FeSe2 nanosheets as a bifunctional platform for synergistic tumor therapy reinforced by NIR-II light

Lan Fang,Jie Wang,Xueliang Ouyang,Binbin Liang,Liying Zhao,Dechun Huang,Dawei Deng

Multi-functionality has been a constant pursuit in the development of next-generation drug carriers, as it will bring the potential for combination therapy by integrating diverse therapeutic modes. In this work, FeSe2 nanosheets (NSs) have been prepared as a bifunctional platform to investigate their use in synergistic cancer therapy. Bifunctional FeSe2 NSs exhibit exceptional Fenton-like activity that generates cytotoxic hydroxyl radical (˙OH) and strong broad photothermal performance including the second-infrared (NIR-II) spectral range, wherein the ˙OH production can be enhanced by NIR-II light irradiation. Furthermore, doxorubicin (DOX) was conjugated onto NSs via a pH-responsive hydrazone bond to achieve preferential drug release in an acidic microenvironment. Upon intratumoral administration, these bifunctional drug-carrying FeSe2 NSs showed an NIR-II irradiation-reinforced strong tumor suppression effect, and no obvious toxicity to normal tissues was observed. This study provides a new paradigm for the design of advanced drug carriers relying on their inherent physicochemical properties.

Simultaneous luminescence enhancement and lifetime tuning of deep UV-NIR upconversion through controlling dopant concentration

Dangli Gao,Yuqian Liang,Jie Gao,Hong Xin,Long Wang,Sining Yun,Xiangyu Zhang

Applications of rare-earth (Re)-doped upconversion (UC) micro/nanocrystals in varying areas have sparked a growing, constant demand for deliberate control over the luminescent profile and lifetime. A general strategy for adjusting UC luminescence is to control the dopant Re 3+ concentration. However, the progress of elevating dopant concentration is significantly limited owing to frequently encountering severe concentration quenching. Herein, by designing UC system using Tm 3+ with super long intermediate energy level lifetime and Yb 3+ with a simple two-level structure as a activator/sensitizer combination, we have achieved the unusually intense deep ultraviolet (UV) UC emissions at 290 nm and 345 nm, near UV emission at 361 nm, visible blue emissions at 452 nm and 478 nm, and near infrared (NIR) emission at 803 nm in NaYF 4 :Yb/Tm microtubes under 980 nm excitation. The luminescence intensity reaches about 56 times and 8 times enhancement for the UV and blue emission, respectively, with the increase of dopant Yb 3+ contents from 20 to 80 mol%. While luminescence lifetimes for all emissions decrease, particularly, for NIR emission at 803 nm, from 1507 μs down to 384 μs. Importantly, upon 980 nm irradiation, these UC NaYF 4 :Yb/Tm microtubes exhibit superior optical performance as a promising candidate of advanced anti-counterfeiting applications and other applications including UV light triggered reactions and multiplexing information storage.

Fe3+, NIR light and thermal responsive triple network composite hydrogel with multi-shape memory effect

Yan-Qin Wang,Yu Zhu,Jing-Hui Wang,Xiao-Na Li,Xiao-Gang Wu,Yi-Xian Qin,Wei-Yi Chen

Recently, artificial intelligence has driven the development of shape memory hydrogels (SMHs) with complicated environmental sensitive abilities. However, current SMHs possess fundamental limitations in terms of their responsive performance and mechanical properties , which severely limits their practical applications. Herein, a triple-network composite hydrogel with Fe 3+ , NIR light, and thermal responsive abilities is fabricated and developed as multi-SMHs, named as PDA/CMC-Fe 3+ /PVA hydrogel. In this hydrogel system, the interpenetrating triple network are formed by chemically-physically co-crosslinked polyvinyl alcohol (PVA) networks, and physically crosslinked carboxymethyl cellulose (CMC)-Fe 3+ network. In addition, polydopamine (PDA), which is introduced into the hydrogel system by in-situ polymerization way, could not only serve as an energy converter to endow the hydrogel with NIR-based responsive ability, but also enhance its mechanical strength . Remarkably, the as-prepared PDA/CMC-Fe 3+ /PVA hydrogels with the optimized compositions exhibit relatively excellent mechanical properties (tensile strength = 163.48 ± 10.42 kPa, elastic modulus  = 45.42 ± 3.96 kPa, toughness = 165.02 ± 7.01 kJ/m 3 ) due to the synergistic effect of the triple-network and PDA doping. Moreover, the composition of the hydrogel is optimized to realize a stable temporary shape and rapid recovery to the permanent shape, which is controlled by Fe 3+ , NIR irradiation, and thermal stimulus. The resultant hydrogels exhibit not only multi-stimulus responsive abilities, but also programmable multi-shape memory properties. The versatile shape memory properties endow potential for the materials to be used in the field of artificial intelligence.

NIR-Driven Water Splitting H2 Production Nanoplatform for H2-Mediated Cascade-Amplifying Synergetic Cancer Therapy

Qi Wang,Yishun Ji,Jinsheng Shi,Lili Wang

As a newly emerging treatment strategy for many diseases, hydrogen therapy has attracted a lot of attention because of its excellent biosafety. However, the high diffusivity and low solubility of hydrogen make it difficult to accumulate in local lesions. Herein, we develop a H2 self-generation nanoplatform by in situ water splitting driven by near-infrared (NIR) laser. In this work, core–shell nanoparticles (CSNPs) of NaGdF4:Yb,Tm/g-C3N4/Cu3P (UCC) nanocomposites as core encapsulated with zeolitic imidazolate framework-8 (ZIF-8) modified with folic acid as shell are designed and synthesized. Due to the acid-responsive ZIF-8 shell, enhanced permeability and retention (EPR) effect, and folate receptor-mediated endocytosis, CSNPs are selectively captured by tumor cells. Upon 980 nm laser irradiation, CSNPs exhibit a high production capacity of H2 and active oxygen species (ROS), as well as an appropriate photothermal conversion temperature. Furthermore, rising temperature increases the Fenton reaction rate of Cu(I) with H2O2 and strengthens the curative effect of chemodynamic therapy (CDT). The excess glutathione (GSH) in tumor microenvironment (TME) can deplete positive holes produced in the valence band of g-C3N4 in the g-C3N4/Cu3P Z-scheme heterojunction. GSH also can reduce Cu(II) to Cu(I), ensuring a continuous Fenton reaction. Thus, a NIR-driven H2 production nanoplatform is constructed for H2-mediated cascade-amplifying multimodal synergetic therapy.

A NIR-triggered multifunctional nanoplatform mediated by Hsp70 siRNA for chemo-hypothermal photothermal synergistic therapy

Bo Zhang,Chen-wen Shao,Kang-Min Zhou,Qin Li,Yong-Tao Duan,Yu-Shun Yang,Hai-Liang Zhu

Recently, hypothermal photothermal therapy (HPTT) seemed essential for the future clinical transformation of cancer optical therapies. However, at a lower working temperature, heat shock proteins (HSPs) seriously affect the anti-tumor effect of HPTT. This work reports a reasonable design of a dual-responsive nanoplatform for the synergistic treatment of chemotherapy and HPTT. We adopted a one-step method to wrap indocyanine green (ICG) into imidazole skeleton-8 (ZIF-8) and further loaded it with the chemotherapy drug doxorubicin (DOX). Furthermore, we introduced Hsp-70 siRNA to block the affection of HSPs at an upstream node, thereby avoiding the side effects of traditional heat shock protein inhibitors. The prepared ZIF-8@ICG@DOX@siRNA nanoparticles (ZID-Si NPs) could significantly improve the stability of siRNA to effectively down-regulate the expression of HSP70 protein during the photothermal therapy, thus realizing the pH-controlled and NIR-triggered release of the chemotherapeutical drug DOX. Moreover, tumors were also imaged accurately by ICG wrapped in ZID-Si nanoparticles. After the evaluation of the in vitro and in vivo photothermal effect as well as the anti-tumor activity, we found that the added Hsp-70 siRNA enhanced the synergistic anti-cancer activity of HPTT and chemotherapy. In summary, this work holds great potential in cancer treatment, and suggests better efficacy of synergistic chemo/HPTT than the single-agent therapy.

NIR-driven fast construction of patterned-wettability on slippery lubricant infused surface for droplet manipulation

Qingqing Rao,Zheming Tong,Lina Song,Abid Ali,Yang Hou,Qinggang He,Jianguo Lu,Xiang Gao,Xiaoli Zhan,Qinghua Zhang

Controllable wettability is highly desired for droplet manipulation technology in biomedical system and industry. Here, a photo-responsive slippery surface (G/NPGS) is fabricated based on the silicon oil and graphene composited polymer network. Under near-infrared (NIR) irradiation, the lubricant oil in the bulk of the substrate can be selectively released to the targeted area and then reversibly erased by cooling treatment. Attractively, discretionary patterned-wettability and on-demand droplet-guiding pathway can be constructed within few seconds (5 ~ 10 s). The droplet motion on this surface can be flexibly and spatiotemporally manipulated. The programmable wettability of G/NPGS can not only used for information transmission but also for droplet pipetting, liquid transportation and manipulation in microplates, microfluidics and microreactors. Moreover, the G/NPGS can be applied for photoresist field to replace the developer solution. These findings provide a novel and facile way to rapidly construct programmable wettability, which is highly significant for liquid manipulation, laboratory-on-a-chip settings and microfluidics.

Electrospun PCL/MoS2 Nanofiber Membranes Combined with NIR-Triggered Photothermal Therapy to Accelerate Bone Regeneration

Ke Ma,Chuanan Liao,Lanli Huang,Ruiming Liang,Jinmin Zhao,Li Zheng,Wei Su

Electrospun nanofiber membranes have been widely used for guided bone regeneration (GBR). For assistance in bone healing, photothermal therapy which renders moderate heat stimulation to defect regions by near-infrared (NIR) light irradiation has attracted much attention in recent years. Combined with photothermal therapy, novel electrospun poly(ε-caprolactone)/molybdenum disulfide (PCL/MoS 2 ) nanofiber membranes are innovatively synthesized as GBR for bone therapy, wherein the exfoliated MoS 2 nanosheets served as osteogenic enhancers and NIR photothermal agents. With the doping of MoS 2 , the mechanical properties of nanofiber membranes got improved with the degradation unaffected. The composite PCL/MoS 2 membranes show enhanced cell growth and osteogenic performance compared with PCL alone. Under NIR-triggered mild photothermal therapy, osteogenesis and bone healing are accelerated by using PCL/MoS 2 nanofiber membranes for growth of bone mesenchymal stem cells in vitro and repair of rat tibia bone defect in vivo. The novel nanofiber membranes may be developed as intelligent GBR in the therapy of bone defects.

Carbon dot/WS2 heterojunctions for NIR-II enhanced photothermal therapy of osteosarcoma and bone regeneration

Bijiang Geng,Hua Qin,Wenwen Shen,Ping Li,Fuling Fang,Xiaokai Li,Dengyu Pan,Longxiang Shen

Osteosarcoma as an aggressive malignant neoplasm is commonly treated by surgical resection, chemotherapy, and radiotherapy, but surgery induces bone defects and chemo/radiotherapy causes severe systemic toxicity. We have designed a novel low-toxic bifunctional 0D/2D heterojunction (HJ) that can be used to generate effective photothermal effects in the second NIR window (NIR-II, 1000–1350 nm) for simultaneous osteosarcoma treatment and bone regeneration. To construct this 0D/2D nanostructure, biocompatible, positively charged NIR-II-responsive carbon dots (CDs) were synthesized by an ultrafast microwave-assisted hydrothermal method, followed by electrostatically driven assembly on both sides of negatively charged WS 2 nanosheets. We found that the HJs exhibited enhanced NIR-II absorption and photothermal conversion efficiency. Due to the deep-tissue penetration ability of 1064 nm laser, complete osteosarcoma ablation was realized at a low power density even when being covered by a 10-mm-thick additional tissue. Furthermore, osteogenic differentiation was remarkably enhanced by mild photothermal effects of CD/WS 2 HJs, which significantly up-regulated bone-related gene expressions, particularly, heat shock protein expression. Designed CD/WS 2 HJs can be used in osteosarcoma treatment and bone-tissue engineering.

A full-spectrum-absorption from nickel sulphide nanoparticles for efficient NIR-II window photothermal therapy

Ziyu Lei,Wenlong Zhang,Bo Li,Guoqiang Guan,Xiaojuan Huang,Xuan Peng,Rujia Zou,Junqing Hu

Near-infrared (NIR) light has been widely applied in the field of photothermal therapy (PTT). Recent advances in the light wavelength for efficient cancer PTT have gradually shifted from the first NIR (NIR-I) biowindow (700–1000 nm) to the second NIR (NIR-II) biowindow (1000–1350 nm) owing to its intrinsic deeper tissue penetration ability and a higher maximum permissible exposure (MPE) value. Herein, we have prepared nickel sulphide (Ni9S8) nanoparticles (NPs) with a full-spectrum-absorption (400 nm–1100 nm) in the NIR region. By a fair comparison, it is found that the PTT using the NPs upon irradiation from an NIR-II (i.e., 1064 nm) laser is more efficient than that from an NIR-I (i.e., 808, 915, and 976 nm) laser. The large mass extinction coefficient value (22.18 L g−1 cm−1) and high photothermal conversion efficiency (46%) at 1064 nm make these NPs promising candidates for NIR-II photo-thermal therapy. This study will benefit future exploration and optimization of nickel-based photoabsorbers utilizing NIR-II light for photothermal applications.

Localized Free Radicals Burst Triggered by NIR-II Light for Augmented Low-Temperature Photothermal Therapy

Boshu Ouyang,Funan Liu,Shuangrong Ruan,Yao Liu,Huishu Guo,Zhengxu Cai,Xiangrong Yu,Zhiqing Pang,Shun Shen

As a novel treatment modality of tumors, hypothermal hyperthermia employed relatively lower temperature (<45 °C) to damage cancer cells with mild toxicity to normal tissues. However, beyond that inducible heat resistance of tumor cells, the discounted therapeutic effect of low temperature hyperthermia was also ascribed to poor penetration of exogenous light stimulation and low accumulation of photothermal agents in tumor sites. Herein, we constructed a multifunctional in situ hydrogel of sodium alginate (ALG) via Ca2+ coordinated with ALG to encapsulate the photothermal agent of Ink and azo initiator of 2,2′-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride (AIPH) for effective tumor treatment. The designed ALG hydrogel was used to improve the therapeutic effect by increased accumulation of Ink and AIPH and avoid potential side-effects caused by the unexpected spread to the surrounding normal tissues. After injection, local low temperature stimulation was generated with near-infrared-II irradiation by a 1064 nm laser, triggering rapid decomposition of AIPH to produce alkyl radicals. The synergistic low temperature photothermal therapy and cytotoxic-free radicals enhanced the apoptosis of tumor cells via physical heat damage and lipid peroxidation. Thus, remarkable inhibition of tumor growth was observed in a subcutaneous colorectal cancer with negligible side effects. Furthermore, the formulation could also exert strong photoacoustic signals, which were utilized to monitor the stability of the composite hydrogel.

Recyclable magnetic Fe3O4@SiO2/β-NaYF4:Yb3+,Tm3+/TiO2 composites with NIR enhanced photocatalytic activity

Zhangxu Chen,Ming-Lai Fu

To extend the activities of TiO 2 to the near infrared (NIR) region and to separate and recycle photocatalysts easily is vital for the high harvest of solar energy for sustainable society. The magnetic NIR activated Fe 3 O 4 @SiO 2 /β-NaYF 4 :Yb 3+ ,Tm 3+ /TiO 2 (FS/UC/T) composites have been synthesized by using the solvothermal method by adding Fe 3 O 4 @SiO 2 and Degussa P25 into the mixed solution of NaOH, ethanol, oleic acid, Re(NO 3 ) 3 and NH 4 F. Moreover, the obtained composites were explored for photocatalytic applications. It was found that FS/UC/T emit UV–vis light under irradiation of 980 nm laser and efficient energy transfer from β-NaYF 4 :Yb 3+ ,Tm 3+ to TiO 2 was verified. The photodegradation experiments for treating methylene blue (MB), rhodamine B (RhB), methyl orange (MO) and colorless phenol were carried out to evaluate the photocatalytic activities of the samples. The photodegradation rates reach 76.62%, 68.48%, 30.05% and 27.16% for MB, RhB, MO and phenol under irradiation of 980 nm laser, respectively, which suggested magnetic FS/UC/T composite is an advanced NIR-driven photocatalyst. Moreover, the magnetic photocatalyst showed easy separation and good stability in the recycled tests. It suggested a promising system to utilize the NIR energy for photocatalysis based on TiO 2 and to utilize the magnetic for separation of photocatalysts, which will contribute to practical environmental remediation in the future.

Fabrication of High-Performance Magnetic Lysozyme-Imprinted Microsphere and Its NIR-Responsive Controlled Release Property

Jinxing Chen,Shan Lei,Yunyun Xie,Mozhen Wang,Jun Yang,Xuewu Ge

The preparation of efficient and practical biomacromolecules imprinted polymer materials is still a challenging task because of the spatial hindrance caused by the large size of template and target molecules in the imprinting and recognition process. Herein, we provided a novel pathway to coat a NIR-light responsive lysozyme-imprinted polydopamine (PDA) layer on a fibrous SiO2 (F-SiO2) microsphere grown up from a magnetic Fe3O4 core nanoparticle. The magnetic core–shell structured lysozyme-imprinted Fe3O4@F-SiO2@PDA microspheres (MIP-lysozyme) can be easily separated by a magnet and have a high saturation adsorption capacity of lysozyme of 700 mg/g within 30 min because of the high surface area of 570 m2/g and the mesopore size of 12 nm of the Fe3O4@F-SiO2 support. The MIP-lysozyme microspheres also show an excellent selective adsorption of lysozyme (IF > 4). The binding thermodynamic parameters studied by ITC proves that the lysozyme should be restricted by the well-defined 3D structure of MIP-lysozyme microspheres. The MIP-lysozyme can extract lysozyme efficiently from real egg white. Owing to the efficient NIR light photothermal effect of PDA layer, the MIP-lysozyme microspheres show the controlled release property triggered by NIR laser. The released lysozyme molecules still maintain good bioactivity, which can efficiently decompose E. coli. Therefore, this work provides a novel strategy to build practical NIR-light-responsive MIPs for the extraction and application of biomacromolecules.
