Alzheimer’s disease (AD) is a progressive neurodegenerative disorder characterized primarily by deterioration in memory, cognition, and learning ability. Its etiology is complex and influenced by multiple factors, including genetics and environment. With advancing research into mitochondrial function and mechanisms, impaired mitophagy has been proposed as a significant mechanism contributing to AD. The ApoE ε4 allele, a high-risk genetic factor for AD, may play a key role in disease pathogenesis by inducing mitophagy dysfunction and apoptosis. From the perspective of APOE gene polymorphisms, this study investigates abnormal changes in mitochondrial function and autophagy in humanized APOE4 mice primary astrocytes under oxidative stress, as well as the regulatory effect of curcumin (Cur) on mitophagy and oxidative stress-induced apoptosis, thereby exploring its potential to ameliorate AD through targeting mitophagy. Mitochondrial function analysis revealed that APOE4 expression reduced the antioxidant capacity and respiratory function of primary astrocytes, leading to mitochondrial membrane damage, intracellular reactive oxygen species (ROS) accumulation, and decreased ATP production. Curcumin effectively protected mitochondrial integrity, reduced the number of damaged mitochondria, improved overall mitochondrial function, and helped maintain mitochondrial homeostasis involving in PINK1/Parkin pathway. Regarding autophagy and apoptosis, curcumin was shown to restore autophagic flux, mitigate autophagy disruption caused by oxidative stress, and reverse early-stage apoptosis. Graphical

Light scattering remains a critical barrier in digital light processing (DLP)-based ceramic additive manufacturing, particularly for achieving high precision, fine resolution, and complex geometries. In this study, we present a novel light scattering mitigation strategy by incorporating a multifunctional reactive additive that combines physical light absorption near 405 nm and chemical free radical scavenging. Curcumin effectively suppresses undesired photopolymerization in non-target regions by neutralizing scattered-light-generated radicals, enabling the fabrication of intricate structures with dimensional fidelity approaching the optical resolution limit (∼2.6 pixels). The strategy operates effectively at low concentrations (0.01 wt%) without requiring process parameter tuning, offering a broad process window and high reproducibility. Furthermore, the strategy is generalizable across diverse ceramic systems, including light-colored ceramics (Al2O3, ZrO2, and SiO2), and dark-colored ceramics (SiC) that are particularly susceptible to scattering. The geometric error of printed SiC components remains below 10% even at high exposure doses of 80 and 100 mJ·cm−2, effectively addressing the long-standing trade-off between curing depth and lateral resolution. This work provides a simple, scalable, and versatile approach for improving resolution in DLP ceramic printing, advancing the reliable production of fine-scale (<50 μm) ceramic architectures for applications in biomedicine, microelectronics, and precision engineering.

To reduce food waste and mitigate health risks from accidentally consuming spoiled food, freshness-indicating technologies are increasingly demanded. However, conventional colorimetric-based freshness-indicating packaging is limited by instability, subtle color changes, and complex production processes. This study presents a curcumin-based ink suitable for eco-friendly polylactic acid (PLA) food packaging films enabling real-time shrimp freshness monitoring via integrated intelligent packaging. The ink comprised curcumin as the indicator, ethyl cellulose (EC) and polyvinyl butyral (PVB) as binders, and polyethylene glycol 400 (PEG 400) to regulate permeability. Excellent printability was demonstrated by fineness, initial dryness and fluidity tests. It also demonstrated good thixotropic, viscosity, and flow curve properties. Printing minimally affected the PLA films’ mechanical and barrier properties. The indicator label showed high sensitivity, rapid response, and excellent reversibility to ammonia vapor. Practical application in monitoring shrimp spoilage at 25 °C and 4 °C revealed a strong correlation between the distinct color transition of the label and the increase in total volatile basic nitrogen (TVB-N) content and pH value, providing a reliable visual warning before obvious spoilage signs appeared. This work provides a viable integrated indicator packaging strategy for developing intelligent packaging, offering significant potential to reduce food waste and enhance supply chain transparency for perishable goods.

In this study, five levulinic acid-based deep eutectic solvents (DESs) were evaluated for the ultrasound-assisted extraction of curcuminoids fromCurcuma longa. The relationship between DES composition, intermolecular interactions, and extraction performance was investigated by combining extraction experiments with spectroscopic characterization and quantum chemical calculations. The levulinic acid–ethylene glycol system at a 1:2 molar ratio exhibited the best performance, achieving an extraction yield of 99.9 mg·g−1. FTIR and1H NMR analyses characterized the interactions within the DES, while SEM observations indicated structural disruption of the turmeric matrix after ultrasound treatment. Molecular analyses suggested that hydrogen bonding and van der Waals interactions contributed to the association between the DES and curcuminoids. These results provide molecular insights into the extraction process and a basis for designing greener solvent systems for curcuminoid extraction.

Petroleum-based packaging materials raise serious environmental and food safety concerns, driving the development of sustainable smart packaging derived from natural polymers. In this study, natural pigment-zein-chondroitin sulfate-epigallocatechin gallate-ι-carrageenan (NZCEC) nanoparticles were fabricated and incorporated into κ-carrageenan-based films to enhance their functional properties. The effects of NZCEC addition on the films' physicochemical stability, intermolecular interactions, antioxidant activity, and responsiveness to ammonia and acetic acid vapors were systematically evaluated, along with their application in preserving mandarin fish ( Siniperca chuatsi ). Scanning electron microscopy revealed good compatibility between κ-carrageenan and moderately loaded NZCEC nanoparticles. Incorporation of NZCEC significantly improved the film's optical properties (opacity increased from 4.10 to 23.32 A mm −1 ) and reduced its water vapor permeability (from 8.23 × 10 −9 to 6.93 × 10 −9 g m −1 s −1 Pa −1 ). Moreover, the antioxidant activity was markedly enhanced (from 1.75 % to 77.65 %), along with notable responsiveness to ammonia and acetic acid. Storage trials with mandarin fish demonstrated that the smart film effectively delayed fish spoilage. Additionally, the film displayed distinct chromatic variations in response to freshness, allowing continuous assessment of food quality. These results establish NZCEC nanoparticle-reinforced κ-carrageenan films as a promising intelligent packaging material with improved stability, responsiveness, and food preservation performance.

Multifunctional hydrogel dressings, with their robust antibacterial and anti-inflammatory properties, represent a great potential in biomaterials for wound care. Injectable and self-healing hydrogels that mimic the natural extracellular matrix of tissues offer a promising strategy for effective wound repair. This study designs a hydrogel based on the formation of a dynamic cross-linking network within the hydrogel matrix through the Schiff base reaction between the synthesized polymers L-hydroxyproline-conjugated chitosan (LHCS) and oxidized pullulan (OPL). The fabricated LP hydrogels preserve the porous interconnected microstructure, excellent self-healing capacity, and injectability, suitable swelling capability, robust mechanical strength, and outstanding tissue adhesion. Moreover, curcumin was encapsulated in Polyether F127 (Pluronic F127) micelles to obtain curcumin loaded PF127 (Cur-PF127) micelles and then incorporated into LP hydrogel to fabricate the composite hydrogels. The composite hydrogels exhibit effective antibacterial properties and antioxidant abilities, admired hemostatic effect and hemocompatibility, excellent cytocompatibility and cell pro-migration ability. In addition, in vivo evaluation confirmed that curcumin loaded hydrogels could facilitate the regeneration of epithelial and hair follicles, increase the collagen production, alleviate the inflammatory response via the downregulation of the tumor necrosis factor-α (TNF-α), and reduce the fibrosis, thus promote the wound repair process. This work offers a promising facile approach for the development of the composite hydrogel as wound dressing in wound healing applications.

The current study developed ethylcellulose (EC) based oleogels with the addition of a surface active ingredient (sorbitan monopalmitate, SP), in order to increase the active loading of curcumin by reducing lipid oxidation , improving curcumin solubility and chemical stability. With the increase in SP content, EC oleogels had more compact gel networks with evenly distributed smaller pores. Rheological analysis revealed that the gels had shear-thinning behavior, and higher concentration of SP contributed to the systems with higher viscosity. The inclusion of SP also worked to reinforce gel strength as determined by frequency sweep, creep recovery and textural analyses. EC oleogels with higher content of SP were capable to hold more liquid oil during centrifugation , and the T 2 relaxation time was much lower as determined by NMR. Peroxide value of the oleogels was significantly lower in the systems with SP, and a SP content of 4% or 6% was effective in inhibiting lipid oxidation during storage. When curcumin was incorporated within the gel networks, its effective concentration was more retained with the addition of SP, as no curcumin crystals were detected by DSC during a 9-day storage test, and curcumin had much higher retention when exposed to UV light for 8 h.

Halloysite nanotubes (HNTs) have a unique tubular structure in nanoscale, and have shown potential as novel carriers for various drugs. Coating the nanotubes with a hydrophilic polymer shell can significantly decrease the toxicity and provide colloidal stability during blood circulation. Here, we synthesized chitosan grafted HNTs (HNTs-g-CS) and investigated their potential as a nano-formulation for the anticancer drug curcumin. The structure and properties of HNTs-g-CS were characterized using water contact angle, zeta-potential, Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), and transmission electron microscopy (TEM) techniques. HNTs-g-CS exhibit a maximum 90.8% entrapment efficiency and 3.4% loading capacity of curcumin, which are higher than those of raw HNTs. HNTs-g-CS also show no obvious hemolytic phenomenon and good stability in serum. The cumulative release ratio of curcumin from HNTs-g-CS/curcumin at cell lysate after 48 hours is 84.2%. The curcumin loaded HNTs-g-CS show specific toxicity to various cancer cell lines, including HepG2, MCF-7, SV-HUC-1, EJ, Caski and HeLa, and demonstrate an inhibition concentration of IC50 at 5.3–192 μM as assessed by cytotoxicity studies. The anticancer activity of this nanoformulation is extremely high in EJ cells compared with the other cancer cell lines. The cell uptake of HNTs-g-CS is confirmed by fluorescence microscopy. Flow cytometric analysis of curcumin loaded HNTs-g-CS shows that curcumin loaded HNTs-g-CS increase apoptosis on EJ cells. The content of ROS created by HNTs-g-CS/curcumin is more than that of free curcumin. All these results suggest that HNTs-g-CS are potential nanovehicles for anticancer drug delivery in cancer therapy.

As a class of highly dynamic organelles, lipid droplets (LDs) are involved in numerous physiological functions, and the changes in polarity of LDs are closely related to a variety of diseases. In this work, we developed two polarity-sensitive fluorescent probes (CC-CH and CC-Cl) based on curcumin analogs. CC-CH and CC-Cl with a donor–acceptor–donor (D–A–D) structure exhibited the property of intramolecular charge transfer (ICT); thus, their fluorescence emissions were significantly attenuated with increasing ambient polarity. Cell experiments indicated that CC-CH and CC-Cl showed excellent photostability, a low cytotoxicity, and a superior targeting ability regarding LDs. After treatment with oleic acid (OA) and methyl-β-cyclodextrin (M-β-CD), the polarity changes of LDs in living cells could be visualized by using CC-CH and CC-Cl. In addition, CC-CH and CC-Cl could monitor polarity changes of LDs in different pathological processes, including inflammatory responses, nutrient deprivation, and H2O2-induced oxidative stress. Therefore, CC-CH and CC-Cl are promising potential fluorescent probes for tracking intracellular LD polarity changes.Keywords:polarity-sensitive;lipid droplet targeted;fluorescent probes;cell imaging

A sensitive voltammetric sensor, a graphene-modified glassy carbon electrode (GR/GCE), for the detection of trace amounts of curcumin is proposed. The GR/GCE was characterized by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The electrochemical behavior of curcumin at the GR/GCE was investigated by CV and liner sweep voltammetry (LSV). The reaction mechanisms of curcumin on the GR/GCE were also investigated and discussed systematically. In addition, the currents measured by LSV presented a good linear relationship with curcumin concentrations in the range of 5.0 × 10−8 to 3.0 × 10−6 mol L−1, with a low detection limit of 3.0 × 10−8 mol L−1. The method was also applied successfully to detect curcumin in a Curcuma longa L. sample with high selectivity and accuracy, as well as good recovery.

Background Using natural polysaccharides from Traditional Chinese Medicine as nanodrug delivery systems have considerable potential for tumor diagnostics and therapeutics. Purpose On the basis of targeted therapy and combining the advantages of natural polysaccharides (angelica polysaccharide, APS) and natural Chinese medicine (curcumin, Cur) to design functionalized nanoparticles to improve the therapeutic through cell membrane encapsulation and immunotherapy . Study Design and Methods Cur-loaded, glycyrrhetic acid (GA)-APS-disulfide bond (DTA)-Cur nanomicelle (GACS-Cur), which were prepared by the dialysis method. GACS-Cur was encapsulated with the membranes from red blood cells (RBCm) termed GACS-Cur@RBCm, which were prepared by the principle of extrusion using a miniature extruder. The developed formulations were subjected to various in vitro and in vivo evaluation tests. Results The resulting APS nanocarriers supported a favorable drug-loading capacity, biocompatibility , and enhanced synergistic anti-hepatoma effects both in vitro and in vivo . After administration in mice, in vivo imaging results showed that the GACS-Cur and RBCm-coated groups had an obvious stronger tumor tissue targeting ability than the control treatment groups. Additionally, the immunomodulatory effect increased IL-12, TNF-α and IFN-γ expression and CD8+ T cell infiltration (1.9-fold) than that of the saline group. Notably, in comparison with hyaluronic acid (HA) nanocarriers , APS nanocarriers possess higher anti-hepatoma efficiency and targeting capabilities and, thus, should be further studied for a wide range of anti-cancer applications. Conclusion Our data demonstrated that APS nanocarriers encapsulated with erythrocyte membrane mighty be a promising clinical method in the development of efficacy, safety and targeting of liver cancer therapy.

New nanocomplexes containing curcumin-loaded casein micelles and surface-coated pectin via electrostatic interaction were fabricated. Curcumin-loaded nanocomplexes prepared at pH 4 showed the most excellent performance than that at pH 3 and pH 2, which had an average size of 266.4 nm (polydispersity index = 0.193) and loading capacity of 93%. The morphology of nanocomplexes exhibited as monodisperse spherical nanoparticles observed by SEM and TEM. The experiment of anti-oxidant activity demonstrated that nanocomplexes tremendously kept the bioactivity of curcumin. Moreover, in vitro release behaviors of curcumin from casein-pectin nanocomplexes indicated that the surface coating layer of pectin obviously delayed the release of curcumin in simulated gastric fluid, and the controlled release ability in simulated intestinal fluid was greatly enhanced. The casein-pectin nanocomplexes exhibited as a promising oral delivery vehicle of bioactive ingredients in food and pharmacy. Graphical abstract
