Scallop ( Patinopecten yessoensis ) male gonad hydrolysates (SMGHs)/ κ -carrageenan (KC)/KCl beads with SMGHs :KC ratios (0:10–5:5) were investigated. SMGHs/KC/KCl-Cur bead (5:5) exhibited the most intact spherical morphology and highest Cur loading content of 0.063 mg/0.1 g bead, ascribing to a shortened T 23 from 1607.9 to 966.4 ms, and red and blueshifts of O H, NH, amide I and II bands. The undetected fingerprint region within 7.82°-28.90° of SMGHs/KC/KCl-Cur beads indicated successful Cur entrapment. Moreover, SMGHs/KC/KCl-Cur beads exhibited shrinkage network backbones and larger void pores as SMGHs increased, with vessel percentage area, total number of junctions, total vessel length decreasing from 52.1, 1446.8, 57931.4 to 39.7, 530.5, 34458.4, and lacunarity increasing from 0.048 to 0.111, respectively. Furthermore, Cur showed approximately 50% release contents in colon phase and above 90% retention rate during 30 days of storage at 4 °C. These results suggested that SMGHs/KC/KCl-Cur beads exhibited sustained-release of Cur and promised stable Cur preservation.

The research of the interaction between proteins and functional active substances in the protein nanoparticles is a research hotspot in the field of food science . Gelatin type had a significant effect on the structural and functional properties of gelatins. Herein, the interaction mechanism of curcumin with four types of gelatins (bovine bone gelatin, BBG; cold water fish skin gelatin, CFG; fish gelatin , FG; and porcine skin gelatin, PSG) and the structural and functional properties of gelatin/curcumin nanoparticles were explored. All four types of gelatins could be applied to prepare gelatin/curcumin nanoparticles with an amorphous state of curcumin. The nanoparticle sizes were not dependent on the molecular weight of gelatins. The stability of gelatin/curcumin nanoparticle solutions was dependent not on the nanoparticle size but on the gelatin type. The binding affinity between gelatins and curcumin was dependent on gelatin type and the number of binding sites was nearly 1. The main binding forces to form gelatin-curcumin nanoparticles were hydrogen bonds, Pi-alkyl interaction, and Pi-anion interaction. The nanoparticulation increased the surface hydrophobicity of BBG, FG, and PSG, whereas it decreased the surface hydrophobicity of CFG. The nanoparticulation increased the surface tension of gelatins and decreased the dynamic interfacial tension of gelatins. The functional properties were dependent on both gelatin type and nanoparticulation. CFG/Cur nanoparticulation could not change the creaming stability, whereas other gelatins/curcumin nanoparticulation significantly increased creaming stability than the corresponding gelatins. This work provided the basic knowledge to illustrate the interaction and application of protein nanoparticles for Pickering emulsion stabilization.

Curcumin is a hydrophobic substance with various beneficial biological activities. Its weak solubility and instability, however, now restrict its usage as a functional food. Herein, the purpose of this work is to fabricate curcumin-loaded bovine serum albumin/carboxymethylpachymaran nanoparticles (BMCNPs) to overcome these challenges. Under optimal conditions, the BMCNPs were spherically shaped with a surface charge of −31.4 mV, polydispersity index of 0.16 and particle size of 158.3 nm were characterized by electron microscopy and dynamic light scattering. The interactions between curcumin, bovine serum albumin (BSA), and carboxymethylpachymaran (CMP) were investigated. BMCNPs had a much better curcumin encapsulation efficiency (74.1%) as well as strong antioxidant activity, storage stability, and ionic strength tolerance. The in vitro release revealed that curcumin was more successfully sustained in the small intestine stage (68.7%) and that BMCNPs were able to prevent it from releasing too early in the stomach (21.4%). Not to mention, the results of the cytotoxicity test performed on the HT-29 and IEC-6 cell lines demonstrated the acceptable biocompatibility of BMCNPs.

Nanocarrier-delivered bioactive compounds are highly desirable for their improved stability and applicability, but their bioavailability is still limited due to the strong mucus and epithelial cell barriers. Herein, a series of self-assembled soy protein nanoparticles (SPNPs) with different mucus permeabilities were prepared and their delivery efficiency upon Curcumin (Cur) encapsulation was evaluated. Results demonstrated that the formed SPNPs-Cur exhibited high compatibility and cellular antioxidant accessibility. Besides, SPNPs enhanced the cellular uptake and transmembrane permeation of Cur, especially promoted the transportation of proto-Cur in addition to Cur metabolites. The SPNPs with the rapid mucus diffusion capacity presented more efficient transcytosis across the Caco-2 cell monolayer, which was mediated by a combination of paracellular and transcellular pathways. This work verified that mucus-permeable soy protein nanoparticles could be a promising delivery system for improving the bioavailability of bioactive compounds.

Curcumin is a hydrophobic polyphenol with various health benefits, but its poor water solubility and stability limit its application in the food and pharmaceutical industries. In this study, a pH-driven method was developed to prepare ovalbumin (OVA)-curcumin nanocomplexes without organic solvents, reducing the environmental impact. The results showed that curcumin was encapsulated in OVA with a high efficiency of over 95% and remained in an amorphous state within the protein molecules. Molecular docking and spectroscopic analyses revealed that curcumin was mainly bound and stabilized by hydrogen bonds and hydrophobic interactions in the hydrophobic core of OVA. The binding strength of the intermediate stage of the pH-driven method was stronger than that of the final stage, due to the exposure of OVA molecular binding sites and the involvement of electrostatic attraction. Furthermore, the potential of polysaccharide blending to enhance the stability of the nanocomplexes was investigated. The results showed that sodium alginate effectively enhanced the photostability and thermal stability of the OVA-curcumin nanocomplexes by co-solubilization and electrostatic complex formation. In addition, simulated in vitro digestion also demonstrated that the pH-driven method combined with polysaccharide blending increased the bioaccessibility of curcumin in nanocomplexes from 17.79% to 52.54%. Therefore, the pH-driven method coupled with polysaccharide blending ensures the encapsulation and stabilization of hydrophobic bioactive substances for use in their delivery systems to improve bioaccessibility.

The fabrication of bio-based epoxy resins having better intrinsic flame retardancy and mechanical performances than petroleum-based epoxy thermosets is important for alleviation of energy and health concerns. In this study, bio-based monomer of diglycidyl ether of curcumin (DGEC) was successfully synthesized from a natural and renewable curcumin through a facile one-step process. The biocompatible epoxy resin obtained was then cured with 4,4′-diaminodiphenyl sulfone (DDS) and compared with petroleum-based diglycidyl ether of bisphenol A (DGEBA). The DGEC/DDS exhibited a higher glass-transition temperature (300 °C) than DGEBA/DDS (215 °C), indicating the outstanding heat resistance. The tensile strength of DGEC/DDS was 65.9 MPa, higher than that of DGEBA/DDS (56.1 MPa). Furthermore, DGEC/DDS showed excellent charring ability with char yield of 50.15% at 800 °C in N 2 , which was approximatively 3.5 times as much as that of the cured DGEBA/DDS (14.68%). Microscale combustion calorimeter (MCC) test demonstrated that the DGEC/DDS possessed lower heat release capacity (HRC, 130 J/gk), peak heat release rate (PHRR, 115.0 W/g) and total heat release (THR, 9.9 kJ/g), which were reduced by 80.2%, 74.8% and 59.4%, respectively, compared to those of DGEBA/DDS (658 J/gk, 457.3 W/g, 24.4 kJ/g). The DGEC/DDS passed the UL-94 test and attained the V-0 rating, revealing the good flame retardancy. This work provided an opportunity to prepare bio-based epoxy resins with better properties than DGEBA, which exhibited promising applications in high heat and flame retardancy fields.

The co-amorphous (CAM) system has attracted widely attention for improving the solubility of poor water solubility drug and provide a formulation strategy for combination administration system. Paclitaxel (PTX) belongs to biopharmaceutics classification system (BCS) class Ⅳ drug, with the poor water solubility and membrane permeability. This study aimed to preparing the CAM system between PTX and curcumin (CUR) by rotary evaporation method. The solid-state characterization of CAM indicated that the PTX and CUR presented as amorphous state, compared with crystalline PTX and CUR, the amorphous system could improve the saturated solubility and dissolution with significantly difference, besides, the PTX absorption permeability increased after amorphization with CUR, and the rat pharmacokinetic study exhibited that the PTX-CUR had a high C max and AUC 0-∞ compared with crystalline CUR and PTX, Furthermore, the amorphous CUR and PTX exhibited the stronger anti-tumor efficacy in A549 cell model in vivo . In addition, PTX-CUR exhibited physical stability under the long-term storage condition. In conclusion, amorphization of PTX and CUR provided a promising mothed to improve the anti-tumor efficacy and oral bioavailability of PTX.

Background Recent efforts have been focused on combining two or more therapeutic approaches with different mechanisms to enhance antitumor therapy. Moreover, nanosize drug-delivery systems for codelivering two drugs with proapoptotic and antiangiogenic activities have exhibited great potential in efficient treatment of cancers. Methods Glycyrrhetinic acid (GA)–modified liposomes (GA LPs) for liver-targeted codelivery of curcumin (Cur) and combretastatin A4 phosphate (CA4P) were prepared and characterized. In vitro cellular uptake, cytotoxicity, cell migration, in vivo biodistribution, antitumor activity, and histopathological studies were performed. Results Compared with unmodified LPs (Cur-CA4P LPs), Cur-CA4P/GA LPs were taken up effectively by human hepatocellular carcinoma cells (BEL-7402) and showed higher cytotoxicity than free drugs. In vivo real-time near-infrared fluorescence–imaging results indicated that GA-targeted LPs increased accumulation in the tumor region. Moreover, Cur-CA4P/GA LPs showed stronger inhibition of tumor proliferation than Cur, Cur + CA4P, and Cur-CA4P LPs in vivo antitumor studies, which was also verified by H&E staining. Conclusion GA-modified LPs can serve as a promising nanocarrier for liver-targeted co-delivery of antitumor drugs against hepatocellular carcinoma.

Acute lung injury (ALI) is a serious illness without resultful therapeutic methods commonly. Recent studies indicate the importance of oxidative stress in the occurrence and development of ALI, and mitochondria targeted antioxidant has become a difficult and hot topic in the research of ALI. Therefore, a sialic acid (SA)-modified lung-targeted microsphere (MS) for ALI therapy are developed, with triphenylphosphonium cation (TPP)-modified curcumin (Cur-TPP) loaded, which could specifically target the mitochondria, increasing the effect of antioxidant. The results manifest that with the increase of microsphere, lung distribution of microsphere is also increased in murine mice, and after SA modification, the microsphere exhibits the ideal lung-targeted characteristic in ALI model mice, due to SA efficiently targeting to E-selectin expressed on inflammatory tissues. Further investigations indicate that SA/Cur-TPP/MS has better antioxidative capacity, decreases intracellular ROS generation, and increases mitochondrial membrane potential, contributing to a lower apoptosis rate in human umbilical vein endothelial cells (HUVECs) compared to H2O2 group. In vivo efficacy of SA/Cur-TPP/MS demonstrates that the inflammation has been alleviated markedly and the oxidative stress is ameliorated efficiently. Significant histological improvements by SA/Cur-TPP/MS are further proved via HE stains. In conclusion, SA/Cur-TPP/MS might act as a promising drug formulation for ALI therapy.

Background The objective of this study was to survey the therapeutic function of curcumin-encapsulated poly(gamma-benzyl l-glutamate)-poly(ethylene glycol)-poly(gammabenzyl l-glutamate) (PBLG-PEG-PBLG) (P) on diabetic cardiomyopathy (DCM) via cross regulation effect of calcium-sensing receptor (CaSR) and endogenous cystathionine-γ-lyase (CSE)/hydrogen sulfide (H 2 S). Methods Diabetic rats were preconditioned with 20 mg/kg curcumin or curcumin/P complex continuously for 8 weeks. The blood and myocardiums were collected, the level of serum H 2 S was observed, and the [Ca 2+ ] i content was measured in myocardial cells, and hematoxylin-eosin, CaSR, CSE, and calmodulin (CaM) expression were detected. Results Both curcumin and curcumin/P pretreatment alleviated pathological morphological damage of myocardium, increased H 2 S and [Ca 2+ ] i levels, and upregulated the expression of CaSR, CSE, and CaM as compared to DCM group, while curcumin/P remarkably augmented this effect. Conclusion PBLG-PEG-PBLG could improve water-solubility and bioactivity of curcumin and curcumin/PBLG-PEG-PBLG significantly alleviated diabetic cardiomyopathy.

The successful treatment of gliomas relies on two important factors, namely therapeutic genes/drugs and efficient delivery vehicles to cross the blood-brain barrier. In this work, a new gene vector was constructed based on polycaprolactone (PCL), glutathione (reduced; GSH)-sensitive polyethylenimine (PEI-SS) and polyethylene glycol (PEG), and modified with a cell-penetrating peptide (tLyp-1); this yielded tLyp-1-conjugated GSH-sensitive biodegradable micelles (PPPt) that mediated enhanced tumour necrosis factor (TNF)-related apoptosis-inducing ligand (pUNO1-hTRAILa) and curcumin (CUR) co-delivery to gliomas. The sensitisation function of CUR was used to explore whether CUR and genes have a synergistic antitumour effect. The results showed that the drug loading efficiency of PPPt was about 8.6%. The PPPt possessed good cytocompatibility and blood compatibility when the concentration was less than 0.06 mg/mL. PPPt strongly condensed pUNO1-hTRAILa to form a nanocomplex and efficiently transfected C6 cells. The drug-release behaviour of PPPt/CUR/pUNO1-hTRAILa was reduction-sensitive and the cellular uptake results indicate that modification of tLyp-1 promoted the internalisation of micelles. The PPPt/CUR/pUNO1-hTRAILa complexes exhibited much better in vitro and in vivo antitumour effects than micelles loaded with CUR or pUNO1-hTRAILa alone. We also used magnetic resonance imaging for in vivo monitoring of the transcranial drug delivery and therapeutic effect of the tumour on rats bearing in situ glioma.

Osteoporosis is an age-related metabolic disease of bone, resulting in bone pain and even bone fragility and brittle fracture . Inhibiting overactive osteoclasts while promoting osteoblast activity is an ideal way to treat osteoporosis. Previous studies have demonstrated that natural compounds , such as curcumin (Cur) have dual roles both in promoting bone formation and inhibiting bone resorption , making them promising candidates for osteoporosis treatment . However, their poor water solubility, high dosage of curative effect and significant toxicity to other organs have largely limited their clinical translations. In this study, a novel method was reported to conjugate Cur and poly(amidoamine) dendrimers (PAD) using hexachlorocyclotriphosphazene (HCCP) as the linkage through a one-pot reaction, forming stable and uniform Cur loaded nanospheres (HCCP-Cur-PAD, HCP NPs). Owing to the hydrophilicity of PAD and hydrophobicity of Cur, HCP NPs can self-assemble into nanoparticles with particle size of 138.8 ± 78.7 nm and display excellent water dispersity . The loading capacity of Cur can reach 27.2% and it can be released from HCP NPs with pH-responsiveness. In vitro experimental results demonstrated that the HCP NPs entered lysosomes by endocytosis and proved dual anti-osteoporosis effects of inhibiting osteoclasts and promoting osteoblasts.
