Building in biologically appropriate multifunctionality in aqueous copper indium selenide-based quantum dots

Xiling Yang,Yun Li,Peisen Zhang,Lingfei Guo,Xiaoqi Li,Yiyang Shu,Kuiyu Jiang,Yi Hou,Lihong Jing,Mingxia Jiao

Advanced nanoplatforms equipped with different functional moieties for theranostics hold appealing promise for reshaping precision medicine. The reliable construction of an individual nanomaterial with intrinsic near-infrared (NIR) photofunction and magnetic domains is much desired but largely unexplored in a direct aqueous synthesis system. Herein, we develop an aqueous phase synthetic strategy for Mn2+ doping of ZnS shell grown on Zn–Cu–In–Se core quantum dots (ZCISe@ZnS:Mn QDs), providing the optimal NIR fluorescence quantum efficiency of up to 18.9% and meanwhile efficiently introducing paramagnetic domains. The relaxometric properties of the water-soluble Mn-doped QDs make them desirable for both the longitudinal and transverse (T1 and T2) magnetic resonance (MR) contrast enhancement due to the shell lattice-doped Mn2+ ions with slow tumbling rates and favoured spin-proton dipolar interactions with surrounding water molecules. Surprisingly, the incorporation of Mn2+ ions into the shell is found to significantly enhance the production of reactive oxygen species (ROS) by combining both the chemodynamic and photodynamic processes upon NIR light irradiation, showing great potential for efficient photo-assisted ablation of cancer cells. Furthermore, a broad-spectrum excitation range beneficial for bright NIR fluorescence imaging of breast cancer has been proven and offers high flexibility in the choice of incident light sources. Multiparametric MR imaging of the brain has also been successfully demonstrated in vivo.

Efficient Degradation of Oxytetracycline in a Novel Plug-In Three-Dimensional Photocatalytic Microreactor

Yubin Wang,Yang Jin,Lianyu Jia,Chenxi Xiong,Jun Li,Ming Chen

The combination of a photocatalytic reaction and microfluidic technology is an increasingly popular method utilized to continuously degrade wastewater. In this work, a novel photocatalytic microreactor with TiO2 loaded on the channel center insert was designed and the photocatalytic degradation process of oxytetracycline (OTC) was investigated. Parameters such as TiO2 loading, OTC initial concentration, light intensity, and ion (NO3– and SO42–) concentrations were systematically investigated. The results show that the degradation efficiency of OTC was 56% under the optimal experimental conditions. Compared with the conventional miniaturized photocatalytic reactors, the reaction rate constant and space–time degradation efficiency of the plug-in three-dimensional photocatalytic microreactor proposed in this work were 1–2 orders of magnitude higher. In addition, dimensionless correlation was obtained to predict the degradation efficiency. This work provides a promising microfluidic technology for the process intensification of photocatalytic reaction and wastewater treatment.

Change in nutrients of Citri Reticulatae ‘Chachi’ in relation to browning during aging

Yitian Zhu,Yan Zhu,Xinyu Liu,Jin Qi

Citri Reticulatae ‘Chachi,’ also called “Guang Chen pi” (GCP), is a geoherb of Citri reticulatae pericarpium(CRP), mainly produced in Xinhui, Guangdong, China. It has been a food and traditional medicine for thousands of years. GCPs are popular green food, and can be divided into three picking times and different aging periods according to edible quality. Despite many pharmaceutical studies, there is little information on edible differences among GCP types. The dynamic changes in dietary value for different aging and picking times were investigated through quantifying polyphenols, soluble proteins, ascorbic acid , reducing sugars, polysaccharides , and β -carotene. The browning index (BI) and pH were also measured. Comprehensive analysis results showed that nutrient contents in brownish and reddish peels were close, and higher than in green peel. With aging time, ascorbic acid and β -carotene decreased, while dissolution of polysaccharides, polyphenols, and proteins increased, likely contributing to the aroma change. At the same time, pH of the system gradually decreased, but BI increased. Mathematical path analysis of related substrates and BI showed the greatest correlation of polyphenols with BI. The browning mechanism likely started with ascorbic acid degradation that caused reduced antioxidation, contributing to further reaction of polyphenols with other compounds to produce browning.

Egocentric processing of items in spines, dendrites, and somas in the retrosplenial cortex

Ning Cheng,Qiqi Dong,Zhen Zhang,Li Wang,Xiaojing Chen,Cheng Wang

Egocentric representations of external items are essential for spatial navigation and memory. Here, we explored the neural mechanisms underlying egocentric processing in the retrosplenial cortex (RSC), a pivotal area for memory and navigation. Using one-photon and two-photon calcium imaging, we identified egocentric tuning for environment boundaries in dendrites, spines, and somas of RSC neurons (egocentric boundary cells) in the open-field task. Dendrites with egocentric tuning tended to have similarly tuned spines. We further identified egocentric neurons representing landmarks in a virtual navigation task or remembered cue location in a goal-oriented task, respectively. These neurons formed an independent population with egocentric boundary cells, suggesting that dedicated neurons with microscopic clustering of functional inputs shaped egocentric boundary processing in RSC and that RSC adopted a labeled line code with distinct classes of egocentric neurons responsible for representing different items in specific behavioral contexts, which could lead to efficient and flexible computation.

In vitro evidence of Piperlongumine as a potential radiosensitizer in cervical cancer cells

Li Shan,Ye Lu,Xiaoli Zhu,Xu Cheng

Background Radiotherapy has been applied for the treatment of cervical cancer (CC), and our current study aims to reveal the mechanism of Piperlongumine on strengthening radiotherapy sensitization in CC. Methods The cell counting kit-8 assay was applied to explore the inhibitory effects of Piperlongumine on the viability of CC cells HeLa and SiHa, and the mechanism of action of Piperlongumine on strengthening radiotherapy sensitization in CC cells. The generation of reactive oxygen species in CC cells after the treatment of different concentrations of Piperlongumine, meanwhile, was observed under a fluorescence microscope, while the assay of flow cytometry was adopted to determine the cell cycle. Further, the content of oxidative stress indicators glutathione (GSH) and thioredoxin reductase (TrxR) and the protein level of DNA damage indicator H2AX was quantified using the enzyme-linked immunosorbent assay (ELISA) and western blotting, respectively. Results The intervention of Piperlongumine evidently reduced the proliferation and survival fraction of CC cells following irradiation at different radiation dosages. Meanwhile, the intervention of Piperlongumine led to increased reactive oxygen species production, while inducing DNA damage (as evidenced by increased H2AX levels), disrupting redox homeostasis (reduced GSH, increased TrxR), and promoting G2/M phase arrest in CC cells. Due to the lack of in vivo validation and the incomplete elucidation of certain mechanisms in this study, future work is needed. Conclusion The current study elucidates the mechanisms of Piperlongumine on strengthening radiotherapy sensitization in CC.

Zhang Wenqin,Cai Xiaozhen,Zhang Xinyi,Zou Shiqi,Zhu Danhong,Zhang Qiulong,Chen Jianmin

Objective To prepare polylactic acid microneedles (PLAMNs) with sustained antibacterial effect to avoid skin infection caused by traditional MNs-based biosensors. Methods Silver nanoparticles (AgNPs) were synthesized using an in-situ reduction process with polydopamine (PDA). PLAMNs were fabricated using the hot-melt method. A series of pressure tests and puncture experiments were conducted to confirm the physicochemical properties of PLAMNs. Then AgNPs were modified on the surface of PLAMNs through in-situ reduction of PDA, resulting in the formation of PLAMNs@PDA-AgNPs. The in vitro antibacterial efficacy of PLAMNs@PDA-AgNPs was evaluated using agar diffusion assays and bacterial liquid co-culture approach. Wound healing and simulated long-term application were performed to assess the in vivo antibacterial effectiveness of PLAMNs@PDA-AgNPs. Results The MNs array comprised 169 tiny needle tips in pyramidal rows. Strength and puncture tests confirmed a 100% puncture success rate for PLAMNs on isolated rat skin and tin foil. SEM analysis revealed the integrity of PLAMNs@PDA-AgNPs with the formation of new surface substances. EDS analysis indicated the presence of silver elements on the surface of PLAMNs@PDA-AgNPs, with a content of 14.44%. Transepidermal water loss (TEWL) testing demonstrated the rapid healing of micro-pores created by PLAMNs@PDA-AgNPs, indicating their safety. Both in vitro and in vivo tests confirmed antibacterial efficacy of PLAMNs@PDA-AgNPs. Conclusions In conclusion, the sustained antibacterial activity exhibited by PLAMNs@PDA-AgNPs offers a promising solution for addressing skin infections associated with MN applications, especially when compared to traditional MN-based biosensors. This advancement offers significant potential for the field of MN technology.

Novel phthalide derivatives: Synthesis and anti-inflammatory activity in vitro and in vivo

Liu Zeng Chen,Jing Wu,Kang Li,Qian Qian Wu,Rui Chen,Xin Hua Liu,Ban Feng Ruan

Phthalide is a promising chemical scaffold and has been proved to show potent anti-inflammatory efficacy. In this study, three series, total of 31 novel phthalide derivatives were designed and synthesized, their anti-inflammatory activities were screened in vitro and in vivo . The anti-inflammatory activity of all the compounds were screened on LPS induced NO production to evaluating their inhibitory effects. Structure-activity relationship has been concluded, and finally 3-((4-((4-fluorobenzyl)oxy)phenyl)(hydroxy)methyl)-5,7-dimethoxyisobenzofuran-1 (3H)-one (compound 9o) was found to be the active one with low toxicity, which showed 95.23% inhibitory rate at 10 μM with IC 50 value of 0.76 μM against LPS-induced NO over expression. Preliminary mechanism studies indicated that compound 9o activated Nrf2/HO-1 signaling pathway via accumulation ROS and blocks the NF-κB/MAPK signaling pathway. The in vivo anti-inflammatory activity shown that compound 9o had obvious therapeutic effect against the adjuvant-induced rat arthritis model.

Breakup dynamics of water-in-water droplet generation in a flow-focusing microchannel

Yinan Liu,Chunying Zhu,Taotao Fu,Xiqun Gao,Youguang Ma

In ATPS, the ultra-low interfacial tension makes the stable generation of droplets very difficult. In this paper, the stable and homogeneous preparation of ATPS droplets is achieved over a wide range of flow rates, and the self-similarity analysis and formation dynamics of the droplets are studied. Formation of ATPS droplets show good self-similarity. The generation process of ATPS droplets can be divided into slow squeezing, fast squeezing and rapid pinch-off stages. In the slow squeezing stage, the dimensionless minimum neck width w m / w c is linear with the time, and the absolute value of the slope increases with the capillary number of continuous phase Ca c . In the rapid squeezing and rapid pinch-off stages, the w m / w c satisfies the power-law relationship with time, and the absolute value of the power-law indexes decrease with increasing Ca c , and the pre-exponential factor of pinch-off stage increases with increasing Ca c .

Selective Cleavage of Methylene Linkage in Kraft Lignin over Commercial Zeolite in Isopropanol

Xiangchen Kong,Chao Liu,Hongyun Zeng,Yuyang Fan,Huiyan Zhang,Rui Xiao

Graphical Depolymerization of Kraft lignin to value-added monomers can significantly enhance the profitability of lignocellulosic biorefinery. In this work, the efficient transformation of highly condensed Kraft lignin to valuable aromatic monomers through stepwise alcoholysis and zeolite-catalyzed methylene linkage cleavage is disclosed. Lignin is an aromatic polymer that constitutes over 30 wt% of lignocellulosic biomass and is the most important source of renewable aromatics in nature. The global paper industry generates more than 70 million tons of Kraft lignin annually. Depolymerization of Kraft lignin to value-added monomers can significantly enhance the profitability of biorefinery. However, the method is impeded by the severe condensation of Kraft lignin during the pulping process, which forms robust C−C bonds and results in low monomer yields. In this study, we present a stepwise approach for producing valuable aromatic monomers from Kraft lignin through the cleavage of both C−O and C−C bonds. The approach initiated with complete cleavage of C−O bonds between lignin units within Kraft lignin through alcoholysis in isopropanol, resulting in a monomer yield of 8.9 %. Subsequently, the selective cleavage of methylene linkages present in the residual dimers and oligomers was achieved with commercial MCM-41 zeolite in the same pot, proceeding with an additional monomer yield of 4.0 %, thereby increasing the total monomer yield by 45 %. This work provides an avenue for increasing the depolymerization efficiency of Kraft lignin.

Dietary proanthocyanidins alleviated ovarian fibrosis in letrozole-induced polycystic ovary syndrome in rats

Yanyuan Zhou,Huan Lan,Zhewen Dong,Wenjing Cao,Zhen Zeng,Jia-Le Song

This study investigated the effects of proanthocyanidins (PCs) on ovarian fibrosis in letrozole-induced polycystic ovary syndrome (PCOS) in rats. The administration of PCs effectively reduced the body weight (BW) and relative ovarian weight in PCOS rats. ELISA results revealed that PCs significantly reduced the level of serum T, LH, LH/FSH in the PCOS group. In addition, qRT-PCR results revealed that treatment with PCs significantly increased the main antioxidant enzymes (Cat, Sod2, Gpx3, Mgst1, Gsta4, Sod1 and Prdx3) in PCOS rats. Also, the expression analysis of proteins by Western blotting revealed that PCs significantly decreased the level of TGF-βR1, p-Smad3, p-Smad2 and Smad4 and reversed the downregulation of Smad7 in PCOS rats. The study suggested that PCs improved ovarian fibrosis in PCOS rats by regulating the serum hormone level, inhibiting oxidative stress and suppressing the activation of the TGF-β1/Smads signaling pathway. Practical applications Currently, plant extracts are being widely used to treat female reproductive and metabolic disorders. Particularly, proanthocyanidins (PCs), the well-known natural polyphenolic compounds, which are a significant source of antioxidants present in many colored fruits, are consumed as fruits as well as a dietary supplement to prevent many disorders. Recent pharmacological studies have reported that PCs have many health beneficial properties, such as antioxidant activity, improving cholesterol homeostasis, blood lipid regulatory properties, microcirculation improvement effect, antitumor activity and anti-aging activity. Despite these properties of PCs, the antifibrosis effect of PCs has not been studied to date. The main purpose of this study was to research the role and the mechanisms of PCs in ovarian fibrosis in PCOS rats.

Two single mutations in carboxylesterase 001C improve fenvalerate hydrolase activity in Helicoverpa armigera

Jing-jing Xu,Yong-mei Chang,Mei Lu,Yuan Tie,Yan-ling Dong,Guang-you Chen,Zhi-qing Ma,Xi-li Liu,Yong-qiang Li

Carboxylesterases (CarEs) usually play critical roles in the detoxification of toxic chemicals and therefore may be involved in insecticide resistance in agricultural pests. Previous work has shown that CarE 001C from Helicoverpa armigera was able to metabolize the isomers of cypermethrin and fenvalerate . In this study, seven mutants of CarE 001C with single amino acid substitution were produced and expressed in the Escherichia coli. Enzyme kinetic analysis indicated that all seven mutations dramatically reduced enzymatic activities toward the generic substrate α -naphthyl acetate, but in vitro metabolism assay showed that two of the mutations, H423I and R322L, significantly improved hydrolase activities toward fenvalerate, with their recorded specific activities being 3.5 and 5.1 nM·s −1 ·mg −1 proteins, respectively. Further, thermostability assay showed that the stability of one mutant enzyme was enhanced. This study will help us better understand the potential of CarEs in insecticide detoxification and resistance in H. armigera .

Quercetin is a promising pancreatic lipase inhibitor in reducing fat absorption in vivo

Jian-Feng Zhou,Wen-Jun Wang,Zhong-Ping Yin,Guo-Dong Zheng,Ji-Guang Chen,Jing-En Li,Ling-Li Chen,Qing-Feng Zhang

The inhibitory activity of quercetin on pancreatic lipase was investigated in both vitro and vivo. Quercetin exhibited inhibitory effect on pancreatic lipase with IC 50 value of 70 μg/mL. Inhibitory kinetics indicated that quercetin was more like a mixed-type (non-competitive) inhibitor of pancreatic lipase. Quercetin induced the endogenous fluorescence quenching of lipase, which suggested the interaction between them. The binding constant and the number of the binding sites were further calculated. Molecular docking results showed that quercetin had a binding site on lipase near the active pocket. The binding of quercetin on lipase would affect its conformation, thereby decreasing the affinity between the substrate and the enzyme . In vivo studies showed that pre-administration with 5 and 10 mg/kg bwt quercetin could significantly reduce fat absorption in rat. The inhibition of quercetin on lipase can last at least 2 h in vivo. Correspondingly, quercetin significantly increased fat excretion in rat feces. The change trends of fat and quercetin content in rat feces were similar.
