Construction and Evaluation of a Novel MAP Immunoassay for 9 Nutrition-and-Health-Related Protein Markers Based on Multiplex Liquid Protein Chip Technique

Jiyong Yin,Jiangping Niu,Junsheng Huo,Jing Sun,Jian Huang,Chaoqun Sun

We attempted to construct and evaluate a novel detection method to realize simultaneous detection based on a multiplex liquid protein chip technique for nine nutrition-and-health-related protein markers to meet the requirement of an accurate, simultaneous and comprehensive analysis of the proteomics of nutrition and health. The lower limits of detection, biological limits of detection and regression equations of serum ferritin (SF), soluble transferrin receptor (sTfR), c-reactive protein (CRP), retinol-binding protein4 (RBP4), apolipoprotein B (ApoB), alpha-fetoprotein (AFP), prealbumin (PA), carcino-embryonic antigen (CEA) and D-Dimmer (D-D) were determined after a series of optimal experiments. Then, the results of the methodological evaluation for this novel method indicated that the accuracies were between 70.12% and 127.07%, the within-run precisions were between 0.85% and 7.31%, the between-run precisions were between 3.53% and 19.07%, the correlation coefficients between this method and other methods were above 0.504 (p< 0.05), and the direct bilirubin (DBIL) of low concentration and the indirect bilirubin (IBIL) of high concentration could not interfere with the detected results of nine indicators. The novel multiplex detection method, which can increase accuracy and improve the ability of comprehensive analysis, can basically meet the requirement of detection and the diagnosis of the proteomics of nutrition and health. Keywords:liquid protein chip;nutrition and health;protein marker;multiplex detection

Vat photopolymerization-based 3D printing of complex-shaped and high-performance Al2O3 ceramic tool with chip-breaking grooves: Cutting performance and wear mechanism

Haidong Wu,Wei Liu,Yuerui Xu,Lifu Lin,Yehua Li,Shanghua Wu

Due to the processing of alumina ceramic cutting tools with complex shapes using traditional methods is difficult and time-consuming, vat-photopolymerization-based 3D printing was adopted to fabricate Al2O3 ceramic cutting tools with grooves for the first time. Subsequently, cutting performance evaluation and wear mechanism analysis were conducted. The relative density, Vickers hardness, and bending strength of the alumina cutting tools were determined. The effects of the cutting speed, feed rate, and cutting depth on the cutting performance and wear mechanism of the cutting tools were systematically investigated. In addition, two commercial cutting tools, namely cemented carbide and ceramic tools without grooves, were used for comparison. The cutting speed has the highest influence on the cutting performance, whereas the cutting depth has the least influence. The cutting performance of the prepared alumina cutting inserts with chip breaker grooves superior to that those without chip-breaking grooves and that of the cemented carbide tools. The wear mechanisms of the prepared alumina cutting tools and commercial tools were determined to be abrasive and adhesive wear, and those of the cemented carbide tools were adhesive wear and breakage. This work opens a new avenue for the future preparation of high-performance and complex-shaped ceramic cutting tools.

Visual detection of the prostate specific antigen via a sandwich immunoassay and by using a superwettable chip coated with pH-responsive silica nanoparticles

Gao Jian Bang,Sann Ei Ei,Wang Xin Yu,Ye Cui,Liu Rui,Gao Zhong Feng

A pH-responsive superwettable chip is described whose surface can switch between superhydrophobic and superhydrophilic. It can be used for the visual detection of the prostate specific antigen (PSA) based on contact angle readout. Magnetic beads were modified with primary antibody against PSA. After immunobinding, gold nanoparticles loaded with secondary antibody labeled with glucose oxidase is added. On addition of glucose, gluconic acid is formed which causes a drop in the local pH value. This results in a wettability switch of the pH-responsive superwettable chip from hydrophobic to hydrophilic. Under the optimized conditions, PSA can be quantified with a 3.2 pg mL −1 limit of detection by analyzing the contact angle and the related color that changes from blue via orange to red. The method is applicable to PSA detection in serum samples and for visual classification by cancer patients and healthy persons. It is also suitable for color-blind and color-weak individuals. Conceivably, this kind of assay can be transferred to the determination of various kinds of other bioanalytes including nucleotide, proteins, and even of ions and small organic molecules, and thus is has a wide scope. Schematic presentation of a pH-responsive superwettable chip coated with silica nanoparticles for the visual detection of prostate specific antigen (PSA) by reading the contact angle. The superwettable chip achieves reliable clinical detection of serum PSA from prostate cancer patients.

Chip-based molecularly imprinted monolithic capillary array columns coated GO/SiO2 for selective extraction and sensitive determination of rhodamine B in chili powder

Haiyun Zhai,Lu Huang,Zuanguang Chen,Zihao Su,Kaisong Yuan,Guohuan Liang,Yufang Pan

A novel solid-phase extraction chip embedded with array columns of molecularly imprinted polymer-coated silanized graphene oxide (GO/SiO 2 -MISPE) was established to detect trace rhodamine B (RB) in chili powder. GO/SiO 2 -MISPE monolithic columns for RB detection were prepared by optimizing the supporting substrate, template, and polymerizing monomer under mild water bath conditions. Adsorption capacity and specificity, which are critical properties for the application of the GO/SiO 2 -MISPE monolithic column, were investigated. GO/SiO 2 -MIP was examined by scanning electron microscopy (SEM) and Fourier transform-infrared spectroscopy. The recovery and the intraday and interday relative standard deviations for RB ranged from 83.7% to 88.4% and 2.5% to 4.0% and the enrichment factors were higher than 110-fold. The chip-based array columns effectively eliminated impurities in chili powder, indicating that the chip-based GO/SiO 2 -MISPE method was reliable for RB detection in food samples using high-performance liquid chromatography. Accordingly, this method has direct applications for monitoring potentially harmful dyes in processed food.

A simple microdroplet chip consisting of silica nanochannel-assisted electrode and paper cover for highly sensitive electrochemiluminescent detection of drugs in human serum

Yi Xiao,Linru Xu,Ping Li,Xiao-Chong Tang,Lian-Wen Qi

This paper describes a simple and miniaturized microdroplet chip (μchip) that is constructed with a silica nanochannel (SNC)-assisted electrode array and a hydrophobic paper cover (SNC&P-μchip). Vertically aligned SNCs with uniform pore size of 2–3 nm in diameter and negatively charged surface can significantly accelerate the mass transport of the positively charged tris(2,2'-bipyridyl) ruthenium (II), resulting in a remarkably enhanced electrochemiluminescence (ECL) signal. The SNC-assisted electrode array was coupled to a low cost paper cover to achieve simultaneous detection of six samples in 1 min. The feasibility and universality of the SNC&P-μchip was evaluated by detecting a series of alkaloidal drugs both in buffers and in human serum. The performance of the SNC&P-μchip was fully validated with respect to linearity (0.9999 >  R  > 0.9939), sensitivity (limits of detection from 1.799 nM to 11.43 nM), and accuracy (recovery rate between 94.38% and 109.12%). The facile and economic SNC&P-μchip shows promising potential for rapid drug detection in complex biofluids.

Real-time detection of mercury ions in environmental water conditions by using a differential phase SPR system equipped with a repeatable SPR chip

Yansheng Liu,Haoteng Wang,Guofu Wang,Xiaobo Jia,Jin Zhou,Hongqi Li,Haixin Chang,Zhicheng Jin,Kang Li,Zhaoxu Li

Contamination with heavy metal ions is one of the most widespread and harmful sources of environmental pollution. In this work, we developed a rapid, reproducible, and highly sensitive biosensor based on the surface plasmon resonance (SPR) effect to detect mercury ions (Hg 2+ ) in DI water, tap water, and rainwater. A homemade differential phase SPR biosensor was designed and constructed, with phase shifts in the SPR interference spectrum extracted using a windowed Fourier transform (WFT) method. Custom software was developed to extract the phase shift values of the SPR system in real-time. The sensing surface of the SPR system is an Au film functionalized with thymine-1-acetic acid (T-COOH), where the thymine group specifically binds to Hg 2+ , forming a stable T-Hg 2+ -T structure. By integrating a microfluidic chip with the sensing surface, the SPR system achieves a detection limit for Hg 2+ of 8.59 × 10 −4  mg/L in DI water, 7.26 × 10 −4  mg/L in tap water, and 8.82 × 10 −4  mg/L in rainwater, with excellent specificity compared to ions like Ca 2+ , Cd 2+ , Co 2+ , Cu 2+ , Fe 2+ , Mg 2+ , and Pb 2+ . Additionally, L-cysteine is used for the regeneration of the SPR sensor, which shows no significant loss of sensitivity after 40 cycles. Therefore, the differential phase SPR biosensor holds great promise for future applications in environmental monitoring, drinking water safety, and healthcare.

Robust and efficient separation of white blood cells from blood using a microfluidic chip with a pair of linearly tapered crossflow filter arrays

Huang Yuanding,Chen Ping,Niu Meng,Peng Weng Kung

Clinical and immunological assays of white blood cells (WBCs) in human peripheral blood are of significance for disease diagnosis and immunological studies. However, separating WBCs from blood with high recovery and high purity remains challenging. In this study, by incorporating a pair of linearly tapered filter arrays, a crossflow filtration-based microfluidic chip was designed and fabricated for separation of WBCs from blood. The implementation of the linearly tapered filter arrays not only ensures a minimal and consistent flow through each sieve, but also achieves a high filtration ratio (~ 19). The validity and robustness of this straightforward design were substantiated through theoretical analysis, simulations, and model microparticle tests. The microfluidic chip achieved an almost perfect (> 99.2%) recovery and a ~ 20-fold enrichment of the targeted 8  μ m particles (as surrogates for WBCs) from undesired 2  μ m particles (as substitutes for red blood cells, RBCs) at flow rates ranging from 50 to 200 μ L/min, irrespective of the filter array length and particle concentration. When applied to a twenty times diluted blood sample, the chip achieved a 96.6% recovery and 19.7-fold enrichment of WBCs, as well as a 95.0% removal of RBCs, at the optimal flow rate of 100 μ L/min. With its simple design, cost-effectiveness, high recovery, substantial enrichment ratio, and considerable throughput, this chip offers an alternative solution that is potentially applicable to scenarios involving the separation of WBCs as well as other particles/cells. A highly sensitive Au@Pd NRs SERS microarray chip based on electrophoretic deposition for detection of GSH in the serum of colorectal cancer patients

Chenxi Xie,Dong Zhang,Yijiang Wu,Yuan He,Yiman Ge,Bin Deng

In this study, a highly sensitive surface-enhanced Raman scattering (SERS) microarray chip was developed for the detection of reduced glutathione (GSH) in the serum of colorectal cancer (CRC) patients. Gold-palladium nanorods (Au@Pd NRs) with peroxidase-like activity were synthesized and assembled into holes on the polydimethylsiloxane (PDMS)-indium tin oxide (ITO) glass substrate via electrophoretic deposition (EPD). The Au@Pd NRs catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to oxidized TMB (oxTMB) in the presence of hydrogen peroxide (H 2 O 2 ), producing a characteristic SERS peak at 1605 cm −1 . Prior to detection, a thiolated GSH aptamer was immobilized on the microarray chip to specifically capture GSH, effectively avoiding interference from other reducing substances in the complex serum matrix. The captured GSH then reduces oxTMB back to TMB, causing signal attenuation proportional to GSH concentration. The limit of detection (LOD) reached 93 pmol L −1 , with a relative standard deviation (RSD) of 6.36% for reproducibility. Clinical serum analysis of 30 healthy individuals and 30 CRC patients demonstrated results consistent with the gold standard testing method of high-performance liquid chromatography (HPLC) (relative error < 8%), confirming the potential of this approach for the effective detection of GSH and early CRC diagnosis. In this study, a highly sensitive surface-enhanced Raman scattering (SERS) microarray chip was developed for the detection of reduced glutathione (GSH) in the serum of colorectal cancer (CRC) patients.

A Bioinspired Three-Dimensional High-Curvature Nano-Interface Integrated Microfluidic Chip for Small Extracellular Vesicles Enrichment and Machine Learning-Assisted Prostate Cancer Precision Diagnosis

Le Wang,Yizhong Liang,Manan Sulaiman,Jiaqi Du,Ming Jiang,Zhihua Wang,Xu Yu,Li Xu

The efficient and unbiased isolation of small extracellular vesicles (sEVs) from complex biological fluids remains a major obstacle for clinical diagnostics. Here, we report a bioinspired microfluidic chip that integrates a three-dimensional high-curvature TiO 2 nano-interface (3D Hic-TiO 2 ) with a biotin-modified artificial insertion peptide (BAIP) modification for rapid enrichment of sEV. Bowl-shaped TiO 2 nanospheres fabricated via electrospray provide topological nanotraps that match the size and curvature of sEVs, enabling efficient size-selective capture. Coupled with BAIP-mediated membrane affinity and herringbone-induced chaotic mixing, the BAIP-TiO 2 -Chip achieved >90% capture efficiency within 5 min. Redox-responsive BAIP variants enabled mild release of intact sEVs for downstream analysis. When the BAIP-TiO 2 -Chip was applied to plasma samples from clinical prostate cancer (PCa) patients, mass spectrometry-based proteomic profiling revealed 110 differentially expressed sEV-associated proteins, including candidates involved in immune regulation and cell adhesion. In parallel, simultaneous quantification of PSA and PSMA mRNAs in sEVs could be achieved. Assisted by machine learning, a boosted decision tree model achieved 80% diagnostic accuracy in distinguishing PCa from benign conditions and healthy donors. This work presents a versatile platform for sEV isolation, enabling both transcriptomic mRNA analysis and proteomic profiling, and provides new molecular insights into PCa for improved early diagnosis.

Multi-chamber petaloid root-growth chip for the non-destructive study of the development and physiology of the fibrous root system of Oryza sativa

Hui Hui Chai,Feng Chen,Shu Jie Zhang,Ya Dan Li,Zhi Song Lu,Yue Jun Kang,Ling Yu

The root system of plants is a major component of their bodies in terms of both function and bulk. The investigation of root system development is greatly assisted by microfluidic devices, which improve the spatial and temporal resolution of observations without destroying tissue. In the present study, a multi-chamber petaloid root-growth chip was developed for studying the development and physiology of root systems that have thin branching structures (i.e., fibrous root systems). The petaloid root-growth chip includes a central seed germination chamber and five root-growth chambers for observing the development of fibrous roots. The proposed device was applied for investigating the root system development of Oryza sativa. The phenotype and growth kinetics of O. sativa root systems grown in the proposed device were compared with those obtained during growth in a conventional conical flask with agar-based medium, and the results indicated that cultivation in the miniaturized device did not delay root system growth in the early stage (≤2 weeks). In addition, the transparent device enabled the non-destructive observation of the developmental and microstructural characteristics of the roots, such as the root caps, root border cells, and root hairs. Moreover, the ability to control the microenvironment in each of the five root-growth chambers individually facilitated the investigation of specific adaptations in the fibrous root growth of single O. sativa seedlings to different drought stresses. Accordingly, five polyethylene glycol (PEG)6000-induced drought stress conditions were established in the five root-growth chambers to investigate the root development of a single O. sativa seedling in the central germination chamber. In situ observations demonstrated that the different PEG6000-induced conditions affected the root growth responses and root microstructural adaptations of the single seedlings in each root-growth chamber. Therefore, the petaloid root-growth microfluidic chip can eliminate the effects of variations in different plant seeds to reveal the responses of plants to different environmental conditions more objectively while concurrently allowing for non-destructive observations at very high spatial and temporal resolution.

Facile fabrication of thermal stable Eu(HCOO)3 red-emitting crystals with high color purity for near-ultraviolet chip triggered white light-emitting diodes

Mingzhou Zhao,Qiong Fu,Peng Du,Yafei Hou,Laihui Luo,Weiping Li

The thermal stable Eu(HCOO) 3 red-emitting crystals were synthesized by a facile hydrothermal route. Both excitation and emission spectra revealed that the prepared crystals can be efficiently pumped by the near-ultraviolet (NUV) light and it can emit brightness red light with admirable color purity of 90.4%. With the help of Judd-Ofelt theory, the optical transition intensity parameters of Eu 3+ ions were evaluated so as to explore its local symmetry performance in the Eu(HCOO) 3 host lattices . The thermal quenching behaviors of the studied samples were investigated through the temperature-dependent emission spectra . Ultimately, a white light-emitting diode (WLED) lamp with good color rendering index (78.2) and correlated color temperature (6386 K) was packaged through coating the tricolor phosphors (BaMgAl 10 O 7 :Eu 2+ blue-emitting phosphor, (Ba,Sr) 2 SiO 4 :Eu 2+ green-emitting phosphor and resultant red-emitting crystals) onto the surface of a NUV chip. These results disclosed the high brightness Eu(HCOO) 3 red-emitting crystals were suitable for NUV chip triggered WLED applications.

Integrated UHPLC-Q-TOF/MS and Liver-on-a-Chip Evaluation of Chemical Composition Changes and Hepatotoxicity Differences in Yaomu Before and After Fermentation

Kexin Ma,Lijun An,Guo Feng,Wei Li,Tingting Liu,Jinxin Hou,Ping Wang,Yibao Jin,Bing Wang,Xie-An Yu

Background: Huafeng Dan (HFD) is a traditional famous medicine from Guizhou Province, commonly used for the treatment of stroke-induced hemiplegia and epilepsy. Yaomu is a key component and serves as the sovereign herb in the formula. Most of the components of Yaomu are toxic Chinese herbal medicines. Traditional fermentation processing methods are required to reduce its toxicity. Purpose: Current studies have not yet systematically analyzed the chemical constituents before and after fermentation. Meanwhile, there is a lack of safety evaluation before and after the fermentation of Yaomu, which can provide a basis for safe clinical medication. Method: Chemical constituents of Yaomu before and after processing were analyzed using UHPLC-Q/TOF-MS to compare compositional changes induced by fermentation. To further screen potential toxic components, representative compounds were selected from these differential compounds based on statistical indicators (such as VIP value), low cost and easy availability, as well as criteria from the literature, and the content changes before and after fermentation were investigated. In vitro toxicity was evaluated using a microfluidic liver organ-on-a-chip model to assess the toxic effects of Yaomu extracts before and after fermentation. Results: Studies have shown that in both positive and negative ionization modes, a total of 361 compounds were annotated in unfermented Yaomu. After fermentation, a total of 350 compounds were annotated. Multivariate statistical analysis revealed significant differences in the chemical composition of Yaomu before and after fermentation. Quantitative analysis demonstrated that the levels of diester-type diterpenoid alkaloids were significantly reduced after fermentation, accompanied by concurrent decreases in lysophosphatidylcholine (LPC) species, compared with unfermented Yaomu. In contrast, the concentrations of amino alcohol-type diterpenoid alkaloids were significantly increased. The microfluidic liver organ-on-a-chip results demonstrated that the post-fermentation extract caused significantly attenuated impairment of hepatocellular function and viability. The in vitro toxicity findings showed good concordance.
