Near-infrared (NIR) photodynamic therapy (PDT) is a promising antitumor strategy under NIR light irradiation to kill cancer cells. Mitochondria has a critical function in sustaining cellular viability and death, which is the ideal organelle for PDT. Here, we reported a tetraphenylporphine (TPP)-conjugated amphiphilic copolymer and an iodinated boron dipyrromethene photosensitizer (BDPI) with high singlet oxygen yield to form nanoparticles (PBDPI-TPP), which could realize mitochondria-targeting and improve the NIR imaging-guided PDT. The as-prepared mitochondria-targeting nanoplatform could show effective subcellular localization and bring about significant irreversible mitochondrial injury for enhanced PDT. Both in vitro and in vivo experiments revealed that the mitochondria-targeting PDT system could achieve a remarkable therapeutic effect, indicating that it is a promising nanoplatform for NIR imaging-guided PDT in cancer therapeutics.

A novel pH-sensitive carborane and cyanine dye conjugated polypeptide and its micelles has been designed and synthesized for potential near infrared fluorescence imaging-guided boron neutron capture therapy (BNCT). The amphiphilic polymer we synthesized with tertiary amine as functional group could self-assemble to micelle nanoparticles at neutral pH, and responsive to acidic solution for the disassembly of the micelles for drug release. The tertiary amine sensitive induced process was observed via dynamic light scattering (DLS) and transmission electron microscopy (TEM). The efficiency of cell killing ability is studied by MTT assays and fluorescence microscope measurement, indicating a new intelligent system for potential NIR imaging-guided therapy.

pH-Sensitive doxorubicin conjugated polymeric micelles entrapped with near infrared (NIR) photosensitizer BODIPY (which works as an imaging agent at the same time) were designed and synthesized by ring opening polymerization of N-carboxyanhydride with mPEG-NH2 as the initiator, following reaction with doxorubicin to form the hydrazone-bond linker for pH responsiveness. Then the NIR dye (BODIPY) was loaded in the micelles for both bioimaging and photodynamic therapy (PDT). A significant cytotoxicity of NIR imaging-guided combined PDT and chemotherapy could be found by MTT assays, which was also confirmed with a fluorescence microscope, indicating a new kind of polymeric nanoparticle for potential theranostic treatment of cancers. In addition, the energy density of the laser for the PDT is extremely low.

Graphical The nanoscale zeolitic imidazole framework-8 (ZIF-8) is adopted to co-assemble a temperature-sensitive free radical initiator (AIPH) and a photothermal molecule (ICG) to realize the NIR-triggered tandem photothermal and oxygen-independent photodynamic therapy. This as-prepared ICG/AIPH@ZIF-8 nanoplatform displays an excellent killing effect of cancer cells in both normoxia and hypoxia. Due to the limited penetration depth of visible light and insufficient oxygen supply in hypoxic tumor tissues, the development of near-infrared (NIR) triggered and oxygen-independent photodynamic therapy (PDT) to generate active radicals is necessary for cancer treatment. Herein, we developed a NIR-triggered photothermal therapy (PTT) and oxygen-independent PDT system based on metal-organic frameworks (MOFs), which integrated indocyanine green (ICG) and 2,2’-azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH) as NIR-triggered photothermal units and temperature-sensitive free radical initiator into ZIF-8, respectively. AIPH was introduced into ZIF-8 via a one-pot strategy with high loading capacity (125 mg g −1 ). Through the irradiation of 808 nm laser, the ICG units could efficiently absorb NIR light to rapidly raise the local temperature to 44 °C within 3 minutes. Meanwhile, the local high temperature further induced the AIPH to continuously generate toxic alkyl radicals (R−C⋅). The combination of PTT and free radical therapy is significantly better than the single treatment mode. More importantly, the NIR-triggered synergistic therapy could induce extensive apoptosis of cancer cells in hypoxic conditions, which sheds new light on the development of MOFs-based photochemical therapy platform in the actual tumor microenvironment.

Background Macrophages with tumor-tropic migratory properties can serve as a cellular carrier to enhance the efficacy of anti neoplastic agents. However, limited drug loading (DL) and insufficient drug release at the tumor site remain the main obstacles in developing macrophage-based delivery systems. In this study, we constructed a biomimetic delivery system (BDS) by loading doxorubicin (DOX)-loaded reduced graphene oxide (rGO) into a mouse macrophage-like cell line (RAW264.7), hoping that the newly constructed BDS could perfectly combine the tumor-tropic ability of macrophages and the photothermal property of rGO. Results At the same DOX concentration, the macrophages could absorb more DOX/PEG-BPEI-rGO than free DOX. The tumor-tropic capacity of RAW264.7 cells towards RM-1 mouse prostate cancer cells did not undergo significant change after drug loading in vitro and in vivo. PEG-BPEI-rGO encapsulated in the macrophages could effectively convert the absorbed near-infrared light into heat energy, causing rapid release of DOX. The BDS showed excellent anti-tumor efficacy in vivo. Conclusions The BDS that we developed in this study had the following characteristic features: active targeting of tumor cells, stimuli-release triggered by near-infrared laser (NIR), and effective combination of chemotherapy and photothermotherapy. Using the photothermal effect produced by PEG-BPEI-rGO and DOX released from the macrophages upon NIR irradiation, MAs-DOX/PEG-BPEI-rGO exhibited a significant inhibitory effect on tumor growth.

Halide double perovskites with near-infrared (NIR) emission are promising for optoelectronic applications. NIR-II (1000–1700 nm) emission, in particular, is attractive due to its strong tissue penetration, high spatial resolution, and low biological light damage risk. However, materials capable of NIR-II emission often require additional sensitizers and suffer from issues such as narrow emission bandwidth and low photoluminescence efficiency. In this work, we report a Re 4+ doping strategy using Cs 2 WCl 6 , a vacancy–ordered double perovskite, to achieve efficient NIR-II emission. Spectroscopic and dynamic measurements reveal energy transfer between the Cs 2 WCl 6 matrix and the Re 4+ centers, resulting in efficient broadband NIR-II emission centered at 1345 nm (FWHM ≈ 87 nm), along with broad excitation ranging from 250 to 850 nm. The optimal NIR-II emission occurs at 1345 nm with a photoluminescence quantum yield (PLQY) of 29.83% when the Re 4+ doping concentration is 1%. This work demonstrates an efficient, sensitizer-free method for achieving broadband NIR-II emission and provides a new material strategy for high–performance double perovskites NIR light sources.

Molecular imaging of tumor biomarkers represents a crucial direction for advancing precision cancer diagnosis and therapy. As a core regulator of tumor angiogenesis, VEGFR2 represents an ideal molecular imaging target. Herein, a novel VEGFR2-targeting peptide (HTMYYHHYQHHLSSSDICLPRWGCLWED, named as VTP) was designed and synthesized by conjugating a peptide K237 (HTMYYHHYQHHL), a flexible tripeptide linker (Ser-Ser-Ser, SSS), and an albumin-binding peptide (ABP, DICLPRWGCLWED). The surface plasmon resonance (SPR) analysis confirmed that VTP had high binding affinity for VEGFR2 ( K D = 15.4 nM). Although indocyanine green (ICG) has been widely used in clinical practice, its short half-life and lack of active targeting ability significantly restrict its potential for intraoperative navigation. In this study, the VEGFR2-targeting peptide VTP was conjugated with ICG to successfully construct a targeted fluorescent probe, ICG-VTP. Owing to ABP modification, this probe exhibits a longer blood half-life and shows accumulation characteristics that are highly positively correlated with VEGFR2 expression levels in multiple tumor models, including ovarian cancer, colorectal cancer, and non-small cell lung cancer. The probe yielded tumor-to-background ratios (TBR) of 2.95, 2.05, and 1.42 in high (SKOV3), moderate (HCT116), and low (A549) VEGFR2-expressing tumors at 10 h post-injection, respectively, confirming its specificity for VEGFR2 imaging. Further, the ICG-VTP probe enabled successful intraoperative fluorescence navigation and precise tumor resection. Safety assessments revealed no significant toxicity at cellular or tissue levels. In summary, the ICG-VTP probe demonstrates strong specificity and superior imaging contrast of VEGFR2-positive tumors, underscoring its high potential to advance biomedical research and clinical translation.

Peroxynitrite anion (ONOO−), a product derived from reaction between reactive oxygen species (ROS) and nitric oxide (NO), is considered to be a more toxic reactive species than most ROS for cancer photodynamic therapy (PDT). To promote the PDT effect, a viable method is to develop rational strategies for efficient ONOO− generation at targeted tumor sites. Herein, a heterostructure nanocomposite containing ZnO-coated lanthanide nanoparticles (LnNPs) is reported for ONOO−-based PDT. In this nanocomposite, Nd3+-doped LnNPs are employed to realize efficient NIR-light-triggered ROS generation by activating the triplet state of chlorin-e6 (Ce6) photosensitizers via a direct lanthanide-to-triplet sensitization mechanism. Meanwhile, ZnO in the composite catalyzes the decomposition of S-nitrosoglutathione (GSNO) to generate NO in the tumor microenvironment. The coupled system allows the combination of photo-induced ROS and NO to produce ONOO−, leading to drastically promoted cancer cell apoptosis and tumor growth inhibition. This study establishes a new apoptosis-inducing PDT agent, which is potentially active in drug resistant malignancies.

As a new generation of near-infrared (NIR) light source, phosphor-converted NIR light emitting diodes (NIR pc-LEDs) have broad application prospects in biological tissue imaging, night vision and chemical composition analysis . However, most NIR emitting phosphors are still limited to short-wavelength, narrow-band emission and low efficiency. Here, based on the multi-site occupation strategy, the NIR phosphor Mg 2 LaTaO 6 :Cr 3+ (MLT:Cr 3+ ) achieves an efficient broadband NIR emission with the full width at half maximum (FWHM) of 203 nm in the range of 600–1300 nm under the excitation of 468 nm blue light. The phosphor MLT:0.01Cr 3+ not only exhibits an excellent internal quantum efficiency (IQE) of 85.6 %, but also has good thermal stability and remains about 64.2 % photoluminescence (PL) intensity of room temperature at 423 K. Furthermore, the MLT:0.01Cr 3+ phosphor was combined with a blue LED chip to fabricate the NIR pc-LED device, and its potential applications were successfully realized in information encryption, bioimaging and the qualitative or quantitative analysis of mixture of ethanol and water.

Phototherapy, including photothermal and photodynamic therapy, has gained extensive attention in the tumor treatment field recently, while synergistic therapy can significantly improve curative effects. However, a complicated photo-responsive nanosystem, different excitation wavelengths, and low tissue depth hindered its actual application. Herein, single near-infrared responsive PEGylated defective MoO2 nanocrystals were fabricated by a green hydrothermal method. The photothermal and photodynamic performances of the samples were presented in detail under a safe power of 1064 nm (NIR-II, 1.0 W cm−2). Interestingly, the photodynamic properties were prompted by the localized surface plasmon resonance (LSPR) photothermal effect obviously, and the collaborative enhancement mechanism was explored in depth. Subsequently, the in vitro cytotoxicity was evaluated on the 4T1 cancer cells under NIR-II irradiation. This work may provide guidance for the facile fabrication of TMOs for NIR-II responsive and enhanced dual-modal phototherapy.

Integration of mechanical and actuation properties with sensory feedback function in soft actuators remains a challenge. Herein, a highly stretchable and electrically conductive hydrogel with fast stimuli-responsive actuating performance is prepared by in situ growth of polyaniline (PANI) nanoparticles in the Pluronic F127 diacrylate (F127DA) cross-linked poly(N-isopropyl acrylamide) (PNIPAM) hydrogel matrix. The obtained PNIPAM/PANI hydrogel exhibits high stretchability (730%) and fast deswelling properties upon heating. Taking advantage of highly efficient photothermal conversion of the embedding PANI, this hydrogel shows a significant temperature rise of ∼50 °C within 150 s and large volume shrinkage of 65% upon near-infrared (NIR) light irradiation. Accordingly, the hydrogel is explored as an underwater actuator, which has achieved programmable deformations to simulate hand-like gestures and Venus flytrap-type locomotion under local NIR irradiation. In addition, the hydrogel is able to translate the local strain into electrical signals. The light-triggered actuation, including bending, weight lifting and moving objects can be monitored by real-time resistance change. These results confirm that actuating and real-time sensory feedback could be integrated in a single homogeneous material, which make it an outstanding candidate in soft actuators and even intelligent biomimetic devices.

Efficient hemorrhage control of severe wound injuries is an urgent medical need, deserving agents with promising blood coagulation and biocompatible characteristics. Current work developed polydopamine (PDA) functionalized porous starch powder (PS-PDA) for emergency bleeding treatment. The micro-morphology and elements, chemical groups, and porosity of PS-PDA were systematically characterized. Its comparison with porous starch (PS) revealed the promising potential of this composite in medical practice. On one hand, PS-PDA showed superior surface area and biomineralization affinity over PS, along with comparable hemo/cyto-compatibility. On the other hand, the photothermal effect of PDA under near Infrared (NIR) light paved the possibility to accelerate blood coagulation in situ. In vivo studies indicated PS-PDA can significantly reduce blood loss and improvement of hemostasis efficiency accompanied by NIR light exposure. These results suggest that this newly developed PS-PDA powder can serve as a promising hemostatic material for bleeding wound control.
