The practical application of near-infrared (NIR) phosphors is hindered due to their limited light emitting region and narrow bandwidth. Here, we report a series of Cr3+-doped olivine phosphors, (Mg1−xLix)(Mg1−xScx)GeO4:Cr3+, with tunable luminescence from NIR-I to NIR II based on crystallographic site engineering. By using a [Li+–Sc3+] unit substituting for a [Mg2+–Mg2+] unit, the NIR luminescence can be largely tuned from 940 nm to 1110 nm, and the corresponding FWHM can be modified from 236 to ∼300 nm. The internal quantum efficiency (IQE) and external quantum efficiency (EQE) of Mg2GeO4:1% Cr3+ were measured to be 48.19% and 16.38%, respectively. The photoluminescence of the phosphors matches well with the absorption of several functional groups. By measuring the NIR absorption of different liquids illuminated by NIR light from our phosphors, qualitative and quantitative analysis can be realized. These results suggest that the super-broad NIR luminescence of (Mg1−xLix)(Mg1−xScx)GeO4:Cr3+ has potential applications as light sources for nondestructive food analysis.

SARS-CoV-2 infection remains a global health threat, yet its pathogenic mechanisms are incompletely understood. Here, we show that viral nucleocapsid protein (NP) is detectable in the serum of patients with SARS-CoV-2 infection independently of viral RNA. Using virus-like particle (VLP) systems and in vitro infection models, we demonstrate that NP is secreted in a vesicle-free form via type I unconventional protein secretion (UPS) pathway. This process is regulated by NP phosphorylation and oligomerization, coordinated by viral structural proteins, and dependent on membrane components including heparan sulfate proteoglycans (HSPGs) and phosphatidylinositol 4,5-bisphosphate (PI (4,5) P 2 ). Secreted NP is preferentially taken up by granulocytes and induces the release of inflammatory cytokines, including IL-6 and TNF-α. Our findings shed a light on the secretion mechanism of this pro-inflammatory NP, highlighting it as a potential therapeutic target for COVID-related systemic inflammation.

Background Among tumors affecting the central nervous system (CNS), glioblastoma multiforme (GBM) is the most aggressive and lethal form. Given the complex mechanisms of the CNS and the blood-brain barrier (BBB), effective therapies for GBM remain limited. To identify potential therapeutic candidates for glioma, we developed a small-molecule library to screen for compounds with potent anti-proliferative effects and BBB permeability. The mechanisms underlying their anti-glioma activity were further elucidated to provide insights into new treatment strategies. Methods A library of small molecules was screened to identify agents that significantly inhibit glioma cells activity. The effects of the lead compound (Dihydrotanshinone I, DHT) on glioma progression were evaluated through a series of experimental approaches, including cell counting kit-8 assays, cell cycle analysis, apoptosis detection, wound healing, transwell migration, reactive oxygen measurement, subcutaneous xenograft mouse models, and intracranial orthotopic tumor models. To elucidate the underlying mechanisms, network pharmacology and transcriptomic analyses were employed. The mechanism of DHT in glioma pathogenesis was further validated using bioinformatics analyses and clinical glioma tissue samples. Results Based on pharmacokinetic evaluation, DHT was found to traverse the BBB, indicating its capacity to reach the cerebral parench. In vitro and in vivo experiments demonstrated that DHT significantly suppresses glioma growth and progression. Mechanistic analyses using network pharmacology and RNA sequencing revealed that DHT induces glioma cell apoptosis by inhibiting the Janus kinase-signal transducer and activator of transcription 3 (STAT3) signaling pathway, increasing intracellular reactive oxygen species levels and triggering intrinsic apoptotic cascades. Furthermore, bioinformatic analyses of clinical cohorts coupled with validation in patient-derived glioma specimens confirmed that elevated STAT3 expression was correlated with unfavorable prognosis and was significantly increased in patients with high-grade glioma. Conclusions This study demonstrates that, in addition to its role as a key contributor to glioma malignancy and a prognostic marker, STAT3 is also a target of DHT, highlighting its potential as a promising therapeutic candidate for treating GBM. A bioactivity repurposing strategy expanded the potential pharmacological applications of sophoridine derivatives. • Sophoridine derivatives modulated the Topo I–DNA catalytic system and exhibited preliminary antitumor activity. • Molecular simulations characterized potential interactions and dynamic behaviors within the Topo I–DNA complex. Sophoridine is a quinolizidine-type natural alkaloid with favorable structural modifiability and diverse biological activities. However, the pharmacological potential of sophoridine derivatives originally optimized for insecticidal activity remains largely unexplored. Here, a bioactivity repurposing strategy was applied to investigate the antitumor potential of these derivatives. Screening across multiple cancer cell models identified A549 cells as a sensitive model, and compounds 5 and 9 were selected as active candidates with IC 50 values of 0.124 μM and 1.396 μM, respectively. Topo I-mediated DNA relaxation assays showed that both compounds interfered with the activity of the Topo I–DNA catalytic system in a concentration-dependent manner. Molecular docking, molecular dynamics simulations, and PMF analyses characterized their potential interactions within the Topo I–DNA complex. Preliminary in vivo evaluation suggested inhibitory effects on tumor growth accompanied by tumor tissue necrotic changes following treatment with compounds 5 and 9 . Collectively, these findings highlight the antitumor potential of sophoridine derivatives and suggest that modulation of the Topo I–DNA system may represent one potential mechanism contributing to their biological effects, providing preliminary insights for the further development of sophoridine-based anticancer candidates. A series of sophoridine-based derivatives were systematically evaluated for their antitumor potential. In vitro antiproliferative assays identified compounds 5 and 9 as active candidates against A549 cells, with IC 50 values of 0.124 μM and 1.396 μM, respectively. Topo I-mediated DNA relaxation assays showed that both compounds interfered with the Topo I–DNA catalytic system in a concentration-dependent manner. Molecular docking, molecular dynamics simulations, and PMF analyses further characterized their potential interactions and dynamic behaviors within the Topo I–DNA complex. Preliminary in vivo evaluation suggested that compounds 5 and 9 exhibited inhibitory effects on tumor growth, accompanied by increased tumor necrotic changes. Collectively, these findings indicate that sophoridine derivatives possess promising antitumor potential, while modulation of the Topo I–DNA system may represent one potential mechanism contributing to their biological effects, providing a basis for further optimization and development of sophoridine-based antitumor candidates. Serum C-terminal telopeptide of type I collagen (CTX-I) is a key bone turnover biomarker in osteoporosis, and its elevated levels are indicative of increased bone resorption. Current techniques for CTX-I measurement suffer from high operational requirements, complex procedures, and high costs. This study focuses on a novel strategy for detecting serum CTX-I concentration to predict the occurrence and severity of osteoporosis. Using silver-capped Fe 3 O 4 (Fe 3 O 4 @Ag) as the substrate material, an artificial antibody for CTX-I was prepared via magnetic core-shell molecularly imprinted polymers (MIPs), and surface-enhanced Raman scattering (SERS) was used as a signal output device. With crystal violet (CV) at 10 −4 M as the reporter molecule, the proposed biosensor exhibited a good linear response over the range of 10 pg/mL to 2 ng/mL (R 2 = 0.986, LOQ = 4.79 pg/mL), covering the normal reference range required for identifying bone turnover. Moreover, the biosensing platform demonstrated resistance to matrix interference in real serum and urine samples. After six reuse cycles, the SERS intensity of CV decreased by only 10.5%, and after 15 days of storage at 4 °C, it decreased by 15.7%. The Fe 3 O 4 @Ag-MIPs were used to detect CTX-I with excellent uniformity (RSD = 4.1%) and repeatability (RSD = 2.7%), and the overall analysis time was 30 min. The proposed biosensing platform was applied to analyze known real serum samples obtained from human. The proposed biosensor was validated using enzyme-linked immunosorbent assay (ELISA), and the developed biosensor showed good correlation with ELISA. The experimental results indicate that the proposed novel biosensing platform is a fast, low-cost, and reliable tool for the rapid detection of the bone biomarker CTX-I in serum or urine. Magnesium Hydride (MgH 2 ) is a promising hydrogen storage material but suffers from a high dehydrogenation temperature and sluggish sorption kinetics, which hinder its practical application. Accordingly, we report the use of a Cu-based metal-organic framework (HKUST-1) derived carbon material as a catalyst for optimizing the MgH 2 hydrogen storage performance. The MgH 2 containing 10 wt% HKUST-1 (denoted as MgH 2 -HKUST-1) exhibits a maximum dehydrogenation temperature of 50 °C, much lower than pure MgH 2 . Additionally, it demonstrates enhanced hydrogenation dynamics: it adsorbs 6.4 wt% H 2 within 240 s when heated to 350 °C, and even at 200 °C, it still captures 5.7 wt% H 2 in 60 min. Moreover, the dehydrogenation activation energy is 85.01 kJ per mole of H 2 , showing a marked reduction compared to pure MgH 2 . Furthermore, the excellent H 2 storage performance perhaps stems from the synergistic interaction of Cu 2+ /Cu + redox reactions, mechanochemical size effects, and in situ carbon scaffolding.

Metal halide scintillators have attracted continuously increasing interest for low-dose and high-resolution X-ray imaging, yet typical luminescent silver halides suffer from severe hygroscopicity and exciton quenching caused by Ag–Ag cluster aggregation. To address these challenges, we design and report mononuclear five-coordinate silver(I) halide complexes (dppb) 2 AgX·DMSO (X = I, Br, Cl) with 1,2-Bis(diphenylphosphino)benzene (dppb) ligand. Halogen substitution enables tunable broadband emission, while the chloride analogue achieves efficient thermally activated delayed fluorescence (TADF) and a high PLQY of 76%. The chloride analogue possesses more rigid AgClP 4 coordination environment compared with its bromide and iodide counterparts, enabling restricts excited-state relaxation, suppresses non-radiative decay, and preserves favorable singlet-triplet energetics and spin-orbit coupling. Meanwhile, the chloride analogue shows superior scintillation with a high light yield of 26631 photons/MeV, and a detection limit of 304.67 nGy air s −1 . Furthermore, a high spatial resolution of 20.2 lp mm −1 for X-ray imaging is achieved based on the transparent Ag-Cl composite gel scintillator screen. This work not only provides a promising design principle for achieving high scintillation in TADF-type Ag(I) halide complexes by halogen regulation, but also provide a new platform for Ag(I) halide complexes in X-ray imaging.

Background Thallium, a highly toxic heavy metal, is widely distributed in the environment. The substance poses a grave threat to human health through contamination of the food chain, drinking water, and environmental exposure. Consequently, the monitoring of thallium levels, particularly in water, is of critical importance. In this study, a novel analytical method was developed for the direct quantification of thallium ions (Tl + ) in aqueous samples. This method is based on the specific supramolecular interaction between cryptand[2.2.2] and Tl + , and it utilizes electrospray ionization tandem mass spectrometry (ESI-MS/MS). Results The method demonstrated remarkable analytical performance, exhibiting a linear range of 2.5 to 100 μg/L (R 2 > 0.999), a limit of quantification (LOQ) of 2.5 μg/L, and a limit of detection (LOD) of 0.8 μg/L. The recovery rates exhibited a range from 88.65% to 118.09%, with relative standard deviations (RSD) falling below 10%, thereby affirming the attainment of satisfactory accuracy and precision. It is noteworthy that this method necessitated only 5 μL of sample and did not require any preliminary separation steps. Despite the inhibitory effects exhibited by complex sample matrices, these effects were effectively mitigated through a straightforward dilution strategy. The method was successfully applied to the analysis of 66 real-world environmental water samples. Significance This work presents a novel tool that enables rapid and reliable detection of Tl + in water samples. Compared with traditional ESI methods for heavy metals, the application of cryptand[2.2.2] makes the developed method more selective and sensitive, since it has strong binding ability and selective to Tl + ion. The study introduces a novel design concept and practical application for the development of supramolecular recognition-based ESI-MS methods for metal ion analysis. These methods hold significant potential for environmental monitoring and related fields.

Developing efficient semiartificial photosynthetic systems is crucial for sustainable solar energy conversion. Photosystem I (PSI) is an ideal photoactive component for such systems due to its ability to perform light-induced charge separation with near-perfect quantum efficiency. We report a high-performance PSI-based biophotocathode by immobilizing PSI in a poly(vinyl)imidazole-Os(bispyridine)2Cl (POs) redox polymer hydrogel within a translucent, three-dimensional (3D) porous indium tin oxide (ITO) scaffold. This hierarchical architecture synergistically enhances the PSI/redox hydrogel loading, promotes light harvesting, and facilitates rapid mediated electron transfer (MET), leading to record-breaking steady-state photocurrent densities of 4068 μA cm–2. An outstanding incident photon-to-current conversion efficiency (IPCE) of up to 61% is observed at a light intensity of 0.14 mW cm–2. To create a self-sustaining system, this advanced photocathode was integrated with a bioanode featuring galactose oxidase that efficiently oxidizes glycerol, a low-cost byproduct of the biodiesel industry. The resulting two-compartment photoelectrochemical (PEC) cell operates as a biophotovoltaic cell, exhibiting an open-circuit voltage of 0.42 V and a maximum power density of 18.5 μW cm–2. This work not only presents a significant advancement in the performance of PSI-based biophotocathodes but also demonstrates a viable pathway for semiartificial photosynthetic systems that simultaneously convert solar energy and valorize a waste stream chemical.

Background Abdominal aortic aneurysm (AAA) is a prevalent and potentially fatal cardiovascular disease, yet the precise therapeutic mechanism of the bioactive compound Senkyunolide I (SEI) against AAA is not fully understood. Methods Utilizing an elastase/BAPN-induced AAA mouse model, this study employed integrated network pharmacology, proteomics (LiP-MS), and biophysical validation (molecular docking, MST) to identify and confirm PARP-1 as a direct target of SEI. The therapeutic effect of SEI was evaluated in vivo by monitoring aortic dilation and histopathology, and its inhibitory effect on TNF-α-induced macrophage migration was assessed in vitro . The functional role of the SEI-PARP-1 axis was further verified using PARP-1 inhibitors and macrophage-targeted PARP-1 overexpression. Results In the elastase/BAPN-induced AAA model, which mimics human AAA progression and rupture risk, SEI treatment attenuated disease progression by reducing aortic dilation, collagen deposition, and elastin degradation. SEI directly bound to and downregulated PARP-1 expression in both AAA tissues and large peritoneal macrophages (LPMs). It dose-dependently suppressed TNF-α-induced LPMs migration in vitro and restored the AAA-induced reduction in peritoneal LPMs proportion in vivo , an effect also seen with pharmacological PARP-1 inhibition. In contrast, macrophage-specific PARP-1 overexpression abolished SEI's protection and promoted LPMs migration into aortic lesions. Conclusion This study demonstrates that SEI attenuates AAA progression by targeting PARP-1 and inhibiting the migration of LPMs from the peritoneal cavity to the aortic lesion. These findings elucidate a novel pharmacological mechanism of SEI and underscore PARP-1 as a promising therapeutic target for AAA.
