To investigate the digestive behavior of extruded starch-polyphenols system, extruded sweet potato starch vermicelli (ESPSV) was used as a model. The multi-scale structure, starch digestibility, polyphenol release, digestive enzyme activity during digestion and their correlation of ESPSV supplemented with matcha (MT), green tea extract (GTE), tea polyphenols (TP) and epigallocatechin gallate (EGCG) (at 1% polyphenol level) were discussed. Results showed that tea products in whatever form could retard starch digestion, with EGCG working best. The predicted glycemic index (pGI) of ESPSV was decreased from 82.50 to 65.46 after adding EGCG. Starch formed larger molecular aggregates with tea products under extrusion, showing a “B + V” type pattern. The order of V-type crystals content was EGCG + ESPSV (1.41) > TP + ESPSV (1.50) > GTE + ESPSV (1.88) > MT + ESPSV (2.62) > ESPSV (3.20). Under external pressure, EGCG, as tea monomer, was more likely to enter the spiral cavity of amylose and form V-type inclusion complex. Notably, polyphenols released during digestion could still reduce digestive enzyme activity , with a 15.53% decrease in EGCG + ESPSV compared to ESPSV. This was verified by correlation analysis, where RDS content (0.961, p < 0.01 ) and pGI (0.966, p < 0.01 ) were highly significantly correlated with the enzyme activity. Furthermore, tea products did not break or even enhance the quality of ESPSV as the final product.

In this study, the blending electrospinning technique was successfully used to prepare poly (vinyl alcohol)/ethyl cellulose/tea polyphenol (PVA/EC-TP) nanofibrous films for the first time. The morphology observation showed that the fiber diameter ranged from 213 ± 40 nm to 432 ± 78 nm. The thermal analysis indicated that PVA/EC-TP composite nanofibrous films had superior thermal stability to pure PVA nanofibrous film. The wettability test showed that the water contact angle of composite nanofibrous films was significantly increased from 22° to 109°. Water stability and water barrier ability analyses indicated the water solubility of composite nanofibrous films was significantly decreased from 100% to 17%, and the water vapor permeability was reduced from 0.71 to 0.42 g⋅mm/m 2 ⋅h⋅kPa. Meanwhile, the PVA/EC-TP composite nanofibrous films exhibited good antioxidant and antimicrobial activity. The application assay showed that the PVA/EC-TP nanofibrous films with high TP content could effectively extend the shelf life of pork by 3 days.

Homology of medicine and food-zizyphi spinosi semen (ZSS) exhibits abundant pharmacological activities, such as sedation, hypnosis and anti-depression. In the present study, the water soluble polyphenols extracted from ZSS via the acid digestion method were named ZSSP, and exhibited significant anti-colorectal cancer (CRC) activity, characterized by restraining cell proliferation, promoting cell apoptosis and increasing chemo-sensitivity of CRC cells. The potential of ZSSP in vivo was further evaluated in an AOM/DSS-induced colitis-associated carcinogenesis (CAC) mouse model. Intriguingly, ZSSP diminished the number and volume of CAC polyps in mice in a dose-dependent manner, and effectively limited the damage of mice organs induced by AOM/DSS. The immunohistochemistry result showed that the elevated CRC early markers in CAC mice, such as COX-II, EMR1, and Ki67, were potently prevented by the ZSSP treatment. Further, the component in ZSSP with the anti-CRC activity was identified as spinosin by the macroporous resin of SP207 and RP-HPLC-MS/MS. Interestingly, during the extraction process, sodium ions were introduced forming spinosin·Na+, which had better water solubility and more remarkable anti-CRC activity than the spinosin. This study provides a new pharmacological property of spinosin derived from ZSS, inhibiting the growth of human CRC cells and colitis-associated CRC in mice, which indicates its potential use as a natural agent against CRC.

In this study, the improvement of gel properties and digestibility of the water-soluble polymer of tea polyphenol (TP)-egg white protein (TEP) under heat induction (HTEP), was studied. Results indicated that the particle size and turbidity of TEP increased with TP concentration, and the absolute value of ζ-potential decreased. After heat induction, the surface hydrophobicity of HTEP decreased with TP concentration, and the degree of protein aggregation increased. Microstructure and T 2 showed that the gel structure became compact and stable, and HTEP had a strong water-binding ability. The ionic and disulfide bonds were the main chemical bonds in HTEP. The hardness and disulfide bond increased, but the digestion of HTEP increased initially and then decreased (caused by the change of gel structure). Infrared spectroscopy indicated the mutual conversion of intermolecular and intramolecular β-sheets. In short, TP could modify egg white gel through forming stable disulfide bonds and dense gel network structures.

Plant polyphenols can form functional coatings on various materials through self-polymerization. In this paper, a series of modified capillary columns, which possess diversity of charge characteristics for modulating electroosmotic flow (EOF), were prepared by one-step co-deposition of gallic acid (GA), a plant-derived polyphenol monomer , and branched polyethyleneimine (PEI). The physicochemical properties of the prepared columns were characterized by Fourier transform infrared spectroscopy (FT-IR), UV–Vis spectroscopy and scanning electron microscopy (SEM). The magnitude and direction of EOF of GA/PEI co-deposited columns were modulated by changing a series of coating parameters, such as post-incubation of FeCl 3 , co-deposition time, and deposited amounts of GA and PEI with different relative molecular mass (PEI-600, PEI-1800, PEI-10000, and PEI-70000). Furthermore, the separation efficiencies of the prepared GA/PEI co-deposited columns were evaluated by separations of small molecules , including organic acids, polar nucleotides, phenols, nucleic acid bases and nucleosides . Results indicated that modulating of EOF plays an important role in enhancing the separation performance and reversing the elution order of the analytes. Finally, the developed method was successfully applied to quantitative analysis of acidic compounds in four real samples. The recoveries were in the range of 73.5%–85.8% for citric acid , benzoic acid , sorbic acid , salicylic acid and ascorbic acid in beverage and fruit samples, 101.6%–104.9% for cinnamic acid , vanillic acid , and ferulic acid in Angelica sinensis sample, while 84.6%–97.8% for guanosine-5′-monophosphate, uridine-5′-monophosphate, cytosine-5′- monophosphate and adenosine-5′-monophosphate in Cordyceps samples. These results indicated that the co-deposition of plant polyphenol-inspired GA/PEI coatings can provide new opportunities for EOF modulation of capillary electrophoresis .

The main purpose of this work was to investigate the influence of typical thermal sterilization approaches (pasteurization, high-temperature sterilization) on the structure, in vitro digestibility , and antioxidant activity of soy protein isolate (SPI)/black soybean seed coat extract (BE) complexes at pH 7.0. The results of zeta potential and particle size demonstrated that the addition of BE was contributed to inhibit protein thermal aggregation. Heat sterilization resulted in protein unfolding revealed by UV–Vis, circular dichroism , and fluorescence spectroscopy analysis. The increase of BE led to the reduction of fluorescence intensity and surface hydrophobicity of SPI. Moreover, the formation of SPI/BE complexes inhibited protein digestion in vitro, while thermal sterilization promoted protein digestion. The SPI/BE complexes showed a strong radical scavenging ability both before and after thermal treatment.

The total antioxidant capacity and total polyphenol content of 9 tea juices were examined following in vitro digestion model. Results show that there was a significant variation in total antioxidant capacity [16,392.30 ± 111.64–22,340.23 ± 46.73 μmol VC/L (DPPH), 3,194.24 ± 14.76–13,795.07 ± 37.46 μmol Trolox/L (ABTS), 2,540.61 ± 59.79–7,951.57 ± 31.91 μmol Trolox/L ferric reducing antioxidant power (FRAP)] and total polyphenol content (265.84 ± 9.52–876.62 ± 6.59 μg GAE/mL). Following the in vitro digestion, most of the juices of antioxidant capacities and 7 juices of the total polyphenol content were decreased. Before the in vitro digestion, a highly positive correlation was found between the total polyphenol content and FRAP value. After the gastric phase of digestion, there was a very strong positive linear correlation between the total polyphenol content and total antioxidant capacity. After the duodenal phase of digestion, the correlation between the total polyphenol content and total antioxidant capacity was very weak, and also, a very weak linear correlation was found between the antioxidant assays.

Novel soybean meal-based biomimetic (STP) adhesives were fabricated via soybean meal (SM) and enhanced by tannic acid (TA) and polyetheylenimine (PEI) (TAPI) co-crosslinking network based on natural polyphenol-inspired chemistry. The multiple physico-chemical interactions (including intermolecular H-bonding and covalent bonding) between the TAPI co-crosslinking system and SM matrices were examined by the Fourier transform infrared spectroscopy, solid-state 13 C nuclear magnetic resonance, X-ray diffraction, thermogravimetric analysis, and scanning electron microscopy. The results showed that a dense, robust, and water-resistant adhesive layer was constructed between network-bound catechol moieties in the TAPI and SM system, endowing the STP adhesive with high wet bonding strength for plywood. As expected, TAPI-modified SM adhesives showed a 156.1% increase in wet bonding strength compared to the control SM adhesive. The adhesion meets standard requirements for interior-use plywood. Both the solid content and residual mass analysis also confirmed that the enhancement in the STP adhesive was attributable to the network crosslinking density and stiffness after integrating the TAPI system. Moreover, the thermal stability of the resultant STP adhesive exhibited a significant improvement. The proposed STP adhesive may be a promising cost-effective and wet-resistant bio-adhesive for the application in the wood composites industry.

In this work, choline chloride (ChCl) deep eutectic solvents (DESs) were evaluated for the pretreatment of palm samples in the analysis of polyphenols, such as protocatechuic acid, catechins, epicatechin, and caffeic acid. During the enrichment step of the pretreatment, eight DESs comprising ChCl with ethylene glycol (EG), glycerol (Gly), xylitol (Xyl), phenol (Ph), formic acid (FA), citric acid (CiA), oxalic acid (OA), or malonic acid (MA), were prepared and applied to the reflux extraction of polyphenols from palm samples. All the DESs exhibited higher polyphenol extraction efficiency than methanol, and the highest extraction efficiency was obtained using ChCl-FA (1:1, mole ratio). For the purification step of the pretreatment, eight ChCl DES-modified adsorbents were prepared by hydrothermal polymerization and packed into solid-phase extraction (SPE) cartridges, and ChCl-Urea, ChCl-Gly, ChCl-FA, and water, were used as eluents. The ChCl DES-modified adsorbents were characterized by Fourier transform infrared spectroscopy, thermal gravimetric analysis, scanning electron microscopy, and Brunauer–Emmett–Teller surface analysis, and the polyphenols were analyzed by mass spectrometry and high-performance liquid chromatograph-ultraviolet detection. The highest purification efficiency was obtained using the ChCl-Ph DES-modified adsorbent as the SPE packing material and ChCl-Urea-H 2 O (1:1:5, mole ratio), ChCl-Gly (1:1, mole ratio), ChCl-FA-H 2 O (1:1:5, mole ratio), and H 2 O as the eluents. Compared to conventional purification processes that employ commercial C 18 or C 8 SPE columns with organic solvents as eluents, the ChCl DES-based SPE purification process successfully avoided the use of expensive commercial SPE columns and organic solvents. Furthermore, it isolated a larger amount of the target compounds under the same experimental conditions, and could be applied over five cycles with good reversibility. This work indicates that DESs as green solvents have great potential for the totally green pretreatment of samples during the enrichment and purification processes.

Background Silicosis, an interstitial lung disease caused by inhalation of silica particles, poses a significant health concern globally. Green tea polyphenol (TP) stands out as a promising therapeutic candidate, yet its specific protective effects and in-depth mechanisms against silicosis have not been thoroughly investigated. Purpose This study aimed to systematically assess the protective potential of TP against silicosis and to elucidate the underlying mechanisms of its action. Methods A combination of physiological, transcriptomic, molecular, and computational techniques was employed. HPLC was used to identify the components of TP, and its antioxidant properties were tested with DPPH and ABTS assays. The effects of TP on lung injury were assessed in silicosis mice using histopathology, qRT-PCR, and western blot. Transcriptomic analysis was applied to explore the differentially expressed genes and pathways in response to TP intervention. In vitro studies with mouse alveolar macrophages (MH-S) examined TP's effects on cell viability, proliferation, apoptosis, and inflammation responses. Integrated qRT-PCR, western blot, immunohistochemistry, and molecular docking were performed to confirm the molecular mechanism underlying the protective effects of TP against silicosis. Results TP effectively attenuated pulmonary inflammation and fibrosis in silicosis mice, as evidenced by significant reductions in inflammation and fibrotic markers. Moreover, TP's therapeutic benefits were linked to its cytoprotective effects on alveolar macrophages, notably its ability to protect MH-S cells from silica particle-induced apoptosis, inhibition of proliferation, and inflammatory response, underscoring its targeted protective effects at the cellular level. Mechanistically, TP exerted its anti-silicosis activity by targeting key pathways implicated in inflammatory responses, notably through the inhibition of the IL-17/NF-κB p65 signaling cascade. Molecular docking simulations corroborated these findings, demonstrating favorable binding affinities between TP's bioactive components (EGC, ECG, and EGCG) and crucial proteins (IL-17A, IL-17F, p65, TNF-α, IL-6, and IL-1β) involved in the IL-17/NF-κB p65 signaling pathway. This pathway inhibition led to a significant decrease in the production of pro-inflammatory cytokines, such as TNF-α, IL-6, and IL-1β, thus mitigated silicosis. Conclusion TP demonstrates efficacy in alleviating silica particle-induced lung injury by suppressing inflammation through the IL-17/NF-κB p65 signaling pathway, underscoring its potential as a valuable natural compound for silicosis management.

Postfermented Pu-erh tea (PE) protects against metabolic syndrome (MS), but little is known regarding its underlying mechanisms. Animal experiments were performed to determine whether the gut microbiota mediated the improvement in diet-induced MS by PE and its main active components (PEAC). We confirmed that PE altered the body composition and energy efficiency, attenuated metabolic endotoxemia and systemic and multiple-tissue inflammation, and improved the glucose and lipid metabolism disorder in high-fat diet (HFD)-fed mice via multiple pathways. Notably, PE promoted the lipid oxidation and browning of white adipose tissue (WAT) in HFD-fed mice. Polyphenols and caffeine (CAF) played critical roles in improving these parameters. Meanwhile, PE remodeled the disrupted intestinal homeostasis that was induced by the HFD. Many metabolic changes observed in the mice were significantly correlated with alterations in specific gut bacteria. Akkermansia muciniphila and Faecalibacterium prausnitzii were speculated to be the key gut bacterial links between the PEAC treatment and MS at the genus and species levels. Interestingly, A. muciniphila administration altered body composition and energy efficiency, promoted the browning of WAT, and improved the lipid and glucose metabolism disorder in the HFD-fed mice, whereas F. prausnitzii administration reduced the HFD-induced liver and intestinal inflammatory responses. In summary, polyphenol- and CAF-rich PE improved diet-induced MS, and this effect was associated with a remodeling of the gut microbiota.

The immunosuppressive tumor microenvironment (ITME) of osteosarcoma (OS) poses a significant obstacle to the efficacy of existing immunotherapies. Despite the attempt of novel immune strategies such as immune checkpoint inhibitors and tumor vaccines, their effectiveness remains suboptimal due to the inherent difficulty in mitigating ITME simultaneously from both the tumor and immune system. The promotion of anti-tumor immunity through the induction of immunogenic cell death and activation of the cGAS-STING pathway has emerged as potential strategies to counter the ITME and stimulate systemic antitumor immune responses. Here we present a bimetallic polyphenol-based nanoplatform (Mn/Fe-Gallate nanoparticles coated with tumor cell membranes, MFG@TCM) which combines with mild photothermal therapy (PTT) for reversing ITME via simultaneously inducing pyroptosis in OS cells and activating the cGAS-STING pathway in dendritic cells (DCs). The immunostimulatory pathways, through the syngeneic effect, exerted a substantial positive impact on promoting the secretion of damage-associated molecular patterns (DAMPs) and proinflammatory cytokines, which favors remodeling the immune microenvironment. Consequently, effector T cells led to a notable systemic antitumor immune response, effectively inhibiting the growth of both primary and distant tumors. This study offers a comprehensive approach to treat OS by mild photothermal therapy and modulating the immune microenvironment, exhibiting considerable potential in addressing the constraints associated with current therapeutic modalities.
