With mercury pollution causes serious threats to ecosystems and human health because of its extreme toxicity and bioaccumulation, the development of efficient removal technologies has become an urgent environmental priority. This study introduces a novel adsorbent thiazolo[5,4- d ]thiazole conjugated microporous polymer (TzTzCMP) for efficient Hg(II) removal from wastewater. TzTzCMP synthesized via condensation of dithiocarbamate and triformyl-phenol, exhibits a coral-like porous structure with a high specific surface area of 436.86 m 2 /g and sulfur-rich active sites containing 15.15 wt% sulfur, enabling exceptional Hg(II) adsorption. Experiments demonstrated rapid kinetics, reaching equilibrium within 10 min, and a maximum adsorption capacity of 1050 mg/g. The adsorption kinetics and isotherm data for Hg(II) on TzTzCMP are well described by the pseudo-second-order and Langmuir models, respectively, indicating that chemical interactions dominate the adsorption process. The material maintained >95 % efficiency over five regeneration cycles and showed good selectivity under coexisting ion interference, attributed to Hg(II) coordination with N/S atoms in TzTzCMP, as confirmed by X-ray Photoelectron Spectroscopy (XPS) analysis and density functional theory (DFT) calculations. Fixed-bed column tests achieved 99.99 % removal, reducing effluent concentration to 1 μg/L and meeting drinking water standards. Combining high adsorption capacity and removal rate, rapid adsorption efficiency, good stability and reusability, TzTzCMP presents a promising solution for industrial Hg(II)-contaminated wastewater treatment.

Non-alcoholic fatty liver disease (NAFLD) is the main cause of chronic liver disease worldwide. Bupleurum is widely used in the treatment of non-alcoholic fatty liver, and saikosaponin D (SSD) is one of the main active components of Bupleurum. The purpose of this study was to investigate the efficacy of SSD in the treatment of NAFLD and to explore the mechanism of SSD in the improvement of NAFLD based on “gut-liver axis”. Our results showed that SSD dose-dependently alleviated high fat diet-induced weight gain in mice, improved insulin sensitivity, and also reduced liver lipid accumulation and injury-related biomarkers aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Further exploration found that SSD inhibited the mRNA expression levels of farnesoid X receptor ( Fxr ), small heterodimer partner ( Shp ), recombinant fibroblast growth factor 15 ( Fgf15 ) and apical sodium dependent bile acid transporter ( Asbt ) in the intestine, suggesting that SSD improved liver lipid metabolism by inhibiting intestinal FXR signaling. SSD can significantly reduce the gut microbiota associated with bile salt hydrolase (BSH) expression, such as Clostridium . Decreased BSH expression reduced the ratio of unconjugated to conjugated bile acids, thereby inhibiting the intestinal FXR. These data demonstrated that SSD ameliorated NAFLD potentially through the gut microbiota-bile acid-intestinal FXR pathway and suggested that SSD is a promising therapeutic agent for the treatment of NAFLD.

Two-dimensional conjugated metal organic frameworks (2D c-MOFs) hold significant promise as electrode materials for alkali metal ion batteries while their electrochemical properties still lack reasonable and effective regulation. Here, two representative 2D c-MOFs (M-HHTQ/M-HHTP, M=Cu or Ni) as positive electrodes are used as models to explore the basic/microscopic principles of their complex storage mechanism in sodium ion batteries (SIBs). It is demonstrated that the energy storage mechanism of 2D c-MOFs is determined by the interaction between coordination covalent bonds and organic linkers. Theoretical calculations and experiment results have jointly demonstrated that the redox potential and theoretical capacity can be regulated based on the valence of M-O bond and the utilization of anions and cations, respectively. As a result, Cu-HHTQ achieves a high discharge voltage at 2.55 V (vs. Na + /Na), a higher stable specific capacity of 208 mAh g −1 at 0.05 A g −1 , and long cyclability with the capacity retention rate of 100% at 1 A g −1 after 2000 cycles.

Costunolide, a sesquiterpene isolated from Vladimiria souliei (Franch.) Ling, is known to exhibit anti-inflammatory, anti-viral, and anti-tumor activities. However, the effects of costunolide on liver injury are poorly understood. The current study aimed to investigate the hepatoprotective effects of costunolide against lipopolysaccharide (LPS) and D-galactosamine-induced acute liver injury (ALI) in mice. The results indicated that costunolide (40 mg/kg) could significantly improve the pathological changes of hepatic tissue, and reduced the LPS and D-galactosamine-induced increases of alanine aminotransferase (from 887.24 ± 21.72 to 121.67 ± 6.56 IU/L) and aspartate aminotransferase (from 891.01 ± 45.24 to 199.94 ± 11.53 IU/L) activities in serum. Further research indicated that costunolide significantly reduced malondialdehyde content (from 24.56 ± 1.39 to 9.17 ± 0.25 nmol/ml) and reactive oxygen species (from 203.34 ± 7.68 to 144.23 ± 7.12%), increased the activity of anti-oxidant enzymes superoxide dismutase (from 153.74 ± 10.33 to 262.27 ± 8.39 U/ml), catalase (from 6.12 ± 0.30 to 12.44 ± 0.57 U/ml), and total anti-oxidant capacity (from 0.64 ± 0.06 to 6.29 ± 0.11 U/ml) in hepatic tissues. Western blot results revealed that costunolide may trigger the anti-oxidative defense system by inhibiting kelch-like ECH-associated protein 1 and nuclear factor-related factor 2 (cytosol), increasing nuclear factor-related factor 2 (nucleus), heme oxygenase-1 and NAD (P) H quinone oxidoreductase 1 activity. Moreover, costunolide significantly decreased the protein expression of proinflammatory cytokines including interleukin 1β, interleukin 6, and tumor necrosis factor. Pretreatment with costunolide could reduce the expression of toll-like receptor 4, myeloid differentiation factor 88, p65 (Nucleus), phosphorylated IκB kinase α/β, inhibitor of nuclear factor kappa-B kinase, inhibitor kappa Bα and prevent the expression of phosphorylated inhibitor kappa B kinase which repressed translocation of p65 from cytoplasm to nucleus. In addition, pretreatment with costunolide also inhibited hepatocyte apoptosis by reducing the expression of B-cell lymphoma 2 associated X, cytochrome C, cysteinyl aspartate specific proteinase 3, cysteinyl aspartate specific proteinase 8 and cysteinyl aspartate specific proteinase 9, and by increasing B-cell lymphoma 2. From the above analysis, the protective effects of costunolide against LPS and D-galactosamine-induced ALI in mice may be attributed to its anti-oxidative activity in nuclear factor-related factor 2 signaling pathways, anti-inflammatory suppression in nuclear factor-kappa B signaling pathways, and inhibition of hepatocyte apoptosis. Thus, costunolide may be a potential therapeutic agent in attenuating LPS and D-galactosamine -induced ALI in the future.

Objectives: To elucidate the role of a novel carbapenem-hydrolysing class D β-lactamase (RAD-1) from Riemerella anatipestifer. Methods: We applied WGS and bioinformatic analysis to screen putative β-lactamase genes in R. anatipestifer SCVM0004. A putative class D β-lactamase gene was cloned into pET24a and transferred into Escherichia coli BL21 (DE3) for antibiotic susceptibility determination and protein purification. Meanwhile, the purified native protein was used to determine the enzymatic activities. Results: A class D β-lactamase, RAD-1, was identified in the genome of R. anatipestifer SCVM0004. It was distinct from all characterized class D β-lactamases (≤42% amino acid sequence identity). Searching in GenBank showed that blaRAD-1 was widely disseminated among R. anatipestifer. Genomic environment analysis indicated that the chromosomal structures of blaRAD-1-located regions were relatively conserved. Expression of RAD-1 in E. coli results in elevated MICs for various β-lactam antibiotics, including penicillins, extended-spectrum cephalosporins, a monobactam and carbapenems. Moreover, kinetic analysis of purified RAD-1 revealed: (i) high-level activity against penicillins; (ii) highest affinity for carbapenems; (iii) moderate hydrolysis of extended-spectrum cephalosporins and a monobactam; and (iv) no activity for oxacillin and cefoxitin. Conclusions: This study identified a novel chromosomally located class D carbapenemase RAD-1 (Bush-Jacoby functional group 2def) in R. anatipestifer SCVM0004. Moreover, bioinformatic analysis confirmed that the RAD-1 was widely prevalent and conserved in R. anatipestifer.

Phospholipase D (PLD) activity plays a critical role in tumor progression, yet its highly sensitive detection remains challenging. In this study, we developed metal halide perovskite nanoprobes via a dual-anchoring strategy that employed co-encapsulation of phospholipid and cholesteryl hemisuccinate (Chs) for the sensitive and selective monitoring of PLD activity. The Chs molecule not only enhances the stability of the phospholipid membrane via its cholesterol moiety but also passivates surface defects of the perovskite through the succinate group, endowing the nanoprobes with exceptional optical properties (PLQY >90 %) and long-term aqueous stability (> 7 days). The nanoprobe operates via a turn-off fluorescence response triggered by PLD-specific hydrolysis of the phospholipid layer, achieving a low detection limit of 0.51 U/L and a wide linear range (5–2000 U/L) with good anti-interference capability. Furthermore, the nanoprobes were successfully applied to in situ imaging of PLD activity in living cells, enabling discrimination between tumor cells with different PLD expression levels. This study not only paves the way for designing novel perovskite-based probes for biomedical applications but also offers a valuable fluorescent imaging tool to monitor tumor progression.

Intercalation pseudocapacitance is the key point to achieving the balance between the volumetric capacitance and the rate performance of energy storage. However, the low pseudocapacitive reactivity of the active center still limits the energy density of the pseudocapacitance devices. Herein, we propose a strategy that regulates the p - d hybridization of confined MXene via the co-doping of B and O atoms (BO-MXene) for outstanding electrochemical performance. Both experiments and density functional theory (DFT) reveal that the inner doped O atoms in low 2 p orbital energy strengthen the hybridization of Ti and electron-deficient B in MXene for high retention of B atoms, weaken the Ti-O covalency in MXene. As a result, the weakened p - d hybridization of Ti and O for the outer surface and strengthened p - d hybridization of Ti and B for the inner, promote the adsorption of protons and electron transfer, respectively. Further, the BO-MXene-based hybrid supercapacitors achieve a significant volumetric energy density of 56.86 Wh L -1 at 25.6 kW L -1 . This electronic structure via tuning the p - d hybridization in MXene proposes a new path for designing proton-confined intercalation pseudocapacitance.

Ammonia (NH3), the second most produced chemical in the world, is predominantly utilized in fertilizer production to underpin modern agriculture and has recently attracted interest as an energy-dense source of renewable energy. Herein, an amorphous CuTe porous nanonets (CuTe PNs) catalyst with p-d orbital hybridization was designed and prepared for NH3 synthesis via the nitrite reduction reaction. The porous and amorphous structure of CuTe PNs provides sufficient channels for molecular penetration and enhances active site accessibility through its abundant defects and vacancies, facilitating electrochemical reactions. The CuTe PNs exhibit excellent nitrite reduction reaction performance (NH3 yield: 633.7 μg h–1 mgcat–1, Faradaic efficiency: 91.41%, NH3 selectivity: 92.2%). This work describes a powerful pathway for the fabrication of p-d orbital hybrid amorphous porous nanonets catalysts and also provides a new possibility for the electrochemical synthesis of NH3.

Phlorizin is the main active ingredient of apple peel and has potential utilization value. Some recent studies have suggested that phlorizin may have antioxidant capacity and protect the liver. The injection of a low dose of D-galactose can cause some changes that resemble accelerated aging in mice. This study explored the protective effects of phlorizin on D-galactose-induced mice and PC12 cells. In this study, ICR mice were divided into a normal group (NOR), a D-galactose model group (D-gal) and phlorizin treatment groups (100 mg kg−1, 200 mg kg−1 and 400 mg kg−1). In addition to the NOR group, four other groups were injected with D-galactose (120 mg kg−1) for 12 weeks. The results showed that phlorizin reduced the decline of strength, coordination and spatial memory caused by aging, increased the activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px), increased total antioxidant capacity (T-AOC), and reduced the content of malondialdehyde (MDA). On the other hand, phlorizin increased the levels of interleukin-2 (IL-2) and acetylcholine (ACh), reduced the release of interleukin-6 (IL-6), aspartate aminotransferase (AST) and alanine aminotransferase (ALT), and decreased the activity of acetylcholinesterase (AChE) in the brain, improved the expression of antioxidant genes related to the nuclear factor E2-related factor 2 (Nrf2) pathway, and reduced the occurrence of morphological lesions in the hippocampus and liver. In addition, phlorizin improved cell viability and reduced the cytotoxicity of D-galactose-induced oxidative stress in PC12 cells. Meanwhile, the protective effect of phlorizin was abolished in Nrf2 gene knockdown PC12 cells. Furthermore, molecular docking showed that phlorizin could bind Keap1 protein, which can interact with Nrf2 protein. Therefore, these results suggest that phlorizin may delay senescence and enhance antioxidant capacity through the Nrf2 pathway.

Sodium tetraborate pentahydrate (STB) was intercalated into graphene oxide (GO) nanosheets to form a nanocomposite (STB@GO). Subsequently, it was self-assembled on a substrate membrane to prepare STB@GO nanofiltration membrane. The properties of the STB@GO powder samples and the nanofiltration membrane were studied using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), contact angle (CA), and zeta potential . When the STB concentration was 1.0 g/L in the cross-linking reaction, the membrane was described as the STB 2 @GO membrane and exhibited a large interlayer space ( d -spacing = 1.347 nm), high hydrophilicity (CA = 22.2 ° ), and high negative potential (zeta = −18.0 mV). Meanwhile, the pure water flux of the membrane was significantly increased by 56.60% than that of the GO membrane. In addition, the STB 2 @GO membrane exhibited a favorable capability for dye rejection,98.52% for Evans blue (EB), 99.26% for Victoria blue B (VB), 91.94% for Alizarin yellow (AY), and 93.21% for Neutral red (NR). Furthermore, the STB 2 @GO membrane performed better in dye separation under various types and concentrations of dye, pH values, and ions in solution. Thus, this study provides a promising method for preparing laminated GO nanofiltration membranes for dye wastewater treatment .

Compared to that of other tumors, various barriers, such as the blood–brain barrier (BBB), enzymatic barriers, and the blood–brain tumor barrier, severely impede the successful treatment of gliomas. Peptide ligands were frequently used as targeting moieties to mediate brain tumor-targeted drug delivery. LWSW (SYPGWSW) is a recently reported quorum-sensing (QS) peptide that is able to efficiently cross the BBB. Even though linear LWSW traverses the BBB in vitro, its in vivo targeting ability has been greatly impaired due to proteolysis. Here, we developed a stable peptide, DWSW (DWDSDWDGDPDYDS), using the retro-inverso isomerization technique to achieve an enhanced antiglioma effect. In vitro studies have demonstrated that both the LWSW and DWSW peptides possessed excellent tumor-homing properties and barrier-penetration abilities, whereas DWSW exhibited exceptional stability in serum and maintained its targeting ability after serum preincubation. In vivo, DWSW-modified probes and micelles accumulated more efficiently in the glioma region in comparison with LWSW-modified probes and micelles because of full resistance to proteolysis in blood circulation. As expected, DWSW-modified paclitaxel (PTX)-loaded micelles (DWSW Micelle/PTX) exhibited the longest median survival time among glioma-bearing nude mice. Our results suggested that the QS peptide appears to be a promising targeting moiety, with potential applications in glioma-targeted drug delivery.

Fish gelatin-chitosan edible films with D-limonene were successfully prepared, which exhibited exceptional mechanical properties and antimicrobial activity. It has been demonstrated that water-soluble chitosan, fish gelatin, and D-limonene could be a candidate precursor to prepare low cost and high-performance edible food packaging material. The results showed that D-limonene in the films could effectively resist the penetration of light and water because of its hydrophobicity. Moreover, the elongation at break (EAB) increased with the addition of D-limonene, which indicated that D-limonene served as a strong plasticizer for the film. Microscopic characterization showed that D-limonene was uniformly distributed in the as-prepared film. And we found that the film exhibited strong antibacterial activity against Escherichia coli (E. coli). All the results indicate that the as-prepared film could be a promising food packaging.
