Intrauterine adhesions (IUAs), a leading cause of uterine infertility, present significant therapeutic challenges, with conventional treatments often yielding unsatisfactory reproductive outcomes. Although mesenchymal stem cell (MSC)-based therapies have demonstrated considerable promise, current biomaterial delivery systems face critical limitations, particularly in addressing the oxidative stress and inflammatory microenvironment of the damaged endometrium that impairs stem cell viability and function. Herein, we developed an injectable antioxidant hydrogel incorporating microspheres loaded with umbilical cord-derived MSCs (TPG hydrogel) for endometrial regeneration and fertility restoration. The TPG hydrogel was formed via dynamic phenylboronic ester crosslinking, endowing it with shear-thinning injectability for minimally invasive administration and potent reactive oxygen species (ROS) scavenging capacity to mitigate oxidative stress. This antioxidant microenvironment protected encapsulated MSCs from oxidative damage, thereby enhancing their therapeutic efficacy. Furthermore, the incorporation of EPLQLKM (E7) peptide-modified gelatin microspheres provided a bioactive niche that promoted cell adhesion and proliferation. Comprehensive in vitro and in vivo studies demonstrated that the TPG hydrogel exhibited dual functionality, including effective ROS scavenging and immunomodulatory effects characterized by the promotion of macrophage polarization toward the anti-inflammatory M2 phenotype. In a rat model of IUAs, the TPG hydrogel significantly enhanced endometrial regeneration and restored fertility. These findings highlight the TPG hydrogel as a promising therapeutic platform for the treatment of endometrial injury and fertility impairment associated with IUAs.

Pepino ( Solanum muricatum ) juice is a nutrient-rich beverage, yet it's limited by poor shelf stability and thermal pasteurization-induced quality loss. So, we developed a novel continuous high-pressure processing (HPP) system and evaluated its effects on the nutritional quality, flavor, and shelf stability of pepino juice. Our results demonstrated that HPP-500 showed superior color stability ( ΔE increase <25%) and achieved >90% PPO and > 88% POD inactivation ( p < 0.05), while retaining higher phenolics (4.44 ± 0.02 mg GAE/mL) and flavonoids (10.17 ± 0.54 mg RE/mL), and enhancing DPPH (10.22%) and ABTS (7.42%) radical scavenging capacity ( p < 0.05) during 28-day storage. Electronic nose and tongue assessments further confirmed markedly flavor retention under HPP-500 ( p < 0.05). HS-SPME/GC–MS and UHPLC-Q-Orbitrap-HRMS revealed a richer profile of volatile and bioactive metabolites, notably phenolic acids, terpenoids, and alkaloids. These findings highlight the industrial potential of continuous HPP in enhancing juice quality and shelf-life.

The injectable hydrogel with self-healing and strong adhesive properties was developed. • Carbon dots derived from Isatis root were synthesized and loaded into the hydrogel. • The hydrogel exhibits pH/ROS-responsive drug release behavior. • The hydrogel exhibits antibacterial activity, antioxidative capability • The hydrogel accelerates infected wound healing. Bacteria-infected wounds are very difficult to treat, largely because the pathological microenvironment involved is dynamic. The requirements for infected wound repair are multifaceted, and conventional hydrogel dressings cannot adequately meet them due to their limited functionality. In this study, we report the construction of a multifunctional, pH/ROS dual-responsive hydrogel based on dynamic Schiff base and borate ester linkages. Oxidized pullulan (OPu) was used to fabricate the hydrogel, and the bioactive polyphenol chlorogenic acid (CA) acted as the crosslinking agent. Next, we synthesized the carbon dots derived from Isatis root (IR-CDs) using the hydrothermal method and subsequently incorporated them into the hydrogel. The hydrogel exhibited excellent injectability and self-healing capability, enabling application at irregular wound sites. Under the acidic and oxidative conditions of infected wounds, the hydrogel gradually dissociated, releasing IR-CDs and CA in a controlled manner. IR-CDs exhibited potent antibacterial activity, while CA and IR-CDs in combination efficiently scavenged excessive reactive oxygen and nitrogen species, therefore promoting tissue regeneration and reducing inflammation. Both in vitro and in vivo evaluations confirmed that the hydrogel was highly biocompatible and provided anti-infective, anti-oxidative, and anti-inflammatory effects during the early stages of healing, followed by an increase in angiogenesis and acceleration of the wound healing process. The hydrogel developed in this study is versatile and can serve as a promising and effective biomaterial platform for managing bacteria-infected wounds.

Dodecyl tannin derivative linked with 1,2,3-triazole was prepared by the click reaction of dodecyl azide and alkynylated tannin. The structure of tannin derivative was identified by FT-IR spectrometer and elemental analyzer, and the surface activity, antioxidant activity and antimicrobial activity of tannin derivative were studied. The surface tension of tannin derivative was significantly reduced because of the introduction of long chain alkyl groups, and the lowest surface tension was 38.87 mN/m at 1.0 mg/mL. The tannin derivative had strong ability to scavenge 1,1-diphenyl-2-picrylhydrazyl radical, the scavenging rate could reach 89.08 % at 0.25 mg/mL. The tannin derivative exhibited strong antibacterial activity against E. coli and S. aureus due to the increased fat-solubility of tannin derivative and the introduction of antibacterial triazole groups in molecular structure of tannin derivative, and the bacteriostatic ratios of tannin derivative against E. coli and S. aureus were 92.16 % and 89.21 % at 2.0 mg/mL, respectively. The tannin derivative can be used as good candidates for antibacterial packaging or antioxidant supplements. Graphical

Silver nanoparticles (AgNPs) have attracted considerable attention owing to their unique biological applications. AgNPs synthesized by plant extract is considered as a convenient, efficient and eco-friendly material. In this work, the aqueous extract of Areca catechu L. nut (ACN) was used as the reducing and capping agents for one-pot synthesis of AgNPs, and their antioxidant and antibacterial activities were investigated. UV (Ultra Violet)-visible spectrum and dynamic light scattering (DLS) analysis revealed that the size of AgNPs was sensitive to the synthesis conditions. The synthesized AgNPs were composed of well-dispersed particles with an small size of about 10 nm under the optimal conditions (pH value of extract was 12.0; AgNO 3 concentration was 1.0 mM; reaction time was 90 min). In addition, scanning electron microscope with energy dispersive X-ray (SEM-EDX), transmission electron microscopy (TEM) and X-ray diffraction (XRD) results further verified that the synthesized AgNPs had a stable and well-dispersed form (Zeta potential value of −30.50 mV and polydispersity index of 0.328) and a regular spherical shape (average size of 15–20 nm). In addition, Fourier transform infrared spectrometry (FTIR) results revealed that phytochemical constituents in ACN aqueous extract accounted for Ag + ion reduction, capping and stabilization of AgNPs. The possible reductants in the aqueous extract of Areca catechu L. nut were identified by high-performance liquid chromatography-electrospray ionization-quadrupole-time of flight-mass spectrometry (HPLC-ESI-qTOF/MS) method. More importantly, the synthesized AgNPs indicated excellent free radical scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH , IC 50 = 11.75 ± 0.29 μg/mL) and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS + , IC 50 = 44.85 ± 0.37 μg/mL), which were significant higher than that of ascorbic acid. Moreover, AgNPs exhibited an enhanced antibacterial activity against six selected common pathogens (especially Escherichia coli and Staphylococcus aureus ) compared with AgNO 3 solution. In a short, this study showed that the Areca catechu L. nut aqueous extract could be applied for eco-friendly synthesis of AgNPs.

Clausena indica (Datz.) Oliv fruit pericarps (CIOPs) is an important agro-industrial by-product rich in active components. In this article, the effects of traditional and green deep eutectic solvents (DESs) on the high-performance liquid chromatography (HPLC) characterization, antioxidant activities, and α-glucosidase-inhibitory activity of phenolic extracts from CIOPs were investigated for the first time. The results showed that ChCl-Gly and Bet-CA had higher extraction efficiency for the total phenolic content (TPC, 64.14–64.83 mg GAE/g DW) and total flavonoid content (TFC, 47.83–48.11 mg RE/g DW) compared with the traditional solvents (water, methanol, and ethyl acetate). LC-Q-Orbitrap-MS/MS was adopted to identify the phenolic compositions of the CIOPs extracts. HPLC-diode array detection (HPLC-DAD) results indicated that arbutin, (–)-epigallocatechin, chlorogenic acid, procyanidin B1, (+)-catechin, and (–)-epicatechin were the major components for all extracts, especially for deep eutectic solvents (DESs). In addition, ChCl-Xyl and ChCl-Gly extracts showed higher antioxidant activities against 2,2-diphenyl-1-picrylhydrazyl (DPPH•), 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid (ABTS+•), ferric reducing antioxidant power (FRAP), reducing power (RP), and cupric ion reducing antioxidant capacity (CUPRAC) than extracts extracted by other solvents. A strong α-glucosidase-inhibiting activity (IC50, 156.25-291.11 μg/ml) was found in three DESs extracts. Furthermore, in silico analysis of the major phenolics in the CIOPs extracts was carried out to explore their interactions with α-glucosidase. Multivariate analysis was carried out to determine the key factors affecting the antioxidant activity and α-glucosidase-inhibiting activity. In short, DES can be taken as a promising solvent for valorization and recovery of bioactive compounds from agro-industrial by-products. The results verified that CIOPs can be used as a prospective source rich in bio-active compounds applied in the food and pharmacy industries.

The modulation of inflammation in tissue microenvironment takes an important role in cartilage repair and regeneration. In this study, a novel hybrid scaffold was designed and fabricated by filling a reactive oxygen species (ROS)-scavenging hydrogel (RS Gel) into a radially oriented poly(lactide-co-glycolide) (PLGA) scaffold. The radially oriented PLGA scaffolds were fabricated through a temperature gradient-guided phase separation and freeze-drying method. The RS Gel was formed by crosslinking the mixture of ROS-responsive hyperbranched polymers and biocompatible methacrylated hyaluronic acid (HA-MA). The hybrid scaffolds exhibited a proper compressive modulus, good ROS-scavenging capability, and cell compatibility. In vivo tests showed that the hybrid scaffolds significantly regulated inflammation and promoted regeneration of hyaline cartilage after they were implanted into full-thickness cartilage defects in rabbits for 12 w. In comparison with the PLGA scaffolds, the neo-cartilage in the hybrid scaffolds group possessed more deposition of glycosaminoglycans and collagen type II, and were well integrated with the surrounding tissue.

This study aimed to design a new-type curcumin (Cur)-based network nanohydrogel (H), which was composed of whey protein isolate nanofibers (WPINs) and pectin (P). Among them, WPINs were prepared with subcritical water at different treatment times (90 min: WPINs-90, 120 min: WPINs-120, and 180 min: WPINs-180). The structural characteristics, rheology and gastrointestinal release kinetics of Cur-based nanohydrogel were investigated. The results showed that WPINs-120-P-H showed the most favorable particle size (659.00 ± 3.90 nm), potential (−30.00 ± 2.00 mV), peak temperature (110.57 °C) and heating stability. The rheological results revealed that each group of nanohydrogels formed strong elastic gels. In addition, WPINs-120-P-H exhibited the highest encapsulation efficiency (98.42 ± 5.00%) of Cur with the strongest network structure. Moreover, the release curved line of Cur showed an initial release of 19.23% in the stomach, and afterwards, a continuous release trend in the small intestine, reaching a final release amount of 84.80%. Interestingly, the release kinetics of Cur was in line with the Weibull equation model, evidencing a continuous and dynamic release process. This study provided a novel way to prepare the WPINs-P nanohydrogel as delivery for Cur, which demonstrated great potential application prospects in the functional food industry.

Acute lung injury (ALI) features vascular endothelial damage, impaired barrier function, and excessive ROS accumulation, which depletes endogenous peroxidases and disrupts redox homeostasis. Transcriptomic comparison between control and ALI model groups verified marked dysregulation of genes linked to oxidative stress, inflammation, and endothelial barrier dysfunction. To date, exogenous enzyme supplementation represents a necessary strategy for treating ALI, yet very few options are available to halt excessive ROS accumulation within the critical therapeutic time window. Here, we developed a nanozyme system (SCP@CD) capable of solving this problem by encapsulating autofluorescent Mn/S-co-doped carbon dots (CDs) into polymer micelles self-assembled via a Schiff base formed by sialic acid-modified chitosan oligosaccharide and protocatechualdehyde. As a targeting ligand, sialic acid enables active targeting of E-selectin on inflamed endothelium, realizing enhanced fluorescence for noninvasive in vivo tracking. Chitosan oligosaccharide linked to sialic acid reduces systemic toxicity, while the Schiff base formed by chitosan oligosaccharide and protocatechualdehyde mediates pH-responsive release via cleavage in acidic inflammatory microenvironments. SCP@CD functions as an exogenous enzyme supplementation via the multi-nanozyme activities of CDs and achieves synergistic anti-inflammatory and endothelial barrier-repairing effects by regulating inflammatory mediators and junctional proteins. This platform offers a novel precision nanomedicine strategy for ALI.

The processing of litchi (Litchi chinensisSonn.) fruits generates substantial amounts of pericarp and seed by-products, which are candidate sources of antioxidant compounds but are underutilized. This study systematically investigated the total phenolic content (TPC) and in vitro antioxidant capacities (FRAP, ABTS, and DPPH values) of pericarp and seed extracts across eight commercial litchi cultivars. Qualitative LC-MS-based phytochemical profiling was further performed on the representative ‘Bingli’ cultivar. The results indicated that litchi seeds consistently exhibited significantly higher TPC and antioxidant activities compared to pericarps. Among the tested pericarps, the ‘Nuomici’ cultivar showed the highest TPC (10.29 ± 0.10 mg GAE/g dw) and antioxidant capacities. The ‘Bingli’ cultivar demonstrated the highest overall antioxidant activity among seeds (e.g., TPC of 33.92 ± 1.75 mg GAE/g dw and FRAP of 373.06 ± 4.64 μmol Fe(II)/g dw). LC-MS-based qualitative screening tentatively annotated several hydroxycinnamic acids and A-type procyanidins as putative phenolic constituents of the ‘Bingli’ litchi pericarp and seed extracts. Subsequent targeted HPLC analysis determined the concentrations of Procyanidin A2, revealing that it had a high concentration in the ‘Xianjinfeng’ and ‘Bingli’ seeds (1298.73 ± 54.87 and 1127.81 ± 67.22 μg/g dw, respectively). These findings indicate cultivar- and tissue-specific variations in litchi fruit by-products and suggest that the seeds of ‘Bingli’ and ‘Xianjinfeng’ are candidate sources of antioxidant phenolics for further investigation, pending validation in food models or biological systems.

High susceptibility of wet Spirulina platensis to post-harvest degradation causes significant nutritional and functional quality losses. Fermentation is a proven strategy to improve bioactive properties but the advancement of this process through integration of predictive modeling analysis remains uninvestigated. Therefore, this study combined response surface methodology (RSM) with Multilayer perceptron artificial neural network (MLP-ANN) modeling to improve the accumulation of phenolic compounds in wet Spirulina beverage via sequential fermentation with Kluyveromyces marxianus and Lactiplantibacillus plantarum . Predicted fermentation conditions (yeast-to-bacteria inoculum ratio of 3.75:1.25, glucose concentration of 8.5%, and Spirulina concentration of 9.5%) suggested the production of 60.56 mg gallic acid equivalents (GAE)/L of total phenolic content (TPC),which was experimentally confirmed at 60.86 mg GAE/L. Both RSM and MLP-ANN models demonstrated strong predictive performance (R 2 > 0.99), but RSM outperformed MLP-ANN by achieving 0.49% of relative error as compared to 0.74% for MLP-ANN. Experimental validation of predictive condition confirmed 76.2%, 119.6%, and 96.69% increase in TPC, total flavonoid content (TFC), and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity respectively. UPLC-MS (ultra-performance liquid chromatography-mass spectrometry) analysis of TPC revealed substantial increases in caffeic acid, kaempferol, and apigenin contents in the fermented beverage. TPC improvement could potentially be attributed to the possible activation of complementary microbial biotransformation pathways including aromatic amino acid metabolism and flavonoid precursor synthesis. An improvement in sensory acceptability and reduction in detectable biogenic amines has also been observed under predicted sequential fermentation conditions. These findings underscore the feasibility of predictive models to improve the functional quality of Spirulina -based fermented beverages.

Alcalase time-specifically dictated hydrolysis property, peptide profiles and multifunction. • Antioxidant and ACE inhibition positively correlated with structural unfolding related hydrolysis efficacy promotion. • DPP-IV inhibition relied more on characteristics of specific peptide motifs. • Activity-guided multivariate and differential peptide profile analysis revealed candidate peptides. Endpoint-based studies failed to clarify time-resolved hydrolysis properties, bioactivities, peptide profiles, and structural characteristics. Current study aimed to explore the correlations of these characteristics during Alcalase hydrolysis of cricket protein (CP) for 30–300 min. Prolonged hydrolysis increased degree of hydrolysis (DH) to 20.3%, with low-molecular-weight peptides (< 2 kDa) accounting for 88.0%–94.0%. Cricket protein hydrolysates (CPHs) showed strongest antioxidant and angiotensin I-converting enzyme (ACE) inhibitory activities at 30 and 300 min, respectively, whereas dipeptidyl peptidase-IV (DPP-IV) inhibition exhibited a bimodal pattern. Structural analyses demonstrated hydrolysis disrupted ordered CP structure, with α -helix+ β -sheet decreasing by 17.4%–26.5%, alongside lowered surface hydrophobicity and increased sulfhydryl exposure. Antioxidant and ACE inhibition were positively associated with structural unfolding and hydrolysis property promotion, whereas DPP-IV inhibition appeared to relate more to peptide-release characteristics. Activity-guided peptidomics and multivariate analyses prioritized LYPL/VYGPL, SGLFDK/PAALGL, and SGPRLH/FAGPS as candidate peptides potentially associated with antioxidant, ACE inhibitory, and DPP-IV inhibitory activities, respectively.
