A NIR-II Laser-Triggered Nb2C-Arg/Gel Composite Multifunctional Hydrogel with Photothermal, ROS Scavenging, and NO Release for Infected Wound Healing

Mi Liu,Weidan Zhao,Denghao Li,Dingyu Cui,Jiali Liu,Qingao Chen,Yan Zhou,Li Jiang,Huaibin Zhou

Infected wound healing remains a significant clinical challenge due to bacterial resistance and excessive inflammation. Herein, a multifunctional hydrogel dressing (Nb2C-Arg/Gel) is developed that integrates two-dimensional Nb2C nanosheets (photothermal agent) and arginine [nitric oxide (NO) donor] into a calcium ion cross-linked sodium alginate matrix. The hydrogel has a relatively high swelling capacity and good ability to scavenge reactive oxygen species (ROS), which are beneficial for absorbing wound exudate and reducing the inflammatory response of wounds. Under irradiation with a low-power NIR-II laser (1064 nm, 1 W cm–2), Nb2C-Arg/Gel achieves rapid photothermal heating (48.6 °C in 1 min), while thermally enhances arginine decomposition-triggered on-demand NO release. This synergistic mechanism provides superior antimicrobial efficacy against Staphylococcus aureus (97.65%) and methicillin-resistant S. aureus (90.05%). In vivo studies on MRSA-infected murine wounds demonstrate that the residual area on day 10 was 2.9%, which accelerated healing compared to that of the control group (17.8%), accompanied by reduced inflammation and enhanced angiogenesis. The excellent biocompatibility of hydrogel is validated by low hemolysis (<5%) and promoted fibroblast proliferation (125% cell viability). Combining with photothermal, NO-mediated antibacterial action, and ROS scavenging to alleviate oxidative stress, this antibiotic-free hydrogel dressing has great potential for clinical application for the treatment of refractory-infected wounds.

Gas–Solid Two-Phase Flow Synthesis Equipment: A New Method for Continuous, Large-Scale Preparation of Metal–Organic Frameworks with No Solvent

Jun Zhao,Rufang Peng

Here, we demonstrated the continuous synthesis of metal–organic frameworks (MOFs) (ZnBTeC/MnBTeC/Ni4[(phen)2(BeTC)]/Co4[(phen)2(BeTC)]) by using a brand new device—gas–solid two-phase flow synthesis equipment. Quantitative conversions occurred to give products at 60 kg h–1 rates. The single-crystal X-ray diffraction validated the precise structure of these MOFs. Also, it should be noted that the reactions were performed under completely solvent-free conditions. In addition, kinetic analysis was carried out to study the reaction mechanisms.

S-nitrosoglutathione functionalized polydopamine nanoparticles incorporated into chitosan/gelatin hydrogel films with NIR-controlled photothermal/NO-releasing therapy for enhanced wound healing

Wenyu Wang,Huan Sheng,Daihong Cao,Fenglian Zhang,Weifen Zhang,Fang Yan,Dejun Ding,Ni Cheng

Nitric oxide (NO) has aroused wide interest in the treating infected wounds due to its characteristic functionalities. However, its utilization is limited due to its volatile properties, high reactivity, direct potential toxicity, and byproducts of NO donors limited its application. Herein, endogenously NO donor S-nitrosoglutathione (GSNO) was connected covalently to polydopamine nanoparticles (PDA-GSNO NPs) to minimize the loss of NO in aqueous medium. Meanwhile, near-infrared (NIR)-controlled NO release and photothermal therapy (PTT) was obtained through the photothermal conversion by PDA. Then chitosan (CS)/gelatin (GE) biocomposite hydrogel films with preferable biocompatibility, surface hydrophilicity, hydroabsorptivity, and mechanical adhesive properties were constructed. By embedding PDA-GSNO NPs into the films, a multifunctional wound dressing was fabricated. Under NIR light irradiation, the combination of PTT, NO-releasing, and CS antibacterial agents can strengthen the in vitro antimicrobial efficacy and in vivo wound healing activities. Meanwhile, the obtained wound dressing presented good biocompatibility. This work outlines an approach for combating bacterial infections and demonstrating the possibility for synergistic NO-releasing wound healing.

Novel Co2+ passivated carbon nanodots with up-conversion effects combined with NH2-MIL-125 for improving photocatalytic NO purification and hydrogen evolution

Youzhou He,Shuang Luo,Yuwei Tan,Mingzuo Chen,Haibo Zhou,Jianning Yu,Linjiang Pu,Yumin Huang,Min Fu,Xingyan Liu

Novel ionic state Co passivated carbon nanodots (Co-CDs) combined with NH 2 -MIL-125 as efficient photocatalysts Co-CDs/NH 2 -MIL-125 were fabricated in a convenient process for the first time. These materials exhibited excellent photocatalytic activity in both NO purification and hydrogen production under visible light irradiation. The NO purification rate of Co-CDs/NH 2 -MIL-125 increased to 46.6%, which was 20.2% and 13.4% higher than that of NH 2 -MIL-125 and CDs/NH 2 -MIL-125, respectively. Moreover, the hydrogen production efficiency of Co-CDs/NH 2 -MIL-125 reached 2261 μmol g −1 , which was 6.86 times and 3.54 times of the values for NH 2 -MIL-125 and CDs/NH 2 -MIL-125, respectively. The corresponding properties characterized by Mott-Schottky, UCPL , and in situ DRIFTS , indicated that the introduction of Co 2+ could not only passivate the CDs to further improve physicochemical properties, but also enhance the photocatalytic activity via the photosensitization and up-conversion effect. This research provides a promising strategy that adding active metal ions passivated CDs in the catalyst platform to improve photocatalytic performance.

Ye Tian,Ke-Ke Wang,Gui-Peng Yang,Pei-Feng Li,Chun-Ying Liu,Riel Carlo O. Ingeniero,Hermann W. Bange

Nitric oxide (NO) is a short-lived intermediate of the oceanic nitrogen cycle, and it is produced by biological and photochemical processes in the ocean. Nitrogen dioxide (NO2) is a reactive atmospheric compound which has not been determined in the ocean so far. Here, we present the setup and validation of a novel continuous underway measurement system to measure dissolved NO and NO2 in the surface ocean. The system consists of a seawater/gas equilibration component coupled to a chemiluminescence detector. It was successfully deployed during a 12 day cruise to the East China Sea in May 2018. Dissolved NO and NO2 surface concentrations ranged from

Time-resolved in situ/operando infrared spectroscopy of Cu-supported zeolite X catalyst during NH3–SCR of NO: A modulated excitation study

Jie Yang,Xinyu Li,Yujia Xu,Lin Chen,Shan Wang,Shan Ren,Junjiang Zhu

Zeolite X was used as support of copper catalyst (Cu/X) for selective catalytic reduction (SCR) of nitrogen oxide (NO) with ammonia (NH 3 ). Modulated excitation in situ/ operando infrared (IR) spectroscopy and phase-sensitive detection (PSD) were applied to study the reaction intermediates that formed on the Cu/X during transient experiments. Changing the gas concentration to periodically create unsteady conditions enabled the selective extraction of the dynamic species that were directly involved in the reaction. The IR results showed that only Lewis acid sites (L X ) existed on zeolite X,and Brønsted acid sites (B) and extra Lewis acid sites (L Cu ) were generated after Cu loading. The transient experiment results indicated that the SCR reactions on Cu/X obeys the Eley − Rideal (E − R) mechanism, in which NH 3 is first adsorbed on the L X and L Cu sites (L X –NH 3 , L Cu –NH 3 ) and then moved to the B sites (B–NH 3 ), where it reacted with gas-phase NO to form NH 2 NO intermediate, which finally decomposed into N 2 and H 2 O. Moreover, it is found that the NH 3 adsorbed on L Cu and B sites exhibits stronger reactivity than that on the L X sites.

Noninterventional target-embolization combined with NO gas therapy for solid tumors by pH-responsive poly(amino acid)s with S-nitrosothiols group

Zhengpeng Chen,Lei Wang,Yuanyuan Jin,Hongyun Guo,Yongdong Zhang,Mingzhong Ma,Xiaobo Wang,Xing Zhou,Ziqiang Lei,Dedai Lu

Gas therapy and embolization are important treatment methods for solid tumors. In order to improve the efficacy of NO therapy and simplify the operation of interventional embolization, we designed and prepared the microacidic-gelled poly[(L-Glutamic acid- ran -L-Tyrosine)- b -L-Threonine- b -L-S-nitrosocysteine]s (PGTTSNCs, obtained by the reaction of tert-butyl nitrite with thiol side groups of poly[(L-Glutamic acid- ran -L-Tyrosine)- b -L-Threonine- b -L-Cysteines]s, PGTTCs.) for noninterventional embolization combined with NO gas therapy for solid tumors. The results of in vivo experiments indicated that PGTTSNCs from PGTTCs-6.4 (the gelled-pH value is 6.4) has significant target-accumulation and therapy effects, and at 10 days post-injection only scars were found at the tumor site. After 20 days, the tumors of model mice were completely disappeared. The method is extremely simple and efficient for solid tumors, which is not only a great innovation of tumors theranostic, but also a qualitative development in nanomaterials .

Novel synthesis of fly-ash-derived Cu-loaded SAPO-34 catalysts and their use in selective catalytic reduction of NO with NH3

Ge Li,Baodong Wang,Qi Sun,Wayne Qiang Xu,Ziran Ma,Hongyan Wang,Daojun Zhang,Jiali Zhou

A combined acid–alkali hydrothermal method was used to prepare fly ash –derived SAPO-34 molecular sieves from a thermal power plant in Inner Mongolia (China). The specific surface area of the prepared fly-ash-derived SAPO-34 molecular sieves was 579 m 2 g −1 , the total pore volume was about 0.27 cm 3 g −1 , and the pore size was 0.56 nm; the molar ratios of Al 2 O 3 :P 2 O 5 :SiO 2 were 1:0.86:0.45. Cu/SAPO-34 catalysts were prepared by impregnation of low-cost fly-ash-derived SAPO-34 molecular sieves as a support and tested in selective catalytic reduction with NH 3 (NH 3 -SCR). Powder X-ray diffraction (XRD), N 2 adsorption–desorption, X-ray photoelectron spectroscopy (XPS), H 2 temperature-programmed reduction (H 2 -TPR), NH 3 temperature-programmed desorption (NH 3 -TPD), electron paramagnetic resonance (EPR), nuclear magnetic resonance (NMR), X-ray fluorescence analysis (XRF) and scanning electron microscopy (SEM) were used for catalyst characterization and investigation of the relationships between the catalyst structure and the catalytic activity. The actual silica:alumina ratio of the molecular sieves did not increase with increasing Cu loading, indicating that increasing the Cu loading does not change the original structure of the SAPO-34 molecular sieves. The XRF and NMR results showed that replacement by Cu results in more Si islands. The molecular sieve acidity decreased because of the increased number of Si islands. The NH 3 -TPD results showed that for the Cu/SAPO-34 catalysts there was a low correlation between the low-temperature activity and the amount of acidic sites. SCR activity is closely related to the location of Cu. The 4.47Cu/SAPO-34 catalyst has the highest isolated Cu 2+ showed the highest NH 3 -SCR activities (> 90%) at 250–350 °C. This work opens up new avenues for recycling fly ash formed in coal-fired power plants (reducing environmental pollution) and developing low-cost SCR catalysts for NO x pollution control.

Formulation optimization and pharmacokinetics evaluation of oral self-microemulsifying drug delivery system for poorly water soluble drug cinacalcet and no food effect

Mengyuan Cao,Xu Xue,Xixi Pei,Yiwen Qian,Lan Liu,Lili Ren,Guoguang Chen

The present research indicated that a new self-microemulsifying drug delivery systems (SMEDDS) were used to reduce the food effect of poorly water-soluble drug cinacalcet and enhance the bioavailability in beagle dogs by oral gavage. Ethyl oleate, OP-10, and PEG-200 was selected as the oil phase, surfactant and co-surfactant of cinacalcet-SMEDDS by the solubility and phase diagram studies. Central Composite Design-Response Surface Methodology was used to determine the ratio of surfactant and co-surfactant, the amount of oil for optimizing the SMEDDS formation. The prepared formulations were further characterized by the droplet size, self-microemulsifying time, zeta potential, polydispersity index (PDI), and robustness to dilution. The in vitro release profile of cinacalcet-SMEDDS was determined in four different release medium and in fasted state and fed state of simulated gastrointestinal fluid. Cinaclcet-SMEDDS were implemented under fed and fasted state in dogs and product REGPARA® was used as a comparison to the prepared formulation in the pharmacokinetics. The result showed the components of SMEDDS, the amount of oil, the ratio of surfactant, and co-surfactant was optimized using solubility, pseudo-ternary phase diagram studies, and response surface methodology. In vitro drug release studies indicated that the cinacalcet-SMEDDS eliminated the effect of pH variability in release medium and variational gastroenteric environments with improved drug release performance. Pharmacokinetic studies revealed that the profiles of cinacalcet-SMEDDS were similar both in the fasted and fed state compared with commercial product, indicating the formulation significantly promoted the absorption, enhanced bioavailability and had no food effect essentially. It is concluded that poorly water-soluble drug cinacalcet was improved in the solubility and bioavailability by using a successful oral dosage form the SMEDDS, and eliminated food effect as well.

Co2+/Co3+ redox reaction in Co-doped Bi4Ti3O12 for enhancing photocatalytic NO removal: Interplay of superoxide radicals and in-situ DRIFTS study

Jiahe Zhai,Hui Wang,Abdelkader Labidi,Long Zeng,Yu Huang,Chuanyi Wang

Removing low-concentration NO in the air by means of photocatalytic technology is a promising approach. Herein, our work pins down the mechanism of cobalt metal doping within the lattice of Bi 4 Ti 3 O 12 (BTO) for the photocatalytic NO conversion. As a result, the Co-BTO exhibits a NO conversion efficiency of 58.3 %, which is substantially higher than that of BTO (30 %). More interestingly, the Co-BTO also shows a lower production of intermediate NO 2 than that of the BTO. These findings demonstrate that within Co-BTO, both Co²⁺ and Co³ ⁺ coexist. The redox cycle between these two cobalt species engenders active sites possessing strong reducing capabilities for O₂ which can be adsorbed and activated at the Co sites, thereby generating superoxide radicals (•O₂⁻). Furthermore, the doping of a certain portion of cobalt broadened light absorption and enhanced the separation efficiency of photogenerated carriers, further facilitating the activation of O₂. Lastly, electron spin resonance (ESR) and in-situ diffuse reflective infrared Fourier transform spectroscopy (in-situ DRIFTS) were respectively utilized to further clarify the active species involved in the photocatalytic oxidation of NO and its adsorption and conversion pathways from a mechanistic standpoint. Overall, this work represents a novel endeavor for the advancement of visible-light-driven photocatalysts usage for environmental remediation against NO pollution.

Constructing sodium alginate/carboxymethyl chitosan coating capable of catalytically releasing NO or CO for improving the hemocompatibility and endothelialization of magnesium alloys

Changjiang Pan,Changpeng Zuo,Jie Chen,Qiuyang Zhang,Linghong Deng,Yang Liu,Pingyun Ding

Although significant progress in developing biodegradable magnesium alloy materials in cardiovascular stents has been achieved recently, they still face challenges such as rapid in vivo corrosion degradation, inferior blood compatibility, and limited re-endothelialization after the implantation. Hydrogel coating that can catalyze the liberation of gas signal molecules offers a good solution to alleviate the corrosion rate and enhance the biocompatibility of magnesium and its alloys. In this study, based on alkaline heat treatment and construction of polydopamine coating on the surface of magnesium alloy, sodium alginate/carboxymethyl chitosan (SA/CMCS) gel was simultaneously covalently grafted onto the surface to build a natural polymer hydrogel coating, and selenocystamine (SeCA) and CO release molecules (CORM-401) were respectively immobilized on the surface of the hydrogel coating to ameliorate the anticoagulant performance and accelerate endothelial cells (ECs) growth by catalyzing the release of endogenous gas signal molecules (NO or CO). The findings verified that the as-prepared hydrogel coating can catalyze the liberation of CO or NO and significantly improve the corrosion resistance of magnesium alloy. At the same time, owing to the excellent hydrophilicity of the hydrogel coating, the good anticoagulant property of sodium alginate, and the ability of CMCS to promote the ECs growth, the modified magnesium alloy could significantly improve the albumin adsorption while preventing the adsorption of fibrinogen, hence significantly augmenting the anticoagulant properties and promoting the ECs growth. Under the catalytic release of NO or CO, the released gas molecules further enhanced hemocompatibility and promoted endothelial cell (EC) growth and the expression of vascular endothelial growth factor (VEGF) and NO of ECs. Therefore, the bioactive coatings that can catalyze the release of NO or CO have potential applications in constructing surface bioactive coatings for magnesium alloy materials used for intravascular stents.

Promotional effect of the TiO2 (001) facet in the selective catalytic reduction of NO with NH3: in situ DRIFTS and DFT studies

Lei Huang,Kaiwen Zha,Supawadee Namuangruk,Anchalee Junkaew,Xin Zhao,Hongrui Li,Liyi Shi,Dengsong Zhang

Surface engineering of TiO2 is significant in the SCR reaction and worth studying. In a previous study, we found that TiO2 nanosheets (TiO2-NS) with the (001) plane exhibited a much better SCR performance than TiO2 nanoparticles (TiO2-NP) with the (101) plane. In this work, in situ DRIFTS and DFT calculations were applied to address the promotional effect of the (001) plane of TiO2-NS in the SCR reaction. The behavior of adsorption and desorption of NOx and NH3 on the two surfaces was studied. It was found that NH3 adsorbed on both TiO2-NS and TiO2-NP was in the form of NH3(g) on the Lewis acid sites. NOx on TiO2-NP were mainly trans-(NO)2 and N2O4 which were non-reactive. Differently, NO on TiO2-NS was mainly in the form of NO2 which was probably due to the high energy and abundant active oxygen species of the (001) facet. The formed NO2 could trigger the subsequent ‘fast SCR’ reaction thereafter promoting the activity. Therefore, TiO2-NS with more (001) facets had better SCR activity than TiO2-NP with the exposed (101) facet.
