Dual-responsive PEG-lipid polyester nanoparticles for siRNA and vaccine delivery elicits anti-cancer immune responses by modulating tumor microenvironment

Zixu Liu,Linxuan Zhao,Yupeng Feng,Qingqing Wang,Nan Dong,Yu Zhang,Tian Yin,Haibing He,Xing Tang,Jingxin Gou,Li Yang

Cancer vaccine-based immunotherapy has great potential; however, vaccine has been hindered by the immunosuppressive tumor microenvironment (TME). In this study, dual-responsive PEG-lipid polyester nanoparticles (PEG BR647-NP) for tumor-targeted delivery were proposed. PEG BR647-NP containing model tumor associated antigen (TAA) OVA and signal transduction and activator of transcription 3 (STAT3) siRNA were delivered to tumor. PEG BR647-NP was internalized by tumor associated dendritic cells (TADCs), where TAA and siRNA were released into cytoplasm via endo/lysosome escape effect. The released OVA was presented by the major histocompatibility complex class I to activate T cells, the released STAT3 siRNA acted to relieve TADCs dysfunction, promote TADCs maturation, improve antigen-presenting ability, and enhance anticancer T cell immunity. Meanwhile, PEG BR647-NP was ingested by tumor cells, and could kill tumor cells by the pro-apoptosis effect of STAT3 siRNA. Moreover, PEG BR647-NP could reduce the proportion of myeloid-derived suppressor cells (MDSCs) and regulatory T cells (Tregs) in tumors and abrogate immunosuppression. The integration of relieved TADCs dysfunction, promoted TADCs maturation, enhanced antigen cross-presentation, abrogated immunosuppression, and improved pro-apoptosis effect can boost the vaccination for tumor immunotherapy. Thus, PEG BR647-NP efficiently delivered vaccine and STAT3 siRNA to tumor, and modulated immunosuppressive TME, provided better antitumor effects.

Effects of Porogen PEG and Pore Structure of PVDF Substrates on the Permeability–Selectivity Trade-off of TFC-NF Membranes

Chenyu Zhu,Xue Zhang,Fuzhi Li,Ruochen Yang,Xuan Zhao

Thin-film composite (TFC) nanofiltration (NF) has been widely utilized to separate monovalent ions from multivalent ions in industrial wastewater treatment. In this study, the pore properties of polyvinylidene fluoride (PVDF) substrates were adjusted by varying the content and molecular weights (MWs) of porogen polyethylene glycol (PEG). The TFC membranes fabricated on different substrates showed distinguishable permeance and selectivity. The PVDF membranes with 2 kDa PEG of 0.5–3.0 wt % and 20 kDa PEG of 0.2 wt % were fit for the substrates of the TFC NF membrane. These substrates are usually characterized by evenly distributed surface pores with porosity of 7–12% and tiny finger-like pores of less than 15 μm length. Such pore structures in the substrates are fit for the storage and migration of amine monomers, resulting in a defect-free PA layer. In a 6 h filtration of mining wastewater using an optimized TFC membrane, the permeance remained around 5 LMH/bar and the separation factor of SO42–/Cl– stayed over 13.39 with a maximum value over 30. This study shed light on improving NF membrane performance by simply adjusting the porogen in the substrate casting solution.

Design, optimization and in vitro-in vivo evaluation of smart nanocaged carrier delivery of multifunctional PEG-chitosan stabilized silybin nanocrystals

Yangyang Liu,Yancai Wang,Juan Zhao

This study explored the feasibility of using smart nanocaged carrier technology for the delivering of multifunctional mPEG-chitosan stabilized silybin nanocrystals and to enhancing the long-term stability, dissolution velocity and bioavailability of the water insoluble drugs. The methoxypoly(ethylene glycol)- b -poly(ε-caprolactone) (mPEG- b -PCL), as nanocaged carrier, was modified with propargyl and azide group and confirmed by FTIR . The methoxypolythylene glycol-grafted chitosan (PEG-CS), as multifunctional stabilizer, were applied for the silybin nanocrystals formulation. Two silybin nanocrystals formulations (PEG-CS nanocrystals and nanocaged nanocrystals) were prepared using anti-solvent precipitation method and optimized by central composite design-response surface model. The transmission electron microscopy , scanning electron microscopy and atomic force microscope revealed small and uniformed morphology. The crystalline state of the nanocrystals was confirmed by X-ray powder diffraction and differential scanning calorimetry . The drug structure was confirmed by Fourier transform infrared spectroscopy. During the long term stability study, the nanocaged nanocrystals were presented remarkable stability. Compared with the silybin solution, the nanocaged nanocrystals and the PEG-CS nanocrystals drug delivery system also exhibited 5.54 and 2.58 fold increasing in the AUC, respectively. Thus, the nanocaged nanocrystals technology with excellent stability, improved dissolution velocity and relative bioavailability was recommended as an efficient and feasible approach for water insoluble drugs delivery.

Improved separation and antifouling properties of PVDF gravity-driven membranes by blending with amphiphilic multi-arms polymer PPG-Si-PEG

Haicheng Jiang,Qi Zhao,Panpan Wang,Jun Ma,Xuedong Zhai

Fabricating high-flux, low-fouling gravity-driven membranes (GDM) has proved to be an extremely challenging work. In this study, a novel tailored amphiphilic multi-arms polymer poly(propylene glycol)-silane-poly(ethylene glycol) (PPG-Si-PEG) was synthesized and blended into Polyvinylidene Fluoride (PVDF) matrix to fabricate GDM via nonsolvent induced phase separation method. The incorporation of multi-arms polymers affected phase separation and surface segregation behaviors, resulting in the formation of more porous structure, which were demonstrated by scanning electron microscopy (SEM). Moreover, the hydrophilic PEG arms in polymers were enriched onto membrane surface through surface segregation and utilized to improve the hydrophilicity of membranes, which were confirmed by Fourier transform infrared (FTIR), X-ray photoelectron spectroscopy (XPS) and contact angle measurement. The separation and antifouling properties of the membranes were evaluated by using gravity as the driving force, employing bovine serum albumin (BSA) as the model foulant. The water flux of membranes increased from 4.6 L m −2  h −1 for the control membrane to 12.1 L m −2  h −1 for PVDF/PPG-Si-PEG∼5% membrane. The antifouling properties of modified membranes were effectively improved in comparison with that of control membrane. The flux recovery ratio, especially for PVDF/PPG-Si-PEG∼5% membrane, was up to 99.4%. This study provided a new method for antifouling modification of GDM.

Design of Janus-like PMMA-PEG-FA grafted fluorescent carbon dots and their nanoassemblies for leakage-free tumor theranostic application

Mingliang Pei,Xu Jia,Peng Liu

Theranostic nanomedicines for imaging guided therapy, simultaneously monitoring the real-time response of therapy, have aroused great interest in cancer research. Here, the Janus-like poly(methyl methacrylate)- b -poly(ethylene glycol)-folic acid block -copolymer-grafted fluorescent carbon dots (CDs-PMMA-PEG-FA) were synthesized via surface-initiated atom transfer radical polymerization (SI-ATRP) of methyl methacrylate (MMA) in ethanol with the modified carbon dots (CDs-Br) as initiator, followed by the PEGylation via click chemistry. Interestingly, they could self-assemble into blank nanoassemblies with hydrodynamic diameter of about 150 nm and Doxorubicin (DOX)-loaded nanoassemblies of about 270 nm with high DOX–loading capacity of 67.2%. The DOX-loaded nanoassemblies could release >80% of DOX at pH 5.0 within 36 h, while that at pH 7.4 was only 2%, demonstrating the excellent on-off behavior on pH stimuli. A possible mechanism was proposed to explore the drug release pathway in vitro. Enhanced anti-cancer efficacy of the CDs-PMMA-PEG-FA/DOX nanoassemblies with folate receptor (FR)-mediated targeting function than the free DOX was revealed by the MTT assays, while the bare CDs-PMMA-PEG-FA nanoassemblies possessed good cytocompatibility. The cellular uptake result showed that the proposed fluorescent theranostic nanomedicines could be effectively internalized into HepG2 cells and release DOX into the cell nuclei.

Tumor-Homing and Penetrating Peptide-Functionalized Photosensitizer-Conjugated PEG-PLA Nanoparticles for Chemo-Photodynamic Combination Therapy of Drug-Resistant Cancer

Xingye Feng,Di Jiang,Ting Kang,Jianhui Yao,Yixian Jing,Tianze Jiang,Jingxian Feng,Qianqian Zhu,Qingxiang Song,Nan Dong,Xiaoling Gao,Jun Chen

The combination of photodynamic therapy (PDT) and chemotherapy holds great potential in combating drug-resistant cancers. However, the major challenge that lies ahead is how to achieve high coloading capacity for both photosensitizer and chemo-drugs and how to gain efficient delivery of drugs to the drug-resistant tumors. In this study, we prepared a nanovehicle for codelivery of photosensitizer (pyropheophorbide-a, PPa) and chemo-drugs (paclitaxel, PTX) based on the synthesis of PPa-conjugated amphiphilic copolymer PPa-PLA-PEG-PLA-PPa. The obtained nanoparticles (PP NP) exhibited a satisfactory high drug-loading capacity for both drugs. To achieve effective tumor-targeting therapy, the surface of PP NP was decorated with a tumor-homing and penetrating peptide F3. In vitro cellular experiments showed that F3-functionalized PP NP (F3-PP NP) exhibited higher cellular association than PP NP and resulted in the strongest antiproliferation effect. In addition, compared with the unmodified nanoparticles, F3-PP NP exhibited a more preferential enrichment at the tumor site. Pharmacodynamics evaluation in vivo demonstrated that a longer survival time was achieved by the tumor-bearing mice treated with PP NP (+laser) than those treated with chemotherapy only or PDT only. Such antitumor efficacy of combination therapy was further improved following the F3 peptide functionalization. Collectively, these results suggested that targeted combination therapy may pave a promising way for the therapy of drug-resistant tumor.

PEG-functionalized ionic liquids: A class of liquid materials for highly efficient extraction of rare earth metals from aqueous solutions

Xiaojia Guo,Wenhui Yao,Yuehua Chen,Jing Fan,Yang Zhao,Jianji Wang

Recycling of rare earth metals is important for their supply risk in the future. In this work, a greener and highly efficient liquid-liquid extraction strategy is presented for rare earth metals from aqueous solution with PEG-functionalized ionic liquids . The factors influencing the solvent extraction process such as phase volume ratio, equilibrium time, pH value of aqueous phase, alkyl chain length of the ionic liquids and temperature of the system were investigated systematically. It was found that under optimal conditions, the single-step extraction efficiency was >   96.8% for the studied 11 kinds of rare earth metal ions Sc(III), La(III), Ce(III), Y(III), Nd(III), Sm(III), Gd(III), Dy(III), Ho(III), Yb(III) and Lu(III). After the extraction, about 100% of the rare earth metals could be stripped from the ionic liquids phases by using dilute aqueous oxalic acid , and the ionic liquids would be recovered and reused in the next extraction process. In addition, the extraction mechanism was investigated in some details, and an ion pair association mechanism was suggested. Since the extraction procedure was performed without saponification , addition of extra extractant, organic diluents or salt, the approach described here has the potential to be used as a sustainable strategy for the separation and enrichment of rare earths from complex matrixes.

Enhanced solar-thermal energy storage performance of NF/Ni-Cu@rGO/PEG composite phase change materials with 3D conductive network

Yuting He,Yongjin Zou,Cuili Xiang,Fen Xu,Yongpeng Xia,Yulian Chen,Shunxiang Wang,Zhicong Hu,Sijia Hao,Lixian Sun

Shape-stabilized composite phase change materials (CPCMs) with high photothermal conversion efficiency, high thermal storage capacity, and excellent thermal conductivity hold immense potential for diverse applications and market growth. This study employed a one-step substitution reaction followed by a thermal reduction technique to successfully affix Ni and Cu nanoparticles onto reduced graphene oxide (rGO) nanosheets , which were subsequently integrated into a nickel foam (NF) matrix. This process resulted in the fabrication of an NF/Ni-Cu@rGO porous carrier, facilitating the adsorption of polyethylene glycol (PEG) and the formation of CPCMs. Introducing Ni-Cu@rGO into the NF matrix enhanced the carrier framework, enabling improved PEG loading and leakage prevention. The hybridization of Ni-Cu@rGO effectively bolstered the interaction between the carrier framework and the phase change material (PCM), resulting in enhanced phase change performance. CPCMs prepared from the NF/Ni-Cu@rGO framework exhibited superior shape stability, higher thermal storage density, and enhanced recyclability than pure PCMs. Optimized CPCMs displayed melting and crystallization enthalpies of 121.13 J·g −1 and 113.44 J·g −1 , respectively. The thermal conductivity of NF/Ni-Cu@rGO/PEG CPCMs significantly improved to 0.4780 W·m −1  K −1 , representing a remarkable 315 % increase over that of PEG alone. Furthermore, the NF/Ni-Cu@rGO/PEG composite demonstrated outstanding photothermal conversion efficiency, thermal cycling stability, and exceptional shape stability. These findings offer promising prospects for its application across various fields.

Hydrophobic Multilayered PEG@PAN/MXene/PVDF@SiO2 Composite Film with Excellent Thermal Management and Electromagnetic Interference Shielding for Electronic Devices

Jiahui Lin,Jintao Huang,Zhanhu Guo,Ben Bin Xu,Yan Cao,Juanna Ren,Hua Hou,Yongshuang Xiao,Mustafa Elashiry,Zeinhom M. El-Bahy,Yonggang Min

With the rapid development of electronic industry, it's pressing to develop multifunctional electromagnetic interference (EMI) shielding materials to ensure the stable operation of electronic devices. Herein, multilayered flexible PEG@PAN/MXene (Ti 3 C 2 T x )/PVDF@SiO 2 (PMF) composite film has been constructed from the level of microstructure design via coaxial electrospinning, coating spraying, and uniaxial electrospinning strategies. Benefiting from the effective encapsulation for PEG and high conductivity of MXene coating, PEG@PAN/MXene composite film with MXene coating loading density of 0.70 mg cm −2 exhibits high thermal energy storage density of 120.77 J g −1 and great EMI shielding performance (EMI SE of 34.409 dB and SSE of 49.086 dB cm 3 g −1 ) in X-band (8–12 GHz). Therefore, this advanced composite film can not only help electronic devices prevent the influence of electromagnetic pollution in the X-band but also play an important role in electronic device thermal management. Additionally, the deposition of nano PVDF@SiO 2 fibers (289 ± 128 nm) endowed the PMF composite film with great hydrophobic properties (water contact angle of 126.5°) to ensure the stable working of hydrophilic MXene coating, thereby breaks the limitation of humid application environments. The finding paves a new way for the development of novel multifunctional EMI shielding composite films for electronic devices.

Clickable and imageable multiblock polymer micelles with magnetically guided and PEG-switched targeting and release property for precise tumor theranosis

Jing Wei,Xiaoyu Shuai,Rui Wang,Xueling He,Yiwen Li,Mingming Ding,Jiehua Li,Hong Tan,Qiang Fu

Targeted delivery of therapeutics and diagnostics using nanotechnology holds great promise to minimize the side effects of conventional chemotherapy and enable specific and real-time detection of diseases. To realize this goal, we report a clickable and imageable nanovehicle assembled from multiblock polyurethanes (MPUs). The soft segments of the polymers are based on detachable poly(ethylene glycol) (PEG) and degradable poly(ε-caprolactone) (PCL), and the hard segments are constructed from lysine- and cystine-derivatives bearing reduction-responsive disulfide linkages and click-active alkynyl moieties, allowing for post-conjugation of targeting ligands via a click chemistry. It was found that the cleavage of PEG corona bearing a pH-sensitive benzoic-imine linkage (BPEG) could act as an on-off switch, which is capable of activating the clicked targeting ligands under extracellular acidic condition, followed by triggering the core degradation and payload release within tumor cells. In combination with superparamagnetic iron oxide nanoparticles (SPION) clustered within the micellar core, the MPUs exhibit excellent magnetic resonance imaging (MRI) contrast effects and T 2 relaxation in vitro , as well as magnetically guided MR imaging and multimodal targeting of therapeutics to tumor precisely, leading to significant inhibition of cancer with minimal side effect. This work provides a safe and versatile platform for the further development of smart theranostic systems for potential magnetically-targeted and imaging-guided personalized medicine.

Permeation enhancer decorated nanoparticles for oral delivery of insulin: manipulating the surface density of borneol and PEG for absorption barriers

Xiaoyu Yang,Jidong Zhang,Jitang Chen,Yunxuan Xie,Tianci Hu,Qin Luo,Tianhao Peng,Han Luo,Linlin Shi,Jiangling Wan,Jianxin Wang,Xiangliang Yang,Jianyong Sheng

Oral protein drugs’ delivery faces challenges due to multiple absorption barriers for macromolecules. Co-administration with permeation enhancers and encapsulation in nano-carriers are two promising strategies to enhance their oral absorption. Herein, the poly(lactic-co-glycolic acid) nanoparticles (PLGA NPs) are decorated with polyethylene glycol (PEG) and a traditional Chinese medicine-derived permeation enhancer borneol (BO) for oral insulin delivery. Compared with a physical mixture of BO and PEG-decorated PLGA NPs, PLGA–PEG–BO NPs significantly facilitate insulin permeation across intestinal epithelia through various transcytosis pathways. The relationship among the BO surface density, physico-chemical properties and multiple barriers penetration ability is further investigated. Increasing the BO density boosts penetration through the epithelial cell layer but reduces enzyme and mucus barrier penetration. When the surface PEG density is at 90% and BO density is at 10%, the NPs possess the strongest overall ability to overcome both the mucus layer barrier and epithelial cell barrier, as illustrated by the highest permeation efficiency through Caco-2/HT29-MTX cell co-cultural monolayers. In diabetic rodents, PLGA–PEG90%–BO10% NPs exhibit high intestinal safety and a substantial hypoglycemic effect, with insulin availability at 6.22 ± 2.30%, double that of orally delivered insulin PLGA–PEG NPs and far superior to a physical mixture with BO. This study reveals the importance of tailored absorption enhancer decoration for oral protein delivery.
