Lateral organic-inorganic hybrid Vis-NIR photodetectors based on GaN nanowires promoting photogenerated carriers transfer

Tao Han,Zexin Wu,Zhilong Deng,Xiaofeng Zhang,Sidi Yang,Cuicui Chen,Jiajia Zhu,Shufang Ding,Chunzhi Jiang

The narrow bandgap of the low-energy near-infrared (NIR) polymer would lead to overlap between adjacent energy levels, which is a major barrier to the preparation of Vis-NIR polymer bulk heterojunction (BHJ) photodetectors with small responsivity and photocurrent. In this study, a high-performance lateral inorganic-organic hybrid photodetector was constructed to eliminate this barrier by combining GaN nanowires (GaN-NWs) with PDPP3T:PC 61 BM-based BHJ. In stage one, high-quality GaN-NWs were synthesized by the catalyst-free CVD method. The mechanism for controlling GaN-NWs morphology by adjusting the NH 3 flow rate was revealed. In stage two, the GaN-NWs with large electron mobility were used to accelerate the transfer of photogenerated carriers in the BHJ layer. Finally, compared with the BHJ device, the BHJ/GaN device demonstrated obvious improvements in responsivity and photocurrent at the wavelength between 400 and 1000 nm. The responsivity and photocurrent increased over 20-fold at the NIR band of 800–900 nm. Besides, owing to the energy level gradient effect, the BHJ/GaN device has a response speed of 7.8/<5.0 ms, which increases over three orders of magnitude than that of the GaN-NWs-based device ( tr / tf : 7.1/10.9 s). Therefore, the novel device structure proposed in this work holds great potential for preparing high-performance Vis-NIR photodetectors.

A bioactivated in vivo assembly nanotechnology fabricated NIR probe for small pancreatic tumor intraoperative imaging

Ren Han,Zeng Xiang-Zhong,Zhao Xiao-Xiao,Hou Da-yong,Yao Haodong,Yaseen Muhammad,Zhao Lina,Xu Wan-hai,Wang Hao,Li Li-Li

Real-time imaging of the tumour boundary is important during surgery to ensure that sufficient tumour tissue has been removed. However, the current fluorescence probes for bioimaging suffer from poor tumour specificity and narrow application of the imaging window used. Here, we report a bioactivated in vivo assembly (BIVA) nanotechnology, demonstrating a general optical probe with enhanced tumour accumulation and prolonged imaging window. The BIVA probe exhibits active targeting and assembly induced retention effect, which improves selectivity to tumours. The surface specific nanofiber assembly on the tumour surface increases the accumulation of probe at the boundary of the tumor. The blood circulation time of the BIVA probe is prolonged by 110 min compared to idocyanine green. The assembly induced metabolic stability broaden the difference between the tumor and background, obtaining a delayed imaging window between 8–96 h with better signal-to-background contrast (>9 folds). The fabricated BIVA probe permits precise imaging of small sized (<2 mm) orthotopic pancreatic tumors in vivo. The high specificity and sensitivity of the BIVA probe may further benefit the intraoperative imaging in a clinical setting.

NIR responsive nitric oxide nanogenerator for enhanced biofilm eradication and inflammation immunotherapy against periodontal diseases

Manlin Qi,Xuan Ren,Wen Li,Yue Sun,Xiaolin Sun,Chunyan Li,Siyao Yu,Lin Xu,Yanmin Zhou,Shuyan Song,Biao Dong,Lin Wang

Periodontal disease is one of the most common oral infectious diseases and closely related to many systemic diseases which is in urgent need of novel and effective drugs. In particular, the problems of deep lesion location and synchronization of antibacterial and anti-inflammatory needs to be addressed. Herein, near infrared (NIR) light triggered nanoplatform with antibacterial and anti-inflammatory dual-functions by antibacterial photodynamic therapy (aPDT), photothermal therapy (PTT) and gas therapy was realized by loading S-nitrosothiols (SNO) and ICG into mesoporous silica-coated gold nanorods . Using a single 808 nm NIR light, the excellent antibacterial efficacy can be achieved not only by aPDT-induced bacteria killing but also by PTT-triggered biofilm dispersal. Furthermore, SNO molecules would also be thermally activated to release nitric oxide (NO) gas molecules at the site of periodontal disease, exerting inflammation inhibition by suppressing the assembly of pro-inflammatory factors and NLRP3 inflammasome. Additionally, such nanoplatform could validly combat pathogenic bacterial biofilm, and modulate the inflammatory immunity, leading to favorable therapeutics on animal models with periodontal diseases. Hence, this design provides an effective solution to solve the problems of deep tissue treatment along with simultaneously antibacterial/anti-inflammatory capabilities, which provides a bright prospect for clinical treatment of periodontal diseases, even extended to other refractory infections in deep sites.

Mitochondrial targeted melanin@mSiO2 yolk-shell nanostructures for NIR-Ⅱ-driven photo-thermal-dynamic/immunotherapy

Yu Zhang,Qi Wang,Yishun Ji,Liyuan Fan,Binbin Ding,Jun Lin,Lili Wang

The challenge of photothermal therapy (PTT) clinical transformation is the limited penetration depth of laser light and the general concern of using synthetic photothermal nanomaterials. Hence, it is of great significance to develop combined cancer therapies based on NIR-II photothermal agents from natural materials. Herein, natural melanin nanoparticles from cuttlefish ink coated with large pore mesoporous SiO 2 were developed to load azodiisobutylimidazoline hydrochloride ( [email protected] ). After modification with (3-carboxypropyl) triphenylphosphonium bromide (CTPP), [email protected] nanoparticles showed enhanced retention in the tumor site and were further delivered to thermally susceptible mitochondria. Under 1064 nm laser irradiation, hyperthermia produced by melanin not only directly leads to cell death but also promotes AIPH release to generate oxygen-independent alkyl free radicals, which could further attack tumor cells. The collaboration of the photothermal effect of melanin and thermodynamic therapy of AIPH could induce a local antitumor immune response and effectively reprogram tumor-associated macrophages from the M2 to M1 phenotype. In combination with anti-PD-1 immune checkpoint blockade, the paralyzed immune system was awakened to inhibit and eliminate secondary metastatic tumors. The NIR-II-driven treatment in this paper can achieve energy conversion processes from NIR-II light to thermal energy and then to chemical energy to perform photothermal-dynamic therapy and immunotherapy. This will overcome the high side effects and insufficient antitumor immune response of PTT and provide a strategy for effective and mild antitumor therapy with clinically approved laser power and safe nanotechnology.

Compositionally complex (Ca, Sr, Ba)ZrO3 fibrous membrane with excellent structure stability and NIR reflectance

Kangkang Yuan,Qing Cui,Jie Li,Xiaotong Jin,Chengshun Li,Xingxiang Ji,Zhongjian Tian,Xinqiang Wang

A compositionally complex (Ca, Sr, Ba)ZrO 3 (CC-CSBZ) fibrous membrane with perovskite structure was successfully prepared via a simple electrospinning method. The formation, phase transformation and microstructure evolution of CC-CSBZ fibers were fully discussed. X-ray diffraction, Raman spectra and transmission electron microscopy analyses confirmed the formation and phase transformation of CC-CSBZ solid solution at high temperatures. Scanning electron microscopy results indicated that a slower grain growth rate and more stable high-temperature microstructure were achieved for CC-CSBZ fibers compared with previous BaZrO 3 and CaZrO 3 fibers. CC-CSBZ fibrous membrane also exhibited excellent flexibility at 1000 °C and high thermal stability at 1200 °C. Furthermore, an average near infrared reflectivity of 98% at least at room temperature was obtained for CC-CSBZ fibrous membrane heat-treated from 800 to 1400 °C. For the slow grain growth rate, excellent high-temperature stability and near infrared reflectance, CC-CSBZ fibrous membrane would be a promising and novel ceramic fiber product for high temperature applications.

NIR-activated nanosystems with self-modulated bacteria targeting for enhanced biofilm eradication and caries prevention

Yunjian Yu,Yufei Zhang,Yijie Cheng,Yuxia Wang,Zeyuan Chen,Haonan Sun,Xiaosong Wei,Zhuang Ma,Jie Li,Yayun Bai,Zhongming Wu,Xinge Zhang

The efficacious delivery of antimicrobial drugs to intractable oral biofilms remains a challenge due to inadequate biofilm penetration and lack of pathogen targeting. Herein, we have developed a microenvironment-activated poly(ethylene glycol) (PEG)-sheddable nanoplatform to mediate targeted delivery of drugs into oral biofilms for the efficient prevention of dental caries. The PEGylated nanoplatform with enhanced biofilm penetration is capable of deshielding the PEG layer under slightly acidic conditions in a PEG chain length-dependent manner to re-expose the bacteria-targeting ligands, thereby facilitating targeted codelivery of ciprofloxacin (CIP) and IR780 to the bacteria after accumulation within biofilms. The nanoplatform tends to induce bacterial agglomeration and suffers from degradation in the acidic oral biofilm microenvironment, triggering rapid drug release on demand around bacterial cells. The self-modulating nanoplatform under near-infrared (NIR) irradiation accordingly displays greatly augmented potency in oral biofilm penetration and disruption compared with drugs alone. Topical oral treatment with nanoplatforms involving synergetic pharmacological and photothermal/photodynamic trinary therapy results in robust biofilm dispersion and efficacious suppression of severe tooth decay in rats. This versatile nanoplatform can promote local accumulation and specific drug transport into biofilms and represents a new paradigm for targeted drug delivery for the management of oral biofilm-associated infections.

NIR light-triggered peroxynitrite anion production via direct lanthanide-triplet photosensitization for enhanced photodynamic therapy

Xi Xiao,Bingzhu Zheng,Qiang Zheng,Zijie Lu,Dong Cen,Xiujun Cai,Xiang Li,Renren Deng

Peroxynitrite anion (ONOO−), a product derived from reaction between reactive oxygen species (ROS) and nitric oxide (NO), is considered to be a more toxic reactive species than most ROS for cancer photodynamic therapy (PDT). To promote the PDT effect, a viable method is to develop rational strategies for efficient ONOO− generation at targeted tumor sites. Herein, a heterostructure nanocomposite containing ZnO-coated lanthanide nanoparticles (LnNPs) is reported for ONOO−-based PDT. In this nanocomposite, Nd3+-doped LnNPs are employed to realize efficient NIR-light-triggered ROS generation by activating the triplet state of chlorin-e6 (Ce6) photosensitizers via a direct lanthanide-to-triplet sensitization mechanism. Meanwhile, ZnO in the composite catalyzes the decomposition of S-nitrosoglutathione (GSNO) to generate NO in the tumor microenvironment. The coupled system allows the combination of photo-induced ROS and NO to produce ONOO−, leading to drastically promoted cancer cell apoptosis and tumor growth inhibition. This study establishes a new apoptosis-inducing PDT agent, which is potentially active in drug resistant malignancies.

Multi-site occupancy high-efficient Mg2LaTaO6:Cr3+ phosphor for application in broadband NIR pc-LEDs

Ziyang Wu,Jinmeng Xiang,Changheng Chen,Zexun Li,Xianju Zhou,Yuehui Jin,Chongfeng Guo

As a new generation of near-infrared (NIR) light source, phosphor-converted NIR light emitting diodes (NIR pc-LEDs) have broad application prospects in biological tissue imaging, night vision and chemical composition analysis . However, most NIR emitting phosphors are still limited to short-wavelength, narrow-band emission and low efficiency. Here, based on the multi-site occupation strategy, the NIR phosphor Mg 2 LaTaO 6 :Cr 3+ (MLT:Cr 3+ ) achieves an efficient broadband NIR emission with the full width at half maximum (FWHM) of 203 nm in the range of 600–1300 nm under the excitation of 468 nm blue light. The phosphor MLT:0.01Cr 3+ not only exhibits an excellent internal quantum efficiency (IQE) of 85.6 %, but also has good thermal stability and remains about 64.2 % photoluminescence (PL) intensity of room temperature at 423 K. Furthermore, the MLT:0.01Cr 3+ phosphor was combined with a blue LED chip to fabricate the NIR pc-LED device, and its potential applications were successfully realized in information encryption, bioimaging and the qualitative or quantitative analysis of mixture of ethanol and water.

NIR-triggered biodegradable MOF-coated upconversion nanoparticles for synergetic chemodynamic/photodynamic therapy with enhanced efficacy

Man Zou,Yajie Zhao,Binbin Ding,Fan Jiang,Yeqing Chen,Ping'an Ma,Jun Lin

The generation of reactive oxygen species (ROS) is often limited by the overexpression of glutathione (GSH) in the tumor microenvironment (TME) and the penetration depth of visible light. In view of this, we designed biodegradable Cu/ZIF-8 shell-coated lanthanide-doped nanoparticles and loaded with a photosensitizer (Rose Bengal), and fabricated a NIR-triggered nanocomposite NaYF4:20%Yb, 2%Er@Cu/ZIF-8/RB@F127 (UCZRF) for the synergistic treatment of CDT and PDT. Interestingly, the UCZRF nanostructures can specifically release RB and Cu2+ in the acidic tumor microenvironment (TME). Cu2+ depletes GSH to effectively change the amount of ROS produced by RB under 980 nm laser irradiation, and Cu+ from the reduction reaction of GSH reacts with excess hydrogen peroxide (H2O2) in the TME to generate hydroxyl radicals (⋅OH) with high oxidation activity. In such a simple nanosystem, we enhance the anti-tumor effect of CDT and PDT by improving the efficiency of ROS generation.

NIR-II Upconversion Photoluminescence of Er3+ Doped LiYF4 and NaY(Gd)F4 Core-Shell Nanoparticles

Feng Qilong,Zheng Wenjing,Pu Jie,Chen Qiaoli,Shao Wei

The availability of colloidal nano-materials with high efficiency, stability, and non-toxicity in the near infrared-II range is beneficial for biological diagnosis and therapy. Rare earth doped nanoparticles are ideal luminescent agents for bio-applications in the near infrared-II range due to the abundant energy level distribution. Among them, both excitation and emission range of Er3+ ions can be tuned into second biological window range. Herein, we report the synthesis of ∼15 nm LiYF4, NaYF4, and NaGdF4 nanoparticles doped with Er3+ ions and their core-shell structures. The luminescent properties are compared, showing that Er3+ ions with single-doped LiYF4 and NaYF4 nanoparticles generate stronger luminescence than Er3+ ions with doped NaGdF4, despite the difference in relative intensity at different regions. By epitaxial growth an inert homogeneous protective layer, the surface luminescence of the core-shell structure is further enhanced by about 5.1 times, 6.5 times, and 167.7 times for LiYF4, NaYF4, and NaGdF4, respectively. The excellent luminescence in both visible and NIR range of these core-shell nanoparticles makes them potential candidate for bio-applications.

Redox/pH/NIR-responsive degradable silica nanospheres with fluorescence for drug release and photothermal therapy

Xinli Li,Mengru Chen,Yanbao Zhao,Lei Sun

In this work, S S bond-doped porous silica nanoparticles with CuS nanocores (CuS/OS) were prepared as drug carriers for chemical-photothermal synergistic therapy and redox/pH/light triple-response drug release. The synthesized CuS/OS carriers have dendritic pore structure with particle size of about 100 nm. Tiopronin (MPG) was used as model drug to evaluate the release performance of the carrier. Chitosan (CS) and green fluorescent curcumin (Cur) were used as plugs to seal the loaded drug, which endows the CuS/OS carrier with pH-responsive release and monitoring properties. The obtained MPG-loaded CuS/OS-CS-Cur has pH/GSH/NIR triple responsive release and redox-responsive degradation. The introduction of CuS nanocores gives the CuS/OS system a good photothermal therapy function. The CuS/OS-CS-Cur and its degradation products have good biocompatibility.

A smart ROS/NIR dual-responsive melanin delivery platform for photoacoustic imaging-guided osteoarthritis therapy

Jihao Ruan,Qili Yu,Haomin Cui,Xing Qin,Limei Qin,Shuai Chen,Dechao Niu,Cunyi Fan

ABSTRACT Oxidative stress is considered an efficient therapeutic target in the progression of osteoarthritis (OA). Unfortunately, the current drug delivery system for the reactive oxidative species (ROS) scavengers is still unsatisfactory due to their insufficient responsiveness and rapid joint clearance. In this study, we design and develop a smart anti-oxidative agent by constructing a dual-responsive hybrid micelle with free radical scavenger melanin in the micellar core and polydopamine on the shell (designated as DHMP/M) for photoacoustic imaging-guided knee OA therapy. The resulting DHMP/M displays a unique long-term controlled-release behavior involving the slow and persistent release under endogenous ROS stimuli and short-burst release capability triggered by exogenous near-infrared (NIR) light. More importantly, DHMP/M possesses excellent free radical scavenging capacity and protective effects in chondrocytes in vitro and in in vivo rat OA models. Additionally, DHMP/M exhibits efficacious photoacoustic (PA) imaging for adjusting the therapy on demand. The RNA-Seq assay of rats further shows a significant reduction of TLR-2 in the OA cartilage, revealing that DHMP/M can protected OA from inflammation via TLR-2/MAPK/Akt signaling. Consequently, the present ROS/NIR dual-responsive melanin delivery platform together with PA imaging-monitoring provides a precise theranostic strategy for OA therapy.
