New insight into enzymatic hydrolysis of peptides with site-specific amino acid d-isomerization

Liang Yan,Yongqi Ke,Yuhe Kan,Dao Lin,Jingkui Yang,Yujian He,Li Wu

The isomerization of l -amino acids in peptides and proteins into d -configuration under physiological conditions would affect the physiological dysfunction and caused protein conformational diseases. The presence of d -amino acids might change the higher-order structure of proteins and triggered abnormal aggregation. In order to better understand this phenomenon and promote degradation, we systematically studied the enzymatic hydrolysis of a series of peptides obtained by replacing l -amino acids in different positions of template peptide KYNETWRSED with d -amino acids under the action of Protease K. The results showed that, compared with normal peptide, isomerization of different amino acids had different effects on the anti-enzymatic hydrolysis of the peptides, especially d -tryptophan at position 6, which significantly inhibited enzymatic hydrolysis. The analysis of the peptide cleavage site revealed that the efficiency of enzymatic hydrolysis mainly depended on the isomerization of the amino acids at a specific site of the peptide cleavage. Further studies showed that the enzymatic hydrolysis of substrates could be facilitated by optimized reaction conditions such as temperature, pH, addition of metal ions, and change of buffer. In this way the accumulation of disease-associated d -amino acid containing polypeptides/proteins could be prevented.

Phlorizin alleviates cholinergic memory impairment and regulates gut microbiota in d-galactose induced mice

Yan-Ling Su,Dong Liu,Yao-Jie Liu,Yang-Lin Ji,Gui-Shan Liu,Ji-Li-Te Wang,Biao Wang,Hao Wang

We explored the effect of phlorizin against cholinergic memory impairment and dysbacteriosis in D-galactose induced ICR mice . The control (CON) group, D-galactose model (DGM) group, and three groups (DG-PL, DG-PM, DG-PH) treated with phlorizin at 0.01%, 0.02%, and 0.04% (w/w) in diets were raised for 12 weeks. Supplementing with phlorizin reversed the loss of organ coefficient and body weight caused by D-galactose. The functional abilities of phlorizin on hippocampal-dependent spatial learning and memory, anti-oxidation, anti-inflammation were also observed. Meanwhile, phlorizin intervention upregulated the gene expression of Nrf2, GSH-PX, SOD1, decreased the gene expression of NF-κB, TLR-4, TNF-α, and IL-1β in the hippocampus , while enhanced the gene expression of JAM-A, Mucin2, Occludin in the caecum. Furthermore, a neurotransmitter of acetylcholine (ACh) was enhanced, while acetylcholinesterase (AChE) activity was inhibited by phlorizin administration. Moreover, phlorizin administration increased short-chain fatty acids (SCFAs) content, and reduced lipopolysaccharides (LPS) levels, which may relate to the rebuilding of gut microbiota homeostasis . Treatment with phlorizin may be an effective intervention for alleviating cognitive decline and gut microbiota dysbiosis.

Antioxidant activity of selenium-enriched Chrysomyia megacephala (Fabricius) larvae powder and its impact on intestinal microflora in D-galactose induced aging mice

Xie Dandan,Jiang Liqin,Lin Yao,Liu Zhenwei

Background The purpose of this study was to assess the antioxidative activity of selenium-enriched Chrysomyia Megacephala (Fabricius) ( C. megacephala ) larvae powder (SCML) and its impact on the diversity and structure of intestinal microflora in a mouse model of D-galactose (D-gal)-induced oxidative damage. Methods Sixty male ICR mice were equally randomized to a normal control (NC) group, a model group, a positive group, a low-dose SCML (L-SCML) group, a mid-dose SCML (M-SCML) group, and a high-dose SCML (H-SCML) group. Animals in NC and model groups received water, animals in the positive group received 40 mg/Kg vitamin E (VE), and those in the three SCML groups received SCML which include 300, 1000 and 3000 μg/Kg selenium (Se) respectively. An oxidative damage model induced by subcutaneous injection of D-gal for 6 weeks via the neck was established. Serum oxidative stress levels and tissue appearance were evaluated. Tissues oxidative stress levels were detected by commercially available kit. Nuclear erythroid 2-related factor (Nrf2) and gut microbiota were determined by western blot and high throughput sequencing 16S rRNA gene respectively. Results An oxidative damage model was established successfully as represented by a significant elevation of malondialdehyde (MDA) and protein carbonylation, and inhibition of the antioxidants including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC) and glutathione (GSH). It was found that oxidative damage and histological alterations were attenuated, the expression of Kelch-like ECH-associated protein (Keap1) was decreased, and the expression of Nrf2 and hemeoxygenase-1 (HO-1) was increased after SCML treatment. In addition, significant changes were observed in the gut microbiota, including Proteobacteria and the ratio of Bacteroidetes to Firmicutes at the phylum level, as well as Helicobacter , Clostridium and Lactobacillus at the genus level. Conclusion SCML exerted an antioxidative effect in vivo, probably by increasing the antioxidant activity and reducing the production of oxidation products via the Nrf2 signaling pathway. SCML could also redress the intestinal flora imbalance induced by oxidative stress. All these findings suggest that SCML could serve as a functional food and natural drug additive to protect the human body against oxidative damage.

Radiation-induced atrial fibrillation is driven by cellular senescence via the GATA4 – NF-κB pathway and rescued by D/Q senolytic therapy

Chao Zhang,Huai Lan,Liming Yu,Jikai Zhao,Tao Huang,Boxuan Sun,Shan Meng,Jinfeng Duan,Jing Liu,Zijun Zhou,Zhishang Wang,Jiahui Li,Xinyi Huang,Huishan Wang

First to report that radiation-induced atrial fibrillation is driven by cellular senescence. • The GATA4-NF-κB-SASP axis is the key signaling pathway mediating this process. • Senolytic therapy with D/Q (Dasatinib+Quercetin) rescues atrial remodeling and reduces AF susceptibility. • Targeting senescence with senolytics offers a promising strategy to prevent radiation-induced AF. Radiation-induced heart disease is a well-recognized complication of thoracic radiotherapy, with atrial fibrillation (AF) being a particularly notable sequela. The mechanisms by which ionizing radiation (IR) heightens susceptibility to AF remain inadequately understood. This study seeks to elucidate the role of cellular senescence and the GATA4-NF-κB signaling pathway in radiation-induced atrial remodeling and the pathogenesis of AF. An in vivo mouse model was developed through localized cardiac irradiation using 20 Gy X-rays. The irradiated mice demonstrated dose-dependent atrial structural and electrophysiological remodeling. Transcriptomic analysis revealed significant enrichment in DNA damage response (DDR), NF-κB signaling, and aging-related pathways. IR exposure induced substantial DNA damage, activated ATM/ATR pathways, and enhanced autophagic flux. These responses culminated in the accumulation of GATA4 and phosphorylation of NF-κB, which drove the expression of the senescence-associated secretory phenotype (SASP), coinciding with increased senescence-associated β-galactosidase (SA-β-gal) activity. In vitro , these crucial findings were recapitulated using irradiated HL-1 atrial cardiomyocytes, whereas the knockdown of GATA4 effectively suppressed both cellular senescence and SASP expression. Notably, treatment with the senolytic combination of dasatinib and quercetin (D/Q) alleviated DNA damage, inhibited excessive autophagy, and suppressed the GATA4-NF-κB pathway in irradiated mice, thereby reducing SASP levels. These improvements resulted in the reversal of atrial structural and electrophysiological remodeling and a marked reduced susceptibility to AF. Overall, our findings demonstrate that IR promotes atrial remodeling by inducing DNA damage-mediated cellular senescence via the GATA4-NF-κB-SASP axis. Targeting this pathway with senolytics such as D/Q thus identifies a promising therapeutic strategy for preventing radiation-induced atrial fibrillation.

β-1,3-d-glucan particles-based “nest” protected co-loaded Rhein and Emodin regulates microbiota and intestinal immunity for ulcerative colitis treatment

Yanli Wang,Siwei Zhong,Ke Yang,Ruifeng Luo,Linxin Dai,Wenzhen Zhong,Yan Ye,Chaomei Fu,Dasheng Lin,Nan Li,Jianping Chen,Chuan Zheng,Shu Fu,Fei Gao

Herein, a β-1,3- D -glucan based yeast cell wall loaded with co-loaded nanoparticles of Rhein (RH) and Emodin (EMO), was developed for the combined treatment of ulcerative colitis (UC) by modulating gut microbiota and the Th17/Treg cell balance. This was achieved through an oral “nano-in-micro” advanced drug delivery system. Specifically, RH was grafted onto the HA chain via disulfide bonds to synthesize a reduction-sensitive carrier material and then used to encapsulate EMO to form nanoparticles with a specific drug ratio (denoted as HA-RH/EMO NPs). As anticipated, HA-RH/EMO NPs were encased within the “nests”-yeast cell wall microparticles (YPs), efficiently reach the colon and then released gradually, this occurs mainly due to the degradation of β-1,3- D -glucan by β-glucanase. Additionally, HA-RH/EMO NPs demonstrated a significant reduction-sensitive effect in GSH stimulation evaluations and a remarkable ability to target macrophages in in vitro cell uptake studies. Notably, HA-RH/EMO NYPs reduced inflammatory responses by inhibiting the PI3K/Akt signaling pathway. Even more crucially, the oral delivery and drug combination methods significantly enhanced the regulatory effects of HA-RH/EMO NYPs on gut microbiota and the Th17/Treg balance. Overall, this research marks the first use of YPs to encapsulate two components, RH and EMO, presenting a promising therapeutic strategy for UC.

Platycodin D inhibits proliferation, migration and induces chemosensitization through inactivation of the NF-κB and JAK2/STAT3 pathways in multiple myeloma cells

Di Wu,Wei Zhang,Ying Chen,Hong Ma,Mengchang Wang

Multiple myeloma (MM) is a malignancy characterized by the proliferation of malignant plasma cells. Platycodin D (PLD) is a triterpenoid saponin that exerts anti-tumour activity through multiple mechanisms. However, the role of PLD in MM remains unknown. Here, we investigated the effect of PLD on MM cell lines NCI-H929 and U266B1, and elucidated the underlying molecular mechanism. Cell Counting Kit-8 assay showed that the proliferation of NCI-H929 and U266B1 cells was significantly decreased after PLD treatment. Transwell assay confirmed that PLD treatment suppressed migration of NCI-H929 and U266B1 cells. Flow cytometry results indicated that the apoptotic rates of bortezomib (BTZ)-treated NCI-H929 and U266B1 cells were markedly increased after PLD treatment. Western blot analysis revealed that bcl-2 expression was decreased, while bax expression was increased in PLD-treated NCI-H929 and U266B1 cells compared with that in BTZ-treated cells. Furthermore, PLD treatment blocked the activation of nuclear factor-kappa B (NF-κB) and Janus kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3) signalling pathways in NCI-H929 cells. Taken together, these data showed that PLD inhibited proliferation and migration, and enhanced chemosensitization to BTZ through inactivation of the NF-κB and JAK2/STAT3 pathways in MM cell lines. These findings indicated that PLD might serve as a novel therapeutic agent for the treatment of MM.

Successive C1–C2 bond cleavage: the mechanism of vanadium(V)-catalyzed aerobic oxidation of D-glucose to formic acid in aqueous solution

Muge Niu,Yucui Hou,Weize Wu,Shuhang Ren,Ru Yang

Vanadium(V)-catalyzed aerobic oxidation in aqueous solution shows high selectivity in the field of C–C bond cleavage of carbohydrates for chemicals with less carbon atoms. However, the pathway of C–C bond cleavage from carbohydrates and the conversion mechanism are unclear. In this work, we studied the pathway and the mechanism of D-glucose oxidation to formic acid (FA) in NaVO3–H2SO4 aqueous solution using isotope-labeled glucoses as substrates. D-Glucose is first transformed to FA and D-arabinose via C1–C2 bond cleavage. D-Arabinose undergoes similar C1–C2 bond cleavage to form FA and the corresponding D-erythrose, which can be further degraded by C1–C2 bond cleavage. Dimerization and aldol condensation between carbohydrates can also proceed to make the reaction a much more complicated mixture. However, the fundamental reaction, C1–C2 bond cleavage, can drive all the intermediates to form the common product FA. Based on the detected intermediates, isotope-labelling experiments, the kinetic isotope effect study and kinetic analysis, this mechanism is proposed. D-Glucose first reacts with a vanadium(V) species to form a five-membered-ring complex. Then, electron transfer occurs and the C1–C2 bond weakens, followed by C1–C2 bond cleavage (with no C–H bond cleavage), to generate the H3COO˙–vanadium(IV) complex and D-arabinose. FA is generated from H3COO˙ that is oxidized by another vanadium(V) species. The reduced vanadium species is oxidized by O2 to regenerate to its oxidation state. This finding will provide a deeper insight into the process of C–C bond cleavage of carbohydrates for chemical synthesis and provide guidance for screening and synthesizing new highly-efficient catalyst systems for FA production.

A UV-curable coating constructed from bio-based phytic acid, D-sorbitol and glycine for flame retardant modification of rigid polyurethane foam

Jinke Yu,Ling Sun,Lan Ding,Yufei Cao,Xiaohui Liu,Yuanlin Ren,Yuesheng Li

When confronted with a fire, the flammability of rigid polyurethane foam (RPUF) makes it extremely easy to burn violently. In this work, a green biomass derived flame retardant coating was synthesized with phytic acid as acid source, D-sorbitol as carbon source and glycine as gas source. A simple UV curing technique was used to improve the flame retardancy of RPUF . Coated RPUF exhibited a limiting oxygen index value of up to 39.7 % and successfully met UL-94 testing criteria with V-0 rating. In comparison to control RPUF, coated RPUF showed an extended ignition time of 3 s, a remarkable reduction of approximately 60 % in the peak heat release rate, and possessed commendable smoke suppression property. The flame retardant mechanism was analyzed, unveiling the emergence of compact char residue on the surface of coated RPUF after burning, accompanied by the liberation of non-combustible gases. Therefore, these results demonstrated that cost-effective UV-curable flame retardant coating forms protective char layer on the surface of RPUF, providing efficient flame retardancy while preserving its excellent properties.

A reliable gold nanoparticle/Cu-TCPP 2D MOF/gold/D-shaped fiber sensor based on SPR and LSPR coupling for dopamine detection

Jinjuan Gao,Wen Yang,Runcheng Liu,Jingwen Feng,Yaru Li,Mingshun Jiang,Shouzhen Jiang

The optical fiber sensor, which exploits surface plasmon resonance (SPR), finds extensive application in biological detection owing to its miniaturized and real-time measurement capabilities. Nevertheless, SPR sensors alone exhibit limited sensitivity when detecting small biological molecules. This paper presents a novel gold nanoparticle (AuNPs)/two-dimensional metal–organic framework (2D MOF Cu-TCPP)/Au/D-shaped fiber SPR sensor for dopamine detection. The composite structure of AuNPs/2D MOF/Au was designed to facilitate the coupling of SPR and local surface plasmon resonance (LSPR), resulting in a substantial enhancement of the intensity of the surface electric field. A 104.61 % increase in sensitivity and a 73.87 % increase in figure of merit (FOM) were achieved by optimizing the optimal deposition concentration of AuNPs/2D MOF . The finite element method was utilized in COMSOL software to demonstrate that the coupling of SPR and LSPR can significantly enhance the sensitivity value (10,000 nm/RIU). Additionally, the sensitization mechanism of the sensor is elucidated by the energy band theory. Moreover, probe DNA was utilized as a linker to capture dopamine molecules, achieving a low limit of detection (LOD) of 1.07 ± 0.07*10 −14 M. These results demonstrated that the proposed sensor enables accurate and highly sensitive detection of dopamine molecules.

Cr(VI) removal during cotransport of nano-iron-particles combined with iron sulfides in groundwater: Effects of D. vulgaris and S. putrefaciens

Xia Liu,Ming Chen,Dengjun Wang,Feng Du,Nan Xu,Wu Sun,Zhaoxiang Han

Iron-based materials such as nanoscale zerovalent iron (nZVI) are effective candidates to in situ remediate hexachromium (Cr(VI))-contaminated groundwater . The anaerobic bacteria could influence the remediation efficiency of Cr(VI) during its cotransport with nZVI in porous media . To address this issue, the present study investigated the adsorption and reduction of Cr(VI) during its cotransport with green tea (GT) modified nZVI (nZVI@GT) and iron sulfides (FeS and FeS 2 ) in the presence of D. vulgaris or S. putrefaciens in water-saturated sand columns. Experimental results showed that the nZVI@GT preferred to heteroaggregate with FeS 2 rather than FeS, forming nZVI@GT-FeS 2 heteroaggregates. Although the presence of D. vulgaris further induced nZVI@GT-FeS 2 heteroaggregates to form larger clusters, it pronouncedly improved the dissolution of FeS and FeS 2 for more Cr(VI) reduction associated with lower Cr(VI) flux through sand. In contrast, S. putrefaciens could promote the dispersion of the heteroaggregates of nZVI@GT-FeS 2 and the homoaggregates of nZVI@GT or FeS by adsorption on the extracellular polymeric substances , leading to the improved transport of Fe-based materials for a much higher Cr(VI) immobilization in sand media. Overall, our study provides the essential perspectives into a chem-biological remediation technique through the synergistic removal of Cr(VI) by nZVI@GT and FeS in contaminated groundwater. Environmental implication The green-synthesized nano-zero-valent iron particles (nZVI@GT) using plant extracts (or iron sulfides) have been used for in situ remediation of Cr(VI) contaminated groundwater. Nevertheless, the removal of Cr(VI) (including Cr(VI) adsorption and Cr(III) generation) could be influenced by the anaerobic bacteria governing the transport of engineered nanoparticles in groundwater. This study aims to reveal the inherent mechanisms of D. vulgaris and S. putrefaciens governing the cotransport of nZVI@GT combined with FeS (or FeS 2 ) to further influence the Cr(VI) removal in simulated complex groundwater media. Our findings provides a chemical and biological synergistic remediation strategy for nZVI@GT application in Cr(VI)-contaminated groundwater.

Effective role of d-sorbitol in enhancing the oxidation stability and crosslink density of tea polyphenol-stabilized ultra-high molecular weight polyethylene

Shah Nouman Ali,Shakir Namra,Gul Rizwan M.,Xu Jia-Zhuang,Li Zhong-Ming

Ultra-high molecular weight polyethylene (UHMWPE) is a versatile polymer with various industrial applications, particularly in orthopedic implants and structural materials. However, its susceptibility to oxidation limits its utility in demanding environments. This study investigates the potential of d -sorbitol (DS), a natural polyol, in enhancing the oxidation stability and crosslink density of UHMWPE when stabilized with tea polyphenols. Tea polyphenols, such as epigallocatechin gallate (EGCG), have been previously evaluated as a promising candidate to stabilize UHMWPE against oxidation without affecting its crosslink density. However, the higher content of EGCG has adverse effect on its mechanical properties. It is hypothesized that addition of DS to EGCG can improve the oxidation and crosslink density of UHMWPE blends. For this purpose, EGCG/DS was blended with UHMWPE at low antioxidant concentrations (≤ 1 wt%), and its comprehensive performance was evaluated. Results demonstrate that the incorporation of d -sorbitol effectively retards oxidation-induced degradation in tea polyphenol-stabilized UHMWPE. The presence of d -sorbitol enhances the crosslink density within the polymer matrix, thereby improving its resistance to oxidative stress. Furthermore, the study elucidates the underlying mechanisms through which DS interacts with EGCG and UHMWPE, elucidating the formation of stable complexes that mitigate oxidative damage. The in vitro biocompatibility of the blends was also very promising for use in artificial joints.

Visible-light-induced polymerization initiated by D-A dyes: towards rational design of efficient light-harvesting photoinitiators by regulation of excited states

Yizhi Shao,Dandan Liu,Qinwei Xia,Zhendong Song,Wei Chen,Ke Sun,Changjiang Zhou,Yangyang Xu,Tao Gong

The elongation of π-conjugation could efficiently enhance the photophysical performances or photochemical reactivties of organic molecules, which adapts to the requests of both designing fluorescent materials and organic photoinitiators (PIs). However, there is no work attempted to design organic PIs with large molar extinction coefficients and long π-conjugation lengths by using hybridized local and charge-transfer (HLCT) method, which is meaningful to obtain high-performance PIs with significant fluorescence/photoluminence properties. What's more, how to overcome the obstacles derived from the unbalance between high triplet concentrations and photoinitiation abilities of organic PIs seems to be crucial, while PI's design from a view of excited state were rarely reported for rational regulation strategy up to now, we thus introduced four chromophores with D-A structures designed by HLCT method, whose triplet states were confirmed by Ultra-fast absorption spectroscopy, into photopolymerzation, where LEDs in various wavelengths were utilized as the light sources in the present work. Noticeably, some of these dyes could furnish high monomer conversions and rapid photopolymerization rates, not only under the irradiation of 405 nm LED, but also long-wavelength light source at 660 nm, which have been confirmed by the photopolymerization kinetics and 3D printing technique. Here, the spin-orbit coupling (SOC) coefficients were carried out as well, to examine their effects on photoinitiation abilities of investigated molecules, which can be regarded as the first attempt to achieving light-harvesting PIs from excited state design. Finally, the corresponding photochemical mechanisms were investigated by steady state photolysis, fluorescence quenching experiments, particularly by frontier orbit energy and distributions to examine the interactions between these dyes with co-initiator/additives in details.
