Understanding the interactions between mercury and microalgae, especially the interactions between inorganic mercury (IHg) and extracellular polymeric substances (EPS, a protective barrier between cells and their external environment), is essential for elucidating mercury’s toxicological mechanisms. Given the inherent cell heterogeneity, a novel analysis system of an online viscoelastic fluid focusing chip-time-resolved analysis inductively coupled plasma mass spectrometry has been developed to investigate the bioaccumulation of HgS nanoparticles and Hg2+ in single Microcystis aeruginosa (M. aeruginosa) cells, exploring the interaction mechanisms between HgS/Hg2+ accumulation in algal cells and EPS. The single-cell analysis results reveal minimal bioavailability of HgS within algal cells, with mercury’s toxicity to M. aeruginosa being species-dependent. Notably, algal cells exhibited more heterogeneity in HgS uptake than in Hg2+ uptake. Under Hg2+/HgS stress, M. aeruginosa cells with EPS removed (EPS-R algal cells) showed an increased level of bioaccumulation of mercury compared to those with EPS (EPS-C algal cells), highlighting the critical role of EPS in mercury bioaccumulation. Overall, the designed viscoelastic fluid microfluidic focusing chip integrates focusing and cleaning functions, featuring easy fabrication, simple operation, low sample loss, and relatively high throughput. Under the optimal conditions, the sample throughput is 1195 min–1 and the cell recovery is 90%. Besides, this research offers novel insights into the interaction mechanisms between Hg2+/HgS and EPS in microalgal cells and unveils the specific toxic effects of Hg2+/HgS on M. aeruginosa at the single-cell level, contributing to a deeper understanding of mercury’s ecological and toxicological impact in aquatic environments.

An analytical technique for selective, simultaneous determination of bisphenol A (BPA) and nonyl phenol (NP) in fish samples was established by a solid-phase extraction chip integrated with array columns of dual-molecularly imprinted polymer-coated silver-modified graphene oxide (Ag/GO-dual-MISPE-chip). Ag/GO dual-molecularly imprinted polymers (Ag/GO-dual-MIPs) were synthesized by in situ polymerization using Ag/GO as the supporting matrix which extracted templates easily. The affinity and specificity on the Ag/GO-dual-MISPE monolithic column were also investigated. The array extraction chip showed high binding capacity and selectivity to BPA and NP, with the imprinting factors of BPA and NP reaching 2.6 and 2.9, respectively. Ag/GO-dual-MIPs were characterized by scanning electron microscopy and Fourier transform-infrared (FT-IR) spectroscopy. As Low as 2.4 ng L −1 BPA and 4.7 ng L −1 NP were detected by high performance liquid chromatography coupled with fluorescence detection. The enrichment factor was 113-fold for BPA and 92-fold for NP. Therefore, the chip-based array columns are feasibly applicable to facile extraction of BPA and NP and effective clean-up of impurities in fish samples.

Foodborne pathogens endanger public health and rapid, sensitive and accurate detection of them is vital. Argonaute stands in the frontline as the next generation nucleic acid detection tool, bearing a few comparable advantages. Centrifugal microfluidic chips (CMCs) use centrifugal force to achieve liquid flowing, mixing and reaction, eliminating complicated designs of valves and pumps. In this study, for the first time, we devised a Thermus thermophilus A rgonaute ( Tt Ago)-powered centrifugal microfluidic chip reaction biosensor for S ample-in- A nswer-out detection of P athogen S. aureus (termed as ASAP) with ultrahigh rapidity and sensitivity in a one-pot fashion. The samples subjected to testing were mixed with a home-made nucleic acid fast extraction reagent and then the mixture was injected into the sample cell of CMC, which was centrifuged down into the reaction cell. The reaction cell was preloaded with both LAMP (Loop-mediated isothermal amplification) and Tt Ago systems. As such, the species-specific nuc genes were amplified by LAMP, while the Tt Ago performed site-specific cleavage to output fluorescent (FL) signals. The sample-to-answer time was 17 min, and the limit of detection (LOD) reached 1 CFU/mL. ASAP was able to perform simultaneous multiplexed detection and up to 16 samples can be detected at the same time. ASAP reduced reagent consumption and minimized the influence of carry-over and cross-contamination. ASAP was capable of detecting S. aureus in foods, but also detecting physiological samples from infect, holding great promise for practical applications. Overall, our work has enriched the Argonaute-powered biosensing and CMC biosensor technology by providing a conceptually novel bacterial detection platform.

We aimed to explore the optimization and application of a bionic system of dynamic co-culture with hepatocytes and renal cells based on the microfluidic chip technique in evaluating emodin, which might replace the conventionally cytological evaluation technique of health food. After optimal experiments, the improved bionic system was composed of human hepatocellular carcinoma cells (HepG2), human renal glomerular endothelial cells (HRGECs), rat tail collagen type I, and gelatin with optimized concentrations (1.3 mg/mL + 7.5%). The applicability of the bionic system indicated that the growth stability was appropriate (CV: 7.36%), and the cell viability of that gradually decreased with the increasing of emodin concentration from 0–100 μM, which statistic significances were at 50 and 100 μM (p< 0.05), and the stained results of dead/live cells also showed the same trend. The LDH level appeared rising trend after decline between 0 μM and 100 μM emodi, and the level of that at 100 μM emodin was significantly higher than that at 25 μM and 50 μM emodin, respectively. The BUN level continuously and significantly declined with the increasing of emodin concentration (p< 0.05). Our research realized the application of this optimized bionic system in evaluating emodin, and provided a useful platform and reference for further in vitro alternative research with regard to evaluating the efficacies of health food in the future. Keywords:bionic system;co-culture of hepatocytes and renal cells;microfluidic chip;emodin;raw material of health food

Conventional fluorescence (FL) biosensors based on intensity quantification mode, are susceptible to individual or detector differences, and background light. In this study, a multi-channel microfluidic chip (D-chip) based on visual fluorescence distance-readout mode was self-made for rapid and simultaneous detection of three bacteria. In the chip, different phages for specifically capturing three kinds of bacterial strains were pre-modified in the channels, respectively. After the sample with multiplex bacteria was pumped into the chip, different target bacteria could be captured by corresponding channels and travel in the flow direction, respectively. Subsequently, novel aggregation-induced emission fluorophore photosensitizer-labeled antimicrobial peptides (AIE@AMPs) signal tags were used to conjugate with all captured bacteria to boost their light emissions. The AIE@AMPs can enhance FL signals after conjugating with bacteria. Consequently, the qualification and quantification of different species of bacteria can be carried out by measuring corresponding luminous channel distances. Under optimized conditions, the D-chip with FL distance readout mode was successfully applied for detecting Escherichia coli O157:H7 ( E. coli 44484) and two Salmonella typhimurium strains (S. T 14028 and S. T 25928) within 30 min. The linear range for E. coli was from 10 2 to 10 6 CFU/mL, with a limit of detection (LOD) of 64 CFU/mL. For two S. T strains , the linear range is from 10 2 to 10 6 CFU/mL, with an LOD of 58 CFU/mL. The multi-channel microfluidic platform based on naked-eye FL distance readout mode shows great potential for practical application in on-site screening of pathogen bacteria.

Recently, sustainable biochar composites created from waste biomass and other benign materials have attracted increasing attention for their promising potential in the removal of organic contaminants from environmental media. A facile approach combining coprecipitation-hydrothermal synthesis with buried carbon pyrolysis was used to produce composites composed of Mg-Al hydrotalcite and hickory-chip/peanut-shell biochar which was tested for its ability to remove anionic Congo red (CR) from aqueous solution. The composites exhibited high specific surface area, high micro-porosity, and abundant oxygen/carbon-containing functional groups. The sorbents derived from hickory and peanut shell exhibited maximum experimental CR adsorption capacities of 1045.6 and 1195.0 mg g -1 , respectively (20 times of pristine biochar) with removal rates of 86.1% and 98.3%, respectively, superior to those of hydrotalcite and many other advanced CR sorbents. The work highlights the potential of hydrotalcite/biochar composites as a promising adsorbent for water treatment and environmental applications.

Self-driven microfluidic systems have attracted significant attention and demonstrated great potential in the field of point-of-care (POC) testing due to their device simplicity, low power consumption , increased portability, and reduced sample consumption. To develop POC detection chips with diverse characteristics that meet different requirements, there is a strong demand for feasible strategies that enable easy operation and reduce processing time. Here, a one-step processing approach using femtosecond laser direct writing technology was proposed to fabricate a capillary-actuated POC microfluidic chip. The driving force of the chip is highly dependent on its surface wettability , which can be easily adjusted by changing the laser processing parameters . This POC microfluidic chip allowed for the detection of intracellular H 2 O 2 through a catalytic reaction system that incorporated 5-aminosalicylic acid -sensitized colloidal TiO 2 nanoparticles and horse radish peroxidase , with integrating semiconductor-based surface-enhanced Raman scattering (SERS) quantitative technique. The concentration of H 2 O 2 was determined by the SERS signal of the catalytic products in the microfluidic chip, resulting in rapid detection with minimal sample consumption. Our method provides a simple, feasible, and alternative strategy for POC testing of H 2 O 2 , with a linear range of 10 −2 ∼10 −6 M and a limit of detection of 0.55 μM. This approach was successfully applied to rapid detection of intracellular H 2 O 2 in MCF-7 breast cancer cells with high sensitivity and minimal sample consumption. Additionally, this study not only demonstrates the exceptional advantages of femtosecond laser processing technology in fabricating diverse microfluidic chips for various applications, but also presents an efficient POC testing strategy for detecting cell signaling molecules .

The fate of stem cells at the single cell level with limited communication with other cells is still unknown due to the lack of an efficient tool for highly accurate molecular detection. Moreover, the conditional sensitivity of biological experiments requires a sufficient number of parallel experiments to support a conclusion. In this work, a microfluidic single cell chip is designed for use with a protein chip to investigate the effect of hydroxyapatite (HAp) on the osteogenic differentiation of human adipose-derived stem cells (hADSCs) in situ at the single cell level. By successfully detecting secretory proteins in situ, it is found that the HAp nanorods enhance osteogenic differentiation at the single cell level. In the chip, the single cell seeding approach confirms the osteogenic differentiation of the hADSCs, which endocytoses HAp, by reducing the influence of the factors secreted by neighboring differentiating cells. Most importantly, more than 7000 microchambers provide a sufficient number of parallel experiments for statistical analysis, which ensure a high level of repeatability of the HAp nanorod-induced osteogenic differentiation. The microfluidic chip comprising single cell culture microchambers with in situ detection capability is a promising tool for research on cell behavior or cell fate at the single cell level.

It is urgent to obtain targeted drugs that selectively bind to pathological targets rather than physiological targets in the early stage of drug screening. G-Quadruplex has become one of the important targets in the development of anti-tumor drugs. However, drugs that target quadruplexes may also bind to dsDNA, which may lead to adverse reactions. In this study, a new three-phase laminar flow chip was constructed to enable the multi-components of a traditional Chinese medicine extract to dynamically and competitively bind with G-quadruplex DNA (on target) and double-stranded DNA (off target), so as to select high-efficiency and low-toxicity anti-tumor drugs. The results showed that there were five compounds in the extracts of Macleaya cordata seeds that exhibited obvious differences in binding to the two targets. Furthermore, the binding constants and modes of four identified alkaloids as they bound to two DNA targets were verified by fluorescence spectra and molecular docking methods. The toxicity to HepG2 and LO2 cells from the four alkaloids was also compared. The results showed that sanguinarine and chelerythrine could be used as candidate drugs with stronger binding to HT24 than DNA26. The chip can also be used for other types of double-target screening of other traditional Chinese medicine extracts or compound libraries.

Highly-intelligent user-friendly sensing strategies are becoming desperately demanded and indispensable in under-developed nations and resources-insufficient areas. Herein, an intelligent sensing platform has been successfully established incorporating smartphone with machine vision and deep learning function, 3D-printed handheld device, and paper microfluidic chip stained with tri-emission ratiometric-fluorescence (FL) carbon dots (TCDs). The intelligent system can quantify zinc ion (Zn 2+ ) and ethylenediaminetetraacetic acid (EDTA) only through a single-step operation on smartphone. TCDs, engendering bright blue and cyan FL as well as weak red one at 370 nm excitation, were prepared hydrothermally using tartaric acid and alizarin carmine. As-prepared TCDs exhibit continuous FL color transformation from blue to purple to blue with the addition of Zn 2+ and EDTA. Subsequently, TCDs-based paper chip is explored to enable field-portable sequential discrimination of Zn 2+ and EDTA. Intriguingly, the proposed sensor is utilized for fast and intelligent quantification of Zn 2+ and EDTA in soil filtrate, plant sap and tap water with a low detection limit of 3.8 μM and 7.9 μM, as well as wide linear correlations of 0–280 µM and 0–560 µM, respectively. It is eagerly anticipated that our methodologies will become the strong impetus for new round of technological transformation for analyte assay.
