A Smart Stimulation–Deadhesion and Antimicrobial Hydrogel for Repairing Diabetic Wounds Infected with Methicillin-Resistant Staphylococcus aureus

Zhongwu Bei,Linghong Zhang,Jianan Li,Qi Tong,Kun Shi,Wen Chen,Yan Yu,Ao Sun,Yang Xu,Jie Liu,Zhiyong Qian

The healing of chronic diabetic wounds is a common and significant challenge in the medical field. Despite extensive efforts, the development of hydrogel dressings with satisfactory functionality remains an ongoing concern. In this study, a multifunctional hydrogel wound dressing (PAN/Ag-PLG) with adhesion, antibacterial, hemostatic, and other properties, which can effectively repair diabetic wounds infected with methicillin-resistant Staphylococcus aureus (MRSA), is presented. The hydrogel dressing is composed of gallic acid (GA)-functionalized polylysine (PL)-reduced silver nanoparticles (Ag-PLG), oxidized hyaluronic acid (OHA), and cross-linked polyacrylic acid grafted with N -hydrosuccinimide ester. Notably, compared to most conventional wound dressing that lack adhesion or are difficult to remove, the prepared hydrogels exhibit excellent adhesion and mild stimulation-triggered detachment. In vitro and in vivo experiments reveal that the PAN/Ag-PLG hydrogel exhibits outstanding biocompatibility and antibacterial properties and promotes diabetic wound repair by reducing oxidative damage and promoting cell migration and angiogenesis. The smart PAN/Ag-PLG hydrogel reported in this study provides an approach for the potential clinical development of painless antibacterial dressings.

Stability and antioxidant activity of 10 isoflavones and anthocyanidins during in vitro digestion

Yaoyao Qi,Leyi Xie,Zeyuan Deng,Bing Zhang,Hongyan Li

The stability and antioxidant activity changes of 10 phytochemicals (isoflavones: genistein, genistin, daidzein, daidzin, formononetin, and anthocyanidins: delphinidin, cyanidin, pelargonidin, malvidin, petunidin) and food representatives (soybeans and blueberries) during in vitro digestion were investigated. Results indicated that the stability of isoflavones and anthocyanidins decreased with digestion. Among the isoflavones, the loss rate of formononetin was the largest, reaching 98.24%, and that of daidzein was the smallest (22.78%). And after intestinal digestion, anthocyanidins degraded into phenolic acids and other compounds, and the loss rate was above 95%. The loss of phenolic compounds in food extract was smaller than the standards, and the loss rate of phenolic compounds in hydrolyzed blueberry extracts (90.66%–96.04%) was higher than that of soybean extract (31.22%–59.74%). Moreover, the antioxidant activity of isoflavones and anthocyanidins decreased after the oral phase. During the gastrointestinal phase, the ABTS, DPPH and FRAP of isoflavones increased, while the ORAC and CAA decreased. The antioxidant activity of anthocyanidins increased in the gastric digestion and decreased in the intestinal phase. The glucoside linkage in the A ring and the hydroxyl methoxylation in the B ring weakened the antioxidant activity of isoflavones, while the hydroxylation and methoxylation of B rings of anthocyanidins could enhance thier antioxidant activity. The changes in antioxidant activity of the two food extracts were consistent with their contents.

Preparation and Characterization of a Zeolite/Chitosan-Based Magnetic Microparticle with Incorporated Ligustrum robustum (Rxob.) Blume Polyphenols for Oil Quality Control

Hao-Xiang Gao,Nan Chen,Qiang He,Bi Shi,Zhi-Long Yu,Wei-Cai Zeng

This study aimed to construct zeolite/chitosan-based magnetic microparticles (MMPs) using ZSM-5 zeolite, Fe3O4, and chitosan with incorporated Ligustrum robustum (Rxob.) Blume polyphenol extract (LRE) for oil quality control. Magnetic characterization and X-ray diffraction analysis demonstrated that the obtained MMPs had a saturation magnetization of 12.07 emu/g and showed magnetic separation ability due to the presence of ferric oxide crystals. Meanwhile, MMPs exhibited good resistance against heating and had a uniform structure with an intermediate diameter of 125.79 μm. Furthermore, scanning electron microscopy and sustained-release analysis indicated that MMPs could well embed LRE and slowly release LRE in weakly polar solvents. Finally, the oil protection experiments demonstrated that MMPs could inhibit the quality deterioration of peanut oils during the frying process and show good recyclability. The findings provide promise for using magnetic microparticles in oil quality control during high-temperature processing in agriculture and food industries.

Arthrospira cell residue valorization: A study on protein hydrolysate production by limited enzymatic hydrolysis

Kefan Ouyang,Qian Chen,Hexiang Xie,Qin Zhang,Liling Tao,Hua Xiong,Yulan Du,Jiangang Yan,Xinliang Mao,Qiang Zhao

Arthrospira cell residue (ACR) is a byproduct of the spirulina industry that can be valorized into protein hydrolysates. In this study, we used alkaline protease to hydrolyze ACR and evaluated the effects of hydrolysis time on the properties and functionalities of the hydrolysates. We measured the degree of hydrolysis (DH), particle size, fluorescence, UV‒visible absorption, surface hydrophobicity , emulsification , solubility, and antioxidant activity of the hydrolysates. The DH reached a maximum of 10.2% ± 0.6% after 240 min. Hydrolysis also altered the structure and conformation of the ACR protein, as shown by changes in fluorescence, UV-visible absorption, and FTIR spectra. Hydrolysis enhanced the solubility, surface hydrophobicity , and emulsifying activity of the hydrolysates but not the emulsion stability. The hydrolysates also exhibited higher antioxidant activity than ACR. These results indicate that enzymatic hydrolysis can convert ACR into valuable protein hydrolysates.

Response surface methodology-optimized extraction of flavonoids from pomelo peels and isolation of naringin with antioxidant activities by Sephadex LH20 gel chromatography

Yuchen Shangguan,Jing Ni,Lili Jiang,Yang Hu,Chuanbo He,Ying Ma,Guohong Wu,Hejian Xiong

In this study, flavonoids were extracted from pomelo peels and naringin was isolated from the flavonoid extract. The effects of extraction parameters, namely, ethanol concentration, solid-to-liquid ratio, and extraction time, on the yield of flavonoids extracted from pomelo peels were analyzed according to the Box-Behnken design of response surface methodology. The experimental conditions for flavonoid extraction were optimized, and naringin was separated from the extracted flavonoids using Sephadex LH-20 column chromatography. Experimental results showed that the influence of factors on the extraction rate of flavonoids from pomelo peels was in the order of ethanol concentration > solid-to-liquid ratio > extraction time, and the optimal extraction parameters were 85% ethanol concentration, 1:20 solid-to-liquid ratio, and 4-h extraction time for extracting flavonoids from pomelo peels. Under these conditions, the yield of flavonoids was 6.07 ± 0.06 mg/g. After three times of extraction, the flavonoid extraction rate reached 96.55%, and the residual naringin in the pomelo peels was 0.017 mg/g, at which point the bitterness in the pomelo peels disappeared. Two components, namely, PF1 and PF2, were separated from the crude flavonoid of pomelo peels through Sephadex LH20 column chromatography. PF2 was identified as naringin by high-performance liquid chromatography tandem mass spectrometry, with a purity of 95.7 ± 0.23%. Both flavonoids and PF2 exhibited good in vitro radicals scavenging activities on DPPH, ABTS, superoxide anion and hydroxyl.

Effects of κ-carrageenan on the emulsifying ability and encapsulation properties of pea protein isolate-grape seed oil emulsions

Wenjuan Wang,Rui Sun,Suping Ji,Qiang Xia

This work investigated the characteristics of pea protein isolate and κ-carrageenan complexes in stabilizing curcumin-loaded emulsions. The complexes structured by electrostatic attraction exhibited biphasic wettability with increased three-phase contact angles close to 90°. Morphological differences in the complexes were the critical factor influencing their emulsifying ability at various pH. As a steric barrier via increasing net negative charge (up to −54.7 ± 2.4 mV) and adsorbed protein content (92.57 %–97.61 %), the interfacial layer could retard droplet coalescence and improve emulsions stability. Rheological tests verified the higher viscoelasticity of emulsions by raising the oil fraction. After 4 weeks of heating treatment, the chemical stability of curcumin was prominently enhanced from 18.6 ± 0.2 % to 64.3 ± 5.7 %. The confirmed synergistic antioxidant activity between grape seed oil and curcumin in emulsions might facilitate the development of specific functional delivery systems in foods.

Effect of hydrochloric acid and citric acid with ultrasound processing on characteristics of superfine-ground pectic polysaccharides from okra (Abelmoschus esculentus L.) peel

Xiao-Jie Yue,Peng-Wei Xu,Yuan Zhu,Shou-Bu Hou,Xiao-Chuan Luo,Bing Zhao

The structural properties and biological activities of okra pectic polysaccharides (OPs) were impacted by various extraction methods. Based on commonly grinding (40, 100 meshes) and superfine grinding okra powders, two extraction solvents (hydrochloric acid, HA; citric acid , CA) were used firstly. Next, the extraction yield, physical and chemical properties, molecular structure and functional properties of OPs were analyzed by non-ultrasonic treatment and ultrasound-assisted superfine grinding method. The outcomes demonstrated that the extraction yield of OPs rose as the particle size of the powder decreased. HA-OPs had higher molecular weight (Mw), apparent viscosity and emulsification ability than CA-OPs. CA-OPs had higher esterification degree (DE), solubility and total sugar content, and higher amounts of rhamnogalacturonan-I (RG-I) segments. Compared with OPs without ultrasound-assisted extraction, ultrasound-assisted superfine grinding extraction exhibited higher sugar content, antioxidant capacity, emulsification ability, lower Mw, DE and apparent viscosity. Finally, the correlation between structure and function of OPs was further quantified. The antioxidant capacity was positively correlated with RG-I content, and negatively correlated with DE and Mw. The emulsification ability was mainly positively correlated with the GlcA of OPs. This study provides a theoretical basis for the development of OPs foods with clear structure-function relationship, which would be instructive for the application of OPs in food and cosmetics.

An antibacterial and anti-oxidant hydrogel containing hyperbranched poly-l-lysine and tea polyphenols accelerates healing of infected wound

Jie Wang,Yiming Sun,Xiaoqing Liu,Yongyuan Kang,Wangbei Cao,Juan Ye,Changyou Gao

Both bacteria-infection and excessive inflammation delay the wound healing process and even create non-healing wound, thus it is highly desirable to endow the wound dressing with bactericidal and anti-oxidation properties. Herein an antibacterial and antioxidation hydrogel based on Carbomer 940 (CBM) and hydroxypropyl methyl cellulose (HPMC) loaded with tea polyphenols (TP) and hyperbranched poly- l -lysine (HBPL) was designed and fabricated. The hydrogel killed 99.9 % of methicillin-resistant Staphylococcus aureus ( MRSA ) and Escherichia coli ( E. coli ) at 10 7  CFU mL −1 , and showed strong antioxidation against H 2 O 2 and 2,2-di(4- tert -octylphenyl)-1-picryl-hydrazyl (DPPH) radicals without noticeable cytotoxicity in vitro . The CBM/HPMC/HBPL/TP hydrogel significantly shortened the inflammatory period of the MRSA-infected full-thickness skin wound of rats in vivo , with 2 orders of lower MRSA colonies compared with the blank control, and promoted the wound closure especially at the earlier stage. The inflammation was suppressed and the vascularization was promoted significantly as well, resulting in reduced pro-inflammatory factors including interleukin-6 (IL-6), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α), and increased anti-inflammatory factors such as interleukin-4 (IL-4) and interleukin-10 (IL-10).

Preparation, stability and digestive properties of tea polysaccharide nanoparticles

Shuyao Li,Na Zhang,Shuyi Li,Xuehua Wang,Xin Cong,Shuiyuan Cheng,Zhenzhou Zhu

This study aimed to investigate the storage stability and functional properties of different sizes of tea polysaccharides, and explore the relationship between sizes and digestive properties, so as to provide reference for the subsequent utilization of tea polysaccharides. In this study, TPS-NPs of about 200 nm and 400 nm were prepared by electrostatic interaction with TPS and EDTA ratios of 6:1 and 100:1, respectively. The structural properties of TPS-NPs were investigated by a series of characterization analyses. The results showed that the major functional groups of TPS-NPs did not change significantly with particle size, but the C O stretching vibrations in the methylated carboxyl group were more pronounced. However, their DPPH free radical scavenging ability and antioxidant activity of TPS were decreased in vitro because of the destruction of the apparent structure of tea polysaccharides nanoparticles during preparation. Stability measurements revealed that TPS-NPs have high storage, temperature, pH and ionic concentration stability, especially for TPS-NPs with a particle size of 200 nm. The digestibility of 400 nm TPS-NPs was improved to 3.89% compared to TPS. The results showed that the TPS-NPs has excellent stability and digestibility, which contributed to the development of TPS applications in the food industry and related products.

Exploring Antioxidant and α-Glucosidase Inhibitory Activities in Mulberry Leaves (Morus alba L.) across Growth Stages: A Comprehensive Metabolomic Analysis with Chemometrics

Wenjie Li,Shenghui Hao,Hengyang Li,Qi An,Lina Yang,Bing Guo,Zijing Xue,Yongli Liu,Long Guo,Yuguang Zheng,Dan Zhang

Metabolic product accumulation exhibited variations among mulberry (Morus albaL.) leaves (MLs) at distinct growth stages, and this assessment was conducted using a combination of analytical techniques including high-performance liquid chromatography (HPLC), gas chromatography–mass spectrometry (GC-MS), and liquid chromatography–mass spectrometry (LC-MS). Multivariate analysis was applied to the data, and the findings were correlated with antioxidant activity and α-glucosidase inhibitory effects in vitro. Statistical analyses divided the 27 batches of MLs at different growth stages into three distinct groups. In vitro assays for antioxidant activity and α-glucosidase inhibition revealed that IC50values were highest at the Y23 stage, which corresponds to the ‘Frost Descends’ solar term. In summary, the results of this study indicate that MLs at different growth stages throughout the year can be categorized into three primary growth stages using traditional Chinese solar terms as reference points, based on the observed variations in metabolite content. Keywords:Morus albaL.;primary metabolites;secondary metabolites;different growth stages;antioxidant activity

Secondary metabolite profiles and bioactivities of red raspberry juice during fermentation with Wickerhamomyces anomalus

Yin Qin,You Luo,Shuyi Qiu,Qunying Zhang,Le Yang

Raspberry contains a broad array of valuable phytochemicals and therefore has positive effects on health. Fermentation can transform these components. The variations in the secondary metabolite profiles and bioactivities of red raspberry juice during Wickerhamomyces anomalus fermentation remain unknown. Herein, the effects of W. anomalus on the secondary metabolites, other constituents (total sugars, reducing sugars, and total acids) and bioactivities of raspberry juice were investigated. The fermentation significantly decreased the level of total sugars, reducing sugars, total acids and anthocyanins, but greatly increased the content of polyphenols and flavonoids. The antioxidant capacity and α-glucosidase inhibitory activity of raspberry juice were markedly enhanced after fermentation, whereas the cholesterol esterase inhibitory activity was reduced significantly. Moreover, 721 secondary metabolites were identified, wherein 437 secondary metabolites (240 upregulated, 197 downregulated) were selected as differential metabolites via targeted metabolomics. A total of 89 secondary metabolites were annotated in the Kyoto Encyclopedia of Genes and Genomes pathways, most of which were matched to biosynthesis of secondary metabolites and metabolic pathways. Phenolic acids were mainly transformed via phenylalanine metabolism and tyrosine metabolism. These results can provide reference for the development of functional raspberry beverages.

Evaluation of biosynthesis parameters, stability and biological activities of silver nanoparticles synthesized by Cornus Officinalis extract under 365 nm UV radiation

Yinghui Wang,Simin Wei,Kang Wang,Zhe Wang,Jinwei Duan,Lin Cui,Huayu Zheng,Ying Wang,Shanshan Wang

Since silver nanoparticles (AgNPs) synthesized by using plant extracts revealed varied biological activities, the green synthesis of AgNPs has attracted considerable attention. Although the green synthesis of AgNPs have been accomplished by using the extracts of Cornus Officinalis, which is a traditional Chinese medicine and exhibits a wide spectrum of phytochemicals. The effects of biosynthesis parameters on reducing reaction, stability and more broad biological activities of so-prepared AgNPs did not been evaluated. In this paper, we firstly assessed the effects of UV radiation, pH, material proportion and radiation times on the green synthesis of AgNPs under 365 nm UV radiation by UV-visible spectrum and dynamic light scattering (DLS) analysis. The results showed that UV radiation could accelerate the formation of AgNPs and influence the average size below pH 7.0, and the size of so-prepared AgNPs were sensitive to the pH and material proportion, but no obvious changes to UV radiation times, offering a size-controlled synthetic method for AgNPs. The further X-ray diffraction (XRD), transmission electron microscopy (TEM) and DLS studies showed AgNPs synthesized at pH 7.0, extract: AgNO3 = 1 : 1 and after 4 h UV radiation were a face-centered cubic (fcc) structure and both spherical and polygonal in shape with average particle size of 64.5 ± 0.3 nm existed in a monodispersed form. Subsquently, the stability of AgNPs was analyzed by zeta potential (−24.8 mV) and the average size measurement after 30 days storage (63.3 ± 0.4 nm), revealing a high degree of stability. Lastly, the investigation of biological activities showed that the biosynthesized AgNPs had potent antioxidant activity, antimicrobial activity against both S. aureus and E. coli as well as anticancer activity against HCT116 and HepG2 cell lines but negligible cytotoxicity against SW620. And the internalization of biosynthesized AgNPs inside the bacterial cell was evaluated by flow cytometric analysis, where the SSC values have significant increase after treating with nanoparticles. These results confirmed that the biosynthesis parameters on the green synthesis of AgNPs by using Cornus Officinalis extract also played pivotal roles and so-prepared AgNPs would be useful for the development of new alternative antioxidant, antimicrobial and anticancer agents in biomedicine.
