Carcinoembryonic antigen (CEA) is an important cancer marker that plays a significant role in achieving low-cost, rapid and highly sensitive clinical detection. In this work, we developed a disposable electrochemiluminescence (ECL) sensor chip based on a screen-printed electrode (SPE) for detecting CEA via ECL technology. An amino-modified Ti3C2 MXene was used as a carrier to successfully prepare a highly efficient ECL probe AuNPs-Ru-Arg@NH2-Ti3C2-MXene by loading with AuNPs-Arg through covalent links and modifying with a ruthenium complex. Upon the addition of CEA, the ECL signal decreased significantly with the increase of CEA, due to the formation of immune complexes at the interface of the electrode. The sensing chip was used to detect CEA in an aqueous solution and found to have a detection limit of 1.5 pg mL−1. The chip was used to determine CEA in the serum of healthy humans and cancer patients, and the results were consistent with those obtained using ELISA. The disposable ECL sensor chip has many advantages including convenience, rapid detection, low cost and easy mass production; thus it has great application potential in clinical cancer diagnosis.

An ultraviolet light emitting diode (UV-LED) array chip as the irradiation source for nano-TiO2 catalyzed photochemical vapor generation (PCVG) was combined with a hollow electrode point discharge microplasma optical emission spectrometer (HEPD-OES) to realize instrumental miniaturization, and utilized for quantitative analysis of trace selenium (Se). Under the irradiation of the UV-LED array chip, Se(IV) and Se(VI) could be effectively converted into volatile species in a miniature quartz coil coating with nano-TiO2 on its inner wall as a photocatalyst. Meanwhile, volatile analyte species were totally guided into the hollow electrode and transported into the inside of micro-discharge plasma for excitation and enhanced sensitivity. Experimental conditions of PCVG and HEPD-OES were investigated in detail using an analytical line of Se at 203.98 nm through the peak area integrated for quantitation. Under optimized experimental conditions of 0.6 mL sample volume, 10% (v/v) acetic acid, 370 nm and 50 W LED, and 75 s UV irradiation, Se(IV) and Se(VI) could be detected with the same LODs, 3.9 μg L−1 and 4.1 μg L−1, respectively. The accuracy of the proposed method was validated by the analysis of the Certified Reference Materials (CRMs, GBW(E)080395 and BWB2427-2016) and real water samples. With the advantages of high compatibility, high efficiency, small size and low power consumption, the integration of UV-LED array chip based PCVG and HEPD-OES would be beneficial to fabricate miniaturized and portable devices for field analysis.

A new technique for simultaneous determination of tetracycline (TC), chlortetracycline (CTC), doxycycline (DC) in egg samples was developed by using radiant-type multi-molecularly imprinted solid-phase extraction (MISPE)-PDMS/glass array chip coated magnetic graphene oxide (Fe 3 O 4 /GO). Three kinds of molecularly imprinted polymers coated Fe 3 O 4 /GO (Fe 3 O 4 /GO-MIPs) were synthesized by in situ polymerization using TC, CTC and DC as single templates, respectively. The Fe 3 O 4 /GO-MISPE monolithic columns exhibited high affinity and specificity to TC, CTC and DC, as detected by high performance liquid chromatography coupled with fluorescence detection. Moreover, the radiant-type array extraction chip showed excellent adsorption capacity and particular selectivity to TC, CTC and DC, with the enrichment factors reaching 86–104 folds. Fe 3 O 4 /GO-MIPs were tested by scanning electron microscopy and Fourier transform-infrared spectroscopy. The limit of detection for TC, CTC and DC was 3.0–5.5 μg kg − 1 . Therefore, the chip-based imprinted monolithic array columns are promising for facile extraction of TC, CTC and DC from egg samples.

A desirable lanthanide-based ratiometric fluorescent probe was designed and integrated into a self-designed Fermat spiral microfluidic chip (FS-MC) for the automated determination of a unique bacterial endospore biomarker, dipicolinic acid (DPA), with high selectivity and sensitivity. Here, a blue emission wavelength at 425 nm was generated in the Fermat spiral structure by mixing the europium (Eu 3+ ) and luminol to form the Eu 3+ /Luminol sensing probe. DPA in the reservoir can be used to specifically bind to Eu 3+ under the negative pressure and transfer energy from DPA to Eu 3+ sequentially via an antenna effect, thus resulting in a significant increase in the red fluorescence emission peak at 615 nm. According to the fluorescence intensity ratio (F615/F425), a good linearity can be obtained with increasing the concentration of DPA from 0 to 200 μM with a limit of detection as low as 10.11 nM. Interestingly, the designed FS-MC can achieve rapid detection of DPA in only 1 min, reducing detection time and improving sensitivity. Furthermore, a self-designed device integrated with the FS-MC and a smartphone color picker APP was employed for the rapid automatic point-of-care testing (POCT) of DPA in the field, simplifying complex processes and reducing testing times, thus confirming the great promise of this ready-to-use measurement platform for in situ inspection.

Early detection of biomarkers has a crucial role in preventing blindness caused by diabetic retinopathy (DR). In this work, we developed a rapid, highly sensitive method for the simultaneous detection of DR-related biomarkers (miR-21 and miR-152) based on an Au nano-hexagon (AuNHs) integrated microarray chip combined with a competitive recognition strategy. The nucleic acid aptamer-modified AuNHs array served as capture substrate, while Au nano-trisoctahedrons (AuNTs) with complementary strands of nucleic acid aptamers mounted on their surface and Raman signaling molecules served as SERS probes (AuNTs@4-MBA@H1 and AuNTs@DTNB@H2). SERS probes carrying Raman signaling molecules (4-MBA and DTNB) were displaced by the target miRNAs in the experiments because they had a higher binding affinity to nucleic acid aptamers than their structural analogs (H1 and H2). This approach led to a notable decrease in the Raman signal. The competitive recognition strategy was straightforward and useful, and required only a one-step dropwise reaction to detect both miRNAs simultaneously and effectively. The microarray chip designed accordingly had the advantages of sensitive, rapid and high-throughput detection, and the detection time was only 8 min. The limits of detection (LOD) for miR-21 and miR-152 were as low as 2.76 × 10−14 M and 1.94 × 10−14 M, respectively. The utility of the method in complex systems was verified by detecting miRNAs in the serum of DR rats, and the accuracy of the method was verified against the detection results of a RT-qPCR method. This method provides a new idea and manner for the detection of miRNAs in serum, which is crucial for the early diagnosis of DR.

A sensitive and selective on chip magnetic immunoassay method, based on a sandwich-type immunoreaction with PbS nanoparticle (NPs) labels in combination with electrothermal vaporization-inductively coupled plasma mass spectrometry (ETV-ICP-MS), was proposed for the determination of carcinoembryonic antigen (CEA). We designed and fabricated a microfluidic chip for magnetic immunoassay, and the prepared iminodiacetic acid modified silica coated magnetic nanoparticles (IDA-SCMNPs) were packed into the central microchannel to form a solid phase column by self-assembly under the magnetic field. After completion of the immunoreaction involving a primary antibody, CEA and a secondary antibody labeled with PbS NPs on a magnetic solid phase packed-column, ETV-ICP-MS was used to determine the concentration of Pb that was released from the captured PbS NPs using an acid-dissolution step. The concentrations of CEA can be correlated with that of Pb. The established method demonstrated a limit of detection of 0.058 μg L−1 for CEA, with a relative standard deviation (RSD) of 6.7% (c = 10 μg L−1, n = 7). A linearity ranging from 0.2 μg L−1 to 50 μg L−1 and a 2-fold enrichment factor (from 60 μL sample solution to 30 μL eluent) were achieved. The proposed method was further validated by analyzing CEA in human serum. The results were in good agreement with those obtained by chemiluminescent immunoassay, which is currently used as a clinical method. Overall, this method offers the advantages of high speed, high sensitivity, high selectivity, low sample/reagents consumption, high integrity and versatility. Moreover, it can be easily applied to other biological and medical assays.

Plasma extracellular vesicles (EVs) have emerged as promising biomarkers for the early diagnosis of glioblastoma. However, rapid and efficient isolation and detection of tumor-specific EV subsets from plasma remains challenging. To address this, we developed an innovative integrated chip based on self-sacrificial metal–organic frameworks and the epidermal growth factor receptor (EGFR) aptamers called ZIF-8@EGFR aptamer (ZIF-8@Apt), which further coupled with a “nano-fuels”-driven DNAzyme amplification strategy. The core functionality of the constructed chip lies in its ability to capture EVs with the ZIF-8@Apt module. Once captured, numerous DNA “nano-fuel” structures are in situ assembled on the EVs’ surfaces. Subsequently, the ZIF-8@Apt@EV@nano-fuels composites are lysed to release Zn 2+ , the EVs’ nucleic acids, and additional nano-fuels for subsequent detection. Finally, a DNAzyme detection system is introduced, where the released nano-fuels activate the DNAzyme, leading to multiple signal amplification aided by Zn 2+ for highly sensitive detection of plasma EVs. This integrated approach effectively enriches and detects target EVs, achieving a significantly enhanced detection sensitivity of 500 particles/μL. In a clinical study involving glioblastoma patients, our chip demonstrated a remarkable ability to distinguish between cancer patients and healthy individuals, achieving an area under the curve (AUC) of 0.958. Moreover, the response signals of the chip can be conveniently read using fluorescence spectroscopy or microscope, offering a rapid, highly sensitive, and high-throughput analysis of plasma EVs for various application scenarios. Overall, this study introduces a comprehensive approach for the rapid isolation and accurate analysis of plasma EVs, holding significant promise for early diagnosis and disease monitoring.

Heavy metal ions pollution is one of the most threatened issues for human health and ecosystem balance. Herein, a promising microfluidic chip with microgel arrays for realizing the simultaneously point of care test to multiple pathogenic heavy metal ions was designed, which consisted of a sample perfusion area in the center (pipe width 200 μm), and four detection chambers (width 8000 μm, length 20000 μm) with microcolumn arrays (radius 100 μm, width 550 μm), all the heights were 100 μm. With simulation of the fluid flow, the flow rate of 5 mL/min made the microgel successfully patterning the precursor in microcolumn. By utilizing different kinds of quantum dots microgels, this chip could achieve specific and on-site quick detection for Pb 2+ , Co 2+ , Cu 2+ and Fe 3+ . Moreover, the gel composition had almost no effect on the performance of quantum dots and owned enough space after losing water to absorb the sample solution for reacting with the analytes. Furthermore, this POCT microfluidic chip was applied in the detection of actual water samples, and the fluorescence quenching of the chip was visible to naked eyes. It possessed the potential to achieve the ions quantitative determination. Therefore, this method realized the simultaneous detection of multiple pathogenic heavy meatal ions rapidly and availably and could further applied in actual water pollution samples with enormous advantages.

Hepatic organoid transplantation is hypothesized to achieve maximal therapeutic benefit when vascular integration coincides with peak metabolic function. However, conventional endpoint-based evaluation methods cannot dynamically track these processes in real time, limiting precise determination of the optimal transplantation window. To address this challenge, we developed a biosensor-integrated organoid-on-a-chip. This platform enables simultaneous, real-time monitoring of vascularization through nitric oxide (NO) and hepatic metabolic function through urea. Under controlled flow conditions, the biosensor-integrated organoid-on-a-chip revealed that flow-induced mechanostimulation synchronizes endothelial and hepatocyte maturation. The biosensors integrated chip further identified a critical day-5 inflection point, corresponding to maximal biomarker levels and defining the optimal transplantation window. Transplantation at this window restored approximately 93% of liver function and lobular architecture in cirrhotic mice, whereas grafts transplanted before or after this window showed 50–70% lower therapeutic efficacy. These findings validate the transplantation window hypothesis and provide a real-time, quantitative framework for quality control in organoid-based regenerative therapies.

Nowadays, cancer researches widely employed organoids as research model. Organoids provide a realistic applicate scene but how to real-time detect and quantified the organoid status is still a bottleneck. To bridge this knowledge gap, we developed an all-in-one microfluidic platform that, for the first time, seamlessly integrates a vascularized tumor organoids-on-a-chip model (VOoC) with a real-time ELISA detection module for advanced tumor research and point of care testing (POCT). The platform features a unique integration of two cutting-edge technologies: (1) VOoC consists of a perfusable self-forming vessel network throughout the tumor organoid, emulates in vivo tumor angiogenesis, and (2) a real-time microfluidic diagnostic chip equipped with ELISA for simultaneously monitoring of tumor biomarker expression levels. We demonstrated the platform's capabilities using cervical cancer organoids and evaluated the drug efficacy of cisplatin and bevacizumab. This coupled system enables simultaneous tumor microenvironment modeling and continuous, real-time monitoring of secreted biomarkers, a capability not achievable with conventional disconnected systems. By combining these two systems, our dual-effect bimodal chip platform holds great promise for advancing personalized medicine and cancer research, offering a powerful tool for both drug screening and quick diagnosis.

By combining the distinctive noninvasive feature with the peculiar complete functional implementation trait, fully integrated raw noninvasive biofluid glucose biosensors offer active and remote glucose monitoring while posing minimal harm or infection risks compared to the traditional invasive manner. However, each previously reported fully integrated raw noninvasive biofluid glucose biosensor is solely focused on single-type raw noninvasive biofluid analysis. Given the diversity and complexity of subjects’ physical conditions, single-type raw noninvasive biofluids are inappropriate to all crowds (e.g., sweat collection/analysis could be inapplicable for dermatopathic subjects). Here, we demonstrate the first example of a universal fully integrated nanoelectronic system with the unique capability to point-of-care and universally monitor diverse raw noninvasive biofluid (i.e., sweat, tears, and saliva) glucose by combining a flexible and disposable microfluidic enzymatic biosensor (named iezSlice) for raw biofluid pump-free sampling and measurement with a customized, handheld, and reusable wireless electronic device (named iezBar) for electrical signal transduction, conditioning, processing, and wireless transmission. We employed the specially designed high-concentration-buffer powder-loaded Kimwipes (HBP-KWs) as the microfluidic channel (microchannel) of iezSlice, guaranteeing a high-accuracy glucose analysis in various raw noninvasive biofluids. We also evaluated the potential utility of the universal fully integrated nanoelectronic system for noninvasive glucose management in healthy and diabetic subjects with the assistance of the proposed volatility-derived blood glucose concentration-free protocol. Although we focus on raw noninvasive biofluid glucose analysis in this work, the universal fully integrated nanoelectronic system may readily realize accurate monitoring of various biomolecules in raw noninvasive biofluids by introducing corresponding bioreceptors.

Shrimp quality deteriorates during storage, making the assessment of freshness vital in the food industry. In this study, a spoilage gas-responsive chip was fabricated to integrate hyperspectral imaging and Raman spectroscopy, enabling the construction of a multimodal nondestructive detection model based on a multi-level data fusion strategy. Three different levels of fusion strategies were analyzed and compared, namely raw-data spectrum fusion, refined-feature spectrum fusion, and cross-model decision fusion. The results demonstrated that the cross-model decision fusion strategy employing adaptive Bayesian weighting for decision-making achieved excellent performance, with Rp 2 , RMSEP, and RPD values of 0.9911, 0.9056, and 10.3730, respectively. In subsequent external validation, the Rv 2 and RMSEV were 0.9352 and 2.1479, respectively, demonstrating significant improvement over the single-modal model and the spectrum fusion models. This study offers a promising approach for rapid, nondestructive freshness detection during shrimp storage.
