TiO2@MOF-919(Fe–Cu) as a sorbent for the extraction of benzoylurea pesticides from irrigation water and fruit juices

Ling Mei,Tiantian Si,Shuai Wang,Jie Zhu,Hao Tang,Xiaojing Liang

The TiO2@MOF-919(Fe–Cu) solid-phase extraction material was prepared by growing MOF-919(Fe–Cu) in situ on three-dimensional radial TiO2 microspheres by a simple solvothermal method. This combination drew on both the resources of good single dispersion and extraction rate, which made it a better extraction material. It was accompanied with high-performance liquid chromatography (SPE–HPLC) for the separation and determination of four benzoylurea pesticides (triflumuron, chlorbenzuron, teflubenzuron and diflubenzuron) in afforestation irrigation water and juice samples (grape, peach and apple juices). Under the optimal conditions, the linearity of the method ranged from 1 to 400 μg L−1 with a correlation coefficient (R2) ≥ 0.9994, while the detection limit was in the range of 0.40–0.56 μg L−1 for the four pesticides. The adopted material showed good reusability and can be used no less than 10 times. The intra-day and inter-day precision were in the range of 1.78–3.24% and 4.06–5.08%, respectively. The proposed method was then successfully applied for the detection of benzoylurea pesticides in the spiked samples with good recoveries (72.3–108.4%) and good precision (5.15%) due to π–π and hydrophobic interactions between the analytes and adsorbent. The results show that the composite had the potential to be used as a SPE adsorbent for the enrichment and extraction of benzene ring structures containing imide groups in actual samples.

Endogenous melatonin generation plays a positive role in chilling tolerance in relation to redox homeostasis in litchi fruit during refrigeration

Jialiang Liu,Jinhua Sun,Yonggui Pan,Ze Yun,Zhengke Zhang,Guoxiang Jiang,Yueming Jiang

The potent role of endogenous melatonin (MEL) in affecting chilling injury (CI) and related chilling tolerance regulatory mechanisms in harvested litchi fruit during refrigeration at 4 °C were investigated in this study. In the first experiment, exogenous MEL exposure exerted an inhibitory impact on the development of CI severity in refrigerated litchi fruit, as indicated by the lower CI index and delayed red color loss of pericarp, whereas treatment with p -chlorophenylalanine ( p -CPA), an inhibitor of MEL biosynthesis, aggravated the degree of CI. The suppression of CI afforded by exogenous MEL was abated under the combined application of exogenous MEL and p -CPA. In the second experiment, the effects of p -CPA alone treatment on changes in biochemical parameters during refrigeration were analyzed. The results showed that p -CPA down-regulated the expressions of MEL biosynthesis genes, including LcTDC , LcT5H and LcSNAT , contributing to reduced generation of endogenous serotonin and MEL. p -CPA treatment accelerated oxidative stress-triggered membrane deterioration, as reflected by increases in relative leakage rate (RLR), reactive oxygen species (ROS) production, lipoxygenase (LOX) activity and malondialdehyde (MDA) accumulation. The membrane damage caused by p -CPA further resulted in loss of subcellular compartmentalization and enzymatic oxidation, as demonstrated by increased activities of polyphenol oxidase (PPO) and peroxidase (POD) in parallel with reduced contents of total phenolics and anthocyanin. Compared with control fruit, lower activities of antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR), were observed in fruit receiving p -CPA. Additionally, p -CPA suppressed the expression levels of genes encoding oxidized protein repair-related enzymes, including LcMsrA1 , LcMsrA2 , LcMsrB1 and LcMsB2 . These results suggest that endogenous MEL might confer a protective role against chilling stress by modulating cellular redox status in harvested litchi fruit during refrigeration.

Development of a chemiluminescence immunoassay for detection of tenuazonic acid mycotoxin in fruit juices with a specific camel polyclonal antibody

Feng Wang,De-Bin Wan,Yu-Dong Shen,Yuan-Xin Tian,Zhi-Li Xiao,Zhen-Lin Xu,Jin-Yi Yang,Yuan-Ming Sun,Bruce D. Hammock,Hong Wang

The natural mycotoxin tenuazonic acid (TeA) in foods is identified as the most toxic mycotoxin among the over 70 kinds of secondary toxic metabolites produced by Alternaria alternata. Some hapten–antibody-mediated immunoassays have been developed for TeA detection in food samples, but these methods show unsatisfactory sensitivity and specificity. In this study, a rationally designed hapten for TeA mycotoxin generated with computer-assisted modeling was prepared to produce a highly specific camel polyclonal antibody, and an indirect competitive chemiluminescence enzyme immunoassay (icCLEIA) was established with a limit of detection of 0.2 ng mL−1 under optimized conditions. The cross-reactivity results showed that several analogs and some common mycotoxins had negligible recognition by the anti-TeA polyclonal antibody. The average recoveries spiked in fruit juices were determined to be 92.7% with an acceptable coefficient of variation, and good correlations between icCLEIA and liquid chromatography tandem mass spectrometry (LC-MS/MS) results were obtained in spiked samples. This developed icCLEIA for TeA detection with significantly improved sensitivity and satisfactory specificity is a promising alternative for environmental monitoring and food safety.

Spiky yolk-shell AuAg bimetallic nanorods with uniform interior gap for the SERS detection of thiram residues in fruit juice

Jian Zhu,Shuang Zhang,Guo-jun Weng,Jian-jun Li,Jun-wu Zhao

By using gold nanorods with silver coating as the sacrificial templates, we prepared spiky yolk-shell AuAg bimetallic nanorods with uniform interior gap via galvanic replacement reaction. The length and number of Au tips of the spiky yolk-shell AuAg nanorods can be tuned simultaneously by altering HAuCl 4 volume. The influence of HAuCl 4 volume and the sliver layer thickness on the SERS activity of spiky yolk-shell AuAg nanorods are studied. When the sliver layer is thin, the interior gap has not been shielded completely and the outer shell has obvious tips, thus the surface-enhanced Raman scattering (SERS) activity has the strongest enhancement with an enhancement factor (EF) of 4.9 × 10 5 . The spiky yolk-shell AuAg nanorods with the strongest SERS activity are used as SERS substrates to detect thiram. The results demonstrate that the SERS intensity increases linearly with the logarithmic concentration of thiram in the range of 10 −3 M to 10 −7 M. The detection limit is as low as 97 nM, which is lower than the maximum pesticide residue limit (29 µM) in fruits stipulated by the US Environmental Protection Agency (EPA). Therefore, the spiky yolk-shell AuAg bimetallic nanorods have important practical application value in pesticide detection.

Hydrophobic deep eutectic solvent-based ultrasonic-assisted liquid-liquid micro-extraction combined with HPLC-FLD for diphenylamine determination in fruit

Shaomin Ma,Xiuyi Jin,Huan Wei,Yue Liu,Ming Guo

In this paper, novel high extraction efficiency hydrophobic eutectic solvents (DESs), n-octanoic acid as a hydrogen bond donor (HBD) and menthol as a hydrogen bond acceptor (HBA), were selected from five hydrophobic DESs to extract trace diphenylamine (DPA) in fruits apple, pear and orange under ultrasonic-assisted liquid-liquid micro-extraction (UA-LLME) technology before high-performance liquid chromatography (HPLC) analysis. Working parameters such as the DESs type, molar ratio, extractant volume, and ultrasonic time of the LLME hydrophobic DESs technology were optimised. Average recoveries between 96% and 108% were obtained on actual samples. This method gave lower detection limit (LOD) than other existing methods due to combining the high-efficiency extraction of hydrophobic DES and high sensitivity of fluorescence detector. This method was sensitive and eco-friendly, and can be used for the determination of trace components in fruits.

Comparative elucidation of bioactive and antioxidant properties of red dragon fruit peel as affected by electromagnetic and conventional drying approaches

Edidiong Joseph Bassey,Jun-Hu Cheng,Da-Wen Sun

The effects of near-infrared (NIRD), mid-infrared (MIRD), far-infrared (FIRD), microwave (MWD), and hot air drying (HAD) on drying kinetic, colour, phytochemical composition, and antioxidant activity of red dragon fruit peel (RDFP) was evaluated. Results indicated that drying methods induced varying microstructural and chemical changes on RDFP, significantly influencing moisture removal rates and phytochemical retention. The lowest drying time was observed for MWD, while MIRD presented the highest drying time. FIRD drying was more favourable for retaining TPC, TFC, betacyanin and betaxanthin, while the ascorbic acid content was better retained during MIRD and NIRD. Enhancements in ABTS, CUPRAC and reducing power were associated with FIRD, and NIRD and MIRD enhanced DPPH and HRSA. Overall, chemical modifications induced by drying improved the phytochemical and antioxidant properties but presented adversative effects on ascorbic acid and DPPH. The study presented an essential background for the optimal drying of RDFP.

Insight into the effects of different ripeness levels on the quality and flavor chemistry of Noni fruit (Morinda citrifolia L.)

Xiaoai Chen,Chunhe Gu,Kexue Zhu,Fei Xu,Zhen Feng,Yanjun Zhang

Different ripeness plays a key role in the final quality and flavor of Noni products, but the variations of quality and flavor during Noni ripening are still unclear. In this study, physicochemical analysis combined with electronic nose, electronic tongue and gas chromatography-mass spectrometry (GC–MS) were used to investigate the effect of different ripeness levels on Noni fruit quality and flavor chemistry. The results showed that the optimum stage of nutrient accumulation is 7–8 ripe (7M−8M), in which especially the higher contents were 144Mg/100 g vitamin C, 11.08 g/kg polyphenols, 15.32 g/kg flavonoids and 2420Mg/kg potassium. Octanoic acid and hexanoic acid with an unfriendly odor began to decrease significantly after 8 ripe (8M), the lowest contents were 256.76 and 231.26 ng/g at 9 ripe (9M). Sour, astringent and fresh tastes are the dominant tastes of Noni fruit. Therefore, the optimum harvesting time of Noni fruit is 8M.

Foodomics revealed the effects of ultrasonic extraction on the composition and nutrition of cactus fruit (Opuntia ficus-indica) seed oil

Xue Li,Baokun Qi,Shuang Zhang,Yang Li

Cactus is a tropical fruit with a high nutritional value; however, little information is available regarding the comprehensive utilization of its byproducts. This study aimed to explore the composition and nutritional value of cactus fruit seed oil (CFO) and reveal the effects of ultrasound-assisted extraction and traditional solvent extraction on oil quality. Foodomics analysis showed that CFO extracted using a traditional solvent is rich in linolenic acid (9c12cC18:2, 57.46 ± 0.84 %), α-tocopherol (20.01 ± 1.86 mg/100 g oil), and canolol (200.10 ± 1.21 μg/g). Compared to traditional solvent extraction, ultrasound-assisted extraction can significantly increase the content of lipid concomitants in CFO, whereas excessive ultrasound intensity may lead to the oxidation of oils and the formation of free radicals. Analysis of the thermal properties showed that ultrasound had no effect on the crystallization or melting behavior of CFO. To further demonstrate the nutritional value of CFO, a lipopolysaccharide (LPS)-induced lipid metabolism imbalance model was used. Lipidomics analysis showed that CFO significantly reduced the content of oxidized phospholipids stimulated by LPS and increased the content of highly bioactive metabolites such as ceramides, thus alleviating LPS-induced damage in C. elegans . Hence, CFO is a functional oil with high value, and ultrasound-assisted extraction is advocated. These findings provide new insights into the comprehensive utilization of cactus fruits.

Glycosylamines-based reactive matrix designed for imaging acidity in Ponkan fruit using matrix assisted laser desorption/ionization mass spectrometry imaging

Ge Ma,Xiaoyong Zhao,Cheng Guo,Shaojia Li,Yaqin Liu,Quan He,Kunsong Chen,Yuanjiang Pan

Current imaging techniques , such as position emission tomography , magnetic resonance imaging and optical imaging , have been developed and applied to imaging acidity in live tissues for years, but they still have some limitations. In the present study, a reliable and interference-free pH imaging method based on matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) was established for the first time. A novel reactive matrix, named glycosyl-3-aminoquinoline (Gly-3AQ), was designed and synthesized here not only to overcome the background interference in the low mass range of MALDI but also provide a new function of acid-responsiveness for the matrix. This matrix was responsive to pH change due to its acid-catalyzed hydrolysis and the hydrolysis product 3-aminoquinoline (3-AQ) has an increase of peak intensity with pH decrease. The Gly-3AQ-based MALDI pH detection method is demonstrated to have good acid-responsiveness in the range of pH 2.0–7.0, and when pH changes 0.5, relative abundance of 3-AQ changes about 1.5 folds. The method showed a very prominent advantage, that is, free-interference when analyzing real samples. Validation for the method revealed pH calibration curve had good linearity (R > 0.9953) with relative standard deviation less than 13.8%. This method was applied to analyze and visualize acidity variation of citrus during development and the changed acidity underwent relative quantitation and statistical analysis. The newly developed reactive matrix has potential application to in-situ MSI of acidity in the biomedical field.

Characterizing the phenolic constituents of baobab (Adansonia digitata) fruit shell by LC-MS/QTOF and their in vitro biological activities

Balarabe B. Ismail,Yunfeng Pu,Lihua Fan,Munir A. Dandago,Mingming Guo,Donghong Liu

Baobab ( Adansonia digitata ) fruit is a part of the baobab tree, a revered multi-purpose tree native to Africa with a myriad of potentials in providing shelter, food, medicine, clothing and as a valuable source of natural antioxidants. A massive quantity of baobab fruit shells (BFS) is generated as a waste from the baobab fruit processing representing significant economic and environmental challenges at the same time an opportunity for its valorization and commercial utilization. The present study explored the potentials of BFS as a source of phenolic compounds. For this purpose, the phenolic constituents of BFS were identified by LC-MS/QTOF analysis. Also, the Total Phenolic Chromatographic Index (TPCI), TPC, TFC, and antioxidant capacity were compared with the baobab seeds and pulp. The antidiabetic potential through α-amylase and α-glucosidase inhibitory activities was also compared with that of acarbose. The LC-MS/QTOF analysis led to the identification of 45 compounds, including quercetin, kaempferol, proanthocyanidins, phenolic acids and their derivatives, several of which had never been reported in baobab fruit. Moreover, the BFS showed higher TPC, TFC, and antioxidant capacity than the baobab seeds and pulp and inhibited α-amylase and α-glucosidase enzymes activities with much higher potency than acarbose. This research demonstrated the promising potentials of BFS as a good source of phenolic compounds that can further be utilized for food and pharmaceutical applications.

Carbon dots and cellulose nanocrystal-incorporated chitosan composite films with enhanced gas selectivity and photodynamic antibacterial properties for fruit preservation

Bingzhen Liu,Kun Wang,Fangfei Sun,Yongliang Zhuang,Hui Li,Lincai Peng

Winter jujube is prone to rapid ripening and deterioration during postharvest storage and transportation. This study developed a multifunctional chitosan (CS) composite film incorporating dialdehyde cellulose nanocrystal (DCNC) and carbon dots (CDs) derived from walnut green husks. DCNC functioned as cross-linking filler, while CDs served as active nanofillers. The CS composite film containing 3 wt% DCNC and 3 wt% CDs (CS/DCNC/CDs-3 %) exhibited significant improvements in UV-blocking, mechanical strength, gas barrier, and thermal stability, attributing to the formation of a dense film structure and synergistic effect of Schiff base and hydrogen bonding interactions among film components. Notably, this composite film demonstrated a high CO 2 /O 2 selectivity of 14.1, facilitating the creation of an ideal atmosphere inside the package. This composite film exhibited remarkable antioxidant activity and displayed potent antibacterial capacity through photocatalytic ROS generation, achieving inhibition rates of 90.8 % against S. aureus and 86.8 % against E. coli . Additionally, this film showed favorable biosafety and biodegradability, addressing food safety and environmental concerns. Postharvest preservation tests confirmed that this film assisted by light exposure effectively regulated physiological metabolism and delayed the ripening of winter jujube, highlighting its potential as an active modified atmosphere packaging material for perishable fruit preservation.

Citrus oil gland and cuticular wax inspired multifunctional gelatin film of OSA-starch nanoparticles-based nanoemulsions for preserving perishable fruit

Ying Xie,Ke Ding,Saiqing Xu,Haishan Xu,Shuai Ge,Xia Chang,Huan Li,Zijun Wang,Zisheng Luo,Yang Shan,Shenghua Ding

Inspired by the citrus oil gland and cuticular wax, a multifunctional material that stably and continuously released the carvacrol and provided physical defenses was developed to address issues of fresh-cut fruits to microbial infestation and moisture loss. The results confirmed that low molecular weight and loose structure of starch nanoparticles prepared by the ultrasound-assisted Fenton system were preferable for octenyl succinic anhydride modification compared to native starch, achieving a higher degree of substitution (increased by 18.59 %), utilizing in preparing nanoemulsions (NEs) for encapsulating carvacrol (at 5 % level: 81.58 %). Furthermore, the NEs-based gelatin (G) film improved with surface hydrophobic modification by myristic acid (MA) successfully replicated the citrus oil gland and cuticular wax, providing superior antioxidant (enhanced by 3–4 times) and antimicrobial properties (95.99 % and 84.97 % against Staphylococcus aureus and Escherichia coli respectively), as well as the exceptional UV shielding (nearly 0 transmittance in the UV region), mechanical (72 % increase in tensile strength), and hydrophobic (WCA 133.63°). Moreover, the 5%NE-G@MA film inhibited foodborne microbial growth (reduced by 50 %) and water loss (controlled below 15 %), extending the shelf life of fresh-cut navel orange and kiwi. Thus, the multifunctional film was a potential shield for preserving perishable fresh-cut products.
