Construction, stability and photodynamic germicidal efficacy of curcumin nanoemulsion stabilised with Maillard conjugate of Wpi-Rha

Hong Zhuang,Xinyu Jiang,Sijia Wu,Xueqian Li,Haiyang Yan

Curcumin has excellent antioxidant and antibacterial properties. The poor solubility and easy degradation limit its application. The present investigation was aimed to prepare curcumin nanoemulsions (Cur-Ne), employing homogeneous and ultrasonication for the encapsulation of curcumin to overcome these problems. Cur-Ne stabilised using whey protein isolate and rhamnose conjugate (Wpi-Rha) was prepared by Maillard reaction. Furthermore, the processing ( i.e . ionic strength, heat and freeze-thaw) and storage stability of Cur-Ne were evaluated. Moreover, the bactericidal capacity of Cur-Ne combined with photodynamic therapy (PDT) technology was estimated. Cur-Ne was prepared successfully with the droplet size of 185.97 ± 6.65 nm, polydispersity index (PDI) of 0.21 ± 0.01, zeta potential of −35.17 ± 0.15mV and entrapment efficiency of 97.0 ± 0.07%. Meanwhile, Cur-Ne was stable at different storage and processing conditions. In addition, the DPPH and ABTS radical scavenging rates of Cur-Ne were 84.86 ± 0.24% and 99.63 ± 0.48%, respectively. At the adding amount of 5 μL of Cur-Ne, the Cur-Ne could deactivate the Escherichia coli and Staphylococcus aureus with the antibacterial rate of 99.05 ± 0.13% and 99.11 ± 0.18%, respectively. Curcumin loaded in nanoemulsion and combined with PDT provide new ideas for food storage and sterilisation.

Cocrystal of curcumin with 4,4′-bipyridine toward improved dissolution: Design, structure analysis, and solid-state characterization

Rui Shen,Jie Zhang,Xi Wang

The prime active ingredient of the herb curcuma longa , curcumin (CUR) has extensive pharmacological activities and high security. However, the low solubility severely restricts its investigation and application as a drug. In this paper, a new cocrystal, CUR–BPY, assembled by CUR and BPY (4,4′-bipyridine) has been synthesized and characterized by various analytical methods. Single-crystal X-ray diffraction reveals that CUR–BPY contains a pentameric BPY–CUR–BPY–CUR–BPY supramolecular architecture connected by O H⋯N hydrogen bonds and further extended to give a 1D loop-like chain through aromatic interactions. Powder dissolution experiment reveals that CUR–BPY exhibits obvious spring phenomenon and about 7 times higher apparent solubility than that of raw curcumin. The improved dissolution performance should be ascribed to the careful selection of BPY coformer and expected O H⋯N weak hydrogen bonding in CUR-BPY cocrystal.

Enhanced curcumin transportation across epithelial barrier by mucus-permeable soy protein nanoparticles-mediated dual transcytosis pathways

Dan Yuan,Qi Li,Qibo Zhang,Feibai Zhou,Qiangzhong Zhao,Mouming Zhao

Nanocarrier-delivered bioactive compounds are highly desirable for their improved stability and applicability, but their bioavailability is still limited due to the strong mucus and epithelial cell barriers. Herein, a series of self-assembled soy protein nanoparticles (SPNPs) with different mucus permeabilities were prepared and their delivery efficiency upon Curcumin (Cur) encapsulation was evaluated. Results demonstrated that the formed SPNPs-Cur exhibited high compatibility and cellular antioxidant accessibility. Besides, SPNPs enhanced the cellular uptake and transmembrane permeation of Cur, especially promoted the transportation of proto-Cur in addition to Cur metabolites. The SPNPs with the rapid mucus diffusion capacity presented more efficient transcytosis across the Caco-2 cell monolayer, which was mediated by a combination of paracellular and transcellular pathways. This work verified that mucus-permeable soy protein nanoparticles could be a promising delivery system for improving the bioavailability of bioactive compounds.

Interaction of curcumin with four types of gelatins in nanoparticles: Mechanism and application for emulsion stabilization

Guangyi Kan,Ye Zi,Cuiping Shi,Yang Tan,Huan Gong,Xichang Wang,Jian Zhong

The research of the interaction between proteins and functional active substances in the protein nanoparticles is a research hotspot in the field of food science . Gelatin type had a significant effect on the structural and functional properties of gelatins. Herein, the interaction mechanism of curcumin with four types of gelatins (bovine bone gelatin, BBG; cold water fish skin gelatin, CFG; fish gelatin , FG; and porcine skin gelatin, PSG) and the structural and functional properties of gelatin/curcumin nanoparticles were explored. All four types of gelatins could be applied to prepare gelatin/curcumin nanoparticles with an amorphous state of curcumin. The nanoparticle sizes were not dependent on the molecular weight of gelatins. The stability of gelatin/curcumin nanoparticle solutions was dependent not on the nanoparticle size but on the gelatin type. The binding affinity between gelatins and curcumin was dependent on gelatin type and the number of binding sites was nearly 1. The main binding forces to form gelatin-curcumin nanoparticles were hydrogen bonds, Pi-alkyl interaction, and Pi-anion interaction. The nanoparticulation increased the surface hydrophobicity of BBG, FG, and PSG, whereas it decreased the surface hydrophobicity of CFG. The nanoparticulation increased the surface tension of gelatins and decreased the dynamic interfacial tension of gelatins. The functional properties were dependent on both gelatin type and nanoparticulation. CFG/Cur nanoparticulation could not change the creaming stability, whereas other gelatins/curcumin nanoparticulation significantly increased creaming stability than the corresponding gelatins. This work provided the basic knowledge to illustrate the interaction and application of protein nanoparticles for Pickering emulsion stabilization.

Coating carboxymethylpachymaran on bovine serum albumin (BSA) nanoparticles for the encapsulation and oral delivery of curcumin

Lan Wang,Jin Mao,Qi Zhou,Qianchun Deng,Lei Zheng,Jie Shi

Curcumin is a hydrophobic substance with various beneficial biological activities. Its weak solubility and instability, however, now restrict its usage as a functional food. Herein, the purpose of this work is to fabricate curcumin-loaded bovine serum albumin/carboxymethylpachymaran nanoparticles (BMCNPs) to overcome these challenges. Under optimal conditions, the BMCNPs were spherically shaped with a surface charge of −31.4 mV, polydispersity index of 0.16 and particle size of 158.3 nm were characterized by electron microscopy and dynamic light scattering. The interactions between curcumin, bovine serum albumin (BSA), and carboxymethylpachymaran (CMP) were investigated. BMCNPs had a much better curcumin encapsulation efficiency (74.1%) as well as strong antioxidant activity, storage stability, and ionic strength tolerance. The in vitro release revealed that curcumin was more successfully sustained in the small intestine stage (68.7%) and that BMCNPs were able to prevent it from releasing too early in the stomach (21.4%). Not to mention, the results of the cytotoxicity test performed on the HT-29 and IEC-6 cell lines demonstrated the acceptable biocompatibility of BMCNPs.

Improving the physicochemical stability and release properties of curcumin using κ-carrageenan/scallop hydrolysates hydrogel beads

Jia-Nan Yan,Yu-Qiao Wang,Zhu-Jun Zhang,Yi-Nan Du,Hai-Tao Wu

Scallop ( Patinopecten yessoensis ) male gonad hydrolysates (SMGHs)/ κ -carrageenan (KC)/KCl beads with SMGHs :KC ratios (0:10–5:5) were investigated. SMGHs/KC/KCl-Cur bead (5:5) exhibited the most intact spherical morphology and highest Cur loading content of 0.063 mg/0.1 g bead, ascribing to a shortened T 23 from 1607.9 to 966.4  ms, and red and blueshifts of O H, NH, amide I and II bands. The undetected fingerprint region within 7.82°-28.90° of SMGHs/KC/KCl-Cur beads indicated successful Cur entrapment. Moreover, SMGHs/KC/KCl-Cur beads exhibited shrinkage network backbones and larger void pores as SMGHs increased, with vessel percentage area, total number of junctions, total vessel length decreasing from 52.1, 1446.8, 57931.4 to 39.7, 530.5, 34458.4, and lacunarity increasing from 0.048 to 0.111, respectively. Furthermore, Cur showed approximately 50% release contents in colon phase and above 90% retention rate during 30 days of storage at 4 °C. These results suggested that SMGHs/KC/KCl-Cur beads exhibited sustained-release of Cur and promised stable Cur preservation.

Curcumin-loaded core-shell biopolymer nanoparticles produced by the pH-driven method: Physicochemical and release properties

Zhenpeng Li,Quanquan Lin,David Julian McClements,Yuying Fu,Hujun Xie,Teng Li,Guowen Chen

In this study, core-shell biopolymer nanoparticles were fabricated for the encapsulation and delivery of curcumin using a pH-driven method. The influences of the coating composition on the physicochemical properties and curcumin release characteristics of the core-shell nanoparticles were studied. Fourier transform infrared spectroscopy and X-ray diffraction analyses indicated that curcumin was encapsulated in an amorphous state inside the nanoparticles. Particle size and ζ-potential measurements indicated that the biopolymer nanoparticles were relatively stable under different environmental conditions: long term storage, heating, pH changes and salt. The DPPH radical scavenging activity of the curcumin was increased after encapsulation within the nanoparticles, whereas the gastrointestinal release of curcumin was prolonged. These results were attributed to the ability of alginate and NaCas to form a thick layer around the nanoparticles, which increased the steric and electrostatic repulsion between them, as well as inhibiting the release of curcumin.

Curcumin-loaded liposomes with the hepatic and lysosomal dual-targeted effects for therapy of hepatocellular carcinoma

Yan Wang,Ruihua Ding,Zan Zhang,Cheng Zhong,Jianyi Wang,Mian Wang

Curcumin can induce cancer cell apoptosis through lysosomal permeabilization pathway. However, the poor selectivity of curcumin restricts its use in the therapy of hepatocellular carcinoma . Because galactose group can recognize ASGPR overexpressed on hepatoma cells and morpholine group can target to the lysosome, they are integrated into a dual-targeted lipid material with low toxicity. The corresponding galactose-morpholine modified liposomes loaded with curcumin (Gal-Mor-LPs) were prepared and evaluated in comparison with conventional liposomes (LPs) and galactose modified liposomes (Gal-LPs). The in vitro and in vivo hepatic targeting capacity of liposomes followed a trend of LPs < Gal-LPs < Gal-Mor-LPs. The endocytosis of Gal-Mor-LPs was competitively inhibited by galactose, which confirmed the galactose modified liposomes entered hepatoma cells via ASGPR-mediated pathway. Gal-Mor-LPs displayed more excellent lysosomal targeting efficacy than LPs and Gal-LPs due to the attraction of acidic lysosome on basic morpholine group of Gal-Mor-LPs. The in vivo tumor inhibition effects of formulations also followed a trend of free curcumin < LPs < Gal-LPs < Gal-Mor-LPs, confirming that hepatic and lysosomal dual-targeting vehicle can improve the antitumor efficacy of curcumin. Moreover, the curcumin-loaded liposomes modified with galactose and morpholine moieties show good biocompatibility in vivo .

Curcumin Ameliorates Copper-Induced Neurotoxicity Through Inhibiting Oxidative Stress and Mitochondrial Apoptosis in SH-SY5Y Cells

Xiang Biao,Li Daowen,Chen Yiqiang,Li Meng,Zhang Yuan,Sun Tun,Tang Shusheng

Impaired homeostasis of copper has been linked to different pathophysiological mechanisms in neurodegenerative diseases and oxidative injury has been proposed as the main mechanism. This study aims to use curcumin, a widely used antioxidative and anti-apoptotic agent, to exert the neuroprotective effect against copper in vitro and illuminate the underlying mechanism. The effect of curcumin was examined by using a cell counting kit-8 assay, flow cytometry, immunofluorescence, spectrophotometer, and western blot. Results revealed that after pretreatment with curcumin for 3 h, copper-induced toxicity and apoptosis show a significant decline. Further experiments showed that curcumin not only decreased the production of ROS and MDA but also increased the activities of the ROS scavenging enzymes SOD and CAT. Moreover, curcumin treatment alleviated the decrease in mitochondrial membrane potential and the nuclear translocation of cytochrome c induced by copper. The protein levels of pro-caspase 3, pro-caspase 9, and PARP1 were up-regulated and the Bax/Bcl-2 ratio was down-regulated in the presence of curcumin. Taken together, our study demonstrates that curcumin has neuroprotective properties against copper in SH-SY5Y cells and the potential mechanisms might be related to oxidative stress and mitochondrial apoptosis.

Preparation and characterization of phosphate-stabilized amorphous calcium carbonate nanoparticles and their application in curcumin delivery

Chaohui Rao,Min Li,Xiaoqing Sun,Meilin Li,Xiaojie Lian,Huifang Wang,Lan Jia,Baolong Niu,Wenfeng Li

Nano-sized amorphous calcium carbonate (ACC) have a great potential for drug transport and drug delivery because of their high drug loading capacity, excellent biocompatibility and biodegradability. However, ACC is the thermodynamically least stable phase of calcium carbonate, and such thermal metastability often transforms it into more stable vaterite or calcite phases, so the inhibition of the phase transformation of ACC is of great significance for its application in drug delivery systems. Herein, phosphate as a morphology controlling additive was introduced to stabilize the metastable ACC. The results showed that the composite nanoparticles (named here the ACCP) with narrow size distribution located at approximately 128 nm (measured by dynamic light scattering) were obtained, when Ca 2+ concentration and C/P were 10 mM and 7/3 respectively, prepared at 30 °C for 10 min. The powdered X-ray diffraction (XRD) results showed that phosphate has an excellent ability to stabilize ACC, and even after storage for 60 days at room temperature, the structure of ACCP remains amorphous. The in vitro drug release tests showed that ACCP nanoparticles had a high curcumin (Cur) loading capacity and a sustained drug release property. Moreover, the 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH) radical scavenging activity assay showed that ACCP-encapsulating strategy could effectively prevent the decomposition of Cur. In vitro cytotoxicity tests demonstrated that the resultants have excellent biocompatibility. Therefore, the as-prepared ACCP nanoparticles are promising for drug delivery applications.

Sensitive analysis of doxorubicin and curcumin by micellar electromagnetic chromatography with a double wavelength excitation source

Yu Xiao,Yu Wanxiang,Han Xiufen,Chen Zuanguang,Wang Shumei,Zhai Haiyun

Doxorubicin has been extensively used to treat cancers, and there are recent findings that the anticancer activities can be enhanced by curcumin. Although the two compounds have native fluorescence, they can hardly be quantified directly simultaneously using the laser-induced fluorescence (LIF) detection method. To avoid complex fluorescence derivatization and introduction of interfering components, a highly sensitive double wavelength excitation source LIF (D-W-Ex-LIF) detector composed of a 445-nm and 488-nm commercial laser diode was constructed to detect them simultaneously. Rhodamine 6G was selected as an internal standard, because its fluorescence can be excited at 445 nm and 488 nm. The native fluorescence of doxorubicin and curcumin and their resolution were enhanced by introducing mixed micelles. The optimal electrophoretic separation buffer was 10 mM borate buffer containing 20 mM Triton X-100, 5 mM sodium dodecyl sulfate, and 30% (v/v) methanol at pH 9.00. Therefore, the developed method was specific, accurate, and easily operable. Its limits of detection for doxorubicin and curcumin in human urine samples were 4.00 × 10 −3 and 1.00 × 10 −2  μg/mL, respectively, and the limits of quantification were 1.00 × 10 −2 and 3.00 × 10 −2  μg/mL, respectively. The recoveries were 94.9–109.1%. Graphical abstract

Preparation of Poloxamer188-b-PCL and Study on in vitro Radioprotection Activity of Curcumin-Loaded Nanoparticles

Lin Xiaona,Shi Yongli,Yu ShaSha,Li Siyi,Li Wenhui,Li Meishuang,Chen Shengxi,Wang Yuanbo,Cong Mei

A novel polymer of poloxamer188-b-PCL was synthesized via a ring-opening polymerization. Fourier transform infrared spectroscopy (FTIR), Raman, and 1H nuclear magnetic resonance (1H NMR) spectra were used to study the structures of obtained poloxamer188-b-PCL. The thermo-stability of poloxamer188 -b-PCL was carried out with a thermal gravimetric analyzer (TGA), and cytotoxicity was obtained using the CCK8 method. Cargo-free and curcumin (CUR)-loaded poloxamer188-b-PCL NPs were fabricated via the solvent evaporation method. The morphology, particle size distribution, and stability of cargo-free NPs were studied with a scanning electron microscope (SEM) and laser particle analyzer. The in vitro radioprotection activity of CUR-loaded NPs was performed. FTIR, Raman, and 1H NMR spectra confirmed that poloxamer188-b-PCL was obtained. TGA curves suggested poloxamer188-b-PCL had better thermo-stability than original poloxamer188. Cell tests suggested that the cargo-free NPs had no cytotoxicity. SEM image showed that the cargo-free NPs were spherical with a diameter of 100 nm. Free radical scavenging experiments proved that CUR-loaded NPs had better antioxidant activity than CUR solutions. CUR-loaded NPs could be detected in all tissues, including liver, kidneys and lung. In summary, this work demonstrated a feasibility of developing an injective formulation of CUR and provided a protection agent in caner radiotherapy.
