This research is aimed to develop a kind of hollow nanosphere based on β-cyclodextrin-grafted α, β-poly(aspartic acid) (β-CD- graft -PAsp) as drug carrier to enhance the stability of camptothecin (CPT) in aqueous media. Firstly, mono(6-(2-aminoethyl) amino-6-deoxy)-β-cyclodextrin (β-CDen) was synthesized by a substitution reaction between mono-6-deoxy-6-(p-tolylsulfonyl)-β-cyclodextrin (6-OTs-β-CD) and ethylenediamine; and then, the five-member rings in poly( l -succinimide) (PSI) were successively opened by β-CDen to obtain β-CD- graft -PAsp. The synthesized β-CD- graft -PAsp was found to form the unique hollow nanosphere with the internal hole of about 17 nm and many β-CD cavities of 0.7 nm interspersed on the shell. The drug-loading behavior of the hollow nanosphere was also evaluated by using CPT as guest molecule of β-CD. It was worth of note that the β-CD- graft -PAsp hollow nanosphere as carrier enhanced the stability of CPT in aqueous media, and the CPT from the β-CD- graft -PAsp hollow nanosphere displayed a sustained release profile and hence resulted in the essential decrease of cytotoxicity to L929 cell line. Furthermore, almost no cytotoxicity of the β-CD- graft -PAsp is desirable for the application of drug carrier. As a result, the β-CD- graft -PAsp hollow nanosphere showed a great potential as the carrier of CPT.

Polymer/bioceramic composite scaffolds have gained importance in treating bone defects. In this paper we described a novel degradable scaffold material for bone tissue repairing prepared by combining poly(lactide-co-glycolide-co-ε-caprolactone) (PLLGC) and 1,4-butanediamine-modified poly(lactide-co-glycolide) (BMPLGA) with nano-biaoactive glass (NBAG) and nano β-tricalcium phosphate (β-TCP). The composite scaffolds were characterized by using Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM) techniques. The FTIR results indicated that the inorganic fillers of NBAG and β-TCP were successfully incorporated into the PLLGC/BMPLGA matrix. The influences of the inorganic filler content and freezing temperature on the porosity and compressive strength of the composite scaffolds were studied. The composite scaffolds showed an interconnected pore structure with the pore size ranging from 20 to 100 μm depending on the freezing temperature and the inorganic fillers. The compressive strength was enhanced with increasing content of the inorganic fillers and the freezing temperature; however, the porosity decreased. We suggest the PLLGC-BMPLGA/NBAG-β-TCP composite scaffolds have great potential for application in bone tissue regeneration.

Background Amyloid β peptide (Aβ) is involved in osteoporosis, but the effects of Aβ on osteoblast and bone formation remain unclear. In this study, we investigated the effect of Aβ on bone formation. Methods An animal model of osteoporosis was established by ovariectomy in C57BL/6 mice. The mice received intraperitoneal injection of Aβ. The effect of Aβ on the osteogenic differentiation of human bone marrow stromal stem cells (hBMSCs) and differentiation of both pre-osteoblasts and pre-osteoclasts in a co-culture system were investigated. Results In the animal study, intraperitoneal injection of Aβ for 8 weeks promoted early and late osteogenic differentiation of hBMSCs. Aβ treatment significantly elevated osterix + (osteoblastic) cells but decreased TRAP + cells (osteoclasts) in the distal femur bone. In vitro study showed that Aβ treatment significantly enhanced matrix mineralization and osteogenic markers (Runx2 and osteocalcin). Aβ treatment activated Wnt/β-catenin signaling in hBMSCs. The effect of Aβ was blocked by DKK1 (a Wnt/β-catenin inhibitor) treatment. In the co-culture system, Aβ treatment significantly increased the ALP activities of MC3T3-E1 cells (pre-osteoblasts) but reduced the TRAP + RAW264.7 cells (pre-osteoclasts). Aβ treatment upregulated TCF1 and OPG proteins in MC3T3-E1 cells. Aβ treatment upregulated IκB-α but downregulated NFATc1protein in RAW264.7 cells. These effects were blocked by XAV-939 (a Wnt signaling antagonist), and then rescued by additional Wnt3a (a Wnt agonist). Conclusion Aβ treatment simultaneously promoted osteogenic differentiation via Wnt/β-catenin signaling, and inhibited osteoclasts differentiation via the OPG/RANKL/RANK system, suggesting Aβ is a positive regulator of osteoblast differentiation and bone formation.

β-Cyclodextrin (β-CD) was modified using octenyl succinic anhydride (OSA) to introduce amphiphilic groups (hydrophilic carboxyl and lipophilic octenyl chains) by esterification under alkaline conditions. The FT-IR results indicated that the OSA-modified β-CD (OCD) showed new absorption peaks of an ester bond and a carboxylate (RCOO–) at 1724 and 1570 cm–1, respectively, confirming the successful preparation of OCD. Then the embedding effects of β-CD and OCD on β-carotene and the emulsifying and antioxidant properties of their inclusion complexes were evaluated. The results of XRD showed that the β-CD (or OCD)/β-carotene inclusion complexes were converted from a cage-type structure to a channel-type structure. AFM and SEM showed that the crystal characteristics and surface morphologies of the inclusion complexes were different from those of the physical mixture. The emulsion stabilized by OCD exhibited smaller droplet sizes and larger zeta-potentials than that stabilized by β-CD. In addition, the inclusion complexes-prepared emulsion exhibited lower POV values and TBARS contents than did the physical mixture. OCD/β-carotene inclusion complexes can improve the physical and oxidative stability of the emulsion, which is of great significance to the food industry.

Chlorothalonil (CHT) suffered from its poor water solubility, which limited the application of CHT and might cause environmental pollution due to the use of organic solvents. In our research, inclusion complexes of hydroxypropyl-β-cyclodextrin (HPβCD) and β-cyclodextrin (β-CD) with CHT were formed to investigate the effect of β-CD and HPβCD on the water solubility, antifungal activity and thermal stability of CHT. The structure and characteristics of the two inclusion complexes were systematically evaluated by nuclear magnetic resonance ( 1 H NMR), scanning electron microscopy (SEM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR) and thermal gravity analysis (TGA). The results of 1 H NMR, FT-IR, SEM and XRD indicated that CHT successfully entered the cavity of β-CD and HPβCD. TGA results proved that the thermal stability of CHT was significantly improved. Molecular modelling demonstrated that cyclodextrin could effectively complex with CHT. In addition, the biological activity test showed that the complexes had better antifungal activity than original CHT. Our research can provide useful information for the more reasonable use of CHT, and cyclodextrin can be more used in agriculture to improve the characteristics of pesticides.

β-1,3-Glucanases have prospective applications in areas such as functional oligosaccharide preparation, plant protection, and breweries. In this study, a glycoside hydrolase (GH) family 17 β-1,3-glucanase (BbGlc17A) from Bacteroidetes bacterium from a microbial mat metagenome from the Great Salt Lake was identified. BbGlc17A catalyzed the hydrolytic conversion of laminarin into β-glucooligosaccharides with polymerization degrees of 3–8. The optimal catalytic conditions of BbGlc17A were pH 6.5 and 30 °C. In addition to its hydrolytic activity, BbGlc17A also exhibited transglycosidase activities, involving catalysis of the formation of new β-1,6-glycosidic bonds. BbGlc17A exhibits the classic (β/α)8 TIM-barrel structure and possesses an elongated catalytic groove, distinguishing it from other typical β-1,3-glucanases, which promote the forward direction of the transglycoside reaction. This effectively highlights the potential of the enzyme to convert β-1,3-glucan into mixed functional oligosaccharides. These results reveal the catalytic properties and the application potential of the GH family 17 β-1,3-glucanase and provide valuable information about the group of carbohydrate-active enzymes in biochemistry.

A novel linear polyester with acid-labile β -thiopropionate groups was synthesized via the direct lipase-catalyzed polycondensation of methyl 3-((2-hydroxyethyl)thio)propanoate (MHETP) using immobilized lipase B from Candida antarctica (CALB). The influences of reaction conditions such as polymerization temperature and time on the molecular weight of the poly( β -thioether ester) (PTE) were studied. Ring-opening and condensation copolymerization of MHETP with ε -caprolactone (CL) led to the synthesis of novel poly( β -thioether ester- co - ε -caprolactone) copolymers , poly(TE- co -CL), with well-defined composition and narrow polydispersity . The random copolymers were characterized by 1 H NMR, 13 C NMR, 2D NMR , GPC , TGA and DSC . The degradation of the poly( β -thioether ester) under acidic conditions was investigated by GPC and 1 H NMR spectroscopy.

Magnetic mesoporous nanoparticles of Fe3O4@mSiO2 were prepared through co-precipitation and tetraethoxysilane hydrolysis at 60 °C and pH 10 with hexadecyl trimethyl ammonium bromide as the template. Hydroxypropyl β-cyclodextrin was modified onto Fe3O4@mSiO2 under sodium citrate and N2 protection via an ultrasound process to increase the catalytic efficiency. The prepared nanoparticles were characterized by Fourier transform infrared spectrometry, X-ray diffraction, scanning electron microscopy and a peroxidase-like activity assay. The results showed that the prepared nanoparticles have magnetic properties, peroxidase-like activity and a loose spherical cluster structure. Compared with Fe3O4@mSiO2 nanoparticles, Fe3O4@mSiO2@HP-β-CD nanoparticles exhibit higher catalytic ability toward both H2O2 and 3,3′,5,5′-tetramethylbenzidine. Fe3O4@mSiO2@HP-β-CD nanoparticles can catalyse β-estradiol (β-E2) oxidation in the presence of H2O2 and be used as a colourimetric sensor for indirect detection of β-E2. A good linear relationship was obtained from 0.8 μM to 16 μM. The limit of detection of the proposed method was 0.2 μM. The visual method was successfully used in the analysis of β-E2 in commercial tablets and animal feeds, with recovery ranging from 92.6% to 110%.

For conventional emulsions used to encapsulate easily degradable bioactive compounds, achieving small droplet size and high encapsulation capacity is a challenging. Pickering emulsions stabilized by self-aggregated chitosan particles may offer high encapsulation efficiency due to the robust mechanical barrier formed by solid particles adsorbed at the oil-water interface. Therefore, the effects of pH, chitosan concentration, oil volume fraction, homogenization pressure, and homogenization cycle on the stability of chitosan Pickering emulsions and the degradation of β-carotene were investigated. Effective interfacial adsorption of chitosan nanoparticles and moderate homogenization intensity facilitated the formation of small emulsion droplets. Unlike conventional emulsions, chitosan Pickering emulsions with smaller droplets provided enhanced protection for β-carotene. This enhancement was primarily attributed to the improved interfacial coverage of chitosan nanoparticles with smaller droplet sizes, which was advantageous for β-carotene protection. The optimal conditions for preparing β-carotene-loaded chitosan Pickering emulsions were as follows: pH 6.5, chitosan concentration of 1.0 wt%, oil volume fraction of 20 %, homogenization pressure of 90 MPa, and 6 homogenization cycles. These findings indicate that chitosan Pickering emulsions are well-suited for encapsulating β-carotene with both small droplet size and high encapsulation efficiency.

Fabricating food packaging films with multifunction (antibacterial, antioxidant, and controlled-release properties) represents a powerful strategy for extending the shelf life and enhancing the safety of perishable fruits. Herein, chitosan (CS) and polyvinyl alcohol (PVA) composite films were loaded inclusion complex (IC) with different concentrations of β-acids/β-cyclodextrin (β-acids/β-CD) or β-acids/hydroxyethyl-β-cyclodextrin (β-acids/HE-β-CD) inclusion complex. Moreover, the films' physical, chemical, and biological properties were systematically investigated. Satisfyingly, these composite films exhibited excellent mechanical properties and thermal stability. As the concentration of the inclusion complex improved, the hydrophobicity of the functionalized films also enhanced (76.50°–108.59°/110.04°). Furthermore, variations in pH were discovered that it could affect β-acids release from films. A significant enhancement in the antioxidant capacity of films could be clearly observed with increased concentrations of the inclusion complex (from 5.56 % to 72.98 %/73.47 %). Additionally, films expressed outstanding antibacterial activity against Gram-positive bacteria ( S. aureus ) (inhibition zone >10.89 mm), with those loading β-acids/β-CD demonstrating a stronger antibacterial effect than those loading β-acids/HE-β-CD. For the preservation of strawberries – the typical representative of perishable fruits, the films significantly extended their shelf life to eight days. As the content of the inclusion complex of β-acids in the film increased, the weight loss dramatically reduced (39.91 % without film vs. 23.12 % with HE-β-CD@CS/PVA film). Overall, the CS/PVA films loading β-acids/β-CD display remarkable multifunctionality and value as a fresh-preserving packaging material with sustained release performance and notable bioactivity.

Antibiotic residues in aquatic environments pose a persistent threat to ecosystems and human health, necessitating efficient remediation strategies. This study presents a supramolecular-engineered type-II heterojunction that integrates β-cyclodextrin (β-CD) surface functionalization with a coral-like BiOIO3/β-Bi2O3 heterostructure for enhanced visible-light photocatalytic degradation of oxytetracycline hydrochloride (OTC). β-CD functionalization simultaneously extends visible-light harvesting through supramolecular sensitization and concentrates OTC molecules at the catalyst surface via host–guest inclusion complexation, which collectively accelerates interfacial charge-transfer kinetics. Concurrently, the staggered type-II band alignment between BiOIO3 and β-Bi2O3 promotes efficient spatial separation of photogenerated carriers, suppressing recombination. The optimized composite (2.5β-CD/BiOIO3/30β-Bi2O3) achieves 87.34% OTC degradation within 60 min, exhibiting a pseudo-first-order rate constant 2.87-fold that of pristine BiOIO3. Comprehensive characterizations confirm the heterojunction architecture and reactive oxygen species evolution. The catalyst exhibits satisfactory photostability after being used 4 times. Degradation pathways are elucidated via high-performance liquid chromatography-mass spectrometry (HPLC-MS), while density functional theory (DFT) calculations of electrostatic potential, Highest Occupied Molecular Orbital (HOMO), and Lowest Unoccupied Molecular Orbital (LUMO) distributions identify the preferential reactive sites of OTC, providing molecular-level mechanistic insight. This work decouples the synergistic contributions of surface supramolecular engineering and heterojunction band engineering, offering a design strategy for photocatalytic remediation of refractory pharmaceutical pollutants.

The transforming growth factor-βs (TGF-βs) are multifunctional cytokines capable of regulating a wide range of cellular behaviors and play a key role in maintaining the homeostasis of the immune system. The TGF-β subfamily, which is only present in deuterostomes, expands from a single gene in invertebrates to multiple members in jawed vertebrates. However, the evolutionary processes of the TGF-β subfamily in vertebrates still lack sufficient elucidation. In this study, the TGF-β homologs are identified at the genome-wide level in the reissner lamprey (Lethenteron reissneri), the sea lamprey (Petromyzon marinus), and the Japanese lamprey (Lampetra japonica), which are the extant representatives of jawless vertebrates with a history of more than 350 million years. The molecular evolutionary analyses reveal that the lamprey TGF-β subfamily contains two members representing ancestors of TGF-β2 and 3 in vertebrates, respectively, but TGF-β1 is absent. The transcriptional expression patterns show that the lamprey TGF-β2 may play a central regulatory role in the innate immune response of the lamprey since it exhibits a more rapid and significant upregulation of expression than TGF-β3 during lipopolysaccharide stimuli. The incorporation of BrdU assay reveals that the lamprey TGF-β2 recombinant protein exerts the bipolar regulation on the proliferation of the supraneural myeloid body cells (SMB cells) in the quiescent and LPS-activated state, while plays an inhibitory role in the proliferation of quiescent and activated leukocytes in lampreys. Furthermore, caspase-3/7 activity analysis indicates that the lamprey TGF-β2 protects SMB cells from apoptosis after serum deprivation, in contrast to promoting apoptosis of leukocytes. Our composite results offer valuable clues to the origin and evolution of the TGF-β subfamily and imply that TGF-βs are among the most ancestral immune regulators in vertebrates.
