Biotransformation of dietary inorganic arsenic in a freshwater fish Carassius auratus and the unique association between arsenic dimethylation and oxidative damage

Di Cui,Peng Zhang,Haipu Li,Zhaoxue Zhang,Wenbao Luo,Zhaoguang Yang

The metabolic process and toxicity mechanism of dietary inorganic arsenic (iAs) in freshwater fish remain unclear to date. The present study conducted two iAs [arsenate (As(V)) and arsenite (As(III))] dietary exposures in freshwater fish crucian carp ( Carassius auratus ). The fish were fed on As supplemented artificial diets at nominal concentrations of 50 and 100 μg As(III) or As(V) g −1 (dry weight) for 10 d and 20 d. We found that the liver, kidney, spleen, and intestine of fish accumulated more As in As(V) feeding group than that in As(III), while the total As levels in muscle were similar between As(V) and As(III) group at the end of exposure. Reduction of As(V) to As(III) and oxidation of As(III) to As(V) occurred in fish fed with As(V) and As(III), respectively, indicating that toxicity of iAs was likely elevated or reduced when iAs was absorbed by fish before entering into human body through diet. Biomethylation to monomethylarsonic acid and dimethylarsinic acid and transformation to arsenocholine and arsenobetaine were also found in the fish. The linear regression analysis showed a positive correlation between secondary methylation index and the malondialdehyde content in tissues, highlighting the vital role of arsenic dimethylation in the oxidative damages in fish.

A Fluorescence Method Based on N, S-Doped Carbon Dots for Detection of Ammonia in Aquaculture Water and Freshness of Fish

Jiaran Zhang,Zeyu Xu,Ce Shi,Xinting Yang

Excessive ammonia can cause the death of fish and the eutrophication of the water environment, so ammonia detection is essential for environmental monitoring. In this study, a highly selective sensing strategy for ammonia detection based on N, S co-doped carbon dots (N, S-CDs) was developed. The as-prepared N, S-CDs exhibited excellent photoluminescence properties and fluorescent stability. N, S-CDs demonstrated fluorescence quenched in the presence of ammonia in the wide linear range of 2–80 mmol/L, and were highly selective towards ammonia over metal ions. Furthermore, a possible fluorescence quenching mechanism is proposed. N, S-CDs were further applied to detection of ammonia in aquaculture water samples and river water samples, showing good practicability with recoveries from 0.93 to 1.27 and relative standard deviations (RSDs) of 0.54% to 17.3%. N, S-CDs were also successfully used to determine the freshness of bighead carps. Keywords:carbon dots;fluorescence;ammonia;aquaculture;freshness

Effects of surfactant type and preparation pH on the droplets and emulsion forms of fish oil-loaded gelatin/surfactant-stabilized emulsions

Ting Zhang,Mengzhen Ding,Ningping Tao,Xichang Wang,Jian Zhong

The simultaneous application of proteins and surfactants is a research hotspot in the field of food emulsions . Herein, we explored the effects of surfactant type and preparation pH on the fish oil-loaded gelatin/surfactant-stabilized emulsions. The results demonstrated gelatin/Span 80-stabilized, gelatin/soybean lecithin (SL)-stabilized, and gelatin/Tween 80-stabilized emulsions consisted of three types of droplets with different sizes, whereas gelatin/sodium dodecyl sulfate (SDS)-stabilized emulsions consisted of one. The relative droplet sizes were: gelatin/Span 80-stabilized emulsions > gelatin/SL-stabilized emulsions > gelatin/Tween 80-stabilized emulsions > gelatin/SDS-stabilized emulsions. The smallest droplet size showed no significant differences in all emulsions. Preparation pH significantly decreased the other two droplet sizes of gelatin/Span 80-stabilized and the largest droplet size of gelatin/SL-stabilized emulsions, whereas had no obvious effect on the droplet sizes of the other two emulsions. The gelatin/Span 80-stabilized, gelatin/SL-stabilized, and gelatin/SDS-stabilized emulsions could be changed into emulsion gels. The gelatin/Tween 80 emulsions could be rehomoginized into liquid emulsions. These differences on the droplets and emulsion forms might be resulted from the electric and steric properties of surfactants on the water/oil interfaces at different pH. This work provides useful information for basic understanding of gelatin/surfactant-stabilized emulsions and suggests gelatin/surfactant-stabilized emulsions can be potentially applied in many types of foods .

N-3 essential fatty acids in Nile tilapia, Oreochromis niloticus: Quantification of optimum requirement of dietary linolenic acid in juvenile fish

Cuiying Chen,Baoli Sun,Xiaoxia Li,Peiyu Li,Wutai Guan,Yingzuo Bi,Qing Pan

To quantify the optimum requirement of dietary linolenic acid (18:3n-3, LNA) for Nile tilapia ( Oreochromis niloticus ), purified ethyl linolenate was added to the basal diet to provide the dietary LNA level of 0.00, 0.10, 0.32, 0.63, 0.98, 1.56 and 2.04% of dry weight, respectively. All diets contained similar amounts of 18:2n-6 (0.61% of dry weight) and 18:1n-9 from purified ethyl linoleate and oleate , respectively. Basal diet without LNA was considered as a control. After 2-week acclimation with a lipid-free diet, 4 replicate groups of 120 juvenile fish with initial body weight of 2.10   g were fed with experimental diets in a recirculated fresh water rearing system for 10   weeks. Feeding rate was 10% of body weight in the first 2   weeks, 8% in the following 4   weeks, and then gradually reduced to 5% in the next 4   weeks. Fish were fed three times daily at 8:30, 12:00 and 17:00. The results showed that fish fed the diet with 0.63% of LNA had significantly higher weight gain (WG) and specific growth rate (SGR) than fish fed the control diet ( P   <   0.05). Feed efficiency (FE) and protein deposition (PD) in fish fed diets with 0.63, 0.98, 1.56 and 2.04% of LNA were significantly higher than those of the control group ( P   <   0.05). Fish muscle fatty acid composition reflected dietary fatty acids. Increasing dietary LNA levels resulted in increased contents of LNA and 22:6n-3, but decreased contents of 20:4n-6 and 20:5n-3. When WG and FE were subjected to broken-line regression analysis, the breakpoint was achieved at dietary LNA levels of 0.45 and 0.64% of dry weight, respectively. Based on those results, the recommended optimum requirement of dietary LNA for juvenile tilapia is 0.45–0.64% of dry weight.

Simultaneous Determination of Trimethoprim and Diaveridine in Tissues of Chicken, Porcine, and Fish by Hydrophilic Interaction Liquid Chromatography–Tandem Mass Spectrometry

Luo Haoshi,Zhang Lifang,Xue Feiqun,Li Ying,Wang Xiaoyang,Fei Chenzhong,Zhang Chong

A sensitive liquid chromatography–tandem mass spectrometry method for simultaneous determination of trimethoprim and diaveridine in edible tissue samples (chicken and porcine muscle; liver, kidney, and fat tissues as well as fish muscle) was developed and validated in this study. Trimethoprim, diaveridine, and the internal standard trimethoprim-D 3 were extracted from samples using acetonitrile. The Oasis MCX Solid-Phase Extraction Cartridge was selected for cleaning up the extracts. Chromatographic separation was performed on a hydrophilic interaction liquid chromatography column with gradient elution. The analytes were determined using triple–quadrupole mass spectrometry in positive electrospray ionization and multiple reaction monitoring mode. The relative recoveries from spiked samples ranged from 82.5 to 117.2 %, with the relative standard deviations generally being below 15.4 %. Limits of detection and quantification for analytes were within 0.3–1 and 1–2 μg kg −1 , respectively. The proposed method was successfully applied to detect trimethoprim and diaveridine in real samples.

Immobilization of lipase onto N-succinyl-chitosan beads and its application in the enrichment of polyunsaturated fatty acids in fish oil

Shuai Cui,Qin-Wei Zhou,Xiao-Ling Wang,Si-Qi Yang,Kai Chen,Zhi-Yuan Dai,Yao-Wen Huang,Tao Zhou

Bovine pancreatic lipase was immobilized onto N -succinyl-chitosan beads by using N -(3-dimethylaminopropyl)- N ′-ethylcarbodiimide (EDC)/ N -hydroxysuccinimide (NHS) as the activating agent. The immobilized lipase was characterized by Fourier transform infrared spectra and scanning electron microscopy. The activity of the immobilized lipase obtained under the optimized immobilization conditions was found to reach 1,480 U/g, with enzyme activity recovery of 74.5%. Compared to the free lipase, thermal stability of the immobilized lipase was greatly improved. The reusability study indicated that the immobilized lipase retained 76.6% of its initial activity after being recycled five times. A preliminary study indicated that the prepared immobilized lipase can effectively enrich the polyunsaturated fatty acid in fish oil (increasing from 45.1 to 67.7%), of which the combined docosahexaenoic acid and eicosapentaenoic acid contents increased from 16.3 to 27.1%. Practical applications To overcome the drawbacks of using free lipase, an effective method was developed by immobilization of lipase onto N -succinyl-chitosan beads using EDC/NHS as the activating agent. The immobilized lipase prepared by this method may find application in the enrichment of polyunsaturated fatty acids of fish oil.

Sulfonated nanoparticles doped electrospun fibers with bioinspired polynorepinephrine sheath for in vivo solid-phase microextraction of pharmaceuticals in fish and vegetable

Junlang Qiu,Guosheng Chen,Fang Zhu,Gangfeng Ouyang

In this study, the biocompatible copolymer Poly(lactic acid-co-caprolactone) (PLCL) doped with sulfonated γ-Al 2 O 3 nanoparticles was used for electrospun on stainless wires. The electrospun fibers were further sheathed by the self-polymerization of norepinephrine, a catecholamine found both in neurotransmitters and mussel adhesive proteins, to improve the surface hydrophilicity and provide a smooth bio-interface. The modified electrospun fibers on stainless wires were developed as novel custom-made solid-phase microextraction (SPME) fibers. These fibers exhibited much higher extraction efficiency compared to the polydimethylsiloxane (PDMS) fibers, especially to the sulfonamides. The custom-made SPME fibers also showed excellent stability with the relative standard deviations (RSDs) of intra-fiber ranged from 1.98% to 9.86% and RSDs of inter-fiber ranged from 4.36% to 15.6%. Moreover, these fibers were also demonstrated to be anti-biofouling and suitable for in vivo sampling. The custom-made SPME fibers were successfully applied to determine the Pharmaceutical concentrations in living fishes and vegetables. The accuracies were verified by the comparison with liquid extraction and the sensitivities were demonstrated to be satisfying with the limits of detection (LODs) ranged from 0.16   ng/g to 5.35   ng/g in fish muscle and 0.02   ng/g to 8.02   ng/g in vegetable stem.

Bioinspired Polyelectrolyte-Assembled Graphene-Oxide-Coated C18 Composite Solid-Phase Microextraction Fibers for In Vivo Monitoring of Acidic Pharmaceuticals in Fish

Junlang Qiu,Guosheng Chen,Shuqin Liu,Tianlang Zhang,Jiayi Wu,Fuxin Wang,Jianqiao Xu,Yan Liu,Fang Zhu,Gangfeng Ouyang

A novel solid-phase microextraction (SPME) fiber was prepared by gluing poly(diallyldimethylammonium chloride) (PDDA) assembled graphene oxide (GO)-coated C18 composite particles (C18@GO@PDDA) onto a quartz fiber with polyaniline (PANI). The fiber surface coating was sequentially modified with bioinspired polynorepinephrine, which provided a smooth biointerface and makes the coating suitable for in vivo sampling. The novel custom-made coating was used to extract acidic pharmaceuticals, and high-performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) was employed for analysis. The custom-made coating exhibited a much higher extraction efficiency than the previously used commercial polydimethylsiloxane (PDMS) and polyacrylate (PA) coatings. The custom-made coating also possessed satisfactory stability (the relative standard deviations (RSDs) ranged from 1.60% to 10.3% for six sampling-desorption cycles), interfiber reproducibility (the RSDs ranged from 2.61% to 11.5%), and resistance to matrix effects. The custom-made fibers were used to monitor the presence of acid pharmaceuticals in dorsal-epaxial muscle of living fish, and satisfactory sensitivities (limits of detection ranged from 0.13 ng/g to 7.56 ng/g) were achieved. The accuracies were verified by the comparison with liquid extraction. Moreover, the novel fibers were successfully used to monitor the presence of acidic pharmaceuticals in living fish, which demonstrated that the custom-made fibers were feasible for possible long-term in vivo continuous pharmaceutical monitoring.

QuEChERS followed by dispersive liquid–liquid microextraction based on solidification of floating organic droplet method for organochlorine pesticides analysis in fish

Xiao-Chen Wang,Bin Shu,Shan Li,Zhao-Guang Yang,Bo Qiu

A fast, sensitive and environmental-friendly method was developed to determine 13 organochlorine pesticides in fish tissue through gas chromatography with electron capture detection. Sample was extracted initially by acetonitrile and then concentrated through dispersive liquid–liquid microextraction based on solidification of floating organic droplet (DLLME–SFO). Dispersive solid phase extraction (d-SPE) was not a separate step but integrated to DLLME-SFO procedure. In order to separate the d-SPE sorbent , water and organic phase well, a modification to the glass centrifuge tube was made to achieving high centrifugal speed. The optimized method was validated with recoveries ranging from 88.1% to 121.2% (with relative standard deviations <15%) at three spiked levels for all of the pesticides. Good linearity was achieved at seven concentration levels from 6.25 to 625   μg   L −1 , which corresponded to 0.001–0.100   mg   kg −1 in the sample. The limits of quantification and the limits of detection for 13 OCPs ranged from 1.94×10 −3 to 4.93×10 −3   mg   kg −1 and 6.50×10 −4 to 1.58×10 −3   mg   kg −1 , respectively. The method was successfully applied to analyze real fish samples.

Detection of amino thiols from fish samples by stable isotope chemical labelling coupled with ultra-performance liquid chromatography-tandem mass spectrometry

Siyuan Wu,Yang Feng,Shengjun Chen,Yongqiang Zhao,Jianchao Deng,Di Wang

The amino thiols are key antioxidants in organisms, and their detection in food is of significant importance. This study developed a new stable isotope chemical labelling coupled with ultra-performance liquid chromatography-tandem mass spectrometry method to detect six amino thiols from fish samples. By the proposed method, amino thiols were labeled after liquid extraction using the stable isotope labeling reagents of iodoacetamide (IAM) and D 4 -IAM. Desirable recovery and linearity of thiols were achieved in fish samples. The limits of detection (LOD) ranged from 0.2 to 6.5 nmol/g, and the detection sensitivity was improved from 2.6 to 74 folds. The cluster analysis revealed that this method successfully discriminated eight fish species and their sources. Additionally, we found that glutathione and cysteine were significantly correlated with the transportation stress status of golden pompano ( Trachinotus ovatus ). Overall, this study presents a sensitive and efficient method for the detection of amino thiols from fish samples.

Hapten design, monoclonal antibody preparation and immunoassay development for direct detection of the toxic metabolite of nitrovin without derivatization from fish

Ghulam Mujtaba Mari,Weilin Wu,M. Farooque Hassan,Ambreen Laghari,Hidayatullah Soomro,Aisha Talpur,Zhanhui Wang,Xuezhi Yu

Nitrovin, classified as a nitrofuran drug, has been banned in numerous countries due to its carcinogenic effect on human health. However, existing analytical techniques for its detection are laborious, requiring an overnight derivatization process lasting 16 h. In this study, novel haptens were designed to prepare antibodies against aminogunidine (AGD), the metabolite of nitrovin, and then developed an indirect competitive enzyme-linked immunosorbent assay (icELISA) method to detect AGD from fish samples without derivatization for the first time. Computer-assisted molecular modeling predicted the efficiency of novel haptens by conformational studies and electronic analysis. The results were consistent with the predicted results in producing high-affinity monoclonal antibody (mAb) against AGD. The half-inhibition concentration (IC 50 ) value of icELISA was 0.80 ng mL −1 for AGD, with a limit of detection of 0.27 μg kg −1 and recoveries of 89.8%–97.2%. The results showed that this new hapten design and synthesis provided a great prospect to obtain high-affinity mAb for AGD. This developed icELISA substantially avoided the conventional and lengthy derivatization process and was an effective method for the direct detection of AGD in fish tissue without derivatization.

Occurrence, bioaccumulation, and ecological and health risks of Cd, Sn, Hg, and Pb compounds in shrimp and fish from aquaculture ponds

Pingxiu Zeng,Hongmei Hu,Yuanchao Wang,Jinhua Liu,Heyong Cheng

Aquaculture organisms may accumulate metals to induce health risks. Compared with the focus on total contents, chemical-specific risk assessment makes reasonable but is rare. Herein, we elucidated occurrence of twelve metal compounds in shrimp and fish (edible muscle, one of major metal-containing and generally targeted organs), water, sediment, and feedstuff from two aquaculture ponds in Zhejiang Province (one of the major aquatic production and consumption areas). We detected Cd(II) (0.6 −71.4 μg kg –1 in 100 % prawn but 63 % fish), methylmercury (MeHg, 0.5 −7.1 μg kg –1 in 100 % fish but 61 % shrimp), Pb(II) (0.4 −1.0 μg kg –1 in 57 % fish and 39 % prawn), and trimethyltin and triethyltin (0.4 −0.7 μg kg –1 ), which were much lower than the maximum limits in China. Pb(II), Cd(II), and Hg(II) up to 0.38 mg kg –1 were main contaminants in sediment while Cd(II) and Pb(II) up to 0.44 mg kg –1 were major contaminants in feedstuff compared with Cd(II), Sn(II), Hg(II), and Pb(II) majored in water at ng L –1 levels. Ecological risks were low in water but high for tributyltin in sediment. Additionally, light bioaccumulation of Cd(II) from sediment for prawn and methylmercury from feedstuff/sediment for crucian and bighead carp was induced. We also found light health risk of triethyl- and trimethyl lead, and Cd(II) (to children) associated with fish/shrimp consumption (edible muscle). This study proved high necessity of chemical-specific assessment, and shall trigger increasing interest to more metallic compounds in a wide range of uncultured and cultured plants and animals.
