Adipose tissue-secreted Spz5 promotes distal tumor progression via Toll-6-mediated Hh pathway activation in Drosophila

KongDu,LiXiaoqin,ZhaoSihua,WangChenliang,CaiZixin,SongSha,GuoYifan,KuangXiaoyu,WangXianping,LiuWenhan,LiuPeng,GuoXiaowei,XuWenyan,WangYirong,ZhaoBin,JinBin,HeLi,MaXianjue

Interorgan communication is vital for tissue homeostasis and health in multicellular organisms, and its disruption can lead to diseases such as cancer. Adipose tissue acts as a key endocrine center, secreting cytokines that influence remote organs. Despite clear links between obesity and increased cancer risk, the underlying mechanisms are unclear. Here, utilizing a Drosophila genetic model combining Gal4-UAS and QF-QUAS tissue-specific transgene expression systems, we reveal that adipose-secreted Spz5 ligand promotes distal epithelial tumor overgrowth and invasion. Mechanistically, Spz5 binds to tumor cell Toll-6 receptors, triggering the degradation of the endocytic adaptor protein AP-2α via Mib1-mediated ubiquitination. Consequently, impaired endocytosis leads to Smoothened (Smo) accumulation on the cell membrane and subsequent activation of the Hedgehog (Hh) pathway. This abnormal Hh activation synergizes with the oncogenic Yorkie (Yki) to drive tumor growth and invasion. Furthermore, tumor-derived Unpaired ligands (Upds) activate the JAK-STAT pathway in the fat bodies, which leads to Hippo pathway-dependent upregulation of spz5 expression. Thus, our study provides insights into the complex regulatory mechanisms by which systemic interorgan communication influences tumor progression.

Biochemical and Functional Characterization of E. coli Aminopeptidase N: A New Role as a 6-Monoacetylmorphine Hydrolase

Xiabin Chen,Yishuang Li,Jianzhuang Yao,Xiaoxuan Li,Hualing Li,Zelin Wu,Qi Hu,Nuo Xu,Tingjun Hou,Jiye Wang,Shurong Hou

6-monoacetylmorphine (6-MAM), a primary active metabolite of heroin that reaches the human brain, plays a crucial role in producing heroin-associated physiological and lethal effects. Therefore, 6-MAM has emerged as a key target for alleviating the adverse consequences of heroin abuse. In this study, the proposed 6-MAM hydrolaseE. coliaminopeptidase N (eAPN) was recombinantly produced, and its biochemical and functional profiles were investigated. eAPN’s biochemical properties, with respect to pH, metal ions, and temperature, and catalytic functions toward peptidase substrates and 6-MAM were thoroughly examined. Extensive experiments reveal that incorporation of an N-terminal His-tag notably affects eAPN’s aminopeptidase activity. This cost-effective recombinant eAPN exhibits favorable thermostability and optimal activity at pH 7.5. Kinetic analysis toward peptidase substrates reveals that eAPN preferentially cleaves peptides following amino acid residues in the order of Ala > Arg >> Met, Gly > Leu > Pro, indicating a preference for small or basic amino acid residues as substrates. Computational and experimental studies have, for the first time, discovered that eAPN is capable of catalyzing the hydrolysis of heroin and 6-MAM, which has shed light on its functional versatility and potential applications. This work elucidates the biochemical properties of eAPN and expands its catalytic functions, thereby laying the groundwork for a deep understanding and further reengineering of eAPN to enhance its activity toward 6-MAM for heroin detoxification.

Supramolecular K⁺ Coordination in Poly(dibenzo-18-crown-6)/SWCNTs Freestanding Electrodes for High-Energy-Density Supercapacitors

Xin Xu,Fengxing Jiang,Xiaofang Liu,Cheng Liu,Congcong Liu,Qinglin Jiang,Haixin Ding,Jingkun Xu,Peipei Liu

The development of freestanding electrodes with high energy density remains a key challenge for potassium-ion supercapacitors. Herein, a supramolecular strategy is presented through the in situ electropolymerization of dibenzo-18-crown-6 (DB18C6) on single-walled carbon nanotubes (SWCNTs) films, fabricating the flexible poly(DB18C6) (PDB18C6)/SWCNTs hybrid electrodes. This design leverages dual charge-storage mechanisms: a conductive SWCNTs network for rapid electron transport and embedded PDB18C6 macrocycles for reversible K + coordination via host-guest interactions, which significantly enhances pseudocapacitance and cycling stability of the prepared film. By optimizing the electropolymerization time, the freestanding electrode achieves a high energy density of 77 Wh kg -1 at a power density of 1595 W kg -1 within an expansive operating window of -1.5 to 1.5 V. Notably, a high capacitance retention of 99.25% is maintained after 15,000 cycles at 10 A g -1 , attributable to the robust K + -crown ether coordination effect. Furthermore, a symmetrical supercapacitor assembled with PDB18C6/SWCNTs electrodes delivers an energy density of 18.75 Wh kg -1 at 1350 W kg -1 . This work underscores the promise of supramolecular coordination chemistry for designing high-performance energy storage devices.

Endophytic Bacillus toyonensis D-6 via producing antifungal volatile organic compounds effectively controls green grape postharvest diseases

Lijun Ling,Wenyue Zhang,Rui Yue,Yijuan Jiang,Fanjin Kong,Rongxiu Mo,Lijun Feng,Yongpeng Zhou

Grapes are susceptible to fungal infections before, during transport, or after harvest, causing significant economic losses. Traditional methods have high equipment costs and are prone to the development of drug resistance. The antifungal potential of volatile organic compounds (VOCs) produced by endophytic bacteria is beginning to be understood. VOCs are ideal for post-harvest disease control in fruits and vegetables, as they can work without direct contact with the fruit. In this study, we isolated pathogenic fungi ( Penicillium crustosum G-4, Alternaria alternata G-5, Arthrinium phaeospermum G-8) from post-harvest rotting green grape and used G-8 as a test organism, screened the endophytic bacterium Bacillus toyonensis D-6 from Typhonium giganteum Engl. that could produce strong antagonistic active VOCs, analyzed the components of its VOCs and screened for the highly effective inhibitory compound 3-methylvaleric acid (3-MA). The inhibitory activities and mechanisms of action of the VOCs and antifungal compounds were investigated. The results showed that the VOCs and 3-MA produced by D-6 inhibited the growth and reproduction of the pathogenic fungi, which mainly exerted antagonistic effects by destroying the normal morphological structure of mycelium and spores and damaging the integrity of cell membranes. In addition, appropriate concentrations of VOCs and 3-MA inhibited the spread of pathogenic fungi on green grapes and effectively delayed the decline in fruit quality. This opens up new possibilities for post-harvest preservation of green grapes and provides a theoretical basis for related applications.

6′-O-Caffeoylarbutin: A Stable and Long-Lasting Skin-Lightening Agent Targeting Melanogenesis Through MITF Pathway Modulation

Zhongyi Chen,Fengkun Xiao,Chunhao Yang,Zhaoxiang Yang,Liping Qu,Feifei Wang

The cosmetic application of arbutin (AR) has recently been challenged due to concerns regarding the skin toxicity associated with its metabolite, hydroquinone (HQ). While the derivatization of AR provides a strategy to promote stability, the multistep and complicated chemical synthesis restricts its industrial production. The present study reported a naturally derived AR derivative, 6′-O-caffeoylarbutin (CA), which is hypothesized to resist hydrolysis to HQ owing to the conjugation effect and steric hindrance, resulting in better stability. To verify this hypothesis, the degradation of CA and AR was investigated using high-performance liquid chromatography with diode-array detection. The skin-lightening substances, AR and caffeic acid, were found in the degradation products of CA under acidic conditions, which also contained lower quantities of HQ compared to that generated by AR. However, CA was not degraded under enzymatic conditions, exhibiting better stability, which might contribute to its prolonged efficacy and reduced toxicity. The long-lasting skin-lightening efficacy of CA was further verified in a zebrafish drug-withdrawal model. Notably, Mechanistic investigations revealed that CA disrupts melanosome maturation and trafficking by modulating the melanocyte-inducing transcription factor-regulated pathway, providing molecular insights into its skin-lightening efficacy. Graphical This study investigates 6′-O-caffeoylarbutin (CA), a natural arbutin (AR) derivative, demonstrating greater stability than AR by resisting hydrolysis into hydroquinone under acidic and enzymatic conditions. In the zebrafish drug-withdrawal model, CA exhibits prolonged skin-lightening efficacy compared to AR. Mechanistic investigations in vitro further reveal that CA's skin-lightening efficacy is achieved by disrupting melanosome maturation via the MITF-regulated pathway.

Weihuang Wang,Xiaomin Wang,Guilin Chen,Liquan Yao,Xiaojuan Huang,Tao Chen,Changfei Zhu,Shuiyuan Chen,Zhigao Huang,Yi Zhang

Antimony sulfide-selenide Sb 2 (S,Se) 3 as a promising absorber material has drawn extensive attention owing to its rewarding photoelectric properties, low toxicity, and earth abundant components. Here, an available and highly effective method, in situ hydrothermal growth accompanied with postselenization, is successfully applied to fabricate Sb 2 (S,Se) 3 solar cells. The rationale for the preparation of Sb 2 S 3 precursors by the hydrothermal method is discussed, and detailed characterization of Sb 2 (S,Se) 3 films selenized with different temperatures is also carried out. Finally, a solar cell with structure of FTO/CdS/Sb 2 (S,Se) 3 /Spiro-OMeTAD/Au is fabricated, which delivers an encouraging 6.14% efficiency with V oc of 732 mV, J sc of 14.6 mA cm −2 , and fill factor of 60.8%. This work provides valuable references for further development and improvement of Sb 2 (S,Se) 3 solar cells.

Ultra-rapid and high-titer biomanufacturing of trehalose 6-phosphate by an in vitro synthetic biology platform

Liu Bohua,Guo Qingqing,Wang Shuo,Shi Ting,Lu Fuping,Zhang Yi-Heng P. Job

Trehalose 6-phosphate (T6P), often referred to as plant insulin, serves as a central signaling molecule that regulates carbon partitioning and sucrose flux in plants, thereby influencing key agronomic traits, such as grain yield and drought resilience. Foliar spraying of T6P has been shown to significantly enhance yield and stress tolerance of numerous grains and vegetables. To circumvent the dependency on coenzymes in natural T6P synthesis, we designed and validated an in vitro new-to-nature, coenzyme-free, minimal enzymatic pathway for the biosynthesis of T6P from maltose and polyphosphate. This three-enzyme cocktail contained maltose phosphorylase, trehalose 6-phosphate phosphorylase, and polyphosphate glucokinase, and did not involve any costly coenzymes, such as ATP or UDP. Through systematic optimization of experimental parameters (including pH, temperature, Mg²⁺, phosphate concentration, and enzyme ratios), a 93% molar yield of T6P was achieved from 10 g/L maltose. The scale-up of this in vitro bioprocess to a 100-mL bioreactor with 200 g/L maltose enabled the production of up to 541 mM T6P (i.e., 252 g/L T6P disodium salt) within two hours, corresponding to a very high volumetric productivity of 126 g/L/h. This study established a scalable and cost-competitive in vitro biomanufacturing of T6P. Chemical intervention based on timed foliar spraying of T6P to cultivated crops offers a simpler and safer agricultural practice compared to genetic modification of crops. A polysaccharide from Arthrospira platensis alleviates pancreatic cancerassociated with negatively regulating galectin-3 and glypican-6 expression

Rakotomalala Manda Heriniaina,Xia Chen,Yulian Wu,Wenfeng Liao,He Fei,Peipei Wang,Kan Ding

Pancreatic cancer, specifically pancreatic ductal adenocarcinoma (PDAC), ranks among the most prevalent malignancies and is a leading cause of cancer-related mortality worldwide. Therefore, there is an urgent need to identify novel anti-pancreatic cancer agents. This study reports a newly identified homogeneous polysaccharide, designated ESPPW, isolated from Arthrospira platensis ( A. platensis ). The molecular weight of ESPPW is estimated at 356 kDa, and it consists predominantly of glucose and rhamnose, with minor amounts of mannose, glucuronic acid, galacturonic acid, galactose, xylose, arabinose, and fucose. ESPPW inhibits the proliferation and migration of PDAC cells both in vitro and in vivo . Mechanistic investigations reveal that ESPPW induces apoptosis through activation of caspase-3 and is associated with upregulation of the tumor-suppressor protein p53. Notably, treatment with 2.8 nmol·L −1 of ESPPW leads to significant time-dependent downregulation of galectin-3 (Gal-3) and glypican-6 (GPC-6). These findings are corroborated by immunohistochemical analysis of tumor xenograft tissues. Furthermore, overexpression of Gal-3 and GPC-6 reverses the pro-apoptotic effect of ESPPW, as indicated by restored cycle regulatory proteins (CDK2) expression. In conclusion, these data demonstrate that ESPPW suppresses PDAC cell growth by promoting apoptosis and disrupting the functional activity of Gal-3 and GPC-6.

Low-dielectric and low-temperature curable fluorinated nano carbon/polyimide composites with 6-aminoquinoline for end capping

Jialin Zhang,Yuying Sui,Jinhui Li,Liang Shan,Fangfang Niu,Guoping Zhang,Rong Sun

One challenge in the high-performance dielectric material of polyimide (PI) is to obtain the excellent comprehensive properties at low curing temperature for advanced IC package. In this work, 6-aminoquinoline (AQL) is introduced through covalent bonds for end capping to obtain a low-temperature curable PI at first. The results demonstrate that with very limited introduce (2.76%) of AQL, the imidization index could reach as high as 1.01 at 200°C. Then, fluorinated nano carbon (FC) is further introduced resulting in the fluorinated nano carbon/polyimide (FCPI) composites, which effectively reduces the dielectric constant (1 wt%, 2.75 @ 1 MHz). Besides, the as-prepared FCPIs also possess excellent thermal property of which FCPI-0.3 exhibited the 5% weight loss temperature of 540°C and the glass transition temperature is also as high as 385°C. Furthermore, the tensile strength of the film is 127 MPa, the elongation at break is around 15.82%, and the Youngs' modulus is about 3.10 GPa proving the superior mechanical property. As a result, the successful preparation of the FCPI nanocomposites not only possess low-temperature curing and low-dielectric performance but also exhibit excellent thermal and mechanical properties, which shows a wide range of application prospects in the field of microelectronics for the advanced electronic package.

6-Shogaol Inhibits Oxidative Stress-Induced Rat Vascular Smooth Muscle Cell Apoptosis by Regulating OXR1-p53 Axis

Liu Jing,Li Bin,Li Wenlian,Pan Taowen,Diao Yunpeng,Wang Fangjun

Apoptosis of vascular smooth muscle cells (VSMCs) is closely related to the pathogenesis of cardiovascular diseases, and oxidative stress is an important cause of VSMCs’ death. Inhibiting VSMCs apoptosis is an effective preventive strategy in slowing down the development of cardiovascular disease, especially for atherosclerosis. In this study, we found that oxidation resistance protein 1 (OXR1), a crucial participator for responding to oxidative stress, could modulate the expression of p53, the key regulator of cell apoptosis. Our results revealed that oxidative stress promoted VSMCs apoptosis by overexpression of the OXR1-p53 axis, and 6-shogaol (6S), a major biologically active compound in ginger, could effectively attenuate cell death by preventing the upregulated expression of the OXR1-p53 axis. Quantitative proteomics analysis revealed that the degradation of p53 mediated by OXR1 might be related to the enhanced assembly of SCF ubiquitin ligase complexes, which is reported to closely relate to the modification of ubiquitination or neddylation and subsequent degradation of p53.

MicroRNA-183 accelerates the proliferation of hepatocyte during liver regeneration through targeting programmed cell death protein 6

Hou Fangxing,Li Xing,Wang Yanfeng,Xiao Xiangzuo

Background Liver regeneration is a highly orchestrated process concerning the modulation of various microRNAs (miRs). miR-183 was recently found to be involved in the process of liver regeneration, that miR-183 was remarkably up-regulated at 2–6 h after partial hepatectomy. Objective This study was aimed to explore the mechanism of miR-183 in on liver regeneration. Methods After partial hepatectomy (PH) or transfection, we measured the changes of miR-183 and programmed cell death protein 6 (PDCD6) levels in rats and the hepatocytes. The histopathology was observed with hematoxylin–eosin staining. The miR-183 mimic and inhibitor plasmids were intravenously injected into rats, and the liver weight/body weight ratio was calculated. The prediction of TargetScan and the validation of luciferase activity assay were employed to confirm the targeting relationship between miR-183 and PDCD6. The viability, apoptosis and cell cycle of transfected rat hepatocyte BRL-3A were determined via MTT and flow cytometry assays. Results MiR-183 expression showed a contrary tendency with that of PDCD6 during liver regeneration. Enhanced miR-183 in rats could notably increase liver/body weight ratio, while its inhibition did conversely. Overexpressed PDCD6, a target of miR-183, repressed the viability and cell cycle in hepatocytes, whereas its silence led to contrary results. Overexpressed miR-183 in BRL-3A cells enhanced cell viability and promoted the cell cycle yet suppressed apoptosis, whereas its inhibition showed contrary results, which were offset by PDCD6. Conclusions Collectively, miR-183 promoted liver regeneration via targeting PDCD6.
