Alginate/PEG based microcarriers with cleavable crosslinkage for expansion and non-invasive harvest of human umbilical cord blood mesenchymal stem cells

Chunge Li,Yufeng Qian,Shuang Zhao,Yuji Yin,Junjie Li

Porous microcarriers are increasingly used to expand and harvest stem cells. Generally, the cells are harvested via proteolytic enzyme treatment, which always leads to damages to stem cells. To address this disadvantage, a series of alginate/PEG (AL/PEG) semi-interpenetrating network microcarriers are prepared in this study. In this AL/PEG system, the chemically cross-linked alginate networks are formed via the reaction between carboxylic acid group of alginate and di-terminated amine groups of cystamine. PEG is introduced to modulate the degradation of microcarriers, which does not participate in this cross-linked reaction, while it interpenetrates in alginate network via physical interactions. In addition, chitosan are coated on the surface of AL/PEG to improve the mechanical strength via the electrostatic interactions. Biocompatible fibronectin are also coated on these microcarriers to modulate the biological behaviors of cells seeded in microcarriers. Results suggest that the size of AL/PEG microcarriers can be modulated via adjusting the contents and molecular weight of PEG. Moreover, the microcarriers are designed to be degraded with cleavage of disulfide crosslinkage. By changing the type and concentration of reductant, the ratio of AL to PEG, and the magnitude of chitosan coating, the degradation ability of AL/PEG microcarriers can be well controlled. In addition, AL/PEG microcarriers can support the attachment and proliferation of human umbilical cord blood mesenchymal stem cells (hUCB-MSCs). More importantly, the expanded hUCB-MSCs can be detached from microcarriers after addition of reductant, which indeed reduce the cell damage caused by proteolytic enzyme treatment. Therefore, it is convinced that AL/PEG based microcarriers will be a promising candidate for large-scale expansion of hUCB-MSCs.

Enhanced interfacial interactions of carbon fiber/epoxy resin composites by regulating PEG-E51 and graphene oxide complex sizing at the interface

Xiaofeng Chi,Muxuan Li,Mei Liang,Yang Chen,Huawei Zou

A method of using modified epoxy resin (PEG-E51) and graphene oxide (GO) as sizing agent in different pH has been proposed and applied to improve the interfacial interactions and bonding of carbon fiber (CF) reinforced epoxy resin-based (EP) composites. The PEG-E51 and GO complex sizing in different pH is prepared and then coated on CF surfaces homogeneously. The sizing layer forms on the fiber surfaces, and multiple GO sheets are introduced successfully surrounding the CFs. The surface morphologies of CFs change distinctly with different pH. The interfacial shear strength increases from 51.36 MPa for bare fiber-reinforced EP composites to 66.62 MPa for composites reinforced by CFs coated with GO only. However, a significant improvement is achieved when the pH is adjusted to 10, making the interfacial shear strength grow up to 77.23 MPa. Furthermore, scanning electron microscopy results and interlaminar shear strength test are in agreement with each other, suggesting better interface bonding of composites by regulating pH of PEG-E51 and GO complex sizing. Besides, the interfacial interaction mechanism in CF reinforced composites is also explored, that is, the positive effects of roughness and specific surface area in complex interfacial layers lead to the improvement of interfacial properties.

RGD/TAT-functionalized chitosan-graft-PEI-PEG gene nanovector for sustained delivery of NT-3 for potential application in neural regeneration

Dongni Wu,Yongnu Zhang,Xiaoting Xu,Ting Guo,Deming Xie,Rong Zhu,Shengfeng Chen,Seeram Ramakrishna,Liumin He

In this study, we prepared a multifunctional gene delivery nanovector containing a chitosan (CS) backbone and polyethylenimine (PEI) arms with arginine-glycine-aspartate (RGD)/twin-arginine translocation (TAT) conjugated via polyethylene glycol (PEG). Branched PEI, with a molecular weight of 2000 Da, was used to achieve a balance between biocompatibility and transfection efficiency, whereas RGD/TAT peptides were conjugated for enhanced targeting ability and cellular uptake. Synthesis of the copolymers was confirmed by characterizing the chemical structure with 1 H nuclear magnetic resonance and Fourier Transform Infrared Spectroscopy (FTIR). The nanovector was biocompatible with cells and showed excellent capability for DNA condensation ; the resulting complexes with DNA were well-formed, and possessed small particle size and reasonable positive charge. Higher gene transfection efficiency, compared to that achieved with PEI (25 kDa), was confirmed in tumor (HeLa cells) and normal cells (293T and NIH 3T3 cells). More importantly, the cells transfected with the chitosan- graft -PEI-PEG/pCMV-EGFP-Ntf3 complex produced sustained neurotrophin-3 with a linear increase in cumulative concentration, which induced neuronal differentiation of neural stem cell and promoted neurite outgrowth . These findings suggested that our multifunctional copolymers might be ideal nanovectors for engineering cells via gene transfection, and could potentially be applied in tumor therapy and regenerative medicine. Statement of Significance We successfully prepared a multifunctional gene delivery nanovector containing branched PEI with a molecular weight of 2000 Da to balance between biocompatibility and transfection efficiency, and RGD/TAT peptides for enhanced targeting ability and cellular uptake. The well-formed CPPP/DNA complexes of small particle size and reasonable positive charges potentially enhanced gene transfection in both tumor and normal cells. More importantly, the CPPP/pCMV-EGFP-Ntf3 complex-transfected 293T cells could produce sustained NT-3 with a constant ratio, which induced neuron differentiation of NSC and promoted neurite outgrowth. Therefore, our study provided an effective strategy for producing neurotrophins by engineering cells with gene delivery, which deserved wide investigation and potential application in regenerative medicine.

pH-sensitive magnetic drug delivery system via layer-by-layer self-assembly of CS/PEG and its controlled release of DOX

Juan Wu,Xi Wang,Binglong Zhu,Qinting He,Fang Ren,Fei Tong,Wei Jiang,Xianghong He

Cancer is one of the biggest killers threat to human life and health and is still difficult to treat mainly due to the lack of targeted drug delivery stages and limitations associated with delivering drugs to targeted cancer tissues. The goal of this work was to develop a magnetic, pH-sensitive formulation for targeted delivery of chemotherapeutic agents to cancer cells. Then the functional drug delivery system (Fe3O4@CS/PEG-DOX) was synthesized by the layer-by-layer (LbL) self-assembly technique. And the drug loading content is calculated to be 19.1%. In addition, the Fe3O4@CS/PEG-DOX exhibited excellent pH-sensitivity, 73.1% DOX was released within 48 h at pH 4.0. Furthermore, all the release behaviors fit the Higuchi model very well and the dissolution of CS/PEG layers played a key role on DOX release from Fe3O4@CS/PEG-DOX. The results of toxicity analysis in human liver hepatocellular carcinoma cells (HepG2) revealed that Fe3O4@CS/PEG-DOX exhibited high anti-tumor activity, while the Fe3O4@CS/PEG nanocomposites were practically non-toxic. Therefore, all the results demonstrated that the Fe3O4@CS/PEG-DOX could have an important impact on the development of targeted intracellular delivery nanodevices for cancer therapy.

A new poly(I:C)-decorated PLGA-PEG nanoparticle promotes Mycobacterium tuberculosis fusion protein to induce comprehensive immune responses in mice intranasally

Xiufen Du,Daquan Tan,Yang Gong,Yifan Zhang,Jiangyuan Han,Wei Lv,Tao Xie,Pu He,Zongjie Hou,Kun Xu,Jiying Tan,Bingdong Zhu

Protein-based subunit vaccine against tuberculosis (TB) is regarded as safer but with lower immunogenicity . To investigate effective adjuvant to improve the immunogenicity of TB subunit vaccine, we modified ploy(I:C) onto PLGA-PEG copolymer nanoparticle with polydopamine to produce a new nanoparticle adjuvant named “PLGA-PEG-poly(I:C)” (NP). M. tuberculosis fusion proteins Mtb10.4-HspX and ESAT-6-Rv2626c (M4) were encapsulated in the nanoparticles to produce the NP/M4 subunit vaccine. The PLGA-PEG/M4 nanoparticle was 200.21 ± 1.07 nm in diameter, and the polydispersity index (PDI) was 0.127 ± 0.02. Following modification with poly(I:C) by polydopamine, the NP/M4 was administered to C57BL/6 female mice intranasally and the immune responses were evaluated. The NP/M4 significantly induced antigen-specific CD4 + T cells proliferation, IL-2 and IFN-γ production. In addition, the NP/M4 could promote the production of antigen-specific IgG, IgG1 , IgG2c in serum, and sIgA in lung washings . Overall, our results indicated that the NP would be a potential TB subunit vaccine adjuvant with the ability to induce strong Th1-type cell-mediated immunity and humoral immune responses .

Comb-shaped polymer grafted with REDV peptide, PEG and PEI as targeting gene carrier for selective transfection of human endothelial cells

Haixia Wang,Qian Li,Jing Yang,Jintang Guo,Xiangkui Ren,Yakai Feng,Wencheng Zhang

If plasmid complexes prepared from targeting carriers have endothelial cell selectivity and high transfection efficiency, they can specifically enhance rapid endothelialization by delivering the corresponding gene plasmids. A high content of functional groups in the carriers benefits high selectivity efficiency. Herein, we have synthesized a comb-shaped polymer bearing several Arg-Glu-Asp-Val (REDV) peptides and poly(ethylene glycol) as a pEGFP-ZNF580 gene carrier with cell-type recognition of human endothelial cells. An amphiphilic block copolymer of poly(2-hydroxyethyl methacrylate)-block-poly(ε-caprolactone)-graft-poly(ethylene glycol)-graft-poly(ethyleneimine) conjugated with REDV peptide (PHEMA-b-PCL-g-PEG-g-PEI-REDV) is synthesized, and nanoparticles of it are prepared by polymer self-assembly. This polycationic PHEMA-b-PCL-g-PEG-g-PEI-REDV carrier effectively condenses pEGFP-ZNF580 plasmid to form REDV-targeted complexes and protects pEGFP-ZNF580 integrity from enzymatic hydrolysis. These REDV-targeted complexes exhibit low cytotoxicity but high transfection efficiency to EA.hy926 cells compared with non-targeted complexes (PHEMA-b-PCL-g-PEG-g-PEI/pEGFP-ZNF580) as demonstrated by MTT assay, fluorescence-activated cell sorting analysis and fluorescence analysis. Furthermore, the relative protein level of endothelial cells transfected by REDV-targeted complexes is higher than that by non-targeted complexes. Therefore, REDV-bearing carriers may have potential as effective and targeting transfer carriers for pEGFP-ZNF580 plasmid, and their complexes can be used in the endothelialization of artificial vascular grafts.

Broadening the debinding temperature window by PEG addition to reduce defects and improve the mechanical properties of vat photopolymerized alumina ceramics

Xinkun Liu,Xiong Gao,Jingyi Chen,Xiaotong Chen,Wenqing Wang,Zhaoliang Qu,Rujie He

PEG400 broadens the debinding temperature window by ∼100 °C, enabling stepwise resin decomposition in VPP alumina. • Early PEG decomposition constructs 3D pores for efficient gas escape, suppressing delamination and cracks. • 10 vol% PEG optimizes defect control, achieving 234 MPa flexural strength (30% improvement) and 97.1% relative density. • Debinding window engineering is a generalizable strategy for defect regulation in light-cured 3D-printed ceramics. To address the critical challenges of narrow debinding window and defect formation in vat photopolymerization (VPP) 3D-printed alumina ceramics with high resin content (as high as 50 vol%), this study introduces polyethylene glycol (PEG400) as an auxiliary organic phase. The concentrated thermal decomposition of the single-resin system, typically occurring within a sharp temperature range of 300–500 °C, was transformed into a stepwise process spanning 200–500 °C by PEG pre-decomposition. This window broadening of ∼100 °C facilitated early gas release through interconnected pore channels formed at low temperatures, thereby alleviating internal pressure and stress accumulation during main resin decomposition. Quantitative X-ray computed tomography analysis confirmed significant defect suppression, with the 10 vol% PEG group (P1) achieving the lowest average defect volume and delamination fraction. Consequently, sintered samples exhibited 30% improvement in flexural strength (234 MPa) and 97.1% relative density. These results demonstrate that broadening the debinding temperature window is an effective strategy for defect control and mechanical property enhancement in VPP ceramics. Download: Download high-res image (227KB) Download: Download full-size image In this study, a PEG-mediated debinding strategy is proposed to address the critical challenges of narrow debinding window and crack formation in conventional single-resin systems. In the traditional system, rapid gas release leads to internal pressure accumulation and defect generation, resulting in a mechanical strength of only 185 MPa. In contrast, PEG decomposition constructs a connected pore network, achieving efficient debinding in a broadened temperature window of 200–500 °C, significantly reducing defects, and ultimately increasing the mechanical strength to 234 MPa, providing a new efficient debinding pathway for high-performance ceramic 3D printing.

Multifunctional anisotropic PEG/P-N Co-doped rGO aerogel phase change composites: Tuning phonon transport for smart thermal and EMI management

Jinhui Hu,Minming Zou,Jue Wang,Shuo Zou,Yue Liu,Yan Ma,Wenjing Chen,Qinling Li,Yinshui He,Xiongxin Jiang,Xiaowu Hu

Addressing the urgent demand for energy conservation, efficient thermal storage, and safety protection in the modern building sector, this study fabricates a temperature-adaptive multifunctional composite phase change material (PEG/RCP). By encapsulating polyethylene glycol (PEG) within high-porosity anisotropic P/N co-doped rGO aerogels prepared via directional freezing, the composite achieves a critical balance between high latent heat (169.6 J·g −1 ) and low thermal conductivity (0.397 W·m −1 ·K −1 ). Comprehensive molecular dynamics simulations reveal that heteroatom doping induces significant phonon spectrum mismatch at the interface. This mismatch substantially enhances interfacial thermal resistance, facilitating a synergistic “heat storage-thermal insulation” effect. The composite exhibits robust flame retardancy with a 53.3% reduction in peak heat release via gas-phase dilution, condensed-phase char formation, and phase change heat absorption. Furthermore, it achieves superior electromagnetic shielding effectiveness (greater than 75 dB) dominated by absorption loss through multiple reflections within the porous framework, interfacial/dipole polarization, and conductive loss. Outdoor building thermal management tests confirm that the PEG/RCP interlayer effectively suppresses indoor temperature fluctuations and delays heat transfer, showcasing exceptional temperature-adaptive thermal regulation performance. Consequently, this work offers a promising strategy for designing advanced materials for multifunctional thermal management in next-generation smart buildings.

Preparation of PEG/P(G-AM-ChCl) Composite Hydrogel via Photo-Polymerization With Ternary Deep Eutectic Solvent and Its Performance Study

Haoyu Lv,Bin Li,Zhigang Hu,Mengjing Zhou,Jiawei Li,Longfei He,Yuzuo Liu,Ming Ma

In this study, a ternary deep eutectic solvent (DES) was prepared using glycerol (Gly), acrylamide (AM), and choline chloride (ChCl) as raw materials, and polyethylene glycol (PEG) with different mass fractions was introduced as a filler into the DES to fabricate PEG/P(G-AM-ChCl) composite hydrogels via photopolymerization. The composite hydrogels were characterized by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). The effects of PEG content on the swelling behavior, water holding capacity, mechanical properties, and fatigue resistance of the composite hydrogels were systematically investigated. The results showed that the addition of PEG promoted the formation of a denser hydrogen bonding network within the hydrogel, effectively reducing the water loss rate, and the equilibrium swelling degree of the composite hydrogels increased to 9.61 with increasing PEG content. The composite hydrogels exhibited excellent mechanical properties and fatigue resistance, with a compressive strength reaching 0.157 MPa after PEG addition, which was 1.25 times higher than that of the PEG-free group. This study provides a simple method for preparing composite hydrogels with high water holding efficiency.

A microenvironment-responsive microsphere–PLGA-PEG-PLGA hydrogel for sustained and coordinated co-delivery of celecoxib and ropivacaine in osteoarthritis therapy

Yuan Hao,Zhang Bingwen,Sun Wen,Liu Haofan,Hu Liandong

In this study, celecoxib (CXB) microspheres (CM) and ropivacaine (RPV) microspheres (RM) were prepared respectively. A poly (D, L-lactide-co-glycolide)-block-poly (ethylene glycol)-block-poly (D, L-lactide-co-glycolide) (PLGA–PEG–PLGA, PPP) was synthesized by copolymerizing glycolide and lactide with polyethylene glycol. CM, RM, and hydroxyapatite (HA) were then incorporated into PPP solution to construct the microsphere–hydrogel system (CRMH hydrogels). CM, RM, and CRMH were systematically characterized in terms of morphology, physicochemical properties, pH-buffering capacity, and degradation behavior. The pH-dependent drug release and release kinetics of the individual CM, RM and the CRMH hydrogels were also investigated. In vitro release studies demonstrated that CRMH hydrogels provided coordinated, sustained release of CXB and RPV at pH 6.2. In a rabbit osteoarthritis (OA) model, intra-articular (IA) administration of CRMH hydrogels achieved sustained drug retention and release within the joint cavity, and effectively promoted articular cartilage repair. Overall, the CRMH hydrogels showed promising synergistic therapeutic potential for OA.

MXene-enabled adsorption-photothermal synergy in starch-PEG/CaCl2 composite hydrogel: A low-cost high-efficiency sorbent for atmospheric water harvesting

Zhijin Wang,Qiongfen Yu,Ming Li,Fen Jiang,Lei Shu,Shengnan Sun,Danya Zhan,Zhongfan Mo,Zhihao Song,Runfang Ma,Meidi Ding,Hui Yao

Sorption-based atmospheric water harvesting (SAWH) offers a solution to global freshwater shortages that is not restricted by geographical conditions. However, in the current research, there are still problems such as the excessively high cost of adsorbents and the fact that photothermal materials only increase the desorption temperature during the desorption stage, without playing a role in the adsorption stage or even having a negative impact on it. This paper choses to use low-cost materials: renewable polysaccharides, potato starch (PS), bulk polymers, polyethylene glycol (PEG) as the framework, low-cost hygroscopic agent (CaCl 2 ) was employed as the moisture sorbing component, and MXene as the photothermal component to prepare high-performance sorbents materials (PPMC) through freeze-drying. MXene integration enabled dual functionality. During adsorption, it simultaneously enhanced kinetics and saturated capacity PPMC. Compared to PPC without MXene, PPMC's initial adsorption rate increased from 6.2 mg/g·min to 13.93 mg/g·min, representing an increase of 125 %, its saturated adsorption capacity increased substantially from 0.93 g/g to 1.47 g/g (59 % enhancement). During desorption, MXene's surface plasmon resonance confers exceptional photothermal conversion capability. Under 0.5 sun illumination (500 W/m 2 ), the material reaches 47 °C within 10 min and stabilizes at 71.1 °C after 2.5 h, enabling rapid vapor liberation. Water production validation using a compact system in arid environments (avg. 34.78 % RH, 19.7 °C) demonstrates successful freshwater harvesting. Post-collection analysis confirms compliance with WHO drinking water standards, which establish a cost-effective strategy for atmospheric water harvesting in resource-constrained scenarios.
