Background The heavy metal ion Cd 2+ is acutely toxic, and excessive concentrations can have adverse effects on human production and life, and even lead to significant public health risks and environmental impacts. There are several mature non-electrochemical methods for heavy metal detection, but these methods are characterized by high cost, which makes it difficult to be applied to the field for timely detection. Therefore, it is necessary to prepare a new electrochemical sensor that is environmentally friendly and capable of detecting Cd 2+ in the environment quickly, easily and sensitively. Results In this study, hydrogen-bonded organic frameworks (HOFs) were synthesized by a simple hydrothermal reaction. The prepared materials consisted of only C, N and O and had a thin lamellar structure. The HOFs were integrated into a novel electrochemical sensor to achieve accurate detection of Cd 2+ ions in real aqueous environments by square wave anodic dissolution voltammetry. The sensor has a wide linear range and a detection limit as low as 0.13 μg/L. Several real water samples, such as tap water, lake water, and e-cigarette digest, were analyzed to simulate the working environment of the sensor, and the results showed that the recoveries of Cd 2+ ranged from 95.75 % to 101.2 %. Significance We pioneered the detection of heavy metal ions Cd 2+ in e-cigarette digestate samples with the innovative use of HOFs as the sensor material, which demonstrated the potential application in electrochemical sensing with extremely low background current value and high sensitivity, providing new ideas for environmental monitoring and public health control.

Enzymatic PET recycling has emerged as a promising green solution in addition to mechanical recycling, but low soluble expression levels of the inherently hydrophobic PET hydrolases hinder large-scale applications. Here, we propose a novel strategy for enhanced production of FastPETase in Escherichia coli using co-expression of molecular chaperones from Ideonella sakaiensis . Co-expression of cognate DnaK and DnaJ chaperones significantly increased soluble FastPETase expression (up to 2.5-fold), surpassing commercial chaperone plasmids. Furthermore, a combinatorial approach employing co-expression of DnaK/DnaJ chaperones and fusion of FastPETase with the VNp6-tag significantly boosted FastPETase secretion, yielding over 2 g/L of target protein in a 5-l bioreactor. Notably, the crude FastPETase in fermentation broth displayed comparable PET hydrolysis effects to the purified enzyme. This work not only provides new insights into the process of chaperones in protein folding but also suggests a novel and efficient strategy for producing recombinant proteins.

Food processing environments contaminated with E. coli O157 :H7 and forming biofilms will be a continuous source of contamination, leading to foodborne illness and food spoilage. Now, the main countermeasure is chemical disinfectants, but the use of disinfectants may cause the development of drug-resistant bacteria and other negative effects. Therefore, there is an urgent need to find novel antimicrobial agents. Quorum-sensing inhibitors (QSI) are a promising strategy to control biofilm formation. This study aimed to screen the optimal QSI of E. coli O157 :H7 from plant-derived small molecules by biofilm and QS inhibitory activities and to validate their effectiveness in combination with disinfectants. In addition, the inhibition mechanism of gallic acid on biofilm formation was further explored. Gallic acid was screened as the QSI of E. coli O157 :H7, and its AI-2 inhibitory activity against LuxS/AI-2 QS system reached 95.54% and 98.89% after treatment with 1/2 MIC gallic acid for 4 h and 6 h, respectively. It inhibited biofilm formation by affecting flagellar assembly, chemotaxis, and expression of adhesion factor-related genes, which interfered with the transition of bacteria from reversible to irreversible adhesion. In beef broth simulation system, the inhibition rate was greater than 90.00% in all gallic acid and benzalkonium chloride combination groups. Therefore, gallic acid can be a potential antimicrobial synergist in the food industry, combined with disinfectants to control E. coli O157 :H7 biofilms while avoiding the development of resistance.

Cyst(e)inase, an engineered human cystathionine-γ-lyase (hCGL), effectively degrades l-Cys and CSSC, inhibiting various tumors in vivo with minimal toxicity. However, its current activity and stability limit its clinical application. To enhance its therapeutic potential, we utilized folding free energy calculations and amino acid site conservation to identify 46 potential mutants. Among them, 17 mutants exhibited improved activity or stability, while six showed a loss of function. Notably, mutant L142E enhanced activity for both substrates, and E349S displayed 2.8 times higher activity toward CSSC and a 12.8 °C increase in Tm, despite reduced activity toward l-Cys. Saturation mutagenesis highlighted the importance of spatial constraints for substrate selectivity. MD simulations suggested that distal mutations stabilize the near-attack conformation, enhancing the activity. In vitro cytotoxicity assays confirmed that the mutants L142E and E349S significantly inhibited gastric and pancreatic cancer cells. This study provides valuable insights for improving therapeutic enzyme design.

It is a high demand but a great challenge to construct all-in-one ionic conductive hydrogels with excellent mechanical performances, transparency, self-adhesion, conductivity and environmental tolerance. Herein, a versatile gel platform was developed via a facile and rapid photopolymerization of acrylic acid (AA) in deep eutectic solvent (DES) with multiple roles simultaneously as water-free solvent, ionic conductor , regulator of hydrogen bond density. Benefiting from the formed crosslinking networks including covalently cross-linked polymer network and multiple dynamic interactions, the resultant PAA-DES gels exhibited high stretchability of 884.7 ± 4.9%, toughness of 31.2 ± 1.4 MJ/m 3 , fatigue resistance and recovery ability, transparency of 93%, self-adhesive strength of 472 kPa. Moreover, the inherent advantages of DES enabled the gels with good ionic conductivity (2.5 ± 0.2 mS/cm), wide working window and biocompatibility . The PAA-DES as self-adhesive ionic sensors not only possessed wide strain range (∼400%) and stability, but also displayed excellent humidity (30–80%), temperature (−40–60 °C), and sweat tolerance which could precisely and steadily discern and monitor human motions and weak physiological signals. Thus, this work not only provides a strategy to construct all-in-one ionic conductive gels with high elasticity, fatigue resilience, transparency, self-adhesion and wide environmental tolerance, but also exploits versatile platform for intelligent gel sensor and e-skin.

Effects of braising duration on volatile organic compounds (VOCs) and lipids in chicken were investigated. Aroma profiles identified by an electronic nose were effective in differentiating braising stages. During braising process, a total of 25 key VOCs were detected in braised chicken, and sample braised for 210 min exhibited the highest level of key VOCs. Additionally, a gas chromatography mass spectrometry fingerprint was established to evaluate the distribution of VOCs throughout the braising process. Partial least square discriminant analysis indicated that 2-heptanone, 3-methyl-2-butanone, octanal, nonanal, butanal, (E)-2-pentenal, 1-octen-3-ol, 1-hexanol, pentanal, hexanal, and 1-pentanol significantly affected flavor characteristics of braised chicken. Furthermore, 88 differential lipids were screened, and glycerolipids metabolic was found to be main metabolic pathway during braising process. Triglycerides (TG) and phosphatidyl ethanolamine (PE), such as TG (16:0/18:1/18:2), TG (18:0/18:1/18:2), TG (18:1/18:2/18:3), TG (18:1/18:1/18:2), PE (O-18:2/18:2), PE(O-18:2/18:1), and TG (16:0/16:1/18:2), played a vital role in the generation of VOCs.

The sustainable recovery of gold from electronic waste (E-waste) remains a significant challenge due to lack of multiple redox active-sites in the existing adsorbents, their high cost, and poor stability in extreme acidic conditions. Herein, we rationally designed the skeleton engineering of poly(organo)phosphazene via substitution reaction of hexachlorocyclotriphosphazene (HCCP) with p-phenylenediamine (DA), 1,4-benzenedithiol (DT), and 4-aminothiophenol (AT), producing poly(p-phenylenediamine-co-cylotriphosphazene) (PDA) with N-redox active-sites, poly(1,4-benzenedithiol-co-cylotriphosphazene) (PDT) containing S-redox active-sites, and poly(4-aminothiophenol-co-cylotriphosphazene) (PAT) holding dual N-S-redox active-sites. The PDA, PDT, and PAT in the N-bromosuccinimide/pyridine (NBS/Py) leaching solution demonstrated extraction efficiency of 83.7 % Au 3+ , 45.5 % Cu 2+, 44 % Ni 2+ , and 83.6 % Au 3+ , 47.4 % Cu 2+ , 48 % Ni 2+ , and 83.1 % Au 3+ , 48.5 % Cu 2+ , 48.8 % Ni 2+ . While, in the aqua regia, these adsorbents showed extraction efficiency of 70.0 % Au 3+ , 14.0 % Cu 2+ , 13.8 % Ni 2+ , and 92.3 % Au 3+ , 6.4 % Cu 2+ , 9.8 % Ni 2+ , and 95.6 % Au 3+ , 13.0 % Cu 2+ , 10.3 % Ni 2+ . In particular, the PAT with the dual N-S-redox active sites achieved maximal Au 3+ extraction efficiency (99.2 %), which is 1.05 and 1.04 times higher than PDA and PDT within 15 min, superb uptakes of 2000 mg/g, excellent selectivity (99.9 %) in a mixture of eight metal ions (Al, Co, Cu, K, Mg, Na, Ni, Mn), 100 % removal capacity and high stability even in pure aqua regia leaching solution. Gold ions adsorbed on these adsorbents, spontaneously reduced, experienced nucleation, and grew into single crystalline nanoplates along (111) planes as well as nanolayers composed of Au-single-atoms and Au-nanoclusters. The density function theory (DFT) calculations and variation in binding energies of N and S indicated that due to the synergy of dual N-S-redox active-sites, the PAT showed enhanced performances and excellent selectivity. For commercial applications, the as-synthesized PDA, PDT, and PAT were highly stable in pure aqua regia leaching solution, showing precisely selective and rapid gold extraction (70 %, 92 %, 96 %, respectively) than N- NBS/Py leaching systems (83 %) with traces of other competing metals. This study proves that skeleton-modulated poly(organo)phosphazene greatly facilitates gold extraction for industrial-scale applications.

Periplocymarin (PPM), a cardiac glycoside, has a narrow therapeutic index, poor tumor selectivity and severe cardiovascular toxicity which hinder its wide clinical applications in cancer treatment. Herein, we report novel redox-responsive prodrug-nanoparticles (MPSSV-NPs) self-assembled by co-nanoprecipitation of PPM-vitamin E conjugate and a PEG derivative of linoleate (mPEG2000-LA) in water. It was found that the characteristics of PPM-vitamin E nanoparticles (PSSV-NPs) were improved through co-nanoprecipitation with increased percentages of mPEG2000-LA. Moreover, the MPSSV-NPs were optimized according to the in vitro release and cytotoxicity study. Furthermore, the optimized MPSSV-NPs dramatically enhanced the circulation time and tumor distribution of PSSV-NPs after single intravenous injection. The in vivo studies in malignant H22-bearing mice revealed that MPSSV-NPs could effectively suppress tumor growth without causing obvious systemic toxicity. Altogether, these results suggested that MPSSV-NPs could offer a safe, multifunctional and viable nanoplatform for cardiac glycosides in cancer treatment.

Emulsion gel is widely used in the delivery of functional components because of its network structure and mechanical properties. In this study, to improve the structural and functional properties of emulsion gels and achieve efficient encapsulation and release of functional components, transglutaminase (TGase) and genipin were employed to crosslink pH-shifting/ultrasonication-modified soybean lipophilic protein (SLP), enabling the simultaneous encapsulation of vitamin E (VE, oil phase) and quercetin (QU, aqueous phase), thereby developing a co-encapsulated emulsion gel system. The impacts of combined modification (pH-ultrasonication) and enzymatic/chemical crosslinking on gel properties were systematically investigated through gel strength analysis, rheological characterization, intermolecular interaction profiling, microstructural observation, and in vitro digestion evaluation. Results demonstrated that pH-shifting combined with ultrasonication exposed more hydrophobic groups in SLP, thereby enhancing intermolecular hydrophobic interactions. Microstructural analysis revealed that TGase and genipin synergistically strengthened network crosslinking density, forming a more compact gel network structure under modified conditions. Furthermore, TGase/genipin crosslinking significantly improved structural characteristics and rheological performance (e.g., water-holding capacity increased by 14.97 %, hardness and viscosity improved), while concurrently enhancing encapsulation efficiencies of both VE and QU. In vitro digestion test confirmed that the maximum release rate reached 97.43 ± 1.50 % (VE) and 90.39 ± 2.43 % (QU) ( p < 0.05). These findings elucidated the potential for constructing emulsion gels via combined modification and crosslinking strategies, providing theoretical guidance for co-encapsulation systems in functional factors delivery applications.

Alcohols can be extensively used in the production of fine chemicals, such as fuel additives, food supplements and pharmaceutical intermediates, which can be acquired by the reduction of biomass-derived aldehyde. In this study, the recombinant Escherichia coli KPADH was genetically constructed through the co-expression of alcohol dehydrogenase from Kluyveromyces polysporus and glucose dehydrogenase from Bacillus subtilis . Biomass-derived aldehydes were biologically converted into alcohols in a benign system. In deep eutectic solvent (DES) choline chloride:glycerol-H 2 O (choline chloride:glycerol, 10 wt %) containing E. coli KPADH cells (0.1 g/mL), HMF (50 mM), glucose (2 equivalents), and NAD + (0.1 μM/mM HMF), bioreduction of HMF into 2,5-bis(hydroxymethyl)furan was implemented, acquiring a high yield of 98.1 % within 8 h. Other biobased compounds, such as isovanillin, vanillin, p -hydroxybenzaldehyde, furfural, and benzaldehyde were also converted into their corresponding alcohols. According to the molecular docking results, HMF had the lowest CDOCKER energy value of -27.422 kcal/mol. It was found that HMF displayed strong binding affinity with E. coli KPADH, resulting in enhanced binding stability and superior catalytic reduction activity. This study demonstrated a green and highly effective biocatalytic method for the synthesis of biobased alcohols.

In addressing the detection of drug resistance inHelicobacter pylori, we have successfully developed an efficient and highly accurate detection methodology. Initially, we designed and fabricated a microarray chip, which underwent finite element analysis for its optical and thermal characteristics. Ultimately, COC material was chosen as the processing material for the chip, ensuring superior performance. Subsequently, we established a comprehensive detection system and validated its performance. Following that, comparative experiments were conducted for detecting drug resistance inH. pylori. The experimental results indicate that our established methodology aligns with the results obtained using the E-test detection kit, achieving a concordance rate of 100%. In comparison to the E-test detection kit, our methodology reduces the detection time to 1.5 h and provides a more extensive coverage of detection sites.

Hydrophobic organic compounds (HOCs) in e-waste disposal sites are difficult to remove effectively. There is little reported about zero valent iron (ZVI) coupled with persulfate (PS) to achieve the removal of decabromodiphenyl ether (BDE209) from soil. In this work, we have prepared the flake submicron zero valent iron by ball milling with boric acid (B-mZVI bm ) at a low cost. Sacrifice experiments results showed that 56.6% of BDE209 was removed in 72 h with PS/B-mZVI bm , which was 2.12 times than that of micron zero valent iron (mZVI). The morphology, crystal form, atomic valence, composition, and functional group of B-mZVI bm were determined by SEM, XRD, XPS, and FTIR, and the results indicated that the oxide layer on the surface of mZVI is replaced by borides . The results of EPR indicated that hydroxyl radical and sulfate radical played the dominant role in the degradation of BDE209. The degradation products of BDE209 were determined by gas chromatography-mass spectrometry (GC-MS), accordingly, the possible degradation pathway was further proposed. The research suggested that ball milling with mZVI and boric acid is a low-cost means of preparing highly active zero valent iron materials. And the mZVI bm has promising applications in improving the activation efficiency of PS and enhancing the removal of the contaminant.
