Background: Schisanlactone E, also known as XueTongSu (XTS), is an active compound extracted from the traditional Tujia medicine Kadsura heteroclita (“XueTong”). Recent studies highlight its anti-inflammatory and antioxidant properties, yet the mechanisms of XTS’s therapeutic effects on Alzheimer’s disease (AD) are unclear. This study aims to elucidate the therapeutic efficacy and mechanisms of XTS in AD.Methods: Ten C57BL/6 mice were assigned to the control group (NC), and twenty APP/PS1 transgenic mice were randomly divided into the model group (M) (10 mice) and the XTS treatment group (Tre) (10 mice). After an acclimatization period of 7 days, intraperitoneal injections were administered over a 60-day treatment period. The NC and M groups received saline, while the Tre group received XTS at 2 mg/kg. Learning and memory abilities were assessed using the Morris Water Maze (MWM) test. Histopathological changes were evaluated using hematoxylin and eosin (HE) and Nissl staining, and immunofluorescence was used to assess pathological products and glial cell activation. Cytokine levels (IL-1β, IL-6, TNF-α) in the hippocampus were quantified by qPCR. 16S rDNA sequencing analyzed gut microbiota metabolic alterations, and metabolomic analysis was performed on cortical samples. The KEGG database was used to analyze the regulatory mechanisms of XTS in AD treatment. Results: XTS significantly improved learning and spatial memory in APP/PS1 mice and ameliorated histopathological changes, reducing Aβ plaque aggregation and glial cell activation. XTS decreased the expression of inflammatory cytokines IL-1β, IL-6, and TNF-α. It also enhanced gut microbiota diversity, notably increasing Akkermansia species, and modulated levels of metabolites such as isosakuranetin, 5-KETE, 4-methylcatechol, and sphinganine. Pathway analysis indicated that XTS regulated carbohydrate metabolism, neuroactive ligand-receptor interactions, and alanine, aspartate, and glutamate metabolism, mitigating gut microbiota dysbiosis and metabolic disturbances. Conclusion: XTS ameliorates cognitive deficits, pathological changes, and inflammatory responses in APP/PS1 mice. It significantly modulates the gut microbiota, particularly increasing Akkermansia abundance, and influences levels of key metabolites in both the gut and brain. These findings suggest that XTS exerts anti-AD effects through the microbial-gut-brain axis (MGBA).

Cholesterol drives E-protein depth redistribution via a matrix-mediated helix tilt adjustment. • Escin-induced peptide localization shifts strictly require the presence of charged DMPS lipids. • Escin is hypothesized to modulate the viroporin conformation via localized helix kinking rather than matrix disruption. • Escin is able to extract cholesterol from lipid membrane. β-Escin is a well-known saponin derived from the horse chestnut tree. Saponins, including escin, exhibit a wide range of biological activities, among which membrane-modifying activity is particularly notable. The latter is key to understanding the molecular mechanisms underlying escin's biological effects, including its antiviral properties. For example, altering membrane properties can strongly influence the life cycle of enveloped viruses such as SARS-CoV-2 by modulating the activity of its envelope protein (E protein). In the present paper, we studied the localization of both escin and the E protein transmembrane domain (ETM) in model membranes with different lipid compositions. We demonstrated that escin can alter ETM localization in membranes containing the negatively charged lipid DMPS, and that escin is also capable of extracting cholesterol from model lipid membranes. Introduction To investigate the impact of different processing methods on the volatile components in Aglaia odorata flower extract (AOFEs). Methods Headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry and electronic nose (E-Nose) analysis were employed to characterize volatiles of extracts obtained by ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE), and heated reflux extraction (HRE). Multidimensional assessment using aroma radar charts, orthogonal partial least squares-discriminant analysis (OPLS-DA), K-means clustering, and relative odor activity value (ROAV) revealed significant processing-dependent variations. Results and discussion The results indicated that the fiber coated with DVB/CAR/PDMS had optimal extraction efficiency. A total of 46 compounds were identified, including eight alcohols, four aldehydes, one acid, 25 terpenes, seven ketones, and one heterocyclic compound. UAE and MAE had 36 and 38 compounds respectively, sharing similar compositional profiles but differing in concentrations, while HRE produced only 25 compounds Sensory evaluation and E-Nose results revealed differences in the aroma profiles of the extracts, with UAE and MAE extracts exhibiting intensified floral and sweet notes, whereas HRE displayed prominent green and spicy characteristics. K-means clustering categorized volatile evolution trends into four distinct subclasses. OPLS-DA identified 13 differential volatiles with variable importance in projection greater than 1, with ROAV analysis further selecting eight key markers, including (1R,7 S )-germacra-4(15),5,10(14)-trien-1 β -ol, α -humulene, copaene, β -cadinene, (E)- β -caryophylene, Γ -cadinene, humulene oxide II, and caryophyllene oxide. These compounds collectively contribute to the sweet and floral attributes of AOFEs. This study elucidated extraction-method-dependent volatile profiles and aroma characteristics, providing theoretical guidance for process optimization and quality enhancement in AOFEs production.

A novel phage HH3 of E. coli was isolated and characterized. • A colorimetric sensor was developed for E. coli detection within 35 min. • The biosensor shows high sensitivity and accuracy in various food and water matrices. • The biosensor exhibited a wide linear range and a low LOD of 3.2 CFU/mL. • It distinguishes viable E. coli from dead cells, beyond conventional methods. A phage HH3-functionalized Ce-UiO-66-NH₂ nanozyme-based colorimetric biosensor was developed for the quantification of viable Escherichia coli ( E. coli ), overcoming the drawbacks of slow detection and inadequate live/dead bacterial discrimination in conventional analytical methods. The proposed platform achieves target detection within 35 min, with a low limit of detection (LOD) of 3.2 CFU/mL and satisfactory spiked recoveries ranging from 82.75% to 122.06%. Mechanistically, Ce-UiO-66-NH₂ exhibits intrinsic oxidase-like activity that directly catalyzes the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to blue-colored oxTMB. The color intensity of this chromogenic reaction is positively correlated with E. coli concentration, enabling quantitative analysis. Benefiting from the specific recognition capability of phage HH3, the biosensor shows negligible responses to non-host bacteria and heat-inactivated E. coli . This distinctive advantage cannot be achieved by antibody- or PCR-based strategies. This nanozyme-phage platform outperforms most reported sensors and possesses promising potential for rapid and selective quantification of viable E. coli in complex food and environmental matrices. Wear and oxidation resistance are among the primary requirements for successful and sustainable total joint replacements (TJRs). Both of these factors are major causes of TJRs failure. To address these issues and enhance the service life of TJRs, the effects of vitamin E (VE) and vitamin E-D-sorbitol (VE-DS), which are antioxidants (AOs), on stabilizing ultrahigh-molecular-weight polyethylene (UHMWPE) are presented. These antioxidants were blended with control-UHMWPE at a concentration of 0.3 % by weight and consolidated by compression molding. Wear occurs on the surface, and restricting the crosslinking to a layer on the surface is desirable, as bulk crosslinking reduces the toughness and fatigue strength of the implant. In this study, surface chemical crosslinking of control-UHMWPE, VE-UHMWPE and VE-DS-UHMWPE with dicumyl peroxide (DCP) is presented. The crosslinked density, crystallinity, wear volume, mechanical performance, biocompatibility and oxidation stability assessed by oxidation induction time (OIT) of the UHMWPE blends were characterized through different techniques. The control-UHMWPE blend showed the highest wear resistance but the lowest OIT, while the VE-DS-UHMWPE blend displayed highest crystallinity, best mechanical performance and the highest OIT, exhibiting the synergistic effect of vitamin E and D-sorbitol on the oxidation resistance of UHMWPE.

Escherichia coli O157:H7 has been reported as an important pathogenic bacteria causing serious infection and economic loss. However, detection of Escherichia coli O157:H7 needs improvement, given its current complexity and sensitivity. Herein, we attempt to build a fluorescence sensing method to detect Escherichia coli O157:H7 with easy operation and high efficiency. The target virulence gene sequences are recognized and cleaved by the CRISPR-Cas9 system, and trigger strand displacement amplification and rolling circle amplification. After amplification reactions, massive products can hybridize with the probes, the fluorescence of which are quenched based on a metal–organic framework platform, leading to the fluorescence recovery at typical excitation/emission wavelengths of 480/518 nm. This method exhibits high sensitivity with the detection limit at 4.0 × 101 CFU mL–1 and a wide range from 1.3 × 102 CFU mL–1 to 6.5 × 104 CFU mL–1. Meanwhile, this assay also shows significant specificity and applies to practical samples with high accuracy. Therefore, our method would have great potential application in bacterial detection, food safety monitoring, or clinical diagnostics.

Surface-growing antibiotic-resistant pathogenic bacteria such as Escherichia coli and Staphylococcus aureus are emerging as a global health challenge due to dilemmas in clinical treatment. Furthermore, their pathogenesis, including increasingly serious antimicrobial resistance and biofilm formation, makes them challenging to treat by conventional therapy. Therefore, the development of novel antivirulence strategies will undoubtedly provide a path forward in combatting these resistant bacterial infections. In this regard, we developed novel biosurfactant-coated nanoparticles to combine the antiadhesive/antibiofilm properties of rhamnolipid (RHL)-coated Fe3O4 nanoparticles (NPs) with each of the p-coumaric acid (p-CoA) and gallic acid (GA) antimicrobial drugs by using the most available polymer common coatings (PVA) to expand the range of effective antibacterial drugs, as well as a mechanism for their synergistic effect via a simple method of preparation. Mechanistically, the average size of bare Fe3O4 NPs was ~15 nm, while RHL-coated Fe3O4@PVA@p-CoA/GA was about ~254 nm, with a drop in zeta potential from −18.7 mV to −34.3 mV, which helped increase stability. Our data show that RHL-Fe3O4@PVA@p-CoA/GA biosurfactant NPs can remarkably interfere with bacterial growth and significantly inhibited biofilm formation to more than 50% via downregulating IcaABCD and CsgBAC operons, which are responsible for slime layer formation and curli fimbriae production in S. aureus and E. coli, respectively. The novelty regarding the activity of RHL-Fe3O4@PVA@p-CoA/GA biosurfactant NPs reveals their potential effect as an alternative multitarget antivirulence candidate to minimize infection severity by inhibiting biofilm development. Therefore, they could be used in antibacterial coatings and wound dressings in the future.IMPORTANCE Antimicrobial resistance poses a great threat and challenge to humanity. Therefore, the search for alternative ways to target and eliminate microbes from plant, animal, and marine microorganisms is one of the world’s concerns today. Furthermore, the extraordinary capacity of S. aureus and E. coli to resist standard antibacterial drugs is the dilemma of all currently used remedies. Methicillin-resistant S. aureus (MRSA) and vancomycin-resistant S. aureus (VRSA) have become widespread, leading to no remedies being able to treat these threatening pathogens. The most widely recognized serotypes that cause severe foodborne illness are E. coli O157:H7, O26:H11, and O78:H10, and they display increasing antimicrobial resistance rates. Therefore, there is an urgent need for an effective therapy that has dual action to inhibit biofilm formation and decrease bacterial growth. In this study, the synthesized RHL-Fe3O4@PVA@p-CoA/GA biosurfactant NPs have interesting properties, making them excellent candidates for targeted drug delivery by inhibiting bacterial growth and downregulating biofilm-associated IcaABCD and CsgBAC gene loci.

Proton exchange membrane water electrolysis (PEMWE) is known as an effective and environmental-friendly method for hydrogen production. However, the process is hindered by the necessity of large loading of high-priced noble metals in the conventional approaches. To resolve this problem, a highly efficient Nafion-based catalyst and proton exchange system are developed, which only need ultralow platinum loading by direct e-beam evaporation deposition. Scanning electron microscopy is performed before and after the deposition, which reveals a very smooth morphology. By assembling the PEMWE device with above-mentioned cathode and conventional iridium black anode, the current density reaches 500 mA cm −2 with a platinum loading of only 0.00379 mg cm −2 at 1.64 V under 80 °C. A mass activity of ≈132 A mg −1 is achieved, which is one order of magnitude higher than previously reported results. Moreover, powered by a commercial silicon solar cell, it realizes a solar-to-hydrogen conversion efficiency up to 8.26%. This work opens up an avenue of fabricating PEMWE with an ultralow catalyst loading, which may have enormous potential for future industrial scale applications.

Extensive utilization and excessive accumulation of bisphenol A (BPA) has posed a serious threat to the whole ecosystem, necessitating the rational design of metal-based catalysts capable of complete mineralization of BPA via electrochemical oxidation. Considering the shortage of metals resources and ever-increasing generation of electronic waste (e-waste), converting e-waste into high-performance catalysts is highly expected to offer an economical and feasible means to achieve the degradation of BPA. To this end, CuAu bimetallic catalysts are in situ electro-codeposited on the carbon cloth (CC) support with the waste central processing unit (CPU) leachate as electrolyte. Significantly, the recovered CuAu/CC composites can be directly applied as an efficient electrocatalyst for the decontamination of BPA via the electro-Fenton process. The electrocatalytic activities of the fabricated CuAu/CC electrodes can be regulated by tuning the reduction potentials during electro-deposition process. Under the optimized operational conditions, 99.5 % of BPA (50 mg/L) is removed within 120 min at a constant cathodic potential of −1.0 V (vs Ag/AgCl) in the CuAu/CC-mediated electro-Fenton system, accompanied by 64.2 % of total organic carbon (TOC) removal, highlighting a satisfactory performance of the BPA electrochemical oxidation. In addition, the proposed CuAu/CC electrode material can be facilely regenerated and reused for at least ten cycles without significant decline in BPA removal efficiency. Our work offers valuable guidance for upcycling of bimetallic alloy catalysts from e-waste and further application in electrochemical decontamination of organic pollutants, achieving the innovative target of “using waste to treat waste.”

A recombinantE. coli, expressing nitrilase fromAcidovorax facilis72W with dual-site expression plasmid pRSFduet (E. colipRSF-AfNit2), was constructed. It showed higher soluble expression of nitrilase than that in the pET21a plasmid. The recombinant nitrilase can efficiently catalyze the hydrolysis of 3-cyanopyridine to nicotinic acid. The whole cells ofE. colipRSF-AfNit2 were immobilized by using sodium alginate/glutaraldehyde/polyethylene imine as the best immobilized reagents. The immobilized cells showed 95% activity recovery and excellent mechanical strength, with improved thermal stability and pH stability. They also retained 82% of initial activity after nearly two months of storage at 4 °C. A semi-continuous packed-bed bioreactor (sPBR) filled with the immobilized cells was studied for efficient production of nicotinic acid. After optimization, the highest space–time yield of 1576 g/(L·d) was obtained on 0.8 M substrate concentration at 2 mL/min of flow rate. The sPBR was repeatedly operated for 41 batches, keeping 100% conversion in the presence of 30 mM CaCl2. Finally, 95 g of nicotinic acid were obtained at 90% yield after separation and purification. The developed technology has potential application value. Keywords:nitrilase;cell immobilization;nicotinic acid;reaction optimization;semi-continuous packed-bed bioreactor

‘Taro-like’ aroma is a pleasant flavor and value-added trait in pumpkin species imparted by unknown key volatile compounds. In this study, we used the electronic nose (E-nose), gas chromatography-mass spectrometry (GC–MS), and GC-Olfactometry (GC-O) to study the aroma profile, volatile compounds, and key contributors, respectively. By E-nose and GC–MS, we found significant differences in the aroma profiles and volatile compounds between fruits from five samples with/without ‘taro-like’ aroma. According to the analysis of differential volatile compounds obtained from GC–MS and the GC-O analysis of the sample with ‘taro-like’ aroma, we found that 2-acetyl-1-pyrroline representing the ‘taro’ odor was only identified in the sample with ‘taro-like’ aroma. Therefore, we conclude that 2-acetyl-1-pyrroline is the key contributor to the 'taro-like' aroma. Moreover, the relationship between 2-acetyl-1-pyrroline and ‘taro-like’ aroma was further verified via other pumpkin samples. Our results provide a theoretical basis for understanding the aroma characteristics of pumpkin fruit.

It is desired and urgently needed to improve the sensitivity of immunochromatography assay (ICA) for hazardous chemicals. In this work, an enhanced ICA strip was established and evaluated for simultaneous, semi-quantitative and quantitative detection of ochratoxin A (OTA) and aflatoxin B 1 (AFB 1 ). The signal strategy based on gold growth on the surface of the E. coli K12 carrier was recommended, which was successfully self-assembled on the enhanced ICA with a double test line pattern. When used as a novel carrier, the E. coli K12 could provide a larger surface area, better biocompatibility and high loading capacity, which was of great help to improve the performance of the ICA. By the naked eye, the semi-quantitative limit of detection (semi-Q-LOD) reached 0.03 ng/mL for both OTA and AFB 1 (17-fold and 33-fold lower than the conventional ICA strip). By the digitized strip reader, the quantitative LODs (Q-LODs) were all identified as 0.01 ng/mL for two mycotoxins (10-fold improvement), with detection ranges of 0.01–0.5 ng/mL for OTA and 0.01–0.2 ng/mL for AFB 1 . Furthermore, the high reliability and applicability of the ICA were confirmed by its good correlation with liquid chromatography-tandem mass spectrometry (LC-MS/MS). The advantages of the improved sensitivity, high efficiency and cost savings had been reflected for the ICA. This study could provide an important reference method for the sensitive, simultaneous, rapid and on-site monitoring of multicomponent contaminants.
