Synergistic indicator labels containing bromothymol blue/red carbon dots for visual real-time fish freshness sensing via colorimetric/fluorescent dual-modal signaling

Lei Zhang,Yuanyuan Jiang,Yufang He,Zili Tang,Mao Zhang,Caiyi Qin,Hongdong Liu,Tao Le

Food freshness indicator labels are designed to assist consumers in identifying food spoilage. This study developed a colorimetric and fluorescent dual-channel indicator label for real-time, visual, and synergistic monitoring of fish freshness, using bromothymol blue (BTB) and red carbon dots (RCDs) as response signals and carboxymethyl cellulose (CMC) as the matrix (CMC-BTB/RCDs). It exhibits significant color and fluorescence responses in the pH range of 5–9 and volatile amine concentrations of 0.05–20 mg/mL. Under daylight, the label transitions from yellow-brown to blue-green (color difference ΔE = 60.47); under 365 nm ultraviolet light, its red fluorescence gradually decreases while the Blue/Red fluorescence (BRF) increases to 5.64. These changes originate from volatile amines released during fish spoilage, which alter the storage environment's pH and changes BTB's molecular structure. This structural change not only triggers the label's color shift but also adjusts its fluorescence intensity via the inner filter effect between BTB and RCDs. In practical use, CMC-BTB/RCDs enables precise, real-time, and non-destructive monitoring of fish freshness at 25 °C and 4 °C, with the label's ΔE and BRF showing a strong correlation with fish total volatile basic nitrogen. Therefore, CMC-BTB/RCDs showed great potential for application as smart packaging in monitoring fish freshness.

A double-network fish gelatin/sodium alginate composite hydrogel as a carrier for the sustained release of a soy-derived osteogenic peptide

Jinpeng Gong,Boya Liu,Tao Huang,Lu Wei,Yupeng Ma,Hao Chen,Jing Gan,Junbo Ge

Osteoporosis, a prevalent metabolic bone disease, poses a significant challenge for bone repair in aged populations. Soy-derived osteogenic peptide (SOP) holds promise but its clinical translation is hampered by rapid enzymatic degradation and poor oral bioavailability, representing a major challenge that necessitates the development of a protective delivery system. To address this, we developed a novel composite hydrogel based on fish gelatin and sodium alginate (SA/FG) as an oral delivery vehicle to overcome these limitations. Systematic characterization revealed that the SA/FG formulation offers distinct advantages: the incorporation of an optimal amount of SA significantly enhanced the mechanical strength and stability of the hydrogel, addressing the common weakness of single-network gelatin hydrogels, while maintaining a high water content (~90%) and a superior rehydration capacity (83.65% in distilled water). In a glucocorticoid-induced zebrafish osteoporosis model, the SOP-loaded SA/FG hydrogel significantly increased skull bone mass by 120.17% and improved larval swimming behavior, markedly outperforming the free SOP group. This study presents the first report of an SA/FG composite hydrogel for effective oral delivery of an osteogenic peptide. The system demonstrates great potential as a functional food or supplement for preventing and treating osteoporosis, offering an innovative strategy to enhance peptide stability and bioavailability through a biocompatible, double-network carrier.

Determination of polyphosphates in fish and shrimp muscles by capillary electrophoresis with indirect UV detection after phosphatase inhibition using high pressure pretreatment

Li Wang,Juan Li,Li Zhang

A method was proposed for the determination of polyphosphates (P 2 O 7 4− , P 3 O 10 5− and (PO 3 ) 3 3− ) in fish and shrimp muscles by capillary electrophoresis with indirect UV detection after phosphatase inhibition using high pressure pretreatment (HPP). Separation parameters were optimized and the effects of HPP on deactivation of enzyme were investigated. Well resolution was obtained with 10   mM adenosine 5′-phosphate (ATP) in 16   mM disodium hydrogen phosphate combining with 32   mM citric acid buffer (pH   =   6.4) containing 0.1   mM cetyltrimethyl ammonium bromide, and phosphatase was effectively inhibited by HPP at 400   MPa for 10   min. Method was validated with LOD (0.23–0.33   mg   g −1 ) and LOQ (0.77–1.0   mg   g −1 ). Recovery of P 2 O 7 4− , P 3 O 10 5− and (PO 3 ) 3 3− in the fortified samples were in the range of 91.6–100.1%, 97.6–104.6% and 79.1–88.1%, respectively, and the corresponding RSD ranged from 1.1% to 6.8%. Finally, the method was applied in the determination of polyphosphates in food samples collected from local markets.

Sodium Alginate Modification as a Promising Strategy for Inducing Oral Immune Tolerance to Fish Gelatin-Based Cell Scaffolds to Control Allergenic Risks

Shuya Wang,Mengqing Shi,Ya Li,Lei Qin,Shuqi Jia,Xiuhan Chen,Siqi Hou,Songyi Lin,Na Sun

Abstract The allergenic risks of fish gelatin-based cell scaffolds remain an unresolved issue. The study proposed a sodium alginate modification strategy to solve allergenic risks by inducing oral immune tolerance. The strategy significantly reduced antibody production, mast cell degranulation, inflammatory mediator release, and Th2 immune response. Peptidomics and bioinformatics prediction revealed that the strategy protected CD4+ T cell epitopes (FTGASGLPG) and CD8+ T cell epitopes (e.g., GATGPTGL). Additionally, intestinal barrier function, integrity, and Th1/Th2/Th17/Treg immune balance were restored. Notably, compared with the positive control group, RORγt expression was suppressed by 73.42%, and FOXP3 expression was increased by 5.34-fold in the 1% group. Gut microbiota analysis suggested that sodium alginate modification significantly downregulated the abundance of Faecalibaculum and Lactobacillus, while increasing Prevotella. It regulates the intestinal microbiota, thereby contributing to immune tolerance induction. Overall, sodium alginate modification represents a promising strategy to promote immune tolerance.

Effect of the non-covalent complex of anthocyanins and whey protein on fish oil emulsions: Evaluation of stability, antioxidant properties, and bioaccessibility

Chen Li,Chao Ai,Boyu Chen,Yanan Zhao,Hui Wang,Lei Chen,Hui Teng

The nutritional efficacy of fish oil is significantly limited by its high susceptibility to oxidative degradation and low bioavailability. In this study, a non-covalent complex composed of anthocyanins (ANC) and whey protein (WP), was rationally designed and utilized to develop a fish oil emulsion system aimed at enhancing oxidative stability and bioaccessibility. The fluorescence spectroscopy (INF), Fourier transform infrared spectroscopy (FTIR) and Far UV circular dichroism (CD) demonstrated specific molecular interactions were generated between ANC and WP. Upon emulsification, the WP-ANC complex at 0.4 wt% acted as an effective emulsifier, yielding emulsions with reduced droplet size and enhanced antioxidative stability. Comparative analysis revealed that WP-ANC stabilized emulsions exhibited superior inhibition of lipid oxidation compared to those stabilized solely by WP, thereby significantly enhancing the stability of fish oil. Under simulated in vitro gastrointestinal digestion, the emulsion formulated with 0.4 % ANC and WP promoted lipid droplet breakdown, suppressed oxidative degradation during digestion, decreased the release rate of free fatty acids, and consequently enhanced the bioavailability of fish oil.

Mitochondrial oxidative stress inhibited Sirt3/Foxo3/PPARα pathway and aggravated copper and zinc co-deficiency-induced hepatic lipotoxicity in a fish model

Zhong Chong-Chao,Zhang Xu,Pantopoulos Kostas,Song Chang-Chun,Yang Hong,Wei Xiao-Lei,Luo Zhi

Copper (Cu) and zinc (Zn) are essential trace elements for terrestrial animals. Studies suggested that single dietary Cu- or Zn- deficiency lead to liver lipid deposition and cause metabolic dysfunction-associated steatotic liver disease (MASLD). However, the mechanisms by which dietary Cu and Zn co-deficiency promote hepatic lipid deposition and metabolism remain unknown. Our study found that, compared to single Cu- or Zn- deficiency, Cu and Zn co-deficiency significantly increased lipid deposition, induced mitochondrial oxidative stress, disrupted mitochondrial structure and function, and inhibited lipolysis in hepatocytes and whole liver tissue. Mechanistically, Cu and Zn co-deficiency-induced lipotoxicity was mediated by sirtuin 3 (Sirt3). Sirt3 inhibited the acetylation and degradation of forkhead box O3 (Foxo3), preventing its binding to the peroxisome proliferator-activated receptor alpha (PPARα) promoter, which accordingly increased lipid deposition. Overall, for the first time, we elucidated the mechanism by which Cu and Zn co-deficiency aggravates hepatic lipotoxicity and identified the critical regulatory role of the Sirt3/Foxo3/PPARα pathway during this process, with the fish as the model.

Allicin-mediated protection of myofibrillar proteins in refrigerated mandarin fish (Siniperca chuatsi): Molecular interactions with endogenous proteases and inhibition of protein degradation

Zhouyi Xiong,Mingqian Zhou,Jihui Wang,Bo Wang,Xuan Chen,Shan Xiao

Endogenous proteases accelerate myofibrillar protein (MP) degradation and quality loss in fish. This study investigated allicin's protective effects on refrigerated mandarin fish ( Siniperca chuatsi ). Allicin treatment significantly inhibited cathepsin B, cathepsin L, and calpain activities and reduced MP degradation, leading to lower MFI values and slower accumulation of TCA-soluble peptides. Molecular docking and enzyme assays demonstrated that allicin binds to key catalytic residues of these endogenous proteases, suggesting a multi-target inhibitory mechanism. SDS-PAGE and histological staining further confirmed slower degradation of MPs and collagen, along with improved maintenance of muscle fiber integrity. CD and intrinsic fluorescence confirmed allicin attenuated α-helix loss, delayed tryptophan exposure, and stabilized sulfhydryl groups, indicating protection against oxidative and conformational damage. Overall, allicin inhibits endogenous proteases and preserves protein conformation, highlighting its potential as a natural preservative to improve fish storage quality.

Inspired with fish scale to manufacture biomimetic MXene derivative for the reinforcement on tribological and mechanical properties of PPS fabric/phenolic composites

Yaohui He,Zhaozhu Zhang,Yanling Wang,Meng Liu,Junya Yuan,Peilong Li,Mingming Yang,Weimin Liu

In recent years, the development of high-performance fabric composites has attracted widespread interest, but the frictional heat accumulation of fabric composites under high-speed has resulted in frictional failure, which has hindered the application of fabric composites. To remedy this deficiency, inspired by multilayer structure of fish scales, biomimetic MXene derivative DOPO-HQ-IPTS@Ti 3 C 2 T x was manufactured in situ, which boosted interfacial compatibility with MXene and provided remarkable enhancement in terms of mechanical performances and tribological properties of fabric composites. The tensile strength of reinforced fabric composites is enlarged by 45%. In addition, the wear rate of the reinforced fabric composites is reduced by 77%. Hence, the biomimetic nanomaterial DOPO-HQ-IPTS@Ti 3 C 2 T x offers a simple and dependable method for obtaining fabric composites with superior overall properties.

Dietary sodium butyrate supplementation improves fish growth, intestinal microbiota composition, and liver health in largemouth bass (Micropterus salmoides) fed high-fat diets

Weijun Chen,Shiyang Gao,Kuo Chang,Xiaoyu Zhao,Ben Niu

The present study evaluated the effect of dietary sodium butyrate (NaBT) supplementation on the growth performance, intestinal microbiota composition, and liver health of largemouth bass Micropterus salmoides fed high-fat diets. Four isonitrogenous and isolipid diets were formulated: a high-fat diet and the high-fat diet supplemented by 0.05% NaBT, 0.1% NaBT, or 0.2% NaBT. Each diet was randomly distributed to triplicate tanks (200 L ) containing 30 fish (initial weight 4.0 ± 0.1 g) per tank. The experimental fish were fed to apparent satiation twice daily for 60 days. The orthogonal polynomial contrasts showed that the weight gain rate (WGR) and specific growth rate (SGR) increased linearly ( P  < 0.05) with increasing dietary NaBT levels (P  < 0.05). No significant differences were found for feed intake (FI), feed conversion ratio (FCR), condition factor (CF), viscerosomatic index (VSI), and hepatosomatic index (HSI). There were no differences in the alpha diversity indices (including Operational Taxonomic Units, Chao index, and Simpson index) of intestinal microbiota between the various dietary groups ( P  > 0.05). For the intestinal microbiota composition, the proportion of Proteobacteria were linearly and cubically increased, whereas the proportion of Tenericutes and Mesomycoplasma were linearly and cubically decreased with the increase of dietary NaBT levels ( P  < 0.05). The increasing dietary NaBT levels led to significantly quadratic decrease of alanine aminotransferase (ALT) ( P  < 0.05), linear and quadratic decrease of aspartate aminotransferase (AST) ( P  < 0.05), and linear, quadratic, and cubic decrease of diamine oxidase (DAO) ( P  < 0.05). Hepatic triglyceride (TG) and total cholesterol (TCHO) contents decreased linearly and quadratically with the increase of dietary NaBT levels ( P  < 0.05). Increasing dietary NaBT levels linearly, quadratically, and cubically decreased the malondialdehyde (MDA) contents ( P  < 0.05). Total superoxide dismutase (T-SOD) and glutathione peroxidase (GPx) activity levels increased linearly with the increasing dietary NaBT levels ( P  < 0.05). As the dietary NaBT levels gradually increased, the relative expression of carnitine palmitoyl transferase (CPT1) and peroxisome proliferator-activated receptor α (PPARα) increased linearly, quadratically, and cubically ( P  < 0.05), while the relative expression of sterol regulatory element binding protein 1 (SREBP 1) decreased linearly, quadratically, and cubically ( P  < 0.05). The relative expression of tumor necrosis factor α (TNF-α) and cysteinyl aspartate specific protease 3 (Caspase 3) decreased linearly and quadratically with the increasing NaBT levels ( P  < 0.05). Pearson correlation analysis showed that fish growth was closely correlated with microbiota composition, hepatic lipid accumulation, hepatic antioxidant-related parameters, and hepatic gene expression ( P  < 0.05). These results indicated that NaBT could be used to alleviate the negative influence of high-fat diets on fish growth, intestinal microbiota homeostasis , and liver health.

Inhibitory effects of some hydrocolloids on the formation of NƐ-(carboxymethyl) lysine and NƐ-(carboxyethyl) lysine in chemical models and fish patties

Xin Xu,Tian Xue,Qingqing Jiang,Daming Fan,Mingfu Wang,Yueliang Zhao

The inhibitory effects of four hydrocolloids, including alginic acid, pectin, carboxymethylcellulose sodium salt, and chitosan on the formation of two typical toxic advanced glycation end products N Ɛ -(carboxymethyl) lysine (CML) and N Ɛ -(carboxyethyl) lysine (CEL) were investigated in chemical models and fish patties. All the four hydrocolloids were found to significantly inhibit CML and CEL formation in chemical models, and among them, pectin and alginic acid were the most effective inhibitors. In roast fish patties, the addition of pectin and alginic acid at 0.9% reduced the content of CML by 43.6% and 44.9%, and CEL by 32.4% and 32.6%, respectively. Moreover, alginic acid plus a polyphenol epigallocatechin gallate (EGCG) showed stronger inhibitory effect against CML and CEL formation than alginic acid alone. Further study showed that alginic acid and pectin decreased the formation of glyoxal (GO) and methylglyoxal (MGO), two key intermediates of CML and CEL, in the chemical models and fish patties. In addition, compared with EGCG, the addition of alginic acid in fish patties showed less impact on color, odor, and sensory properties. These findings suggest that alginic acid is a potent inhibitor against the formation of AGEs with less impact on food quality.

Bimetallic Au@Pt Nanocrystal Sensitization Mesoporous α-Fe2O3 Hollow Nanocubes for Highly Sensitive and Rapid Detection of Fish Freshness at Low Temperature

Jiabin Shen,Shanshan Xu,Cheng Zhao,Xiaopeng Qiao,Haiquan Liu,Yong Zhao,Jing Wei,Yongheng Zhu

In this work, we present a new metal oxide semiconductor gas sensor for detecting trimethylamine (TMA) by bimetal Au@Pt-modified α-Fe2O3 hollow nanocubes (NCs) as sensing materials. The structure and morphological characteristics of Au@Pt/α-Fe2O3 were evaluated through multiple analyses, and their gas-sensitive performance was investigated. Compared with the pristine α-Fe2O3 NC sensor, the sensor based on Au@Pt/α-Fe2O3 NCs exhibited faster response time (5 s) and higher response (Ra/Rg = 32) toward 100 ppm TMA gas at a lower temperature (150 °C). Furthermore, we also assessed the Au@Pt/α-Fe2O3 NC sensor for detecting the freshness of Larimichthys crocea which have been observed by headspace solid-phase microextraction and gas chromatography–mass spectrometry. The high performance of the Au@Pt/α-Fe2O3 NCs is attributed to the special hollow morphology with a high specific surface area (212.9 m2/g) and the synergistic effect of the Au@Pt bimetal. The Au@Pt/α-Fe2O3 sensor shows promising application prospects in estimating seafood freshness on the spot.

Au/Ag Bimetallic Nanocuboid Superlattices Coated with Ti3C2 Nanosheets for Surface-Enhanced Raman Spectroscopy Detection of Fish Drug Residues in Pond Water

Yuhui Miao,Kuo Yang,Shenfei Zong,Zhuyuan Wang,Yiping Cui

Applying nanomaterials to highly sensitive, fast, and accurate detection of hazardous chemicals in water remains to be a great challenge. Herein, a Ti3C2-functionalized superlattice SERS substrate was developed, which can identify multiplex fish drug residues in pond water with a high sensitivity and reliable reproducibility. Specifically, a superlattice substrate was fabricated by vertically self-assembled bimetallic nanocuboids (BNCs) through the Marangoni effect, which exhibited an excellent SERS activity. Furthermore, the modification of the Ti3C2 layer endows the substrate with both an increased SERS signal and an enhanced adsorption for multiple fish drugs, including crystal violet (CV), malachite green (MG), and methylene blue (MB). The limit of detection (LOD) was as low as 10–12 M, while the signal fluctuation was as small as ∼5.9% between batches. More importantly, a trace amount of multiplex fish drug residues was detected in natural pond water, demonstrating a convincing practicality of such a Ti3C2-functionalized superlattice SERS substrate.
