Assessment of the ‘taro-like’ aroma of pumpkin fruit (Cucurbita moschata D.) via E-nose, GC–MS and GC-O analysis

LI Junxing,Miao Aiqing,ZHAO Gangjun,Liu Xiaoxi,Wu Haibin,Luo Jianning,Gong Hao,Zheng Xiaoming,Deng Liting,Ma Chengying

‘Taro-like’ aroma is a pleasant flavor and value-added trait in pumpkin species imparted by unknown key volatile compounds. In this study, we used the electronic nose (E-nose), gas chromatography-mass spectrometry (GC–MS), and GC-Olfactometry (GC-O) to study the aroma profile, volatile compounds, and key   contributors, respectively. By E-nose and GC–MS, we found significant differences in the aroma profiles and volatile compounds between fruits from five samples with/without ‘taro-like’ aroma. According to the analysis of differential volatile compounds obtained from GC–MS and the GC-O analysis of the sample with ‘taro-like’ aroma, we found that 2-acetyl-1-pyrroline representing the ‘taro’ odor was only identified in the sample with ‘taro-like’ aroma. Therefore, we conclude that 2-acetyl-1-pyrroline is the key contributor to the 'taro-like' aroma. Moreover, the relationship between 2-acetyl-1-pyrroline and ‘taro-like’ aroma was further verified via other pumpkin samples. Our results provide a theoretical basis for understanding the aroma characteristics of pumpkin fruit.

Efficient and sustainable photocatalytic inactivation of E. coli by an innovative immobilized Ag/TiO2 photocatalyst with peroxymonosulfate (PMS) under visible light

Jialin Jia,Stefanos Giannakis,Dong Li,Boyin Yan,Tao Lin

A novel catalytic system for effective photocatalytic inactivation of Escherichia coli ( E. coli ) was constructed by anchoring Ag nanoparticles (AgNPs) on silane coupling agent (SCA) pretreated TiO 2 nano-tube arrays (Ag/SCA/TiO 2 NTAs). Morphology and structural analyses revealed that SCA could disperse AgNPs evenly on TiO 2 NTAs, thus inducing a superior surface plasmon resonance (SPR) effect. Ag/SCA/TiO 2 NTAs catalyst exhibited excellent inactivation performance when in the presence of peroxymonosulfate (PMS) and visible light (VL), with 6-log E. coli was completely inactivated within 60 min, which was 5.3, 12.5 and 13.2 times higher than that of Ag/SCA/TiO 2 NTAs/VL, PMS/VL and Ag/SCA/TiO 2 NTAs/PMS/dark systems, respectively. Additionally, the photocatalyst exhibited a highly reusable property, with the inactivation performance almost unchanged after ten cycles of uses with minimal Ag leaching. The inactivation mechanism analysis demonstrated that both radical (SO 4 •- , OH) and non-radical (h + , 1 O 2 ) pathways involved in E. coli inactivation, and SCA played a pivotal role in the production of reactive species. Chloride ions (Cl − ) greatly enhanced the inactivation efficiency, while bicarbonate (HCO 3 − ) and phosphate (H 2 PO 4 − ) showed an inhibitory effect. Humic acid (HA) displayed a dual effect on inactivation performance, where the low concentration of HA facilitated the bacteria inactivation, while the higher dose suppressed bacteria inactivation. Moreover, the system exhibited excellent inactivation performance in tap water . This work first used SCA as the binder to fix AgNPs on TiO 2 NTAs for VL photocatalytic inactivation of bacteria with the assistance of PMS, which was expected to provide some insights into the practical treatment of drinking water .

Dual-mode immunochromatographic assay based on dendritic gold nanoparticles with superior fluorescence quenching for ultrasensitive detection of E. coli O157:H7

Yanna Shao,Zhengzheng Wang,Jihang Xie,Zhenjun Zhu,Ying Feng,Shubo Yu,Liang Xue,Shi Wu,Qihui Gu,Jumei Zhang,Qingping Wu,Juan Wang,Yu Ding

We presented a colorimetric/fluorescent dual-mode immunochromatographic assay (ICA) for the sensitive detection of Escherichia coli O157:H7. The use of polydopamine (PDA)-modified gold nanoparticles (AuNPs) with broadband absorption allowed for excellent colorimetry signals for the ICA detection. Moreover, the absorption spectrum of PDA-AuNPs significantly overlaps with the excitation and emission spectra of ZnCdSe/ZnS quantum dots (QDs), resulting in effective quenching of the QDs fluorescence due to the inner filter effect. The fluorescence intensity changes induced by PDA-AuNPs were utilized for the sensitive detection of E. coli O157:H7, achieving a detection limit of 9.06 × 10 1 CFU/mL, which was 46-fold lower than that of traditional AuNPs-based immunoassay. The proposed immunosensor exhibited the recovery rate between 80.12% and 114.69% in detecting actual samples, indicating its reliability and satisfactory accuracy. This study provides insights into dual-mode signal outputs and the ICA development for food safety applications.

Visible light-activated 1-D core-shell paramagnetic Fe-Ag@AgCl as an innovative method for photocatalytic inactivation of E. coli

Cui Yanping,Thathsarani Nimesha,Peng Liang,Gao Ya,Lei Lidan,Zhou Zhengwei,Liang Lili,Shi Xiaoyan

Innovative paramagnetic one-dimensional (1-D) core-shell Fe-Ag@AgCl visible light-driven photocatalysts are synthesized through a template-assisted electrodeposition method trailed by FeCl 3 in-situ oxidation. The metallic nature of Fe-Ag@AgCl is confirmed through scanning electron microscopy (SEM) and crystal nature through X-ray diffraction (XRD). The controllable diameter of Fe-Ag is obtained through the selection of hollow size of the polycarbonate (PC) template. Electron impedance spectroscopy (EIS) confirms through the introduction of Fe to the Ag core that has prolonged the recombination of electron and hole. Escherichia coli ( E. coli ) are employed as the target bacteria to evaluate the photocatalytic disinfection performances. A total of 1.30 mg of Fe-Ag@AgCl is proved to be able to completely inactivate 10 7  CFU (colony forming units)/mL after 120 min of visible light irradiation. The transition electron microscopy (TEM) confirms the stability of the material after the photo reaction. As Fe-Ag@AgCl possesses magnetic properties, the material is recovered through the application of an external magnetic field. SEM images and results of 3D emission extraction matrix (EEM) depict that the bacteria cell death is caused by membrane permeability changes caused by the reduction of membrane associated proteins.

A convenient chemiluminescence detection for bisphenol A in E-waste dismantling site based on surface charge change of cationic gold nanoparticles

Yingying Qi,Jiahuan He,Fu-Rong Xiu,Xuan Yu,Xiang Gao,Yifan Li,Yongwei Lu,Zhiqi Song

The crude disassembling for electronic waste (E-waste) has resulted in serious pollution of the environment in dismantling site. Convenient detection of single contaminant in dismantling site is very important to evaluating environmental pollution caused by single component during dismantling process and environment supervision. Herein, a novel chemiluminescence (CL) assay was constructed to detect ubiquitous pollutant bisphenol A (BPA) in dismantling site of E-waste based on surface charge change of cationic gold nanoparticles arising from the interaction of BPA and aptamer. The detection limit for BPA is 6.2 × 10 −2  ng/mL, which is far lower than the requirements of Chinese Health Standards (0.05 mg/kg). The sensitivity can be attributed to the strong affinity between anti-BPA aptamer and BPA, and the direct and sensitive signal response of nanoparticles' microstructure for CL reaction. The high specificity of affinity between anti-BPA aptamer and BPA led to high selectivity of BPA detection, thus resisting the complexity of real environment. So, this analysis was successfully applied for the detection of BPA in the surface soil of E-waste dismantling site.

Small molecule nAS-E targeting cAMP response element binding protein (CREB) and CREB-binding protein interaction inhibits breast cancer bone metastasis

Min Jiang,Yufei Yan,Kai Yang,Zhuochao Liu,Jin Qi,Hanbing Zhou,Niandong Qian,Qi Zhou,Tianqi Wang,Xing Xu,Xiangshu Xiao,Lianfu Deng

Bone is the most common metastatic site for breast cancer. The excessive osteoclast activity in the metastatic bone lesions often produces osteolysis. The cyclic-AMP (cAMP)-response element binding protein (CREB) serves a variety of biological functions including the transformation and immortalization of breast cancer cells. In addition, evidence has shown that CREB plays a key role in osteoclastgenesis and bone resorption. Small organic molecules with good pharmacokinetic properties and specificity, targeting CREB-CBP (CREB-binding protein) interaction to inhibit CREB-mediated gene transcription have attracted more considerations as cancer therapeutics. We recently identified naphthol AS-E (nAS-E) as a cell-permeable inhibitor of CREB-mediated gene transcription through inhibiting CREB-CBP interaction. In this study, we tested the effect of nAS-E on breast cancer cell proliferation, survival, migration as well as osteoclast formation and bone resorption in vitro for the first time. Our results demonstrated that nAS-E inhibited breast cancer cell proliferation, migration, survival and suppressed osteoclast differentiation as well as bone resorption through inhibiting CREB-CBP interaction. In addition, the in vivo effect of nAS-E in protecting against breast cancer-induced osteolysis was evaluated. Our results indicated that nAS-E could reverse bone loss induced by MDA-MB-231 tumour. These results suggest that small molecules targeting CREB-CBP interaction to inhibit CREB-mediated gene transcription might be a potential approach for the treatment of breast cancer bone metastasis.

Enhanced paracellular and transcellular paclitaxel permeation by chitosan–vitamin E succinate–N-acetyl-l-cysteine copolymer on Caco-2 cell monolayer

Lian He,Zhang Tianhong,Sun Jin,Pu Xiaohui,Tang Yilin,Zhang Youxi,He Zhonggui

The aim of this study was to evaluate the underlying mechanism of enhanced oral absorption of paclitaxel (PTX)-loaded chitosan–vitamin E succinate– N -acetyl- l -cysteine (CS–VES–NAC) nanomicelles from the cellular level. In aqueous solution, CS–VES–NAC copolymer self-assembled into the polymeric nanomicelles, with the size ranging from 190 to 240 nm and the drug loading content as high as 20.5 %. Cytotoxicity results showed that the PTX-loaded nanomicelles exhibited the similar effect to PTX solution (PTX-Sol) on Caco-2 cells, but no toxicity observed for blank CS–VES–NAC nanomicelles. The cellular uptake of PTX was significantly increased by CS–VES–NAC nanomicelles, compared with that of PTX-Sol, due to the possible escapement of P-glycoprotein (P-gp) efflux pumps by endocytosis pathway. Confocal laser scanning microscope (CLSM) images also confirmed CS–VES–NAC nanomicelles could be effectively internalized by Caco-2 cells. More importantly, P app value of PTX-loaded CS–VES–NAC nanomicelles was 2.3-fold higher than that of PTX-Sol, and the efflux ratio decreased by more than 10.8-fold for the nanomicelles. As a consequence of opening of tight junctions and P-gp inhibition induced by free CS–VES–NAC copolymer, the P app value of PTX was almost increased up to 19.5-fold. All the results indicate that CS–VES–NAC copolymer hold great promises as nanocarrier for antitumor drug oral delivery by improving paracellular and transcellular permeation.

Enhanced Oral Delivery of Paclitaxel Using Acetylcysteine Functionalized Chitosan-Vitamin E Succinate Nanomicelles Based on a Mucus Bioadhesion and Penetration Mechanism

He Lian,Tianhong Zhang,Jin Sun,Xiaohong Liu,Guolian Ren,Longfa Kou,Youxi Zhang,Xiaopeng Han,Wenya Ding,Xiaoyu Ai,Chunnuan Wu,Lin Li,Yongjun Wang,Yinghua Sun,Siling Wang,Zhonggui He

In addition to being a physiological protective barrier, the gastrointestinal mucosal membrane is also a primary obstacle that hinders the oral absorption of many therapeutic compounds, especially drugs with a poor permeability. In order to resolve this impasse, we have designed multifunctional nanomicelles based on the acetylcysteine functionalized chitosan-vitamin E succinate copolymer (CS-VES-NAC, CVN), which exhibit marked bioadhesion, possess the ability to penetrate mucus, and enhance the oral absorption of a hydrophobic drug with a poor penetrative profile, paclitaxel. The intestinal absorption (Ka = 0.38 ± 0.04 min–1, Papp = 0.059 cm·min–1) of CVN nanomicelles was greatly improved (4.5-fold) in comparison with paclitaxel solution, and CLSM (confocal laser scanning microscope) pictures also showed not only enhanced adhesion to the intestinal surface but improved accumulation within intestinal villi. The in vivo pharmacokinetics indicated that the AUC0–t (586.37 ng/mL·h) of CVN nanomicelles was markedly enhanced compared with PTX solution. In summary, the novel multifunctional CVN nanomicelles appear to be a promising nanocarrier for insoluble and poorly permeable drugs due to their high bioadhesion and permeation-enhancing capability.

Efficient biosynthesis of rare natural product scopolamine using E. coli cells expressing a S14P/K97A mutant of hyoscyamine 6β-hydroxylase AaH6H

Yue-De Cao,Yu-Cai He,Hao Li,Guo-Yin Kai,Jian-He Xu,Hui-Lei Yu

Hyoscyamine 6 β -hydroxylase (H6H, EC 1.14.11.11), an α-ketoglutarate dependent dioxygenase catalyzes the hydroxylation of (−)-hyoscyamine and the subsequent epoxidation of 6 β -hydroxyhyoscyamine to form scopolamine, a valuable natural alkaloid. In this study, random mutagenesis and site-directed saturation mutagenesis were used to enhance the hydroxylation activity of H6H from Anisodus acutangulus ( Aa H6H). A double mutant, Aa H6H M1 (S14P/K97A), showed a 3.4-fold improved hydroxylation activity compared with the wild-type enzyme, and the in vivo epoxidation activity was also improved by 2.3 times. After 34   h cultivation of Escherichia coli cells harboring Aa h6h m1 in a 5-L bioreactor with a working volume of 3   L, scopolamine was produced via a single-enzyme-mediated two-step transformation from 500   mg   L −1 (−)-hyoscyamine in 97% conversion, and 1.068   g of the product were isolated, corresponding to a space-time yield of 251   mg   L −1   d −1 . This study shows that the protein engineering of some key enzymes is a promising and effective way for improving the production of rare natural products such as scopolamine.

Control of cell proliferation in E-jet 3D-printed scaffolds for tissue engineering applications: the influence of the cell alignment angle

Tong Liu,Ruiying Huang,Juchang Zhong,Yikun Yang,Zhikai Tan,Weihong Tan

The ideal 3D scaffold for biological applications has not yet been designed. Our aim is to better match the scaffold performance through fine control of the fabrication process. Here, we applied electro-hydrodynamic jet (E-jet) 3D printing technology using poly-(lactic-co-glycolic acid) (PLGA) solution to construct scaffolds for tissue engineering applications. We fabricated different scaffolds of 1 : 1, 1 : 2 and 1 : 3 aspect ratios and tested the biological compatibility of the 2D and 3D printed scaffolds with fibroblasts. The scaffolds were used to culture fibroblasts, the main cellular components of loose connective tissue. The results show that the E-jet printed scaffolds could guide and improve cell growth. These scaffolds were able to support cellular alignment and proliferation. The cell angle was consistent with the longitudinal direction of the scaffolds, potentially enhancing the wound healing performance. Thus, the potential of this flexible technology for the 3D printing of scaffolds for the engineering of tissues is extensive.

Deficiency of Calpain-1 attenuates atherosclerotic plaque and calcification and improves vasomotor dysfunction in Apolipoprotein E knockout mice through inhibiting inflammation

Youming Shi,Wenjiao Gu,Yujie Wei,Shuangyi Li,Shining Zhang,Ying jiang,Chuanjian Chen,Ting Liu,Limei Shuai,Xiaochun Zhou,Futian Tang

Purpose Atherosclerosis (AS) and atherosclerotic calcification (AC) are closely related to the cardiovascular diseases, largely due to its induction of vasomotor dysfunction. We previously reported that Calpain-1 inhibitor attenuated AS and AC. The present study was designed to investigate the effects and potential mechanisms of Calpain-1 knockout (Calpain-1 KO) in Apolipoprotein E KO (ApoE KO) mice on AS, AC, aortic vasomotor function as well as the liver dysfunction. Methods We hybridized ApoE KO mice with Calpain-1 KO mice to obtain ApoE /Calpain-1 double KO (A×C DKO) mice. The formation of AS and AC was evaluated and liver function was determined. Aortic vasomotor function was assessed. Contents of TNF-α, IL-6, IL-18, IL-1β and NO and the activity of AST, ALT, ALP and eNOS in serum were quantified. The mRNA expression of CD68, SR-A, CD36, PPAR-γ, LXR-α, ABCA1, BMP-2, OPN, ALP and Runx2 in aorta and/or liver were measured. Results The results showed that in comparison to C57 mice, ApoE KO mice demonstrated the significant increases in the areas of AS and AC, the increases in the mRNA expression of CD68 in the aorta, the increases in the AST, ALT and ALP activity in serum. ApoE KO mice also showed the dysfunction of the aortic contraction induced by phenylephrine and of the relaxation induced by acetylcholine. However, compared with ApoE KO mice, A×C DKO mice exhibited the significant attenuation of AS and AC and the downregulation of mRNA expression of CD68 in aorta. A×C DKO mice revealed the reduction of AST, ALT and ALP activity in serum, the improvements in aortic contraction and relaxation as well as the increases in eNOS activity and NO content in serum. A×C DKO mice also showed the decreases in the contents of TNF-α, IL-6, IL-18 and IL-1β in serum. The mRNA expression of CD68 in aorta, SR-A and CD36 in both aorta and liver of A×C DKO mice was downregulated, while that of PPAR-γ, LXR-α, and ABCA1 was upregulated in comparison of ApoE KO mice. In addition, the mRNA expression of BMP-2, OPN, ALP and Runx2 in aorta of A×C DKO mice was downregulated in comparison of ApoE KO mice. Conclusion The results suggested that deficiency of Calpain-1 attenuated the formation of AS and AC and improved vasomotor and liver dysfunction in ApoE KO mice through anti-inflammation, and modulation of the mRNA expression of genes related to AS and AC.

Characterization of volatile compound differences of Shaoxing Huangjiu aged for different years using GC-E-Nose, GC–MS, and GC-IMS

Chen Ziqiang,Ma Mingtao,Chen Xingguang,Peng Zhengcong,Liu Hua,Lu Jian,Wu Dianhui

The process of aging was crucial in enhancing the aroma quality of Huangjiu. Gas chromatography electronic nose (GC-E-Nose), gas chromatography-mass spectrometry (GC–MS), and gas chromatography-ion mobility spectrometry (GC-IMS) were employed to detect the variations in aroma among five samples aged for varying years (5–11). A comprehensive analysis revealed the presence of 85 compounds in the Shaoxing Huangjiu, twenty of which were identified as crucial contributors to its flavor profile through odor activity values analysis. The volatile aroma components of Huangjiu exhibited notable variations among different aging years. Additionally, the concentration and diversity of key aroma compounds increased with prolonged aging. At the same time, quantitative descriptive sensory analysis showed that aging could enhance the ester and acid aroma of Huangjiu. Moreover, based on variable importance for the projection, seven potential aroma markers such as diethyl succinate, ethyl butyrate, and furfural were screened out, which were the major compounds that caused the flavor differences of aged Huangjiu. The results offer valuable theoretical support for illustrating and regulating the quality of Huangjiu during its aging process.
