Regarded as one of the hallmarks of tumorigenesis and tumor progression, the evasion of apoptotic cell death would also account for treatment resistance or failure during cancer therapy. In this study, a Ca2+/Cu2+ dual-ion “nano trap” to effectively avoid cell apoptosis evasion by synchronously inducing paraptosis together with apoptosis was successfully designed and fabricated for breast cancer treatment. In brief, disulfiram (DSF)-loaded amorphous calcium carbonate nanoparticles (NPs) were fabricated via a gas diffusion method. Further on, the Cu2+-tannic acid metal phenolic network was embedded onto the NPs surface by self-assembling, followed by mDSPE-PEG/lipid capping to form the DSF-loaded Ca2+/Cu2+ dual-ions “nano trap”. The as-prepared nanotrap would undergo acid-triggered biodegradation upon being endocytosed by tumor cells within the lysosome for Ca2+, Cu2+, and DSF releasing simultaneously. The released Ca2+ could cause mitochondrial calcium overload and lead to hydrogen peroxide (H2O2) overexpression. Meanwhile, Ca2+/reactive oxygen species-associated mitochondrial dysfunction would lead to paraptosis cell death. Most importantly, cell paraptosis could be further induced and strengthened by the toxic dithiocarbamate (DTC)–copper complexes formed by the Cu2+ combined with the DTC, the metabolic products of DSF. Additionally, the released Cu2+ will be reduced by intracellular glutathione to generate Cu+, which can catalyze the H2O2 to produce a toxic hydroxyl radical by a Cu+-mediated Fenton-like reaction for inducing cell apoptosis. Both in vitro cellular assays and in vivo antitumor evaluations confirmed the cancer therapeutic efficiency by the dual ion nano trap. This study can broaden the cognition scope of dual-ion-mediated paraptosis together with apoptosis via a multifunctional nanoplatform.

The levels of Cu 2+ and alkaline phosphatase (ALP) are the important indicators of the developed stage of the relative diseases. Herein, a binary ratio-fluorescent and smartphone-assisted visual strategy basing on 4′-aminomethyl-4, 5′, 8-trimethylpsoralen (AMT) and the oxidation of o-phenylenediamine was developed. Under the action of Cu 2+ , the fluorescent molecule, 3-diaminophenazine (DAP) formed which can act as a fluorescent acceptor of the ratio-fluorescent sensor. The emission spectrum of AMT overlapped with the excitation spectrum of DAP and, thus, it can act as the fluorescent donor of the ratio-fluorescent sensor. With the increasing concentration of Cu 2+ and ALP, the fluorescent intensity of AMT decreased and the fluorescent intensity of DAP increased. The dual-emission reverse change ratio-fluorescent sensor realized the sensitive detection Cu 2+ and ALP with the detection limits of 2 nM and 0.03 U/mL, respectively. In addition, the acceptable recoveries were obtained when the Cu 2+ and ALP in spiked samples were detected. Furthermore, the relative activity of ALP was assessed by increasing the concentrations of the inhibitor Na 3 VO 4 and IC 50 of 25 μM was obtained. Importantly, the target concentration-dependent color change of DAP allowed us to utilize R/B ratio values to design the smartphone-assisting visual detection model of Cu 2+ and ALP activity with the detection limits of 0.1 μM and 0.18 U/mL. This simple, flexible, dual-mode sensor strategy has a potential for disease diagnosis and drug screening. Graphical abstract

Trypsin is a serine protease playing a key role in regulating pancreatic exocrine function and can be applied as a marker for the diagnosis of pancreatitis. In this work, a convenient and sensitive fluorescent assay was developed toward trypsin. Hydrogen peroxide slowly oxidized a non-fluorescent o-phenylenediamine (OPD) into a fluorescent product 2,3-diaminophenothiazine (DAP) under the catalytic from copper ions. After the introduction of bovine serum albumin (BSA), the combination of BSA with copper ions formed a peroxidase mimic and significantly accelerated the reaction rate. As an efficient protease, trypsin cleaved the lysine and arginine residues in BSA. This destroyed the binding between Cu 2+ and BSA, and brought in a reduction of the catalytic effect. The accompanying decrease in fluorescence provided a response to trypsin in the range of 0.01–600 ng/mL, with a detection limit of 0.007 ng/mL. The scheme had a good selectivity and was successfully applied to the detection of real samples.

Developing a fluorescence sensor capable of precisely identifying highly toxic nerve agents has become a focus of research, given that they are easily available and can be used by terrorists to threaten the security of the world. In this work, we designed a ratiometric fluorescence sensing platform with Cu 2+ -modified UiO-66-NH 2 (Cu 2+ @UiO-66-NH 2 ) and o-phenenediamine (OPD) as signal indicators and successfully used for the liquid and gas detection of the nerve agent simulant diethyl chlorophosphate (DCP). UiO-66-NH 2 obtained oxidase activity by simple Cu 2+ modification, which catalyzed the oxidation of OPD to give a yellow fluorescent product 2, 3-diaminophenazine (DAP). In this detection system, the fluorescence of UiO-66-NH 2 was greatly enhanced by blocking the ligand metal charge transfer (LMCT) process inside the framework by DCP. At the same time, the amino group of DAP was phosphorylated by DCP, and the attenuation of DAP fluorescence was observed by the static quenching effect. In addition, the Cu 2+ @UiO-66-NH 2 /OPD loaded agarose hydrogel device can be used to detect DCP vapor with naked eyes and showed satisfactory detection result (LOD: 6.65 ppb).

A novel 3-dimensional graphene oxide/2-mercaptothiazoline (3D GO-MT) aerogel was prepared by using GO and MT which contains heteroatoms (nitrogen-, sulfur-) through a facile hydrothermal assembly process and a following freeze-drying technology . The effects of hydrothermal reaction temperature and GO-to-MT mass ratio on the physicochemical properties of the composites were investigated. The removal of Cu 2+ from aqueous solution by 3D GO-MT aerogels was evaluated. The results showed that the composite (GO-MT 1:3/100 °C ) with GO-to-MT mass ratio of 1:3 prepared at 100 °C had the highest adsorption capacity for Cu 2+ (0.5299 mmol g −1 ). In a bicomponent system of Cu 2+ /Pb 2+ , GO-MT 1:3/100 °C exhibited higher adsorption selectivity for Cu 2+ . And the distribution coefficients K of GO-MT 1:3/100 ° C for Cu 2+ /Y 3+ , Cu 2+ /Nd 3+ in the bicomponent systems were 0.6224 and 0.1253, respectively. The morphology and composition of GO-MT 1:3/100 °C , as well as the adsorption mechanism for Cu 2+ were investigated by scanning electron microscopy, Brunauer-Emmett-Teller, elemental analysis, Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy. The presence of nitrogen and sulfur on MT heterocycle promoted the possible assembly of MT with GO through π-π stacking and hydrogen bonding interactions. Additionally, the existence of these heteroatoms in MT dominated the metal-chelate affinity of 3D GO-MT aerogel for selective adsorption of Cu 2+ . The fitted data of kinetics and thermodynamics showed that the adsorption process followed linear pseudo-second-order kinetics and linear Langmuir isotherm model. The adsorption process is non spontaneous and endothermic. The adsorption mechanism of GO-MT 1:3/100 °C for Cu 2+ involves coordination, cation-π electron interaction, electrostatic attraction and other intermolecular interactions. 3D GO-MT 1:3/100 °C can be used as an adsorbent material for selective adsorption/enrichment of Cu 2+ in aqueous solution.

Development of sustainable biomaterials from wasted resources has been widely advocated recently. In this study, chitin was extracted from Hericium erinaceus residues and used as the main framework to in situ synthesize series of magnetic chitin hydrogels for Cu 2+ removal. Results showed that the extracted chitin with 1.56 × 10 5 g/mol molecular weight and 51.83% degree of acetylation was completely composed of glucosamine. After introduction of Fe 3 O 4 particles, the magnetic chitin hydrogels exhibited preferable adsorption capacity for Cu 2+ than its native form. Besides, the magnetic chitin hydrogels showed paramagnetic behavior and improved mechanical property but the slight reduced swelling and thermal performance. Moreover, the swelling, mechanical, magnetic and adsorption properties were found to be Fe 3 O 4 content-dependent, indicating its specific tunable performance. Therefore, this work provided a new alternative renewable resource for chitin and facile procedure to prepare magnetic chitin hydrogels as a promising candidate material for heavy metal removal system.

A novel label-free fluorescent sensor for histidine was developed based upon Cu 2+ -specific DNAzyme, hybridization chain reaction(HCR) and triplex DNA . Cu 2+ can bind to the histidine, in the presence of histidine, leading to the inhibition of the cleavage of substrate strand of Cu 2+ -dependent DNAzyme, then the intact substrate strand trigger the HCR between H1 and H2. The HCR product can be recognized by triplex-forming oligonucleotide (TFO) through triplex formation and reported by the fluorescence of berberine , the fluorescence intensity of the sensing system was proportional to the concentration of histidine during the range of 5.7–455 nmol L −1 , with a detection limit of 2.0 nmol L −1 .

The sensitive and rapid quantification of lysozymes (Lys) is a critical goal in clinical practice. Herein, a novel, label-free and sensitive turn ON–OFF chemiluminescent (CL) probe for Lys was developed based on modulating the biocatalytic ability of the Cu2−xSe nanoparticles. The synthesized Cu2−xSe nanoparticles could significantly enhance the chemiluminescence signal of the luminol–H2O2 system owing to their high copper deficiency, where a very bright blue CL emission was directly observed by the naked eye in darkness and it could last for a long period of up to 36 minutes. However, Lys could induce the aggregation of the Cu2−xSe nanoparticles due to the electrostatic attraction, which could be monitored by SEM and dark-field imaging. The aggregated Cu2−xSe nanoparticles with lower copper deficiency density could suppress their catalytic ability and turn off the chemiluminescence of luminol–H2O2. The established ON–OFF CL strategy exhibited a selective response to protein Lys and showed a quantitative relationship between the changes in the CL signals and the concentration of Lys over a wide range of 0.5–5000 ng mL−1, with a low detection limit of 0.36 ng mL−1. This study provides a promising strategy for detecting biomolecules by exploiting their specific interactions with nanomaterials.

Three enhanced fluorescence probes based on Rhodamine B-Schiff base structure were synthesized for detecting Cu 2+ . The corresponding detection limits were found to be 0.25 μM, 0.15 μM and 0.18 μM. Binding ratio and binding sites were determined by Job's and nuclear magnetic titration experiments. The binding constants obtained by the Benesi-Hildebrand equation to be 341.0 M −0.5 ,1.8 × 10 4 M −1 , and 265.4 M −0.5 , respectively. As isomers, the different effects of probes on Cu 2+ detection were researched. By adjusting the position and the size of the substituent group, the effects of binding sites and steric hindrance on the complexation ratio, response time and detection limit were discussed. Optimal spatial combination structure with Cu 2+ was obtained through energy calculation. Detection mechanism of Rhodamine B ring opening based on the complex of the Schiff base with Cu 2+ was confirmed. E. coli staining and detection of real water samples had expanded their applications.

In this work, we proposed a convenient fluorescent assay for the detection of quinolones based on their inhibition towards the oxidation of o-phenylenediamine (OPD) by copper ions (Cu2+). Free Cu2+ can oxidize OPD into fluorescent 2,3-diaminophenzine followed by the formation of copper nanoparticles (CuNPs), which would further catalyze the oxidation of OPD. When quinolones were added, their strong binding affinity to Cu2+ inhibited the oxidase-like activity of Cu2+. So, the fluorescence intensity of the Cu2+–OPD system decreased obviously. The determination of quinolones can be achieved by using this fluorimetric assay without the use of complicated instruments and protocols. As demonstrated, this assay offered a highly sensitive readout for quinolones including nalidixic acid, cinoxacin, ciprofloxacin, and moxifloxacin in the linear ranges of 1.0–100 nM, 0.5–100 nM, 1.0–100 nM, and 0.1–100 nM, respectively. In addition, the developed assay was successfully applied to the direct analysis of quinolones in real amples.

Stimuli-responsive nanomedicines have become a recent research focus as a candidate for cancer treatment because of their effectiveness, sensibility, and minimal invasiveness. In this work, a novel nanosystem is developed based on Cu 2− x S@MnS core–shell nanoparticles (CSNPs) in which the Cu 2− x S core serves as a photosensitizer to generate hyperthermia and reactive oxygen species (ROS), and the MnS shell is used in H 2 O 2 -responsive O 2 production. Cu 2 −x S@MnS CSNPs with an independent core and shell ratio are synthesized by a controllable hot-injection method, resulting in an optimal photothermal (PT) effect with a PT conversion efficiency of up to 47.9%. An enhanced photodynamic (PD) effect also occurs in an H 2 O 2 environment. More significantly, in vivo experiments demonstrate that Cu 2 −x S@MnS CSNPs can mediate tumor shrinkage in both HeLa tumor cell line-derived xenograft (CDX) and head and neck squamous cell carcinoma (HNSCC) patient-derived xenograft (PDX) models, with the capability of being used as a T1-enhanced magnetic resonance (MR) contrast agent. These results suggest the great potential of as-prepared Cu 2 −x S@MnS CSNPs as photo/H 2 O 2 -responsive therapeutic-agents against tumors, even in a complicated and heterogeneous environment, thus promoting the clinical translation of nanomedicine.

Synthesis and engineering of nanostructures with multiple components for the acceptable biosensing platform is a hot topic research. Herein, flower-like nonstoichiometric copper chalcogenides nanocrystals (Cu 2− x S y Se 1− y NCs) loaded with rhodamine 6G (R6G) were used as a selective and sensitive off-on fluorescent probe for biothiols based on inner filter effect (IFE). The Cu 2 −x S y Se 1− y showed the strong interband transition band in the UV–vis region and broad localized surface plasmon resonance (LSPR) in the near-infrared region which exhibited good light-harvesting features, and thus can show sufficient absorption of the excitation or emission light of fluorescent R6G with high fluorescence quenching ability. When the biothiols (glutathione, homocysteine and cysteine) were added, the interband transition bands of Cu 2− x S y Se 1− y NCs were suppressed, resulting in the fluorescence of R6G switched on. There was a good linear relationship between fluorescent recovery of R6G and the concentrations of glutathione, homocysteine and cysteine over the range of 1.0–150 μM, 1.0–200 μM, 1.0–150 μM with a detection limit of 32.0 nM, 34.0 nM and 31.0 nM, respectively. The established strategy exhibited the selective response to biothiol compounds and was also been applied for detecting biothiols in the human serum samples.
