The extraction, characterization and antioxidant activity of polysaccharides fromChoerospondias axillarisleaves were investigated in the present study. Two purified polysaccharide fractions, CALP-1 and CALP-2, were isolated from crudeChoerospondias axillarisleaf polysaccharides (CALP) by DEAE-52 cellulose chromatography and Sephadex G-100 column chromatography. The characteristics of CAL-1 and CALP-2 were determined by using High-performance Gel Permeation Chromatography (HPGPC), High-Performance Anion-Exchange Chromatography, HPAEC (HPAEC-PAD) and Fourier transform infrared spectroscopy (FTIR). CALP-1 with molecular weight of 11.20 KDa was comprised of Rhamnose, Arabinose, Galactose, Glucose, Xylose, Mannose and galacturonic acid in a molar ratio of 5.16:2.31:5.50:27.18:1.00:0.76:1.07. CAL-2 with molecular weight of 8.03 KDa consisted of Rhamnose, Arabinose, Galactose, Glucose, and galacturonic acid at a ratio of 1.38:3.63:18.84:8.28:1.45. FTIR revealed that CALP-1 and CALP-2 were acidic polysaccharides. The antioxidant activity of crude CALP, CALP-1 and CALP-2 was evaluated in vitro. The fraction CALP-2 was demonstrated to be of polysaccharide nature containing a large percentage of Galactose but no Xylose and Mannose. The antioxidant activity assays showed that CALP-1 and CALP-2 exhibited antioxidant and scavenging activities on hydroxyl and DPPH radicals in vitro. Compared with pure polysaccharide, crude CALP exhibited stronger anti-oxidant activities. These results will provide a better understanding ofChoerospondiasaxillaris leaf polysaccharide and promote the potential applications ofChoerospondiasaxillaris leaf polysaccharide in the pharmacological field and as a natural antioxidant. Keywords:polysaccharide;Choerospondias axillarisleaves;purification;characterization;antioxidant

The complex structure of rice straw is such that its bioconversion requires multiple physical and chemical pretreatment steps. In this study, it was found that a large amount of soluble polysaccharides (SPs) are formed during the pretreatment of straw. The yield of NaOH-based SPs (4.8%) was much larger than that of ball-milled SPs (1.5%) and H 2 SO 4 -based SPs (1.1%). For all the pretreatments, the ratio of phenolic compounds to saccharides (P/S) for each type of SPs increased upon increasing the concentration of ethanol in the order of 90% > 70% > 50%. The yield of NaOH-based SPs was much higher than that of acid-based and ball-milled SPs. The changes in the 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS), ferric reducing antioxidant power assay (FRAP), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) of SPs follow the same rule, i.e., the higher the P/S ratio, the higher the antioxidant values of the SPs. The flow cytometry and laser scanning microscopy results show that the P/S ratio can significantly influence the effect of SPs on microbial growth and cell membrane permeability. Upon varying the ethanol concentration in the range of 50–90%, the P/S ratio increased from 0.02 to 0.17, resulting in an increase in the promoting effects of the SPs on yeast cell growth. Furthermore, H 2 O 2 , NAD + /NADH, and NADP + /NADPH assays indicate that SPs with a high P/S ratio can reduce intracellular H 2 O 2 and change the intracellular redox status.

An edible film based on soluble soybean polysaccharide (SSPS) and gelatin incorporating curcumin was fabricated as active packaging material for soybean oil . The effect of curcumin (Cur) on structural and physical properties of SSPS/gelatin films were investigated. Scanning electron microscopy revealed that curcumin was evenly distributed in the SSPS/gelatin matrix. Hydrogen bonding interactions between SSPS/gelatin and curcumin were unveiled by Fourier-transform infrared spectroscopy. The addition of curcumin (0 −0.20 %) significantly increased the tensile strength , temperature of maximum mass loss rate, yellow index, transparency value and water contact angle from 2.3 to 5.5 MPa, 249–252 °C, 2.04–183.07, 0.51–1.63, and 70.7–98.6°, respectively. The DPPH and ABTS radical scavenging activities of the SSPS/gelatin film containing 0.20% curcumin were 42.6 % and 99.6 %, respectively, which were remarkably higher than those of neat SSPS/gelatin film. The SSPS/gelatin/Cur composite films showed antibacterial activity against Escherichia coli and Staphylococcus aureus . The heat-sealed SSPS/gelatin/Cur pouch of soybean oil showed quick-dissolving property upon contact with hot water. Moreover, peroxide value results indicated that the SSPS/gelatin/Cur pouch could effectively delay the soybean oil oxidation under accelerated storage conditions. Therefore, the SSPS/gelatin/Cur composite films may be served as active packaging materials for edible oils .

Antarctic krill is a crucial marine resource containing plenty of high-valued nutrients. However, krill oil as a single product has been developed by the current solvent extraction with high cost. From the perspective of comprehensive utilization of Antarctic krill, this study proposed a novel two-step enzymolysis-assisted extraction in attempt to produce value-added oil and enzymolysate simultaneously. After two-step chitinase/protease hydrolysis, the lipid yield increased from 2.09% to 4.18%, reaching 112% of Soxhlet extraction. The method greatly improved the yields of main components while reducing the impurity content without further refining. After optimization, the oil contained 246.05 mg/g of phospholipid, 80.96 mg/g of free eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), and 0.82 mg/g of astaxanthin. The by-product enzymolysate was abundant in water-soluble proteins (34.35 mg/g), oligopeptides (13.92 mg/g), amino acids (34.24 mg/g), and carbohydrates (5.79 mg/g), which was a good source of functional nutrients. In addition, both oil and enzymolysate showed high antioxidant capacity. This novel method could simultaneously provide oil and enzymolysate amounting for 58.61% of dried krill. Keywords:Antarctic krill oil;enzymolysis;protease;chitinase;extraction

Introduction: Sea buckthorn (Hippophae rhamnoides) seed oil is rich in unsaturated fatty acids, and is thus susceptible to oxidation and rancidity. Microencapsulation technology allows the effective protection of active substances, thereby prolonging the deterioration time and shelf life. Methods: In this study, H. rhamnoides microcapsules were prepared using a spray-drying method, and the microencapsulation parameters were optimized. The morphological characteristics, structural parameters, and stability of the microcapsules were determined using scanning electron microscopy, Fourier transform infrared spectroscopy, thermogravimetric analysis, differential scanning calorimetry, and oil oxidation stability testing. Results: Based on encapsulation efficiency (EE, %) and the particle size (D50) of the microcapsules, the optimal preparation conditions were characterized as a wall material consisting of soy protein isolate and soybean polysaccharide (2:3), a wall concentration of 15%, a core-to-wall ratio of 1:3, and an inlet temperature of 160°C. Under these optimal conditions, the encapsulation efficiency was 95.30 ± 2.67%, with a yield of 57.03 ± 3.71% and a particle size of 7.96 ± 1.04 μm.Discussion: Furthermore, the effectiveness of microencapsulation in protecting the biological activity of H. rhamnoides seed oil was confirmed by an antioxidation test. Thus, the results of this study showcase the successful microencapsulation of H. rhamnoides seed oil, thereby significantly improving its stability.

In this paper, Lepista sordida polysaccharides (LSP) were separated from Lepista sordida (L. sordida) mainly using the Ultrasonic-Micro Wave Synergy Extraction (UMSE) method and purified by graded alcohol precipitation. Three polysaccharide components: 40%-LSP-UMSE, 60%-LSP-UMSE, and 80%-LSP-UMSE were obtained and further analyzed the physicochemical properties, structural characteristics, and antioxidant activity. And the effects on the proliferation of Lactobacillus casei of three polysaccharide components were studied. The characteristic absorption peaks and the β-glycosidic bond of three polysaccharide components were the direct expression at UV 200 nm using UV and FT-IR spectroscopy. The three polysaccharide components were mainly composed of glucose, mannose, galactose, and ribose using high-performance liquid chromatography (HPLC) analysis. The antioxidant activity study revealed that the polysaccharides obtained by the UMSE method had better antioxidant activity compared to the traditional “Hot Water Extraction (HWE)” method. In addition, the polysaccharide components promoted the proliferation of L. casei to some extent. 40%-LSP-UMSE, 80%-LSP-UMSE as the carbon source had better acid production than the control inulin. Three LSP-UMSE used as a carbon source compared with glucose for culturing L. casei could significantly improve its tolerance to bile salts. Results are helpful to develop the bioactive polysaccharides from Lepista sordida and beneficial to develop a unique health and functional product in the future.

Industrial hemp is an important economic crop. The extract of its leaves is well-known as cannabidiol full-spectrum oil (CFSO) and has great application potential. But the chemical composition of CFSO is still not clear, which limits its efficient application. Therefore, this work aimed to explore the phytochemical profile and composition-activity relationship of CFSO. The results showed that a total of 12 cannabinoids were identified by HPLC-ESI-MS and they were CBD-C 1 , CBGVA, CBGA, CBGV, CBDVA, CBG, CBDV, CBDA, CBD, CBD-C 4 , CBN, and CBT. CBD (559.3 mg/g) was the most abundant component, followed by CBDV (302.4 mg/g) and CBN (31.7 mg/g). Besides, the content of CBDV, CBD, and CBN of different polar extracts increased with the rise of ethanol concentration. The TPC and TFC of the extracts also increased with the rise of ethanol concentration. The extract of 90 % EtOH had the highest TPC (949.21 ± 2.37 mg GAE/g extract) and TFC (267.05 ± 1.21 mg RE/g extract) while the extract of 0 % EtOH displayed the lowest TPC (244.89 ± 2.13 mg GAE/g extract) and TFC (46.26 ± 0.85 mg RE/g extract). Furthermore, as ethanol concentration increased, the cytotoxicity, antioxidant, and antibacterial activities of the extracts had a significant increasing trend. And the correlation analysis demonstrated that the antioxidant activity and cytotoxicity of the extracts were closely related with the phenolic compounds and flavonoids, especially cannabinoids. These findings could provide data support for the application of CFSO.

The pathological aggregation of some proteins is claimed to be highly related to several human diseases, such as β-amyloid 1–42 (Aβ 42 ) to Alzheimer's disease (AD), islet amyloid polypeptide, and insulin to type 2 diabetes mellitus. Therefore, it is in desperate need to develop effective methods for detection of protein aggregates and inhibition of abnormal aggregation. Herein, to construct all-in-one probe with both diagnosis and treatment potentials for protein aggregation diseases, Congo red (CR), a classical staining reagent with red fluorescence signal output for protein aggregates, is deliberately adopted to react with three different reductive carbon sources and ammonium persulfate to generate three CR-derived carbon dots (CDs). The obtained CDs exhibit the capabilities of turn-on red fluorescence imaging of protein aggregates, and/or inhibition of protein aggregation as well as scavenging of free radicals. Among them, CA-CDs, using citric acid as the reductive carbon source, demonstrate the superiority to the other two studied CDs in integrating all of these functions, and particularly exert excellent cytoprotection effect against toxic Aβ 42 species, possessing tremendous potential in diagnosis and treatment of AD for future study. The present study paves a new way to develop all-in-one CDs for the protein disease research.

Castanopsis is diffusely spread in tropical and subtropical regions and is an important nectar source plant in China. The Castanopsis honey (CH) is characterized by its bitter taste. However, its composition and functions remain unclear. In this study, the physicochemical parameters, chemical composition, and antioxidant capacity of CH were comprehensively investigated, with the anti-inflammatory effects of the Castanopsis honey extract (CHE) evaluated based on the RAW 264.7 cell inflammatory model. The results revealed a high level of quality in CH based on the quality standards. Among a total of 84 compounds identified in CH, 5 high response compounds and 29 phenols were further quantified by UPLC-Q/TOF-MS. The high content of phenylethylamine (117.58 ± 64.81 mg kg−1) was identified as a potential marker of CH. Furthermore, the CH showed evident antioxidant activities, and the anti-inflammatory activities of CHE were observed to inhibit the release of nitric oxide (NO) and reduce the content of tumor necrosis factor alpha (TNF-α) and improve the content of interleukin-10 (IL-10) by regulating the NF-κB pathway. Our study indicates that CH has sound physicochemical properties and biological activities with a high level of quality, providing strong experimental evidence to support the further economic and agricultural development and application of CH.

Selenium-enriched peptide (SP) is a new type of selenium supplement with unique bioactivities; however, its instability jeopardizes its industrial applications. Hence, a system stabilizing and delivering SP is a prerequisite to overcome this defect. This study developed a microcapsule, dextran–whey protein isolate–selenium-enriched peptide (DX–WPI–SP), encapsulating SP within the WPI–DX glycation product via electrospraying technology, and its properties were characterized. Results showed that DX–WPI–SP considerably surpassed the physical mixture of WPI, DX, and SP, with respect to antioxidant capacity, thermal stability, and cell cytotoxicity. The in vitro gastrointestinal digestion results showed that over 42% of the SP in DX–WPI–SP survived the gastric fluids and was then released in the intestinal phase. The stability of SP in long-term storage was also observed after its encapsulation in DX–WPI–SP, signifying its stability in this delivery system. This study demonstrated that the DX–WPI–SP microcapsule is a valid strategy to stabilize SP in intestinal delivery.

Ethnopharmacological relevance Syringa Pubescens Turcz. (SP), a member of the Oleaceae family, is a species of plant known as Syringa . Flowers, as the medicinal part, are commonly used in the treatment of hepatitis and tonsillitis. Aim of the study: The research was the first to assess the antioxidant and anti-inflammatory potential of different parts of SP flowers (SPF) in vitro . The most promising fraction was ethyl acetate fraction of SP flower (SPFEA). The antioxidant, anti-inflammatory and analgesic activities of SPFEA were further studied, and the chemical components were identified. Methods HPLC was used to identify the major components in various fraction of SPF. DPPH and ABTS + radical scavenging assays as well as FRAP test and β-carotene bleaching test were employed to assess the antioxidant potential of SPF fraction in vitro . The inhibitory effect on NO production in LPS-treated RAW264.7 cells and heat-induced protein denaturation test were used to evaluate the anti-inflammatory potential of SPF fraction. Further analysis of the biological activity of SPFEA was performed. Acute toxicity test was conducted to assess the toxicity of SPFEA. The anti-inflammatory effect was assessed by utilizing xylene induced ear edema model, carrageenan-induced foot edema model and peritonitis model in vivo . The analgesic effect of SPFEA was evaluated using hot plate test, tail immersion test, formaldehyde test as well as acetic acid-induced abdominal writhing pain experiment in vivo . In carrageenan induced foot edema model, ELISA kits were employed to measure levels of inflammation factors (NO, TNF-α, IL-6, COX-2, IL-1β) in foot tissue as well as MDA, CAT, SOD, GSH-PX levels in liver tissue. Results HPLC results showed that there were significant differences in bioactive substances among different fractions of SPF, and SPFEA was rich in bioacitve components. Compared with other fractions of SPF, SPFEA exhibited better antioxidant and anti-inflammatory abilities. The 3000 mg/kg SPFEA group in mice had no obvious side effects. The xylene-induced ear edema model, carrageenan-induced foot edema and peritonitis models demonstrated that the SPFEA had significant anti-inflammatory effect. Moreover, inflammation factors including NO, TNF-α, IL-6, COX-2, IL-1β were significantly reduced in SPFEA groups in foot tissue induced by carrageenan. Additionally, SPFEA effectively decreased liver tissue oxidative stress levels (MDA, SOD, GSH-PX and CAT). The bioactivities of SPFEA demonstrated a clear dose-dependent relationship. The results of the hot plate test, tail immersion test, formaldehyde test and acetic acid-induced abdominal writhing pain experiments indicated the SPFEA possessed an excellent analgesic effect, and this effect was in dose-dependent manner. Conclusion The study provides a scientific foundation for understanding the pharmacological action of SPFEA. It has been indicated that SPFEA has excellent antioxidant, analgesic and anti-inflammatory effects.

To intensively investigate the antioxidant property of flavonoids and their different phenolic hydroxyl groups, we selected six types of typical flavonoids and carried out experimental analysis and quantum chemical calculations. By calculating the Mulliken charges, frontier molecular orbitals, molecular dipole moments, and interaction model hydrogen bond lengths of the compounds, the theoretical analysis results were compared with the free radical scavenging assays. Results show that the three flavonoids with different numbers of hydroxyl groups on the B ring are ranked in antioxidant activity as myricetin > quercetin > kaempferol. The glycoside linked in the C ring has a great effect on bioavailability, and the antioxidant activity is ranked as quercetin > isoquercetin > rutin. Besides, the results of the novel interaction model calculations show that quercetin with an meta -hydroxy group on the B ring structure has stronger antioxidant capacity than morin with an ortho -hydroxy group. The theoretical evaluation method established in this work is expected to provide new ideas and references for the evaluation of antioxidant properties of natural active substances from the electronic structure perspective.
