL against E. coli and S. aureus , respectively. The signal intensity of •O 2   −  induced by 10-Ag@ZIF-8 was 4.38 times higher than that of ZIF-8. As the addition of Ag@ZIF-8 increased, the antibacterial performance of melamine-impregnated paper improved. Considering the antibacterial property and economy of melamine-impregnated paper, 0.20 wt% was the optimal amount for 10-Ag@ZIF-8 decorated blockboard. This study proposes a novel approach for synthesizing MOF-based melamine-impregnated paper with antibacterial properties, aiming to reduce bacterial spread, minimize disease risks, and contribute to lowering overall healthcare costs.

Molecularly imprinted Photoelectrochemical sensor for Escherichia coli based on Cu:ZIF-8/KZ3TTz heterojunction

Xiaoqing Wang,Xufeng Zang,Laiyi Deng,Fei Tan,Xingbo Liu,Zhiguo Zhang,Bo Cui,Yishan Fang

Herein, a signal stable molecularly imprinted photoelectrochemical (MIP-PEC) sensing platform was designed to sensitively detect Escherichia coli by incorporating polythiophene film with Cu: ZIF-8/KZ3TTz heterojunction . Attributed to the formation of a staggered type II heterostructure between KZ3TTz and Cu: ZIF-8 semiconductors, the Cu: ZIF-8/KZ3TTz heterojunction exhibited stable and significant cathode PEC response. Impressively, selective MIP film was grown on the surface of Cu: ZIF-8/KZ3TTz/GCE by electro-polymerization of 2,2-Dimethyl-5-(3-thienyl)-1,3-dioxane-4,6-dione (DTDD) in the presence of E. coli . After removing E. coli , more electrons were transferred to the electrolyte solution through the imprinting cavity on the MIP film, which was eliminated by O 2 in the electrolyte, causing further enhancement of the cathode PEC response. On the contrary, when the imprinted cavity was filled with E. coli , the cathodic PEC response gradually decreased due to steric hindrance effect. The sensor showed excellent linearity in the range of 10 1 to 10 8  CFU/mL with a detection limit of 4.09 CFU/mL (S/N = 3). This strategy offered a novel approach for pathogenic bacteria detection in food safety and environmental monitoring

Efficient Low-Pressure CO2 capture via ZIF-8 modified by deep eutectic solvents

Duoyong Zhang,Chen Zhang,Xinqi Zhang,Yongqiang Tian,Xusheng Cheng,Xuancan Zhu,Liwei Wang

To enhance CO 2 sorption efficiency under low-pressure conditions and effectively compete with N 2 sorption in flue gas, modifications are implemented aimed at improving the adsorption capacity and selectivity of ZIF-8, renowned for their robust stability. Deep eutectic solvent (DES) comprising tetraethylammonium chloride (TEAC), tetrapropylammonium chloride (TPAC), and tetrabutylammonium bromide (TBAB) as hydrogen-bond acceptors, along with ethanolamine (MEA) as the hydrogen-bond donor, were meticulously prepared for this purpose. ZIF-8 underwent modification through DES loading, resulting in the distinctive emergence of a “core-membrane” structure. Characterization revealed that the DES effectively adhered to the surface of ZIF-8 in a membrane-like structure, without altering the chemical structure or pore size of ZIF-8. The most significant enhancement in sorption capacity was observed with TPAC&MEA modification. Considering the presence of N 2 partial pressure in the flue gas, at 0.05 and 1 bar, the CO 2 adsorption capacities reached 1.92 and 3.03 mmol g −1 , respectively, increasing 71.11 and 4.00 times compared to pristine ZIF-8. The ZIF-8 exhibited a CO 2 /N 2 separation coefficient of 72.77, which increased to a maximum of 997.50 after DES modification. Additionally, the modified material demonstrated exceptional cyclic and thermal stability during testing. This study significantly elevates the CO 2 adsorption capacity and selectivity of solid materials within the low-pressure range, providing pivotal support for CO 2 capture and decarbonization efforts.

Combined Photothermal/Combination Chemotherapeutic Approach Using Zif-8-Supported Artesunate and Gold Nanorods

Xing Li,Jun Wang,Wanzhen Li,Feiyang Xu,Yuan Gao,Ting Cui,Kai Yang,Weiwei Zhang,Fei Ge,Yugui Tao

Tumors pose a significant threat to human well-being, presenting substantial risks to lives. Conventional approaches to cancer treatment exhibit limited efficacy in eliminating the disease, leading to undesirable outcomes and an increased likelihood of disease relapse. The application of chemotherapy and photothermal therapy has recently shown an increasing trend, owing to their limited negative effects and exceptional specificity. Chemotherapy can potentially induce a rapid reduction in tumors during initial treatment. However, it carries the risk of inducing drug resistance, ultimately contributing to tumor recurrence. Combination therapy offers more advantages, than single chemotherapy. Photothermotherapy and photodynamic therapy have gained significant public interest as innovative approaches to medical treatment. Nonetheless, gold nanorods demonstrate notable potential as exceptional photothermal agents in photothermal therapy. An acidic microenvironment is commonly found in tumors. With this factor considered, a tumor-specific cleavable material is used to encapsulate the drug for facilitating controlled release exclusively within the tumor microenvironment. The combination of dequalinium chloride and artesunate increases its selectivity toward mitochondria, enhancing its effectiveness as a chemotherapy agent. The gold nanorods were successfully employed to encapsulate the drug with ZIF-8, enabling precise transportation to the tumor location while preserving its stability. The therapeutic effectiveness of the treatment on mouse models with NCI-H1299 tumors was highly satisfactory, also demonstrating high biological safety, revealing its potential capabilities in cancer therapy. It provides a research basis for nanomaterials to inhibit tumors.

Rui Deng,Jibin Wu,Baokang Zhu,Gaoqing Song,Tao Zhou,Musheng Yang,Lingling Pan,Jian Wang,Xuan Zou,Ziquan Lv,Xiaobao Jin,Yinghua Xu,Xuemei Lu,Shuiqing Gui

Hepatocellular carcinoma (HCC) has emerged as one of the most prevalent and highly malignant tumor malignancies worldwide and is characterized by poor prognosis and high mortality. Therefore, effective medications are urgently needed to improve therapeutic effects with HCC. Exosomes are nanoscale vesicles that can transfer cellular information. Owing to their excellent biocompatibility, exosomes are frequently utilized as drug carriers for cancer treatment. Targeted drug delivery systems that modify exosomes to increase targeting have emerged as promising therapeutic platforms in response to the lack of precise targeting by natural exosomes. In this study, HTPP-Exo was successfully constructed by modifying the exosome membrane by connecting sulfhydryl group of HTPP with maleimide in DSPE-PEG2000-Mal, and the antimicrobial peptide M1–8 was included for the first time through co-incubation, thus successfully establishing the HTPP-Exo-M1–8 targeted drug delivery system. The targeting ability of HTPP-Exo-M1–8 was verified by evaluating its in vitro and in vivo targeting of HepG2 cells, and its therapeutic ability was evaluated by its inhibitory effect on HepG2 cells and in situ liver cancer in mice. The results showed that HTPP-Exo-M1–8 targeted drug delivery system could target HepG2 cells in vivo and in vitro , and the survival rate of HepG2 cells was 45 % after HTPP-Exo-M1–8 treatment for 48 h. After in vivo treatment, the volume of HCC in HTPP-Exo-M1–8 group (270.4 mm 3 ) was much smaller than that in control group (969.5 mm 3 ), which achieved the expected purpose of HCC treatment. Further research showed that the targeted drug delivery system HTPP-Exo-M1–8 could be used to treat HCC by inhibiting the lysosome–autophagosome fusion in HepG2 cells. Additionally, there was no obvious toxicity to healthy cells or tissues when HTPP-Exo-M1–8 was used. Overall, the exosome-based targeted drug delivery system HTPP-Exo-M1–8 can treat HCC by targeting and penetrating tumor cells, which provides a new strategy for the treatment of HCC.

Hongju Cheng,Md All Amin Newton,Mia Rajib,Qinchen Zhang,Weihong Gao,Zan Lu,Yuansheng Zheng,Zijian Dai,Jie Zhu

Designing wound dressings necessitates the crucial considerations of maintaining a moist environment and implementing effective bacterial control. Furthermore, developing a three-dimensional framework emulating the extracellular matrix (ECM) confers advantages in fostering cellular migration and proliferation. Inspired by this, hydrogel/nanofiber composites have been demonstrated as promising materials for wound dressings. The composites also overcome the disadvantages of poor mechanical properties and rapid release of traditional pure hydrogels. In this study, we constructed a calcium alginate hydrogel/polylactic acid nanofiber (CAH/PLANF) composite with an interpenetrated network. Additionally, the synthesis of zeolitic imidazolate framework-8 (ZIF-8) incorporated into the composite system endowed the system with enhanced mechanical properties and photodynamic antibacterial attributes. The obtained composite patch (ZIF-8@CAH/PLANF) exhibited excellent swelling, strong mechanical properties, low cytotoxicity, and durable photodynamic antibacterial effect with an antibacterial efficacy of higher than 99.99%. Finally, bacterial infection and wound healing properties were investigated in vivo, and the ZIF-8@CAH/PLANF patch was proven to have the ability to fight infection and accelerate wound healing.

Biotin-decorated celastrol-loaded ZIF-8 nanoparticles induce ferroptosis for colorectal cancer therapy

Jie Chen,Yan Liu,Yanting You,Ruisi Zhou,Yanchun Li,Xinghong Zhou,Xiaohu Chen,Ying Yang,Jinxiang Chen,Hiu Yee Kwan,Xiaoshan Zhao,Yuyao Wu

Celastrol (CEL) has garnered significant interest for its anti-tumour properties and potential for colorectal cancer (CRC) treatment. However, its clinical use is constrained by its limited bioavailability and potential toxicity. Herein, a biotin-decorated CEL nano-drug delivery system using a zeolitic imidazolate framework (ZIF-8) as a carrier was synthesized, named CEL@ZIF-8@BIO. It exhibits excellent water solubility, efficient loading of CEL, and a high release rate in acidic environments. In vitro experiments demonstrated that CEL@ZIF-8@BIO inhibited proliferation, induced cell cycle arrest in G0/G1 phase, increased ROS production and reduced mitochondrial membrane potential in CRC cells. RNA-Seq analysis indicated that the anticancer mechanism of CEL@ZIF-8@BIO may be linked to ferroptosis as indicated by a significant increase in Fe 2 +  levels, oxidative stress, lipid peroxidation, and mitochondrial dysfunction in CRC cells. Furthermore, these effects could be reversed by ferroptosis inhibitor. In vivo research revealed that CEL@ZIF-8@BIO significantly inhibited the growth of CRC tumours and reduced the toxicity associated with CEL treatment. Taken together, the CEL@ZIF-8@BIO nano-drug delivery system shows great promise for CRC therapy due to its improved efficacy and reduced toxicity.

AuNPs-based lateral flow strip genotoxicity biosensor by sensitive quantification of 8-oxodGuo

San-Jun Shi,Rong-Fu Huang,Zi-Yan Fan

Development of rapid and sensitive methods for screening of large number of chemical pollutants with potential genotoxicity is in urgent demands. In this work, we reported a chemical genotoxicity sensor based on the lateral flow strip (LFS) quantification of the 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxodGuo). The proposed sensing strategy introduced human 8-oxoguanine DNA glycosylase (hOGG 1) and human apyrimidinic endonuclease (APE 1) to convert 8-oxodGuo to strand break. Then, the DNA conjugated gold nanoparticles (AuNPs) complexes could not be captured on test line (T-line), showing visible red color fading. Additionally, the mean grey value of the optical density in the T-line zone was analyzed by using a mobile phone camera and Image J software to quantify 8-oxodGuo. Under the optimized experimental conditions, the LFS DNA biosensor could detect the lowest concentration of 8-oxodGuo down to 0.2 pmol. To demonstrate its universal applications, in vitro generation of 8-oxodGuo by Rose Bengal under irradiation and Fe 2+ -mediated Fenton reagent were successfully measured. The detection limit of oxidative DNA damage induced by Rose Bengal or Fenton reagent could be down to 0.01 μM or 1 nM Fe 2+ /4 nM H 2 O 2 , respectively. The 10-fold higher sensitivity for Fenton reagent was attributed to much more severe damage than Rose Bengal, producing both DNA strand break and nucleobases conversion. The results demonstrated that the LFS DNA biosensor could detect oxidative DNA damage for the first time, showing its potential application for assessment of the genotoxicity of abundant chemical pollutants.

Bimetallic ZIF-8 from Hydroxide Double Salts for Efficient Cu2+ Removal in Wastewater

Yixin Chen,Zexi Chen,Sheng Chu

Metal–organic frameworks (MOFs) hold significant potential for applications in gas adsorption and separation, catalysis, chemical sensing, and drug delivery. Zeolitic imidazolate frameworks (ZIFs) are a type of MOF composed of metal ions and imidazolate ligands, structurally similar to zeolite structures. ZIF-8, a widely studied ZIF material, is composed of zinc ions (Zn2+) and 2-methylimidazole as fundamental building blocks featuring unique porous structures, a high specific surface area, and excellent thermal and chemical stability. This study introduces a rapid room-temperature synthesis method for bimetallic ZIF-8 through the hydroxide double salt (HDS) precursor. Various characterization techniques confirmed that the synthesized bimetallic ZIF-8 exhibits uniform particle size and high crystallinity. Experimental results indicate that the HDS precursor provides numerous active sites, facilitating rapid nucleation and resulting in uniformly sized bimetallic ZIF-8 particles. By optimizing the ultrasonic time of HDS, the concentration of 2-methylimidazole, and the reaction time, the synthesis conditions were refined, producing bimetallic ZIF-8 particles with an average size of 135.0 nm and a minimum polydispersity index (PDI) of 0.024. Additionally, the copper ion adsorption performance was evaluated, with the synthesized bimetallic ZIF-8 showing the highest adsorption capacity of 1196.82 mg/g at pH 6, demonstrating its effectiveness in heavy metal removal.

Ting Liu,Kangkang Huang,Yuxia Yang,Shengwu Wen,Jingxian Zhang,Suiping Deng,Shaozao Tan,Langhuan Huang

Bacterial infection is the most common risk factor that causes the failure of implantation surgery. Therefore, the development of biocompatible implants with excellent antibacterial properties is of utmost importance. In this study, NIR light-driven AgBiS2@ZIF-8 hybrid photocatalysts for rapid bacteria-killing were prepared. AgBiS2@ZIF-8 exhibited excellent photocatalytic activity due to the rapid transfer of photoelectrons from AgBiS2 to ZIF-8, resulting in abundant reactive oxygen species (ROS) to kill bacteria. Meanwhile, AgBiS2@ZIF-8 exhibited a noteworthy photothermal effect, which could effectively convert NIR light into heat. Subsequently, the NIR light-driven antibacterial activity of AgBiS2@ZIF-8/Ti against S. aureus and E. coli was studied. The experimental results showed that AgBiS2@ZIF-8 displayed enhanced photodynamic therapy (PDT) and photothermal therapy (PTT) performance. Under irradiation with 808 nm NIR light for 10 min, AgBiS2@ZIF-8/Ti could effectively eliminate 98.55% of S. aureus in vitro, 99.34% of E. coli in vitro and 95% S. aureus in vivo. At the same time, AgBiS2@ZIF-8/Ti had good biocompatibility. Therefore, AgBiS2@ZIF-8/Ti showed potential as an antibacterial material, which provided a strategy to fight polymicrobial infections.
