On-site quantitation of xanthine in fish and serum using a smartphone-based spectrophotometer integrated with a dual-readout nanosensing assay

Ningxia Xu,Meng Xiao,Zipei Yu,Baohui Jin,Mengsu Yang,Changqing Yi

Rapid and quantitative biochemical analysis at points-of-need is imperative for food safety inspection. This work reports on: 1) a stand-alone smartphone-based “two-in-one” spectrophotometer (the SAFS) installed with a self-developed application (the SAFS-App) which can precisely collect both absorption spectra and fluorescence spectra in a reproducible manner within 5 s; and 2) a straightforward protocol for xanthine detection using fluorescent carbon nanodots and silver nanoparticles. The assay performed with the SAFS demonstrates high specificity towards xanthine, and a linear range of 1–60 μM with LODs of 0.38 and 0.58 μM for colorimetric and fluorometric readouts, respectively. The reliability and robustness of the SAFS are validated by on-site quantitation of xanthine in fish and serum samples, with comparable accuracy to HPLC method. More importantly, the SAFS presents itself as an appealing device which is accessible to everyone through the Internet of Things and can be tailored for diverse point-of-care testing applications.

Design and fabrication of Zn@ZnO@Ag chip for Raman scattering analysis of norfloxacin in liquid milk, fish and animal feeds

Hanqiang Zhang,Xuejing Fan,Yangyue Ding,Lunzhao Yi,Kun Ge,Ying Gu

A novel and easy-to-prepare plasmonic nanoparticles doped semiconductor substrate—Zn@ZnO@Ag chip with ultra-high surface-enhanced Raman scattering (SERS) activity was fabricated for label-free, rapid and sensitive analysis of norfloxacin. The Zn@ZnO array was synthesized by surface oxidation at low temperature, followed by in-situ reduction to form leaf-like AgNPs on Zn@ZnO array without extra reducing agent, thus fabricating a Zn@ZnO@Ag chip. The ultra-high SERS activity is attributed to the synergistic effect of semiconductor characteristics of ZnO and surface plasmon resonance properties of leaf-like AgNPs. The possible enhancement mechanism was verified by density functional theory simulations. The proposed SERS method showed a wide linear range (3.0–500.0 μg/L) and low limit of detection (0.8 μg/L) for norfloxacin analysis. High sensitivity, good selectivity and acceptable recoveries (82.7–113.6%) in real sample analysis were obtained. This study offers a promising SERS chip-based platform for norfloxacin detection in the field.

Development and evaluation of a semi-automatic single-step clean-up apparatus for rapid analysis of 18 antibiotics in fish samples

Shuhe Mu,Tengwang Teng,Ruidong Zhou,Huan Liu,Huiwu Sun,Jincheng Li

Here we report a novel semi-automatic pre-treatment apparatus to fulfill a single-step clean-up aim for rapid quantification of 18 antibiotics in fish samples. Trimethoprim and 17 sulfonamides were extracted from fish samples by acetonitrile solution based on ultrasonic-assisted extraction. Cheap analytical filtration syringe (CAFS) clean-up column was used to fulfill single-step clean-up of the extract which was set on the developed semi-automatic clean-up apparatus. This semi-automatic clean-up apparatus is quite convenient for single-step purification by precisely controlling the flow rate of the extracts, in which up to 10 samples can be purified in parallel at one time. The time needed to clean sample was only about 2 min per 10 samples. Type of compatible purification column ranged from 1 mL (inner diameter × length, 5.6 × 55 mm) to 12 mL (inner diameter × length, 15.6 × 83 mm). The apparatus also did not need additional gas source such as nitrogen or air. After purification, the analytes were simultaneously separated and quantified by high-performance liquid chromatography/quadrupole-linear ion trap tandem mass spectrometry (HPLC-QTRAP-MS/MS). The limits of detections ranged from 0.5 to 1.0 μg kg −1 , and the limits of quantifications ranged from 1.0 to 5.0 μg kg −1 . All the recoveries ranged from 81.39% to 116.01%. Intra-day and inter-day relative standard deviations (RSDs) were lower than 14% for all analytes.

Random forest algorithm-enhanced dual-emission molecularly imprinted fluorescence sensing method for rapid detection of pretilachlor in fish and water samples

Chenxi Liu,Jingxin Liao,Yong Zheng,Ying Chen,Hongsheng Liu,Xizhi Shi

A sensitive and efficient fluorescence sensor based on dual-emission molecularly imprinted polymers (Dual-em-MIPs) was successfully developed using the random forest (RF) machine-learning algorithm for the rapid detection of pretilachlor. SiO 2 coatings on red-emitting CdSe/ZnS quantum dots (r-SiO 2 @QDs) as intermediate light-emitting components are non-selective for pretilachlor, whereas molecularly imprinted layers coated with blue-emitting nitrogen-doped graphene quantum dots (N-GQD S ) are selective. Fluorescence images of the Dual-em-MIPs were acquired. The red (R), green (G), and blue (B) color values of the image were analyzed using an RF algorithm, and the classifier was trained using 103 fluorescent images for automatic analyses. Under optimized conditions, an excellent linear relationship between the sensor and pretilachlor was obtained in the range of 0.001–5.0 mg/L (R 2 , 0.9958). Additionally, the satisfactory recoveries of Dual-em-MIPs ranged between 92.2 % and 107.6 % for the real samples, with a relative standard deviation (RSD) under 6.5 %. The satisfactory recoveries of the RF model based on the fluorescence sensor were 84.2–108.2 % with the RSD under 6.4 %. Overall, the proposed fluorescence sensor based on Dual-em-MIPs and machine learning methods was successfully used to determine pretilachlor in the environment and in aquatic products.

A Novel Rare Earth and Covalent Organic Framework Composite for Rapid and Highly Sensitive Electrochemical Analysis of Sulfadiazine in Fish Muscle

Jiajia Han,Zicong Liao,Guosheng Chen,Junlang Qiu,Fang Zhu

The misuse of sulfadiazine (SFZ) has led to great hazard to the environment and human safety; therefore, a simple, rapid, and sensitive method to detect sulfadiazine is urgently needed. Herein, we report a simple fabrication method for rare earth vanadate samarium (SmV)-doped covalent organic framework COFTDBA-TTLnanocomposites (SmV/COFTDBA-TTL), which were used to construct a sulfadiazine (SFZ) electrochemical sensor. The synergistic effect arising from the combination of SmV and COFTDBA-TTLaccelerates the charge transfer kinetics, along with the creation of more surface-active sites that benefit effective detection. Compared with other electrochemical sensors, this electrochemical sensor exhibits low detection limit (2.40 nM), wide linear range (7.32–12.0 μM), good reproducibility (RSD = 0.823%), and stability (RSD = 3.60%). It provides a novel method and theoretical basis for the application of rare earth COF-based electrochemical sensors to detect environmentally destructive pollutants. Keywords:sulfadiazine;covalent organic framework;rare earth vanadate;electrochemical sensor;fish muscle

A Simple Method for the Determination of Pharmaceutical and Personal Care Products in Fish Tissue Based on Matrix Solid-Phase Dispersion

Shouchun Xiao,Shengchen Lv,Zheng Cheng,Caiwei Hu,Pengxi Li,Fang Nan,Xueke Liu,Donghui Liu,Zhiqiang Zhou,Peng Wang

A simple and effective pretreatment method based on matrix solid-phase dispersion was developed for the determination of pharmaceutical and personal care products (PPCPs) and their metabolites in fish by high-performance liquid chromatography tandem mass spectrometry. The type and amount of dispersant, adsorbent, and eluting solvent were optimized by a single-factor experiment and Box–Behnken design. Under the optimal conditions with 2.5 g of Florisil as a dispersant, 500 mg of C18 as an adsorbent, and 5 mL of acetonitrile as an eluting solvent, the recoveries ranged from 70.4 to 99.9% with relative standard deviations less than 10.5%, and the limits of quantitation ranged from 0.13 to 1.01 μg/kg. The developed method was successfully applied to detect PPCPs in marketed fish, and five PPCPs, including triclocarban, sulfadiazine, sulfadimidine, sulfamethoxazole, and carbamazepine, were detected at trace levels. The proposed method, which has the advantages of short analysis time, less solvent consumption, and high sensitivity, can be used for the determination of trace PPCPs in fish.

Identification of Cytochrome P450 Isozymes Involved in Enantioselective Metabolism of Fipronil in Fish Liver: In Vitro Metabolic Kinetics and Molecular Modeling

Siqi Wu,Huizhen Li,Jing You

Fipronil has been frequently detected in waterways worldwide at concentrations that threaten aquatic organisms, yet the metabolic behavior of fipronil enantiomers in aquatic organisms is largely unknown, which is of significance in enantioselective toxicity evaluation. We quantitatively identified the specific cytochrome P450 (CYP) isozymes involved in metabolizing fipronil enantiomers in tilapia by combining in vitro metabolic kinetic assays and molecular docking. Inhibition studies suggested that CYP1A enzyme was the main isoform catalyzing metabolism of fipronil and that CYP3A contributed in a limited way to the metabolism in fish liver S9. Both the dissipation rate constant and the maximum metabolic velocity of R -(−)-fipronil were greater than those of S -(+)-fipronil in tilapia liver S9, suggesting that tilapia selectively metabolized R -(−)-fipronil. The CYP1A1 isozyme exhibited the highest binding capacity to R -(−)-fipronil and S -(+)-fipronil (binding energy ‒9.39 and ‒9.17 kcal/mol, respectively), followed by CYP1A2 (‒7.30 and ‒6.94 kcal/mol, respectively) and CYP3A4 (‒7.16 and ‒6.91 kcal/mol, respectively). The results of in vitro metabolic assays and molecular docking were consistent, that is, CYP1A, specifically CYP1A1, exhibited a higher metabolic capacity to fipronil than CYP3A, and fish liver S9 selectively metabolized R -(−)-fipronil. The present study provides insight into the enantioselective metabolic behavior and toxicological implications of the in vitro metabolic kinetics of fipronil in fish. Chain length-dependent antioxidant activity of phosphatidylcholine complexes of alkyl gallates in fish oil and emulsions: a reverse micelle interfacial mechanism

Qian Wang,Qingxiang Zhang,Fujun Liu,Weiyuan Niu,Xiangbo Zeng,Chuan Li,Xinmiao Wang,Dayong Zhou,Fawen Yin

Non-monotonic chain length-dependent antioxidant activity of alkyl gallate-PC complexes in fish oil. • Short-chain complexes (C2) quench aqueous initiators at the hydrophilic interface. • Long-chain complexes (C12) intercept lipophilic radicals in the reverse micelle leaflet. • Excess PC micelles relocate long-chain complexes to the O/W interface for enhanced efficacy. In bulk oils, phosphatidylcholine (PC) and trace water form reverse-micelle interfaces that concentrate hydroperoxides and pro-oxidant metals, promoting oxidation. PC complexes of gallic acid and selected C1-C16 alkyl gallates were evaluated in fish oil and emulsions. Activity peaked at C2 (ethyl gallate-PC, EG-PC) and C12 (dodecyl gallate-PC, DG-PC) rather than changing progressively with chain length. After accelerated oxidation, EG-PC and DG-PC showed the lowest peroxide values (25.27 and 24.53 mmol/kg oil) and TBARS values (7.00 and 6.89 mg MDA/kg oil), while preserving polyunsaturated fatty acids and suppressing aldehyde formation. Spectroscopy supported complexation, whereas cryo-TEM, CLSM, and SAXS supported reverse-micelle formation. We propose that PC anchors gallates at this interface, the short-chain EG-PC localizes near the hydrophilic region to quench aqueous-phase initiators, whereas the long-chain DG-PC partitions into the lipophilic region to intercept chain-propagating radicals. In emulsions, excess PC micelles further relocate DG-PC to the interface, enhancing its efficacy. Download: Download high-res image (263KB) Download: Download full-size image

Crosslinked fish scale gelatin/alginate dialdehyde functional films incorporated with carbon dots derived from pomelo peel waste for active food packaging

Yongshi Li,Junxian Yang,Linping Sun,Bingzhen Liu,Hui Li,Lincai Peng

A multifunctional and environmentally friendly composite film was developed by incorporating pomelo peel-derived carbon dots (PCDs) into a fish scale gelatin (FSG)/alginate dialdehyde (ADA) biopolymer matrix. ADA was used to reinforce the physicomechanical properties of the FSG film via Schiff base crosslinking. PCDs with strong antioxidant and antimicrobial activities were synthesized via a hydrothermal method . The effect of various PCDs content on the surface morphological, physicochemical, and functional characteristics of the composite films was investigated. The results showed that the introduction of PCDs into the FSG/ADA matrix effectively reinforced the mechanical performance, enhanced the water vapor and water resistance, increased UV-light blocking, conferred fluorescence properties, and improved the thermal properties of the composite films. Under 3 wt% PCDs content, the FSG/ADA/PCDs-3 % composite film not only presented significant antioxidant capacity with a radical scavenging rate of 91.71 % for DPPH and approximately 100 % for ABTS , but also exhibited excellent antimicrobial ability against bacteria and fungi. Results of a preservation experiment showed that the prepared FSG/ADA/PCDs-3 % film preserved the physiological qualities of strawberries post-harvest and extended their shelf-life to 7 days at room temperature. Overall, the fabricated FSG/ADA/PCDs composite films are promising for use in eco-friendly active food packaging.

Adsorption characteristics of polythiophene for tetracyclines and determination of tetracyclines in fish and chicken manure by solid phase extraction-HPLC method

Piaoyang Li,Danmei Rao,Yimei Wang,Xiaorong Hu

Porous polythiophene materials were successfully synthesized by external-crosslinker method and applied as sorbents in solid-phase extraction. A method for the determination of tetracycline antibiotics (tetracycline/ oxytetracycline/doxycycline) in fish and chicken manure was proposed by combining solid phase extraction-ultraviolet visible spectrophotometer (SPE-UV) and solid phase extraction-high performance liquid chromatography (SPE-HPLC). The effects of material dosage, extraction time, washing time, eluent type and dosage, and the reusability of polythiophene sorbent on extraction rate were investigated. With the use of SPE-HPLC method, satisfied linearity for three tetracyclines antibiotics was obtained in the range of 0.004–1 mg/L. The detection limits (LODs) were ranged from 0.0012 to 0.0027 mg/L and the relative standard deviations (RSDs) were 3.5–7.3% (n = 5). The recoveries in actual fish samples and chicken manure samples were 91.1–114.1% and 69.5–93.2%, respectively. No tetracycline antibiotics were detected in fish samples and no doxycycline was detected in chicken manure samples. However, oxytetracycline and tetracycline were detected in two chicken manure samples with the contents of 5.12, 6.58 mg/kg and 6.08, 7.72 mg/kg, respectively. This method was simple, sensitive and suitable for the determination of tetracyclines in practical samples.

Histamine detection in fish samples based on indirect competitive ELISA method using iron-cobalt co-doped carbon dots labeled histamine antibody

Yi-Fang Li,Zheng-Zhong Lin,Cheng-Yi Hong,Zhi-Yong Huang

Due to complex matrixes and specific reagent deficiency, the rapid detection of histamine is still a challenge to date. Based on the high peroxidase-like activity of iron-cobalt co-doped carbon dots, an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) was established for histamine detection using the mimic enzyme labeled with histamine antibody (His-Ab). Through the competitive binding of the labeled His-Ab to solid-phase and sample antigens, histamine content was detected with a linear range of 2.5–150 μg mL −1 . The detection limit based on 3σ/K was 0.50 mg kg −1 , which was much lower than those of commercial His-kit and HPLC methods. The ic-ELISA method was applied to histamine detection in fish samples with the recovery of (103.4 ± 0.5)%, which was in accord with those of commercial His-kit and HPLC methods. The results indicated that the established ic-ELISA method was suitable for rapid detection of histamine in fish samples with high accuracy, sensitivity and stability.

A fish scale-like magnetic nanomaterial as a highly efficient sorbent for monitoring the changes in auxin levels under cadmium stress

Qingqing Ding,Hui Chen,Chuanhui Huang,Qiaomei Lu,Ping Tong,Wenmin Zhang,Lan Zhang

Sorbents with high surface utilization and good dispersibility are of great importance for the extraction performance of magnetic solid-phase extraction (MSPE). In this study, a fish scale-like magnetic nanomaterial (Co@Co3O4/OCN) was synthesized, which can be used as a highly efficient MSPE sorbent due to its strong magnetism, special morphology, doping of N element, numerous micro–mesopore cavities and organic functional groups (hydroxyl and carboxyl). Furthermore, a Co@Co3O4/OCN-based MSPE method for monitoring the changes in the levels of three auxins (indole-3-acetic acid, indole-3-propionic acid and 3-indole butyric acid) was successfully established. Wide linear ranges (1.0–1000.0 pg mL−1) with good correlation coefficients (R > 0.9992), low limits of detection (LODs, 0.2–4.0 pg mL−1) and satisfactory repeatability (RSD ≤5.9%, n = 3) were obtained. Using the developed method, various growth parts and different growth periods of plants under Cd stress were monitored. The results showed that auxins in various parts of plants showed differential response under Cd stress, and there was a threshold for the changes in auxin levels against Cd stress. This indicates that the developed fish scale-like Co@Co3O4/OCN nanomaterial has a good application prospect for enriching small molecular targets containing hydroxyl and carboxyl groups.
