Nanoscale cationic micelles of amphiphilic copolymers based on star-shaped PLGA and PEI cross-linked PEG for protein delivery application

Wang Jun,Li Shunying,Chen Tingting,Xian Wenjiao,Zhang Huiwu,Wu Lei,Zhu Wenting,Zeng Qingbing

To enhance the bioavailability of protein therapeutants and improve the stability of storage and delivery, a series of branched amphiphilic block copolymers consisting of cholic acid (CA) initiated poly(D,L-lactide-co-glycolide) (CA-PLGA) and water-soluble polyethyleneimine cross-linked polyethylene glycol (PEI-PEG) denoted as CA-PLGA-b-(PEI-PEG) were synthesized and characterized. CA-PLGA-b-(PEI-PEG) presented low cytotoxicity by MTT and cck-8 assay. The cationic CA-PLGA-b-(PEI-PEG) micelles (diameter about 100 nm and zeta potential 34–61 mV) were prepared through self-assembly method, and complexed with insulin via electrostatic interaction to obtain nanoscale micelle/insulin complexes. The micelle/insulin complexes-loaded CA-PLGA microspheres (MIC-MS, 10.4 ± 3.85 μm) were manufactured by employing a double emulsion (W 1 /O/W 2 ) method. The in vitro insulin release behavior and in vivo hypoglycaemic effect of MIC-MS on streptozotocin (STZ) induced diabetic rats were compared with those of the insulin-loaded CA-PLGA microspheres (INS-MS, 7.8 ± 2.57 μm). The initial burst in vitro release of MIC-MS was markedly lower than that of INS-MS ( P  < 0.01), and the pharmacological availability of MIC-MS via subcutaneous administration was 148.9% relative to INS-MS. Therefore, the cationic CA-PLGA-b-(PEI-PEG) micelles can effectively increase the bioavailability of insulin in CA-PLGA microspheres and can be considered as a potential protein carrier.

Synthesis of a small molecular polydentate polymer IHP-MP and its application to cement or graphene oxide dispersion via strong interactions between adsorption groups and interfaces

Ma Jianfeng,Zhao Hongxia,Shang Yazhuo,Peng Changjun,Liu Honglai,Zheng Shuzhen,Wang Tao,Qi Shuai,Ran Qianping

Abstract The presence of a highly alkaline environment or large amounts of Ca 2+ in concrete exerts a great influence on the dispersion of graphene oxide (GO) particles. Therefore, the development of a highly efficient GO dispersant to adapt to complex concrete environments is important. In this work, novel phosphate-modified small molecular polydentate polymers were synthesized and used as efficient dispersants for cement and high-concentration GO solutions. The chemical structure of the polymers was confirmed by 1 H NMR and IR spectroscopy, and the effect of adsorption groups on the adsorption ability of polymers was studied by using adsorption experiments and theoretical calculations. Results showed that the dispersant inositol hexaphosphate–methoxy polyether (IHP-MP) features a high adsorption equilibrium constant and strong adsorption ability. IHP-MP exhibited not only good water-reducing and slump-retaining properties but also remarkable cement hydration-retarding effects due to its numerous phosphate groups. IHP-MP could be used as an auxiliary GO dispersant with the comb polymer PCE to effectively reduce the viscosity and improve the stability of GO solutions. SEM, TGA and IR characterization indicated that IHP-MP strongly interacts with the surface groups of GO particles. Graphic Abstract

Efficient and stable Ni-supported novel porous Ca-Mg-Al complex oxide catalyst for catalytic pyrolysis of oleic acid into gasoline-kerosene rich production using methanol as a hydrogen donor: Synthesis methods and process optimization

Jida Wang,Feng Shi,Can Liu,Yi Lu,Xu Lin,Defa Hou,Jirong Li,Dechao Wang,Zhifeng Zheng,Yunwu Zheng

In this study, a series of cost-effective and eco-friendly Ca-Mg-Al and Ni-modified Ca-Mg-Al catalysts were successfully synthesized with different methods (coprecipitation, impregnation and mechanical mixing method) and applied to catalytic pyrolysis vapor upgrading of oleic acid (OA) to produce gasoline-kerosene rich production using methanol as a hydrogen donor . In addition, the effects of the process parameters (catalyst type, OA to methanol ratio, pyrolysis/catalytic temperature, catalyst loading, feedstock injection volume) on the product yield and selectivity were explored, and the reuse properties and deactivation mechanism were also evaluated. Active Ni species coupled with methanol were more conducive to enhancing the catalyst activity and OA conversion and obviously increased the selectivity toward the gasoline-kerosene product. The impregnation method was favorable for the formation of NiAl 2 O 4 species, and coprecipitation method suppressed the formation of NiAl 2 O 4 species. The enhanced activity and deoxygenation selectivity of the coprecipitation method (NiCaMgAl) at lower pyrolysis temperatures was attributed to the abundant higher specific surface area and pore size, higher active metal oxide (Ni 2+ ) and well-active Ni dispersion with a smaller particle size, as well as the lower acidity and partially oxidized Ni, which suppressed the NiAl 2 O 4 spinel phase formation compared with the Ni/Ca-Mg/Al (impregnation) and Ca-Mg-Al (mixing) catalysts, which was beneficial to the decarbonylation/ decarboxylation reaction and inhibited the hydrocracking reaction of C C. Therefore, 100% conversion, 97.12% hydrocarbon yield, 54.53% gasoline and 58.80% kerosene content over NiCaMgAl catalyst were obtained under optimal conditions (pyrolysis/catalytic temperature = 450/500°C, catalyst loading = 1.5 g, feedstock injection volume = 0.1 ml/min and OA to methanol ratio = 3:1). Moreover, NiCaMgAl catalyst also demonstrated good reusability and hydrothermal stability performance due to the lower amount of coke deposition with a lower graphitized structure, which suppressed sintering.

Dual modes reinforced silk adhesives for tissue repair: Integration of textiles and inorganic particles in silk gel for enhanced mechanical and adhesive strength

Lu Liu,Enling Hu,Haoyu Qiu,Qian Xu,Kun Yu,Ruiqi Xie,Fei Lu,Qi Wang,Bitao Lu,Qing Li,Guangqian Lan

Skin wound healing in dynamic environments remains challenging. Conventional gels are not ideal dressing materials for wound healing due to difficulties in completely sealing wounds and the inability to deliver drugs quickly and precisely to the injury. To tackle these issues, we propose a multifunctional silk gel that rapidly forms strong adhesions to tissue, has excellent mechanical properties, and delivers growth factors to the wound. Specifically, the presence of Ca 2+ in the silk protein leads to a solid adhesion to the wet tissue through a chelation reaction with water-trapping behavior; the integrated chitosan fabric and CaCO 3 particles ensure enhanced mechanical strength of the silk gel for better adhesion and robustness during wound repair; and the preloaded growth factor further promoted wound healing. The results showed the adhesion and tensile breaking strength were as high as 93.79 kPa and 47.20 kPa, respectively. MSCCA@CaCO 3 -aFGF could remedy the wound model in 13 days, with 99.41 % wound shrinkage without severe inflammatory responses. Due to strong adhesion properties and mechanical strength, MSCCA@CaCO 3 -aFGF can be a promising alternative to conventional sutures and tissue closure staples for wound closure and healing. Therefore, MSCCA@CaCO 3 -aFGF is expected to be a strong candidate for the next generation of adhesives.
