The Novel Organic Arsenical PZ2 Exhibits Potent Anti-T-ALL Activity via ROS-Dependent Apoptosis and GABARAPL1-Induced Autophagic Cell Death

Xian Zhang,Zhuping Chen,Yujiao Liu,Dongdong Zhang

Objective:T-cell acute lymphoblastic leukemia (T-ALL) is a highly aggressive hematological malignancy with poor overall prognosis. This study systematically evaluates the anti-leukemic therapeutic potential and underlying redox-driven mechanisms of PZ2, a novel organic arsenic compound, in T-ALL.Methods:The cytotoxicity of PZ2 against T-ALL cell lines and normal cells was assessed to determine its therapeutic window. RNA-seq was utilized to map transcriptomic alterations. Intracellular redox homeostasis, mitochondrial membrane potential (MMP), cell cycle, and apoptosis were evaluated using flow cytometry and fluorescence microscopy. Ultrastructural changes and key protein expressions were analyzed via TEM, RT-qPCR, and immunoblotting. In vivo efficacy was validated using a T-ALL cell-line-derived xenograft (CDX) mouse model.Results:PZ2 exhibited superior cytotoxicity and an improved safety profile compared to inorganic arsenic trioxide (ATO). Transcriptomic data indicated strong enrichment in apoptosis and autophagy–lysosome pathways. PZ2 induced profound mitochondrial oxidative stress, leading to ROS accumulation, MMP loss, and subsequent activation of caspase-dependent apoptosis—effects that were completely rescued by ROS scavengers. Furthermore, PZ2 triggered G2/M phase arrest and activated autophagic flux (upregulatedGABARAPL1and LC3-II, degraded p62). Notably, pharmacological inhibition of autophagy rescued PZ2-induced apoptosis, revealing a pro-death autophagic mechanism. In vivo, PZ2 significantly suppressed tumor growth and prolonged survival without overt systemic toxicity.Conclusions:PZ2 exhibits potent anti-T-ALL activity that may involve a dual mechanism: ROS-dependent mitochondrial apoptosis and pro-death autophagy potentially associated withGABARAPL1. This novel organic arsenical may represent a promising and potentially safer alternative for leukemia therapy, and these findings suggest a potential interplay between oxidative stress and autophagic cell death.

Wei-Tong-Xin decoction targets CD8+ T cell-driven colitis via gut microbiota and linoleic acid modulation: an integrated omics study

Fei Lin,Jinyu Wang,Yue Han,Dehong Zhang,Bo Wu,Tingxu Yan,Ying Jia

PE fraction of WTX alleviates colitis via microbiota-linoleic acid-CD8⁺ T cell axis. • PE enriches Bacteroidota , suppresses Proteobacteria and linoleic acid metabolism. • ALOX5/PTGS2 downregulation reduces pathogenic CD8⁺ T-cell infiltration. • Luteolin, Isoalantolactone, Citreorosein identified as key PE constituents. Background Wei-Tong-Xin (WTX), derived from the Hua Tuo Zhong Cang Jing , has been demonstrated to exert therapeutic effects against gastric ulcers and gastritis, with well-established anti-inflammatory properties. However, its potential role in intestinal inflammation has not yet been investigated. Purpose To evaluate the therapeutic efficacy of WTX in dextran sulfate sodium (DSS)-induced colitis in mice and to elucidate its underlying pharmacological mechanisms while identifying its active components. Methods Mice with DSS-induced colitis were treated with WTX extracts. The primary active fraction of WTX was identified by screening intestinal injury and hematological indicators. Integrated 16S rRNA sequencing, serum metabolomics, and network pharmacology analyses were employed to investigate the therapeutic mechanisms. Specifically, serum metabolomics identified linoleic acid (LA) metabolism as the major metabolic pathway regulated by the petroleum ether (PE) fraction, whereas network pharmacology linked PE-derived constituents to colitis-related targets and identified ALOX5 and PTGS2 as key nodes connecting PE with LA metabolism. These findings were subsequently validated using LA supplementation and pseudo-germ-free mouse models. Results The PE fraction significantly alleviated colitis symptoms, including reduced disease activity index scores, attenuation of colon shortening, improved intestinal mucosal barrier integrity, and decreased CD8⁺ T-cell infiltration in the spleen, colon, and mesenteric lymph nodes. PE also reshaped the gut microbiota by modulating the abundance of Bacteroidota and Proteobacteria, reduced the levels of unsaturated fatty acids, particularly those involved in LA metabolism, and downregulated the expression of ALOX5 and PTGS2. Fecal microbiota transplantation produced comparable therapeutic effects, whereas LA supplementation reversed the protective effects of PE. Conclusion PE alleviates colitis by remodeling the gut microbiota, promoting the dominance of Bacteroidota, inhibiting LA metabolism through the ALOX5/PTGS2 pathway, and suppressing the activation of pathogenic CD8⁺ T cells. These effects collectively preserve intestinal barrier integrity and mitigate the progression of colitis. Download: Download high-res image (348KB) Download: Download full-size image

Dual regulation of metabolism and immune contact by a self-reinforcing hydrogel enhances CAR-T-mediated residual tumor clearance and surveillance

Nan Yang,Linlin Tao,Xiaoxiao Hu,Chengqian Shang,Nian Huang,Qiaoqiong Lin,Fangnan Lv,Zhiyong Sun,Chengcheng Gu,Yunfei Lian,Wei Xu,Meirong Huo

CAR-T therapy holds immense potential for eradicating residual tumors and restoring postoperative immune surveillance. However, nutrient deprivation and impaired immune contact within the postoperative tumor microenvironment (PTME) severely hinder the therapeutic efficacy of CAR-T cells. Herein, we developed a self-reinforcing hydrogel system that dually modulated metabolism and immune contact to reprogram the PTME. Specifically, the c-Myc inhibitor JQ1 and bovine serum albumin (BSA)-decorated nanoparticles loaded with the actin cytoskeleton inhibitor NP-G2–044 (BPG) were encapsulated into a reactive oxygen species (ROS)-sensitive hydrogel (JQ1 + BPG@gel). Upon implantation into the surgical cavity, JQ1 + BPG@gel underwent ROS-responsive degradation, continuously releasing agents. JQ1 not only suppressed glycolysis in tumor cells, thereby redirecting nutrients toward CAR-T cells and alleviating lactic acid-induced immunosuppression, but also upregulated the gradients of chemokines CCL5 and CXCL10, thus providing a strong driving force for CAR-T cell infiltration. Notably, JQ1-induced glycolysis inhibition significantly enhanced the tumor uptake of BSA, establishing a self-reinforcing internalization process for BPG. The increased intracellular accumulation of BPG remodeled actin cytoskeleton, enhancing the spatial co-localization between T cells and tumor cells while reducing the steric hindrance imposed by complex cellular topology, thereby facilitating the formation of immune contact and tumor lysis. This hydrogel system improved the anti-recurrence efficacy of CAR-T therapy by 9-fold compared with monotherapy while establishing durable immune surveillance via memory T-cell differentiation within lymphoid organs. This research offers an innovative strategy to improve postoperative tumor cure rates and potentiate CAR-T immunotherapy.

Mixing Surface Tracking Method: A Lagrangian Fluid Tracking-Based Approach for CFD Micromixing Characterization and Its Application in T-Shaped Microreactors

Tianheng Liu,Feng Hong,Renjie Gu,Lixiong Wen

The mixing surface tracking (MS-T) method, a reaction-free approach based on Lagrangian trajectories of fluid elements in CFD simulation, was developed to characterize micromixing under steady-state turbulent flow in T-shaped microreactors. By defining mixing surfaces (MSs) as assemblies of fluid elements of equal mixing time, the evolution of MSs─which visually represented both fluid motion and concentration changes in the mixing process─was quantified using mixing intensity (IM). The micromixing time (tm) was defined as the mixing time required for IM to reach 0.95. The tm values obtained by the MS-T method showed good agreement with those derived from a conventional I–/IO3– reaction system (Villermaux/Dushman method) combined with the incorporation model, confirming the reliability of the MS-T method on micromixing evaluation.

Microvascular endothelial scavenger receptor class B type I protects against heart failure with preserved ejection fraction by inhibiting T-cell cardiotropism

Yufei Wu,Xiaomei Yang,Yu Bai,Chenze Li,Peng Wang,Qing Xu,Hui Li,Xiaoli Rao,Yangkai Xu,Jie Chen,Huanhuan Cao,Qi Zhang,Mingming Zhao,Rui Zhan,Xue Fan,Yuedong Hou,Jie Liu,Hong S Lu,Tianyun Wang,Wei Dong Gao,Linzhang Huang,Han Xiao,Lingyun Zu,Alan Daugherty,Mingguo Xu,Lemin Zheng

Cardiac microvascular endothelial cells (CMECs) dysfunction is a well-recognized mediator of heart failure with preserved ejection fraction (HFpEF), but the underlying mechanism remains unclear. Here we find that scavenger receptor class B type I (SR-B1) is predominantly expressed in CMECs and decreased significantly in HFpEF. Endothelial-specific SR-B1 deficiency exacerbates cardiac pathological remodeling and diastolic dysfunction in HFpEF, which can be prevented by endothelial SR-B1 reconstitution through adeno-associated virus serotype 1 (AAV1)-mediated delivery in endothelial-specific SR-B1-deficient mice. Single-cardiac-endothelial-cell transcriptomics and lineage-tracing system reveal that inflammatory CMECs subcluster activation is responsible for the deteriorating HFpEF progression induced by endothelial SR-B1 loss, rather than endothelial-to-mesenchymal transition. Mechanistically, SR-B1 loss drives increased CXCL10 secretion, which orchestrates CMECs activation and CXCR3-positive T-cell cardiotropism to promote diastolic dysfunction—a process associated with endothelial IRF1 activation. Most importantly, the SR-B1-CXCL10-CXCR3 axis is activated in human HFpEF cardiac tissue, and the elevated CXCL10 level in plasma is independently associated with a higher HFpEF prevalence. This study uncovers that activation of the SR-B1–CXCL10-CXCR3 axis in CMECs aggravates HFpEF pathogenesis through the accumulation of CXCR3-positive T-cells in hearts. This study demonstrates that SR-B1 deficiency in cardiac microvascular endothelial cells (CMECs) exacerbates heart failure with preserved ejection fraction (HFpEF) by driving the CXCL10-CXCR3 signaling, thereby linking inflammatory CMEC activation to CXCR3 + T-cell recruitment. This study demonstrates that SR-B1 deficiency in cardiac microvascular endothelial cells (CMECs) exacerbates heart failure with preserved ejection fraction (HFpEF) by driving the CXCL10-CXCR3 signaling, thereby linking inflammatory CMEC activation to CXCR3 + T-cell recruitment.

Investigation of Density and Viscosity for Ternary Blends of n-Butyl Ether + Methyl Laurate + n-Nonane at T = 293.15–323.15 K

Dan Li,Shasha Liu,Yuhan Zhuang

Densities and viscosities of ternary mixtures of n-butyl ether + methyl laurate + n-nonane along with their corresponding binary subsystems (n-butyl ether + methyl laurate, n-butyl ether + n-nonane, and methyl laurate + n-nonane) were measured over the entire composition range at T = 293.15–323.15 K and ambient pressure (p = 0.1 MPa). These systems were selected to serve as model mixtures for ternary fuel blends of biodiesel. The excess molar volume (VmE) and viscosity deviation (Δη) of the ternary and binary mixtures were derived from the experimental data. The Redlich–Kister, Cibulka, and Nagata–Tamura equations were used to correlate the ternary VmE and Δη. The Kohler, Jacob–Fitzner, Tsao–Smith, and Toop models were used to calculate the ternary VmE. The results provide fundamental thermodynamic data that are essential for optimizing the formulation and predicting the performance of ether–biodiesel–diesel blends.

Lycopene mitigates T-2 toxin-induced systemic inflammation and oxidative stress in association with gut microbiota and SCFAs regulation in mice

Adam Saber Y.,Ennab Wael,Zhu Cuipeng,Yuan Long,Ahmed Abdelkareem A.,Essa Mohamed Osman Abdalrahem,Husien Hosameldeen Mohamed,Saleh Ahmed A.,Kim In Ho,Liu Hao-Yu,Cai Demin

T-2 toxin is a trichothecene mycotoxin produced by Fusarium species and can cause disease after contaminated food or feed ingestion. This study aimed to investigate the impact of lycopene (Ly) on systemic inflammation, oxidative stress, and dysbiosis in mice exposed to T-2 toxin. A total of 20 male BALB/c mice (6 weeks old, weighing 23.5 ± 0.32 g) randomly divided into four groups ( n  = 5): Control (Cont), Ly, T-2, and T-2 + Ly groups. Findings revealed that Ly enhanced the body weight, feedd, and water intake that have been reduced by T-2. Ly increased the villus height (VH) and VH: crypt depth (CD) and decreased CD and villus width (VW) in ileum compared to the T-2 group. In alpha diversity, Staphylococcus and Corynebacterium were the most prevalent in Cont group; Staphylococcus , and Lactobacillus , were the most prevalent in T-2 group; Candidatus-Arthromitus and Staphylococcus were the most prevalent in T-2 + Ly group. Moreover, T-2 mice had significantly ( p  < 0.05) greater levels of interleukin-1 beta (IL-1β), interferon gamma (IFN-γ), Interleukin-17 (IL-17), and tumor necrosis factor-alpha (TNF-α) than Cont, while the Ly significantly ( p  < 0.05) lowered their concentration. The T-2 mice had significantly ( p  < 0.05) higher levels of reactive oxygen species (ROS) and malondialdehyde (MDA), while their catalase (CAT),, glutathione (GSH), adenosine triphosphate (ATP), and superoxide dismutase 1(SOD1) activities were significantly ( p  < 0.05) lower than Cont. Ly significantly ( p  < 0.05) restored their concentration. Moreover, there was a significant reduction ( p  < 0.05) of hexanoic, butyric, acetic, pentanoic, and Heptanoic acids in the T-2 mice compared to Cont. In contrast, treatment with Ly was found to significantly restore their levels compared to T-2 group. We conclude that Ly administration in the context of T-2–induced toxicity may help attenuate systemic oxidative stress and inflammation, possibly through modulation of gut microbiota and fecal short-chain fatty acids (SCFAs).

Reprocessable, high-performance, and degradable bio-based polyhydroxyurethane vitrimers from a Musk T diamide dicyclocarbonate and a vanillin-glycerol epoxy adduct

Yanyan Wang,Zeyun Wu,Jingbo Zhao

Research on biobased polyhydroxyurethane (PHU) vitrimers has attracted increasing attention due to their sustainable development. In this paper, new biobased amide-containing PHU vitrimers (PHUAs) with imine (IM) dynamic covalent bonds mainly were prepared from an ethylene brassylate (Musk T) diamide dicyclocarbonate, a vanillin – glycerol epoxy adduct as the crosslinker, and 1,12-dodecanediamine. The diamide and aromatic vanillin segments imparted excellent mechanical properties to PHUAs. IM bonds provide prominent reprocessability at low reprocessing temperature (120 °C) and good degradability in 2 M HCl–THF (v:v,1:4) mixture solution. PHUAs exhibit excellent mechanical properties with the maximum tensile strength of 60.1 MPa and excellent twice reprocessing efficiency of 87.7 – 97.8%. Although PHUAs contain IM and hydroxyurethane dual dynamic covalent bonds, the IM bonds play the major role in dynamic exchange due to low stress relaxation temperatures (50–70 °C) and low E a (52.73–80.34 kJ/mol) of PHUAs. This study establishes a promising strategy to synthesize strong degradable PHU vitrimers with prominent reprocessing and degradability from renewable substances.

Density and Viscosity of the Ternary System Pinane + n-Dodecane + Methyl Laurate and Corresponding Binary Systems at T = 293.15–333.15 K

Panxi Wu,Lifeng Zhang,Shujun Zhao,Chenchen Wang,Yongsheng Guo,Wenjun Fang

Biofuel may be an important component of aviation fuel for its significant advantages. In the present work, two biofuel samples, methyl laurate and pinane, are selected for blending research with n-dodecane, a typical model fuel of aviation kerosene. Densities and viscosities for the ternary system pinane (1) + n-dodecane (2) + methyl laurate (3) and corresponding binary systems have been measured at the temperatures from 293.15 to 333.15 K and atmospheric pressure. The excess molar volume (VmE) and viscosity deviation (Δη) of binary systems have been calculated based on experimental data and fitted to the Redlich–Kister equation, while those of the ternary system have been fitted to Nagata–Tamura, Redlich–Kister, Cibulka, and Singh semiempirical equations, respectively. The values of the excess molar volume are all positive for all investigated systems, while the viscosity deviations are all negative. The results are discussed from the perspective of intermolecular force and the structure. Furthermore, the densities and viscosities of both binary and ternary mixtures are satisfactorily correlated using the Jouyban–Acree model.

Tumor-secreted IFI35 promotes proliferation and cytotoxic activity of CD8+ T cells through PI3K/AKT/mTOR signaling pathway in colorectal cancer

Li Peisi,Zhou Dawang,Chen Dongwen,Cheng Yikan,Chen Yuan,Lin Zhensen,Zhang Xi,Huang Zhihong,Cai Jiawei,Huang Wenfeng,Lin Yanyun,Ke Haoxian,Long Jiahui,Zou Yifeng,Ye Shubiao,Lan Ping

Background A large proportion of the patients with cancer do not respond to immunotherapies. Recent studies suggested an important role for tumor-infiltrating cytotoxic T lymphocytes (CTL) in enhancing response to immunotherapy. Here, we aim to identify gene that induce proliferative and cytotoxic states of CD8 + T cells, and to investigate its effect on CAR-T cells against colorectal cancer. Methods Correlation between the expression of IFI35 with the activation and cytotoxicity of CD8 + T cells was assessed with TCGA and proteomic databases. Then we constructed murine colon cancer cells over-expressing IFI35 and tested their effect on anti-tumor immunity in both immunodeficient and immunocompetent mouse models. Flow cytometry and immunohistochemistry were performed to assess the immune microenvironment. Western blot analysis was used to identify the potential down-stream signaling pathway regulated by IFI35. We further investigated the efficacy of the rhIFI35 protein in combination with immunotherapeutic treatment. Results The transcriptional and proteomic analysis of the activation and cytotoxicity of CD8 + T cells in human cancer samples demonstrated that IFI35 expression is correlated with increased CD8 + T cell infiltration and predicted a better outcome in colorectal cancer. The number and cytotoxicity of CD8 + T cells were significantly increased in IFI35-overexpressing tumors. Mechanistically, we identified that the IFNγ-STAT1-IRF7 axis stimulated IFI35 expression, and that IFI35-mediated regulation of CD8 + T cell proliferation and cytotoxicity was dependent on PI3K/AKT/mTOR signaling pathway in vitro. Furthermore, IFI35 protein enhanced the efficacy of CAR-T cells against colorectal cancer cells. Conclusion Our findings identify IFI35 as a new biomarker that can enhance the proliferation and function of CD8 + T cells, as well as increase the efficacy of CAR-T cells against colorectal cancer cells. Resveratrol Enhances the Radiosensitivity by Inducing DNA Damage and Antitumor Immunity in a Glioblastoma Rat Model under 3 T MRI Monitoring

Liping Qian,Lihua Mao,Weixing Mo,Rong Wang,Yunlong Zhang

Glioblastoma (GBM) is the most common intracranial tumor with characteristic of malignancy. Resveratrol, a natural originated polyphenolic compound, has been reported to act as a potential radiosensitizer in cancer therapy. Magnetic resonance imaging (MRI) is the first choice for the diagnosis, pathological grading, and efficacy evaluation of GBM. In this study, MRI was applied to observe whether resveratrol could intensify the anti-GBM tumor effect by enhancing antitumor immunity during radiotherapy. We established an intracranial C6 GBM model in SD rats, treated with radiation and resveratrol. The increased body weight, the inhibition on mortality, and tumor volume in radiated- GBM rats were further enhanced by resveratrol addition, while the pathological damage of brain was alleviated. The modulation of radiation on inflammation, cell cycle, and apoptosis was strengthened by resveratrol; and Ki-67, PD-L1, and cell cycle- and apoptosis-related protein expressions were also improved by cotreatment. Besides, cotreatment attenuated DNA damage and induced G0/G1-phase cell arrest of GBM rats, accompanied with the changed expression of ATM-AKT-STAT3 pathway-related proteins. Moreover, the percentages of CD3+CD8+T cells and IFN-γ+CD8+T cells were enhanced, while (CD4+CD25+Foxp3)/CD4+T cells were decreased by radiation or resveratrol, which was strengthened by cotreatment. The modulation effect of cotreatment on CD3, Foxp3, and IFN-γ levels was also stronger than radiation or resveratrol alone. To conclude, resveratrol enhanced the effect of radiotherapy by inducing DNA damage and antitumor immunity in the intracranial C6 GBM.

Development of two immunochromatographic test strips based on signal amplification and selenium nanoparticles for the rapid detection of T-2 mycotoxin

Xianrui Chen,Zhipeng Gao,Tingting Long,Jianhua Xie,Xiujiang Li,Zhibing Huang

Conventional immunochromatographic test strips (ICSs) based on gold nanoparticle (AuNP) probes offer limited sensitivity. Here, AuNPs were separately labeled with monoclonal or secondary antibodies (MAb or SAb). In addition, spherical, homogeneously dispersed, and stable selenium nanoparticles (SeNPs) were also synthesized. By optimizing the preparation parameters, two ICSs based on the dual AuNP signal amplification (Duo-ICS) or SeNPs (Se-ICS) were developed for the rapid detection of T-2 mycotoxin. The detection sensitivities of the Duo-ICS and Se-ICS assays for T-2 were 1 ng/mL and 0.25 ng/mL, respectively, which were 3-fold and 15-fold more sensitive, respectively, than a conventional ICS. Furthermore, the ICSs were applied in the detection of T-2 in cereals, which requires higher sensitivity. Our findings indicate that both ICS systems can be used for rapid, sensitive, and specific detection of T-2 toxin in cereals and potentially other sample types.
