Understanding the interactions between mercury and microalgae, especially the interactions between inorganic mercury (IHg) and extracellular polymeric substances (EPS, a protective barrier between cells and their external environment), is essential for elucidating mercury’s toxicological mechanisms. Given the inherent cell heterogeneity, a novel analysis system of an online viscoelastic fluid focusing chip-time-resolved analysis inductively coupled plasma mass spectrometry has been developed to investigate the bioaccumulation of HgS nanoparticles and Hg2+ in single Microcystis aeruginosa (M. aeruginosa) cells, exploring the interaction mechanisms between HgS/Hg2+ accumulation in algal cells and EPS. The single-cell analysis results reveal minimal bioavailability of HgS within algal cells, with mercury’s toxicity to M. aeruginosa being species-dependent. Notably, algal cells exhibited more heterogeneity in HgS uptake than in Hg2+ uptake. Under Hg2+/HgS stress, M. aeruginosa cells with EPS removed (EPS-R algal cells) showed an increased level of bioaccumulation of mercury compared to those with EPS (EPS-C algal cells), highlighting the critical role of EPS in mercury bioaccumulation. Overall, the designed viscoelastic fluid microfluidic focusing chip integrates focusing and cleaning functions, featuring easy fabrication, simple operation, low sample loss, and relatively high throughput. Under the optimal conditions, the sample throughput is 1195 min–1 and the cell recovery is 90%. Besides, this research offers novel insights into the interaction mechanisms between Hg2+/HgS and EPS in microalgal cells and unveils the specific toxic effects of Hg2+/HgS on M. aeruginosa at the single-cell level, contributing to a deeper understanding of mercury’s ecological and toxicological impact in aquatic environments.

To conveniently monitor and remove toxic Hg 2+ and S 2− in food samples, Au Ag quantum-dot nanozyme (Au-Ag@FA QDs) was identified. The strong synergistic effect among folic acid (FA), Ag and Au endowed Au-Ag@FA QDs with superior peroxidase-mimic activity to accelerate 3,3′,5,5′-tetramethylbenzidine (TMB) redox with K m / V max of 0.28 mmol·L −1 /6.40 × 10 −8 mol·L −1 ·s −1 . Trace Hg 2+ or S 2− could selectively alter its peroxidase-like activity and inherent fluorescence performance, with obvious hyperchromic/“turn-on” fluorescent effect for Hg 2+ or hypochromic/“turn-off” one for S 2− . And red-green-blue (RGB) values analyzed from smartphones changed regularly with c Hg 2+ or c S 2− . Under the optimized conditions, Au-Ag@FA QDs were successfully applied for multi-mode UV–vis, fluorescent, and RGB monitoring of Hg 2+ or S 2− in food samples. The detection limits were 7.38 × 10 −7 /5.74 × 10 −8 /4.93 × 10 −6 g·L −1 and 1.99 × 10 −6 /4.48 × 10 −8 /5.79 × 10 −6 g·L −1 respectively. Importantly, over 93.8 % Hg 2+ or 90.5 % S 2− could be removed by simple centrifugal-separation after detection. The selective recognition mechanisms to Hg 2+ and S 2− were also proposed.

Biosensor based on DNA hybridization holds great potential to get higher sensitivity as the optimal DNA hybridization efficiency can be achieved by controlling the distribution and orientation of probe strands on the transducer surface. In this work, an innovative strategy is reported to tap the sensitivity potential of current electrochemiluminescence (ECL) biosensing system by dispersedly anchoring the DNA beacons on the gold nanoparticles (GNPs) array which was electrodeposited on the glassy carbon electrode surface , rather than simply sprawling the coil-like strands onto planar gold surface. The strategy was developed by designing a “signal-on” ECL biosensing switch fabricated on the GNPs nanopatterned electrode surface for enhanced ultra-sensitivity detection of Hg 2+ . A 57-mer hairpin-DNA labeled with ferrocene as ECL quencher and a 13-mer DNA labeled with Ru(bpy) 3 2+ as reporter were hybridized to construct the signal generator in off-state. A 31-mer thymine (T)-rich capture-DNA was introduced to form T–T mismatches with the loop sequence of the hairpin-DNA in the presence of Hg 2+ and induce the stem-loop open, meanwhile the ECL “signal-on” was triggered. The peak sensitivity with the lowest detection limit of 0.1 nM was achieved with the optimal GNPs number density while exorbitant GNPs deposition resulted in sensitivity deterioration for the biosensor. We expect the present strategy could lead the renovation of the existing probe-immobilized ECL genosensor design to get an even higher sensitivity in ultralow level of target detection such as the identification of genetic diseases and disorders in basic research and clinical application.

A near-infrared (NIR)-excitable ratiometric fluorescent nanosensor, CS-UCNP@mSiO 2 –RBPA, was developed for the selective and sensitive detection of Hg 2+ . The nanosensor is constructed by grafting a rhodamine B-derived probe (RBPA) onto mesoporous silica-coated core–shell NaYF 4 :Yb,Er upconversion nanoparticles (UCNPs). Upon 980 nm excitation, the UCNPs exhibit characteristic green emission at ~ 543 nm and red emission at ~ 655 nm. In the presence of Hg 2+ , the spirolactam ring of RBPA opens specifically, producing a yellow-emitting species that enables efficient Förster resonance energy transfer (FRET) from the UCNP donor. This leads to a Hg 2+ concentration-dependent quenching of green emission and a synchronous enhancement of yellow fluorescence at ~ 584 nm, with the red emission acting as a stable internal reference. The nanosensor shows outstanding selectivity toward Hg 2+ against other competing metal ions and performs optimally at neutral pH. It presents a good linear response in the Hg 2+ concentration range 0–70 µM via UV–vis absorption, and achieves an ultra-low detection limit of 14 nM based on the ratiometric fluorescence signal (I 584 /I 655 ). Benefiting from NIR excitation, ratiometric self-calibration and excellent specificity, this fluorescent nanoplatform holds great potential for practical applications in environmental monitoring and biological analysis.

Split crRNA with Cas12a (SCas12a)-powered biosensor has shown sensitivity and accuracy in nucleic acid detection, but its application to metal ion target detection still needs to be explored. By integrating an SCas12a-mediated cascade strand displacement reaction (CSDR) with a phosphorothioate-modified hairpin G-quadruplex (psHG4) reporter, a portable “turn-on” colorimetric/photothermal system was engineered for the precise and sensitive detection of Hg2+ (termed as SCas12a/CSDR-psHG4). Target Hg2+ triggers SCas12a-mediated CSDR, and then activates SCas12a’s nuclease activity. Activated SCas12a cleaves the loop region of psHG4, thereby liberating the previously silenced psG4 sequence, which subsequently assembles into a psG4/hemin complex in the presence of K⁺ and hemin. This complex catalyzes the typical oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB), generating the colorimetric and photothermal product TMBox. Record the RGB value of the solution with a portable smartphone and track the temperature change using a thermometer. This colorimetric/photothermal biosensor displayed a linear range of 1 pmol/L to 100 nmol/L and 100 fmol/L to 10 nmol/L, with a detection limit of 0.5 pmol/L and 60 fmol/L, respectively. Utilizing the SCas12a technique, the SCas12a/CSDR-psHG4 presents a fresh method for identifying metal ion targets, along with significant prospects for enhancing human health protection and environmental surveillance.

A water-soluble and colorimetric fluorescent probe with a large Stokes shift (139 nm) for rapidly detecting Hg2+, namely Hcy-mP, was synthesized by using an indole derivative and 2,4-dihydroxybenzaldehyde as starting materials. This probe demonstrates good selectivity for Hg2+ over other metal ions including Ag+, Pb2+, Cd2+, Cr3+, Zn2+, Fe3+, Co2+, Ni2+, Cu2+, K+, Na+, Mg2+, and Ca2+ in aqueous solution. With the increase in concentration of Hg2+, the color of the solution changed from pale yellow to pink and the fluorescence intensity decreased slightly. When 5-equivalents of EDTA were added to the solution with Hg2+, the fluorescence intensity of this probe was restored. The probe has been applied to the detection of Hg2+ in real water samples. Moreover, this probe was confirmed to have low cytotoxicity and excellent cell membrane permeability. The effect of Hcy-mP–Hg2+ towards living cells by confocal fluorescence was also investigated.

We have developed an efficient strategy for synthesizing a strongly coupled Au/Fe3O4/GO hybrid material to improve the catalytic activity, stability, and separation capability of Au nanoparticles (NPs) and Hg2+. The hybrid material can be synthesized by the direct anchoring of Au and Fe3O4 NPs on the functional groups of GO. This approach affords strong chemical attachments between the NPs and GO, allowing this hybrid material to ultrasensitively detect Hg2+ in aqueous solutions with a detection limit as low as 0.15 nM. In addition, the deposition of Hg0 on the surface of Au/Fe3O4/GO could be quickly (within 30 min) and efficiently (>99% elimination efficiency) removed by the simple application of an external magnetic field and then Au/Fe3O4/GO could be subsequently reused at least 15 times, with the elimination efficiency remaining high (>96%).

We have constructed a multifunctional nanoprobe with sensing and imaging properties by using hollow mesoporous silica coated upconversion nanoparticles (UCNPs) and Hg2+ responsive ruthenium (Ru) complex. The Ru complex was loaded into the hollow mesoporous silica and the UCNPs acted as an energy donor, transferring luminescence energy to the Ru complex. Furthermore, polyethylenimine (PEI) was assembled on the surface of mesoporous silica to achieve better hydrophilic and bio-compatibility. Upon addition of Hg2+, a blue shift of the absorption peak of the Ru complex is observed and the energy transfer process between the UCNPs and the Ru complex was blocked, resulting in an increase of the green emission intensity of the UCNPs. The un-changed 801 nm emission of the nanoprobe was used as an internal standard reference and the detection limit of Hg2+ was determined to be 0.16 μM for this nanoprobe in aqueous solution. In addition, based on the low cytotoxicity as studied by CCK-8 assay, the nanoprobe was successfully applied for cell imaging and small animal imaging. Furthermore, when doped with Gd3+ ions, the nanoprobe was successfully applied to in vivo magnetic resonance imaging (MRI) of Kunming mice, which demonstrates its potential as a MRI positive-contrast agent. Therefore, the method and results may provide more exciting opportunities to afford nanoprobes with multimodal bioimaging and multifunctional applications.

In the present work, we proposed a novel naked-eye and turn-on fluorescence sensor based on the 5,10,15,20-(4-sulphonatophenyl) porphyrin (TPPS 4 )-Hg 2+ system with short assay time and high sensitivity to detect glutathione (GSH) in real samples. The TPPS 4 was the first time to detect GSH because of its high fluorescence quantum yield. The initial fluorescence from TPPS 4 was quenched by Hg 2+ with an electron transfer process. However, when put a certain concentration of GSH into this system, the fluorescence sensor was turned to the “on” state, which was related to a competitive affinity of Hg 2+ to GSH and the N atoms of porphyrin ring. Under the optimal conditions, the naked-eye detection limit was 0.43 nM and the limit of detection (LOD) was arrived to 16 fM (S/N = 3) for GSH. The linear regions are obtained by two ranges varying from 0 to 28.5 μM and from 38.15 to 61.87 μM. Furthermore, it was confirmed that the TPPS 4 -Hg 2+ fluorescence probe has excellent selectivity, high sensitivity and quick fluorescence response for GSH.
