Construction of Redox-Responsive PEG Cloaked Mesoporous Silica Core-Shell Nanoparticles: Examination of Prostate Cancer Cells Proliferation and Apoptosis Induction

Jin Rui,Gong Weijian,Wang Dongzhao,Du Yangyang,Nie Guoyan,Zhu Mingting,Yu Yong

Regulating the release of drugs and limiting their accumulation in healthy tissues is highly beneficial. This study employed core mesoporous silica (MS) nanoparticles, encapsulated with Vincristine sulphate (VC) and cloaked with non-porous shell silica modified with the disulfide bonds (SS). The MS was cloaked with polyethylene glycol (PEG) through disulfide bonds (PEG/NH 2 -MS@VC-SS). We examined the physicochemical characteristics of MS, encompassing XRD patterns, FTIR spectra, SEM, TEM, DLS, and zeta potential. The drug release pattern was assessed using different glutathione (GSH) levels. The nanoparticle cell uptake of the cell lines was evaluated. Additionally, the nanoparticle’s cell survival and ability to trigger apoptosis in the PC3 cell lines was assessed by MTT assay. The NPs-treated PC3 cell lines were imagined through fluorescent microscopy using acridine orange/propidium iodide (AO/PI) staining, 4′,6-diamidino-2-phenylindole (DAPI), and propidium iodide (PI) staining. Our study demonstrated that the release of VC from the PEG/NH 2 -MS@VC-SS was responsive to redox conditions. Outcomes showed increased cellular absorption and effective apoptosis induction, highlighting the promising capability of these redox-response DDSs for prostate cancer treatment.

High-throughput UPC2-MS/MS for elucidating the molecular-weight-dependent excretion kinetics of PEG polymers with 10–17 subunits

Zhang Jiarui,Liu Jiale,Lai Jintian,Deng Yue,Feng Shuang,Zhou Xinyue,Shi Meiyun,Yin Lei

Individual polyethylene glycol (PEG) oligomers’ in vivo fate is crucial for evaluating polymer-based therapeutic safety. Herein, a high-throughput UPC 2 -MS/MS method was developed for the oligomer-resolved excretion kinetics of PEG600 ( n  = 10–17) in rats. By optimizing supercritical CO 2 chromatography and utilizing ammonium adducts ([M+NH 4 ] + ), baseline separation of eight oligomers was achieved within 2.4 min, significantly enhancing efficiency for bioanalysis. This platform significantly enhances analytical capacity, enabling the efficient processing of large-scale biological sample sets required for mass balance studies. Application of this method revealed distinct molecular-weight-dependent elimination. Specifically, 72-h urinary recovery showed a complex non-linear relationship, peaking at n  = 10 (71.9%) and n  = 16 (72.7%). Simultaneously, total cumulative recovery (urine and feces) decreased progressively from >95% ( n  = 10, 11) to 75.3% ( n  = 17). This reduction suggests enhanced tissue sequestration or metabolic biotransformation for larger oligomers. This high-resolution profiling uncovers subtle elimination differences obscured in bulk analysis, providing critical pharmacokinetic insights for PEG excipients. Graphical abstract

Multifunctional SIS-PEG-AC bio-patch with enhanced adhesion, sustained anti-inflammatory activity, and regenerative properties to promote lung repair

Heng Li,Deguang Wang,Zixun Wang,Wenjie Chen,Yuxin Guo,Han Liang,Yanqiu Zhao,Qi Wang,Yiqian Huang,Wei Jing,Pengfei Wei,Bo Zhao,Binbin Guan,Min Zhang,Jing Yu,Xudong Xiang

Postoperative complications, such as air leakage, inflammation, and pleural adhesion, present considerable clinical challenges following lung resection procedures. These complications critically affect pulmonary recovery. In this study, we report a novel multifunctional self-adhesive bio-patch SIS-PEG-AC, which synergistically combines the biological activity of small intestinal submucosa (SIS), the effective sealing air leaks provided by polyethylene glycol (PEG) coating, and the anti-inflammatory effects of asiaticoside (AC). Compared to SIS, which exhibited limited tissue adhesion (0.73 kPa), the SIS-PEG-AC patch demonstrated a 30-fold increase in adhesiveness, achieving robust tissue adhesion (23.31 kPa) and significantly enhanced reparative capacity. AC delivered from the PEG hydrogel coating in a sustained manner, promoting the polarization of macrophages to M2 phenotype, to suppressing pro-inflammatory cytokine expression, thereby establishing a pro-regenerative ecological environment. The results of 28-day implantation on rat lung injury and 70-day implantation on pig lung wedge resection showed, the SIS-PEG-AC achieved excellent wound sealing, minimized postoperative pleural adhesions and significantly accelerated lung tissue regeneration. Compared with traditional synthetic polymers, the SIS-PEG-AC patch exhibited reduced adverse reactions, superior host tissue integration, and heightened control of the regenerative microenvironment, providing a promising approach to mitigate postoperative complications. Overall, this multifunctional patch demonstrates considerable clinical potential in lung repair, offering an effective strategy for thoracic surgical interventions.

Swelling Behaviour of Polystyrene Microsphere Enhanced PEG-Based Hydrogels in Seawater and Evolution Mechanism of Their Three-Dimensional Network Microstructure

Chen Zhang,Yuhong Qi,Zhanping Zhang

To understand the microstructure evolution of hydrogels swollen in seawater, freeze-drying technology was used to fix and preserve the swollen three-dimensional microstructure. By this method, we revealed the swelling behavior of hydrogels in seawater, and elucidated the mechanism of the swelling process. Meanwhile, we also used Fourier-transform infrared spectroscopy; laser confocal microscopy; field emission scanning electron microscopy, and swelling performance tests to research the structure and properties of PS-PEG hydrogels, before and after seawater swelling, and analyzed the structure and properties of PEG-based hydrogels with different contents of polystyrene microspheres. Results showed that PS-PEG hydrogels went through three stages during the swelling process, namely ‘wetting-rapid swelling-swelling equilibrium’. Due to the capillary effect and hydration effect, the surface area would initially grow tiny pores, and enter the interior in a free penetration manner. Finally, it formed a stable structure, and this process varied with different content of polystyrene microspheres. In addition, with the increase of polystyrene microsphere content, the roughness of the hydrogel before swelling would increase, but decrease after swelling. Appropriate acquisition of polystyrene microspheres could enhance the three-dimensional network structure of PEG-based hydrogels, with a lower swelling degree than hydrogels without polystyrene microspheres.

Novel MoS2/montmorillonite hybrid aerogel encapsulated PEG as composite phase change materials with superior solar-thermal energy harvesting and storage

Qijing Guo,Hao Yi,Feifei Jia,Shaoxian Song

Phase change materials (PCMs) offer significant advantages in energy conversion and storage by facilitating the storage and release of thermal energy during phase transition processes. However, challenges such as leakage during PCM phase transitions and poor light absorption properties have constrained their application in the field of photothermal energy storage. In this study, Montmorillonite (Mt) and molybdenum disulfide (MoS 2 ) has been used to design and synthesize hybrid aerogels (MoS 2 /Mt) boasting high mechanical strength and excellent photothermal conversion performance. These aerogels are then used to encapsulate polyethylene glycol (PEG) to prepare composite PCMs with outstanding solar-thermal conversion and storage performances. The results show that the synthesized MoS 2 /Mt-PEG composite PCMs exhibit high enthalpies of melting and solidification of 169.16 J/g and 170.78 J/g, respectively, while the aerogel supporting material has a high compressive modulus of 1.96 MPa. Moreover, the composite material displayed excellent thermal stability and leakage resistance after undergoing 30 melting-cooling cycles. Furthermore, the incorporation of MoS 2 imparted outstanding light absorption properties to the MoS 2 /Mt-PEG composite, resulting in a high light absorption and photothermal conversion-storage efficiency of 93.4 % and 96.47 %, respectively. Synthesized composite PCMs also demonstrate outstanding performance in solar-thermal-electricity conversion, achieving a voltage output of 458 mV under illumination conditions and maintaining a sustainable voltage output even after removing the light source. Thus, the composite PCMs prepared in this work can meet the requirements of high enthalpy, effective leakage prevention, efficient solar-thermal conversion and solar-thermal-electricity conversion performance, thereby presenting potential applications in practical solar energy collection, conversion, and storage.

Dual-responsive PEG-lipid polyester nanoparticles for siRNA and vaccine delivery elicits anti-cancer immune responses by modulating tumor microenvironment

Zixu Liu,Linxuan Zhao,Yupeng Feng,Qingqing Wang,Nan Dong,Yu Zhang,Tian Yin,Haibing He,Xing Tang,Jingxin Gou,Li Yang

Cancer vaccine-based immunotherapy has great potential; however, vaccine has been hindered by the immunosuppressive tumor microenvironment (TME). In this study, dual-responsive PEG-lipid polyester nanoparticles (PEG BR647-NP) for tumor-targeted delivery were proposed. PEG BR647-NP containing model tumor associated antigen (TAA) OVA and signal transduction and activator of transcription 3 (STAT3) siRNA were delivered to tumor. PEG BR647-NP was internalized by tumor associated dendritic cells (TADCs), where TAA and siRNA were released into cytoplasm via endo/lysosome escape effect. The released OVA was presented by the major histocompatibility complex class I to activate T cells, the released STAT3 siRNA acted to relieve TADCs dysfunction, promote TADCs maturation, improve antigen-presenting ability, and enhance anticancer T cell immunity. Meanwhile, PEG BR647-NP was ingested by tumor cells, and could kill tumor cells by the pro-apoptosis effect of STAT3 siRNA. Moreover, PEG BR647-NP could reduce the proportion of myeloid-derived suppressor cells (MDSCs) and regulatory T cells (Tregs) in tumors and abrogate immunosuppression. The integration of relieved TADCs dysfunction, promoted TADCs maturation, enhanced antigen cross-presentation, abrogated immunosuppression, and improved pro-apoptosis effect can boost the vaccination for tumor immunotherapy. Thus, PEG BR647-NP efficiently delivered vaccine and STAT3 siRNA to tumor, and modulated immunosuppressive TME, provided better antitumor effects.

Effects of Porogen PEG and Pore Structure of PVDF Substrates on the Permeability–Selectivity Trade-off of TFC-NF Membranes

Chenyu Zhu,Xue Zhang,Fuzhi Li,Ruochen Yang,Xuan Zhao

Thin-film composite (TFC) nanofiltration (NF) has been widely utilized to separate monovalent ions from multivalent ions in industrial wastewater treatment. In this study, the pore properties of polyvinylidene fluoride (PVDF) substrates were adjusted by varying the content and molecular weights (MWs) of porogen polyethylene glycol (PEG). The TFC membranes fabricated on different substrates showed distinguishable permeance and selectivity. The PVDF membranes with 2 kDa PEG of 0.5–3.0 wt % and 20 kDa PEG of 0.2 wt % were fit for the substrates of the TFC NF membrane. These substrates are usually characterized by evenly distributed surface pores with porosity of 7–12% and tiny finger-like pores of less than 15 μm length. Such pore structures in the substrates are fit for the storage and migration of amine monomers, resulting in a defect-free PA layer. In a 6 h filtration of mining wastewater using an optimized TFC membrane, the permeance remained around 5 LMH/bar and the separation factor of SO42–/Cl– stayed over 13.39 with a maximum value over 30. This study shed light on improving NF membrane performance by simply adjusting the porogen in the substrate casting solution.

Integration of PEG-conjugated gadolinium complex and superparamagnetic iron oxide nanoparticles as T1–T2 dual-mode magnetic resonance imaging probes

Yang Li,Fu Shengxiang,Cai Zhongyuan,Liu Li,Xia Chunchao,Gong Qiyong,Song Bin,Ai Hua

The T1−T2 dual-mode probes for magnetic resonance imaging (MRI) can non-invasively acquire comprehensive information of different tissues or generate self-complementary information of the same tissue at the same time, making MRI a more flexible imaging modality for complicated applications. In this work, three Gadolinium-diethylene-triaminepentaaceticacid (Gd-DTPA) complex conjugated superparamagnetic iron oxide (SPIO) nanoparticles with different Gd/Fe molar ratio (0.94, 1.28 and 1.67) were prepared as T1–T2 dual-mode MRI probes, named as SPIO@PEG-GdDTPA0.94, SPIO@PEG-GdDTPA1.28 and SPIO@PEG-GdDTPA1.67, respectively. All SPIO@PEG-GdDTPA nanocomposites with 8 nm spherical SPIO nanocrystals showed good Gd3+ chelate stability. SPIO@PEG-GdDTPA0.94 nanocomposites with lowest Gd/Fe molar ratio show no cytotoxicity to Raw 264.7 cells as compared to SPIO@PEG-GdDTPA1.28 and SPIO@PEG-GdDTPA1.67. SPIO@PEG-GdDTPA0.94 nanocomposites with r1 (8.4 mM−1s−1), r2 (83.2 mM−1s−1) and relatively ideal r2/r1 ratio (9.9) were selected for T1–T2 dual-mode MRI of blood vessels and liver tissue in vivo. Good contrast images were obtained for both cardiovascular system and liver in animal studies under a clinical 3 T scanner. Importantly, one can get high-quality contrast-enhanced blood vessel images within the first 2 h after contrast agent administration and acquire liver tissue anatomy information up to 24 h. Overall, the strategy of one shot of the dual mode MRI agent could bring numerous benefits not only for patients but also to the radiologists and clinicians, e.g. saving time, lowering side effects and collecting data of different organs sequentially.

Cai Mingzhong,Liu Rong,Xu Caifeng,Huang Bin

Pd(OAc) 2 /XPhos–SO 3 Na in a mixture of poly(ethylene glycol) (PEG-400) and water is shown to be a highly efficient catalyst for the cyanation of aryl chlorides with potassium ferrocyanide. The reaction proceeded smoothly at 100 or 120 ºC with K 2 CO 3 or KOAc as base, delivering a variety of aromatic nitriles in good to excellent yields. The isolation of the crude products is facilely performed by extraction with cyclohexane and more importantly, both expensive Pd(OAc) 2 and XPhos–SO 3 Na in PEG-400/H 2 O system could be easily recycled and reused at least six times without any apparent loss of catalytic efficiency. Graphical Abstract Palladium-catalyzed cyanation of aryl chlorides with potassium ferrocyanide leading to aryl nitriles by using Pd(OAc) 2 /XPhos–SO 3 Na/PEG-400/H 2 O as a highly efficient and recyclable catalytic system is described.

Facile, catalyst-free, and high enthalpy PEG-based phase change composite for use in photothermal conversion and waste heat recovery

Zhigang Liu,Shang Gong,Xiaolong Li,Xiuli Chen,Xinpeng Hu,Ying Li,Xiangyu Yan,Hu Wang,Yi Li,Xiang Lu

The poor shape stability, low thermal storage density, and complex process confronted in fabricating high-performance phase change composites (PCCs) via the chemical cross-linking technology. Herein, PEG-based shape stability PCC (SSPCC) with high enthalpy and good shape stability was obtained using a facile one-pot method without additional catalysts. The reliable three-dimensional cross-linked structure formed by the interaction of epoxy resin and curing agent binds the movement of PEG molecular chains , which simultaneously in coupled with the stronger hydrogen bonding between PEG and cross-linked products to endow the as-prepared SSPCC with excellent shape stability, thermal stability, durability, and high thermal storage density (160.8 J/g). Moreover, the Raman spectra clearly presented the interaction force and phase morphology distribution between PEG and the skeleton structure. SSPCC with the high relative enthalpy efficiency and melt enthalpy can play an essential role in photothermal applications, while the facile manufacturing process free from additional organic solvents or high temperature, highlighting its green and sustainable features. Additionally, the photothermal transport and thermostatic effects of the SSPCCs show great potential for application in waste heat recovery, which can maintain higher temperatures for a long time. Totally, such SSPCC with reliable and distinguished phase change properties ensure a wide range of potential applications, while the easy-to-prepare and low-cost fabrication method provides an inspiring strategy for alternative SSPCC development.

The Modified Exenatide Microspheres: PLGA-PEG-PLGA Gel and Zinc-Exenatide Complex Synergistically Reduce Burst Release and Shorten Platform Stage

Jiang Wenjing,Gao Xiangjun,Wang Qiuli,Chen Yang,Li Dan,Zhang Xiaoyan,Yang Xinggang

The existing exenatide microspheres have the problem of burst release in the early stage, and minimal release in the middle stage which makes it difficult to achieve effective blood drug concentration (platform period). In this study, the modified exenatide microspheres were constructed to address the aforementioned issues. Poly(D,L-lactic-co-glycolic acid) (PLGA) and triblock copolymer with sol-gel conversion characteristics (PLGA-PEG-PLGA gel) were introduced as carriers to prepare microspheres. The hot gel characteristics and hydrophilicity of PLGA-PEG-PLGA gel were utilized to decline the burst release and shorten the platform period. Simultaneously, zinc acetate and exenatide were combined to generate an insoluble complex to further reduce the burst release. Herein, we prepared three types of exenatide microspheres using the solvent evaporation method and investigated their characterization as well as in vitro and in vivo release. According to the experimental findings, the modified exenatide microspheres, i.e., PLGA-PEG-PLGA gel and PLGA co-loaded zinc-exenatide insoluble complex microspheres (Zn-EXT-Gel-MS), had smooth and rounded surfaces, with a particle size of 24.7 μm, and the encapsulation rate reached 89.43%. And it was released for 40 days in vitro , behaving better than the other two microspheres in terms of release behavior. When this product was administered subcutaneously to rats, it produced a comparatively constant plasma exenatide concentration that lasted for 24 days and superior bioavailability than the exenatide microspheres (EXT-MS). The creation of modified exenatide microspheres may serve as a heuristic method for other long-acting medications. Graphical Abstract Schematic diagram of the synthesis process and release curves of three types of exenatide microspheres in vitro and in vivo

MiRNA320a Inhibitor-Loaded PLGA-PLL-PEG Nanoparticles Contribute to Bone Regeneration in Trauma-Induced Osteonecrosis Model of the Femoral Head

Zhang Ying,Li Chuan,Wei Qiushi,Yuan Qiang,He Wei,Zhang Ning,Dong Yiping,Jing Zhenhao,Zhang Leilei,Wang Haibin,Cao Xiangyang

BACKGROUND: This study aimed to explore the effect of a nanomaterial-based miR-320a inhibitor sustained release system in trauma-induced osteonecrosis of the femoral head (TIONFH). METHODS: The miR-320a inhibitor-loaded polyethylene glycol (PEG)- Poly(lactic-co-glycolic acid) (PLGA)- Poly-L-lysine (PLL) nanoparticles were constructed using the double emulsion method. The TIONFH rabbit model was established to observe the effects of miR-320a inhibitor nanoparticles in vivo . Hematoxylin–eosin staining and microcomputed tomography scanning were used for bone morphology analysis. Bone marrow mesenchymal stem cells (BMSCs), derived from TIONFH rabbits, were used for in vitro experiments. Cell viability was determined using the MTT assay. RESULTS: High expression of miR-320a inhibited the osteogenic differentiation capacity of BMSCs in vitro by inhibiting the expression of the osteoblastic differentiation markers ALP and RUNX2. MiR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticles were constructed with a mean loading efficiency of 1.414 ± 0.160%, and a mean encapsulation efficiency of 93.45 ± 1.24%, which released 50% of the loaded miR-320a inhibitor at day 12 and 80% on day 18. Then, inhibitor release entered the plateau. After treatment with the miR-320a inhibitor nanoparticle, the empty lacunae were decreased in the femoral head tissue of TIONFH rabbits, and the osteoblast surface/bone surface (Ob.S/BS), osteoblast number/bone perimeter (Ob.N/B.Pm), bone volume fraction, and bone mineral density increased. Additionally, the expression of osteogenic markers RUNX2 and ALP was significantly elevated in the TIONFH rabbit model. CONCLUSION: The miR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticle sustained drug release system significantly contributed to bone regeneration in the TIONFH rabbit model, which might be a promising strategy for the treatment of TIONFH.
